Total calcium, total protein, albumin and globulin content were determined in 49 samples of mammary secretions from 37 crossbred and Thoroughbred mares. The mares were divided into three categories: Group 1--post partum samples from spontaneous full term Thoroughbred deliveries (n = 20); Group 2--pre- and post partum samples from spontaneous full term Thoroughbred deliveries (n = 6); Group 3--pre- and post partum samples from induced premature deliveries in crossbred mares (n = 11). Calcium concentrations of the mammary secretions proved useful in predicting full term and also in assessment of the chances of foal survival in prematurely induced parturition. Mean (+/- se) calcium levels in full term mares that foaled spontaneously (Group 1) were 10.6 +/- 2.7 mmol/litre on the day of parturition. However lower values than this did not necessarily mean parturition would not take place. In Group 2 mares, calcium levels were 4.1 +/- 0.8 mmol/litre just before foaling and rose to 9.2 +/- 1.5 mmol/litre within 20 mins of parturition; an increase of 124 per cent. Group 3 mares, induced prematurely, showed poor foal survival if pre-partum calcium levels were less than 3 mmol/litre. Total protein, albumin and globulin concentrations showed a similar trend but the increase just before or during parturition was much less marked.  相似文献

The status of the mare and foetus in relation to readiness for birth was assessed by measurement of the electrolytes sodium, potassium and calcium in mammary secretions pre-partum. Sixteen Thoroughbred mares were allowed to foal spontaneously and the ionic status of their mammary secretions was measured over three to five weeks pre-partum. From these measurements, a scoring system was developed where an ionic score of 35 points or more suggested that the mare was within 24 h of foaling. On the basis of this ionic score, 10 pony mares were induced with either oxytocin or fluprostenol and assessment of foal maturity was made by physical, behavioural and physiological criteria. Eight pony mares, induced when the ionic score was 35 points or more, delivered full term foals; two mares were induced when their scores were 30 and 20 points and delivered a full term and slightly immature foal respectively. These results suggest that foetal maturity may be related to electrolyte concentrations in mammary secretions and that an ionic score of 35 points or more may indicate that induction would be successful in terms of maturity of the newborn foal.  相似文献

The aim of this study was to test whether low dose oxytocin i.v. injection once a day to mares diagnosed as being ready for birth by mammary secretion calcium strip test measurements could be used as a reliable method to induce parturition and/or predict the mare would not foal during the following night if parturition did not occur within 2 h of treatment. Fifty-one near-term Haflinger mares were used and a single injection of 2.5 iu oxytocin was given between 1700 and 1900 h, including 10 mares used as controls which were administered a placebo. Administration of oxytocin resulted in the delivery of a normal foal within 120 min in 95% of mares. Twenty-four out of 38 (63%) treated animals foaled in response to the first oxytocin injection, 9 out of 38 (24%) in response to the second injection and 3 out of 38 (8%) in response to the third treatment. Two out of 38 (5%) treated mares foaled during the night irrespective of treatment whereas 7 out of 10 (70%) control mares foaled during the night. It was concluded that the major advantage of injecting a daily low dose of oxytocin appears to be that such a low dose induces delivery only in mares carrying a mature fetus and which are ready to foal.  相似文献

AIMS: To test published models for predicting time of foaling using data derived from prepartum mammary secretions and to develop a new model based on concentrations of mammary secretion constituents that accurately predicts time of foaling in Thoroughbred mares. METHODS: Concentrations of sodium, potassium, calcium, citrate and lactose were measured in prepartum mammary secretions of 20 Thoroughbred mares in the 2 weeks before expected date of foaling. Models to predict time of foaling were fitted to data based on absolute concentration, change in concentration, and percentage change in concentration of mammary secretion constituents in relation to actual intervals to foaling. RESULTS: Concentrations of potassium, calcium, citrate and lactose increased, and concentration of sodium decreased as foaling approached but variation between mares was large. Models to predict time of foaling based on percentage change in electrolyte concentrations were less accurate than those based on absolute concentration and change in concentration. When data from this study were fitted to two previously published models, the statistical sensitivity, specificity, and positive and negative predictive values of both models were lower than those originally reported. CONCLUSIONS: The use of prepartum equine mammary secretion electrolyte concentrations for prediction of time of foaling is unreliable, due to large variation in both absolute concentrations and change in concentrations between mares. Models that use a combination of mammary secretion electrolytes and physical and behavioural factors may better predict foaling than those based on mammary secretions alone.  相似文献

