The present study explores the effect of dietary sodium propionate on mucosal immune response and expression of antioxidant enzyme genes in zebra fish (Danio rerio). Six hundred healthy zebra fish (0.42 ± 0.06 g) supplied, randomly stocked in 12 aquariums and fed on basal diets supplemented with different levels of sodium propionate [0 (control), 5, 10 and 20 g kg?1] for 8 weeks. At the end of the feeding trial, mucosal immune parameters (TNF-α, IL-1β, Lyz), antioxidant enzyme (SOD, CAT) as well as heat shock protein 70 (HSP70) gene expression were measured. The results revealed feeding on sodium propionate significantly up-regulated inflammatory response genes (TNF-α, IL-1β, Lyz) in a dose-dependent manner (P < 0.05). However, antioxidant enzyme genes significantly down-regulated in the treated group compared with control (P < 0.05). Also, HSP70 gene expression was higher in the liver of fish fed the basal diet and deceased with elevation of sodium propionate levels in the diet. These results showed beneficial effects of dietary sodium propionate on mucosal immune response as well as the antioxidant defense of zebra fish.  相似文献

The distribution and expression of lymphocystis disease virus (LCDV) vaccine, on the basis of DNA vaccine (pEGFP-N2-LCDV0.6 kb) construction, were analyzed in tissues of the Japanese flounder by PCR, RT-PCR and fluorescent microscopy. Results from PCR studies indicated that the vaccine-containing plasmids were distributed in injected muscle, muscle located opposite the injection site, hind intestine, gill, spleen, head kidney, liver and gonad 7 days after vaccination. However, these vaccine-containing plasmids disappeared by 90 days following vaccination. Fluorescent microscopy observations revealed that green fluorescence appeared in muscle, muscle located at the opposite side of the injection site, hind intestine, gill, spleen, head kidney and liver of fish 36 h after vaccination, and that green fluorescence did not appear in control tissue. The green fluorescence became weaker at 60 days post-vaccination, however, it remained detectable in the spleen 90 days post-vaccination. Results from RT-PCR studies indicated that the Mcp gene is expressed in all tissues of vaccinated fish 7-20 days after vaccination. These results demonstrate that the DNA vaccine is distributed and expressed in different tissues of vaccinated fish, and therefore, may have provided an antigen producing specific immune response.  相似文献

In this study, efficacy of three antigenic preparations from the fish pathogen Aeromonas hydrophila was evaluated as vaccine candidate in rohu Labeo rohita. The rabbit anti-rohu globulin conjugate was prepared in rabbit using sera obtained from healthy adult rohu. Three antigens, namely formalin inactivated A. hydrophila (FAH), FAH mixed with Freund’s incomplete adjuvant in a 1:1 (v/v) ratio (FAH + A), and extra cellular product were prepared. Protection of rohu against A. hydrophila infection was tested at days 10, 20, and 30 post-vaccination. At the end of the vaccination, fish were challenged with A. hydrophila (dose: 1 × 107 cfu ml?1) and relative per cent of survival was recorded up to 60th day of post-challenge. Study of cellular immunological parameters including antigen-specific leucocyte proliferation, antigen-specific nitric oxide production, and superoxide anion production exhibited significantly higher (p < 0.05) values on 10th and 20th day of post-vaccination compared with control. Among all the antigenic groups, FAH + A showed most significant responses compared with the other groups and produced maximum antibody on 30th day following vaccination. Among the humoral immune responses, lysozyme activity showed almost similar trend like cellular parameters. The result of the challenge study showed a higher level of survival in all the vaccinated groups, especially in FAH + A group. Our results suggest that vaccine FAH + A can effectively protect rohu against A. hydrophila infection and could offer an appropriate strategy to prevent this infection in rohu farms.  相似文献

Cholecystokinin (CCK) is a multi-functional brain–gut peptide in fish and mammals. To investigate the role of CCK in appetite regulation in fish, a 770-bp full-length cDNA sequence of CCK gene was obtained by RT-PCR and rapid amplification of cDNA ends methods in grass carp Ctenopharyngodon idellus. Homology analysis showed that the CCK cDNA sequence of grass carp had the highest similarity (90 %) to that of goldfish Carassius auratus and a higher similarity (>70 %) to those of other teleosts than to mammals. The PCR amplification using genomic DNA identified that the CCK gene of grass carp was comprised of three exons and two introns. Real-time quantitative PCR was used to detect CCK mRNA expression in adult tissues. High levels of gene expression were found in the hypothalamus and pituitary; moderate levels in the intestine, muscle and white adipose tissue; and low levels in other tissues. During early development (i.e., fertilized eggs to 35-day post-hatching larvae) the levels of CCK mRNA expression were higher during embryonic developmental stages than during post-hatch larval stages. Fasting decreased CCK mRNA expression levels in the brain and intestine, whereas refeeding resulted in an increase of expression. The results suggest that CCK mRNA expression has obvious tissue specificity and may have a role in feed intake regulation in grass carp.  相似文献

