用 8头荷斯坦黑白花奶牛 ,在饲粮中添加过瘤胃蛋白和丙二醇 ,研究其对乳牛瘤胃发酵、产乳量及乳成分的影响。结果表明 ,过瘤胃蛋白+丙二醇可显著提高丙酸浓度 (P <0 0 5) ,并且产乳量较对照组有增加的趋势。乳脂率、乳蛋白率、乳糖率及无脂固形物率 ,并未因添加过瘤胃蛋白和丙二醇而受到显著影响 (P >0 0 5)。乳蛋白质量、乳脂量及无脂固形物量 ,过瘤胃蛋白 +丙二醇组较对照组有增加的趋势 ,但差异均不显著 (P >0 0 5)。  相似文献

A three-phase laboratory procedure suitable for predicting protein degradability in the rumen and digestibility of undegraded protein is reported. In the first phase the feed was incubated with starch and buffered rumen fluid. In the incubation mixture the viability of protease-active bacteria was checked by anaerobic culturing, whereas changes in protease activity were monitored by azocasein degradation. In the second and third phase rumen undegradable protein (UDP) was digested with pepsin and pancreatin, respectively. The measurements showed that 63.2, 5.2 and 4.7% of the crude protein of green lucerne was decomposed by rumen fluid, pepsin and pancreatin, respectively. Degradability of the crude protein of extracted sunflower meal was 68.3, 17.7 and 5.5% in the three phases, respectively. Repeated determination yielded crude protein degradabilities of 66.7, 27.1 and 5.1% for the three phases, respectively.  相似文献

本文旨在研究不同剂量的瘤胃细菌培养物对荷斯坦奶牛瘤胃发酵及菌体蛋白(BCP)含量的影响。试验采用单因素试验设计,选用4头装有永久性瘤胃瘘管,体况相近的荷斯坦奶牛,取其瘤胃液混匀,添加不同剂量的瘤胃细菌培养物进行体外培养,分别为:对照组(0 CUF/g),试验Ⅰ组(添加量为3%,4.4×108CUF/g),试验Ⅱ组(添加量为5%,7.8×108CUF/g),试验Ⅲ组(添加量为10%,1.2×109CUF/g),每组分别设2、4、8、12、24、36、48 h 7个指标测定时间点,每个时间点3个重复。结果表明:(1)添加瘤胃细菌培养物使瘤胃液pH有所降低,但始终在6.5~7.0,对瘤胃内环境无不良影响;(2)添加3%的瘤胃细菌培养物显著降低氨态氮(NH3-N)的含量(P<0.05),显著增加了培养液BCP的浓度(P<0.05);(3)添加3%的瘤胃细菌培养物可一定程度增加总挥发性脂肪酸(TVFA)和乙酸浓度,但对其他各挥发性脂肪酸及乙酸/丙酸比值无显著影响。由此可见,添加3%剂量的瘤胃细菌培养物可改变瘤胃的发酵环境,提高BCP浓度。  相似文献

以4只安装永久性瘤胃瘘管的白山羊作为采集瘤胃液的供体,用体外法研究了4种不同蛋白质来源(玉米-豆粕型、玉米-豆粕-棉粕型、玉米-豆粕-菜粕型、玉米-棉粕-菜粕型)的日粮对不同培养时间的发酵底物的干物质和蛋白质消化率、产气量、瘤胃pH值和NH3-N质量浓度的影响。结果表明:经24 h人工瘤胃培养后,干物质和蛋白质的消化率由高到低顺序依次为:玉米-豆粕型>玉米-豆粕-棉粕型>玉米-豆粕-菜粕型>玉米-棉粕-菜粕型。发酵底物产气量由高到低依次为:玉米-豆粕-菜粕型>玉米-豆粕-棉粕型>玉米-豆粕型>玉米-棉粕-菜粕型。瘤胃NH3-N质量浓度由高到低依次为:玉米-豆粕-棉粕型>玉米-豆粕-菜粕型>玉米-豆粕型>玉米-棉粕-菜粕型。 各组的瘤胃pH值基本一致,处于正常瘤胃生理范围。  相似文献

