To determine and compare the prevalence of trypanosome infections in different livestock species (cattle, pigs and goats) in areas where game animals are scarce and livestock constitute the main food source of tsetse, a survey was conducted on the plateau of the Eastern Province of Zambia in Katete and Petauke districts where Glossina morsitans morsitans is the only tsetse species present. Blood was collected from a total of 734 cattle, 333 goats and 324 pigs originating from 59 villages in both districts and was examined using the buffy coat method and the PCR-RFLP as diagnostic tools. The prevalence of trypanosome infections differed substantially between livestock species. Using microscopic diagnostic methods, trypanosome infections were detected in 13.5% of the cattle and 0.9% of the pigs. All goats were parasitologically negative. The PCR-RFLP analyses increased the trypanosomiasis prevalence to 33.5, 6.5 and 3.3% in cattle, pigs and goats respectively. The majority of the infections (91.2%) were due to Trypanosoma congolense. The presence of a trypanosome infection in cattle and pigs resulted in a significant decline in the packed cell volume. The outcome of the study clearly shows that despite the availability of goats and pigs, cattle seem to be the major livestock species affected by the disease in trypanosomiasis endemic areas. The high proportion of infections in cattle could be partly attributed to their higher availability and attractiveness to tsetse.  相似文献

A longitudinal study was conducted within a cattle ranch in Gabon to determine the diminazene aceturate (Berenil) index (DAI) in a group of Zebu, raised under low tsetse density; this measure providing an assessment of trypanosomiasis risk. The objective was to evaluate the trypanosomiasis pressure thus informing trypanosomiasis control methods and cattle management. Twenty female adult Zebu were monitored for 24 weeks during the dry season. Blood samples were collected on a weekly basis and subjected to parasitological and haematological analysis (n?=?480), using the buffy-coat method and the packed cell volume value (PCV), respectively, infected animals were treated with a single intramuscular injection of diminazene aceturate (8 mg/kg). Twenty-nine single infectious events were recorded and a DAI of 1.45 was calculated. Two trypanosome species were identified: Trypanosoma congolense (96.2%) and Trypanosoma vivax (3.8%). The mean PCV value of the infected animals was lower (26.6) compared to non-infected animals (32.0). This study shows that DAI may be a useful tool to assess trypanosomiasis. However, this is a time-consuming method that may be improved by using randomly selected sentinel animals to adapt the chemoprophylactic schemes, hence decreasing the costs and the drug resistance risk.

A study to assess the influence of re-invasion of Glossina pallidipes on the epidemiology of bovine trypanosomosis was conducted in Southeast Uganda. A total of 1,992 cattle were screened in villages, with (949) and without G. pallidipes (1043) for trypanosomosis using a combination of the BCT and HCT methods. The prevalence of trypanosomosis (15.5%), Trypanosoma brucei infection (1.4%), T. congolense infection (7.2%), T. vivax infection (5.3%) and mixed infection (1.6%) in cattle in villages with was significantly higher than in those without G. pallidipes: trypanosomosis (7.1%), T. brucei infection (0.6%), T. congolense infection (2.0%), T. vivax infection (3.3%) and mixed infection (1.2%) (overall trypanosome infection, chi2=35.5, d.f.=1, P<0.05; T. brucei infection, chi2=8.06, d.f.=1, P<0.05; T. congolense infection, chi2=22.8, d.f.=1, P<0.05 and T. vivax infection, chi2=6.4, d.f.=1, P<0.05). Infections of Trypanosoma congolense were predominant in cattle in villages with G. pallidipes, while T. vivax infections were predominant in cattle in villages without. In all villages, T. brucei infections were fewer than either T. congolense or T. vivax infections. The risk of transmission of T. brucei, T. congolense and T. vivax infections was 3, 2.7 and 1.6 times, respectively, higher in villages with G. pallidipes than in those without, despite the presence of G. f. fuscipes in either set of villages. The mean PCV (28.27+/-0.41, 95% CI) and mean herd size (3+/-0.46) of cattle in villages with G. pallidipes were significantly (P<0.05) lower than in those in villages without (mean PCV, 29.48+/-0.34; mean herd size, 4+/-0.72). It is evident that presence of G. pallidipes brings about an increase in the prevalence of T. congolense, which causes a more severe disease in cattle than other species of trypanosomes. This is a rare case of a re-invasion of a tsetse species whose disease transmission capability calls for refocusing of the traditional national tsetse and trypanosomosis control strategies to contain it.  相似文献

