Ascaridia dissimilis, a roundworm in turkeys, has been noted with increased frequency in commercial turkeys. Because infected turkeys can shed A. dissimilis ova in their feces, the potential exists for the external surface of turkey eggshells to be contaminated with A. dissimilis ova. The objectives of this study were to determine the presence of and recover A. dissimilis ova on the external surface of the turkey egg. In Experiment 1, turkey eggs were collected from naturally infected flocks, and eggs were processed by a sodium hydroxide procedure to recover any A. dissimilis ova on the external egg surface. In Experiment 2, the external surface of the turkey eggs was inoculated with 116 A. dissimilis ova/g feces, and eggshells were sampled every 3 days until 28 days of incubation to assess the recovery of A. dissimilis ova from the eggshell. In Experiment 1, of the 36 eggs examined from a flock naturally infected with A. dissimilis, one egg had an A. dissimilis ovum on its external eggshell surface. Experiment 2 demonstrated that A. dissimilis ova can be recovered from the external egg surface after a 28-day incubation period in the incubator. Ova recovery declined from an average of 62 A. dissimilis ova/turkey egg at day 2 of incubation to an average of 3 A. dissimilis ova/turkey egg at day 28 of incubation.  相似文献

The lateral transmission of Histomonas meleagridis in turkeys was studied in floor pens without the presence of Heterakis gallinarum. Battery-reared poults (120) were transferred at 2 wk of age to concrete-floored floor pens with fresh pine shavings litter (40/group). One group received no exposure. In other groups, either 10% or 25% of the birds were inoculated per cloaca with cultured H. meleagridis (200,000/bird) and placed in the pens as seeder birds. Inoculated birds died at 10-18 days postinfection (PI) showing typical liver and cecal lesions of histomoniasis. Birds in the high-exposure group died of histomoniasis beginning 16 days PI and continuing to 100% mortality by day 23 PI. Birds in the low-exposure (LE) group died beginning on day 19 PI and continuing through day 31 PI. All but one LE bird alive on day 31 PI had severe liver and cecal lesions of histomoniasis at necropsy. There was no evidence of histomoniasis in unexposed birds. No cecal worms (H. gallinarum) were found at necropsy of dead birds or in unexposed birds at the end of the experiment. Even though H. gallinarum is the only known reservoir for H. meleagridis, these results suggest that lateral transmission of histomoniasis through a flock can occur readily through normal contact between uninfected birds and infected birds and their droppings in the total absence of cecal worms.  相似文献

During the 1984 to 1986 spring hunting seasons in Connecticut, viscera from 300 hunter-killed wild turkeys and blood samples from live-trapped wild turkeys were examined in order to establish a health profile on the State's wild turkey population. Seven species of endoparasites were recovered from 224 (75%) of 300 birds: Metroliasthes lucida, Ascaridia dissimilis, Heterakis gallinarum, Syngamus trachea, Capillaria species, Trichomonas gallinarum, and Eimeria species. The most prevalent parasites were A. dissimilis (52%) and M. lucida (37%). Although some turkeys harbored high intensities of these two helminths, there were no associated gross or microscopic lesions nor body weight changes. The prevalence of S. trachea, H. gallinarum, Capillaria and Eimeria species, which are potential pathogenic parasites in domestic and wild turkeys, was very low (less than 3%). Blood samples from 19 live-trapped wild turkeys were negative for hemoprotozoa and antibodies to 15 common bacterial and viral agents. Serum samples from 82 birds were negative for Mycoplasma gallisepticum and Mycoplasma synoviae. The survey indicates that the wild turkey population of Connecticut presently has little evidence of common infectious diseases and minimal prevalence of potentially pathogenic parasites.  相似文献

