Objective   To investigate the practical application and image quality of transcutaneous ultrasonography across the right flank for the diagnosis of mid- to late-gestation in cattle.
Procedure   Dairy cows of known pregnancy status were used to establish criteria for the identification of mid- to late-stage pregnancy by transcutaneous ultrasonography. Factors involved in the practical application of transcutaneous ultrasound for routine use in pregnancy diagnosis in dairy cattle were also examined.
Results   Transcutaneous ultrasound, using a 3.5-MHz sector transducer applied over the caudodorsal right flank fold, allowed rapid and detailed visualisation of bovine pregnancy (i.e. fetus, fetal fluids, placentomes and/or fetal membranes) and could be used to establish criteria for the diagnosis of pregnancy. The commonly available cattle holding facilities of the herringbone dairy, rotary herringbone dairy, AI race or crush, and walk-through dairy all allow safe access to the right flank of a dairy or beef cow. However, transcutaneous scanning over the right flank is difficult in modern rotary turnstile dairies.
Conclusion   A 3.5-MHz sector transducer applied over the caudodorsal right flank fold allows detailed visualisation of the bovine fetus in most types of cattle-holding facilities. Further investigation of the sensitivity and specificity of this technique over the entire gestation period and its possible use in ageing of the bovine fetus is merited.  相似文献

Objective   To determine bovine fetal characteristics significantly associated with increasing gestational age as measured via transcutaneous ultrasonography over the right flank.
Methods   The length of gestation at date of pregnancy diagnosis via transcutaneous and transrectal ultrasonography was determined for 224 dairy cattle by estimation from subsequent calving dates. A separate dataset was created for each measurable fetal characteristic (i.e. thoracic diameter, abdominal diameter, umbilical diameter, placentome length and placentome height) and risk factors significantly associated with gestational age at pregnancy diagnosis within each dataset, including the fetal characteristic, were identified.
Results   Abdominal diameter was the most frequently observed fetal characteristic and thoracic diameter was the least. Gestational age at pregnancy diagnosis (d) was significantly associated with fetal thoracic diameter (P < 0.01), abdominal diameter (P < 0.01) and umbilical diameter (P = 0.02) when measured via transcutaneous ultrasound. Within each model, sire breed, dam breed, dam age and/or calf sex were also significantly associated with gestational age. Gestational age at pregnancy diagnosis was not significantly associated with either placentome height or length (P > 0.05).
Conclusion   Fetal thoracic diameter, abdominal diameter and umbilical diameter were found to be significantly associated with gestational age between approximate days 73 to 190 of gestation. Transcutaneous ultrasonography may prove a useful method of estimating gestational age in the absence of accurate breeding records.  相似文献

Ultrasonography allows early and reliable pregnancy detection in several domestic animals. Transrectal sonography can be recommended in horses and cattle, transrectal or transcutaneous procedures in sheep, goats and pigs while transcutaneous ultrasound scanning is appropriate in dogs and cats. Three periods of time can be distinguished in the diagnosis of early pregnancy by ultrasound: the earliest phase, where signs of pregnancy can be found in some cases, but accuracy of diagnosis is very low; the succeeding period, where reliable diagnosis is possible, but results are often difficult to achieve and examination can be time consuming; the third phase in which ultrasound diagnosis is very accurate even under practice conditions and can be efficiently carried out on a large scale basis in breeding management. To achieve high accuracy in determining pregnancies a suitable ultrasound scanner with a good image quality and considerable expertise are required. Under field conditions ultrasonic pregnancy diagnosis should be possible: from Day 14 in the horse, from Day 25-28 in cattle, from Day 25 (transrectal), resp. 35 (transcutaneous) in sheep, goats and swine and from Day 24-25 (transcutaneous) in the dog and cat. Before this period of time reliable diagnosis is sometimes possible: between Day 11 and 13 in the horse, between Day 20 and 25 in cattle, between Day 20 and 25 (transrectal), resp. between Day 30 and 35 (transcutaneous) in sheep and goats, between Day 20 and 25 (transrectal) and Day 22 and 35 (transcutaneous) in swine and between Day 19-20 and 24-25 in the dog and cat. Earlier sonographic indications of pregnancy are not sufficiently reliable for large scale accurate pregnancy diagnosis.  相似文献

AIM: To define the incidence rate of pregnancy loss and risk factors for those losses in pasture-fed dairy cattle in the Waikato region of New Zealand.