In order to verify whether the low impact on livestock production in endemic areas is related to a low number of trypanosome strains circulating in livestock, 37 Trypanosoma congolense isolates collected from cattle in 11 sites in an endemic trypanosomiasis area in Eastern Zambia were characterised for genotype variability using a modified amplified fragment length polymorphism technique (AFLP). Isolates were further cloned to evaluate the occurrence of mixed infections in individuals. The results obtained revealed a high genotype diversity (94.6%) among these isolates. Apart from one site, all isolates gave different AFLP profiles in each of the sites. When clones were compared, three (8%) of the 37 isolates had mixed infections. These results indicate the circulation of a high number of strains in this trypanosomiasis endemic area despite the low impact the disease has on livestock production.  相似文献

The protective efficacy of isometamidium chloride (ISMM) and diminazene aceturate (DIM) against Trypanosoma brucei, Trypanosoma congolense and Trypanosoma vivax infections in cattle under a suppressed tsetse population was assessed in southeast Uganda. A total of 66 and 57 trypanosome-infected cattle were treated with ISMM and DIM, respectively together with 177 trypanosome-free animals not treated were followed for 12 months, checked every 4 weeks. There was no statistical difference in the mean time to infection with any trypanosome species in animals treated with ISMM or DIM. However, the mean time to trypanosome infection was significantly longer for treated animals than controls. The mean time to infection with each of the three trypanosome species differed significantly, with the average time to T. vivax infection the lowest, followed by T. congolense and then T. brucei. The protective efficacy of DIM was as good as that of ISMM; implying curative treatments against trypanosomosis are sufficient for combination with tsetse control. Isometamidium chloride or DIM had the highest impact on T. brucei and T. congolense infections in cattle.  相似文献

Summary A micromodification of the enzyme-linked immunosorbent assay (ELISA) was evaluated for its potential application in the immunodiagnosis of bovine trypanosomiasis. Serum samples from infected and non-infected Zebu cattle and samples from Friesian cattle with experimental infections were examined for the presence of trypanosomal antibodies. There were significant differences between the microELISA values obtained with samples from infected and non-infected cattle. During the course of infection microELISA values were found to fluctuate and the antibody response varied in individual animals. The test did not distinguish between infections withTrypanosoma brucei, T. vivax andT. congolense. There were no cross-reactions between trypanosome antigens and serum samples from cattle infected withT. theileri, Theileria parva, Th. mutans, Th. annulata, Babesia divergens andAnaplasma marginale.  相似文献

Laboratory experiments and field observations clearly show that tsetse flies can be carriers of mixed trypanosome infections. Question remains how easy it is for the tsetse fly to acquire such a mixed infection during the first bloodmeal. This is of particular importance in the epidemiology of Trypanosoma brucei s.l., often a cryptic infection and difficult to transmit to non-teneral tsetse flies. To determine the transmission rate of T. brucei as part of a mixed infection, teneral Glossina morsitans morsitans were fed once on cattle with a mixed (Trypanosoma brucei brucei/Trypanosoma congolense) or single (T. brucei) infection. Of the 140 flies fed on animals with a mixed infection and examined 30 days later, 4 had a metacylic T. brucei infection, 29 a T. congolense infection and 13 a mixed T. brucei/T. congolense infection. There was no significant difference between the transmission rate of T. brucei as a single or as part of a mixed infection. The high proportion of mixed T.b. brucei/T. congolense infections was explained best by a model implying that if a fly is refractory to T. congolense, it is also refractory to T.b. brucei and vice versa. Hence, results suggest that the transmission of T.b. brucei is affected mainly by the vectorial capacity of flies and not by concurrent trypanosome infections in the host.  相似文献