Between 2004 and 2008, 338 samples from 156 German turkey, chicken, and peacock flocks with suspected histomonosis (histomoniasis) were sent to the Institute for Poultry Diseases of the Free University Berlin. Most samples were from ceca or livers; the other samples were organ pools or were taken from other organs or the environment. In 108 samples from 65 flocks, histomonal DNA was detected by polymerase chain reaction (PCR). Tetratrichomonas gallinarum DNA was found in 5.3% of investigated samples from flocks infected with Histomonas meleagridis and in 27.4% of investigated samples from flocks that were not infected with H. meleagridis. For subtyping of the strains, the C-profiling method, a method used to analyze the internal transcribed spacer-1 (ITS-1) of the rRNA gene, was modified to be more specific for H. meleagridis. Results showed the presence of more than the three subtypes described so far. There was no clear correlation between the subtype and the host. By C-profiling the clonal cultures, heterogeneous ITS-1 sequences were shown to probably result from intragenomic differences between rRNA genes.  相似文献

The spread of Histomonas meleagridis infections through groups of turkeys in the absence of the cecal worm vector (Heterakis gallinarum) was studied in a battery cage model. Battery-reared poults were exposed at 2 wk of age by commingling with infected birds into cages that had the floor lined with paper. One treatment received no exposure, whereas other birds were commingled with two, three, or four birds/cage (25%, 37.5%, or 50%) inoculated per cloaca with cultured H. meleagridis (200,000/bird). Inoculated birds died at 7-13 days postinoculation (DPI) showing typical liver and cecal lesions of histomoniasis. By 14 DPI, 87.5% of the directly inoculated birds died or had severe lesions of histomoniasis. Turkeys commingled with two, three, or four infected birds became infected at the rate of 72%, 80%, or 75%, respectively. In another experiment, two birds/cage (25%) were inoculated with Histomonas from culture and allowed to commingle with other birds for 1, 2, 3, or 4 days. Two of 12 (16.7%) birds had minor cecal lesions after contact with inoculated birds for 1 day, but 87.5%-100% became infected if inoculated birds remained in the cage for 2-4 days. Contemporaneous inoculation with cecal coccidia (Eimeria adenoeides) as a predisposing factor in blackhead infections was studied using the model. Turkey poults directly inoculated with Histomonas were allowed to commingle for 5 days with uninoculated birds that had received inoculation with 0, 10(3), or 10(4) sporulated oocysts. The coccidian infection appeared to interfere with transmission of blackhead infection by 7 DPI, as suggested by lessened severity of cecal lesions and a lower percentage of infected birds. These studies confirm that histomoniasis is transmitted readily from directly exposed young turkeys to others in the absence of the cecal worm vector, and that this phenomenon can be reproduced in battery cages as an experimental model.  相似文献

After its discovery in 1893 in Rhode Island, blackhead disease was reported across the continent and soon in many other countries. It decimated the turkey industry in New England and followed production like a faithful shadow. Blackhead disease causes high mortality in turkeys, sometimes approaching 100% of a flock. In chickens, the mortality may be 10%-20% with high morbidity, although many outbreaks pass unnoticed. Early workers identified Histomonas meleagridis, a protozoan related to Entamoeba histolytica, Giardia lamblia, and Trichomonas, as the causative agent. Like many other parasites, its life cycle is complex, involving as an intermediate host, the common cecal worm Heterakis gallinarum. The necessity for bacteria for Histomonas to become virulent in the turkey and chicken, notably Escherichia coli, Bacillus subtilis, and Clostridium spp., was discovered by research in gnotobiotic birds. Changes in management brought the disease under control, although it remained the first cause of mortality in turkeys until modern antihistomonal products were developed after WWII. The ban of nitroimidazole products in the United States and Europe was followed by an upsurge in reported cases in turkeys and chickens. Immunization is not an option for prevention, as birds do not reliably become resistant to reinfection after suffering a primary exposure. Recent research demonstrated that histomoniasis could spread rapidly through a flock of turkeys by direct contact, probably involving the phenomenon of cloacal drinking. Direct transmission was not demonstrated for chickens, stressing dependence on H. gallinarum as the source of infection. The lack of suitable treatment drugs or vaccines emphasizes the importance of prevention by worm control and management.  相似文献

