The objective of this study was to evaluate the effect of reduced-calorie avocado paste on lipid serum profile, insulin sensitivity, and hepatic steatosis in rats fed a hypercholesterolemic-high fructose diet. Thirty five male Wistar rats were randomly separated in five groups: Control group (ground commercial diet); hypercholesterolemic diet plus 60 % fructose solution (HHF group); hypercholesterolemic diet plus 60 % fructose solution supplemented with avocado pulp (HHF+A group); hypercholesterolemic diet plus 60 % fructose solution supplemented with reduced-calorie avocado paste (HHF+P group); and hypercholesterolemic diet plus 60 % fructose solution supplemented with a reduced-calorie avocado paste plus fiber (HHF+FP group). The A, P, and FP were supplemented at 2 g/kg/d. The study was carried out for seven weeks. Rats belonging to the HHF group exhibited significantly (P?≤?0.05) higher total cholesterol, triglycerides, and insulin levels in serum as well as lower insulin sensitivity than the control group. Supplementation with reduced-calorie avocado paste showed a significant (P?≤?0.05) decrease in total cholesterol (43.1 %), low-density lipoprotein (45.4 %), and triglycerides (32.8 %) in plasma as well as elevated insulin sensitivity compared to the HHF group. Additionally, the liver enzymes alanine aminotransferase and aspartate aminotransferase decreased significantly in the HHF-P group (39.8 and 35.1 %, respectively). These results are likely due to biocompounds present in the reduced-calorie avocado paste, such as polyphenols, carotenoids, chlorophylls, and dietary fibre, which are capable of reducing oxidative stress. Therefore, reduced-calorie avocado paste attenuates the effects of a hypercholesterolemic-high fructose diet in rats.  相似文献

Increased seaweed consumption may be linked to the lower incidence of metabolic syndrome in eastern Asia. This study investigated the responses to two tropical green seaweeds, Ulva ohnoi (UO) and Derbesia tenuissima (DT), in a rat model of human metabolic syndrome. Male Wistar rats (330–340 g) were fed either a corn starch-rich diet or a high-carbohydrate, high-fat diet with 25% fructose in drinking water, for 16 weeks. High-carbohydrate, high-fat diet-fed rats showed the signs of metabolic syndrome leading to abdominal obesity, cardiovascular remodelling and non-alcoholic fatty liver disease. Food was supplemented with 5% dried UO or DT for the final 8 weeks only. UO lowered total final body fat mass by 24%, systolic blood pressure by 29 mmHg, and improved glucose utilisation and insulin sensitivity. In contrast, DT did not change total body fat mass but decreased plasma triglycerides by 38% and total cholesterol by 17%. UO contained 18.1% soluble fibre as part of 40.9% total fibre, and increased magnesium, while DT contained 23.4% total fibre, essentially as insoluble fibre. UO was more effective in reducing metabolic syndrome than DT, possibly due to the increased intake of soluble fibre and magnesium.  相似文献

This study was designed to investigate the effects of long-term feeding of chitosan on plasma glucose and lipids in rats fed a high-fructose (HF) diet (63.1%). Male Sprague-Dawley rats aged seven weeks were used as experimental animals. Rats were divided into three groups: (1) normal group (normal); (2) HF group; (3) chitosan + HF group (HF + C). The rats were fed the experimental diets and drinking water ad libitum for 21 weeks. The results showed that chitosan (average molecular weight was about 3.8 × 105 Dalton and degree of deacetylation was about 89.8%) significantly decreased body weight, paraepididymal fat mass, and retroperitoneal fat mass weight, but elevated the lipolysis rate in retroperitoneal fats of HF diet-fed rats. Supplementation of chitosan causes a decrease in plasma insulin, tumor necrosis factor (TNF)-α, Interleukin (IL)-6, and leptin, and an increase in plasma adiponectin. The HF diet increased hepatic lipids. However, intake of chitosan reduced the accumulation of hepatic lipids, including total cholesterol (TC) and triglyceride (TG) contents. In addition, chitosan elevated the excretion of fecal lipids in HF diet-fed rats. Furthermore, chitosan significantly decreased plasma TC, low-density lipoprotein cholesterol (LDL-C), very-low-density lipoprotein cholesterol (VLDL-C), the TC/high-density lipoprotein cholesterol (HDL-C) ratio, and increased the HDL-C/(LDL-C + VLDL-C) ratio, but elevated the plasma TG and free fatty acids concentrations in HF diet-fed rats. Plasma angiopoietin-like 4 (ANGPTL4) protein expression was not affected by the HF diet, but it was significantly increased in chitosan-supplemented, HF-diet-fed rats. The high-fructose diet induced an increase in plasma glucose and impaired glucose tolerance, but chitosan supplementation decreased plasma glucose and improved impairment of glucose tolerance and insulin tolerance. Taken together, these results indicate that supplementation with chitosan can improve the impairment of glucose and lipid metabolism in a HF-diet-fed rat model.  相似文献

