The major purpose of this investigation was to determine the effect of prednisolone and megestrol acetate in cats on the adrenal cortisol response to exogenous adrenocorticotrophic hormone during drug administration at dose rates employed for management of some inflammatory feline dermatoses. Prednisolone (at least 2 mg/kg/day) and megestrol acetate (5 mg/cat/day) were each administered orally to seven cats from days 1 to 16. Three additional cats received no therapy. Basal and stimulated cortisol concentrations, food and water intake, hematology, blood biochemistry, urinalyses, and hepatic and cutaneous histology were studied in all cats before, during, and two weeks following the end of treatment. Cats given prednisolone or megestrol acetate had significant suppression of stimulated cortisol levels on day 8. This change was more marked on day 15, when the suppression in cats given megestrol acetate was also significantly more severe than in those receiving prednisolone. Recovery of adrenal reserve was considered present on day 30 in six of seven cats given prednisolone, but in only three of seven receiving megestrol acetate. Eosinopenia, glycosuria and hepatocyte swelling from glycogen deposition were occasionally recorded in treated cats of both groups, providing additional circumstantial evidence for glucocorticoid activity of megestrol acetate in cats. It is advised that abrupt withdrawal of prednisolone or megestrol acetate therapy be avoided in this species to reduce the chance of precipitating clinical signs of hypoadrenocorticism, even after treatment for as little as one week.  相似文献

This study evaluated the effect of two combination products containing febantel, pyrantel, and praziquantel (FPP) for the treatment of Giardia species in experimentally infected kittens. In experiment 1, five kittens were administered the United States (US) formulation of FPP at doses of 37.8 mg/kg, 7.56 mg/kg, and 7.56 mg/kg, respectively, PO, q24h, for 5 days and four kittens remained as controls. In experiment 2, five kittens were administered the European formulation of FPP at the doses of 12.5 mg/kg, 12 mg/kg, and 4.16 mg/kg, respectively, PO, q24h, for 5 days and four kittens remained as controls. In experiment 3, six kittens were administered the US formulation of FPP at 56.5 mg/kg, 11.3 mg/kg, 11.3 mg/kg, respectively, PO, q24h, for 5 days and five kittens remained as controls. Thirteen days after treatment, kittens testing negative for Giardia species cysts were administered 20 mg/kg methylprednisolone acetate, IM, weekly for a maximum of two injections. Feces were analyzed for Giardia species cysts using a direct immunofluorescence test. After experiment 3, four of the six treated kittens, but no control kittens, remained negative for Giardia species after the administration of methylprednisolone acetate.  相似文献

Daily doses between 30 and 35 mg megestrol acetate were administered to 4,000 heifers over roughly two weeks and produced highly synchronising effects which, however, were lower in response to lower doses. Rectal checks were applied to the ovaries of heifers which had received megestrol acetate and showed that here the number of mature follicles was higher than recorded from animals that had been given chlormadinone acetate. Megestrol acetate, applied in granular form blended into mixed fodder, proved to be the most favourable variant of application. Synchronisation of oestrus on the basis of the above approach was found to yield per animal savings between 4.30 and 22.70 roubles under practice conditions.  相似文献

Heinz body formation was examined in kittens, in response to consumption of a variety of diets. A commercial salmon-based diet containing 16.5 mg of nitrite, 39 mg of histamine, and 210,000 IU of vitamin A/kg of diet (dry-matter basis) was found to induce Heinz body formation. Purified experimental diets--containing nitrite up to 405 mg/kg; histamine, 50 mg/kg; histamine 50 mg/kg plus nitrite, 45 mg/kg; or vitamin A, 250,000 IU/kg--failed to induce Heinz body formation. The effect of propylene glycol (PG) on Heinz body formation was examined by giving groups of 6 kittens purified diets containing 5 or 10% PG for 12 weeks. Two additional kittens were fed a commercial soft-moist diet containing PG for 12 weeks. All kittens fed PG developed Heinz bodies, with peak values for erythrocytes containing Heinz bodies being: 28% for kittens of the 10% PG group; 20% for kittens of the 5% PG group; and 36% for kittens of the soft-moist diet group. Kittens did not develop anemia or methemoglobinemia. Heinz body percentage required 6 to 8 weeks to decrease to the pretreatment value of less than 1% after diets containing PG were discontinued. 51Chromium-labeled erythrocytes were used to evaluate erythrocyte survival in 4 kittens of the 10% PG-fed group and in 4 control kittens. Kittens with Heinz body formation induced by 10% PG had significantly (P less than 0.001) decreased erythrocyte-survival, compared with that for controls, with half-life of 8.3 days for kittens of the PG group, compared with 12.6 days for kittens of the control group.  相似文献

