This study assessed the effects of five different transport periods on physiometabolic responses and gas exchange in ostriches. It included 138 ostriches that were assigned to five experimental groups. G1 included 78 birds that were set aside as a reference group (RG). Each one of the four remaining experimental groups included 20 ostriches, which were transported in the following manner: G1 on one occasion for a period of 1 h; G2 on one occasion for 2 h; and G3 on one occasion for 3 h; G4a was made up of the ostriches from G1 but they were shipped on a second occasion, for 2 h (making a total of 3 h); finally, G4b was that same group, but after a third transport period, on this occasion during 3 h (for a total of 6 h). Groups G2 and G3 presented the most marked blood alterations (P < 0.05), including an increase in pH, hypocapnia (27.8 ± 0.80 mmHg), hypernatremia (171.75 ± 1.84 mmol/L), hypocalcemia (0.95 ± 0.03 mmol/L), and hyperglycemia (224.05 ± 3.94 mg/dL). Also, group G3 presented the lowest hematocrit values (26.5 ± 0.47 HTC%). Therefore, transporting ostriches with no prior experience for 3 h caused the most pronounced physiometabolic changes.  相似文献

Weaning of beef calves is usually done abruptly and early compared to the natural weaning of the species, and is associated with simultaneous exposure of calves to a range of social and environmental stressors. Behavioural and physiological responses to weaning indicate detrimental effects on the welfare of these animals. The development and assessment of weaning methods aiming at reducing or avoiding this problem must be supported by scientific knowledge of the morphological, physiological and psychological mechanisms involved in the establishment, maintenance and braking of the cow-calf bond. Solutions also depend on the understanding of the various stressors associated with weaning, among which are the change in diet, cessation of nursing, separation from the dam, the change to a new spatial environment and the need for social reorganization following removal of the adults from the group. This review discusses these issues and assesses the effectiveness of the methods so far proposed for improving the welfare of beef calves during the weaning period.  相似文献

T-cells have been implicated both, in promoting and reducing viral replication during lentivirus infection. CD8+ lymphocytes are believed to be important in controlling viral load through direct killing of virus-infected cells and by secretion of inhibitory chemokines and cytokines. To evaluate the role of CD8+ T-cells in the induction and control of the primary phase of a lentivirus infection, we have used a non-T-cell tropic lentivirus, maedi-visna virus (MVV), to study the initial pathogenesis and subsequent immune responses in sheep depleted in vivo of CD8+ cells. Sheep were depleted of CD8+ cells in both blood and efferent lymph for up to 14 days. No difference in MVV replication was observed in either the draining efferent lymph or lymph node of these sheep. Surprisingly, these animals displayed a normal induction of pCTL whereas the virus-specific proliferative responses were reduced. This could reflect either that a proportion of functional CD8+ lymphocytes remained in these animals, as suggested by the appearance of pCTLs, or that CD8+ cells are not required for control of primary MVV infection.  相似文献

The human–animal relationship can influence how an animal responds to situations involving humans and affect animal well-being and production. The objectives of this research were to (1) determine if there is a relationship between the behavioral reactivity of New Zealand dairy heifers toward humans and the behavioral response to cluster placement during milking and milk production measures and (2) determine if this relationship changes as heifers become habituated to the milking process. A total of 150 primiparous heifers were randomly selected from 3 commercial dairy farms. Approximately 1 month before calving, 2 behavioral tests, avoidance distance in response to an approaching human and exit speed from a restraint, were performed on 50 heifers at each farm. For each heifer, behavioral observations were recorded during the morning milking period (during the first and sixth weeks of lactation). Behaviors recorded during cluster placement at milking included flinching, stepping, and/or kicking (FSK). Milk yields, milking durations, and average milk flow rates were recorded for each individual heifer during the first and sixth weeks of lactation. Avoidance distance and exit speed were positively correlated (P < 0.001). The FSK response during cluster placement was negatively (P < 0.01) correlated with exit speed during week 6 of lactation, but there was no relationship (P > 0.05) between avoidance distance and FSK. Avoidance distance was positively (P < 0.005) correlated with milk flow rates during weeks 1 and 6 of lactation and milk yields (P < 0.01) during week 6 of lactation. However, there was no relationship (P > 0.05) between exit speed and milk yields, durations, or flow rates. Furthermore, the FSK response was negatively (P < 0.05) correlated with milk yield during weeks 1 and 6 of lactation. Therefore, the behavioral response of heifers toward humans and the milking process appears to be related to milk production measures.  相似文献

