Objective To investigate tissue residues of two longacting oxytetracycline (OTC) preparations in cattle.
Design A randomised drug residue trial.  

Animals


Two hundred and forty beef cattle in 24 groups of ten.
Procedure Two blind-coded 200 mg/mL OTC preparations were used in five treatment regimens of various combinations of injection sites (from one to five) and administrations (one or two). Five cattle from each group were slaughtered at 21, 30 and 60 days after injection and the injection site, urine, kidney and diaphragm muscle analysed for residues.
Results The OTC concentration exceeded the maximum residue limit in kidney in animals slaughtered 21 days after treatment, which is the prescribed withholding period. Concentration at the injection site was much greater than the maximum residue limit 30 days post-treatment, but not 45 days post-treatment. The residue was smaller when OTC had been injected in multiple sites. There was no difference between the two OTC preparations.
Conclusion A review of the maximum injection volume, site of injection and the withholding period is needed for long-acting OTC formulations.  相似文献

Objective To assess the persistent activity of injectable formulations of abamectin and doramectin against gastrointestinal nematodes of cattle.
Design Controlled slaughter study assessing residual efficacy.
Procedure Nematode-free calves were treated with abamectin or doramectin (each at a dose of 200 μg/kg) and infections then induced with repeated doses of infective larvae of Trichostrongylus axei, Haemonchus placei, Ostertagia ostertagi and Cooperia species. The duration of challenge ranged from 14 to 28 days. The calves were slaughtered at either 38/39 or 45/46 days after the treatments and nematodes recovered from the gastro-intestinal tract.
Results Significant reductions in numbers of O ostertagi occurred for both abamectin and doramectin treatments (> 93%) relative to counts in untreated calves, when challenge was administered up to 21 days after treatment. For T axei and Cooperia spp significant reductions occurred when the challenge occurred for 14 days after treatment (99%). Although differences from untreated animals were not significant, the results for H placei suggested high efficacy (> 85%) for up to 21 days for doramectin and up to 28 days for abamectin.
Conclusion There was no significant difference between abamectin and doramectin for any parasite at any challenge point, indicating that there is equivalent persistent activity of doramectin and abamectin against important gastrointestinal nematodes of cattle.  相似文献

Investigations were conducted on several small neighbouring beef cattle and sheep farms that were found to be contaminated with polychlorinated biphenyls (PCBs). The residues detected contained about 62% w/w chlorine and soil concentrations ranged from 0.35 to 1.9 mg/kg. Beef cattle and sheep grazing the contaminated land had PCB concentrations in their fat of 0.3 to 1.7 mg/kg and 0.19 to 0.45 mg/kg, respectively. The concentration of PCBs in the caudal fat of cattle was 0.3 to 2 times the concentration of PCBs in the land they were grazing and was positively related to stocking rates. PCBs were present in milk fat from cattle at about the same concentration as in caudal fat, and lactation appeared to contribute to decontamination. Wool grease from sheep contained about 1 to 2 times the concentration of PCBs in subcutaneous fat. Calves of contaminated cows also became contaminated. The half-life for decontamination of PCBs in a group of 8 young female beef cattle was calculated at 13 months. PCBs were detected in pasture samples at concentrations ranging from less than 0.01 mg/kg to 0.12 mg/kg. However, PCBs were not detected in any sample of hay cut from these paddocks. In 4 sheep fed a sole ration of this hay, traces of PCBs were detected in their fat following 89 days of feeding. Grazing livestock for meat production on land contaminated with PCBs is not recommended because livestock readily acquire residues, and PCBs persist in soil and livestock for long periods. Grazing sheep for wool production on land contaminated with PCBs may be an option.  相似文献

The single intradermal comparative test was used with both avian and bovine tuberculin. Three cattle infected with bovine leukosis virus (BLV) were used as a source of infection. BLV-positive and susceptible animals were tuberculin tested alternately. Fifteen susceptible calves and 15 susceptible sheep were tested. A further three calves and three sheep were used as controls; the needles of the tuberculin syringes were deliberately contaminated with blood from the BLV-infected cattle, before being used in the test. Whereas all three calves and the three sheep inoculated intradermally with contaminated needles developed BLV infections, all of the other 30 animals have remained serologically negative to BLV for 10 months. Transmission of BLV with needles contaminated with BLV-infected blood was prevented by wiping the needles with absorbent cotton wool.  相似文献