Recurrent outbreaks of histomoniasis in flocks of 4-to-6-week-old white leghorn pullets is reported. In a typical outbreak, 5% of the pullets were stunted and listless with unkempt feathers. Mortality ranged from 2 to 3%, and the cull rate was approximately 2%. Ceca of affected chickens contained caseous cores. Histological examination of the ceca revealed fibrinonecrotic ulcerative granulomatous typhlitis associated with numerous histomonad trophozoites in the cecal wall. The outbreaks of histomoniasis were associated with heavy parasitism of pullets by Heterakis gallinarum. Litter had not been completely removed from the houses for the past 10 years, providing a constant source of Histomonas-infected larvated Heterakis ova.  相似文献

Third- and fourth-stage Ascaridia dissimilis larvae were isolated from commercial white turkey intestinal scrapings from two farms that were experiencing high mortality. Lesions consisted of a necrotic-like enteritis that was most severe in the jejunum. Subsequent bacteriological isolation yielded heavy growth of Escherichia coli and Clostridium perfringens. The rate of mortality declined rapidly when the turkeys were administered 18 ppm fenbendazole for 7 days.  相似文献

Outbreaks of Pasteurella anatipestifer infections in California turkey flocks were investigated and found to have a seasonal distribution, with a peak incidence in fall, coinciding with peak Culex mosquito populations. An experiment was conducted to test the hypothesis that mosquitoes may serve as vectors for P. anatipestifer infections in turkeys. Four 7-week-old turkey poults were exposed for 7 days to mosquitoes captured from turkey barns during a field outbreak of P. anatipestifer serotype 1 infection. One turkey developed serum antibodies to serotype 1, detectable by enzyme-linked immunosorbant assay, and was resistant to an intravenous inoculation of P. anatipestifer serotype 1 at 4 weeks postexposure. Giemsa-stained blood smears from this bird and from three 7-week-old turkeys inoculated intravenously with P. anatipestifer revealed the presence of rod-shaped bacteria in or on the surface of host erythrocytes. No such rod-shaped bodies were found on erythrocytes of an uninoculated control turkey.  相似文献

Hepatic foci are a serious economic problem for most turkey-producing areas in the United States. Current estimates indicate that as much as 43% of the flocks sent to slaughter may experience condemnations because of hepatic foci. The present experiments were designed to duplicate naturally occurring lesions with Ascaridia dissimilis. Newly hatched poults were placed on fresh litter and given feed containing either 500 embryonated A. dissimilis ova/bird/day (from day of hatch) or no ova, in three experiments. Hepatic foci were reproduced in exposed poults in all three experiments, indicating that A. dissimilis is directly involved in the etiology of hepatic foci.  相似文献

Histomoniasis (histomonosis, infectious enterohepatitis, or blackhead) is a disease of turkeys on litter or range caused by the protozoan Histomonas meleagridis, a parasite of worms, primarily spread in feces, in Heterakis gallinarum (cecal worm) eggs, or in Eisenia foetida (earthworms). In this trial, Natustat (Alltech, Inc., Nicholasville, KY), a proprietary plant-derived product, was used at 1.925 kg/tonne and compared with nitarsone in male hybrid turkey diets to 42 days of age on histomonad infected litter (day 7) from broiler breeders. Infected nonsupplemented and uninfected nonsupplemented control groups were also included. Natustat and nitarsone significantly improved 28- and 42-day feed conversion ratios and lowered 28- and 35-day cecal and liver lesion scores compared with infected nonsupplemented turkeys. The body weight at 42 days was greater in the Natustat and nitarsone supplemented groups than in the infected nonsupplemented group.  相似文献