Advancing maize crop maturity is associated with changes in ear‐to‐stover ratio which may have consequences for the digestibility of the ensiled crop. The apparent digestibility and nitrogen retention of three diets (Early, Mid and Late) containing maize silages made from maize of advancing harvest date [dry matter (DM) contents of the maize silages were 273, 314 and 367 g kg?1 for the silages in the Early, Mid and Late diets respectively], together with a protein supplement offered in sufficient quantities to make the diets isonitrogenous, were measured in six Holstein–Friesian steers in an incomplete Latin square design with four periods. Dry‐matter intake of maize silage tended to be least for the Early diet and greatest for the Medium diet (P = 0·182). Apparent digestibility of DM and organic matter did not differ between diets. Apparent digestibility of energy was lowest in the Late diet (P = 0·057) and the metabolizable energy concentrations of the three silages were calculated as 11·0, 11·1 and 10·6 MJ kg?1 DM for the Early, Medium and Late diets respectively (P = 0·068). No differences were detected between diets in starch digestibility but the number of undamaged grains present in the faeces of animals fed the Late diet was significantly higher than with the Early and Mid diets (P = 0·006). The apparent digestibility of neutral‐detergent fibre of the diets reduced significantly as silage DM content increased (P = 0·012) with a similar trend for the apparent digestibility of acid‐detergent fibre (P = 0·078). Apparent digestibility of nitrogen (N) was similar for the Early and Mid diets, both being greater than the Late diet (P = 0·035). Nitrogen retention did not differ between diets. It was concluded that delaying harvest until the DM content is above 300 g kg?1 can negatively affect the nutritive value of maize silage in the UK.  相似文献

The purpose of this study was to evaluate the hypolipidemic effect of a methanolic extract from Opuntia joconostle seeds fed to mice in a hypercholesterolemic diet. Acute toxicity of the methanolic extract was investigated by an established method. Phenolic composition and antioxidant activity were determined by high-performance liquid chromatography and DPPH, respectively. The total phenolic content of Opuntia joconostle seeds was 47.85?±?1.29 mg gallic acid equivalents/g dry weight. The main phenolic compounds were identified as quercetin, rutin, and cafeic acid. Percent inhibition of DPPH+ was 49.76?±?0.49 %. The oral LD50 for the methanolic extract from the Opuntia joconostle seeds was >5,000 mg/kg BW. Mice fed a hypercholesterolemic diet for six days exhibited significantly (P?≤?0.001) higher plasma lipid levels than mice fed a normal diet. Remarkably, supplementation with methanolic extract from Opuntia joconostle at doses of 1, 2, and 5 g/kg body weight significantly (P?≤?0.001) prevented the increase in total cholesterol, low-density lipoprotein cholesterol, triglycerides level, and atherogenic index. Similar concentrations of the HDL cholesterol were observed in both treated and control groups. A significant dose-dependent reduction in lipid levels was noted for treated groups compared to the hypercholesterolemic group. We attribute this result to the seeds’ phenolic composition. This methanolic extract has potential to be included in short-term hypercholesterolemia treatment regimens as it exhibits hypolipidemic activity with no apparent toxic manifestations.  相似文献

