The aim of this work was to verify whether polymorphisms in candidate genes for litter size segregate in Italian Large White (ITLW) pigs. We genotyped 120 sows that belonged to six different farms for 10 single nucleotide polymorphisms (SNPs) of 10 different genes. Polymorphisms in the chosen genes had already been associated with litter-size traits in other pig populations and were candidates for function and/or chromosomal location. The results indicated that the CLGN, pDAZL, and RFN4 SNPs were not segregating in the genotyped samples. The remaining seven markers were polymorphic with minor allele frequencies ranging from 0.10 (AFP) to 0.48 (RBP4). Because of the observed genetic variabilities in the investigated loci, the polymorphisms in the AFP, BMPR1B, CXCL10, ESR2, GNRHR, MAN2B2, and RBP4 genes can be considered suitable markers for association studies with litter-size traits in ITLW pigs.

We estimated genetic parameters in Landrace and Large White pig populations for litter traits at farrowing (total number born, number born alive, number stillborn, total litter weight at birth (LWB), and mean litter weight at birth) and those at weaning (litter size at weaning (LSW), total litter weight at weaning (LWW), mean litter weight at weaning (MWW), and survival rate from farrowing to weaning). We analyzed 65,579 records at farrowing and 6,306 at weaning for Landrace, and 52,557 and 5,360, respectively, for Large White. Single‐trait and two‐trait repeatability animal models were exploited to estimate heritability and genetic correlation respectively. Heritability estimates of LSW were 0.09 for Landrace and 0.08 for Large White. Genetic correlations of LSW with MWW were –0.43 for Landrace and –0.24 for Large White. Genetic correlations of LSW with LWW and LWB ranged from 0.5 to 0.6. The genetic correlation of MWW with LWW was positive, but that with LWB was negligible. The results indicate that utilizing LWW or LWB could improve LSW efficiently, despite the antagonistic genetic correlation between LSW and MWW.  相似文献

Liver metabolism is affected by nutrients. The aim of this study was to explore the effects of low‐protein diets (17% crude protein, CP) supplemented with branched‐chain amino acids (BCAAs), including leucine (Leu), isoleucine (Ile) and valine (Val), on hepatic amino acid profile and lipid metabolism in growing pigs. The ratio of Leu : Ile : Val in all groups was 1 : 0.51 : 0.63 (20% crude protein, CP), 1 : 1 : 1 (17% CP), 1 : 0.75 : 0.75 (17% CP), 1 : 0.51 : 0.63 (17% CP) and 1 : 0.25 : 0.25 (17% CP) respectively. Results revealed that compared to the positive control group (1 : 0.51 : 0.63, 20% CP), the low‐protein diets significantly augmented the concentrations of most essential amino acids and non‐essential amino acids (< .05), with the greatest values observed in the 1 : 0.25 : 0.25 group. Moreover, relative to the control, the low‐protein diets with the Leu : Ile : Val ratio ranging from 1 : 0.75 : 0.75 to 1 : 0.25 : 0.25 markedly downregulated the mRNA abundance of acetyl‐CoA carboxylase (ACC), lipoprotein lipase (LPL) and fatty acid‐binding protein 4 (FABP‐4) (< .05), and upregulated the mRNA expression of hormone‐sensitive lipase (HSL), peroxisome proliferator‐activated receptor‐g coactivator‐1α (PGC‐1α), uncoupling protein 3 (UCP3) and liver carnitine palmitoyltransferase 1 (L‐CPT‐1) (< .05). Therefore, our data suggest that protein‐restricted diets supplemented with optimal BCAA ratio, that is, 1 : 0.75 : 0.75–1 : 0.25 : 0.25, induce a shift from fatty acid synthesis to fatty acid oxidation in the liver of growing pigs. These effects may be associated with increased mitochondrial biogenesis.  相似文献