A technique for the rapid field assessment of African animal trypanosomosis (AAT) was developed during studies in the Gambia. This involved gathering indigenous information from rapid-appraisal questionnaires addressed to local informants, the results of single tsetse surveys and evaluations of the prevalence of trypanosome infections in village cattle. Local informants included livestock owners and herdsmen and trained personnel such as livestock assistants. The answers to the questionnaires were weighted in order to translate them into semi-quantitative ranked estimates (zero, low, medium, high or very severe) of the severity of AAT problems. A similar ranking was also defined for tsetse and prevalence data in the Gambia. The three assessment methods generally gave complementary results leading to similar conclusions about the severity of tsetse-trypanosomosis problems in a survey area; inconsistencies usually suggested that additional information was needed. The rankings of AAT intensity were used to develop management guidelines for minimising the impact of AAT at different levels through control interventions or improved livestock management. The methodology was designed to provide reliable, up-to-date and cost-effective assessments of AAT problems. Emphasis was placed on the importance of the involvement, priorities and perceptions of village livestock owners and herdsmen in making these assessments.  相似文献

The prevalence of trypanosomosis, mean packed cell volume and anti-trypanosomal antibody levels in village cattle of different age groups (<0.5 year, 0.5–2 years, >2–5 years and >5 years) in the areas with tsetse control were compared with those of corresponding age groups in areas without tsetse control in Tororo, southeast Uganda. The prevalence of trypanosomosis in cattle in the age groups of 0.5–2 years, >2–5 years and >5 years in the areas with tsetse control was significantly lower than in cattle in similar age groups in the areas without tsetse control (p<0.5). Trypanosoma vivax was the most predominant Trypanosoma species in the areas with tsetse control, while T. congolense was the most predominant species in the areas without tsetse control. The mean Trypanosoma antibody levels in cattle in the age groups <0.5 year, 0.5–2 years and >2–5 years in the areas with tsetse control were significantly lower than those of the similar age groups in the areas without tsetse control (p<0.5). The mean PCV values for cattle in the age groups 0.5–2 years, >2–5 years and >5 years from the areas with tsetse control were significantly higher than those of the similar age groups in the areas without tsetse control. Tsetse control appeared to have a considerable impact on the prevalence of trypanosomosis, distribution of Trypanosoma species, specific antibody levels and the packed cell volume of cattle in the different age groups.  相似文献

Trypanosome infection rate in cattle at Nguruman was investigated in a study conducted in 1984-1986. Shifting pastoralism significantly reduced trypanosome infections in cattle. The cattle were more heavily infected with Trypanosoma congolense (16.5%) than Trypanosoma vivax (4.95%) and Trypanosoma brucei (0.19%). Trypanosoma theileri was observed only once among the cattle examined. Mixed trypanosome infections in cattle were observed to be 2.75% and 0.014% for T. congolense/T. vivax and T. congolense/T. brucei, respectively. The duration of infection in the cattle was 55 days for T. congolense and 79 days for T. vivax. High infections in cattle were observed 2 months after the rains, which were concomitant with high tsetse densities.  相似文献

Teneral tsetse flies infected with either Trypanosoma brucei or T. vivax were fed on healthy cattle. Blood samples collected daily from the cattle were examined by microscopy for the presence of trypanosomes, in thick smear, thin smear and in the buffy coat (BC). All the cattle fed upon by infected tsetse developed a fluctuating parasitaemia. DNA was extracted from the blood of these cattle and subjected to polymerase chain reaction (PCR) using oligonucleotide primers specific for T. brucei or T. vivax. The PCR products unique to either T. brucei or T. vivax were identified following amplification of DNA from the blood samples of infected cattle, whereas none was detectable in the DNA from the blood of the cattle exposed to non-infected teneral tsetse. In a concurrent set of experiments, one of the oligonucleotide primers in each pair was biotinylated for use in PCR-ELISA to examine all the blood samples with this assay. Both the PCR and the PCR-ELISA revealed trypanosome DNA in 85% of blood samples serially collected from the cattle experimentally infected with T. brucei. In contrast, the parasitological assays showed trypanosomes in only 21% of the samples. In the blood samples from cattle experimentally infected with T. vivax, PCR and PCR-ELISA revealed trypanosome DNA in 93 and 94%, respectively. Microscopy revealed parasites in only 63% of the BCs prepared from these cattle. Neither PCR nor PCR-ELISA detected any trypanosome DNA in blood samples collected from the animals in the trypanosome-free areas. However, both assays revealed the presence of trypanosome DNA in a number of blood samples from cattle in trypanosomosis-endemic areas.  相似文献