An isotope dilution method using [U-(13)C] glucose infusion and a glucose clamp approach were applied to determine the effects of supplemental Cr and cold exposure on blood glucose turnover rate and tissue responsiveness and sensitivity to insulin in eight sheep. The daily profiles of blood metabolites and hormones were also determined. The sheep consumed diets containing either 0 or 1 mg of Cr/kg from a high-Cr yeast and were exposed from a thermoneutral environment (20 degrees C) to a cold environment (0 to 4 degrees C) for 9 d. The experiment used a crossover design. Body weight was lost (P = 0.02) during cold exposure, regardless of Cr supplementation. Blood glucose turnover rate and the maximal glucose infusion rate did not differ between diets, but both were higher (P = 0.0004 and P = 0.0001, respectively) during cold exposure than in the thermoneutral environment. The plasma insulin concentration at half-maximal glucose infusion rate changed with neither diet nor environment. Plasma concentrations of glucose and NEFA increased (P < 0.05) during cold exposure for both diets. In sheep, Cr supplementation, 1 mg/kg of diet as high-Cr yeast, has little influence on blood glucose metabolism and insulin action, whereas cold exposure enhances both without further modification by Cr supplementation.  相似文献

KRT2.13 is a type II keratin wool intermediate filament (IF) protein. Extensive variation was revealed in the 5′ untranslated region (UTR) of the ovine KRT2.13 gene (KRT2.13) using polymerase chain reaction – single strand conformational polymorphism (PCR‐SSCP) analysis. Nine unique PCR‐SSCP patterns were obtained with individual sheep having either one (homozygous), or a combination of two (heterozygous) of these patterns. Seven of the amplicons that produced the apparently homozygous patterns were successfully sequenced (GenBank FJ217670 – FJ217676), revealing eight single nucleotide insertions, 10 single nucleotide substitutions, a nucleotide deletion and a 16 nucleotide insertion that occurred in only one of the sequences. The seven sequences showed between 85% and 95% homology to the previously identified KRT2.13 sequence (GenBank X72379). This study emphasizes the power of PCR‐SSCP analysis in genotyping, as this extensive variation was found in only 100 sheep, of a variety of breeds. Since variation in the 5′UTR of genes may affect their expression, this genetic variation needs to be further studied to establish its role if any, in influencing gene expression and consequently wool traits.  相似文献

Different head–neck positions (HNPs) are used in equestrian sports and are regarded as desirable for training and competition by riders, judges and trainers. Even though some studies have been indicative of hyperflexion having negative effects on horses, this unnatural position is frequently used. In the present study, the influence of different HNPs on physical and psychological stress parameters in the ridden horse was investigated. Heart rate (HR), heart rate variability (HRV) and blood cortisol levels were measured in 18 horses. Low frequency (LF) and high frequency (HF) are power components in the frequency domain measurement of HRV which show the activity of the sympathetic and parasympathetic nervous system. Values were recorded at rest, while riding with a working HNP and while riding with hyperflexion of the horse's head, neck and poll. In addition, rideability and behaviour during the different investigation stages were evaluated by the rider and by an observer. Neither the HR nor the HRV showed a significant difference between working HNP (HR = 105 ± 22/min; LF/HF = 3.89 ± 5.68; LF = 37.28 ± 10.77%) and hyperflexion (HR = 110 ± 18; LF/HF = 1.94 ± 2.21; LF = 38.39 ± 13.01%). Blood cortisol levels revealed a significant increase comparing working HNP (158 ± 60 nm ) and hyperflexion (176 ± 64 nm , p = 0.01). The evaluation of rider and observer resulted in clear changes of rideability and behavioural changes for the worse in all parameters collected between a working HNP and hyperflexion. In conclusion, changes of the cortisol blood level as a physical parameter led to the assumption that hyperflexion of head, neck and poll effects a stress reaction in the horse, and observation of the behaviour illustrates adverse effects on the well‐being of horses during hyperflexion.  相似文献