This study aimed to examine the benefits of different amounts of omega-3 (n-3) polyunsaturated fatty acids from fish oil (FO) on lipid metabolism, insulin resistance and gene expression in rats fed a high-fructose diet. Male Wistar rats were separated into two groups: Control (C, n = 6) and Fructose (Fr, n = 32), the latter receiving a diet containing 63% by weight fructose for 60 days. After this period, 24 animals from Fr group were allocated to three groups: FrFO2 (n = 8) receiving 63% fructose and 2% FO plus 5% soybean oil; FrFO5 (n = 8) receiving 63% fructose and 5% FO plus 2% soybean oil; and FrFO7 (n = 8) receiving 63% fructose and 7% FO. Animals were fed these diets for 30 days. Fructose led to an increase in liver weight, hepatic and serum triacylglycerol, serum alanine aminotransferase and HOMA1-IR index. These alterations were reversed by 5% and 7% FO. FO had a dose-dependent effect on expression of genes related to hepatic β-oxidation (increased) and hepatic lipogenesis (decreased). The group receiving the highest FO amount had increased markers of oxidative stress. It is concluded that n-3 fatty acids may be able to reverse the adverse metabolic effects induced by a high fructose diet.  相似文献

Aronia melanocrpa fruit juice (AMFJ) used in our experiment was very rich in phenolic substances (709.3 mg gallic acid equivalents/100 ml juice). Anthocyanins (106.8 mg cyanidin-3-glucoside equivalents/100 ml juice) were the main flavonoid group. The aim of this study was to assess the influence of AMFJ on plasma lipids and lipoprotein profile, and histopathology of liver and aorta in rats with dietary-induced hyperlipidemia. AMFJ was administered by gavage for 30 days at doses of 5, 10 and 20 ml/kg body weight to rats fed a standard diet (SD) or a 4% cholesterol-containing diet (4% ChD). The 4% ChD caused a significant elevation of plasma total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) and triglycerides (TG). AMFJ did not significantly influence plasma lipids in rats fed the SD and significantly hindered the elevation of plasma TC, LDL-C and TG in rats fed the 4% ChD. High-density lipoprotein cholesterol (HDL-C) levels were not significantly influenced either by the 4% ChD or by AMFJ. Neither the cholesterol feeding, nor AMFJ treatment induced any histopathological changes in rat liver and aorta. In conclusion, AMFJ showed an antihyperlipidemic effect in rats with hyperlipidemia and could be valuable in reducing this factor of cardiovascular risk.  相似文献

In the present study, the objective was to evaluate the long-term metabolic impact of adding lactic acid to a bread-based diet in obese, hyperinsulinaemic Zucker (fa/fa) rats. All diets were based on a white wheat bread, and the lactic acid was added either prior to, or after the baking process. In addition, a diet with addition of Lactobacillus plantarum 299v was included to investigate the possible impact of probiotic lactic acid bacteria, in the absence of lactic acid. The intervention period was fourteen days and oral glucose tolerance tests (OGTTs) were performed before and after the intervention. Glucose, insulin and glucagon were measured during both OGTTs. Other parameters studied were blood lipids (total cholesterol and triglycerides) and liver cholesterol. The intervention period with the wheat bread baked in the presence of lactic acid improved glucose tolerance as judged from a 51% reduction (P=0.007) in the total glycaemic area. In contrast, there was no such improvement with the diet where lactic acid was added after baking or with addition of probiotic bacteria. No differences were seen between groups in insulin, blood lipids or liver cholesterol following the intervention. It is concluded that bread baked in the presence of lactic acid improves glucose metabolism in obese and hyperinsulinaemic Zucker rats.  相似文献

The present investigation was undertaken to study the effect of guargum on plasma cholesterol levels of overweight adults. The consumption of 15 g guargum with normal diet for six weeks produced significant reduction in plasma total cholesterol, LDL-cholesterol and total cholesterol to HDL-cholesterol ratio. Objective: This study is to evaluate the anti-obese effects of glucosamine (GLC) and chitosan oligosaccharide (COS) on high-fat diet-induced obese rats. Methods: The rats were randomly divided into twelve groups: a normal diet group (NF), a high-fat diet group (HF), Orlistat group, GLC high-, middle-, and low-dose groups (GLC-H, GLC-M, GLC-L), COS1 (COS, number-average molecular weight ≤1000) high-, middle-, and low-dose groups (COS1-H, COS1-M, COS1-L), and COS2 (COS, number-average molecular weight ≤3000) high-, middle-, and low-dose groups (COS2-H, COS2-M, COS2-L). All groups received oral treatment by gavage once daily for a period of six weeks. Results: Rats fed with COS1 gained the least weight among all the groups (P < 0.01), and these rats lost more weight than those treated with Orlistat. In addition to the COS2-H and Orlistat groups, the serum total cholesterol (CHO) and low-density lipoprotein cholesterol (LDL-C) levels were significantly reduced in all treatment groups compared to the HF group (P < 0.01). The various doses of GLC, COS1 and COS2 reduced the expression levels of PPARγ and LXRα mRNA in the white adipose tissue. Conclusions: The results above demonstrated that GLC, COS1, and COS2 improved dyslipidemia and prevented body weight gains by inhibiting the adipocyte differentiation in obese rats induced by a high-fat diet. Thus, these agents may potentially be used to treat obesity.  相似文献