A cross-sectional study was carried out from November 2008 to February 2009 in Guraferda and Sheko districts of Bench Maji Zone, South Western Ethiopia. The objective of the study was to determine the prevalence of bovine trypanosomosis and the density of its vectors. An overall prevalence of trypanosome infection in the study area was 4.4%. Trypanosoma congolense (36.36%) was the dominant trypanosome species followed by Trypanosoma vivax (18.18%) and Trypanosoma brucei (9.09%). Mean packed cell volume value of parasitemic animals (21.8%) was significantly (P < 0.05) lower than that of aparasitemic animals (27.7%). Biconical and NGU traps were deployed for 72 h, and the result indicated Glossina pallidipes followed by Glossina fuscipes as the only tsetse fly species caught in the study area along with other biting flies like Stomoxys and Tabanus. The apparent density of tsetse flies was 2.83 flies trap−1 day−1. NGU trap caught more of G. pallidipes while biconical trap caught more G. fuscipes, and the difference was significant (P < 0.05). Although the current study indicated low prevalence of trypanosomosis in the study area, the impacts of trypanosomosis on cattle production and productivity should not be neglected. Therefore, attention should be given to control the disease and also the vector.  相似文献

The effect of challenging cattle, chronically infected with Trypanosoma brucei brucei, with T. congolense on the development of the T. b. brucei infection was investigated. For this purpose, nine experimental animals were first infected with T. b. brucei through the bites of infected tsetse flies. Once the T. b. brucei had developed into a chronic infection, that was difficult to detect using routine parasitological diagnostic tools, seven of the experimental animals were challenged by tsetse flies infected with T. congolense. Two of the animals infected with T. b. brucei were kept as control. The infection with T. congolense resulted in a sudden increase in the parasitaemia of T. b. brucei. In the T. b. brucei control animals, on the other hand, the parasitaemia remained below the level of detection. The epidemiological repercussions of this increase in the parasitaemia of T. b. brucei after infection with T. congolense are discussed.  相似文献

Four populations of Trypanosoma congolense and Trypanosoma brucei brucei were isolated from cattle under different management practices and environments in Zambia. All four isolates had varied responses to both diminazene aceturate (Berenil) and isometamidium chloride (Samorin) as curative drugs in infected mice. Trypanosomes from a traditionally managed herd in a high-tsetse-challenge area had the strains most resistant to Berenil, with maximum curative dose of 45 mg kg-1 body weight. Another isolate from a high-tsetse-challenge area was evidently resistant both to Berenil at 40 mg kg-1 and to Samorin at 4 mg kg-1. The strains most susceptible to both Berenil and Samorin were from a commercially managed herd of cattle under medium tsetse challenge. They responded to recommended cattle standard doses of 3.5 mg kg-1 or 7 mg kg-1 Berenil and to as little as 0.25 mg kg-1 Samorin. It is evident that trypanosome strains resistant to Berenil and/or partially resistant to Samorin exist, and that both T. congolense and T. b. brucei are implicated.  相似文献

Summary Investigations of acute trypanosomiasis in a group of 1,200 cattle provided strong evidence for mechanical transmission by haematophagous flies other than tsetse. Although recently introduced cattle may have had previous contact with tsetse flies the incident occurred in an area remote from their known range. Mortality exceeded 6% before the outbreak was controlled by chemotherapy. Mechanically transmittedTrypanosoma vivax infection may be of widespread occurrence in eastern Ethiopia.

