Lavandula angustifolia (lavender) is a small woody perennial grown for essential oil, which is steam distilled from flowers. To potentially improve size of flowers and oil yield we produced and characterised autotetraploid plants. L. angustifolia seed germinated in the presence of the mitotic spindle inhibitor colchicine at concentrations of 125 mg l−1 or less resulted in plants carrying sports with larger flowers. Propagation of two sports gave rise to putative polyploid cultivars C3/2 and C6/24. Direct chromosome counts in root tip cells of seedlings from four common cultivars of L. angustifolia and the seed lot from which C3/2 and C6/24 were derived was 50 whereas C3/2 and C6/24 had greater than 90 chromosomes indicating they were autotetraploid. Ploidy level assessed by flow cytometry (FCM) of nuclei showed that 12 cultivars of L. angustifolia had similar nuclear DNA content whereas C3/2 and C6/24 had double the amount of DNA confirming autotetraploidy. The genome size (1C-value) of a diploid L. angustifolia cultivar was estimated by FCM to be 0.90 (±0.07) pg. Morphological characteristics were measured in autotetraploid and control plants. Autotetraploids had thicker peduncles, larger flowers and larger seeds than diploids. Scanning electron microscopy revealed peltate glandular trichomes were larger in the tetraploids relative to diploids. Both tetraploid and diploid cultivars had complex non-glandular trichomes on leaves and sepals and two different types of capitate glandular trichomes were identified on leaves. Autotetraploid lavenders represent useful germplasm both for commercial oil production and future breeding.  相似文献

Numerous cytogenetic studies have been performed on alfalfa (Medicago sativa L. 2n= 4x= 32), but only a few have examined the somatic chromosomes. Because the major factor limiting the study of somatic chromosomes of M. sativa is their rather uniform morphology, characterization was attempted using the C-banding technique instead of the traditional Feulgen method. Chromosome morphology and position of the heterochromatic bands resulted in satisfactory characterization of the chromosomes and provided further evidence of the autotetraploid origin of the species. It was possible to identify the homologous chromosomes and arrange them into eight groups of four. The ideogram of the basic genome of eight chromosomes was constructed. The impact of these results on cytogenetic studies and breeding research in alfalfa is discussed.  相似文献

To address the issue of genome evolution in autopolyploids and particularly to investigate whether rapid sequence elimination also occurs in autopolyploids as in allopolyploids, amplified fragment length polymorphism (AFLP) fingerprinting was employed to examine a large number of genomic loci in F1 hybrids between two different autotetraploids of Arabidopsis thaliana accessions, namely Ler and Col. Using this approach, perfect additivity in the F1 hybrids was found between the newly‐formed autopolyploids when compared with their parental lines. Using flow cytometry, the study was extended in a quantitative manner, in which the nuclear DNA contents in one autotetraploid A. thaliana accession Ler, was determined. The increase in genome size of the autotetraploid line was additive. Taken together, no evidence was found for genome size reduction due to autopolyploidization of A. thaliana. The results indicating that there was no DNA loss in autotetraploid A. thaliana suggest that a different type of genome evolution may occur in autopolyploids during the initial stages of their formation when compared with allopolyploids.  相似文献

Summary Viable monohaploids with 12 chromosomes in their somatic cells were obtained from an autotetraploid cultivar of Solanum tuberosum (2n=4x=48) by inducing female parthenogenesis successively in the tetraploid cultivar and in dihaploids from that cultivar. Both dihaploids and monohaploids were induced using the diploid S. phureja clones IvP 35 and IvP 48 as pollinators. The average frequencies of dihaploids and monohaploids in 1973 were 2.0 and 0.14 per berry respectively.Non-homologous chromosome associations (bivalents, trivalents and even quadrivalents) were observed at metaphase I of meiosis in pollen mother cells of the two monohaploids studied.The occurrence of non-homologous associations of chromosomes during meiosis is discussed. In addition the potential significance of monohaploids for basic research andfor breeding of potatoes is considered.  相似文献

