Equine influenza virus (EIV) surveillance is important in the management of equine influenza. It provides data on circulating and newly emerging strains for vaccine strain selection. To this end, antigenic characterisation by haemaggluttination inhibition (HI) assay and phylogenetic analysis was carried out on 28 EIV strains isolated in North America and Europe during 2006 and 2007. In the UK, 20 viruses were isolated from 28 nasopharyngeal swabs that tested positive by enzyme-linked immunosorbent assay. All except two of the UK viruses were characterised as members of the Florida sublineage with similarity to A/eq/Newmarket/5/03 (clade 2). One isolate, A/eq/Cheshire/1/06, was characterised as an American lineage strain similar to viruses isolated up to 10 years earlier. A second isolate, A/eq/Lincolnshire/1/07 was characterised as a member of the Florida sublineage (clade 1) with similarity to A/eq/Wisconsin/03. Furthermore, A/eq/Lincolnshire/1/06 was a member of the Florida sublineage (clade 2) by haemagglutinin (HA) gene sequence, but appeared to be a member of the Eurasian lineage by the non-structural gene (NS) sequence suggesting that reassortment had occurred. A/eq/Switzerland/P112/07 was characterised as a member of the Eurasian lineage, the first time since 2005 that isolation of a virus from this lineage has been reported. Seven viruses from North America were classified as members of the Florida sublineage (clade 1), similar to A/eq/Wisconsin/03. In conclusion, a variety of antigenically distinct EIVs continue to circulate worldwide. Florida sublineage clade 1 viruses appear to predominate in North America, clade 2 viruses in Europe.  相似文献

An inactivated equine influenza virus (EIV) vaccine and a live equine herpesvirus type 1 (EHV-1) vaccine are usually administered concurrently to Thoroughbred racehorses in Japan. The objective of this study was to evaluate whether concurrent administration of an inactivated EIV vaccine and a live EHV-1 vaccine in Thoroughbred racehorses influences the antibody response against EIV. We compared the antibody response against EIV in horses administered both vaccines on the same day (Group A; n = 27) and the response in horses administered an inactivated EIV vaccine first and then a live EHV-1 vaccine 1–2 weeks later (Group B; n = 20). In both groups, geometric mean hemagglutination inhibition (HI) titers against A/equine/Ibaraki/1/2007 and A/equine/Yokohama/aq13/2010 increased significantly after EIV vaccination. However, the percentage of horses that showed a twofold increase or greater in HI titers against A/equine/Yokohama/aq13/2010 was significantly higher in Group B (75%) than in Group A (37%; P = .02). These results suggest that the concurrent use of an inactivated EIV vaccine and a live EHV-1 vaccine reduced the immune response against EIV to some extent, and it would be better to use these vaccines consecutively, especially for naïve horses or horses whose vaccination history is incomplete.  相似文献

A 13‐year‐old Quarter Horse mare was presented to the Ontario Veterinary College Teaching Hospital with chronic progressive respiratory noise and exercise intolerance. Severe respiratory stridor associated with a laryngeal mass was seen on upper airway endoscopy. Ultrasonography revealed an intra‐cartilagenous mass and allowed for a guided biopsy. Histological analysis of the mass characterised a neuroendocrine tumour and the mare was subjected to euthanasia due to the poor prognosis. Ultrasound examination was paramount in making the diagnosis and facilitating the biopsy. To our knowledge, neuroendocrine tumours have not previously been reported in the equine larynx.  相似文献