Absorption by sheep of dieldrin from contaminated soil   总被引:1,自引:0,他引:1  
Objective To study the accumulation of dieldrin residues in sheep from ingestion of contaminated soils was studied in two experiments.
Design A controlled feeding study of sheep fed contaminated soils of different type at varying intervals.
Animals and procedure Thirty-four 2-years-old wethers were divided into four groups (one control sheep only) and fed water-soluble dieldrin or soil contaminated with aldrin and dieldrin at varying intervals in the first study. In a second study 34 similar sheep were divided into four treatments with one being a control. Sheep were fed sandy, high clay or high organic matter soils with similar dieldrin and aldrin concentrations.
Results In the first study the concentration of dieldrin in the body fat of sheep dosed with dieldrin-contaminated soil was about half that in the body fat of sheep dosed with an equivalent amount of water-soluble dieldrin. The concentration of dieldrin was almost the same in sheep fed 500 μg of total dieldrin per day as it was in sheep fed 5000 μg every tenth day, over a 50-day period. In the second experiment sheep accumu-lated nearly three times as much pesticide from a soil with a high organic matter content, and about four times as much from a soil with a high clay content, as from a sandy soil with the same dieldrin content, over a 100-day period. The half-life of dieldrin in the fat of all sheep varied between 96 and 116 days after sheep ceased ingesting contaminated soil.
Conclusions Dieldrin concentrations in the fat of sheep that consume dieldrin contaminated soil fall within 10 days of removal from the source of contamination. However, dieidrin accumulates in the wool of sheep that consume dieldrin-contaminated soil.  相似文献

Psoroptes ovis was not transmitted by natural contact to susceptible cattle which were exposed to infested, ivermectin-treated cattle 6, 12, 14, 16, and 18 days after treatment was given. However, clinical scabies did develop in 2 calves naturally exposed to P ovis-infested, ivermectin-treated calves at 10 days after treatment was given subcutaneously (200 micrograms/kg). Psoroptes ovis was transmitted to stanchioned cattle manually exposed to 200 to 300 ml of hair and skin scrapings from infested, ivermectin-treated cattle at 6, 10, 12, 14, and 16 days after treatment was given subcutaneously (200 micrograms/kg). Scabies did not develop in cattle exposed to skin scrapings obtained from infested, treated cattle at 18 and 20 days after they were treated with ivermectin. The 14-day isolation of P ovis-infested, ivermectin-treated cattle from susceptible cattle recommended by the US Department of Agriculture, although marginal, is adequate under natural conditions to prevent transmission of scabies from treated to noninfested cattle.  相似文献

Transmission of Mycobacterium avium subspecies paratuberculosis, the organism responsible for paratuberculosis (or Johne's disease) in ruminants, occurs through the faecal-oral route. As M. a. paratuberculosis has been isolated from rabbit faeces, cattle grazing rabbit faecal contaminated pasture may thus be at risk.A herd of 57 beef cattle was monitored on a farm in Perthshire, throughout the 1999 'grazing year', to investigate whether the cattle avoided rabbit faecal contaminated pasture and thus the potential for disease transmission. Grazing was measured every two days over eight rotations by sward heights on 40 marked treatment plots (0.5 m x 0.5 m) to which 0, 10, 50 and 250 rabbit faecal pellets were added. Cattle were also monitored by an active transponder system which enabled individual animals contacting two plots per field rotation (one control and one contaminated) to be recorded. During the monitored grazing year, grazing pressure was low with a net mean sward offtake of 18% of sward height per rotation. There were no significant differences between rabbit faecal treatments (0, 10, 50 and 250 pellets) with respect to the height or proportion of sward removed, or between the numbers of contacts made by cattle on contaminated and uncontaminated plots. Over 90% of all the cattle contacted contaminated plots, indicating that the potential for disease transmission was widespread among the herd.To our knowledge, this is the first reported instance of a lack of avoidance by grazing cattle towards swards contaminated with faeces, and implies that the potential for transmission of paratuberculosis from rabbit contaminated pasture is high.  相似文献

