Powdery mildew is one of the most common and severe diseases of cucurbits, causing heavy yield losses in all growing areas when not successfully controlled. Two different fungal species, Podosphaera xanthii and Golovinomyces orontii, are generally recognized as causal agents. The results of monitoring carried out in 2016 and 2018 confirmed that P. xanthii is the exclusive pathogen causing cucurbit powdery mildew (CPM) in southern Italy. P. xanthii is a bipolar heterothallic fungus; a PCR-based method for distinguishing MAT1-1 and MAT1-2 idiomorphs was applied for assessing mating type distribution in fungal populations present on cucurbits in different sites. The idiomorph MAT1-2 was prevalent over the MAT1-1 in 2016, whereas they were approximately in a 1:1 ratio in 2018; this finding corroborated the hypothesis that the MAT1-1 idiomorph was more recently introduced in the area. Cyflufenamid-resistant isolates were widespread in commercial greenhouses and field plantings even though use of this fungicide had been drastically reduced by the farmers 1 year before the monitoring due to the effectiveness losses observed in CPM control. Occurrence of cyflufenamid resistance and its impact on efficacy were evaluated in a field trial comparing different fungicide spray schedules. Cyflufenamid-resistant isolates were detected even at the first appearance of symptoms on leaves, increasing over time. Isolates resistant to cyflufenamid showed a resistance factor as high as 900. Generally, P. xanthii was better controlled when cyflufenamid was used in integrated strategies rather than in spray schedules based on the exclusive use of the fungicide.  相似文献

Powdery mildew of hop (Podosphaera macularis) may cause economic loss due to reductions in cone yield and quality. Quantitative estimates of crop damage from powdery mildew remain poorly characterized, especially the effect of late season disease management on crop yield and quality. Field studies in Washington State evaluated cone yield, bittering acid content and quality factors when fungicide applications were ceased at different stages of cone development. The incidence of cones with powdery mildew was linearly correlated with yield of cones, bittering acids and accelerated cone maturation. In cultivar Galena, the cumulative effect of every 1% increase in cones powdery mildew incidence was to reduce alpha‐acid yield by 0·33%, which was due to direct effects on cone yield but also indirect effects mediated by dry matter. In the more susceptible cultivar Zeus, alpha‐acid yield was increased 20% by controlling powdery mildew through the transition of bloom to early cone development compared to ceasing fungicide applications at bloom: additional applications provided only modest improvements in alpha‐acid yield. In both cultivars, the impact of powdery mildew on aroma characteristics and bittering acid content were less substantial than cone yield. The damage caused by powdery mildew to cone colour and alpha‐acid yield, as well as the effectiveness of fungicide applications made to manage the disease, appears inseparably linked to dry matter content of cones at harvest. Realising achievable yield potential in these cultivars requires control of the disease through early stages of cone development and harvest before maturity exceeds c. 25% dry matter.  相似文献

Diversity of powdery mildew pathogens infecting pea (Pisum sativum) in the US Pacific Northwest was investigated using both molecular and morphological techniques. Phylogenetic analyses based on rDNA ITS sequences, in combination with assessment of morphological characters, defined two groups of powdery mildews infecting pea. Group I (five field samples and three glasshouse samples) had ITS sequences 99% similar to those of Erysiphe pisi in GenBank and exhibited simple, mycelioid type of chasmothecial appendages typical of E. pisi. Erysiphe pisi is normally considered as the powdery mildew pathogen of pea. Group II (four glasshouse samples and two field samples) had ITS sequences 99% similar to those of E. trifolii and produced chasmothecia with dichotomously branched appendages similar to those of E. trifolii. There are fourteen nucleotide differences in the ITS region between the two groups. The correlation of rDNA ITS sequences with teleomorphic features for each of the two groups confirms their identity. Repeated samplings and artificial inoculations indicate that both E. pisi and E. trifolii infect pea in the US Pacific Northwest. Erysiphe trifolii is not previously known as a pathogen of pea. The existence of two distinct powdery mildew species infecting pea in both glasshouse and field environments may interfere with the powdery mildew‐resistance breeding programmes, and possibly explains putative instances of breakdown of resistance in previously resistant pea breeding lines.  相似文献

