Tick-borne fever (TBF) is caused by the rickettsiae Ehrlichia phagocytophila and is a common disease in sheep in tick (Ixodes ricinus) infested areas in Norway. Earlier investigations have shown that some sheep could remain infected for several months after the primary infection. In this study, the persistence of E. phagocytophila after experimental infection was investigated in 2 age groups of lambs. Six lambs (1-2 weeks old) and 14 lambs (6-8 months old) were inoculated intravenously with an ovine strain of E. phagocytophila and thereafter examined clinically (including daily body temperature recording) and by haematological and serological (E. equi antibodies) methods for the next 4 months. At the end of this period, the lambs were examined for a TBF infection by blood smear investigation and blood inoculation studies. The infection was demonstrated in 19 (95%) of the 20 lambs.  相似文献

The significance of tick-borne fever (TBF) and other tick-borne diseases of British sheep are reviewed. Experimental and field studies were carried out to clarify the role of TBF as a pathogen per se and as a predisposing factor in other diseases. Experimental TBF infection caused anorexia and depression in two- to three-week-old lambs, which under the stress of a hill environment could alone be a cause of mortality. Nine out of 10 lambs experimentally inoculated with Staphylococcus aureus during the febrile phase of a TBF reaction developed pyaemic lesions compared with four out of 20 lambs inoculated with S aureus alone. Specific pathogen-free lambs inoculated with an aerosol of Pasteurella haemolytica serotype A1 during a TBF reaction showed more severe clinical signs and had more extensive pathological changes at necropsy than control lambs given P haemolytica alone. Dual infection with TBF and louping-ill virus showed that not only were dually infected sheep more susceptible to louping-ill but almost all of them succumbed to a haemorrhagic syndrome involving a systemic mycotic infection with Rhizomucor pucillus. None of eight sheep given louping-ill virus alone developed this syndrome. Field studies indicated that morbidity and mortality in lambs in south-west Scotland could be markedly reduced by dipping and long acting antibiotic prophylaxis. Lamb groups in which both of these were carried out incurred losses of only 0.6 per cent compared with 10.3 per cent in control groups. Anaplasma phagocytophilum infects a wide variety of host species and causes the diseases tick-borne fever (TBF) in ruminants and granulocytic anaplasmosis in humans, horses and dogs. TBF in sheep has become one of the more prevalent tick-borne diseases in some regions of Europe. A. phagocytophilum infection modifies host gene expression and immune response. The objective of this research was to characterize differential gene expression in sheep experimentally and naturally infected with A. phagocytophilum by microarray hybridization and real-time RT-PCR. The results of these studies demonstrated in sheep the activation of inflammatory and innate immune pathways and the impairment of adaptive immunity during A. phagocytophilum infection. The characterization of the genes and their expression profiles in sheep in response to A. phagocytophilum infection advances our understanding of the molecular mechanisms of pathogen infection and the pathogenesis of TBF. Collectively, these results expand current information on the mammalian host response to A. phagocytophilum infection.  相似文献

以家蚕真菌病病原为受试菌,对抗真菌药物氟康唑(FCZ)和盐酸特比萘芬(TBF·HCl)进行了体外筛选,研究了盐酸特比萘芬对家蚕真菌病的防治效果及其稳定性。试验结果表明,盐酸特比萘芬的体外抑菌、杀菌效果优于氟康唑,盐酸特比萘芬对家蚕病原真菌的最小抑菌浓度范围为0.0625~0.2726μg/mL,最小杀菌浓度范围为0.125~4μg/mL。生物试验表明,盐酸特比萘芬对家蚕真菌病防治效果良好,200μg/mL时防治效果达100%,且对家蚕生长发育、茧质无不良影响。稳定性试验表明盐酸特比萘芬长期稳定性良好。  相似文献

Background

Anaplasma phagocytophilum (formerly Ehrlichia phagocytophila) is an obligate intracellular bacterium causing the disease tick-borne fever (TBF) in domestic ruminants. An effective vaccine against the infection has been demanded for livestock by sheep farmers and veterinary practitioners for years.

Findings

In the present study, we immunized lambs with an inactivated suspension of 1 × 108 killed A. phagocytophilum organisms mixed with adjuvant (Montanide ISA 61VG; Seppic). Twelve 9-months-old lambs of the Norwegian White Sheep breed were used. A full two-dose series of immunization was given subcutaneously to six lambs with a 4 week interval between injections. One month after the last immunization, all lambs were challenged with the homologous viable variant of A. phagocytophilum. After challenge, all lambs showed clinical responses for several days, although the immunized lambs reacted with an anamnestic response, i.e. significant reduction in infection rate and a significantly higher antibody titer.