Fifty-six mares of lighthorse breeding were utilized in a controlled management scheme for induced daytime foaling. All mares had pre-foaling mammary secretions sampled for evaluation of water hardness (ppm) or calcium carbonate content (ppm). Sampling began 10 days prior to expected foaling date for each mare and was performed once daily for 3 days followed by twice daily until foaling occurred. Samples were diluted 1:6 in distilled water and tested by each of 3 methods: Sofchek™ Test Strips4, Predict-A-Foal™ Test Kit5 and Titrets™ Calcium Hardness Test Kit6. Mares were then either induced (i) to foal (no.= 33) according to a decision point of readiness for birth as indicated by pre-foaling mammary secretion testing (≥250 ppm water hardness by Sofchek™ test, ≥ 250 ppm calcium carbonate content by Titrets test, ≥ 4 color bar changes by Predict-A-Foal test), or allowed to foal spontaneously (s) (no.=23). There were no differences (p>0.05) in the mean pre-foaling mammary secretion test values (μi=293ppm, μs=329ppm; μi=4.1, μs=4.2; and μi=281ppm, μs=298ppm; for Sofchek, Predict-A-Foal and Titrets, respectively) between mare groups at the time of foaling (time=0) for any of the 3 testing methods employed. Mean intervals to foaling after reaching decision points of readiness for birth were different (p<0.05) between Sofchek and Titrets test for both mare groups, but only in the induced-to-foal group between Sofchek and Predict-A-Foal tests. Probabilities of 79%, 53% and 59% were calculated for mares foaling spontaneously within 24 hours of reaching the decision points used in this trial of readiness for birth on the initial occasion for Sofchek, Predict-A-Foal and Titrets™ tests, respectively. Each test was determined to have the ability to predict readiness of approaching parturition and found to be easily applicable to field use. No knowledge regarding the peripartum changes in mammary secretions in the jenny are presently available in literature. In the mare, instead, several studies report the role of these changes as indicators of foetal readiness for birth and impending parturition. This experiment was designed to determine calcium, sodium, potassium concentrations, and the value of sodium/potassium ratio in mammary fluids during prepartum in the jenny. Samples were daily collected by hand milking, after mammary gland size increased noticeably, from 17 Martina Franca jennies. Prepartum mammary secretions were analysed every other day between day 10 and day 2 antepartum, and then once a day from the day before to the day of parturition. Calcium concentration showed a significant increase between day 10 and day 6 antepartum and then between day 6 and days 4 and 2. Afterwards, another statistical significant increase was observed at parturition. Sodium concentration significantly decreased from day 10 to day 2 prepartum. Potassium concentration significantly increased between day 10 and day 8 before parturition, then showed a further increase at day 4, followed by none significant changes until foaling. All jennies showed a reversal in sodium/potassium ratio between 2 days antepartum and the day before. In conclusion, the evaluation of mammary fluid calcium concentrations and the reversal of sodium/potassium ratio could be used as good indicators of foetal maturity in the jenny. As far as the prediction of parturition is concerned, the reversal of sodium/potassium ratio is the best parameter, since it was detected 48–24 h before parturition in all considered animals.  相似文献