Reasons for performing study: During the 2007 Australian equine influenza (EI) outbreak, an accelerated primary course 14 day intervaccination schedule was proposed, but not widely implemented. Expert opinion was divided as to the efficacy of such a schedule given the lack of published data. This study determined the level and duration of humoral immunity following administration of a recombinant canarypox‐vectored vaccine (ALVAC‐EIV) with a primary intervaccination interval of 14 days and booster at 105 days. Objectives: To examine whether protective levels of immunity of adequate duration were achieved following a primary course reduced from a minimum interval of 28 to 14 days. Antibody responses to 2 H3N8 American lineage virus strains (including A/equine/Sydney/6085/2007) were assessed and compared to previous challenge studies using ALVAC‐EIV at conventional intervaccination intervals. Methods: Fourteen Thoroughbred horses and 2 ponies from a rural racehorse training property in Victoria, Australia, were vaccinated with ALVAC‐EIV on Days 0, 14 and 105. Serial blood samples were collected over the next 32 weeks and tested with haemagglutination inhibition and single radial haemolysis (SRH) in full assays to evaluate the serological response. Results: All horses and ponies responded to the accelerated ALVAC‐EIV vaccination schedule. Mean SRH antibodies remained above those consistent with clinical protection for the duration of the study period. All vaccinates demonstrated high SRH antibodies 14 days following V2, thereby achieving 100% herd immunity to homologous viral challenge. Conclusions: An accelerated vaccination schedule conferred long‐lasting protective antibody levels despite a >50% reduction in the recommended V1–V2 interval. Potential relevance: High levels of rapidly acquired herd immunity are critical in containing an outbreak of such a highly contagious pathogen as EIV. In a strategic vaccination programme, it is important that horses remain protected for sufficient time to allow control programmes to succeed. An accelerated 14 day primary course intervaccination interval and booster at 105 days achieves both of these objectives.  相似文献

为建立马流感血清学ELISA诊断方法,本研究以马流感病毒中国分离株A/马/新疆/07(H3N8)通过SPF鸡胚培养和增殖,收取含病毒尿囊液经蔗糖密度梯度离心纯化后作为ELISA包被抗原,首次在我国建立了检测H3N8亚型马流感抗体的间接ELISA诊断方法。试验的最佳反应条件为:最佳抗原稀释度7μg/mL,封闭液5%脱脂乳,血清稀释度1∶100,二抗稀释度1∶10000,稀释液PBS(pH7.4),血清反应时间1.5h,二抗反应时间1h。通过本方法对555份临床样品进行检测并与血凝抑制(HI)试验检测结果比较,证明本方法特异、敏感,具有良好的稳定性和可重复性,适于马流感的流行病学调查和监测工作。  相似文献

Equine respiratory viral infections cause significant worldwide disease and economic loss. Common causes include equine influenza virus (EIV) and equine herpesviruses-1 and -4 (EHV-1 and -4), and risk of exposure to these agents may be highest in young horses commingling at sales and competitive events. A surveillance study was conducted at two horse shows and two Thoroughbred sales to determine whether horses shed EHV-1, EHV-4, or EIV on arrival, or 2-4 days later, and whether shedding was associated with identifiable risk factors. Real-time polymerase chain reaction assays were used to detect EHV-1, EHV-4, and EIV nucleic acid in nasal swabs obtained from 369 horses at the four events. In response to evidence of clinical disease, 82 additional horses were sampled at two farms providing horses for one of the sales. On arrival at the events, shedding of EHV-1 was detected in 3.3%, EHV-4 in 1.1%, and EIV in 0.8% of horses. EHV-1 was detected at low levels, and EHV-1 and EHV-4 detection was not associated with clinical disease. EIV was detected only in horses at a Thoroughbred sale, in association with an outbreak of respiratory disease traced back to regional farms. On arrival at events, horses younger than 2 years had a significantly greater risk of shedding EHV-1 compared with older horses; no other significant risk factors associated with viral shedding were identified. Thus, there is a risk of exposure to EIV, EHV-1, and EHV-4 at equine events, and horses and events should be managed to mitigate this risk.  相似文献