The importance of preparation technique, culture media and incubation time in the embryonation of the infective egg stages of the intestinal nematode parasite Heterakis gallinarum was studied. Mature H. gallinarum worms were isolated from the caeca of infected chickens and separated by sex. In a first experiment intact female worms were kept for the development of their eggs in four different media (0.5% formalin, 2% formalin, 0.1 N sulphuric acid, 0.1% potassium dichromate) and incubated under constant temperature (20-22 degrees C) for 2, 4, 6 or 8 weeks. Afterwards the body of the worms were ruptured and the numbers of unembryonated and embryonated eggs were determined using a McMaster egg counting chamber, and the percentage of embryonated eggs was calculated. After 8 weeks of incubation in 0.5% formalin, 0.1 N sulphuric acid or 0.1% potassium dichromate 27.6%, 26.7% and 29.4% of the eggs, respectively, embryonated into third stage larvae (p > 0.05). In contrast, incubation in 2% formalin resulted in an embryonation of 18.6% only (p < 0.05). In a second experiment H. gallinarum eggs were directly harvested from worm uteri and cultivated afterwards in different media (2% formalin, 0.1 N sulphuric acid, 0.1% potassium dichromate) at 20 to 22 degrees C for 6 weeks. An incubation of isolated eggs in 2.0% formalin or 0.1% potassium dichromate during 6 weeks resulted in a significantly higher percentage of embryonation in comparison to the incubation of intact worms (first experiment). The results suggest that preparation technique, media and time of incubation has an essential influence on the development rate of H. gallinarum eggs.  相似文献

During the second half of the year 2000, we have diagnosed a megabacterial-associated proventriculitis in 13 laying hens and 4 cocks from 14 different flocks and one turkey. All birds were submitted for necropsy because of progressive runting, increased mortality and poor laying performance in the herds. Routine diagnostic workup included necropsy, histology, bacteriology, parasitology and virology. At necropsy, the proventricular of the birds were enlarged. The walls were thickened and the mucosa covered with cloudy, grey-white mucus. Petechial haemorrhages and ulcerations predominantly at the proventricular-ventricular junction and sloughing of the necrotic koilin layer of the ventriculus were inconsistently present. Megabacteria (approximately 40-50 by 2-4 microns, gram-positive to gram-labile, rod-shaped organisms) were present in large numbers in touch preparations of the proventricular mucosal surface. In some of the preparations, the megabacteria showed a branching pattern similar to fungal hyphae. By histopathology, all birds showed moderate to marked, diffuse lympho-plasmacytic proventriculitis. Heterophilic exudation was present in the terminal portion of the proventriculus, where the greatest numbers of the organism were found. Megabacteria were present in the mucus covering the mucosal surface and the lumina of the superficial proventricular crypts, occasionally invading the luminal epithelium. Megabacterial infection was in general associated with other diseases such as avian tuberculosis, salmonellosis, coccidiosis, chlamydiosis and various other bacterial and parasitic infections. Only in one hen no other concurrent infectious organism was detected. The turkey had histomoniasis. Wild birds were regarded as probable source of infection, since all affected birds were kept under conditions allowing contact to wild birds and we have diagnosed megabacterial infections in wild-living green finches earlier.  相似文献

1. A study was conducted to evaluate the effects of ochratoxin A (OA) on broiler chicks challenged with Salmonella gallinarum. 2. One hundred and seventy-six 1-d-old broiler chicks were divided into two groups of 88 chicks each, with one group fed on a control mash diet and the other given a mash diet containing 2 ppm OA. On d 14, each group was further subdivided into two groups with one group infected with S. gallinarum and the other uninfected. 3. Following S. gallinarum inoculation on d 14, 4 birds from each group were killed at 1, 2, 3, 5, 7, 10, 14 and 21 d post inoculation. 4. S. gallinarum infection caused dullness, depression, weakness, increased thirst, droopy wings, ruffled feathers and greenish-yellow diarrhoea. S. gallinarum infection in the absence of OA caused 11.5% mortality which increased to 28.8% in the presence of OA. 5. Decreased body weight and reduced feed intake were observed in chicks fed on the diet containing OA. S. gallinarum infection also reduced the body weights of chicks, with the effects being more marked in chicks receiving OA. The OA diet led to increased serum levels of aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, uric acid and creatinine, and decreased levels of total proteins, albumin, globulins, calcium and phosphorus. S. gallinarum infection did not cause significant alteration in any of the serum biochemical parameters. 6. Mortality and the severity of S. gallinarum infection in broiler chicks were increased by the presence of OA in the diet.  相似文献