Objectives To compare the survival of larvae of a dung-breeding fly in the faeces of cattle treated either with an injectable formulation of abamectin, or with oral or injectable formulations of ivermectin.
Design Replicated bioassays were conducted on larvae of the bush fly, Musca vetustissima, using faeces collected before and at intervals after drug treatment.
Animals Two cows and their calves were allocated to each of three drug treatments and dosed according to individual weights.
Procedures Differences in the proportions of larvae pupariating were used as measures of the toxicity of drug residues.
Results Development of fly larvae was inhibited in all faeces collected 1 to 4 days after treatment. In cattle treated with oral ivermectin, there was reduced larval survival in faeces collected 8 and 16 days after treatment, but by day 32, survival was equivalent to that recorded in the faeces of untreated cattle. With injectable ivermectin, there was no survival at day 8, limited survival at day 16 and, at day 32, survival was not significantly affected. With injectable abamectin, survival was completely suppressed until day 32, at which time the number of pupariating larvae did not differ significantly from that recorded in faeces from untreated animals.
Conclusion The oral formulation of ivermectin is eliminated more rapidly than the injectable formulation and, as a consequence, is likely to be less harmful to dung-feeding insects. Abamectin and ivermectin appear to equally toxic larvae of M vetustissima.  相似文献

Objective — This report describes our experience with a new technique for stabilization of mandibular fractures in cattle using a pinless external fixator.
Study Design — Mandibular fractures were stabilized with a pinless external fixator, which is a modification of a unilateral AO/ASIF (Association for the Study of Internal Fixation)-fixator in which pins are replaced with bone clamps.
Animals — Seven dairy cattle between 1 and 5 years of age.
Methods — Fracture stabilization was achieved by using the pinless external fixator. Postoperative management included intravenous administration of sodium chloride-glucose and antibiotics. A suspension of ruminal contents from a normal cow together with linseed and grass pellets was administered with a gastric tube.
Results — Three to 10 days after surgery six of seven cows masticated comfortably. The only failure was a yearling with a 10-day-old open infected fracture. This animal was slaughtered 9 days after surgery because of additional problems. In six cases there was enough callus formation 33 to 54 days after surgery to stabilize the fracture. The fixation devices were removed under heavy sedation. The major complication was bone sequestration at the fracture site, which required additional treatment.
Conclusion — The pinless fixator has proven satisfactory for external stabilization of unilateral horizontal ramus fractures of the mandible in cattle.
Clinical Relevance — The technique provides good stability without penetration of the medullary cavity and damage to the tooth roots. Other advantages of the technique include ease of application, minimal surgical trauma, and the short surgical time for application.  相似文献

Objective To assess the toxicity to insects of drug residues excreted in cattle faeces following treatment with deltamethrin.
Design Bioassays were performed on one species of dung-breeding fly ( Musca vetustissima ) and two species of dung beetle ( Onthophagus binodis and Euoniticellus fulvus ).
Animals Cattle on properties near Kangaroo Valley, Canberra and Gundagai were treated with pour-on formulations of deltamethrin. Untreated animals acted as controls.
Procedures Faeces from treated and untreated cattle were inoculated with newly emerged fly larvae or fed to adults of two species of dung beetle. Percentage survival and duration of development provided measures of the toxicity of deltamethrin residues in faeces.
Results Residues of deltamethrin were excreted in concentrations sufficient to inhibit survival of larvae of M vetustissima for 1 to 2 weeks after treatment. Peak concentrations of 0.4mg deltamethrin/kg dry weight of faeces occurred 3 days after treatment and were sufficient to kill adult beetles for at least twice this period. With one of two formulations tested, there was evidence of a reduction in dung beetle fecundity and an increase in the duration of juvenile development. A model of the effect of deltamethrin on the breeding success of dung beetles in the field suggests that a single treatment, applied when most of the population is in a non-parous condition, may cause up to 75% reduction in beetle acitivity by the end of the season. Multiple treatments at 10 or 21 day intervals may drive local populations towards extinction.
Conclusion Depending on the time and frequency of treatment, the effect of deltamethrin on insects in cattle faeces may range from negligible to catastrophic.  相似文献