Verticillium dahliae, a soilborne plant pathogen, causes wilt disease in many important crops. We reported previously that the mating type gene MAT1-2-1 is spread to isolates of this asexual fungus. However, we did not determine whether V. dahliae is homothallic or heterothallic because the opposite mating type gene, MAT1-1-1, had not been identified. In the present study, we identified the MAT1-1-1 gene from an isolate lacking MAT1-2-1 and the mating type idiomorphs of V. dahliae. Each isolate we tested contained either the MAT1-1 or MAT1-2 idiomorph, indicating that the asexual fungus V. dahliae is potentially heterothallic.  相似文献

Lettuce (Lactuca sativa) is the major leafy vegetable that is susceptible to powdery mildew disease under greenhouse and field conditions. Quantitative trait loci (QTLs) for resistance to powdery mildew under greenhouse conditions were mapped in an interspecific population derived from a cross between susceptible L. sativa cultivar Salinas and the highly susceptible L. serriola accession UC96US23. Four significant QTLs were detected on linkage groups LG 1 (pm‐1.1), LG 2 (pm‐2.1 and pm‐2.2) and LG 7 (pm‐7.1), each explaining between 35 to 42% of the phenotypic variation. The four QTLs are not located in the documented hotspots of lettuce resistance genes. Alleles for the disease resistance at the four QTLs originated from both parents (two from each), demonstrating that even highly susceptible accessions may provide alleles for resistance to powdery mildew. These QTLs appeared to operate during limited periods of time. Results of the field trials with F2:3 and F3:4 families derived from a Soraya (moderately resistant) × Salinas cross demonstrated effective transfer of resistance to powdery mildew in this material. An integrated rating approach was used to estimate relative levels of resistance in 80 cultivars and accessions tested in a total of 23 field and greenhouse experiments. Generally, very low resistance was observed in crisphead‐type lettuces, while the highest relative resistance was recorded in leaf and butterhead types. Comparison of two disease assessment methods (percentage rating and the area under the disease progress steps, AUDPS) for detection of QTLs shows that the two approaches complement each other.  相似文献

A temperature‐driven, mechanistic model predicting the development of Erysiphe necator chasmothecia in vineyards was developed and validated in 38 vineyards in the Po Valley (northern Italy), Baden‐Württemberg (Germany), and South Australia between 2005 and 2011. The model, which begins operating when the first ascocarp initials are formed, predicts on a daily basis the proportions of chasmothecia at the yellow, brown and black maturity stage. The initialization date was estimated with an iterative procedure that minimized the residuals of predicted versus observed values. In all vineyards, a drop to more favourable temperatures for ascocarp production over 2–4 days in the week or in the 2 weeks before the model initialization date probably triggered chasmothecia production. Model predictions provided a good fit of observed data (coefficients of determination, model accuracy, efficacy and efficiency were all ≥0·90), with some overestimation. When predicted production of black chasmothecia (on leaves) was compared with observed dispersal of chasmothecia from vines, lack of splashing rain was probably the main cause of overestimation. When observed numbers of yellow, brown or black chasmothecia on leaves were compared with model predictions, removal of the developing chasmothecia by rainfall was probably the main cause of overestimation. Inclusion of the effect of rainfall on the removal of immature and mature chasmothecia from the powdery mildew colonies could improve the model. The model could be used to time the application of fungicides or biocontrol agents for reducing ascocarp formation and reducing primary inoculum in the following season.  相似文献

Leaf blotch is a globally important disease of barley crops and other grasses that is caused by at least five host‐specialized species in the fungal genus Rhynchosporium. The pathogen R. commune (specialized to barley, brome‐grass and Italian ryegrass) has long been considered to reproduce only by asexual means, but there has been accumulating evidence for recombination and gene flow from population genetic studies and the detection of complementary MAT1‐1 and MAT1‐2 isolates in a c. 1:1 ratio in the field. Here, it is demonstrated that 28 isolates of the closely related species R. agropyri (on couch‐grass) and R. secalis (on rye and triticale), collected from Europe, were also either of MAT1‐1 or MAT1‐2 genotype and that the distribution of mating types did not deviate significantly from a 1:1 ratio. Evidence is then provided for MAT1‐1‐1 and MAT1‐2‐1 gene expression during mycelial growth for all three species. By contrast, 27 isolates of the more distantly related R. orthosporum (on cocksfoot) and R. lolii (on Italian and perennial ryegrasses) from Europe were exclusively of the MAT1‐1 genotype, and expression of the MAT1‐1‐1 gene could not be detected during mycelial growth. These data suggest that cryptic sexual cycles are more likely to exist for R. commune, R. agropyri and R. secalis than for either R. orthosporum or R. lolii. A phylogenetic analysis of partial MAT1‐1 idiomorph sequences resolved these five species into two distinct groups (R. commune, R. agropyri and Rsecalis versus R. orthosporum and R. lolii) but provided only limited resolution within each group.  相似文献

