Data on enzyme polymorphism and blood group polymorphism were examined with special reference to molecular evolution, by using a statistic that depends on neither population structure nor other ecological factors. Evidence is provided for the existence of a renal erythropoietic factor, devoid of vasopressor activity, which upon interaction in vitro with normal serum yields erythropoietin. When undialyzed serum is used, erythropoietin inactivation develops in the incubation mixtures, and this inactivation appears to be dependent on an enzymatic component in preparations of the factor and on ions in serum. 相似文献

A tuberculin carbohydrate fraction, GAE, in sensitized animals induced a delayed type of skin reactivity and inhibited the migration of macrophages but failed to stimulate lymphocyte transformation in vitro. Tuberculin protein-containing fractions were active in each test. These results show that in vitro lymphocyte transformation is not necessarily a corollary of delayed type hypersensitivity. 相似文献

B-type lamins, the major components of the nuclear lamina, are believed to be essential for cell proliferation and survival. We found that mouse embryonic stem cells (ESCs) do not need any lamins for self-renewal and pluripotency. Although genome-wide lamin-B binding profiles correlate with reduced gene expression, such binding is not directly required for gene silencing in ESCs or trophectoderm cells. However, B-type lamins are required for proper organogenesis. Defects in spindle orientation in neural progenitor cells and migration of neurons probably cause brain disorganizations found in lamin-B null mice. Thus, our studies not only disprove several prevailing views of lamin-Bs but also establish a foundation for redefining the function of the nuclear lamina in the context of tissue building and homeostasis. 相似文献

A 37-kilodalton protein is synthesized at higher rates in the peripheral and central nervous system of newborn rats than in adult animals. As a specific response to denervation, the synthesis of the 37-kilodalton protein is increased in the mature peripheral and central nervous system; however, this protein accumulates only in the peripheral nervous system. The differences in accumulation of the protein correlate with the apparent differences in the ability of peripheral and central axons to regenerate. The synthesis of the 37-kilodalton protein is inhibited when proper innervation or reinnervation is established. 相似文献

A single genetic alteration, a guanine-to-cytosine transversion, is responsible for the acquisition of malignant properties by K-ras genes of two human tumor cell lines established from carcinomas of the bladder (A1698) and lung (A2182). As a consequence, arginine instead of the normal glycine is incorporated into the K-ras-coded p21 proteins at amino acid position 12. This mutation creates a restriction enzyme polymorphism that can be used to screen human cells for transforming K-ras genes. This approach was used to identify the mutational event responsible for the malignant activation of a K-ras oncogene in a squamous cell lung carcinoma of a 66-year-old man; this point mutation was not present in either the normal bronchial or parenchymal tissue or in the blood lymphocytes. Hence, malignant activation of a ras oncogene appears to be specifically associated with the development of a human neoplasm. 相似文献

Traditional reconstructions of the prehistory and early history of Southeast Asia contain two periods of contact between Southeast Asia and the West, these being the beginnings of the so-called Dongson Culture and the first historic state of Funan. There has been controversy as to whether the Western contacts which gave rise to the "Dongson Culture" came directly to north Vietnam around 800 B.C. or whether they were filtered through Chou China and reached north Vietnam about 300 B.C. In either case, primarily decorative patterns, ultimately from the European Bronze and Early Iron Age, and bronze-working came in together. The "Dongson" patterns spread over much of Southeast Asia and are still being used today in some areas. Research by historians and geographers indicated that the Kingdom of Funan existed somewhere in coastal Mainland Southeast Asia. From this work it was apparent that Funan was in some way connected with the trade between China and the West during the first millennium A.D. up until the end of Funan around 600 A.D. In both of these models of contact Southeast Asians were the passive recipients of whatever came from the West. 相似文献

The distribution in chicken retina of the alpha subunit of transducin, the guanine nucleotide--binding protein that couples light-dependent activation of rhodopsin with activation of guanosine 3',5'-monophosphate phosphodiesterase, was determined with the aid of a specific antiserum. alpha-Transducin was found in rod photoreceptor cells but was not detected in cones. These results show that rods and cones differ with respect to alpha-transducin content and suggest that the processes of phototransduction may differ correspondingly in rods and cones. 相似文献

Erythropoietin (Epo), the hormone that stimulates red blood cell production, is synthesized in the kidney and liver in response to hypoxia. The human hepatoma cell line Hep3B regulates its production of Epo in a physiologic manner. Either hypoxia or cobalt chloride markedly increases expression of Epo mRNA as well as production of biologically active and immunologically distinct Epo protein. New protein synthesis is required before the induction of increased levels of hypoxia- or cobalt-induced Epo mRNA. Hypoxia, cobalt chloride, and nickel chloride appear to stimulate Epo production through a common pathway. The inhibition of Epo production at low partial pressures of oxygen by carbon monoxide provides evidence that a heme protein is integrally involved in the oxygen-sensing mechanism. This hypothesis is further supported by the finding that when heme synthesis is blocked, hypoxia-, cobalt-, and nickel-induced Epo production are all markedly inhibited. A model is proposed in which a ligand-dependent conformational change in a heme protein accounts for the mechanism by which hypoxia as well as cobalt and nickel stimulate the production of Epo. 相似文献

Glacial refuge areas are expected to harbor a large fraction of the intraspecific biodiversity of the temperate biota. To test this hypothesis, we studied chloroplast DNA variation in 22 widespread European trees and shrubs sampled in the same forests. Most species had genetically divergent populations in Mediterranean regions, especially those with low seed dispersal abilities. However, the genetically most diverse populations were not located in the south but at intermediate latitudes, a likely consequence of the admixture of divergent lineages colonizing the continent from separate refugia. 相似文献

Patients with cerebellar damage are known to exhibit deficits in the temporal control of movements. We report that these deficits are restricted to discontinuous movements. Cerebellar patients exhibited no deficit in temporal variability when producing continuous, rhythmic movements. We hypothesize that the temporal properties of continuous movements are emergent and reflect the operation of other control parameters not associated with the cerebellum. In contrast, discontinuous movements require an explicit representation of the temporal goal, a function of the cerebellum. The requirement for explicit temporal representation provides a parsimonious account of cerebellar involvement in a range of tasks. 相似文献