El incidente ocurrió en un área remota del habitat de la tsetse, aunque un grupo de ganado recientemente introducido, pudo tener algun contacto previo con éstas últimas. La mortalidad excedió el seis por ciento, antes que el brote se controlara. La transmisión puramente mecánica de la tripanosomiasis en Etiopía, podría tener más importancia que la que actualmente se le da.

La mortalité a dépassé 6 p. 100 avant que le foyer n'ait pu être contrôlé par la chimiothérapie. La transmission mécanique de l'infection àT. vivax est probablement largement répandue dans l'Est éthiopien.

Ten Boran (Bos indicus) cattle were used to study the stress responsiveness of the hypothalamic-pituitary-adrenal (HPA) axis during trypanosome infection. Five cattle were infected with Trypanosoma congolense IL 1180 by tsetse challenge and five cattle served as controls. All infected animals developed acute trypanosomiasis. Insulin-induced hypoglycaemia (50 per cent of pre-insulin glucose concentration) was used as a stress factor. Acute hypoglycaemia was observed in three infected and three control animals after insulin challenge. Two animals from each group either did not respond or responded slowly. Hypoglycaemia in infected animals completely failed to induce an HPA axis response, while in control animals an HPA axis response was indicated by a significant increase in plasma adrenocorticotrophic hormone (ACTH) and cortisol concentrations (P less than 0.01). The results show that trypanosomiasis in Boran cattle can cause a decrease in the stress responsiveness of the HPA axis as indicated by a blunted ACTH/cortisol response to insulin-induced hypoglycaemia.  相似文献

This review describes research conducted from 1900–1983 on trypanosomiasis due to Trypanosoma evansi in Indonesia. Clinical signs and post‐mortem findings in horses, cattle, buffaloes, pigs and dogs, experimental transmission tests to establish possible surra vectors in Indonesia, and research on chemotherapy and chemoprophylaxis are discussed.  相似文献

Foot and mouth disease (FMD) is the most economically important disease of livestock that still affects extensive areas of the world. This study described the use of participatory appraisal tools such as pair-wise ranking, matrix scoring and proportional piling to assess the perception of livestock keepers about the clinical signs and epidemiological features of cattle diseases with particular emphasis on FMD. Strong agreement among informant groups (W?=?0.710; P?=?0.000) in pair wise ranking indicated that the diseases were common problem to all the selected districts. Matrix scoring of disease indicators/signs also showed strong agreement (W?=?0.504 to 0.955; P?=?0.000) that implied significant current veterinary knowledge by Afar pastoralists. Age specific means of annual incidence rates estimated by proportional piling were significantly (P?<?0.05) different but negatively correlated for FMD, CBPP and pasteurellosis. However, correlation was not significant for anthrax, blackleg and other important diseases. Age specific means of annual mortality was significantly (P?<?0.05) but negatively correlated for FMD, CBPP, pasteurellosis and other important cattle diseases with correlation coefficients of ?0.72, ?0.81, ?0.8 and ?0.55, respectively. However, correlation (r?=?0.12) was not significant for anthrax and not correlated at all for blackleg. This study indicated that pastoralists have detailed knowledge about their livestock health problems, and hence the combined use of participatory appraisal and conventional methods is essential for an ultimate disease control strategy.  相似文献

In a survey for bovine trypanosomiasis blood smears from 1,617 cattle at 72 sites were examined. Trypanosomes were found in 93 cattle, representing 16% of the cattle in herds in which trypanosomiasis was confirmed. Of the positive cattle 56% had infections with T. congolense, 17% T. vivax and 2.2% T. brucei. Five cattle had mixed infections and in 18 cattle the species was not identified. Sera from 1,352 cattle were tested using microelisa. Ten out of 16 sites, at which no trypanosomes were found in blood smears and at which trypanocides were in use, had over 15% seropositive cattle compared with five of 19 sites at which trypanocides were not in use. It was concluded that the microelisa was a useful aid to the diagnosis of bovine trypanosomiasis and that there is a need for accurate records of drug use and livestock movements to be kept. The serious risk of drug resistant strains of trypanosomes emerging due to the uncontrolled use of trypanocides is emphasised.  相似文献