A study was designed to model the fluctuations of nine specific element concentrations in mammary secretions from periparturient mares over time. During the 1992 foaling season, serial samples of mammary secretions were collected from all 18 pregnant Arabian mares at the Michigan State University equine teaching and research center. Non-linear regression techniques were used to model the relationship between element concentration in mammary secretions and days from foaling (which connected two separate sigmoid curves with a spline function); indicator variables were included for mare and mare parity. Element concentrations in mammary secretions varied significantly during the periparturient period in mares. Both time trends and individual variability explained a significant portion of the variation in these element concentrations. Multiparous mares had lower concentrations of K and Zn, but higher concentrations of Na. Substantial serial and spatial correlation were detected in spite of modeling efforts to avoid the problem. As a result, p-values obtained for parameter estimates were likely biased toward zero. Nonetheless, results of this analysis indicate that monitoring changes in mammary-secretion element concentrations might reasonably be used as a predictor of impending parturition in the mare. In addition, these results suggest that element concentrations warrant attention in the development of neonatal milk-replacement therapies. This study demonstrates that non-linear regression can be used successfully to model time-series data in animal-health management. This approach should be considered by investigators facing similar analytical challenges.  相似文献

The purpose of this study was to observe the changes in body temperature before parturition using a wireless temperature monitoring device (WTMD) and to evaluate the usefulness of body temperature measurements using a digital rectal thermometer (DRT) and a microchip transponder thermometry device (MTTD) for predicting parturition in mares. The body temperatures using a WTMD at 0 hr and -1 hr were significantly different from those at the same time on Days 1-5 (P<0.01). The temperature differences between the morning of Day 0 and at -3 hr, -2 hr, -1 hr and 0 hr using the DRT and MTTD showed a significant drop compared with the temperature differences between the morning and evening of Days 1-5 (P<0.05). Furthermore, when the cutoff value of the temperature differences between the morning and other times was set to ≤0, the sensitivities of the DRT and MTTD in the evening of Day 0 and at -3 hr were 43% and 100% and 71% and 86%, respectively. The results suggested that monitoring the body temperature differences between morning and within 3 hr before the time of parturition is a valuable method for predicting parturition in mares. Conversely, this method would be more useful in predicting parturition when used in combination with other observations such as the mammary gland size and waxing of the teat ends because it has nearly a 20% probability of false-positive results prior to the day of parturition.  相似文献

Changes in mare mammary secretion composition were characterized prior to and after foaling. Eighteen mares were used for collection of mammary secretions from 23 days prepartum through 44 days postpartum. Concentrations of lactose, total protein, IgG and lysozyme (activity assay) were determined. Considerable variabilty was observed among mares. Mean lactose concentrations by mare were lower (P<.05) in the prepartum period compared with the postpartum period. Mean total protein and immunoglobulin G concentrations by mare were higher (P<.05) in the prepartum period compared with the postpartum period. Mean lysozyme activities by mare tended to be lower (P<.08) during the prepartum period compared with the postpartum period. Mammary secretion protein profiles, characterized by SDS-PAGE, were consistent with changes in concentrations of specific proteins measured in the secretions. Dynamic changes occurring in mammary secretions during the transition from prepartum accumulation of colostrum to postpartum production of milk include factors like IgG and lysozyme which have protective roles in the neonatal foal.  相似文献

The objective was to determine if induced mammary inflammation at cessation of milking influenced growth of gram-positive mastitis pathogens in mammary secretions, particularly during early involution. Growth of all mastitis pathogens evaluated was similar in cell-free fat-free mammary secretions from LPS-infused and control glands. These data indicate that intramammary infusion of LPS at cessation of milking did not alter growth of gram-positive mastitis pathogens in mammary secretion during the nonlactating period. Stage of lactation and the nonlactating period influenced bacterial growth and marked differences between bacteria and among strains of a bacterial species were observed. Staphylococcus aureus grew well in secretions collected during late lactation, but growth decreased during early- and mid-involution and increased again in secretions obtained near parturition. Streptococcus agalactiae and Strep. uberis grew better in mammary secretion obtained during involution than in secretions collected during late or early lactation. Streptococcus dysgalactiae grew well in mammary secretions at all time periods. These data demonstrate the variability of mastitis pathogen growth during physiologic transitions of the bovine udder.  相似文献