METHODS: Cows (n=2,004) from 10 pasture-fed, spring-calving dairy herds in the Waikato were enrolled following confirmation of pregnancy 29–45 days after insemination, for inseminations that occurred within the first 16 days of the seasonal breeding period. Transrectal ultrasonographic examinations for pregnancy were conducted at approximately 6, 8, 10, 14 and 22 weeks gestation, and subsequent calving data were recovered. Pregnancy loss was defined as having occurred when a confirmed pregnancy was not rediagnosed, when gross abortion was detected, or when a cow calved <265 days after the confirmed conception date. Data were analysed using reverse stepwise logistic regression and Cox's proportional hazards analysis.

RESULTS: A total of 128 (6.4%) pregnancy losses were detected. The incidence rate was higher in early compared to late gestation (10.9 vs 2.8 losses/10,000 cow-days between Weeks 6–10 vs Weeks 10–14, respectively; p<0.001). Higher rates of loss were associated with the occurrence of clinical mastitis (Hazards ratio (HR)=1.57; p=0.071), being treated for anoestrus (HR=1.69; p=0.007), and in cows that had calving-to-conception intervals ≤63 days compared with those that had calving-to-conception intervals >92 days (HR=2.49; p=0.06). In addition, the rate of pregnancy loss differed between herds (p=0.05).

CONCLUSIONS: The highest rate of pregnancy loss occurred in early gestation. Clinical mastitis, anoestrus and calving late in the calving season were risk factors for pregnancy loss.

CLINICAL RELEVANCE: Pregnancy diagnosis using ultrasonography can be undertaken from 28 days post-insemination. However, due to the high rate of pregnancy loss at this stage of gestation, herdowners need to be warned of possible losses, and cows should be re-examined to confirm pregnancy before certification of pregnancy status is given.  相似文献

The use of ultrasonography for pregnancy diagnosis in the bitch   总被引:1,自引:0,他引:1
Twenty-three Greyhound bitches housed at 3 breeding kennels were examined for pregnancy via transabdominal palpation and ultrasonography. Pregnancy was timed from the calculated day of ovulation (day of ovulation = day when first pup was whelped--63), and from a single breeding date (day 0). Starting on day 10 after ovulation, 9 bitches were monitored every 3 days by ultrasonography only, to determine gestational vesicle sizes during gestation and the time when fetal movements and heartbeats could be first detected. The other 14 bitches were examined by ultrasonography and transabdominal palpation on the same day every week, starting on postovulation day 19, to compare the effectiveness of the 2 methods of pregnancy determination. Parturition was the final determinant of pregnancy status. The earliest correct diagnosis of pregnancy was at 18 days after ovulation, but fetal movements and heartbeat could not be identified until days 28 and 35, respectively. Estimation of fetal numbers by ultrasound or palpation was not reliable when there were more than four in the litter. Pregnancy and nonpregnancy were correctly determined by both methods in an increasingly greater number of bitches as gestation progressed, but ultrasonography was more accurate at all stages; on days 19 to 22, 26 to 30, 34 to 38, and greater than 40 after ovulation, correct diagnoses were made in 33%, 42%, 50%, and 75% of the bitches by palpation and in 42%, 67%, 75%, and 83% of the bitches by ultrasonography. The most common error was failure to detect pregnancy by palpation or ultrasonography in bitches with small litters and tense abdominal muscles.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The objective of the present study was to compare two commercially available blood-based pregnancy tests, namely BioPRYN, an ELISA for pregnancy-specific protein B (PSPB), and an ELISA for pregnancy-associated glycoprotein (PAG), for early pregnancy diagnosis in dairy cattle using transrectal ultrasonography as a gold standard. Transrectal ultrasonography was conducted 26-58 days after artificial insemination (AI) in 197 cattle from 19 farms. Concurrently, a blood sample was collected for determination of serum PSPB and PAG. Transrectal palpation was performed approximately 120 days after AI to verify that pregnancy was maintained. For PSPB and PAG, there were no significant differences (P>0.05) in sensitivity (98.0 and 97.8%), specificity (97.1 and 91.2%), positive predictive values (99.3 and 97.8%), negative predictive values (91.9 and 91.2%) and accuracy (97.8 and 96.4%). In conclusion, the two blood pregnancy assays were equally efficacious and were highly accurate (based on transrectal ultrasonography as the gold standard).  相似文献