Background: Myocarditis is thought to occur secondary to equine influenza virus (EIV) infections in horses, but there is a lack of published evidence. Hypothesis/Objectives: We proposed that EIV challenge infection in ponies would cause myocardial damage, detectable by increases in plasma cardiac troponin I (cTnI) concentrations. Animals: Twenty‐nine influenza‐naïve yearling ponies: 23 were part of an influenza vaccine study (11 unvaccinated and 12 vaccinated), and were challenged with 108 EID50 EIV A/eq/Kentucky/91 6 months after vaccination. Six age‐matched healthy and unvaccinated ponies concurrently housed in a separate facility not exposed to influenza served as controls. Methods: Heparinized blood was collected before and over 28 days after infection and cTnI determined. Repeated measures analysis of variance, chi‐square, or clustered regression analyses were used to identify relationships between each group and cTnI. Results: All EIV‐infected ponies developed clinical signs and viral shedding, with the unvaccinated group displaying severe signs. One vaccinated pony and 2 unvaccinated ponies had cTnI greater than the reference range at 1 time point. At all other times, cTnI was <0.05 ng/mL. All control ponies had normal cTnI. There were no significant associations between cTnI and either clinical signs or experimental groups. When separated into abnormal versus normal cTnI, there were no significant differences among groups. Conclusions and Clinical Importance: This study demonstrated no evidence of severe myocardial necrosis secondary to EIV challenge with 108 EID50 EIV A/eq/Kentucky/91 in these sedentary ponies, but transient increases in cTnI suggest that mild myocardial damage may occur.  相似文献

A 5-year-old Mangalarga mare was referred to the Veterinary Teaching Hospital of the Veterinary Medical School in Botucatu with a complaint of a persistent 2-month ovarian follicle which had grown progressively to 20 cm in diameter. Ultrasonography showed a circular, cystic structure in the right ovary, whose wall was thickened and interior was filled with anechoic content. Ovarian neoplasia was suspected, and a unilateral ovariectomy was performed through the paralumbar fossa, using general anesthesia through inhalation. Histopathological evaluation concluded that the ovarian enlargement was caused by a fibrothecoma. The present report describes features of fibrothecoma, which is extremely rare in the equine species.  相似文献

The prevalence and epidemiology of important viral (equine influenza virus [EIV], equine herpesvirus type 1 [EHV-1] and EHV-4) and bacterial (Streptococcus equi subspecies equi) respiratory pathogens shed by horses presented to equine veterinarians with upper respiratory tract signs and/or acute febrile neurological disease were studied. Veterinarians from throughout the USA were enrolled in a surveillance programme and were asked to collect blood and nasal secretions from equine cases with acute infectious upper respiratory tract disease and/or acute onset of neurological disease. A questionnaire was used to collect information pertaining to each case and its clinical signs. Samples were tested by real-time PCR for the presence of EHV-1, EHV-4, EIV and S equi subspecies equi. A total of 761 horses, mules and donkeys were enrolled in the surveillance programme over a 24-month study period. In total, 201 (26.4 per cent) index cases tested PCR-positive for one or more of the four pathogens. The highest detection rate was for EHV-4 (82 cases), followed by EIV (60 cases), S equi subspecies equi (49 cases) and EHV-1 (23 cases). There were 15 horses with double infections and one horse with a triple infection. The detection rate by PCR for the different pathogens varied with season and with the age, breed, sex and use of the animal.  相似文献

During 2007, large outbreaks of equine influenza (EI) caused by Florida sublineage Clade 1 viruses affected horse populations in Japan and Australia. The likely protection that would be provided by two modern vaccines commercially available in the European Union (an ISCOM-based and a canarypox-based vaccine) at the time of the outbreaks was determined. Vaccinated ponies were challenged with a representative outbreak isolate (A/eq/Sydney/2888-8/07) and levels of protection were compared. A group of ponies infected 18 months previously with a phylogenetically-related isolate from 2003 (A/eq/South Africa/4/03) was also challenged with the 2007 outbreak virus. After experimental infection with A/eq/Sydney/2888-8/07, unvaccinated control ponies all showed clinical signs of infection together with virus shedding. Protection achieved by both vaccination or long-term immunity induced by previous exposure to equine influenza virus (EIV) was characterised by minor signs of disease and reduced virus shedding when compared with unvaccinated control ponies. The three different methods of virus titration in embryonated hens’ eggs, EIV NP-ELISA and quantitative RT-PCR were used to monitor EIV shedding and results were compared. Though the majority of previously infected ponies had low antibody levels at the time of challenge, they demonstrated good clinical protection and limited virus shedding. In summary, we demonstrate that vaccination with current EIV vaccines would partially protect against infection with A/eq/Sydney/2888-8/07-like strains and would help to limit the spread of disease in our vaccinated horse population.  相似文献