Histomonas meleagridis infection of turkeys is usually accompanied by a severe disease with unspecific clinical symptoms but with distinct pathological lesions in the ceca and liver. In the literature some macro- and microscopic evidence of the spread of histomonads to the other organs has been provided. The aim of the present investigations was to use real-time polymerase chain reaction (PCR) to demonstrate the dissemination of H. meleagridis DNA to different organs after natural and experimental infection of meat turkeys. Samples from several organs were collected from a meat-turkey flock, which proved to be naturally infected with histomoniasis, and examined for histomonad DNA by real-time PCR. Histomonad DNA was detected in all investigated ceca, livers, spleens, kidneys, and pooled brain swabs. Additionally it was found in 75% of investigated samples from bursae of Fabricius, in 50% of investigated duodenums, and in 40% of investigated jejunum samples. After experimental intracloacal infection of 3-wk-old turkey poults with 147,500 histomonads, similar samples were collected from all turkeys that died. After a 3-wk observation period the surviving birds, as well as the noninfected control group, were euthanatized and samples were taken. During the entire experimental period, 10 birds out the 20 infected birds died. Histomonad DNA was detected in all investigated ceca, livers, lungs, and hearts (100%) and almost all kidneys (90%) and bursae of Fabricius (80%). On the other hand, only 30% of examined spleens and 10% of brain samples revealed positive results. Surviving infected birds were euthanatized and necropsied; histomonad DNA was found in one out of 10 livers but not in any ceca. Also, histomonad DNA could not be detected in examined cecal and lung samples from the noninfected control group.  相似文献

The major helminth parasites found in wild, semi-wild and golden Sovereign stock guinea fowl were Heterakis gallinarum, Ascaridia galli, Capillaria caudinflata, Raillietina tetragona and R. echinobothrida, while Eimeria species was the most important gastro-intestinal protozoan parasite. The incidence of the latter was higher in the semi-wild stock than in the wild stock. Necropsy of dead guinea-fowl indicated that A. galli, H. gallinarum and Eimeria species were indeed responsible for their deaths, especially in the young birds. Parasites found in blood smears were Leucocytozoon sp., Plasmodium sp. and Aegyptianella pullorum. The only tick found, Argas persicus, was on a few semi-wild stock, while lice of genus Damalinia were found only on wild birds.  相似文献

An indirect fluorescent antibody test was used to detect Bluetongue virus (BTV) antigen in tissue smears prepared from laboratory-infected Culicoides variipennis. Insects were infected by intrathoracic inoculation and virus was detected in head, thorax and abdomen, in general increasing in titre over a period of 9 days, at which time the experiment was terminated. Reliable positive results by fluorescence were obtained from the fifth day post infection in head, thorax and abdomen smears. Since high titres of virus were found in the head, smears could therefore be prepared from the head alone, leaving the remainder of the insect for further tests.The technique iis useful in experiments involving the screening of large numbers of insects for possible BTV infection, although further work would be necessary to establish the reliability of the method for routine use.  相似文献

Bobwhites given heterakid eggs but no Sevin became infected with cecal histomonads, but there was no pathological histomoniasis. Quail given 50 microgram of Sevin (10 microgram/day) behaved normally, but at necropsy they had slightly discolored livers. Quail given various doses of heterakid eggs and Sevin (Sevin increasing from 2.5 to 50 microgram) and those given various doses of heterakid eggs and 10 microgram/day of Sevin developed pathological histomoniasis and mortality rates of 36 and 63%, respectively.  相似文献