This study was designed to establish the factors, if any, which could affect plasma pregnancy-associated glycoprotein-1 (PAG-1) expression in a study population of 87 pregnant, high-producing dairy cows. The factors examined were: semen providing breed (Holstein-Friesian vs Limousin), outcome of gestation (male vs female newborn, and singleton vs twin pregnancies), lactation number, milk production at pregnancy diagnosis, plasma progesterone concentration, season of gestation (warm period, March–November vs cool period, December–February), and day of gestation (40, 90, 120, 150, 180 and 210). Pregnancy was diagnosed by transrectal ultrasound on day 40 post-insemination and by palpation per rectum on days 90, 120, 150, 180 and 210. Blood samples were collected from each animal immediately before each pregnancy diagnosis. The relative contributions of the different factors on PAG-1 concentrations were evaluated by GLM repeated measures analysis of variance. No significant effects of the herd, foetal sex, milk production, lactation number and plasma progesterone concentrations were observed. In contrast, twin pregnancy, the use of Limousin semen and conception during the cool period were correlated with significantly increased plasma PAG-1 concentrations throughout gestation. Our data indicate that both cow well-being during early placental development, determined in our conditions by reduced heat stress when conception occurred in the cool season, and crossbreed pregnancies lead to improved PAG-1 production throughout the gestation period.  相似文献

AIMS: To determine the sensitivity (Se) and specificity (Sp) of pregnancy diagnosis using transrectal ultrasonography and an ELISA for pregnancy-associated glycoprotein (PAG) in milk, in lactating dairy cows in seasonally calving herds approximately 85–100 days after the start of the herd’s breeding period.

METHODS: Paired results were used from pregnancy diagnosis using transrectal ultrasonography and ELISA for PAG in milk carried out approximately 85 and 100 days after the start of the breeding period, respectively, from 879 cows from four herds in Victoria, Australia. A Bayesian latent class model was used to estimate the proportion of cows pregnant, the Se and Sp of each test, and covariances between test results in pregnant and non-pregnant cows. Prior probability estimates were defined using beta distributions for the expected proportion of cows pregnant, Se and Sp for each test, and covariances between tests. Markov Chain Monte Carlo iterations identified posterior distributions for each of the unknown variables. Posterior distributions for each parameter were described using medians and 95% probability (i.e. credible) intervals (PrI). The posterior median estimates for Se and Sp for each test were used to estimate positive predictive and negative predictive values across a range of pregnancy proportions.

RESULTS: The estimate for proportion pregnant was 0.524 (95% PrI?=?0.485–0.562). For pregnancy diagnosis using transrectal ultrasonography, Se and Sp were 0.939 (95% PrI?=?0.890–0.974) and 0.943 (95% PrI?=?0.885–0.984), respectively; for ELISA, Se and Sp were 0.963 (95% PrI?=?0.919–0.990) and 0.870 (95% PrI?=?0.806–0.931), respectively. The estimated covariance between test results was 0.033 (95% PrI?=?0.008–0.046) and 0.035 (95% PrI?=?0.018–0.078) for pregnant and non-pregnant cows, respectively. Pregnancy diagnosis results using transrectal ultrasonography had a higher positive predictive value but lower negative predictive value than results from the ELISA across the range of pregnancy proportions assessed.