To study the substitution of fish oil by vegetable oils in fish diets, juveniles Senegalese sole (Solea senegalensis) were fed diets (56 % crude protein, 12 % crude lipid) containing either linseed (100LO) or soybean (100SO) oils in comparison with a 100 % fish oil-based diet (100FO) for 90 days. Samples of muscle, liver, and intestine were collected for biochemical analysis and for glucocorticoid receptor-related genes, including GR1 and GR2, and the associated heat shock proteins HSP70, HSP90AA, and HSP90AB. Besides, basal levels of plasma cortisol were also determined. After the feeding period, a stress test, consisting on 5 min of net chasing, was applied to a selected population of each dietary group. Total replacement of fish oil by vegetable oils did not induced changes in fish growth and performance, but affected fatty acid profile of muscle, liver, and intestine, reflecting those tissues the characteristic fatty acids of each type of dietary oil. A tendency to conserve the ARA/EPA ratio could be observed in the different tissues, despite of the level of these fatty acids in diet. Chasing stress induced an increase of muscle GR1 and a reduction in intestinal GR2 relative expressions at any of the experimental diets assayed. In liver, chasing stress induced an increase in both GR1 and GR2 gene expression in fish fed fish oil diets. Similarly, chasing stress induced an increase of muscle HSP70 and decrease of HSP90AB in liver at any of the experimental diet assayed. Besides, vegetable oils decreased the expression of HSP70 in intestine, being the relative expression of liver HSP90AA increased by the inclusion of linseed oil in the diet, at any of the experimental conditions assayed.  相似文献

Diet is the primary source of iron (Fe) for freshwater fish, and the absorption of Fe is believed to occur via the Nramp family of divalent metal transporters (also called DMT1). Presently, the homeostatic regulation of dietary Fe absorption in fish is poorly understood. This study examined the gastrointestinal mRNA expression of two Nramp isoforms, Nramp-β and Nramp-γ, in the freshwater rainbow trout (Oncorhynchus mykiss), following exposure to elevated dietary Fe [1,256 mg Fe/kg food vs. 136 mg Fe/kg food (control)] for 14 days. The physiological performance, plasma Fe status and tissue-specific accumulation of Fe were also evaluated. In general, the mRNA expression level of Nramp was higher in the intestine relative to the stomach. Interestingly, fish fed on a high-Fe diet exhibited a significant induction in Nramp expression after 7 days, followed by a decrease to the level observed in control fish on day 14. The increase in Nramp expression correlated with the elevated gastrointestinal and plasma Fe concentrations. However, the hepatic Fe concentration remained unchanged during the entire exposure period, indicating strong homeostatic regulation of hepatic Fe level in fish. Fish appeared to handle increased systemic Fe level by elevating the plasma transferrin level, thereby enhancing the Fe-binding capacity in the plasma. Overall, our study provides new interesting insights into the homeostatic regulation of dietary Fe uptake and handling in freshwater fish.  相似文献

Sesamin (S) is a known lipid modulator and has been shown to increase the conversion of α-linolenic acid (ALA) to docosahexaenoic acid in rainbow trout (Oncorhynchus mykiss) fed vegetable oil mixtures including linseed oil. In this study, we evaluated the effects of S supplementation in linseed oil-based diets, content of α- and γ-tocopherols, fatty acid (FA) composition, as well as the gene expression of lipid-related genes. Fish with an average weight of 36.5 g were fed different combinations of commercial linseed oil (LO), purified linseed oil triacylglycerols (TAG) with polar fraction removed and a mixed linseed-sunflower oil (6:4 v/v) (MO). S was added at 0.58 g 100?1g feed and fed to the fish for a period of 58 days. Expression of PPARα was downregulated in white muscle of fish fed S containing diets (P < 0.05). The expression of PPARβ1A was not affected by S supplementation except where TAG oil was used. The expression of PPARβ1A declined significantly in TAG + S fed group (P < 0.05), which indicates that some minor compounds in linseed oil might suppress the effect of S on the expression of PPARβ1A. The expression of PPARγ(long) declined in LO + S and MO + S fed group (P < 0.05). The β-oxidation-related genes CPT1 and ACO were upregulated by vegetable oils compared to fish oil. S decreased percentage of ALA in white muscle of fish fed LO + S (P < 0.05). The increased desaturation index and the decreased ALA levels suggest that S may increase the biosynthesis of highly unsaturated FA in rainbow trout.  相似文献