There are some differences in reproductive features between the Chinese Meishan (CM) pig and Large White (LW) pig. The aim of the present study was to investigate the quantitative changes and ultrastructural features of ciliated cells in the various regions of the CM and LW pig oviduct during the follicular and luteal phases of the oestrous cycle. In the fimbrial and ampullar epithelia at the follicular phase, the ciliated cells were more plentiful than in the isthmic region in both pigs. In the CM pigs, there was a striking decrease in the percentage and cell height of ciliated cells in the luteal phase compared with the follicular phase. Although similar quantitative changes were observed in the LW pig oviduct, these changes were less dramatic than that in the CM pig oviduct. In both pigs, the percentage and cell height of ciliated cells in the isthmus were unchanged between the follicular and luteal phases. The ultrastructure of ciliogenic and ciliated cells was observed. In the fimbrial and ampullar epithelia during the follicular phase, most of the ciliated cells showed normal morphology, having many elongated cilia and mitochondria, but in the CM pig oviduct during the luteal phase many of the ciliated cells possessed immature cilia and swollen mitochondria. Cells undergoing ciliogenesis were frequently observed in the fimbriae and ampulla, and occasionally in the isthmus. Cytoplasmic protrusions containing variable numbers of ciliary axonemal complexes occurred in the fimbrial and ampullar epithelium in the CM pigs at the luteal phase, suggesting that deciliation of cells occurs by membrane-bound cilia packets forming at the apices of cells and pinching off. These results demonstrate that there are regional variations in the cyclic changes associated with the oviductal ciliated cells of the pigs, while there are marked morphometrical and ultrastructural changes in oviductal ciliated cells of the CM pigs compared with that of the LW pigs.  相似文献

Atmospheric ammonia in animal housing is reported to have adverse effects on livestock performance and animal health. Previous experiments have found that 75 ppm ammonia reduced the production performance and altered body fat distribution quality of broilers. In this study, we examined the body fat distribution, serum metabolites and lipid metabolism gene expression of broiler exposed to ammonia. A total of 400 chickens were randomly allocated to four groups with four replicates and received ammonia treatments at 0, 25, 50 and 75 ppm, respectively, for 3 weeks. The average daily feed intake and weight gain were decreased when broiler was exposed to ammonia concentration exceeding 50 ppm (< .05). The increased abdominal fat and reduced thickness of subcutaneous adipose were found in broilers of 75 ppm group (< .05). When ammonia exceeded 50 ppm, the content of fat in breast muscle of broiler was increased, and when ammonia was higher than 25 ppm, the fat in liver was increased (< .05). It showed that the fat content in liver was a sensitive index for broilers exposed to ammonia. Furthermore, ammonia exposure had no significant effect on total cholesterol and triglyceride in serum, but significantly increased the relative mRNA expression of acetyl‐CoA carboxylase (= .046) and malic enzyme in liver (= .038), which indicated that ammonia exposure may increase the de novo fat synthesis in liver. In addition, ammonia increased the high‐density lipoprotein cholesterol (= .02) and activity of hepatic lipase in serum (< .001), which indicated that ammonia exposure may improve the transportation of cholesterol to liver. To conclude, our results indicated that ammonia exposure might increase the de novo fat synthesis in liver and increased the transportation of cholesterol to liver. In addition, the concentration of ammonia in poultry house should be limited lower than 25 ppm based on the variation of hepatic fat content.  相似文献

The allelic frequencies of PRKAG3 gene (the RN gene) have been investigated in several pig breeds. R200Q mutation appear only in Hampshire pigs, whereas V199I mutation is most abundant in Iberian, Porco Celta or Bizaro, and less in breeds selected for muscularity as Duroc, Landrace and Pietrain. A thorough study of phenotypic effects of V1991 has been performed in a Duroc × Landrace‐Large White cross. 199I homozygous pigs show increased pH24 values in ham homogenates and loin (0.14 and 0.16 pH units, respectively) compared to 199V homozygous ones. Meat of 199I homozygous pigs exudates 42.6% less fluid and is darker (2.46 ‘L’‐value units). 199I homozygous pigs are fatter (4.2 mm more backfat thickness) and contain less muscle mass in ham (1.0 percentage points) and shoulder (2.7 percentage points), than 199V homozygous ones. 199I homozygous pigs contain 7.3% less protein in the belly and 8.5% more fat in shoulder muscle mass than 199V homozygous pigs. 199I homozygous pigs have also superior functional properties: better gelling (22.8% larger G′ value) and emulsion capacities (14 percentage points less of total exuded fluid), and higher curing yield in the belly (6 percentage points more). These data support the adipogenic character of the V199I mutation. The advantages and disadvantages of selecting any of the two PRKAG3 alleles for position 199 are discussed.  相似文献