An eosin B staining procedure for use with confocal microscopy (WE‐CLSM) was used to examine the mature embryo sacs in 24 typical autotetraploid (2n = 4x = 48) indica/japonica hybrids and to compare with their original diploid (2n = 2x = 24) indica/japonica hybrids in rice. Four of the 24 hybrids showed >80% embryo sac fertility, and 13 hybrids over 70%. The average embryo sac fertility of the 24 autotetraploid indica/japonica hybrids was 68.08%, which was 17.33% higher than that in diploid hybrids. When compared with the diploid hybrids, 79.17% of the autotetraploid hybrids had higher embryo sac fertility. A diversity of abnormal embryo sacs occurred in autotetraploid indica/japonica hybrids. Embryo sac abnormalities were similar for both autotetraploid and diploid hybrid, but their frequencies were different. Some novel abnormal types were found in autotetraploid hybrids, such as enlarged egg apparatus. The embryo sac fertilities and frequencies of various abnormalities in autotetraploid hybrids varied with parental genotype and environment. The average seed set of the autotetraploid hybrids were higher than their diploid hybrids in different growing seasons.  相似文献

Summary The species Solanum chacoense BITT. (2n=2x=24) is a tuber-bearing, self-incompatible species which is important both for breeding and for genetic research. It crosses readily with most other tuber-bearing Solanum species including the common potato S. tuberosum (2n=4x=48). Gametophytic incompatibility hampers research in and utilization of this species. Doubling the chromosome number by colchicine makes it a self-compatible autotetraploid. By crossing selfed progeny of 4x-S. chacoense with a number of haploid-inducing diploid Solanum species a high yield of different dihaploid S. chacoense individuals (2n=24) could be obtained from one originally diploid clone: S. chacoense CPC 1153. More than 160 haploids showing a large variability were identified. The average haploid frequency was 53.7 per 100 berries. Most hybrid plants (70–100%) from four 4x × 2x crosses studied were tetraploid. The frequency of triploid hybrids was low (0–10%). Haploid-inducing capacity of fifteen male parents used in this study varied from 0–141 haploids per 100 berries. Careful examination of 156 haploids revealed 15 viable aneuhaploids (2n=25, 26, and 27), i.e. 9.4%. The potential value of these aneuhaploids is discussed.About 50% of the haploids were sufficiently male fertile to use them in crossing. A few of them set berries after selfing. Five aneuhaploids including the one with 27 chromosomes were successfully crossed as females with a diploid hybrid clone.  相似文献

Summary Conversion of a system of free pairing between four homologous chromosomes of an autotetraploid to the restricted two by two pairing of an allopolyploid requires differentiation of orginally homologous chromosomes. Structural rearrangements can induce partial differentiation, as can naturally occurring differences in the attraction system itself. Combination of the two was found to be very effective, but it is extremely difficult to combine the correct rearrangements with the correct attraction differences in alle chromosomes simultaneously.  相似文献

The agronomic performance and chromosome behavior of autotetraploid sorghum [Sorghum bicolor (L.) Moench] inbreds and F1 hybrids were investigated. Autotetraploids have possible commercial use and can serve as bridging species in crosses to natural tetraploids. Autotetraploid sorghum inbreds (2n = 40) had higher kernel weight, seed yield, and protein and amino acid content than their diploid counterparts. Stalk height and panicle length of the autotet-raploids, however, were not different from those of the diploids, and seed set of autotetraploids was lower in early generations. Seed set of autotetraploid Ft hybrids was related to and higher than that of the autotetraploid parental inbred lines. Frequency of quadrivalents, anaphase I and II laggards, and quartet micronuclei varied among autotetraploids. Frequency of quadrivalents did not appear to be an important factor in seed set nor was it significantly associated with frequency of laggards. Aneuploids (2n = 39, 41) had higher percentages of laggards and micronuclei than did euploids (2n = 40). Methods of pollination significantly affected seed set in autotetraploid S4 lines. Differences in seed set among autotetraploid lines indicated the feasibility of selecting for lines with high seed set and utilizing those lines for hybrid production.  相似文献