At 40 to 75 days of pregnancy, 20 multiparous mares of Quarter Horse breeding were blocked by expected foaling date and assigned to one of two treatment groups such that average age, body weight, body condition and percent body fat were similar between groups. Body condition was evaluated using a scoring system based on visual appraisal and palpable fat cover, with scores ranging from 1 to 9. Percent body fat was estimated by ultrasonographic scanning of rump fat thickness. The two treatment groups were: 1) control—fed to maintain and foal at a moderate condition (condition score 5.5 to 7.0) and 2) obese— fed to achieve and foal at an extremely high degree of body fatness (condition score 9). Condition score (p<0.001), body weight (p<0.01) and percent body fat (p<0.05) were greater for obese compared to control mares prior to and following parturition. Gestation was not affected by treatment. Parturition was observed and video taped from a raised platform located outside of a foaling stall. The duration of stages II and III of parturition, all intermediate times, the interval from birth of the foal to standing and nursing, and the degree of cervical and vaginal bruising incurred during parturition were not different between treatment groups. Foal birth weights and placenta weights did not differ between treatment groups, however, the nonfoal/non-placenta weight loss was greater in obese compared to control mares (33.2 vs 17.4 kg, SE = 3.6, p<0.05). Excessive body fat stored during gestation in the multiparous mare was not detrimental to parturition.  相似文献

Undiluted uterine secretion was used to determine the concentration of total protein and the accumulated volume of uterine secretion after a bacterial inoculation in mares susceptible and resistant to chronic uterine infection (CUI). The uterus of 6 susceptible and 5 resistant mares was inoculated with 5 x 10(6) Streptococcus zooepidemicus on the third day of estrus. Using a tampon inserted in the uterus, secretions were sampled at 5, 12, 24, and 36 hours after inoculation, followed by intrauterine lavage with phosphate buffered saline solution. The concentration of protein was determined in the undiluted secretion as well as in the uterine washing and the total amount of accumulated uterine secretion was calculated. Protein concentrations in plasma were compared before and after absorption by the tampon. Protein concentration of plasma before and after absorption by the tampon did not differ. Mares susceptible to CUI accumulated significantly (P less than 0.001) more fluid in the uterus than mares resistant to CUI, and uterine washings from the resistant mares were significantly (P less than 0.05) more dilute than those from the susceptible mares. Significant differences in protein concentrations between susceptible and resistant mares were not found. It was concluded from this study that the described method to sample undiluted uterine secretion was practical and reliable for the analysis of protein concentration. Various concentrations of uterine secretions in washings from susceptible and resistant mares emphasizes the importance in using undiluted uterine secretions or dilution markers in washings when intrauterine products are analyzed.  相似文献

Blood and mammary secretions were obtained from cows throughout the dry period. Quantitative and qualitative assays were performed to determine the cell types and cell distributions at weekly intervals from day of dry off until parturition. The total cell counts in secretions increased during involution and remained at high levels until a few weeks prepartum. The macrophages were the predominant cell type in mammary secretions whereas the numbers of lymphocytes were always less than neutrophils or macrophages. Enriched mononuclear cell populations derived from blood and mammary secretions were also evaluated using "T-cell rosette" assays. Changes observed in the relative distribution of three T-cell subsets in secretions did not reflect the dynamics of the cells in the peripheral blood. T-cell subsets that predominated in mammary secretions were the EN+ EAET+ and EN-EAET+ phenotypes. Distinct patterns of migration or differentiation of T-cell subsets were suggested by the changes of subsets observed in mammary secretions collected throughout the dry period.  相似文献