试验采用3×3拉丁方设计,3头安装永久性大口径瘤胃瘘管的青年母牛饲喂3种不同精饲料的日粮,3种精饲料分别为豆粕+玉米(日粮1),猪血粉+玉米(日粮2)和豆粕+小麦(日粮3)。精料日喂量按活牛体重的1%喂给,干玉米秸等量组成的日粮配给。试验为3个试验周期,每一周期为37d,其中适应期26d,在早晨采食后2、4、8、16和24h,分别5次实施瘤胃掏空,全量测定瘤胃物存留量。结果表明:日粮1在采食后2~8h瘤胃干物质、液体和NDF的存留量最低,与其他两种日粮相比差异显著(P<0.05);日粮1和日粮3瘤胃液氨氮存留量在采食后2~8h始终高于日粮2(P<0.05),但日粮3在采食4h后瘤胃液氨氮开始下降,而日粮1至采食后16h开始下降,日粮1瘤胃液氨氮数量采食后2~8h处于高峰状态,说明瘤胃微生物在瘤胃环境长时间一直保持稳定的降解能力;日粮1和日粮3在采食2~4h后处于较高的总挥发性脂肪酸(VFA)瘤胃存留量,日粮1在采食后8~16h瘤胃存留量降低,与日粮2和日粮3相比差异显著(P<0.05),日粮1对瘤胃VFA的快速移除,意味着瘤胃液的快速流出以及可能解释瘤胃固体物的快速移除。因此,日粮1和日粮3在牛瘤胃洗涤纤维降解和瘤胃液产物代谢模式发挥不同作用。  相似文献

选用3头安装永久性瘤胃瘘管的健康荷斯坦产奶牛为试验动物,选取8种北方地区奶牛常用蛋白质饲料原料为试验样品,每个样品设3个重复,用尼龙袋法分别测定各个样品的粗蛋白质(CP)、中性洗涤纤维(NDF)和酸性洗涤纤维(ADF)在瘤胃内的动态降解率和有效降解率。结果表明:(1)所选8种蛋白质饲料原料中葵花粕的粗蛋白质(CP)有效降解率均高于其他粗饲料,达到61.17%以上,酒精粕的粗蛋白质(CP)有效降解率最低,仅为19.03%。(2)在饲喂基础上,玉米蛋白粉所含的瘤胃降解蛋白质(RDP)的比例最高,达到30.99%以上,而酒精粕所含的比例最低,仅为5.52%。(3)豆粕所含的中性洗涤纤维(NDF)的有效降解率最高,达到50.74%以上,而菜籽粕的最低,仅为11.06%。(4)玉米蛋白粉所含的酸性洗涤纤维(ADF)的有效降解率最高,达到59.23%以上,而酒精粕的最低,仅为11.27%。  相似文献

An experiment was conducted to test the hypothesis that the supplementation of crude protein (CP) results in rumen acidosis and increased histamine concentrations in dairy cows. Six ruminally fistulated, non-pregnant dry cows were fed three experimental rations in a replicated 3 × 3 Latin square design. The CP contents in the low-CP, the high rumen undegradable protein (high-RUP) and the high rumen degradable protein (high-RDP) rations were 112, 259 and 266 g/kg dry matter (DM) respectively. The cows were fed 7.7 kg DM of the concentrates and 2.7 kg DM of rice straw. High levels of RDP in the ration significantly increased the ammonia, total volatile fatty acid (VFA) and histamine concentrations in the rumen fluid. However, supplemental CP, whether degradable or undegradable, did not significantly affect the pH of rumen fluid. Plasma urea nitrogen concentration was higher in both high-RDP and high-RUP in relation to cows fed the low-CP ration. The rise in ruminal histamine concentrations was physiologically non-relevant, most likely because rumen pH was not affected by supplemental CP at the installed level of DM intake. Therefore, it can be concluded that the issue of supplemental CP, rumen pH and ruminal histamine concentrations has not yet been settled. Further research is warranted to understand these relationships.  相似文献