This report describes an 11‐year‐old Percheron mare that presented with signs of colic and was ultimately diagnosed with bacterial peritonitis of unknown origin. Bacterial culture of a peritoneal fluid sample isolated 2 Clostridial species, one of which was strongly suspected to be Clostridium haemolyticum. The horse was markedly hypoalbuminaemic at presentation, leading to the development of low oncotic pressure and ventral oedema. The mare was administered a low molecular weight/low molar substitution hydroxyethyl starch solution in conjunction with other therapies that resulted in marked improvement of clinical signs. The purpose of this report is to describe the clinical findings associated with equine peritonitis associated with C. haemolyticum, a rarely identified pathogen in the horse. Secondly, this report serves to describe the beneficial effects of tetrastarch administration in a clinical case with severe hypoalbuminaemia and ventral oedema.  相似文献

The hemagglutination inhibition test is used by many diagnostic and surveillance laboratories for detection of antibodies to influenza viruses. It is well known that the hemagglutination inhibition test is affected by nonspecific inhibitors present in equine serum. Several serum treatments are in use to remove these inhibitors, including treatment with kaolin. Discrepant results were observed in the authors' laboratories when using kaolin treatment before testing equine sera for antibodies against equine influenza virus (EIV) subtype-1 (H7N7). It is demonstrated here that kaolin treatment leads to false positive results when testing for antibodies against EIV subtype-1, as compared to other standard serum treatments (trypsin-periodate, receptor-destroying enzyme). Against EIV subtype-2 (H3N8), however, false positive results were not evident. Trypsinperiodate and receptor-destroying enzyme (RDE) treatments appear to be superior to kaolin for removal of nonspecific inhibitors from equine serum and should be used for serological diagnosis and surveillance of equine influenza virus.  相似文献

Behavior problems in horses are often thought to be due to the horse’s bad attitude, which commonly results in them being punished. However, when these cases are carefully worked up, there may be an underlying physical problem, and pain may play an important role. A 5-year-old Dutch Warmblood mare was presented for rearing and flipping over backward. The mare had been repeatedly examined by an equine veterinarian and a nonveterinarian equine dentist who found no abnormalities. Several different bits were tried, and side reins were used to keep her head down. The mare was whipped every time she reared. None of these measures helped. On presentation to our equine teaching hospital, no abnormalities were detected on routine physical examination. The behavior was observed when the mare was lunged; as soon as a small amount of tension was put on the lunge rein, the mare shook her head horizontally for approximately 1-2 seconds, reared, and flipped over onto her back. Neurological examination revealed that the mare was slow to resume a normal stance when her hind legs were placed in an abnormal position. Dental examination and endoscopy of the upper airways proved unremarkable. Radiography of the atlanto-occipital joint area revealed fractures of the first and second cervical vertebrae and fracture fragments, which are both suggestive for the presence of cervical instability and the possible presence of spinal cord compression. These suggestions were further supported by identification of delayed postural reflexes during neurological examination. The mare was deemed unfit for dressage and was successfully kept for breeding. Horses are frequently punished for “bad behavior.” These behaviors are frequently due to underlying physical problems and not a “nasty” horse. This case highlights the importance of careful history taking and a thorough veterinary examination with the use of further diagnostics (if necessary) when dealing with equine behavior problems.  相似文献

This report describes the successful treatment of immune‐mediated thrombocytopenia (IMTP) with azathioprine in a pregnant mare that had been refractory to therapy with dexamethasone. The interesting features of this case included the presence of dermatitis, a clinical presentation not previously associated with IMTP in the equine literature and the maintenance of pregnancy and delivery of a healthy normal foal despite immunosuppressive therapy with azathioprine during critical stages of early gestation.  相似文献