Twelve mares were allowed to foal naturally, after which they were monitored to study uterine involution. Starting on day 3 after parturition, the internal genital tract was examined per rectum manually and ultrasonographically every other day for changes in uterine characteristics and ovarian activity. By day 5, gravid and nongravid uterine horns were similar in size, and by day 7, uterine fluid was absent. On day 7 after parturition, endometrial biopsy samples were obtained for histologic evaluation, and uterine swab specimens were obtained for microbiologic culture. Uterine swab specimens from 10 of 12 mares had slight bacterial growth. The uteri of 8 of the 12 mares were histologically involuted by day 7. All mares ovulated 7 to 12 days after parturition. Concentrations of 13,14-dihydro-15-keto-PGF2 alpha (PGFM) were measured in jugular plasma samples obtained daily for 21 days after parturition. Concentrations of PGFM were low by the day after parturition, and there was no significant correlation between uterine involution and PGFM concentrations in these mares. All 12 mares were bred at the first estrus after parturition, and 9 became pregnant.  相似文献

Tear production was evaluated in 39 horses and 29 ponies using Schirmer tear test strips to determine whether diurnal or weekly fluctuations occur, whether location of strip placement has an effect, if values are the same for both eyes in an animal and whether sex, age, stabling vs. pasture and winter vs. summer had an effect. There was no test in which the raw score was less than 10 mm, although there were many occasions where tear wetting exceeded 35 mm. Analysis of the raw (continuous) scores by linear regression provided no evidence that signalment, housing or season or location of strip placement affected results. The distribution of tear test scores for a 'population' of eyes did not differ when the right eye was compared with the left eye or when the same eye was compared at different times on the same day. Individual test wetting values for opposing eyes measured at the same time, and also wetting values for the same eye measured at different times on the same day sometimes differed substantially. In winter maximum tear wetting exceeded 35 mm more frequently in the STT I than in the STT II even in housed horses and ponies, but there was no consistent significant difference. There appears to be wide variability in the STT I in normal horses and ponies.  相似文献

Ten dairy cows were infused intramammarily near drying off with concanavalin A (conA) or phytohemagglutinin (PHA). Mammary secretions were collected during physiologic transitions of the udder and were used in an in vitro microbiological assay to determine growth inhibition of mastitis pathogens. As mammary involution progressed, in vitro growth inhibition of Klebsiella pneumoniae, Escherichia coli, and Streptococcus uberis increased. Mammary secretions from conA- and PHA-treated glands had significantly increased bacterial growth inhibition. Secretions contained significantly increased concentrations of lactoferrin and a decreased citrate:lactoferrin molar ratio earlier in the dry period than did control mammary secretions. Greatest bacterial growth inhibition was observed in mammary secretions obtained 7 days before parturition. However, differences in secretion composition or bacterial growth inhibition were not found between conA- or PHA-treated and control udder halves during the prepartum period. Bacterial growth inhibition by mammary secretion decreased markedly during early lactation. A highly significant positive correlation was found between bacterial growth inhibition and concentrations of lactoferrin, serum albumin, and immunoglobulin G. A highly significant negative correlation was found in the citrate:lactoferrin molar ratio during early involution and the peripartum period.  相似文献

The objective of this study was to investigate the protein, amyloid A3 (AA3), in equine colostrum and early milk. We hypothesized that AA3 was consistently present in equine colostrum and early milk, that no correlation existed between serum and colostrum concentrations of this protein in individual mares at parturition and that colostrum/milk concentrations of this mammary protein may be affected by age, breed, length of gestation and/or induction of parturition. Thirty-eight peripartum mares and seven non-pregnant, non-lactating mares were included in the study. Mean serum concentrations of this protein in the pregnant and non-pregnant mares were consistent with previous reports. Amyloid A3 was found in all colostrum and early milk samples at consistently higher concentrations than in peripartum maternal serum. There was no correlation between serum AA and colostrum AA3 concentrations at parturition. Age and breed effects were not significant. Increased gestation length and induction of parturition were associated with decreased colostrum and milk AA3 concentrations. We conclude that AA3 is consistently present in equine colostrum and early milk. The production of this protein in the mammary gland is likely to be under different stimulus to the production of serum AA, and may have protective effects in the neonatal intestine.  相似文献