Conclusion

Immunization with inactivated A. phagocytophilum did not protect lambs TBF.  相似文献

The use of synthetic pyrethroids in pour-on formulations reduced tick infestations and the incidence of tick-associated diseases in lambs more than dipping in organophosphate acaricides. Though the use of pyrethroids did not prevent the lambs from being infected with tick-borne fever (TBF), the incidence of lambs with lameness (tick pyaemia) or lambs suddenly found dead (Pasteurella haemolytica septicaemia), which often are seen in association with TBF, was reduced. The use of pyrethroids for three years did not seem to affect the prevalence of TBF.  相似文献

Tick-borne fever (TBF) is a rickettsial disease of domestic and wild ruminants in temperate climates. It is characterized by high fever and severe leukopenia. In the present study, the possible difference in the severity of disease in sheep and goats was investigated by inoculating one group of eight goats and one group of eight sheep with the Old Sourhope (OS) strain of Ehrlichia (Cytoecetes) phagocytophila. All sheep and goats experimentally infected with E. phagocytophila reacted with fever and rickettsiaemia, but there were significant differences between goats and sheep in the severity of clinical disease, the duration and magnitude of fever, the magnitude of rickettsiaemia and the patterns of reduction in the number of total leucocytes. Sheep reacted with fever significantly earlier than goats and the febrile period lasted for a significantly longer period. In contrast, the magnitude of rickettsiaemia was significantly higher in goats than in sheep. Infection with TBF was characterized by a transient increase in the number of neutrophils, which was quickly followed by an acute reduction in the number of lymphocytes and a prolonged reduction in the number of neutrophils in both sheep and goats. In both groups of animals, infection with TBF was also characterized by significant reductions in the total number of red blood cells (RBCs), thrombocytes and packed cell volume (PCV) and the concentration of haemoglobin (Hb). However, the mean corpuscular volume (MCV) and mean corpuscular Hb (MCH) were significantly increased in sheep only.  相似文献

Four cows from North-West Germany have been diagnosed with tick-borne fever (TBF) based on the demonstration of morulae in neutrophilic granulocytes in their blood smears, positive signals in real-time PCR specific for Anaplasma phagocytophilum using DNA extracted from their buffy coats, and demonstration of specific antibodies in their sera using a commercially available immunofluorescence assay. Clinical findings included high fever, decreased milk production, lower limb edema with stiff walking, eye and nasal discharge, and depression. These signs developed about a week after the animals had been brought to the pasture for the first time in their life. All cows recovered after 5-15 days, although DNA of A.phagocytophilum could be detected by real-time PCR up to 6 weeks after onset of the disease. Considering the known prevalences of A.phagocytophilum in ticks in Germany and its detection in dogs and horses, we think that underdiagnosing of TBE in cattle is highly likely. Therefore TBF should be taken into account as differential diagnosis in case of high fever and/or a sudden decrease in milk production in pastured animals.  相似文献

The effect of tick-borne fever (TBF) on the plasma disposition of sulphadimidine (SDM) and its metabolites in goats was studied. In uninfected goats, SDM was extensively metabolised mainly by hydroxylation, glucuronidation and to a minor extent by acetylation. In TBF infected goats the hydroxylation of SDM into 6-methylhydroxysulphadimidine (SCH2OH) as well as into 5-hydroxysulphadimidine (SOH) was markedly reduced (-57.6 and -63.6 per cent, respectively). An unidentified metabolite (metabolite X) was detected, which was largely glucuronidated in the uninfected goats. In the TBF infected goats the glucuronide derivatives of the X metabolite and of SOH were barely detectable. In TBF infected goats the plasma concentration of the N4-acetylated metabolite (N4-SDM) was decreased to a lesser extent (-22.1 per cent) than the hydroxy metabolites. Due to the diminished metabolism the elimination half-life of SDM was increased 1.8 times and the total sulphonamide body clearance was diminished compared with findings in the control experiments.  相似文献

Lambs aged 2 weeks were inoculated with a tick-borne fever (TBF) stabilate on Day 0 and Staphylococcus aureus-contaminated ticks were applied on Day 5. Tick pyaemia was produced experimentally for the first time using Ixodes ricinus as a mechanical vector of S. aureus. Lambs aged 18 weeks were rechallenged with a homologous strain of TBF, and S. aureus-infected ticks applied 5 days later. No significant changes were noted at post-mortem examination.  相似文献

In Europe, clinical manifestations due to A. phagocytophilum have been recorded in sheep, goat, cattle, horse, dog, cat, roe deer, reindeer and human. However, seropositive and PCR-positive mammalian have been detected in several other species. Investigations indicate that the infection is prevalent in Ixodes ricinus areas in most countries in Europe. A. phagocytophilum infection may cause high fever, cytoplasmatic inclusions in phagocytes and severe neutropenia, but is seldom fatal unless complicated by other infections. Complications may include abortions, and impaired spermatogenesis for several months. However, the most important aspect of the infection at least in sheep is its implication as a predisposing factor for other infections. Factors such as climate, management, other infections, individual conditions etc. are important for the outcome of the infection. A. phagocytophilum may cause persistent infection in several species. Based on the 16S rRNA gene sequences several variants exist. Different variants may exist within the same herd and even simultaneously in the same animal. Tick-borne fever (TBF) is a rickettsial disease of domestic and wild ruminants in temperate climates where the hard tick Ixodes ricinus is present. The disease is characterized by a high temperature and severe leukopenia. In the present study, the effects of TBF on the activity of serum alkaline phosphatase (ALP), and on the concentrations of plasma zinc, iron, total bilirubin, urea, creatinine and albumin were investigated by inoculating one group of eight sheep and one group of eight goats with the Old Sourhope (OS) strain of Ehrlichia (Cytoecetes) phagocytophila. All goats and sheep experimentally infected with E. phagocytophila reacted with fever, rickettsiaemia and leukopenia. The leukopenia was due to an acute lymphocytopenia and prolonged neutropenia. In both groups of animals. TBF was characterized by significant reductions in the activities of serum ALP and concentrations of plasma zinc, iron and albumin. However, there were significant increases in the concentrations of plasma total bilirubin, urea and creatinine in both species of animals. The reductions in ALP and iron were significantly more pronounced in sheep than in goats.  相似文献