Summary Tetraploid Bromus ciliatus L. is a North American bromegrass that has been placed in the Pnigma section of Bromus. The objective of this study was to characterize the genome of tetraploid B. ciliatus by cytogenetic methods and compare it to the genomes of other species included in the section Pnigma. All the plants of the accession (USDA PI 232214) selected for chromosome counting were tetraploids (2n = 28). The mean 2C nuclear DNA content for tetraploid B. ciliatus was 19.13± 0.07 pg as determined by flow cytometry which is significantly greater than the tetraploid DNA content of B. inermis Leyss. (11.74± 0.16 pg). C-banding procedures were used to identify individual mitotic chromosomes and to develop a karyotype for B. ciliatus. The genome of the tetraploid B. ciliatus consisted of 16 median chromosomes, eight submedian chromosomes, and four chromosomes with satellites which included one pair with a large satellite and one pair with a small satellite. The general pattern of the distribution of constitutive heterochromatin in B. ciliatus was quite different than the other bromegrasses that have been analyzed to date. Except for two pairs of chromosomes, all chromosomes in tetraploid B. ciliatus had telomeric bands on one or both arms. Some of the chromosomes with telomeric bands had centromeric bands that were located at one or both sides of the centromere and intercalary bands which were generally absent in the other bromegrass species. It was possible to identify all chromosomes of tetraploid B. ciliatus and to match the pairs of homologous chromosomes by using chromosome lengths, arm length ratios and C-banding patterns. The results of this study indicate that tetraploid B. ciliatus has different genomes than the European species evaluated to date in the section Pnigma.  相似文献

为获得高产优质的不结球白菜四倍体苏州青,本研究使用浓度分别为0.1%,0.2%及0.3%的秋水仙素溶液,点滴处理二倍体苏州青幼苗植株的子叶生长点,进行5次处理后,通过形态学和细胞学方法筛选鉴定四倍体苏州青植株,并比较二、四倍体苏州青的农艺性状及品质差异。结果表明:秋水仙素浓度为0.2%时加倍效果最好。形态解剖学鉴定结果表明:四倍体在株型、叶片、花、种荚、种子大小都有较明显的变大趋势;四倍体植株叶片气孔较二倍体变大,气孔密度变小;同时四倍体植株花粉粒比二倍体更膨大,更不规则;细胞学鉴定结果表示,四倍体植株染色体数为40条,二倍体植株为20条;通过流式细胞仪鉴定,四倍体植株荧光强度约为二倍体的2倍。通过品质比较分析,结果显示四倍体苏州青的各项生理指标含量较于二倍体均有不同。通过农艺性状比较,单株质量、叶宽、叶厚都有一定程度的增加。本研究利用秋水仙素操作技术,抑制有丝分裂后期纺锤丝的形成,实现染色体加倍,创制不结球白菜新种质,为选育四倍体不结球白菜提供理论基础和种质资源。  相似文献

用8个微卫星位点,63个等位基因,研究了8个栽培型紫花苜蓿品种(Medicago sativa L.),320个单株群体内和群体间的遗传变异。首次提出用四倍体方法统计紫花苜蓿微卫星位点的等位基因频率,以等位基因频率为基础,得到遗传杂合度、有效等位基因数、标准遗传距离和NJ(Neighbor Jointing)聚类图。结果表明这8个材料在群体内和群体间均具较高变异。通过0/1法和四倍体方法得到的遗传杂合度之间的显著性检验(P<0.01),表明四倍体方法更适于紫花苜蓿微卫星多态性研究。  相似文献

Summary An autotetraploid population of caraway (Carum carvi L.) was produced by treating the growing points of young plants with a 0.2% colchicine solution. Selection on morphology, pollen size and pollen shape, followed by a selection on seed size proved to be an effective way to identify the mixoploid C0 plants. Three percent of the treated plants produced a tetraploid progeny. Moderate selection in the C1 and the C2 population resulted in an improvement of the average seed-set to the amount of 50% of the diploid control, with a range of 21–78%. The 1000-kernel weight of the tetraploid material was 7 g, compared to 3 g in the diploid control. The oil content of the autotetraploid seed was increased with 35.6%; the increase in carvone amounted to 6.9%. The results of the experiment indicate that breeding work in autotetraploid caraway may result in a new cultivar.  相似文献