The objective of this research was to determine the scope of equine fescue toxicosis, current procedures to control the disease, and to gain veterinary insight to possible new therapies. A survey was developed with help form experts in the area of fescue toxicosis. From a list of members of the American Veterinary Medical Association (AVMA), a sample was obtained by selecting large animal/equine veterinarians from states where fescue toxicosis is common. These states included: Alabama, Arkansas, Georgia, Illinois, Indiana, Kentucky, Missouri, Mississippi, North Carolina, Ohio, South Carolina, Tennessee, Virginia, and West Virginia. One-fifteenth of the veterinarians (515 total) were mailed questionnaires; 207 responded. Veterinarians were grouped into regions based on latitude. Region 1 included veterinarians practicing north of 39.0° latitude; Region 2 included veterinarians practicing between 36.5° and 39.0° latitude; and Region 3 included veterinarians practicing south of 36.5° latitude.A majority of horse pasture in the area of the survey is comprised of at least 50% fescue, with approximately 70% of the pasture being at least partially comprised, of fescue.Almost 50% of the brood mares in the area of the survey are exposed to endophyte-infected fescue. Many gravid brood mares in the area of the survey were suffering from acute and subacute symptoms, with over 43% warranting some kind of management technique to prevent toxicity from occurring. A large majority of veterinarians in the area of the survey feel that fescue toxicosis is, to some level, a problem. Current management techniques can be tedious and expensive. New management techniques and/or therapies are needed to handle the problems associated with equine fescue toxicosis. A large majority of veterinarians in the area of the survey feel an effective drug therapy would be valuable in treating equine fescue toxicosis. If a drug therapy were developed, a daily feed additive or an oral gel would be the most popular method of dosing with veterinarians.As a result of trends noticed in the rates of equine fescue toxicosis in different areas, statistical analyses were conducted on data generated from this survey along regions that seemed to have different infection rates. There was no difference (P>0.05) in levels of fescue in pastures, mare exposure to the endophyte, and mare expression of acute or subacute symptoms between regions 2 and 3. There was a difference (P<0.05) between region 1 and regions 2 and 3 for all of the factors tested except for number of mares seen by veterinarians.  相似文献

Summary

The prevalence of antibodies to various viruses was investigated in a series of serum samples collected from horses in the Netherlands between 1963 and 1966 and from 1972 onwards. Neutralizing antibodies to equine rhinopneumonitis virus, equine arteritis virus and to equine rhinovirus types 1 and 2 were detected in respectively 76%, 14%, 66% and 59% of the equine serum samples tested.

The observed incidence of serum samples positive to equine adenovirus in the complement fixation test was 39%. Precipitating antibodies to equine infectious anaemia virus were detected only in serum samples from two horses imported from abroad. Haemagglutination inhibiting antibodies to Myxovirus influenzae A / equi‐1, M. Influenzae A / equi‐2, and Reovirus types 1, 2, and 3 were present in respectively 82%, 50%, 10%, 33% and 3.6% of the serum samples tested.

The most frequently observed incidence of antibodies to the various equine respiratory viruses occurred in the groups of horses having repeatedly contact with other horses.  相似文献

Equine influenza and equine rhinopneumonitis are among the Office International des Epizooties or the World Organisation for Animal Health notifiable, contagious respiratory diseases. Although vaccination of horses in Israel against equine influenza virus (EIV) and against equine herpesvirus (EHV) is routinely performed, information regarding the occurrence and the epidemiology of the diseases is lacking. We hereby attempt to determine seroprevalence and rate of infection for EHV-1 and 4 and for EIV in horses distributed throughout Israel and describe demographic and environmental risk factors associated with seroprevalence. Despite the fact that last reported isolation of EIV in Israel occurred in 2007, we found a 26.4% (29/110) (95% confidence interval [CI]: 18.18–34.62) seroprevalence for H3N8, a 16.4% (18/110) (95% CI: 9.49–23.31) for H7N7, and a 6.4% (7/110) (95% CI: 1.83–10.97) rate of seroconversion for H3N8, suggesting current and active circulation of EIV in horses in Israel. Age, housing management type, and type of farm activity were significantly associated with seroprevalence, with activities allowing exposure to new horses positively associated with seroprevalence to EIV and an only pasture housing management negatively associated with seroprevalence. No association was detected between other demographic variables (gender, breed, and color) and environmental factors (climatic regions). Seroprevalence to EHV-1 and 4 were very low (<1%) and very high (>99%), respectively, raising questions regarding the appropriate vaccination guidelines. Our findings of the occurrence of EIV in horses in Israel imply an underdiagnosis of this virus in this country and warrant further investigation as to the strains that circulate in this region and their accordance with the current vaccine strains.  相似文献