Objective To estimate the specificity of an absorbed enzyme-linked immuno-sorbent assay kitd for Johne's disease (JD) when used in mature cattle populations resident in northern Australia.
Design Blood samples were collected from beef cattle in northern Queensland, the Northern Territory and northern Western Australia, and from dairy cattle in northern Queensland. The specificity of a serological test for JD was estimated by testing the blood samples with an absorbed ELISA kit. Further samples were collected from cattle with positive ELISA results to determine the presence or absence of infection with Mycobacterium avium subsp paratuberculosis .
Procedure During 1995 and 1996, blood, tissue and gut contents were collected from beef cattle at abattoirs in Queensland and the Northern Territory; and blood and faecal samples were collected from dairy cattle in herds assessed to be most at risk for JD in northern Queensland. The blood samples were tested using an absorbed ELISA kit. Tissues and gut contents from beef cattle that had positive ELISA results were cultured for M avium subsp paratuberculosis , and tissues were examined histo-logically. Faecal samples from dairy cattle with positive ELISA results were cultured for M avium subsp paratuberculosis .
Results Estimates of specificity for this absorbed ELISA in mature northern Australian cattle were 98.0% (97.0 to 98.8%, 95% CI) in beef cattle, and 98.3% (96.7 to 99.3%, 95% CI) in dairy cattle.
Conclusion Estimates of specificity in this study were lower for beef cattle from the Northern Territory and northern Western Australia and for dairy cattle from northern Queensland than those quoted from studies on cattle in southern Western Australia. This should be considered when serological testing using the JD ELISA is carried out on northern Australian cattle.  相似文献

Objective To determine if an association existed between sickness, mortality and bullers in a western Canadian feedlot.
Design A retrospective epidemiological study.
Animals 78,445 male cattle that entered a 24,000-head feedlot in western Canada from 1991 to 1993.
Procedure Animal health records for bullers were collected and analysed to see if they were at greater risk of sickness and mortality than other steers, and to see if pens with a high prevalence of bullers also had a high prevalence of sickness and mortality.
Results The prevalence of bullers increased with increasing age of cattle on arrival at the feedlot (R = 0.36; P < 0.001). Sickness and mortality decreased with increasing age of cattle on arrival. However, sickness and mortality in bullers relative to other steers actually increased with increasing age on arrival suggesting an interaction existed between sickness and bullers. Bullers were significantly (P < 0.05) more likely to get sick and to die than other steers. In all cases, there was a strong temporal association between sickness and bullers, with sickness and bullers mostly occurring within the first 30 days of the feeding period. On average, pens of cattle with a high prevalence of bullers did not have a correspondingly high prevalence of sickness or mortality.
Conclusion This study suggests that sickness is an effect modifier of dominance behaviour and therefore bullers in feedlot steers. Bullers should always be checked for signs of sickness and treated accordingly. Further research is needed to investigate the effects of sickness on dominance behaviour in pens of feedlot cattle.  相似文献

The aim of the experiment was to study whether bovine herpesvirus 1 (BHV1) marker vaccine batches known to be contaminated with bovine virus diarrhoea virus (BVDV) type 1 could cause BVD in cattle. For this purpose, four groups of cattle were used. The first group (n = 4 calves, the positive control group), was vaccinated with vaccine from a batch contaminated with BVDV type 2. The second group (n = 4 calves, the negative control group), was vaccinated with vaccine from a batch that was not contaminated with BVDV. The third group (n = 39 calves), was vaccinated with a vaccine from one of four batches contaminated with BVDV type 1 (seronegative experimental group). The fourth group (n = 6 seropositive heifers), was vaccinated with a vaccine from one of three batches known to be contaminated with BVDV type 1. All cattle were vaccinated with an overdose of the BHV1 marker vaccine. At the start of the experiment, all calves except those from group 4 were seronegative for BVDV and BHV1. The calves from group 4 had antibodies against BVDV, were BVDV-free and seronegative to BHV1. After vaccination, the positive control calves became severely ill, had fever for several days, and BVDV was isolated from nasal swabs and white blood cells. In addition, these calves produced antibodies to BVDV and BHV1. No difference in clinical scores of the other groups was seen, nor were BVDV or BVDV-specific antibody responses detected in these calves; however, they did produce antibodies against BHV1. The remainder of each vaccine vial used was examined for the presence of infectious BVDV in cell culture. From none of the vials was BVDV isolated after three subsequent passages. This indicates that BVDV was either absent from the vials or was present in too low an amount to be isolated. Thus vaccination of calves with vaccines from BHV1 marker vaccine batches contaminated with BVDV type 1 did not result in BVDV infections.  相似文献