CONCLUSIONS AND CLINICAL RELEVANCE: Pregnancy diagnosis using transrectal ultrasonography and ELISA for PAG in milk had similar Se but differed in predictive values. Pregnancy diagnosis in seasonally calving herds around 85–100 days after the start of the breeding period using the ELISA is expected to result in a higher negative predictive value but lower positive predictive value than pregnancy diagnosis using transrectal ultrasonography. Thus, with the ELISA, a higher proportion of the cows with negative results will be non-pregnant, relative to results from transrectal ultrasonography, but a lower proportion of cows with positive results will be pregnant.  相似文献

This study analyses anomalous cases of gestation ending in pregnancy loss during the early foetal period and their effect on progesterone and plasma pregnancy-associated glycoprotein-1 (PAG-1) concentrations. Data derived from a large-scale ultrasound pregnancy diagnosis programme in high producing dairy cows. Over a 3-year period (2004–2007), a very low incidence (0.5%: 15 of 3094) of anomalous pregnancies was recorded. The results revealed that the following anomalies were detected on days 35–41 of gestation in cows carrying singletons with one single corpus luteum: embryo death in eight cows (0.3%); and embryo in the uterine horn contralateral to the corpus luteum in seven cows (0.2%). All these animals suffered pregnancy loss during the early foetal period. In cows carrying dead embryos, no signs of conceptus degeneration were observed on pregnancy diagnosis. Amnion size (approximately 25 mm diameter) and uterine horn fluid contents were estimated to be similar to those of the normal pregnant cows in this period. In the contralateral gestations, live embryos were observed in all ultrasound checks before pregnancy loss. Uterine fluid contents increased in the two cows in which gestation continued for more than a week. In the cases of embryo death but not in those of contralateral gestation, a drop in PAG-1 levels was noted prior to pregnancy loss. Two cows carrying dead embryos increased with time allantoic fluid contents. The PAG-1 values increased with time in one cow bearing a dead embryo (from 2.31 to 6.79 ng/ml) and in two of the contralateral gestations (from 1.66 to 2.33 ng/ml and from 0.39 to 6.79 ng/ml, respectively). Results of this study indicate that the foetal membranes continue to undergo some activity following embryo death, and that contralateral pregnancy may determine failure of the gestation process.  相似文献

Early and accurate diagnosis of nonpregnant sows and gilts has the potential to increase reproductive efficiency and the financial income in pig production by reducing non-productive days per sow per year. The objectives of this study were to compare the efficiency of pregnancy diagnosis between Doppler Echo+ and real time ultrasonography (RTU) and to compare the efficiency by using RTU at different days post-mating (days 17 to 24) under commercial conditions. In the 1st study, using crossbreed sows and gilts (n=107), pregnancy diagnoses were done with Doppler Echo+ and then with RTU. Between 28 and 65 days of gestation, Doppler Echo+ had 85% sensitivity and 32% specificity, and efficiency was 73%. In the 2nd study, sows (n=142) were scanned for pregnancy diagnosis between 17 and 24 d post-mating (PD1) and reconfirmed between 38 and 45 days of gestation (PD2). After 21 days of gestation, RTU had over 90% sensitivity and 45% specificity, and 70% efficiency. Accuracy between PD1-farrowing was 75.5% and between PD1 and PD2 was 80.6%. In the 3rd study, sows were diagnosed pregnant by RTU (n=151) at 17 to 24 days of gestation or A-mode ultrasound (n=172) at 28 to 30 days of gestation. There were no significant differences in conception rate (P > 0.09) and farrowing rate (P > 0.67) between both groups. Hence, there was no improvement in fertility and farrowing rate by using RTU instead of A-mode ultrasound under commercial conditions.  相似文献

