Silicon is required for normal growth and development in the chick when a low silicon diet is fed in a trace element controlled environment. Day-old deutectomized cockerels fed a purified amino acid diet showed significantly retarded growth and development within 2 to 3 weeks. Chicks fed the same diet plus a silicon supplement showed 50 percent higher growth and normal development. Silicon meets the criteria for an essential trace element. 相似文献

The identity of nicotinic receptor subtypes sufficient to elicit both the acute and chronic effects of nicotine dependence is unknown. We engineered mutant mice with a4 nicotinic subunits containing a single point mutation, Leu9' --> Ala9' in the pore-forming M2 domain, rendering a4* receptors hypersensitive to nicotine. Selective activation of a4* nicotinic acetylcholine receptors with low doses of agonist recapitulates nicotine effects thought to be important in dependence, including reinforcement in response to acute nicotine administration, as well as tolerance and sensitization elicited by chronic nicotine administration. These data indicate that activation of a4* receptors is sufficient for nicotine-induced reward, tolerance, and sensitization. 相似文献

Entry into mitosis in eukaryotes requires the activity of cyclin-dependent kinase 1 (Cdk1). Cdk1 is opposed by protein phosphatases in two ways: They inhibit activation of Cdk1 by dephosphorylating the protein kinases Wee1 and Myt1 and the protein phosphatase Cdc25 (key regulators of Cdk1), and they also antagonize Cdk1's own phosphorylation of downstream targets. A particular form of protein phosphatase 2A (PP2A) containing a B55δ subunit (PP2A- B55δ) is the major protein phosphatase that acts on model CDK substrates in Xenopus egg extracts and has antimitotic activity. The activity of PP2A-B55δ is high in interphase and low in mitosis, exactly opposite that of Cdk1. We report that inhibition of PP2A-B55δ results from a small protein, known as α-endosulfine (Ensa), that is phosphorylated in mitosis by the protein kinase Greatwall (Gwl). This converts Ensa into a potent and specific inhibitor of PP2A-B55δ. This pathway represents a previously unknown element in the control of mitosis. 相似文献

Soybean plants deprived of nickel accumulated toxic concentrations of urea (2.5 percent) in necrotic lesions on their leaflet tips. This occurred regardless of whether the plants were supplied with inorganic nitrogen or were dependent on nitrogen fixation. Nickel deprivation resulted in delayed nodulation and in a reduction of early growth. Addition of nickel (1 microgram per liter) to the nutrient media prevented urea accumulation, necrosis, and growth reductions. This evidence suggests that nickel is essential for soybeans and possibly for higher plants in general. 相似文献

beta-Arrestins, originally discovered in the context of heterotrimeric guanine nucleotide binding protein-coupled receptor (GPCR) desensitization, also function in internalization and signaling of these receptors. We identified c-Jun amino-terminal kinase 3 (JNK3) as a binding partner of beta-arrestin 2 using a yeast two-hybrid screen and by coimmunoprecipitation from mouse brain extracts or cotransfected COS-7 cells. The upstream JNK activators apoptosis signal-regulating kinase 1 (ASK1) and mitogen-activated protein kinase (MAPK) kinase 4 were also found in complex with beta-arrestin 2. Cellular transfection of beta-arrestin 2 caused cytosolic retention of JNK3 and enhanced JNK3 phosphorylation stimulated by ASK1. Moreover, stimulation of the angiotensin II type 1A receptor activated JNK3 and triggered the colocalization of beta-arrestin 2 and active JNK3 to intracellular vesicles. Thus, beta-arrestin 2 acts as a scaffold protein, which brings the spatial distribution and activity of this MAPK module under the control of a GPCR. 相似文献

The E6AP ubiquitin-protein ligase (E3) mediates the human papillomavirus-induced degradation of the p53 tumor suppressor in cervical cancer and is mutated in Angelman syndrome, a neurological disorder. The crystal structure of the catalytic hect domain of E6AP reveals a bilobal structure with a broad catalytic cleft at the junction of the two lobes. The cleft consists of conserved residues whose mutation interferes with ubiquitin-thioester bond formation and is the site of Angelman syndrome mutations. The crystal structure of the E6AP hect domain bound to the UbcH7 ubiquitin-conjugating enzyme (E2) reveals the determinants of E2-E3 specificity and provides insights into the transfer of ubiquitin from the E2 to the E3. 相似文献

Normal corn germplasm can be used to improve popcorn (Zea mays L.) through 1-2 backcrosses with popcorn as recurrent parents. Popping characteristics of derived lines from popcorn × dent or flint corn crosses play a determinant role in popcorn breeding. Advanced backcross QTL methods can effectively combine QTL identification and plant breeding. 220 selected BC2F2 families developed from a cross between Dan 232, a dent corn inbred line, and N04, an elite popcorn inbred line, were evaluated for three popping characteristics, popping volume, flake size, and popping rate, under two environmental conditions. Using composite interval mapping, a total of 10 significant QTLs were detected, and of these, 2 to 4 QTLs were identified for each trait. Six QTLs had favorable alleles contributed by Dan 232. Comparison with the 15 QTLs detected in the F2:3 families showed that 3 QTLs were the same in both populations. The QTLs should be redetected in generations developed through severe selection. Improved N04 and near isogenic lines could be developed from this BC2F2 population through selfing or another 1 to 2 backcrosses with N04. 相似文献