The effect of a daily dose of 30 mg Monensin on rumen fermentation pattern and degradation of the crude protein fraction of soybean meal was investigated with sheep. Degradation parameters for soybean meal were measured by the nylon bag technique. Monensin increased propionic acid percentages in the rumen, with a concomitant decrease in acetic- and butyric acid proportions, thus confirming earlier work. Ammonia N concentration in the rumen was also lowered, indicating an inhibitory effect on rumen protein breakdown. However, no effect on degradation rate, degradability and effective degradability (only with one sheep) could be observed. Possible reasons for these findings are discussed. Amino acid analysis on the residues in incubated bags revealed slight changes in amino acid composition, but these changes were considered to be nutritionally unimportant.  相似文献

The objective of this study was to investigate rumen available soluble, insoluble and total protein (CP), estimated structural (SC) and non‐structural carbohydrate (starch: ST), degradation characteristic ratios and hourly effective degradation of six barley varieties during three consecutive growth years (2003, 2004, 2005). The magnitude of the differences was determined between the varieties and growth years. Measured degradation kinetics included soluble fraction (S), undegradable fraction (U), lag time (T0) and rate of degradation (Kd) of the insoluble but degradable fraction (D). Rumen available soluble, insoluble, and total N, SC and ST and the rumen degradation characteristic ratios were determined using the Tamminga rumen degradation ratio system. Characteristics of the hourly effective degradation between N and carbohydrate (CHO) among the six barley varieties for 3 years were also studied. The degradation ratios included were total rumen available N and carbohydrate ratio (FN/FCHO), rumen available soluble N and carbohydrate ratio (SN/SCHO), and rumen available insoluble N and carbohydrate ratio (EN/ECHO). Results show that both the barley variety and growth year had a significant effect on degradation kinetics (S, D, U, T0 and/or Kd). Differences in the ratio of FN/FCHO among varieties ranged from 16.6 to 19.0 g/kg (p < 0.01). There was no difference in SN/SCHO (p > 0.05) with an average of 4.9 g/kg. The difference in the EN/ECHO ratio tended to be significant among the varieties (p = 0.069) ranging from 18.4 to 21.3 g/kg. Differences in the hourly effective degradation between N and CHO were relatively small at shorter incubation times (2–4 h) However, as the length of rumen incubation increased (12–24 h), much larger differences in the rate of effective hourly degradation were observed. In conclusion, both barley variety and growth year had significant effects on rumen degradation kinetics. The mean FN/FCHO ratio of 17.2 (16.6–19.0) was observed for the six barley varieties in this experiment. All barley varieties exhibited a less optimal rumen fermentation ratio (17.2 < optimum: FN/FCHO = 25 to 33 g N per kg CHO). The large differences in the degradation kinetics, characteristics ratios and hourly effective degradation among barley varieties and growth years may help to explain some of the large variations that are seen in cattle performance.  相似文献

Recently a new super‐genotype of oat has been developed in the Crop Development Center called CDC SO‐I (‘SuperOat’: low lignin and high fat). In a previous study, we evaluated total metabolizable protein using a TDN‐based model‐NRC‐2001 which is popular in North America. However, the TDN‐based NRC model is not accepted universally. The objectives of this study were to use a complex non‐TDN based model, the DVE/OEB system, to evaluate potential nutrient supply to ruminants from the SuperOat in comparison with two normal varieties of oats (CDC Dancer and Derby) in western Canada. The quantitative predictions were made in terms of: (i) truly absorbed rumen synthesized microbial proteins in the small intestine; (ii) truly absorbed rumen undegraded feed protein in the small intestine; (iii) endogenous protein in the digestive tract; (iv) total truly absorbed protein in the small intestine; and (v) protein degraded balance. Results showed that using the DVE/OEB system to predict the potential nutrient supply, it was found that the SuperOat had similar truly absorbed rumen synthesized microbial protein levels (61, 63, 59 g/kg DM, P > 0.05, for SuperOat, CDC Dancer and Derby, respectively), higher truly absorbed rumen undegraded feed protein than CDC Dancer (22 vs. 17 g/kg DM P < 0.05, for SuperOat, CDC Dancer, respectively), but similar to Derby (22 vs. 21 g/kg DM; P > 0.05), and similar endogenous protein (16, 16, 18 g/kg DM; P > 0.05). Total truly absorbed protein in the small intestine is only numerically higher in the SuperOat (67 vs. 65, 62 g/kg DM, P > 0.05, for CDC Dancer and Derby, respectively). However, the protein degraded balance was significantly different (P < 0.05) with a positive value for the SuperOat (7.0 g/kg DM) and negative values for two normal varieties (?1.5, ?6.8 g/kg DM for CDC Dancer and Derby, respectively). In conclusion, the model predicted significantly different protein degradation balance. The SuperOat had positive degradation balance but other two normal varieties had negative protein degraded balance However, the SuperOat had similar total absorbed protein value to the two normal varieties of oats.  相似文献