Eighteen cyclic Shiba goats were used in this study. Estrus was synchronized with a single injection of 125 microg of a synthetic analogue of prostaglandin F(2)alpha (PGF(2)alpha) after detection of at least one corpus luteum by B-mode ultrasonography. Blood samples were collected from each animal on days 0, 7 and 21 post-mating for progesterone assay. Animals in estrus were either allowed to be mated by fertile bucks twice during estrus (group I; n=12) or not at all (group II; n=6). Ultrasonographic examinations were performed transrectally or transabdominally using a real-time B-mode scanner equipped with a 7.5 or 5 MHz transducer. All animals exhibited estrus 56.0 +/- 2.7 h after injection of PGF2alpha. The results show that the accuracy of the progesterone assay in diagnosing pregnancy on day 21 after mating was 80% for pregnancy and 100% for non-pregnancy, retrospectively. Ultrasonographic examinations showed that gestational sac and embryos heartbeats were detected on days 20.2 +/- 0.6 and 24.3 +/- 0.7 of gestation, respectively. Placentomes were detected on day 35.4 +/- 1.0 of gestation as small nodules (0.7 +/- 0.2 cm in size). At two months pregnancy, skeletal structures like skull, thorax and long bones were clear. Biparietal diameter of the skull and length of long bones could be used as an estimate of gestational age. The accuracy of detection of fetal number using real-time B-mode ultrasonography was 91.7% on day 60 of gestation. In conclusion, progesterone assay at day 21 post-mating (cut-off value, 1 ng/ml) can be used for pregnancy diagnosis in goats. However, B-mode transrectal ultrasonography was more efficient due to detection of embryo and confirmation of its viability by heartbeats. In addition, fetal number and gestational age could be determined only by ultrasonography.  相似文献

In recent years, the interest in equine foetal gender determination (FGD) during gestation increased remarkably. Ultrasonographic FGD can be performed in two different periods during gestation. The earliest examination can take place at a gestational age of 60–70 days, whereby the genital tubercle is used to differentiate between male and female foeti. The time window of the second approach is wider (120–210 days), and there are more characteristics to take into consideration. In this article, the feasibility and accuracy of ultrasonographic FGD in mid‐ to late gestation are evaluated. One hundred twenty‐one mares from different breeds with a pregnancy stage between 120 and 270 days were examined once, using B‐scale ultrasonography (Esaote MyLab™ClassC). None of the mares were sedated nor shaved, and the procedure was completed within 15 min. Diagnosis was firstly based on the gonads. The final judgement was made based on all visible foetal reproductive organs. In three cases with a pregnancy stage beyond 257 days, FGD was not possible. All of the examined mares in which a FGD could be performed gave birth to a healthy foal. In 98% of the examinations (116/118), the diagnosis was correctly made. In both cases of misdiagnosis, only one characteristic was seen during the procedure and wrongly interpreted. Beyond 210 days of pregnancy, the extremities can preclude a good visualization of the inguinal region. In conclusion, equine FGD in mid‐ to late gestation is an accessible and accurate technique, although a good ultrasound device is a prerequisite and experience and expertise is necessary.  相似文献