A comparative analysis of the elasticity, microstructure and thermal stability of fibres (thickness ranging from 43.4 to 189.4 µm) isolated from pineapple leaves (PALF), coconut coir (COIR), banana leaf-stem (BAN) and oil palm empty fruit bunch (OPEFB) reported in this study. Statistical analysis of the mechanical properties derived from tensile test to rupture reveals significant differences (P<0.05) in the fibre strength (σ), stiffness (E) and extensibility (parameterized by the strain to rupture, ?). It is observed that COIR fibres yield the smallest strength, σ (=99.8±22.5 MPa), and stiffness, E (= 0.5±0.1 GPa), while PALF fibres yield the largest σ (=639.5±301.6 MPa) and E (=7.1±3.1 GPa); PALF fibres exhibit the smallest ? (=0.11±0.03) but OPEFB fibres yield the largest ? (=2.0±1.3). From scanning electron micrographs, it is observed that cellulose fibril rupture predominates in OPEFB, COIR and BAN fibres; a large proportion of the cellulose fibrils fail by pullout in PALF fibres. Thermogravimetric analysis reveals that all fibres are thermally stable up to 250 °C; the fibre residue ranges from 30 to 80 wt% after heating to 500 °C. In particular, BAN experiences the highest weight loss and PALF experiences the lowest weight loss. The findings lend to a simple approach for determining the performance of the composites by assessing the type of natural fibres for reinforcing polymeric matrices.  相似文献

Acidogenic diets, commonly measured by the potential renal acid load (PRAL), have been linked with metabolic diseases including insulin resistance, hepatic dysfunction, and cardiometabolic risk. Vegan diets are linked to low dietary acid loads, but the degree of adherence to a vegan diet to demonstrate this benefit is unknown. This study compared the change in PRAL and urine pH of omnivores who followed a vegan diet for either 2, 3, or 7 days over one week. Healthy adults were recruited from a campus population and randomly assigned to one of the three groups: VEG7 (vegan diet followed for seven consecutive days); VEG3 (vegan diet followed for three evenly spaced days over one week); or VEG2 (vegan diet followed for two evenly spaced days over one week). Gender, age, and body mass index did not differ between groups (overall: 21.8 ± 2.4 y and 24.4 ± 5.6 kg/m2). Following the one week intervention, outcome measures did not vary between the VEG2 and VEG3 groups, and these groups were collapsed for the final analyses. The 24-h urine pH was raised after seven consistent days of vegan diet adherence and was unchanged after 2–3 days of vegan diet adherence over the course of a week (+0.52 ± 0.69 and ?0.02 ± 0.56 respectively, p = 0.048). However, dietary PRAL scores fell significantly in both dietary groups during the 7-day trial. Since low dietary PRAL scores have been related to improve metabolic parameters, adoption of a vegan diets for several days per week should be explored as a diet strategy to lower disease risk.  相似文献

In two experiments with young healthy volunteers the effects of a high in take of dietary fibre on blood lipids and intestinal transit time were investigated. In the first experiment the effects were studied of a high-fibre diet, in which half of the dietary fibre was provided by vegetables and fruits and the rest came from bread and other cereal products. In the second trial the effects of dietary fibre derived from vegetables and fruits were compared with those of isolated citrus pectin and wheat bran. It is concluded that—at least in short-term controlled experiments—fibre-rich food-stuffs have only a small (vegetables and fruits) or no (bran) favourable effect on the level of serum cholesterol. However, in uncontrolled circumstances, a ‘natural’ high fibre diet may through its low fat and cholesterol content indirectly reduce the concentration of serum cholesterol. A high-fibre diet with vegetables and fruits as well as a diet with bran shorten the intestinal transit time and enhance faeces production.  相似文献