The ash dieback fungus, Hymenoscyphus fraxineus, a destructive, alien pathogen of common ash (Fraxinus excelsior), has spread across Europe over the past 25 years and was first observed in the UK in 2012. To investigate the relationship of the pathogen's population structure to its mode of arrival, isolates were obtained from locations in England and Wales, either where established natural populations of ash had been infected by wind‐dispersed ascospores or where the fungus had been introduced on imported planting stock. Population structure was determined by tests for vegetative compatibility (VC), mating type and single‐nucleotide polymorphisms (SNPs). VC heterogeneity was high at all locations, with 96% of isolate pairings being incompatible. Frequencies of the MAT1‐1‐1 and MAT1‐2‐1 idiomorphs were approximately equal, consistent with H. fraxineus being an obligate outbreeder. Most SNP variation occurred within study location and there was little genetic differentiation between the two types of location in the UK, or between pathogen populations in the UK and continental Europe. There was modest differentiation between UK subpopulations, consistent with genetic variation between source populations in continental Europe. However, there was no evidence of strong founder effects, indicating that numerous individuals of H. fraxineus initiated infection at each location, regardless of the route of pathogen transmission. The ssRNA virus HfMV1 was present at moderate to high frequencies in all UK subpopulations. The results imply that management of an introduced plant pathogen requires action against its spread at the continental level involving coordinated efforts by European countries.  相似文献

A recent study on fungal mating type genes revealed two MAT alleles within homothallic Sclerotinia sclerotiorum differentiated by an inversion, Inv? (inversion negative) and Inv+ (inversion positive). An analysis of mating type in closely related S. minor was conducted to shed light on the evolution of this MAT inversion. Inv? and Inv+ MAT alleles were identified in S. minor and were characterized. Both MAT alleles in S. minor were flanked by APN2 and SLA2, and consisted of two idiomorphs fused as in other homothallic ascomycetes. However, in the Inv+ MAT, the 3·6 kb MAT region was inverted relative to the Inv? MAT. Except for the inversion, both Inv? and Inv+ MAT in S. minor were equal in size and identical in nucleotide sequence. The MAT inversion in Inv+ S. minor was at exactly the same place as in Inv+ S. sclerotiorum and affected three of four MAT genes: MAT1‐1‐1 was truncated and MAT1‐2‐4 and MAT1‐2‐1 were inverted. Unlike S. sclerotiorum, expression of MAT genes did not differ between Inv? and Inv+ S. minor. The 250 bp inverted repeat motif that flanked the inverted MAT region in S. sclerotiorum and believed responsible for the MAT inversion was also found in S. minor, but was 256 bp. Depending on the MAT genes, 93–96% nucleotide identity was observed between Sclerotinia species. Both Inv+ and Inv? MAT S. minor and S. sclerotiorum isolates were commonly found in lettuce fields of Arizona along with MAT heterokaryons.  相似文献