Pregnancy diagnoses were performed under farm conditions in 201 cows (Hungarian Red Pied and crossbreds with Holstein) between 21 and 70 days after last insemination date, using a linear-array real-time ultrasound scanner with a 3.0 MHz rectal transducer. Identification of (a) sharply demarcated black area(s) and/or an embryo within the uterus were used as criterions for pregnancy. Pregnancy was confirmed by rectal examination at 6 and/or 8 weeks after A.I. From the total number of correct positive diagnoses (n = 129), incorrect positive diagnoses (n = 3, made on days 36, 40 and 44 after A I), correct negative diagnoses (n = 62) and incorrect negative diagnoses (n = 7, made on days 25, 28, 28, 29, 30, 31, and 33 after A I), a sensitivity of 94.8%, a specificity of 95,3%, a positive predictive value of 97.7% and a negative predictive value of 89.8% were calculated. From these preliminary results it was concluded that real-time ultrasound scanning is a useful and reliable technique for early pregnancy diagnosis in cows. It is suggested that a combination of milk/plasma progesterone estimation between days 21 and 24 and real-time ultrasound scanning between days 25 and 45 would allow the confirmation and quantification of (late) embryonic mortality in the cow.  相似文献

It is useful to determine the gestational age in sheep to provide essential information for effective flock management practices. The aims of this study were to evaluate the efficacy and practical aspects for using two ultrasound techniques, transabdominal and transrectal, in determining the gestational age in Württemberg ewes. Monitoring of embryo and fetus developmental stages during the ewes’ gestation was carried out with real time ultrasound using a transabdominal convex probe, frequency 3.5 MHz, and a transrectal linear probe, frequency 7.5 MHz. The size of the embryonic vesicle during the period from the 23rd to the 38th day of gestation can be used as a confirmational indicator of gestational age when the transrectal probe is used. The occipital nasal diameter correlated with the gestational age with both transabdominal (P < 0.05) and transrectal probes (P < 0.01) from the 46th to the 63rd day of gestation. The biparietal diameter of the fetal head measured by transabdominal probe during the period from the 46th to the 63rd day of gestation correlates with gestation age (P < 0.05). The diameter of the fetal eye orbit monitored by transrectal probe from the 46th to the 63rd day of gestation also correlated well with gestational age (P < 0.05).  相似文献

AIMS: To record the prevalence of gross abnormalities of the reproductive tract in culled New Zealand dairy cows, to determine how accurately farmers classify the pregnancy status of their animals and to establish if this was influenced by method of pregnancy diagnosis. METHODS: The reproductive tracts from 1134 cull dairy cows were examined after slaughter and evisceration for the presence of gross abnormalities, ovarian activity and pregnancy at a commercial abattoir. The farmers that had submitted these animals for slaughter were surveyed for information about the farm and herd from which each cow was derived and to establish whether the farmer believed each cow to be pregnant or not. The method that had been used to determine pregnancy status was recorded for each animal. RESULTS: Gross abnormalities were evident in 5.7% of reproductive tracts. Ovarian activity (presence of follicles 5 mm diameter and/or a corpus luteum) was apparent in 88% of non-pregnant cows. Pregnancy was detected in 39% of cows, of which 2.3% carried twins. The pregnancy status evident at slaughter varied from that reported by farmers in 7.0% of the 954 cows for which farmers were able to provide information. Of the cows that had been examined by palpation or ultrasound per rectum prior to slaughter, 10.3% that were recorded as non-pregnant by farmers were pregnant, and 3.2% of those recorded as pregnant were not. Of the cows that had not been examined, 3.8% of those recorded as nonpregnant by farmers were pregnant while 10.4% of those recorded as pregnant were not. There was no apparent association between gross genital tract abnormalities or ovarian activity and the misclassification of pregnancy status. Amongst cows that were pregnant at slaughter the foetus was significantly smaller in cows that had been recorded as 'not pregnant' after palpation or ultrasound examination than in cows that had been recorded as 'not pregnant' on the basis of farmer observation only. CONCLUSIONS: The prevalence of gross abnormalities of the reproductive tract was comparable to that reported in similar studies overseas. Farmer observation as a method of pregnancy detection overestimates pregnancy rate. Pregnancy status may be misclassified or misrecorded following palpation or ultrasound examination of cattle per rectum. Accurate classification of pregnancy status is dependent on the method and timing of pregnancy diagnosis and on minimising errors of diagnosis, cow identification and recording.  相似文献