Background: Commercially available cardiac troponin I (cTnI) assays developed for use in humans have not yet been validated for use in cattle.
Hypotheses: The ADVIA Centaur TnI-Ultra immunoassay can be used for the detection of bovine cTnI. In healthy cattle, serum cTnI is undetectable or is present only in trace amounts.
Methods: Purified bovine cTnI and cTnI-free bovine serum were used for the evaluation of assay performance including intra- and inter-assay precision, sensitivity, interference, linearity, and recovery. Effects of storage at 23, 4, −20, and −80 °C for 2 days, and at −20 and −80 °C for 7 and 14 days and repeated freeze-thaw cycles on recovery of cTnI were analyzed. Serum cTnI concentrations in 30 healthy dairy cows were determined.
Results: Intra- and inter-assay precisions (mean ± SD) were 4.48 ± 2.26 and 13.36 ± 6.59%, respectively. The assay demonstrated linearity at 0.5, 2, 15, and 30 ng/mL cTnI. Mean recovery was 100.81, 85.26, 87.72, and 114.42%, respectively. Skeletal muscle homogenate added to serum of known cTnI concentration did not alter the concentration of the analyte ( P > .05). Concentration of cTnI significantly decreased when samples were stored at 4 and 23 °C for 2 days ( P < .05). Repeated freeze-thaw cycles and storage at −20 °C for 7 days had no significant influence on cTnI concentration ( P > .05). Serum cTnI concentration in healthy cattle was ≤0.03 ng/mL.
Conclusion and Clinical Importance: ADVIA Centaur can be used reliably for the detection of serum cTnI concentration in cattle.  相似文献

Brucella abortus infection in 14 farm dogs   总被引:1,自引:0,他引:1  
Fourteen dogs were obtained from 10 farms with Brucella-infected cattle and were studied for periods ranging from 2 to 81 days. At necropsy, Brucella abortus biovar 4 was isolated from all 14 dogs. Mandibular, medial retropharyngeal, tracheobronchial, and mesenteric lymph nodes yielded the highest rate of recovery. Urogenital infection with active shedding was seen in a single aged bitch. Fecal samples (291 from 13 dogs) were B abortus culture negative. Ten dogs monitored serologically over time had standard tube agglutination test titers that fluctuated between 1:50 with incomplete reaction and greater than or equal to 1:200 with positive reaction. At necropsy, the magnitude of seroconversion was not directly related to the number of culture-positive tissues. The maximal duration of infection, based on the time interval between the last possible exposure to infected cattle and recovery of the organism at necropsy, was 464 days. If it were assumed that infection occurred at about the same time as seroconversion in the cattle herd of origin, maximal observed duration of infection was 539 days. The actual maximal duration of infection could not be determined from this study, but would have exceeded the values reported here. The data indicate that dogs have the potential to infect cattle and could pose a threat for longer duration of disease transmission than had previously been assumed. Although the risk of transmission appears small, infection of human beings or cattle associated with Brucella-infected dogs would cause unfavorable political and economic consequences, particularly in low-incidence areas. Removal of contact dogs from infected herds should prevent such transmission.  相似文献

Thirteen primiparous and 41 multiparous Holstein-Friesian cattle were used to study the relationship between maternal plasma progesterone (P4) and estrone sulfate (E1S) concentrations and the prevalence of dystocia. The calvings in 4 heifers and 30 cows were normal (eutocia), while the calvings in 9 heifers and 11 cows were difficult (dystocia). Neither the concentrations of P4 nor E1S were different between the groups with eutocia and dystocia from days 90 to 270 of pregnancy. However, a few days prior to parturition, eutocial cows and heifers showed a sharp decline of plasma P4, while dystocial cattle did not show such a remarkable decline of P4 concentration. Plasma P4 levels in dystocial cows a few days antepartum were significantly higher than in eutocial animals (P<0.05 or P<0.01). Prepartum E1S concentrations were significantly lower (P<0.05) in dystocial than eutocial cattle during the prepartum period from days 6 to 1 in heifers and from days 3 to 1 in cows. These results suggest that insufficient production of E1S and delayed regression of the corpora lutea are possible causes of dystocia in cattle.  相似文献