Gender determination of the equine fetus using transabdominal ultrasonography was studied in 20 mares. One group of 10 research mares was scanned repeatedly every 2 weeks from 100 days gestation to parturition, while the second group of 10 client mares was subjected to echography once during mid-gestation. In males, the penis and/or prepuce was observed on 71 occasions from 102 days to 258 days gestation. On cross-sectional views, the male external genitalia had a round shape with parallel linear echogenic foci up to approximately 140 days gestation and then appeared triangular. Fetal testes were oval in shape in frontal view and had an homogeneous ultrasonographic appearance. Females were diagnosed on 23 occasions from 118 days to 227 days gestation based on the presence of the mammary glands and teats. Fetal ovaries appeared homogeneous with a characteristic circular echo from 100 days to 134 days gestation. Gender identifications (n = 98) based on the presence of the penis and/or prepuce in males and mammary glands and teats or fetal gonads in females were all correct, in agreement with the sex of the foals at birth. The optimal window of time was defined in both sexes as 100 to 220 days gestation. Thereafter, it was increasingly difficult to identify the anatomical structures cited above. Fetal sex was mainly determined using the transabdominal approach (87/98). However, the transrectal approach was useful in cases in which fetuses were either in posterior presentation or located very high in the mares abdomen. Good quality diagnostic scanners used typically in equine reproduction and equipped with a 5.0 MHz probe can be used for this procedure up to 160 days gestation, after which a 3.5 MHz transducer is often necessary due to increasing fetal size.  相似文献

Two experiments were conducted to determine when a placental source of progestin was sufficient for maintaining pregnancy in the mare. In the first study, embryos were transferred into ovariectomized mares and pregnancy was maintained with altrenogesta Altrenogest treatment was terminated at either day 100 (n=6) or day 150 (n=6). Twelve ovarian-intact mares were assigned to a second experiment on day 100 of gestation. On day 160 of gesta- tion, these mares were assigned to one of three treatments: 1) ovariectomy on day 160 and given altrenogest to day 200 (n=4); 2) ovariectomy on day 180 and given altrenogest to day 250 (n=4); or 3) ovariectomy on day 200 and given altrenogest to day 300 (n=4). Blood samples were collected every 2 weeks from all mares in both experiments from day 100 to parturition and assayed for concentrations of pro- gestins. Pregnancy loss from day 100 to parturition was not different among groups in either experiment. Serum con- centrations ofprogestins in ovary-intact mares were greater (P<0.05) than those in ovariectomized pregnant mares until day 130, after which they were similar. Serum con- centrations of progestins in the ovariectomized pregnant mares rose gradually from day 100 until near parturition. Serum concentrations of progestins in the ovary-intact pregnant mares declined from day 100 to day 157, did not vary significantly from day 157 to day 245, then rose until near parturition. A study was conducted to determine the persistence of antibiotic preparations for use in nonlactating cows in bovine mammary secretions following intramammary infusion at cessation of milking. Five commercially available antibiotic formulations were evaluated using 311 cows. All quarters of each cow were sampled once only during the nonlactating period and most cows were sampled at or near parturition. Antibiotic residues were detected qualitatively by the Bacillus stearothermophilus disc assay. Great variation between different antibiotics in persistence in mammary secretion was observed. In general, mammary secretions from most mammary glands infused with cloxacillin or penicillin-dihydrostreptomycin were positive at 28–35 days after infusion and some were positive at 42–49 days after infusion. On the other hand, <13% of mammary secretions at 7 days after infusion of novobiocin and 50% of mammary secretions at 14 days after infusion of penicillin-novobiocin were positive for antibiotics. Cephapirin benzathine persisted for about 21 days after infusion. Some samples that were positive for antibiotics after initial testing were negative following heating of samples, suggesting that component(s) of dry secretion can inhibit growth of B. stearothermophilus and influence the interpretation of results. Colostrum samples from all quarters except one were negative for antibiotics. These data suggest that nonlactating-cow antibiotic formulations persist primarily during the early to mid-nonlactating period. Based upon present methods of formulation, it would appear that antibiotic preparations for use in nonlactating cows most likely provide little protection during the periparturient period, at a time when mammary glands are highly susceptible to new intramammary infections.  相似文献