A total of 1,936 pigs were transported through 8 trips (8.4 ± 0.5 hr) from 2 grow-to-finish farms to a commercial slaughter plant, all located in Southern Brazil. On each trip, a sub-sample of each load (36 barrows/load, weighing 118.9 ± 9.8 kg) was randomly allocated into one of the following loading densities during transport: 200 kg/m2 (D200), 235 kg/m2 (D235), and 270 kg/m2 (D270). Behavioral recordings of postures and activities were made during transportation and lairage using video-cameras. At slaughter, blood samples were collected to assess the concentrations of creatine kinase (CK) and lactate. Carcass weights and skin lesion scores were assessed on-line, and meat quality was evaluated in the longissimus thoracis (LT) muscle by assessing pH, color, and drip loss. During transportation, the proportion of animals lying down was higher (P < 0.05) in D200 and D235 groups compared with D270. The proportion of sitting animals during transportation was higher (P < 0.01) in D270 compared with D200. In lairage, D200 and D235 pigs stood more compared with D270 (P = 0.01), while the proportion of lying pigs was higher (P = 0.02) for D270 pigs compared with D200 and D235. The frequency of drinking bouts in lairage was higher (P < 0.05) for D200 group compared with D235 and D270. The levels of CK were lower (P < 0.05) in D200 pigs transported compared with D270. Lesion scores tended to be higher (P = 0.06) in D270 carcasses compared with D200 and D235. A tendency for lower (P = 0.10) pH1 values in the LT muscle of D270 pigs compared with D200 pigs was also found. Based on the results of this study, the application of lower loading densities (≤235 kg/m2) in the truck allows pigs to have sufficient space to rest, travel more comfortably and arrive less fatigued at the slaughter plant.  相似文献

Safety and efficacy of a protocol of alternating 1‐(2‐chloroethyl)‐3‐cyclohexyl‐1‐nitrosourea (CCNU; 70 mg m?2) and vinblastine (3.5 mg m?2), and prednisone (1–2 mg kg?1; CVP) in dogs with mast cell tumours (MCT) were evaluated. A total of 17 dogs had nonresectable MCTs and 35 received CVP as adjunctive treatment to locoregional control of metastatic MCTs or grade III MCTs. Neutropenia with fever occurred in 8% of dogs after treatment with vinblastine and in 2% after treatment with CCNU. Persistent elevation of serum alanine transaminase, suggestive of hepatotoxicity, occurred in 9% of the dogs. Response rate in dogs with nonresectable MCTs was 65%; five achieved a complete response (median, 141 days) and six achieved a partial response (median, 66 days). Overall median progression‐free survival (PFS) time in dogs treated in the adjuvant setting was 489 days. Dogs with grade III MCTs had shorter PFS compared with dogs with metastatic grade II MCTs (190 days versus 954 days; P < 0.001). Phase III studies are needed to provide reliable information about the comparative efficacy of this protocol.  相似文献

The effect of presenting a companion's face picture on social isolation stress was assessed by saliva sampling in cows. Three Japanese Black test cows and another companion cow were used. At 09.00 hours, one of the test cows was housed in an experimental pen with the companion in a neighboring pen. At 11.30 hours, the test cow was socially isolated by taking the companion out the neighboring pen. At 12.00 hours, the test cow was exposed to one of four treatments until 15.00 hours; presenting a blank picture (P‐Blank), a white‐ground picture of the companion's face (P‐Com) or an unfamiliar Holstein's face (P‐Hol), or the companion in the neighboring pen and a blank picture (Com). The cows were randomly exposed to all treatments. The cows' saliva were sampled for measuring salivary cortisol and chromogranin A concentrations (Sa‐Cort, Sa‐ChA) at 11.30, 12.00, 12.30 and 14.00 hours. At 12.00 hours, Sa‐ChA was higher than that at 11.30 hours (P < 0.01). At 12.30 hours Sa‐ChA of Com and P‐Com were lower than that of P‐Blank (P = 0.09, P = 0.08). At 14.00 hours Sa‐Cort of Com was lower than that of P‐Blank (P < 0.01), but Sa‐Cort of P‐Com was higher than that of P‐Blank (P < 0.01). It is suggested that P‐Com might decrease isolation stress only a short time, but afterward induced stress in cows.  相似文献