This study was carried out in order to determine the influence of nitrogen excess in rumen and deficit of digestible organic matter in a feed ration for dairy cows on hypomagnesaemia at the beginning of green forage feeding. The balance of nitrogen and energy metabolism in rumen was determined using the rules of evaluation of the nutritional value of fodder (PDI system) according to the Institut National de la Recherche Agronomique (INRA), taking into consideration a potential protein synthesis from nitrogen by microorganisms in rumen (MPRN) and microbial protein synthesized from digestible organic matter (MP(RE)). It was found that an increase in the amount of rumen degradable protein (RDP) and MPRN-MPRE balance leads to a significant increase of urea nitrogen in serum (p < 0.05). An increase of total MPRN-MPRE balance in rumen results in a decrease of Mg concentration in serum. It has been demonstrated that in cows fed green forage at the end of spring (2nd half of May to 1st half of June) when the MPRN-MPRE balance is not higher than 187 g, adding 7 g or 25 g of MgO/cow/day to the same extent successfully prevents hypomagnesaemia.  相似文献

The aim of the present study was to examine the effects of feeding Fusarium toxin-contaminated wheat to dairy cows on nutrient utilization in the rumen and on duodenal flow of deoxynivalenol (DON), zearalenone (ZON) and their metabolites. Six dairy cows fitted with a large rumen cannula and a simple T-shaped cannula at the proximal duodenum was used in two experiments. The experiments included a control period in which the uncontaminated control wheat was fed and a period in which the control wheat was replaced by the Fusarium toxin-contaminated wheat (8.05 and 7.15 mg DON/kg and 0.26 and 0.1 mg ZON/kg in Expts 1 and 2 respectively). The wheat portion of the daily ration amounted to 50% on a dry matter (DM) basis and rations were completed with hay or grass silage. Five of the six cows were non-lactating and the total daily DM-intake ranged between 4 and 12 kg. The pH-values and the concentration of volatile fatty acids in ruminal fluid were not significantly influenced by feeding the contaminated wheat. In contrast, the postprandial ammonia concentration was consistently higher when the mycotoxin-contaminated wheat was fed. Moreover, the flow of microbial protein and utilizable protein at the duodenum were reduced at the same time. The concentrations of DON and ZON and of their metabolites in freeze-dried duodenal digesta were either not detectable or negligible during the control periods whereas distinct concentrations were measured during the periods where the contaminated wheat was fed. DON was nearly completely metabolized to de-epoxy-DON and the flow at the duodenum ranged between 4% and 28% of DON-intake. The ZON metabolites alpha-zearalenol (ZOL) and beta-ZOL were recovered at the duodenum beside the parent toxin ZON. Their recovery as a percentage of ZON-intake ranged between 43% and 132%. In conclusion, feeding of Fusarium toxin-contaminated wheat altered the ruminal protein utilization. The question of whether this effect was a result of the mycotoxin being present in the rumen or of Fusarium growth-related structural (cell wall) changes of the wheat grain needs to be clarified. The low recovery of DON at the duodenum would indicate either a nearly complete degradation of the molecule in the rumen or an absorption by the mucosa of the rumen, whereas the higher ZON recovery would suggest a lower degradation of the parent toxin in the rumen and/or recovery of some bile-originating entero-hepatic cycling ZON/metabolites.  相似文献

