本研究分别用含有标记基因和编码小麦麦谷蛋HMW亚单位的基因的两种质粒,对小麦品种(Triticum aestivum)进行相互轰击而生产出了6个转基因小麦品系,然后研究了转基因的结合、遗传和表达。结果发现转基因的插人数在1-15个范围。在转基因位点由插入DNA隔开的分散基因组区域内发现了大量的质粒复制。然而有迹象表明,被导入的质粒复制物是按连环状排列的,并且显示为质粒重排列和平截形状。 相似文献

At present, transgenic plants are globally grown. Availability of a reliable regeneration system predominantly from a single transformed cell is the prerequisites for gene transfer, but regeneration is still a key problem (Wenzel, 2006). The application of genetic modification technology in plants is closely related to the efficiency of the regeneration protocol. Shoot formation is oilen enhanced by the combination of auxins and cytokinins. TDZ (Thidiazuron, N-phenyl-N'-1,2,3-thiodiazol-5-ylurea), a non-purine, has been shown to promote differentiation of organized centers of growth in cultured tissues at much lower concentrations, and shoot regeneration occurs with an efficiency comparable to or greater than that of other cytokinins (Fiola et al., 1990). By addition of TDZ, a series of plants which were difficult to culture in vitro or less sensitive to plants growth regulators obtained somatic embryos and regenerated plants, some of them have become the eminent transformation system for genetic engineering. TDZ has been reported to be very efficient in stimulating adventitious shoot production in several recalcitrant plants. TDZ is considered as a potential growth regulator for in vitro shoot regeneration and somatic embryogenesis of several crops. 稻瘟病是水稻三大病害之一,长期的生产实践证明,水稻抗瘟性品种的选育和利用是防治稻瘟病行之有效的措施。溶菌酶是一个广泛分布的酶家族,并且具有几丁质酶的活力,能够分解细菌或真菌细胞壁组分中多糖的糖苷键,从而抵御病原菌的侵染。本文以转溶菌酶基因水稻的10个株系当代及其后代为研究对象,对其农艺性状以及它们对稻瘟病的抗性水平变化进行研究,并利用PCR检测来研究外源基因在转基因株系各世代中的分离规律。结果表明:在R0、R1代群体中农艺性状、外源基因分离情况比较大,在R2代群体中只有个别株系分离较大。在R0代群体中单株之间的表现差异可能是由外源基因插入的位点和拷贝数不同而造成的,R1代表现出来的群体差异要比当代更大,在R1代中的差异可能是由于当代植株中的外源基因是杂合型,分离世代造成的,在R2代群体中由于外源基因趋于纯合,群体中株系相差小,同对照水平接近。与未转化株系比较,转基因植株进行了苗瘟、叶瘟和穗茎瘟的鉴定,在转基因的两个品系中溶菌酶基因的插入整合提高了转基因株系的抗稻瘟病的能力。 相似文献

A partial male-sterility system was found in a released guar, Cyamopsis tetragonoloba (L.) Taub. (2n = 2x = 14), breeding line, TX76-3285. Partially male-sterile plants produce some functional pollen, but with suitable genetic markers may be used to facilitate crossing in breeding programs. In addition, self fertilization allows partially male-sterile plants to be propagated sexually as homozygotes, once placed in breeding lines. The objective of the present study was to determine the inheritance of a partial male-sterility system in guar. Partial male-sterility appeared to be controlled by two genes, with one gene acting in a dominant epistatic manner and the second gene by incomplete dominance. The trait was expressed on every raceme beginning at about 4 to 6 weeks after planting in field-grown plants. The gene symbols, “pms 1” and “Pms 2” are proposed for the new trait. Segregation data indicated that alleles for pubescence segregated independently of alleles for partial male-sterility. Partial male-sterility in guar may be useful in the production of hybrid seed for breeding and genetic research. 相似文献

W6基因是由普通小麦地方品种小白麦cDNA文库中克隆获得的ERF类转录因子, 属于ERF IV家族。将W6基因构建在由组成型CaMV35S强启动子控制的双子叶转化载体pBI121上, 通过农杆菌介导法将其转入烟草新华1号。利用RT-PCR技术分析W6基因的过量表达对下游抗性基因表达水平的影响, 并测定高盐胁迫下转基因烟草的SOD酶活性和叶绿素含量。结果显示, 所检测的转基因烟草阳性株中W6基因均有不同程度的表达, 其耐盐性明显提高。W6基因的表达诱导并增强了含有GCC box的PR(pathogenesis-related)基因和含有DRE/CRT元件的COR/RD基因的表达。在高盐胁迫下, 转基因烟草的SOD酶活性和叶绿素含量明显高于对照。W6基因既能诱导抗病相关的PR基因表达又能诱导与提高非生物胁迫能力的COR/RD基因表达, 可能与生物胁迫和非生物胁迫相关, 证实转录因子W6参与了耐盐胁迫相关基因的调节, W6基因的过表达提高了转基因烟草的耐盐性。 相似文献

Resistance to early flowering is an important attribute of lettuce cultivars adapted to tropical conditions. Lettuce cultivars
may vary widely in the number of days necessary from sowing to inflorescence formation and flowering. In this paper, we report
on the polygenic inheritance of the number of days to flowering in two crosses among contrasting lettuce cultivars – Vitória
× Brasil-303 and Babá × Elisa. F1 and F2 seed were obtained for each cross, and used to estimate broad sense heritabilities. F3 families were obtained from randonly sampled F2 plants, and used to estimate narrow sense heritabilities based on parent-offspring regression. Trials were carried out in
plastic greenhouses in Campos dos Goytacazes, in the Northern part of the State of Rio de Janeiro, Brazil. Broad and narrow
sense heritabilities for days to first anthesis were 0.737 and 0.489 for the cross Vitória × Brasil-303, and 0.818 and 0.481
respectively, for the cross Babá × Elisa. In both crosses, both early and late-flowering transgressive segregants were obtained.
Genetic gains based on selection of late flowering transgressive segregants in the F2's were estimated to be 10.2 days in Vitória × Brasil-303 cross, and 8.7 days in the Babá × Elisa cross.

转基因植物商业化生产10年以来,尽管遇到各种各样的阻力,仍以前所未有的速度发展。长期积累的数据表明,在经济利益上,采用转基因植物的农民获得了显著的经济效益。由于采用转基因植物,农药特别是高毒农药使用量减少,减少了农田环境和水环境的化学毒素毒物的残留。由于转基因抗虫植物虫害减少,在粮食产品中有害微生物及其毒素毒物残留大大降低。这表明,在现有技术基础上,转基因植物商业化有利于农民生活质量的提高和环境的改良。 相似文献