The effect of soil solarization and Trichoderma harzianum on induced resistance to grey mould (Botrytis cinerea) and powdery mildew (Podosphaera xanthii) was studied. Plants were grown in soils pretreated by solarization, Tharzianum T39 amendment or both, and then their leaves were inoculated with the pathogens. There was a significant reduction in grey mould in cucumber, strawberry, bean and tomato, and of powdery mildew in cucumber, with a stronger reduction when treatments were combined. Bacillus, pseudomonad and actinobacterial communities in the strawberry rhizosphere were affected by the treatments, as revealed by denaturing gradient gel electrophoresis fingerprinting. In tomato, treatments affected the expression of salicylic acid (SA)‐, ethylene (ET)‐ and jasmonic acid (JA)‐responsive genes. With both soil treatments, genes related to SA and ET – PR1a, GluB, CHI9 and Erf1 – were downregulated whereas the JA marker PI2 was upregulated. Following soil treatments and B. cinerea infection, SA‐, ET‐, and JA‐related genes were globally upregulated, except for the LOX genes which were downregulated. Upregulation of the PR genes PR1a, GluB and CHI9 in plants grown in solarized soil revealed a priming effect of this treatment on these genes' expression. The present study demonstrates the capacity of solarization and T. harzianum to systemically induce resistance to foliar diseases in various plants. This may be due to either a direct effect on the plant or an indirect one, via stimulation of beneficial microorganisms in the rhizosphere.  相似文献

This study was designed to assess the reliability of grapevine leaf bioassays for predicting disease resistance on fruit in the field. The efficacy of various grapevine quantitative trait loci (QTLs) for conferring resistance to downy and powdery mildew was evaluated in bioassays and in a 2‐year field experiment for downy mildew. The resistance genes studied were inherited from Muscadinia rotundifolia (Rpv1 and Run1) and from American Vitis species through cv. Regent (QTLRgP and QTLRgD). In bioassays, genotypes carrying Run1 blocked powdery mildew development at early stages. Genotypes combining Run1 with QTLRgP displayed no greater level of resistance. For downy mildew, genotypes carrying Rpv1 and/or QTLRgD were more resistant than the susceptible cv. Merlot, and showed a high level of leaf resistance in the field (<10% severity). Disease levels on bunches were much higher than those on leaves, with a high variability between Rpv1 genotypes (1–48%). A Bayesian decision theory framework predicted that an OIV‐452 threshold of 5 in leaf bioassays allowed accurate selection of grapevine genotypes (P = 0·83) with satisfactory disease severity on bunches. Therefore, this study validates that the use of early bioassays on leaves, as currently performed by grapevine breeders, ensures a satisfactory level of resistance to downy mildew of bunches in the field.  相似文献

Potato early dying (PED) is a disease complex primarily caused by the fungus Verticillium dahliae. Pectolytic bacteria in the genus Pectobacterium can also cause PED symptoms as well as aerial stem rot (ASR) of potato. Both pathogens can be present in potato production settings, but it is not entirely clear if additive or synergistic interactions occur during co‐infection of potato. The objective of this study was to determine if co‐infection by V. dahliae and Pectobacterium results in greater PED or ASR severity using a greenhouse assay and quantitative real‐time PCR to quantify pathogen levels in planta. PED symptoms caused by Pectobacterium carotovorum subsp. carotovorum isolate Ec101 or V. dahliae isolate 653 alone included wilt, chlorosis and senescence and were nearly indistinguishable. Pectobacterium wasabiae isolate PwO405 caused ASR symptoms including water‐soaked lesions and necrosis. Greater Pectobacterium levels were detected in plants inoculated with PwO405 compared to Ec101, suggesting that ASR can result in high Pectobacterium populations in potato stems. Significant additive or synergistic effects were not observed following co‐inoculation with these strains of Vdahliae and Pectobacterium. However, infection coefficients of V. dahliae and Ec101 were higher and premature senescence was greater in plants co‐inoculated with both pathogens compared to either pathogen alone in both trials, and Vdahliae levels were greater in basal stems of plants co‐inoculated with either Pectobacterium isolate. Overall, these results indicate that although co‐infection by Pectobacterium and V. dahliae does not always result in significant additive or synergistic interactions in potato, co‐infection can increase PED severity.  相似文献