Polar stratospheric clouds (PSCs) play a key role in stratospheric ozone depletion. Surface-catalyzed reactions on PSC particles generate chlorine compounds that photolyze readily to yield chlorine radicals, which in turn destroy ozone very efficiently. The most prevalent PSCs form at temperatures several degrees above the ice frost point and are believed to consist of HNO(3) hydrates; however, their formation mechanism is unclear. Results of laboratory experiments are presented which indicate that the background stratospheric H(2)SO(4)/H(2)O aerosols provide an essential link in this mechanism: These liquid aerosols absorb significant amounts of HNO(3) vapor, leading most likely to the crystallization of nitric acid trihydrate (NAT). The frozen particles then grow to form PSCs by condensation of additional amounts of HNO(3) and H(2)O vapor. Furthermore, reaction probability measurements reveal that the chlorine radical precursors are formed readily at polar stratospheric temperatures not just on NAT and ice crystals, but also on liquid H(2)SO(4) solutions and on solid H(2)SO(4) hydrates. These results imply that the chlorine activation efficiency of the aerosol particles increases rapidly as the temperature approaches the ice frost point regardless of the phase or composition of the particles. 相似文献

【目的】研究控制小麦株高的数量性状位点(QTL)。【方法】利用SSR和AFLP分子标记构建连锁图谱,在3种不同试验环境(2003~2004年北京、2004~2005年北京和河南安阳)下分析百农64×京双16组合的218个F2:3株系群体的株高。【结果】构建了由158个分子标记(100个SSR标记和58个AFLP标记)位点组成的遗传连锁图谱,覆盖了除1D连锁群外小麦全基因组的3 114 cM;检测到3个控制株高的QTL,分别位于2B、4D和6A染色体上,贡献率分别为7.3%~11.5%,7.4%~12.9%和5.7%~11.3%。【结论】3个株高QTL位点在不同环境下表现稳定,其紧密连锁的分子标记可用于矮秆、半矮秆小麦的标记辅助育种。 相似文献

The distribution of H-2 isoantigens on the plasma membrane of lymphocytes and thymocytes was studied with ferritin-conjugated alloantibody. The spatial arrangement of the membrane components which possess H-2 isoantigens is markedly altered by the attachment of a second antibody, directed toward mouse gamma globulin or ferritin; the second anitibody appears to cause the aggregation of the sensitized H-2 antigens into discrete zones on the cell membrane. Other membrane antigens may be equally affected by indirect labeling techniques and, thus, topographic analysis of the antigenic structure should be done with antibody that is directly conjugated. 相似文献

Organic dianions have been stabilized by (M(C(5)(CH(3))(5))(2))(+), where M is iron or cobalt. This has allowed the structural and spectroscopic characterization of these dianions. The structure of (M(C(5)(CH(3))(5))(2))(2)(+) (DDQ)(2-), where DDQ is 2,3-dichloro-5,6-dicyanobenzoquinone, has been determined by x-ray crystallography. The structure of (DDQ)(2-)is consistent with ab initio molecular orbital calculations that suggest a localizd as opposed to a delocalized (aromatic) ring structure. 相似文献

Traditional pest control approaches rely mostly on the experience of farmers, which may not be effective due to lack of scientific information regarding the environment where crops grow. Farmers can initiate a more effective integrated pest management program when precise and quantified results of forecasting pest population outbreaks are provided. Previous studies generally utilize long-term data to predict pest populations, but such a prediction approach might not be useful for farmers who grow fruit and vegetables with shorter life cycles. This paper therefore proposes an interval type-2 fuzzy logic system (IT2FLS) with short-term data to forecast the population dynamics of the oriental fruit fly (OFF, Bactrocera dorsalis (Hendel)) and the tobacco cutworm (TC, Spodoptera litura (Fabricius)). Two automatic monitoring systems are used to collect the data of the population dynamics of OFFs and TCs and the environmental parameters in farming areas. A univariate fuzzy time series forecasting model with difference-based intervals (UFTSFM_DI) and a bivariate fuzzy time series forecasting model with difference-based intervals (BFTSFM_DI) are developed, and integrated into the proposed IT2FLS. It is found that the BFTSFM_DI model yields better performances of forecasting OFF and TC populations when the atmospheric temperature data are employed. With the forecasting results, farmers will have a better understanding of the population dynamics of the OFF and TC in farming areas, so they can take proper measures, such as bagging their fruits and spraying pesticides, before pest outbreaks occur. 相似文献