In an experiment, involving twelve male cattle (initially 235 kg live weight), the effects of applying lactic acid bacteria [Lactobacillus plantarum; 109 colony-forming units (g fresh silage)?1] to grass silage, immediately prior to that silage being fed, on dry-matter (DM) intake of the silage, degradability of nitrogen (N) and fibre in the rumen, total tract digestibility and composition of rumen fluid in the animals were examined. A grass silage, which had been made from the primary growth of a predominantly perennial ryegrass sward, was offered as the sole diet. The inoculant was applied to the silage at the rate of 2 g of freeze-dried powder reconstituted in 12 ml of water (kg fresh silage)?1 immediately prior to that silage being fed and an equivalent amount of water was applied to the silage in the control treatment. The two diets were compared in a change-over design. The silage was well preserved, having a pH and concentrations of ammonia N and butyrate of 3.72, 74 g (kg total N)?1 and 0.11 g (kg DM)?1 respectively. Application of the inoculant significantly increased true protein, acid-insoluble N and water-soluble carbohydrate concentrations (P < 0.001) in the diet. Silage DM intake was not significantly increased (P= 0.072) by this of inoculant treatment, which had no significant effect (P > 0.05) on rumen degradability or total tract digestibility of DM, N, neutral detergent fibre or modified acid detergent fibre. Rumen pH, ammonia concentration or the molar proportions of volatile fatty acids were not altered (P>0.05) by inoculant treatment. It is concluded that application of the inoculant to the silage prior to silage being fed did not significantly affect silage DM intake, total tract digestibility, or degradability or fermentation in the rumen of cattle offered grass silage as the sole diet. It is also concluded that the results of this experiment provide no evidence that the mode of action of L plantarum, applied as an additive to grass at ensiling in previous studies, is through ‘direct’ effects in the rumen.  相似文献

The effects of diets including restructured meats (RM) containing Wakame or Nori on total liver glutathione status, and several antioxidant enzyme gene expressions and activities were tested. Six groups of ten male growing Wistar rats each were fed a mix of 85% AIN-93 M diet and 15% freeze-dried RM for 35 days. The control group (C) consumed control RM, the Wakame (W) and the Nori (N) groups, RM with 5% Wakame and 5% Nori, respectively. Animals on added cholesterol diets (CC, CW, and CN) consumed their corresponding basal diets added with cholesterol (2%) and cholic acid (0.4%). Alga and dietary cholesterol significantly interact (P < 0.002) influencing all enzyme expressions but not activities. The cholesterol supplement decreased most enzyme expression and activity. W-RM vs. C-RM increased (P < 0.05) expression of GPx, GR, Mn-SOD, and Cu,Zn-SOD and decreased that of catalase. N-RM vs. C-RM increased (P < 0.05) expression of catalase and Mn-SOD. GR activity increased in W-RM rats while SOD activity increased, but that of Se-GPx decreased in N animals. W-RM increased total and reduced glutathione and decreased the redox index. CN diet induced significantly lower plasma cholesterol levels (P < 0.001) than the CW diet. In conclusion, Nori-RM is a hypocholesterolemic food while Wakame-RM is an antioxidant food. This should be taken into account when including this kind of RM as potential functional foods in human.  相似文献

Two rapeseed protein concentrates (RPC) supplemented with zinc oxide were fed to male and female rats at two levels providing approximately half or all of the protein in a purified basal diet containing 20% protein (N×6.25). Control groups were given either a purified basal diet with casein or a laboratory chow diet. After 16 weeks, there were no significant (P<0.05) differences in body weight except the male rats fed the casein control diet had higher body weights than did other male rats. No significant differences were observed in organ weights including thyroids, hematology, bone marrow differential counts, serum glucose, urea nitrogen, total protein, albumin/globulin ratio, cholesterol, transaminases or alkaline phosphatase. The levels of serum vitamins A and E of females fed some of the RPC diets were less than in those fed the caseincontrol diet, but equal to those in females fed laboratory chow diet. The study was conducted on eleven healthy non-anaemic adolescent girls of 16 to 18 years of age. Balance studies were conducted in two trials of three weeks each on low and high fibre diets. High fibre diet contained 25 g Isabgol husk in addition to low fibre diet. The mean diet and nutrient intakes of the subjects were approximately the same during both trials. Addition of Isabgol husk to low fibre diet significantly (P0.05) increased faecal excretion of zinc, copper and manganese and lowered their apparent retention. The serum levels of these trace minerals decreased significantly (P0.05). Thus the high level of Isabgol has undesirable effect on trace minerals.  相似文献