OBJECTIVE: To determine whether passive transfer of immunity affects results of diagnostic tests for antibodies against FIV in kittens born to vaccinated queens. DESIGN: Experimental trial. ANIMALS: 12 specific-pathogen-free queens and their 55 kittens. PROCEDURE: Queens were vaccinated with a whole-virus FIV vaccine prior to breeding. Serum was obtained from the queens on the day of parturition and from the kittens on days 2 and 7, then weekly until results of tests for antibodies against FIV were negative for 2 consecutive weeks. Milk was collected from the queens daily for the first week and then weekly. Serum and milk were tested for antibodies against FIV with 2 commercial assays. RESULTS: Antibodies against FIV were detected in serum obtained from the queens on the day of parturition and in the milk throughout lactation. All kittens tested positive for antibodies against FIV at 2 days of age. At 8 weeks of age, 30 (55%) kittens tested positive with 1 of the commercial assays, and 35 (64%) tested positive with the other. All kittens tested negative for antibodies against FIV by 12 weeks of age. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that kittens readily absorb antibodies against FIV in colostrum from vaccinated queens and that these antibodies may interfere with results of commercially available tests for FIV infection past the age of weaning. Currently licensed diagnostic tests for FIV infection are unable to distinguish among kittens with antibodies against FIV as a result of infection, passive transfer from infected queens, and passive transfer from vaccinated queens.  相似文献

Cats were given megestrol acetate (MA, 5 mg once daily for 14 days), subcutaneous proligestone (PRG, 100 mg on two occasions one week apart) or subcutaneous saline (1 ml as for PRG). In cats given saline (n = 6) basal cortical concentrations, cortisol concentrations after adrenocorticotrophic hormone (ACTH) administration and fasting blood glucose concentrations did not change significantly during the following seven weeks. Cats given MA (n = 7) developed suppression of basal and ACTH-stimulated cortisol concentrations and fasting hyperglycaemia during treatment. Effects on cortisol persisted for two weeks after MA dosage ceased. In cats given PRG (n = 7), basal cortisol concentrations were reduced overall, but only three cats had persistently suppressed post-ACTH cortisol concentrations. Adrenal suppression continued for 14 weeks in one of these and for at least 22 weeks in two cats. Fasting blood glucose concentrations were unchanged in PRG-treated cats.  相似文献

OBJECTIVES: To determine the efficacy of ronidazole (RDZ), tinidazole (TDZ), and metronidazole (MDZ) against Tritrichomonas foetus in vitro and of RDZ for treatment of feline naturally occurring or experimentally induced T. foetus infection. ANIMALS: A cat naturally infected with T. foetus infection and diarrhea. Ten specific-pathogen-free (SPF) kittens. PROCEDURE: RDZ, TDZ, and MDZ were tested for activity against 3 different feline isolates of T. foetus in vitro. RDZ then was administered to a naturally infected cat at 10 mg/kg PO q24h for 10 days. SPF kittens were infected orogastrically with feline T. foetus and treated with either placebo or RDZ (10 mg/kg PO q12h for 14 days). Cats with relapsing infection or those receiving placebo were treated subsequently with RDZ (either 30 or 50 mg/kg PO q12h for 14 days). Feces were examined for T. foetus by direct microscopy, culture, and polymerase chain reaction (PCR) testing weekly. RESULTS: Both RDZ and TDZ killed T. foetus at concentrations >0.1 microg/mL in vitro. In the naturally infected cat, RDZ abolished diarrhea and T. foetus infection for 85 days after treatment, at which time infection and diarrhea relapsed. Retreatment with RDZ eradicated diarrhea and T. foetus infection for over 407 days. In experimentally induced infection, RDZ at 10 mg/kg caused initial improvement, but infection relapsed in all 5 cats 2 to 20 weeks after treatment. At 30 or 50 mg/kg, 10/10 cats were negative for T. foetus infection for follow-up durations of 21 to 30 weeks after treatment. CONCLUSIONS AND CLINICAL RELEVANCE: Oral administration of RDZ at 30 to 50 mg/kg q12h for 14 days resolved diarrhea and eradicated infection (on the basis of polymerase chain reaction [PCR] testing) in 1 naturally infected cat and 10 experimentally inoculated cats receiving a different isolate of T. foetus.  相似文献