The concept of feeding anionic salts in late gestation is widely used to prevent milk fever in dairy cows. While the effects of these diets on renal Ca excretion and tissue responsiveness towards parathyroid hormone have clearly been demonstrated, data on a potential impact on gastrointestinal Ca absorption are conflicting. Therefore, the aim of this study was to investigate the influence of feeding a diet negative in dietary cation–anion difference (DCAD) on ruminal mineral concentrations, fermentation products, electrophysiological properties of rumen epithelia and Ca flux rates. For this purpose, sheep were kept for 3 weeks on diets that were either positive or negative in DCAD. The induction of a compensated hyperchloremic metabolic acidosis could be demonstrated by increased plasma Cl and enhanced concentrations of ionised Ca, while plasma concentrations of and base excess were decreased with the low DCAD diet. Neither transmural potential differences nor fermentation products were affected, but ruminal concentrations of Cl and Mg as well as the relation of ionised to total Ca were increased. Ussing chamber experiments revealed alterations of electrophysiological parameters and an increase in the electroneutral component of Ca flux rates from the mucosal to the serosal side of rumen epithelium. As plasma calcitriol concentrations were not affected, it can be concluded that the administration of anionic salts results in a vitamin D‐independent stimulation of ruminal Ca transport.  相似文献

In the Japanese macaque, semen has been collected by electro-ejaculation (EE), using the higher voltage stimuli compared to other species including genus Macaca. Semen coagulates immediately after ejaculation, which makes difficult to produce high-quality semen for artificial insemination. Recently, semen collection using urethral catheterization (UC) has been reported in carnivore and this technique may allow semen collection without coagulation in a less invasive manner. Further, the temporal preservation temperature and cooling rate of semen during cryopreservation affect post thawing sperm quality. In this study, to improve semen quality and quantity, as well as the animal welfare, semen collection was performed by EE with high (5–15 V) or low (3–6 V) voltage, UC and a combination of the two (EE-UC). It has been suggested that a high voltage is necessary for semen collection, but 10 V stimulation was effective enough and 15 V is for additional sperm collection. Also, liquid semen was collected by EE-UC and this could increase the total number of sperm. Further, to improve the post thawing sperm motility, semen was kept at four temperatures (4, 15, 25 and 37°C) for 60 min, and processed with two cooling procedures (slow cooling before second dilution and fast cooling after second dilution). Holding semen at 25°C and fast cooling after the second dilution maintained progressive motile sperm rate. The present results will contribute to the improvement of semen collection and animal welfare of Japanese macaques.  相似文献

The effect of yeast addition on intake and digestive utilization of pasture was studied in ovines under restricted time of access to forage. Eighteen wethers housed in metabolic cages and fed fresh forage (predominantly Lotus corniculatus) were randomly assigned to three treatments: forage available all day (AD); forage available only 6 h/day (R) and forage available only 6 h/day plus live Saccharomyces cerevisiae yeast (RY). Feed intake and digestibility, feeding behaviour, kinetics of passage, ruminal pH and ammonia concentration, nitrogen balance and microbial nitrogen synthesis (MNS) were determined in vivo, and ruminal liquor activity of animals was evaluated in vitro. Restricted animals consumed less than those fed all day but achieved more than 75% of the intake and spent less time ruminating (p = 0.014). Although animals without restriction consumed more feed, they had a lower rate of passage (p = 0.030). The addition of yeast did affect neither intake nor feeding behaviour, but increased digestibility. Organic matter digestibility tended to increase 11% by yeast addition (p = 0.051), mainly by a rise in NDF (27%, p = 0.032) and ADF digestibility (37%, p = 0.051). Ingested and retained N was lower in restricted animals, as MNS (p ≤ 0.045). The use of yeasts did not significantly change the N balance or MNS, but retained N tended to be higher in supplemented animals (p = 0.090). Neither ruminal pH nor ammonia concentrations were affected by the restriction, but restricted animals had a lower ruminal activity evidenced by a lower volume of gas (p = 0.020). The addition of yeast overcame this limitation, noted by a higher volume of gas of inocula from supplemented animals (p = 0.015). Yeast addition emerged as a useful tool to improve digestibility of forage cell walls in ovines under restricted time of access to forage.  相似文献