The present study was performed to investigate the possible effects of wheat germ supplementation on lipid metabolism in humans. Ten free-living adult subjects participated in the study. None was obese or diabetic. They all presented an, hypercholesterolemia (from 6.58 to 9.50 mM), associated in 6 over 10 cases to an hypertriglyceridemia (from 1.70 to 5.00 mM). The subjects were studied in three consecutive periods, during which they first were on their usual diet (first week), they then ingested a daily supplement of 30 g wheat germ (4 weeks) and then they returned to their usual basal diet (4 weeks follow-up). Dietary records were obtained for 7 and 3 consecutive days before and during wheat germ supplementation, respectively. Fasting blood samples were taken at the end of each period. After 4 weeks of wheat, germ intake, glycemia did not change while total plasma cholesterol significantly decreased (paired Student'st test,p0.05) from 7.80 to 7.15 mM. LDL and HDL cholesterol values did not show marked changes, but VLDL cholesterol significantly dropped by 40.6%. Thus, the plasma/HDL total cholesterol ratio was significantly lower. Apoprotein B and A1 decreased. In the hypertriglyceridemic subjects, this was accompanied by a significant reduction of plasma triglycerides (1.64 vs. 2.68 mM) and a marked drop of VLDL triglycerides (–51%). Taken as a whole, the present results obtained in humans are very close to those previously obtained in the rat [40] and point out that wheat germ may play a beneficial role in the dietary management of hyperlipidemia.Part of the data communicated in this paper was presented at the Fibre 90 Conference, 17–20 April 1990, Norwich, UK.  相似文献

The link between dietary fats and cardiovascular diseases has necessitateda growing research interest in palm oil, the second largest consumedvegetable oil in the world. Palm oil, obtained from a tropical plant, Elaeis guineensis contains 50% saturated fatty acids, yet it does notpromote atherosclerosis and arterial thrombosis. The saturated fatty acidto unsaturated fatty acid ratio of palm oil is close to unity and it containsa high amount of the antioxidants, -carotene, and vitamin E. Although palm oil-based diets induce a higher blood cholesterol level thando corn, soybean, safflower seed, and sunflower oils, the consumption ofpalm oil causes the endogenous cholesterol level to drop. Thisphenomenon seems to arise from the presence of the tocotrienols and thepeculiar isomeric position of its fatty acids.The benefits of palm oil to health include reduction in risk of arterialthrombosis and atherosclerosis, inhibition of endogenous cholesterolbiosynthesis, platelet aggregation, and reduction in blood pressure. Palm oilhas been used in the fresh state and/or at various levels of oxidation.Oxidation is a result of processing the oil for various culinary purposes.However, a considerable amount of the commonly used palm oil is in theoxidized state, which poses potential dangers to the biochemical andphysiological functions of the body. Unlike fresh palm oil, oxidized palm oilinduces an adverse lipid profile, reproductive toxicity and toxicity of thekidney, lung, liver, and heart. This may be as a result of the generation oftoxicants brought on by oxidation. In contrast to oxidized palm oil, red orrefined palm oil at moderate levels in the diet of experimental animalspromotes efficient utilization of nutrients, favorable body weight gains,induction of hepatic drug metabolizing enzymes, adequatehemoglobinization of red cells and improvement of immune function. However, high palm oil levels in the diet induce toxicity to the liver asshown by loss of cellular radial architecture and cell size reductions whichare corroborated by alanine transaminase to aspartate transaminase ratioswhich are higher than unity.The consumption of moderate amounts of palm oil and reduction in thelevel of oxidation may reduce the health risk believed to be associated withthe consumption of palm oil. Red palm oil, by virtue of its-carotene content, may protect against vitamin A deficiency andcertain forms of cancer.  相似文献