Interleukin-10 (IL-10) is an anti-inflammatory cytokine and negatively regulates cell-mediated immunity (CMI) induction by inhibiting cytokine production in type 1 T helper cells. IL-10 genes have been isolated from several fish, and inflammatory cytokine inhibition by IL-10 has been well examined. However, a CMI regulator of IL-10 in fish has not yet been identified. In this study, we cloned the IL-10 gene in amberjack Seriola dumerili and analyzed its function using its recombinant protein (rIL-10). In an in vitro culture experiment, gene expression of inflammatory cytokines was suppressed in leukocytes incubated with rIL-10 compared with cells that only received Nocardia seriolae stimulation. This result suggests amberjack IL-10 has conserved function as an inflammatory cytokine inhibitor. Bactericidal activity of amberjack cells against intracellular pathogen stimulation was decreased in a rIL-10 dose-dependent manner. Furthermore, a significant reduction in the T-bet/GATA-3 ratio was observed in N. seriolae living cell (LC)?+?rIL-10-injected fish. Taken together, these results suggest amberjack rIL-10 suppresses CMI induction both in vitro and in vivo. In addition, the number of IgM+ cells among spleen leukocytes in N. seriolae?+?rIL-10-injected fish was higher than in only N. seriolae LC, suggesting that Th2-dominant immunity was induced by adding rIL-10.  相似文献

The immunomodulatory effect of garlic–mineral oil as a modified adjuvant was investigated in rohu, Labeo rohita along with formalin-killed Aeromonas hydrophila. The garlic extract was not having any proteolytic effect on the tested antigen as marked in SDS-PAGE. The efficacy of graded levels of modified adjuvant formulation was assessed by measuring serum hemolysin and agglutination titers as well as level of protection against A. hydrophila challenge in the immunized fish at days 10 and 30 post-vaccination (dpv). The higher hemolysin and bacterial agglutinin titers along with significantly lower mortality against A. hydrophila challenge in modified adjuvant-based vaccinated fish, particularly at low level of garlic incorporation, indicated the pronounced immunoadjuvant role of this modified adjuvant formulation in rohu.  相似文献

Aquaculture practices bring several stressful events to fish. Stressors not only activate the hypothalamus–pituitary–interrenal-axis, but also evoke cellular stress responses. Up-regulation of heat shock proteins (HSPs) is among the best studied mechanisms of the cellular stress response. An extract of the prickly pear cactus (Opuntia ficus indica), Pro-Tex, a soluble variant of TEX-OE®, may induce expression of HSPs and reduce negative effects of cellular stress. Pro-Tex therefore is used to ameliorate conditions during stressful aquaculture-related practices. We tested Pro-Tex in zebrafish (Danio rerio), common carp (Cyprinus carpio L.) and yellowtail kingfish (Seriola lalandi) exposed to aquaculture-relevant stressors (thermal stress, net confinement, transport) and assessed its effects on stress physiology. Heat shock produced a mild increase in hsp70 mRNA expression in 5-day-old zebrafish larvae. Pro-Tex increased basal hsp70 mRNA expression, but decreased heat-shock-induced expression of hsp70 mRNA. In carp, Pro-Tex increased plasma cortisol and glucose levels, while it did not affect the mild stress response (increased plasma cortisol and glucose) to net confinement. In gills, and proximal and distal intestine, stress increased hsp70 mRNA expression; in the distal intestine, an additive enhancement of hsp70 mRNA expression by Pro-Tex was seen under stress. In yellowtail kingfish, Pro-Tex reduced the negative physiological effects of transport more efficiently than when fish were sedated with AQUI-S®. Overall, our data indicate that Pro-Tex has protective effects under high levels of stress only. As Pro-Tex has potential for use in aquaculture, its functioning and impact on health and welfare of fish should be further studied.  相似文献