Over several years, the mass of hair shed weekly by a normal Alsatian bitch varied cyclically. Base-line values were less than 5 grams, while the maximum loss recorded was over 70 grams. Moults (and, by implication, the growth of new hairs which must have preceded them) were related to the bitch's oestrous cycle, always occurring shortly before or coincidentally with the onset of pro-oestrus, even when megestrol acetate was administered to prevent ovulation or to re-time the oestrous cycle. Fluctuating levels of ovarian hormones over-rode any influence upon hair cycles of seasonal changes in photoperiod or temperature.  相似文献

The effect of a low-selenium diet on thyroid hormone metabolism was investigated in growing kittens. Twelve specific-pathogen-free kittens with ages ranging from 16 to 18 weeks were divided into two groups of equal number with equal sex distribution in each group. One group was fed a yeast-based low-selenium diet (0.02 mg Se/kg diet) while the other group was fed the same diet supplemented with Na2SeO3 at 0.4 mg Se/kg diet for 8 weeks. Food intake, body weight and body weight gain were not affected by the low-Se diet during the study period. However, kittens given the low-Se diet had significantly reduced plasma selenium concentration and glutathione peroxidase activity. Plasma total thyroxine (T4) increased and total 3,5,3'triiodothyronine (T3) decreased significantly in kittens fed the low-Se diet at the end of the study. These results suggest that type I deiodinase in cats is a selenoprotein- or a selenium-dependent enzyme.  相似文献

The efficacy of a modified live-virus intranasal vaccine and a killed-virus adjuvanted parenteral vaccine in inducing protective immunity against feline viral rhinotracheitis (FVR) was evaluated in kittens with and without maternally derived FVR antibodies. The intranasal vaccine was given as a single dose to kittens 5 weeks old, and the parenteral vaccine was administered in 2 doses at 5 and 7 weeks of age. Seroconversion was delayed for 5 to 10 days in kittens with maternally derived antibodies, but occurred in all vaccinated kittens by 8 weeks of age. When virulent FVR virus was given, both vaccines provided satisfactory protection against disease but did not prevent infection. The results indicated that the modified live-virus intranasal vaccine or the killed-virus adjuvanted parenteral vaccine can be used successfully in kittens with residual maternally derived FVR antibodies.  相似文献

Long-term studies of megestrol acetate and chlormadinone acetate in 100 female dogs are in progress. Doses of zero, one, 10 and 25 times the expected human dose of megestrol acetate and 25 times the expected human dose of chlormadinone acetate (on a milligram per kilogram body weight basis) are being given daily. During the first 4 years, eight dogs from each of the five groups were killed. The principal gross findings included enlarged uteri with mucoid material in the lumina, mammary development in dogs given middle and high doses of megestrol acetate and chlormadinone acetate, and thickened gallbladder walls in dogs given high doses of each. Histologic evaluation showed inhibition of ovulation for progestogen-treated dogs and suppression of ovarian follicular development with the high doses. Cystic endometrial hyperplasia was slight in the low-dose dogs and moderate to severe in most of the high-dose dogs; a few also had ulcerative endometritis and pyometra. The mammary glands of dogs given the middle and high doses produced lobules, acini, and secretion exceeding natural metestrus. Slight to marked cystic mucinous hyperplasia occurred in the gallbladders of most dogs given the high doses. Tow high-dose megestrol dogs had clinical signs and microscopic pancreatic, renal, and ocular changes indicative of diabetes mellitus.  相似文献