Powdery mildew (Erysiphe cruciferarum) is an important disease in oilseed rape crops worldwide, but of sporadic importance in most southern Australian crops. Six Brassica napus cultivars were exposed to E. cruciferarum simultaneously in four plant age cohorts. First symptoms of powdery mildew appeared 9 days after inoculation (dai) on the oldest plants [42 days after seeding (das)], but 44 dai in the youngest plants that were exposed to inoculum from sowing, although final disease severity did not differ with the plant age at exposure. The maximum level of pod peduncle infestation was unaffected by plant age (= 0.37) or cultivar (= 0.28). The effect of temperature was also investigated. The development of disease on plants was slower and final severity reduced at a day/night temperature 14/10 °C compared with 22/17 °C. In vitro, maximum growth of germ tubes from conidia of E. cruciferarum was at 15–20 °C and survival of conidia reduced by temperatures >30 °C. The results explain the sporadic nature of powdery mildew outbreaks in winter‐grown oilseed rape in Australia, where slow rates of infection occur when seasonal colder prevailing winter conditions coincide with the presence of younger plants, together curtailing rapid disease development until temperatures increase in late winter/early spring. These results explain why epidemics are most severe in the two warmer cropping regions, viz. the northern agricultural region of Western Australia and New South Wales. This study suggests that with increases in winter temperatures under future climate scenarios, earlier and more severe powdery mildew outbreaks in Australia will be favoured.  相似文献

Barley (Hordeum vulgare) was grown in different climatic environments with elevated [CO2] (700 vs 385 ppm), [O3] (60/90 vs 20 ppb) and temperature (24/19 vs 19/12°C day/night) as single factors and in combinations, to evaluate the impact of these climatic factors on photosynthesis and susceptibility to powdery mildew and spot blotch disease. No significant increase in net CO2 assimilation rate was observed in barley grown under elevated [CO2] at ambient temperature. However, this rate was positively stimulated under elevated temperature together with a slightly higher potential quantum efficiency of PSII, both at ambient and elevated [CO2], suggesting that photosynthesis was not limited by [CO2] at ambient temperature. When growing under elevated temperature or [O3], infection by the biotrophic powdery mildew fungus decreased, whereas disease symptoms and growth of the toxin‐secreting hemibiotrophic spot blotch fungus increased compared to ambient conditions, implying that climate‐induced changes in disease severity could be linked to the trophic lifestyle of the pathogens. Elevated [CO2] decreased powdery mildew infection but had no effect on spot blotch disease compared to ambient condition. However, the effect of elevated [CO2], [O3] and temperature did not act in an additive manner when combined. This led to a surprising disease development in the combination treatments, where powdery mildew infection increased despite the individual reducing effect of the climatic factors, and spot blotch disease decreased despite the individual promoting effect of temperature and ozone, emphasizing the importance of conducting multifactorial experiments when evaluating the potential effects of climate change.  相似文献

Grosmannia alacris is a fungus commonly associated with root‐infesting bark beetles occurring on Pinus spp. The fungus has been recorded in South Africa, the USA, France, Portugal and Spain and importantly, has been associated with pine root diseases in South Africa and the USA. Nothing is known regarding the population genetics or origin of G. alacris, although its association with root‐infesting beetles native to Europe suggests that it is an invasive alien in South Africa. In this study, microsatellite markers together with newly developed mating type markers were used to characterize a total of 170 isolates of G. alacris from South Africa and the USA. The results showed that the genotypic diversity of the South African population of G. alacris was very high when compared to the USA populations. Two mating types were also present in South African isolates and the MAT1‐1/MAT1‐2 ratio did not differ from 1:1 (χ2 = 1·39, = 0·24). This suggests that sexual reproduction most probably occurs in the fungus in South Africa, although a sexual state has never been seen in nature. In contrast, the large collection of USA isolates harboured only a single mating type. The results suggest that multiple introductions, followed by random mating, have influenced the population structure in South Africa. In contrast, limited introductions of probably a single mating type (MAT1‐2) may best explain the clonality of USA populations.  相似文献

This study aimed to assess the extent and distribution of Fusarium graminearum species complex (FGSC) diversity in rice seeds produced in southern Brazil. Four species and two trichothecene genotypes were detected among 89 FGSC isolates, based on a multilocus genotyping assay: F. asiaticum (69·6%) with the nivalenol (NIV) genotype, F. graminearum (14·6%) with the 15‐acetyldeoxynivalenol (ADON) genotype, and F. cortaderiae (14·6%) and F. meridionale (1·1%), both with the NIV genotype. Seven selected F. asiaticum isolates from rice produced NIV in rice‐based substrate in vitro, at levels ranging from 4·7 to 84·1 μg g?1. Similarly, two F. graminearum isolates from rice produced mainly 15‐ADON (c. 15–41 μg g?1) and a smaller amount of 3‐ADON (c. 6–12 μg g?1). One F. meridionale and two F. cortaderiae isolates did not produce detectable levels of trichothecenes. Two F. asiaticum isolates from rice and two from wheat (from a previous study), and one F. graminearum isolate from wheat, were pathogenic to both crops at various levels of aggressiveness based on measures of disease severity in wheat spikes and rice kernel infection in a greenhouse assay. Fusarium asiaticum and the reference F. graminearum isolate from wheat produced NIV, and deoxynivalenol and acetylates, respectively, in the kernels of inoculated wheat heads. No trichothecene was produced in kernels from inoculated rice panicles by any of the isolates. These findings constitute the first report of FGSC composition in rice outside Asia, and confirm the dominance of F. asiaticum in rice agroecosystems.  相似文献