An experiment was carried out in red porgy, Pagrus pagrus (Teleostei, Sparidae), to assess the effects of a 14-day fasting period, followed by refeeding to apparent satiation, on the contents of digestive enzymes (total proteases, and particularly pepsin, trypsin, chymotrypsin, carboxypeptidases A and B; amylase and lipase). Two fish groups were considered: one (indicated as fasted/refed group) was fasted for 14 days and then refed during further 7 and 15 days, and the other was fed throughout the study and was taken as a control group. The measured enzymatic values showed that fasting resulted in a generalized, not significant decrease, of the activity of digestive enzymes. Refeeding caused a significant increase for most of the assayed enzymes: total proteases both in the middle and distal intestine, pepsin in the stomach, trypsin in the middle intestine, and amylase and lipase in the proximal intestine. Nevertheless, the detection in the fasted/refed fish of enzymatic values still lower than those measured in the control fish suggested that fish experiencing short-term fasting were partially impaired in their digestive capacity.  相似文献

Groupers bred for mariculture are of high economic value in Taiwan. Unfortunately, these fish are highly susceptible to bacterial infection, and the use of antibiotics to control disease has had the unintended consequence of generating antibiotic-resistant bacteria. Herein, we examined whether it is possible to enhance grouper immunity through application of Bacillus coagulans, a probiotic bacterial species. In order to study its effects on immunity, B. coagulans was isolated from lactobacillus mixtures and administered live to grouper (Epinephelus coioides) and zebrafish (Danio rerio). Fish were fed for 30 days on a diet consisting of B. coagulans (104, 106, 108, or 1010 colony-forming units (cfu) of B. coagulans in 50 ml of media) mixed with 50 g of eel powder. After 30 days, disease resistance against Vibrio vulnificus (204) infection and expression of immune-related genes were determined. We found that oral administration of B. coagulans significantly enhanced expression of several immune-related genes, including myeloid differentiation factor (MyD)88, interleukin (IL)-1β, tumor necrosis factor (TNF)-2, and IL-1β in grouper, and Toll-like receptor (TLR) 4, TNF-α, TRAM 1, and nuclear factor (NF)-κB in zebrafish. Treatment with B. coagulans significantly reduced mortality at 24, 48, 72, and 96 h after challenge with V. vulnificus (204). In vitro studies revealed that B. coagulans possesses bactericidal activity against Enterococcus faecalis (10066), Staphylococcus aureus subsp. (10780), Streptococcus agalactiae (S47, S48, 819), Escherichia coli (10675), Klebsiella oxytoca (13985), Pseudomonas aeruginosa (19660), and V. vulnificus (204, 12905, YJ016). On average, 107 colony-forming units of B. coagulans were required for bactericidal activity. In conclusion, our results indicate that dietary intake of B. coagulans enhances expression of immune-related genes and protects grouper and zebrafish against V. vulnificus (204) infection.  相似文献

We studied accumulation and biochemical effects of microcystin-LR (MCLR) in Odontesthes hatcheri after dietary administration of the cyanobacteria Microcystis aeruginosa (1.3 μg MCLR/g body mass, incorporated in standard fish food). After 12 h, MCLR content in liver did not differ between fish fed with crushed or intact cells, demonstrating O. hatcheri’s capacity to digest cyanobacteria and absorb MCLR. In the second experiment, fish received toxic cells, non-toxic cells, or control food; MCLR accumulation was monitored for 48 h. Protein phosphatase 1 (PP1), catalase (CAT), glutathione-S-transferase (GST) activities, and lipid peroxidation (as MDA) were measured in liver and intestine. Methanol-extractable MCLR was determined by PP1 inhibition assay (PPIA); extractable and protein-bound MCLR were measured by Lemieux oxidation-gas chromatography/mass spectrometry (GC/MS). MCLR accumulated rapidly up to 22.9 and 9.4 μg MCLR/g in intestine and liver, respectively, followed by a decreasing tendency. Protein-bound MCLR represented 66 to ca. 100 % of total MCLR in both tissues. PP1 activity remained unchanged in intestine but was increased in liver of MCLR treated fish.CAT and GST activities and MDA content were significantly increased by MCLR only in liver. We conclude that O. hatcheri is able to digest cyanobacteria, accumulating MCLR mostly bound to proteins. Our data suggest that this freshwater fish can be adversely affected by cyanobacterial blooms. However, the rapid decrease of the detectable MCLR in both tissues could imply that sublethal toxin accumulation is rapidly reversed.  相似文献