In Brazil, great milk productivity was achieved after the implementation of a genetic improvement program. However, reproductive efficiency is still far from optimal, possibly due to the high number of undiagnosed disorders that may affect fertility. The aim of this study was to evaluate occurrences of the main reproductive disorders in dairy goats in southeastern Brazil. Data were collected between January 2015 and May 2017 from 23 commercial herds of different breeds, with goats ranging from 8 months to 12 years of age. Transrectal ultrasound exams were performed in 2680 goats. A total of 14.8% of the does showed a disorder in the reproductive tract: hydrometra (10.0%), ovarian follicular cysts (2.3%), gestational loss (1.5%), and hydrosalpinx (1.1%). This was the first study evaluating reproductive disorders in live animals that used a high number of Brazilian dairy goats. Considering that all these diseases affect fertility to different degrees, the performance of transrectal ultrasonography exams twice a year is strongly suggested, in order to guide precocious treatment or discard the animal as soon as possible, thus reducing economic losses in dairy goat farming.  相似文献

Objective— To assess agreement between ultrasonography (transcutaneous and transrectal) and standing radiography in horses with fractures in the pelvic region and disorders of the coxofemoral joint.
Study Design— Case series.
Animals— Warmblood horses (n=23) and 2 ponies.
Methods— Medical records (1999–2008) of equids with pelvic or coxofemoral disorders that had pelvic radiography and ultrasonography were retrieved and results of both techniques compared.
Results— Radiography and ultrasonography each identified equal numbers of fractures of the tuber coxa (n=4), ilial shaft (2), ischium (3), femoral neck (2), and osteoarthritis/osis of the coxofemoral joint (6). Fractures of the ilial wing (4) were only identified by ultrasonography not by standing radiography. Of 9 acetabular fractures, 3 were identified on radiographs only, 5 were identified with both modalities. One pubic fracture was identified using ultrasonography and radiography. One acetabular and 1 pubic fracture were only diagnosed on necropsy.
Conclusions— We found reasonable agreement (73%; 24/33) between ultrasonography and standing radiography for diagnosis of pelvic–femoral disorders. Ultrasonography was more useful for ilial wing fractures and radiography for acetabular fractures.
Clinical Relevance— Ultrasonography is a rapid, safe imaging technique for detecting disorders of the pelvic region with a high diagnostic yield and is a preferred initial approach in horses with severe hindlimb lameness.  相似文献

This study was designed to establish possible factors affecting plasma pregnancy-associated glycoprotein (PAG) concentrations during early pregnancy in high producing dairy cows with live fetuses. Blood samples were obtained on days 35, 42, 49, 56 and 63 of gestation from 80 lactating cows in two herds carrying live fetuses. Radioimmunoassay systems were used to determine PAG (RIA-497 and RIA-706) and progesterone concentrations. We evaluated the effects on PAG concentrations of herd, lactation number, sire of fetus, day of gestation, fetus number, plasma progesterone and milk production at each time point established, along with possible paired interactions. Mean milk production per cow approached 41kg during the study period. PAG concentrations were not affected by herd, lactation number or plasma progesterone concentration. Significant positive effects on PAG concentrations were shown by the gestation day, and the interaction between day of gestation and twin pregnancy. Significant differences between bulls and a significant negative correlation between milk production and PAG values on day 63 of pregnancy were also detected. Proportions of blood samples showing undetectable PAG levels and false negative diagnoses throughout the study period were significantly higher (P<0.001) using the RIA-497 system (2.5% and 5.3%, respectively) compared to RIA-706 (0% and 0.8%, respectively). Our findings suggest that PAG concentrations during the early fetal period are related to the day of gestation, milk production, number of fetuses and sire of fetus in high producing dairy cows. Under our working conditions, the RIA-706 method was better at detecting plasma PAG molecules than the RIA-497 system.  相似文献