为比较繁殖性状主效基因对不同品系大白猪产仔性能的影响,以便在不同品系大白猪选种实际应用提供依据,本试验采用PCR-RFLP技术,分别对促卵泡素亚基β(FSHβ)、催乳素受体(PRLR)、视黄醛结合蛋白(RBP4)、核受体辅激活蛋白Ⅰ(NCOA1)、瘦素(LEP)等5种与繁殖性状相关候选基因的基因型频率以及不同基因型的总产仔数和产活仔数在英系、美系大白猪群体中进行了检测与关联分析。结果显示:NCOA1和LEP基因多态位点分别在英系和美系大白猪中未检测到BB基因型,其他基因多态位点均存在AA、AB、BB等3种基因型,在美系大白猪群体中5个基因多态位点均符合Hardy-Weinberg平衡定律(P>0.05);而在英系大白猪群体中除在FSHβ、NCOA1、LEP基因多态位点均处于Hardy-Weinberg平衡状态(P>0.05)外,在PRLR、RBP4基因多态位点处于Hardy-Weinberg不平衡状态(P<0.01)。各基因不同基因型与产仔性状关联分析结果表明,无论是英系大白猪还是美系大白猪,FSHβ基因多态位点在窝总产仔数和产活仔数中均表现为BB型为有利基因型,在美系大白猪中BB型较AA型总产仔数高1.6961头(P<0.05),产活仔数高1.696 9头(P<0.05),但在英系大白猪中不存在显著差异(P>0.05);LEP基因在英系大白猪中BB型为有利基因型,分别较AA型母猪总产仔数高0.790 7头、产活仔数高1.409 3头(P<0.05),而在美系大白猪中不存在显著差异(P>0.05);PRLR、RBP4、NCOA1基因对产仔数不存在显著差异(P>0.05)。  相似文献

Impact of MYOD family genes on pork traits in Large White and Landrace pigs   总被引:2,自引:0,他引:2  
Summary Porcine myogenic differentiation genes ( MYOD ) family play a key role in growth and muscle development and are therefore considered as candidate genes for meat production traits. The objective of the study was to investigate the polymorphisms at four loci belonging to the MYOD genes family and analyse their associations with variation in meat production traits in Czech pig breeds. To verify the associations between the polymorphisms and the selected meat traits, altogether 254 pigs, including full- and half-sibs, of Large White and Landrace breeds were tested. The studied meat characteristics were weight of neck, loin, shoulder and ham, lean meat content (LMC), backfat thickness, intramuscular fat (IMF), remission, dry matter content and test daily gain. Statistically significant associations were observed between MYOG gene and fat and neck weight, and between MYF5 gene and IMF and LMC. High significant differences were observed between genotypes AA and AB of MYOD1 in IMF and between genotypes AB and BB of MYF5 in loin weight.  相似文献

Genetic parameters for sow stayability were estimated from farrowing records of 10,295 Landrace sows and 8192 Large White sows. The record for sow stayability from parity k to parity k + 1 (k = 1, …, 6) was 0 when a sow had a farrowing record at parity k but not at parity k + 1, and 1 when a sow had both records. Heritability was estimated by using single-trait linear and threshold animal models. Genetic correlations among parities were estimated by using two-trait linear–linear and single-trait random regression linear animal models. Genetic correlations with litter traits at birth were estimated by using a two-trait linear–linear animal model. Heritability estimates by linear model analysis were low (0.065–0.119 in Landrace & 0.061–0.157 in Large White); those by threshold model analysis were higher (0.136–0.200 & 0.110–0.283). Genetic correlations among parities differed between breeds and models. Genetic correlation between sow stayability and number born alive was positive in many cases, implying that selection for number born alive does not reduce sow stayability. The results seem to be affected by decisions on culling made by farmers.  相似文献

We estimated genetic parameters for number born alive (NBA) from the first to the seventh parities in Landrace and Large White pigs using three models. Analyzing 55,160 farrowing records for 12,677 Landrace dams and 43,839 for 10,405 Large White dams, we used a single‐trait animal model to estimate the heritability of NBA at each parity and a two‐trait animal model and a single‐trait random regression model to estimate the genetic correlations between parities. Heritability estimates of NBA at each parity ranged from 0.08 to 0.13 for Landrace and from 0.05 to 0.16 for Large White. Estimated genetic correlations between parities in all cases were positive. Genetic correlations between the first and second parities were slightly lower than those between other neighboring parities. Genetic correlations between more distant parities tended to be lower, in some cases <0.8. The results indicate the necessity to investigate the applicability of evaluating NBA at different parities as different traits (e.g., the first and later parities), although a repeatability model might still be reasonable.  相似文献

This study was carried out to evaluate the nutritional effects of rice feeding and carnitine addition to a diet for broiler chicks. Thirty‐six male 10‐day‐old broiler chicks were assigned to one of the following four treatment groups: corn‐based diet (corn group), rice‐based diet (rice group), and each diet with added carnitine (100 ppm). The experimental period was 2 weeks. Rice feeding resulted in significantly higher growth performance (body weight gain and feed efficiency) compared to corn feeding. Carnitine addition also resulted in higher growth performance. Breast muscle and thigh muscle weight (g) were significantly higher in broiler chicks fed rice and those fed diets with added carnitine. Liver mRNA expression of IGF‐I was significantly higher in broiler chicks fed rice compared to those fed corn. There was no significant difference in mRNA expression of muscle atrogin‐1 or liver CPT‐I between broiler chicks fed rice and those fed corn, not between broilers chicks fed diets containing carnitine or not. Overall, these results show that rice feeding and carnitine addition improve the growth performance of broiler chicks by increasing mRNA expression of liver IGF‐I. In addition, carnitine action is not affected by different cereals (corn and rice).  相似文献