An experiment was performed to evaluate the effect of extrusion and carbohydrate addition on rumen degradation and intestinal digestion of raw legume seeds and solvent extracted soya bean meal (SBM) protein. Whole soya beans (WSB) without or with maize added (75:25) (WSB-M), peas, lupins and SBM were extruded at 140 degrees C. Protein rumen degradation and intestinal digestibility of unprocessed and extruded protein sources were measured by in sacco and mobile bag procedures, respectively, in two dairy cows cannulated in rumen and duodenum. Between 12 and 15 polyester bags with 4 g of each protein source were incubated in rumen for 12 h and the residues, pooled by feed, were introduced into the duodenum in small nylon bags after pre-incubation in a pepsin solution, and recovered from faeces the day after. Extrusion significantly (p < 0.001) reduced N degradation of all protein sources, from 98.1%, 91.6%, 90.5% and 64.8% to 53.1%, 73.8%, 70.3% and 44.2% for peas, lupins, WSB and SBM respectively. The addition of maize to WSB strengthened the effect of extrusion on rumen N degradation, from 88.2% to 52.6%. Residues from rumen incubation of extruded feeds showed a higher (p < 0.001) intestinal N digestibility except for SBM (87.0%, 82.9%, 66.3%, 85.0% and 97.2%, and 99.1%, 95.8%, 96.8%, 97.8% and 98.7%, respectively, for non-extruded and extruded, peas, lupins, WSB, WSB-M and SBM). In conclusion, the extrusion of studied legume seeds and SBM promotes a clear and significant increase of their metabolizable protein value, particularly in peas, and the inclusion of a source of carbohydrates before extrusion increase this response.  相似文献

Vitamin E (Vit. E) is discussed to influence ruminal biohydrogenation. The objective of this study was to investigate the influence of a Vit. E supplementation on rumen fermentation characteristics, ruminal microbial protein synthesis as well as ruminal organic matter fermentation. Furthermore, we aimed to investigate the influence of Vit. E supplementation on short‐chain fatty acids (SCFA) and protozoa concentrations in the rumen and, in addition, on transfer rates of middle‐chain and long‐chain fatty acids into the duodenum in lactating dairy cows. Eight rumen and duodenum fistulated German Holstein cows were assigned to either a group receiving 2,327 IU/d Vit. E (138.6 IU/kg DM DL‐α‐tocopherylacetate; = 4) or a control group (23.1 IU/kg DM;= 4). Neither ruminal protein synthesis nor organic matter fermentation was influenced by treatment. Vit. E did not act on the concentrations of short‐chain fatty acids and protozoa in rumen fluid. Duodenal flow of C13:0 (1.3 versus 0.2 g/d, = 0.014) and iso‐C14:0 (1.0 versus 0.5 g/d, = 0.050) was higher in the Vit. E group. We observed a trend for higher duodenal flows for C12:0 (1.6 versus 0.9 g/d, = 0.095) and anteiso‐C15:0 (12.2 versus 8.9 g/d, = 0.084). Transfer rate of C12:0 tended to be higher in the Vit. E group (125.61 versus 73.96, = 0.082). No other transfer rates were affected by treatment. Further studies are necessary to investigate the influence of Vit. E on rumen microbiota and their fatty acid production as well as on the impact of different doses of Vit. E supplementation on variables of protein synthesis efficiency.  相似文献

The effect of virginiamycin in incubations of rumen fluid with carbohydrate or protein substrate was investigated. In carbohydrate incubations, methane production was partially inhibited while propionate proportions increased. Total microbial growth was slightly decreased, but net microbial growth was considerably lower. Protein degradation was slightly lowered after addition of virginiamycin. These effects were compared with results obtained with other rumen manipulating additives.  相似文献

Ruminant animals are able to convert plant materials (grain and the human‐indigestible portion of carbohydrates) to milk and meat. In this conversion, most of the plant materials are digested by rumen fermentation and are changed to short‐chain fatty acids, microbial cells, and methane, which is released into the atmosphere. The relationships among feed, rumen fermentation, and milk production are poorly understood. Here we report a novel indicator of characteristics of rumen fermentation, theoretical turnover rate (TTOR) of the rumen liquid fraction. The TTOR was calculated from the presumed rumen volume (PRV) which is estimated by dividing the methane yield by the methane concentration of rumen fluid. The formula for the TTOR is: TTOR = PRV/body weight0.75. Our present analyses confirm that the TTOR as an indicator is capable of connecting feed, rumen fermentation, and milk production, because dry matter intake/TTOR showed a strong correlation with milk yield/TTOR. In addition, the TTOR may be related to ruminal pH, as we observed that the ruminal pH decreased as the TTOR increased. We propose that the TTOR is a factor characterizing rumen fermentation and a good indicator of the productivity of ruminants and dysbiosis of the rumen microbiome.  相似文献