Hybridization followed by pedigree selection was carried out between two autotetraploid strains of fenugreek (Trigonella foenum-graecum L.). Morphological and cytological investigations were carried out in two autotetraploid parents, their F1 hybrids and F6 lines derived from F1 after selfing. Cytomorphological data of F6 lines were compared with those of F1, hybrids and of their parents. The hybrid plants were intermediate with respect to different vegetative traits, however, they showed increased seed set as compared to their autotetraploid parents. The mean pod number per plant in F6 was one and half times as high as one of the high yielding tetraploid parents and approximately twice as high as that of F1 hybrid. Meiosis in F6 was more regular than in F, and the parents. Thus, hybridization followed by pedigree selection is helpful not only in regularizing the meiotic behavior, but vigour and seed fertility have also improved in autotetraploid fenugreek.  相似文献

Summary In interspecific crosses between diploid and autotetraploid cytotypes of Hordeum vulgare L. (cultivated barley) and H. bulbosum L. (bulbous barley grass), several kinds of hybrid endosperm, vegetative tissue and generative tissue were studied. In all these tissues, mitotic disturbances occurred which resulted in chromosome elimination. The chromosomes of H. bulbosum were eliminated more frequently than those of H. vulgare.Differences between the three types of tissue might have been due to differences between their mitotic rhythms. Differences between various kinds of the same type of tissue were discussed in relation to the ratio of the total number of genomes in embryo, endosperm and maternal tissue, and the ratio of the genomes of the parental species within the hybrid nuclei. For both ratios assumptions as to the genetic value of each genome were taken into account.The cause of the disturbances was probably genic disharmony between the parental genomes.  相似文献

Chromosome duplication—autopolyploidization—may affect plant morphology and breeding systems, ultimately enabling the production of improved genotypes. In this study, the autotetraploid lines obtained from the self-incompatible diploid Hylocereus monacanthus and the autooctapolyploid lines obtained from the self-compatible tetraploid H. megalanthus were studied and compared with the donor accessions. The resulting H. monacanthus autotetraploids exhibit lower fruit weight, seed number, and pollen viability than the donor plant, but it has larger pollen grains. Although the resulting H. megalanthus autooctaploids had larger pollen grains and lower pollen viability compared with the donor plant, only aborted fruits were obtained from these lines. The most valuable change observed was the breakdown of the self-incompatibility system in the H. monacanthus autotetraploid lines. This research provides important information on the horticultural value of vine cacti autopolyploid lines.  相似文献

Alfalfa cultivar development will be enhanced by breeding strategies whichutilize the full potential of autotetraploid population genetic structures. Thisstudy evaluates the effectiveness of an allelic selection scheme, which wasdeveloped to overcome limitations of inbreeding depression and to exploitgeneral and specific combining ability effects in autotetraploid populations.Allelic selection entails the minimization of non-additive genetic effects byselecting among full-sib families (F1) which are at uniform levels ofheterozygosity. Such F1 lines are developed by crossing individuals fromtwo unrelated random mating populations. Selected F1 lines wereintercrossed to form an improved population. Eight random matingpopulations of alfalfa were developed to study the effectiveness of allelicselection. Selection for increased dry matter yield resulted in alfalfapopulations with 38 percent greater yield than the parent populations. Twocycles of intercrossing, among selected F1 lines, did not dissipate the gainfrom selection. This result has important implications for synthetic cultivardevelopment in which a major limitation is the decline in productivity withadvancing generations of seed increase. A positively correlated response toselection for dry matter yield was observed for plant height and stemdiameter. The results of this research indicate that continued testing of theallelic selection scheme is warranted and could have a significant impact onthe breeding of autotetraploid alfalfa, particularly for synthetic cultivardevelopment.  相似文献

Festulolium is a hybrid between Festuca and Lolium species that has valuable agronomic traits from both grass species. The purpose of our breeding program is to produce hexaploid festulolium that introduces tolerance to summer depression into Italian ryegrass (Lolium multiflorum) by crossing it with tall fescue (Festuca arundinacea). However, we found the DNA ploidy of hexaploids was not stable and was reduced in successive generations. We aimed to find out how to obtain stable high-ploidy festulolium. F1 hybrids of L. multiflorum and F. arundinacea were produced. The F3 generation was produced from putative hexaploid F2 individuals by open pollination. The F4 to F6 generations were obtained by polycrossing. The DNA ploidy levels of F2 to F6 individuals were estimated by flow cytometry. Cytological characteristics of the F5 and F6 individuals were investigated by FISH and GISH. The DNA ploidy level of hexaploid festulolium was reduced and stabilized at almost the same level as a tetraploid. Seed fertility was inversely correlated with an increase in ploidy level. GISH revealed no preferential Lolium transmission. FISH with a telomere probe revealed that counting the exact number of chromosomes in festulolium was difficult. DNA ploidy level was strongly correlated with the number of chromosomes.  相似文献