The objective of this study was to evaluate the long‐term survival rates, clinical response, and lung gross and microscopic changes in pigs treated intratracheally with lipopolysaccharide of Escherichia coli 0111:B4 (LPS‐Ec). Healthy pigs were randomly allocated to three groups: (i) no‐LPS‐Ec (n = 1), (ii) LPS‐Ec‐T1 (1 mg/mL, 10 mL/pig) (n = 7), and (iii) LPS‐Ec‐T2 (0.5 mg/mL, 10 mL/pig) (n = 6). Two pigs from each dose group were euthanized at 24 (n = 3 for T1), 48 and 144 h post‐LPS‐Ec challenge. LPS‐Ec‐treated animals showed macroscopic lesions in middle lobes of the lung. A reversible recruitment of macrophages and neutrophils was observed at 24, 48, and 144 h post‐LPS‐Ec challenge. The highest cellular infiltration level was observed at 24 h after challenge. The highest clinical scores were evident in both experimental dose levels within 3 and 5 h after LPS‐Ec administration. Administration of LPS‐Ec, under the conditions evaluated, can be used to induce a reproducible model of acute pulmonary inflammation in pigs.  相似文献

Data from a French experimental herd recorded between 1990 and 1997 were used to estimate genetic parameters for individual birth and weaning weight, as well as litter size of Large White pigs using restricted maximum likelihood (REML) methodology applied to a multivariate animal model. In addition to fixed effects the model included random common environment of litter, direct and maternal additive genetic effects. The data consisted of 1928 litters including individual weight observations from 18 151 animals for birth weight and from 15 360 animals for weaning weight with 5% of animals transferred to a nurse. Estimates of direct and maternal heritability and proportion of the common environmental variance for birth weight were 0.02, 0.21 and 0.11, respectively. The corresponding values for weaning weight were 0.08, 0.16 and 0.23 and for litter size 0.22, 0.02 and 0.06, respectively. The direct and the maternal genetic correlations between birth and weaning weight were positive (0.59 and 0.76). Weak positive (negative) genetic correlations between direct effects on weight traits and maternal effects on birth weight (weaning weight) were found. Negative correlations were found between direct genetic effect for litter size and maternal genetic effects on all three traits. The negative relationship between litter size and individual weight requires a combined selection for litter size and weight.  相似文献

This study examined the influence of adding different amounts of maternal dietary l ‐carnitine and two fat types on fatty acid (FA) composition and the expression of lipid metabolism‐related genes in piglets. The experiment was designed as a 2 × 2 factorial with two fat types (3.5% soyabean oil, SO, and 3.5% fish oil, FO) and two levels of l ‐carnitine (0 and 100 mg/kg) added to the sows' diets. A higher proportion of n‐3 polyunsaturated fatty acids (PUFA) and a lower ratio of n‐6/n‐3 PUFA in sow milk and piglet tissues were observed in the FO groups than in the SO groups. Adding l ‐carnitine increased the proportion of C16:1 in sow milk and decreased n‐3 PUFA in piglet subcutaneous fat. Hepatic peroxisome proliferator‐activated receptor α (PPAR‐α) was more abundantly expressed in piglets from the FO groups than from the SO groups (p < 0.05), whereas stearoyl‐CoA‐desaturase (SCD), sterol regulatory element binding protein‐1 (SREBP1) and ?6‐desaturase (D6D) genes were less expressed in the FO groups compared with piglets from the SO groups. The expression of fatty acid synthase (FAS) genes was decreased in the SO groups with l ‐carnitine compared to that of the other dietary treatments. No differences among dietary treatments were observed with regard to the expression of acetyl‐CoA carboxylase (ACC). In conclusion, FO and l ‐carnitine supplementation in sows affect FA composition and hepatic gene expression in piglets.  相似文献