Ontogenetic development of the digestive tract and associated organs in long snout seahorse Hippocampus guttulatus juveniles was morphologically and histologically examined from the time of release from the male’s pouch until 72 h after the first meal. When released from the male’s pouch, juvenile seahorses are small adult replicates. This means that unlike other teleost fish larvae, the first developmental phase has already taken place, and juveniles are morphologically prepared and able to feed on live prey immediately following parturition. At this stage, the buccopharynx, oesophagus, and intestine already appear to be fully developed. The intestine is divided into the midgut and hindgut by an intestinal valve, and intestinal villi are visible in the midgut. When fed with DHA-Selco® enriched Artemia, H. guttulatus juveniles developed a severe condition of overinflation of the gas bladder. The continuous overinflation of the gas bladder forced air into the gut (48 h after the first meal), resulting in overinflation of both the gut and the gas bladder (72 h after the first meal), and death occurred within 120 h after the first meal. When fed natural copepods, H. guttulatus juveniles continued a normal feeding activity with no signs of intestinal disorders, and the gas bladder and intestine maintained their normal shape. This is the first study to positively associate gas bladder overinflation of juvenile seahorses with nutritionally unbalanced diets, and not to gas supersaturation alone. It is therefore necessary to develop more adequate feed and/or enrichment products to improve the survival of juvenile seahorses in captivity.  相似文献

The present study examined the influence of extract obtained from the interior of cells of yeast, Saccharomyces cerevisiae (NuPro®), on the growth, proximate composition, nonspecific cellular and humoral defense mechanisms, and protection against furunculosis in pikeperch (Sander lucioperca). It was noted that eight weeks of supplementation with 40 g of NuPro® kg?1 feed (containing approximately 0.22 % exogenic nucleotides) enhanced pikeperch innate immunity. Increased serum lysozyme activity and total Ig levels (P < 0.05) were noted in the NuPro®-fed group, which indicated the activation of nonspecific humoral defense mechanisms. Brewer’s yeast extract also stimulated nonspecific cellular defense mechanisms as indicated by the increased metabolic activity of blood phagocytes and pronephric macrophages, and in the increased intracellular killing activity of phagocytes and proliferative responses of T and B lymphocytes (P < 0.05). Additionally, reduced mortality (15 %) after the in vivo challenge with pathogenic Aeromonas salmonicida suggested that NuPro® activated nonspecific protection against furunculosis in pikeperch. The experimental feed did not have a significant impact on the growth or proximate composition of the fish (P > 0.05).  相似文献

This study was undertaken to assess the effects of feeding European seabass (Dicentrarchus labrax) canola oil-added diets on growth, health status and liver and intestine histomorphology. Seabass (56.18 ± 0.16 g initial body weight) were fed one of three fish meal-based diets with ~48 % crude protein and ~16.0 % lipid, combining fish oil (FO) and canola oil (CO) for 12 weeks. The diets contained: zero (control, CTRL), 45 (CO50) or 63 (CO70) g CO kg?1 assigned in triplicates to three dietary groups. The results indicated that neither dietary oil type (FO or CO) nor CO level adversely affected (P > 0.05) the growth, feed utilization or major blood constituents’ composition as an indicator of the overall health status of fish. Despite the CO diets influence on head kidney macrophage activity being unappreciable (P > 0.05), there was a reducing trend with an increase in CO level incorporation. The CO70 diet induced a minor fat infiltration in hepatocytes and leucocytes infiltration, hyperplasia of the basal nuclei and supranuclear vacuolization of the enterocytes of the distal intestine. The present observations suggest that it is possible to incorporate up to 63 g CO Kg?1 in the feed for European seabass juveniles without major negative effects on growth, health status or liver and intestinal histomorphology.  相似文献

Probiotic supplementation in fish aquaculture has significantly increased in the last decade due to its beneficial effect on fish performance. Probiotic use at early stages of fish development may contribute to better face metamorphosis and weaning stress. In the present work, we studied the influence of Shewanella putrefaciens Pdp11 supplementation on growth, body composition and gut microbiota in Senegalese sole (Solea senegalensis) during larval and weaning development. S. putrefaciens Pdp11 was incorporated using Artemia as live vector (2.5 × 107 cfu mL?1) and supplied to sole specimens in a co-feeding regime (10–86 DAH) by triplicate. Probiotic addition promoted early metamorphosis and a significantly higher growth in length at 24 DAH larvae. S. putrefaciens Pdp11 also modulated gut microbiota and significantly increased protein content and DHA/EPA ratios in sole fry (90 DAH). This nutritional enhancement is considered especially important after weaning, where significantly higher growth in length and weight was observed in probiotic fish. Moreover, a less heterogeneous fish size in length was detected since metamorphosis till the end of weaning, being of interest for sole aquaculture production. After weaning, fish showed significantly higher growth (length and weight) and less variable lengths in fish when supplemented with probiotics. Both the enhancement of nutritional condition and the decrease in size variability associated with probiotic addition are highly interesting for sole aquaculture production.  相似文献