Background: Original rumen digesta, rumen liquid and solid fractions have been frequently used to assess the rumen bacterial community. However, bacterial profiles in rumen original digesta, liquid and solid fractions vary from each other and need to be better established.Methods: To compare bacterial profiles in each fraction, samples of rumen digesta from six cows fed either a high fiber diet(HFD) or a high energy diet(HED) were collected via rumen fistulas. Rumen digesta was then squeezed through four layers of cheesecloth to separate liquid and solid fractions. The bacterial profiles of rumen original digesta, liquid and solid fractions were analyzed with High-throughput sequencing technique.Results: Rumen bacterial diversity was mainly affected by diet and individual cow(P 0.05) rather than rumen fraction. Bias distributed bacteria were observed in solid and liquid fractions of rumen content using Venn diagram and LEf Se analysis. Fifteen out of 16 detected biomarkers(using LEf Se analysis) were found in liquid fraction, and these 15 biomarkers contributed the most to the bacterial differences among rumen content fractions.Conclusions: Similar results were found when using samples of original rumen digesta, rumen liquid or solid fractions to assess diversity of rumen bacteria; however, more attention should be draw onto bias distributed bacteria in different ruminal fractions, especially when liquid fraction has been used as a representative sample for rumen bacterial study.  相似文献

试验研究了不同产地(吉林、九三、林口)豆粕(SBM)和不同产地(集贤、牡丹江、肇东)玉米酒精糟(DDGS)营养成分、CP的瘤胃降解规律及RUP的小肠消化率。试验选用3头装有永久性瘤胃瘘管的干奶期中国荷斯坦奶牛,利用尼龙袋原位培养法研究不同产地豆粕和DDGS在奶牛瘤胃中CP的降解规律。每个尼龙袋每种饲料装5 g,在瘤胃中培养0、2、4、8、16、24和48 h。将每个尼龙袋每种饲料装75 g瘤胃培养16 h,瘤胃未降解残渣强饲给公鸡,从而测定豆粕和DDGS的IDP。吉林豆粕中的CP和SP含量最高,林口豆粕最低,而九三豆粕居中。林口豆粕中NDIP和ADIP含量为29.10%和2.52%,在三种豆粕中处于最高。三种DDGS的CP、NDIP和ADIP含量的变化趋势一致,均为肇东地区最高,其次为集贤、牡丹江。除了慢速降解部分(b)的降解速率(c)外,豆粕和DDGS的其它降解参数均差异不显著。九三豆粕IDP为73.37%,显著高于其它两种豆粕(P<0.05)。集贤DDGS的IDP显著高于牡丹江DDGS(P<0.05),而肇东DDGS居中。由此得出结论,除了营养指标受到产地影响外,豆粕和DDGS的瘤胃降解率和小肠消化率均不受产地影响。  相似文献

Currently, knowledge regarding the ecology and function of bacteria attached to the epithelial tissue of the rumen wall is limited. In this study, the diversity of the bacterial community attached to the rumen epithelial tissue was compared to the rumen content bacterial community using 16S rRNA gene sequencing, PCR-DGGE, and qRT-PCR analysis. Sequence analysis of 2785 randomly selected clones from six 16S rDNA (~1.4kb) libraries showed that the community structures of three rumen content libraries clustered together and were separated from the rumen tissue libraries. The diversity index of each library revealed that ruminal content bacterial communities (4.12/4.42/4.88) were higher than ruminal tissue communities (2.90/2.73/3.23), based on 97% similarity. The phylum Firmicutes was predominant in the ruminal tissue communities, while the phylum Bacteroidetes was predominant in the ruminal content communities. The phyla Fibrobacteres, Planctomycetes, and Verrucomicrobia were only detected in the ruminal content communities. PCR-DGGE analysis of the bacterial profiles of the rumen content and ruminal epithelial tissue samples from 22 steers further confirmed that there is a distinct bacterial community that inhibits the rumen epithelium.