Cashew powdery mildew is presently the most important disease of cashew trees in all Brazilian growing regions. Although it was described over a century ago, it had never threatened the Brazilian cashew industry until the first decade of the 21st century. Morphological and pathogenic evidence indicated the possibility of different pathogen species being involved in early and late types of cashew powdery mildew. This study was designed to elucidate this issue by comparing two different powdery mildew fungi occurring on cashew plants in Brazil according to the morphological characteristics, phylogenetic relationships with closely related powdery mildew fungi and pathogenic relationships. Based on morphology, molecular phylogenetics and pathogenicity on cashew, it was shown that two species of powdery mildew specimens are without question associated with cashew trees. One species, which infects young immature tissues such as shiny leaves, flowers and young fruits, is Erysiphe quercicola, while Erysiphe necator is associated exclusively with mature leaves. This is the first report of both E. quercicola and E. necator causing cashew powdery mildew, and the first detection of E. necator on cashew.  相似文献

Management of grape powdery mildew (Erysiphe necator) and other polycyclic diseases often relies on calendar‐based pesticide application schedules that assume the presence of inoculum. An inexpensive, loop‐mediated isothermal amplification (LAMP) assay was designed to quickly detect airborne inoculum of E. necator to determine when to initiate a fungicide application programme. Field efficacy was tested in 2010 and 2011 in several commercial and research vineyards in the Willamette Valley of Oregon from pre‐bud break to véraison. In each vineyard, three impaction spore traps were placed adjacent to the trunk. One trap was maintained and used by the grower to conduct the LAMP assay (G‐LAMP) on‐site and the other two traps were used for laboratory‐conducted LAMP (L‐LAMP) and quantitative PCR assay (qPCR). Using the qPCR as a gold standard, L‐LAMP was comparable with qPCR in both years, and G‐LAMP was comparable to qPCR in 2011. Latent class analysis indicated that qPCR had a true positive proportion of 98% in 2010 and 89% in 2011 and true negative proportion of 96% in 2010 and 64% in 2011. An average of 3·3 fewer fungicide applications were used when they were initiated based on spore detection relative to the grower standard practice. There were no significant differences in berry or leaf incidence between plots with fungicides initiated at detection or grower standard practice plots, suggesting that growers using LAMP to initiate fungicide applications can use fewer fungicide applications to manage powdery mildew compared to standard practices.  相似文献

The application of silicon to the roots or leaves reduces the severity of powdery mildew (Podosphaera xanthii) in melon but the latter treatment is less effective. This study compared key biochemical defence responses of melon triggered by P. xanthii after root or foliar treatment with potassium silicate (PS). Treatments consisted of pathogen‐inoculated or mock‐inoculated plants supplied with PS via roots or foliarly, as well as a non‐treated control. The activity of defence enzymes and the concentration of phenolic compounds, lignin and malondialdehyde were determined from leaf samples at different time points after inoculation. Pathogen‐inoculated plants irrigated with PS showed both an accumulation of silicon and primed defence responses in leaves that were not observed in pathogen‐inoculated plants either sprayed with PS or not treated. These responses included the anticipated activity of peroxidase and accumulation of soluble phenols, the activation of chitinase and repression of catalase, and the stronger activation of superoxide dismutase, peroxidase and β‐1,3‐glucanase. Moreover, the lignin concentration increased in response to inoculation, whereas the malondialdehyde concentration decreased. For the foliar treatment, however, only an increase in lignin deposition was observed compared with the control plants.