This experiment investigated the effects of supplementing the maternal diet with linseed oil (LSO) and soya bean oil (SBO) on immunoglobulins, the fatty acid composition and hepatic expression of lipid metabolism‐related genes in piglets. Multiparous sows (twenty‐four per diet) were fed on diets containing a supplement of either SBO or LSO during last week of gestation and lactation. The results indicated that supplementation of maternal diet with LSO could improve the weaning weight of piglets and average daily gain (ADG) (p < 0.05). The concentration of immunoglobulin G (IgG) and immunoglobulin A (IgA) was enhanced in sow plasma, colostrum and milk by the addition of LSO (p < 0.05). In addition, the concentration of 18: 3n‐3 fatty acids was higher in the milk of LSO sows. Meanwhile, maternal supplementation with LSO increased the levels of plasma IgG, IgA and the tissues n‐3 polyunsaturated fatty acid (PUFA) in piglets (p < 0.05). Correspondingly, the mRNA expression levels of hepatic ?5‐desaturase (D5D) and ?6‐desaturase (D6D) were higher, and fatty acid synthase (FAS) was lower in piglets from LSO‐fed sows when compared with that in the SBO group. In conclusion, LSO supplementation of the maternal diet increases immunoglobulins, modifies the fatty acid composition and affects the gene of D5D and D6D expression of piglets.  相似文献

Glutamine metabolism is an important metabolic pathway for cancer cell survival, and there is a critical connection between tumour growth and glutamine metabolism. Because of their similarities, canine mammary carcinomas are useful for studying human breast cancer. Accordingly, we investigated the correlations between the expression of glutamine metabolism‐related proteins and the pathological features of canine mammary tumours. We performed immunohistochemical and western blot analysis of 39 mammary tumour tissues. In immunohistochemical analysis, the expression of glutaminase 1 (GLS1) in the epithelial region increased according to the histological grade (P < .005). In the stromal region, complex‐type tumours displayed significantly higher GLS1 intensity than simple‐type tumours. However, glutamate dehydrogenase expression did not show the same tendencies as GLS1. The western blot results were consistent with the immunohistochemical findings. These results suggest that the expression of GLS1 is correlates with clinicopathological factors in canine mammary tumours and shows a similar pattern to human breast cancer.  相似文献

The effect of breed [Creole (CR) vs. Large White (LW)] on performance and physiological responses during acclimation to high ambient temperature was studied in 2 experiments involving 24 (12/breed) growing pigs each. Pigs were exposed to 24 degrees C for 10 d (d -10 to -1) and thereafter to a constant temperature of 31 degrees C for 16 d (d 1 to d 16) in Exp. 1 and for 20 d (d 1 to d 20) in Exp. 2. For both experiments, the temperature change was achieved over 4 h on d 0. The first experiment began at 105 d of age, and the average BW of CR and LW pigs was 36.6 +/- 2.5 kg and 51.7 +/- 3.0 kg, respectively. The second experiment was designed to compare both breeds at a similar BW (about 52 kg on d 0). Pigs were individually housed and given ad libitum access to feed. At 24 degrees C, ADG was lower (P < 0.01) in CR than in LW (602 vs. 913 g/d and 605 vs. 862 g/d in Exp. 1 and 2, respectively), but the ADFI was not affected by breed (190 and 221 g x d(-1) x kg(-0.60) in Exp. 1 and 2, respectively). Short-term thermoregulatory responses during the 4-h transition from 24 to 31 degrees C (d 0) were analyzed according to a linear plateau model to determine the break point temperature, above which rectal temperature (RT), cutaneous temperature (CT), and respiratory rate (RR) began to change. The CT increased linearly with temperature increase (0.22 degrees C/ degrees C) and was less (P < 0.05) in CR than in LW (by -0.3 degrees C on average). In both experiments, the break point temperature for RT was not affected by breed (27.6 degrees C on average), whereas for RR it was greater (P < 0.05) in CR than in LW (27.5 vs. 25.5 degrees C, P < 0.01). On average, ADFI declined by about 50 g x d(-1) x kg(-0.60) from d -1 to d 1 (P < 0.01), and thereafter at 31 degrees C, it gradually increased (23 g x d(-1) x kg(-0.60); P < 0.05), suggesting an acclimation to high exposure. This response was not influenced by breed. After the day that marked the beginning of the acclimation response (i.e., the threshold day), RR, CT, and RT declined over the duration of exposure to 31 degrees C (P < 0.05) in both experiments. During this period, RT and CT were less in CR than in LW pigs (39.6 vs. 39.9 degrees C and 37.9 vs. 38.2 degrees C, respectively; P < 0.05), whereas RR was not affected by breed. The threshold day at which RT began to decline was less in CR than in LW pigs (0.18 vs. 1.17 d and 0.39 vs. 0.93 d in Exp. 1 and 2, respectively; P < 0.05). In conclusion, this study suggests that short- and long-term physiological reactions during heat acclimation differed when CR and LW pigs were compared at the same age or BW.  相似文献