Late blight (LB), caused by the oomycete Phytophtohra infestans, is one of the most destructive diseases of tomato (Solanum lycopersicum) and other Solanaceae species. Current disease control and prevention strategies are not sufficient to control the disease in tomato. Recent germplasm screening experiments led to the identification of a new source of resistance (PI 270443) in the tomato wild species S. pimpinellifolium. This study was conducted to identify genomic regions associated with LB resistance in this accession. A large F2 population (n = 986) derived from a cross between PI 270443 and a LB-susceptible tomato breeding line (NCEBR-2) was screened for LB resistance using a highly aggressive isolate of P. infestans. Twenty-five of the most resistant and 29 of the most susceptible, but surviving F2 individuals were identified based on disease evaluations conducted in the F2 and F3 progeny populations. The selected individuals were genotyped with 153 DNA markers located across the 12 tomato chromosomes. A selective genotyping approach led to the identification of two genomic regions on tomato chromosomes 1 and 10 associated with LB resistance in PI 270443. Identification of two genomic regions associated with resistance was consistent with a previous estimate of the number of LB resistance genes in this accession. Research is currently underway to fine map the two resistance genes and incorporate them into new tomato breeding lines and hybrid cultivars.  相似文献

为发掘枳橙同源四倍体资源,从而为柑橘砧木遗传改良提供新种质,本研究通过初步的形态学观察,从具多胚性的柑橘重要砧木资源枳橙实生苗群体750个单株中初步筛选出具典型多倍体特征的植株24株,其中流式细胞仪鉴定为枳橙四倍体22株,群体四倍体自然发生率达2.93%。下表皮气孔分析表明枳橙四倍体气孔密度显著低于二倍体对照。对获得的四倍体植株进行了SSR分子鉴定,19对引物扩增结果均显示枳橙四倍体与其二倍体对照的带型完全一致,表明获得的四倍体枳橙均为同源四倍体。本研究表明将形态学初选与流式细胞仪快速鉴定及SSR分析相结合是从多胚性柑橘资源实生群体获得同源四倍体的简便有效途径,本研究发掘的同源四倍体枳橙是柑橘砧木遗传改良相关研究的良好资源。  相似文献

Chromosome morphology was studied in diploid cultivars of Tulipa fosteriana and T. gesneriana (2n = 2x = 24) and triploid Darwin hybrids (2n = 3x = 36) developed from interspecific crosses of T. gesneriana and T. fosteriana. Chromosomes were arranged in the karyotype according to decreasing total length. Based on our karyotypic analysis, we propose that median chromosomes may serve as markers for diploid genotypes. Discriminant analysis with respect to total chromosome length and short arm length showed a significant difference between the size of the larger median chromosomes of T. gesneriana and T. fosteriana Comparison of median chromosome length in Darwin hybrid tulips showed that two larger chromosomes and one smaller chromosome were derived from T. gesneriana and T. fosteriana, respectively. This finding was clearly and unambiguously confirmed by simultaneous hybridization of differentially labeled genomic probes of T. fosteriana and T. gesneriana to metaphase chromosomes of the triploid cultivar ‘Yellow Dover’, thereby enabling us to distinguish between the 24 chromosomes derived from T. gesneriana and 12 chromosomes derived from T. fosteriana. Thus, genomic in situ hybridization and median chromosome analyses can be useful to identify the genome constitution of triploid Darwin hybrid tulips. In addition, their hybridity was readily verified by flow cytometry using vegetative tissue of Darwin hybrid tulips. Our results clarify the process of Tulipa cultivar formation and will be useful for interspecific hybridization breeding.The first and second author have contributed equally to this paper  相似文献