A medication error resulted in two kittens being treated with azathioprine (12 and 12.5 mg/kg) instead of azithromycin for 2 weeks. On clinical examination, the kittens were febrile, weak, and had oronasal hemorrhage. Complete blood cell counts indicated severe bone marrow suppression. Treatment consisted of multiple transfusions, antibiotics, and granulocyte colony-stimulating factor. One of the kittens responded to therapy and had a complete recovery. The other kitten was treated for 40 days with no clinical response before dying. Both kittens also contracted Mycoplasma hemofelis infection from a contaminated blood transfusion.  相似文献

The site within the hypothalamic-pituitary axis at which cortisol acts to inhibit luteinizing hormone (LH) secretion was investigated in female pigs. Six ovariectomized, hypophysial stalk-transected (HST) gilts were given 1 microgram pulses of gonadotropin releasing-hormone (GnRH) iv every 45 min from day 0 to 12. On days 6-12, each of 3 gilts received either hydrocortisone acetate (HCA; 3.2 mg/kg body weight) or oil vehicle im at 12-hr intervals. Four ovariectomized, pituitary stalk-intact gilts served as controls and received HCA and pulses of 3.5% sodium citrate. Jugular blood was sampled daily and every 15 min for 5 hr on days 5 and 12. Treatment with HCA decreased serum LH concentrations and LH pulse frequency in stalk-intact animals. In contrast, serum LH concentrations, as well as the frequency and amplitude of LH pulses, were unaffected by HCA in HST gilts and were similar to those observed in oil-treated HST gilts. We suggest that chronically elevated concentrations of circulating cortisol inhibit LH secretion in pigs by acting at the level of the hypothalamus.  相似文献

Six cats (Nos. 1-6) were inoculated intramuscularly with (1 x 10(6)) and orally (5 x 10(5)) tachyzoites of Neospora caninum. Three (Nos. 1-3) of the six cats were given 40 mg/kg methylprednisolone acetate 7 days before and on the day of inoculation with N. caninum tachyzoites, and three cats (Nos. 4-6) were not given methylprednisolone acetate. Two of the cats (cat Nos. 1 and 2) given methylprednisolone acetate died suddenly. Cat No. 1 died 8 days post-inoculation, and cat No. 2 died 16 days post-inoculation. Cat No. 3 was euthanatized 21 days post-inoculation. Cat No. 1 had lesions of gram-positive bacterial septicemia. Necrotizing encephalitis, myelitis, disseminated skeletal muscle necrosis, hepatic necrosis, interstitial pneumonia, and renal tubular necrosis were the main lesions in cat Nos. 2 and 3. The cats that were not given methylprednisolone acetate remained clinically normal except for slight weight loss in cat No. 6. All three of these cats were euthanatized 55 days post-inoculation. Mild myositis and encephalitis were noted on microscopic examination of tissues from these three cats. Neuromuscular lesions were not seen in six control cats (Nos. 7-12) not inoculated with N. caninum and euthanatized 21 or 22 days after administration of the first two doses of methylprednisolone acetate (40 mg/kg), given at a weekly interval.  相似文献