Four groups of lambs aged 1 week, 4 weeks, 1/2 year and 1 year old respectively were inoculated with Ehrlichia phagocytophila infected blood. Clinical signs, temperature reactions, haematological changes and parasitaemia were more moderate in lambs inoculated with E. phagocytophila at the age of 1 week than those recorded in the older animals. The clinical response to tick-borne fever (TBF) appears to be more severe with increasing age of the lambs. The lymphocyte reactivity to mitogens was reduced in the TBF infected lambs, and was most pronounced in lambs in the 3 older age groups.  相似文献

Sheep is apparently fairly resistant against infections with Listeria monocytogenes (Lm). Grønstøl (unpublished) found that a high proportion of healthy sheep are carriers of Lm. In such animals Lm may cause clinical disease when resistance is lowered. Tick-borne fever (TBF) is known to reduce the resistance against several infections (Øverås 1972). Anaplasma phagocytophilum is a Gram-negative, obligate intracellular tick-transmitted bacterium that replicates in neutrophils. It causes tick-borne fever (TBF) in sheep and cattle, but also elicits febrile disease in humans as well as in other domestic animals such as dogs, horses, and cats. Although increasingly recognized in Europe, the first laboratory-confirmed case of TBF in cattle from Germany has been published only recently. We here present the unusual case of an intrauterine transmission of A. phagocytophilum in a calf from northern Germany. To our knowledge, this is the first report of such an event occurring under field conditions in cattle.  相似文献

Use of real-time ultrasonography in meat animal improvement programs has increased largely as a result of the perceived accuracy of such instruments in measuring longissimus muscle area (LMA) and fat depth (BF). The objective of this study was to examine operator effects for ultrasonic determination of BF and LMA in market-weight cattle, sheep and pigs. The day prior to slaughter, five persons scanned 45 pigs, 32 sheep and 18 cattle at the last rib using an A-mode Ithaco Model 731AM101 body composition meter (ABF) and a Johnson and Johnson Ultrasound 210DX B-mode scanner fitted with a 3.0 MHz probe. All operators measured their own B-mode recordings (OBF, OLMA); an independent, experienced technician also obtained measurements from all recordings (TBF, TLMA). Species x machine x operator interaction effects were detected (P less than .001) for ultrasonic BF. Within-species analyses also revealed that the machine x operator interaction was important (P less than .001) for all species. Analyses of ABF, OBF, TBF, OLMA and TLMA detected species x operator interactions (P less than .001) for all traits except TLMA (P less than .20). Assuming a random model with animal, operator and residual effects, variance components were estimated by maximum likelihood for each species. Repeatabilities, calculated as the intraclass correlation among animals for BF and LMA scanned and interpreted by different operators, were .13, .21 and .51 (OBF) and .28, .15 and .29 (OLMA) for cattle, sheep and pigs, respectively, indicating the importance of operator effects. Repeatabilities of TBF were .90, .42 and .68 and of TLMA were .19, .55 and .65 for cattle, sheep and pigs, respectively, indicating the importance of image interpretation in determining operator effects associated with use of the 210DX B-mode scanner. Technical training standards must, therefore, be established for operators of such equipment.  相似文献

Anaplasma phagocytophilum, the causative agent of tick-borne fever (TBF) in sheep and cattle and human granulocytic anaplasmosis, has the unique ability to infect and multiply within neutrophils, eosinophils and monocytes, cells at the frontline of the immune system. Infection with A. phagocytophilum is also characterized by severe leukopenia due to lymphocytopenia, neutropenia and thrombocytopenia lasting for several days. By itself TBF does not cause high mortality rates but infected animals are more susceptible to other secondary infections, pregnant animals may abort and there is a severe reduction in milk yield in dairy cattle. The susceptibility to secondary infections can be attributed to the leukopenia that accompanies the disease and the organism's adverse effects on lymphocyte and neutrophil functions. One of its fascinating features is that it infects and actively grows in neutrophils by employing an array of mechanisms to subvert their bactericidal activity. These include its ability to inhibit phagosome-lysosome fusion, to suppress respiratory burst and to delay the apoptotic death of neutrophils. It is also able to survive within an apparently immune host by employing a complex mechanism of antigenic variation.  相似文献