Objective To determine the minimum alveolar concentration (MAC) of isoflurane in cattle.
Study design Prospective study.
Animals Sixteen healthy adult female Holstein-Friesian cattle weighing 612 ± 17 kg (× ± SEM) and aged 5.7 ± 0.9 years old.
Methods The unsedated cattle were restrained in right lateral recumbency using a rope harness technique. Anaesthesia was induced with isoflurane (ISO) in oxygen via a face mask connected to a large-animal, semiclosed anaesthetic circle system. Each cow was intubated with a cuffed orotracheal tube (25 mm ID). Inspired and end-tidal ISO were monitored using a calibrated infra red analyser with a methane filter. The MAC of ISO that prevented gross purposeful movement in response to a tail and dewclaw clamp was determined. The time from the start of ISO administration to intubation, the time interval between discontinuance of ISO and the time the animal regained sternal recumbency, were recorded. Time to standing and quality of recovery were also recorded.
Results The time from the start of ISO administration to tracheal intubation was 18.68 ± 2.77 minutes. The MAC of ISO in these cattle was 1.27 ± 0.03% (1.14 ± 0.01% corrected to sea level). Time to sternal recumbency after 90 ± 16 minutes of anaesthesia from intubation was 4.60 ± 0.58 minutes and time to standing was 6.70 ± 1.02 minutes. All cattle were extubated when they regained sternal recumbency.
Conclusion The MAC of isoflurane in these cattle was 1.27 ± 0.03% (1.14 ± 0.01% corrected to sea level). ISO provided a smooth induction to, and rapid recovery from, anaesthesia.
Clinical relevance Knowledge of the MAC of ISO in cattle will facilitate its appropriate clinical use.  相似文献

Objective To determine practices for the control of cattle ticks on dairy farms in Queensland, the attitudes of farmers to tick infestations and to identify opportunities for and barriers against the introduction of non-chemical methods of tick control.
Design A survey of 199 dairy farmers from tick-infested parts of Queensland was undertaken by 20 dairy advisers and stock inspectors from October 1996 to June 1997. The sample was a proportional, random selection of dairy farms from four regions. A personal interview was conducted with each farmer and answers to 134 questions were obtained.
Results and conclusions Most farmers were not concerned by cattle ticks on their own farms, although they believed that ticks are important to the dairy industry. They were most concerned about the development of chemical resistance by cattle ticks. Inadequate facilities and lack of motivation appeared to be the factors most limiting to improving the methods of control. Most farmers claimed to have only small numbers of ticks at worst. Although a control program recom mended by the Queensland Dairyfarmers' Organisation was well regarded by farmers, few had adopted it. Many farmers saw no need to implement a strategic control program.  相似文献

The changes in concentration of heptachlor epoxide (HCE) and oxychlordane (OCD) were studied in the tissues of beef cattle that had previously grazed pasture contaminated with commercial heptachlor. In 25 cows and heifers monitored over 488 days, the mean concentration of HCE decreased from 22.0 to 0.08 mg/kg and of OCD from 5.68 to 0.18 mg/kg. The respective half-lives were estimated to be 66 and 92 days. In a controlled feeding experiment, nine steers were subjected to one of three dietary regimes; a high plane of nutrition for 81 days then a low plane for 140 days; a moderate plane of nutrition for the total period; and a low plane of nutrition for 81 days then a high plane for 140 days. Approximately five months later similar treatments were applied for 101 and 94 days. The rate of decline of HCE and OCD was consistently greatest in animals on the high plane of nutrition and lowest in those on the low plane. Lactation did not appear to have a major effect on the rate of decline in the tissue concentrations of these compounds. No significant (P greater than 0.05) relationships were found between the concentrations of HCE or OCD in the subcutaneous fat and either milk or blood at any stage of the experiment. At slaughter, from 18 to 24 months after the commencement of the experiments, significant correlations (P less than 0.05) were found among concentrations of HCE and OCD in subcutaneous fat, renal fat and fat from the bone marrow.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