Adrenal and/or thyroid gland function tests were evaluated in horses at various times during short-term therapy with phenylbutazone, stanozolol, and boldenone undecylenate. There were no significant treatment or time effects on mean basal plasma cortisol concentrations in horses during treatment with the following: phenylbutazone, given twice daily (4 to 5 mg/kg, IV) for 5 days; stanozolol, given twice weekly (0.55 mg/kg, IM) for 12 days; boldenone undecylenate, given twice weekly (1.1 mg/kg, IM) for 12 days; or nothing. There was no significant effect of phenylbutazone treatment on the changes in plasma cortisol concentration during the combined dexamethasone-suppression adrenocorticotropic hormone (ACTH)-stimulation test. Plasma cortisol concentration was significantly decreased from base line at 3 hours after dexamethasone administration and was significantly increased from base line at 2 hours after ACTH in all horses (P less than 0.05). Likewise, the stimulation of basal plasma cortisol concentrations at 2 hours after administration of ACTH (P less than 0.05) was not affected by treatment with stanozolol or boldenone undecylenate. There were no significant treatment effects on mean basal plasma concentrations of thyroxine (T4) or triiodothyronine (T3) among horses during the following treatments: stanozolol, given twice weekly (0.55 mg/kg, IM) for 12 days; boldenone undecylenate, given twice weekly (1.1 mg/kg, IM) for 12 days; or nothing. The objective of this study was to evaluate the effects of in utero and postnatal exposure of a high‐protein (HP; n = 9) or moderate‐protein (MP; n = 16) diet on growth, and serum metabolite, ghrelin and leptin concentrations during the first 4 months of life in kittens. It was hypothesized that blood indices would be modified due to diet. Blood samples were collected from kittens at 4, 8, 12 and 16 weeks of age. Kittens were weaned at 8 weeks of age onto the same diet as the dam. Body weight was measured weekly from birth and daily food intake for each litter was recorded post‐weaning. Serum concentrations of urea nitrogen, total protein and triglycerides were greater (P < 0.05) in kittens fed the HP diet. Serum cholesterol concentrations were greater (P < 0.05) in MP‐fed kittens at 4 weeks of age. Moderate‐protein fed kittens tended to have greater (P < 0.10) serum ghrelin concentrations. Leptin concentrations were not affected by diet, but changed over time (P < 0.05). Our data indicate that diet and age of kittens affect circulating concentrations of peptides important in appetite regulation. Further research testing the effects of in utero and early postnatal nutrient exposure on feline obesity risk in adulthood is needed.  相似文献

A commercial feline leukemia virus (FeLV) vaccine was evaluated in a natural exposure system. All kittens were negative for FeLV antigen on two enzyme-linked immunosorbent assay (ELISA) tests and one indirect immunofluorescence antibody (IFA) test before vaccination or exposure. Twenty-three kittens were vaccinated subcutaneously at nine and 12 weeks of age. The vaccinated kittens and 14 unvaccinated littermates were housed in an infected environment starting at 14 weeks. The kittens were exposed for 24 weeks by living in a large room with one feline leukemia virus-positive, asymptomatic adult cat for each five kittens. Sixty-four percent of the unvaccinated kittens and 70% of the vaccinated kittens became infected as determined by ELISA. Forty-three percent of unvaccinated kittens and 39% of vaccinated kittens died. There was no difference between the infection and mortality of vaccinated kittens that developed antibodies to anti-FeLV glycoprotein 70-envelope antigen and those that did not. Consideration should be given to evaluation of feline leukemia virus vaccines using "street" virus in a natural exposure system.  相似文献

Five milligram of megestrol acetate were orally used over ten days during mating periods to synchronise oestrus in caracool sheep. Eighteen days were allowed to elapse from the last administration of progestagen before 1,000 I.U. PMSG were injected to boost fertility. The test reported in this paper had been applied to a large sheep herd over three years. The following conclusions were drawn: Synchronised occurrence of heat in the mating period was not reduced by oral administration of 5 mg megestrol acetate over ten days for a period of three years (87.1 to 95.5 per cent). The fertilisation rate in response to first insemination was between 68.8 and 73.1 per cent. Birth to lambs was given by 85-92 per cent of the ewes after two inseminations. No fertility occurred in 1.5-3.1 per cent of all animals.  相似文献

High concentrations of retinoids occur in some commercial cat food formulations as a result of the use of animal liver as an ingredient. Our objective was to study the teratogenic potential of dietary vitamin A in cats. We investigated the incidence of birth defects in kittens of queens given diets with retinyl acetate concentrations of 6000, 306000, or 606000 retinol equivalents (RE)/kg diet (control, 306K, or 606K groups, respectively) for approximately 3 years [1 RE=1 micro g retinol=3.3 International Units (IU)]. Each group comprised 12-15 age-matched, nulliparous domestic short-haired queens that were exposed to toms. There were a total of 396 kittens born in 97 litters. Pregnancy rate, number of kittens per gestation and gestations per year were not significantly different among treatment groups. A total of 2, 5 and 11 malformed kittens occurred in the control, 306K and 606K groups, respectively. Malformations included cleft palate, cranioschisis, foreshortened mandible, stenotic colon, enlarged heart and agenesis of the spinal cord and small intestine, which are typical foetal defects consistent with ingestion of excess retinoids in other species. This study demonstrated that a concentration of 306000 RE/kg diet has a potential for causing birth defects in the kittens.  相似文献
