The present study was conducted to evaluate whether supplementation of semen extender with glutathione (GSH) can maintain the quality of frozen-thawed canine spermatozoa. Eighteen ejaculates were obtained from 5 dogs and placed in extender (20% egg yolk, Tris, citric acid, lactose, raffinose, antibiotics and 6.5% glycerol) containing 0 (control), 2.5, 5, 7.5 or 10 mM GSH. The samples were cooled to 4 C and then frozen in liquid nitrogen vapor. Motility parameters of the sperm were evaluated at 0, 1, 2, 3, 4, 12 and 24 h after thawing. Sperm motility was higher in the 5 mM GSH group than in the control or 2.5 and 10 mM GSH groups; this effect was observed at 1 to 24 h after thawing (P < 0.05). The 5 mM GSH group had a higher sperm viability index at 12 and 24 h after thawing compared with the other groups (P < 0.05). Acrosome integrity, evaluated at 4 h after thawing, was greater in two of the GSH-treated groups (5 and 10 mM) compared with the control. Lipid
peroxidation (LP) levels immediately after thawing were lower in the 5 and 10 mM GSH groups compared with the control, while those at 12 h after thawing did not differ significantly. Frozen-thawed semen in the 5 mM GSH group was used for transcervical insemination of 4 bitches, resulting in delivery of 5 puppies from 2 bitches. These results indicate that supplementation of semen extender with 5 mM GSH was effective in improving motility, longevity and acrosomal integrity and inhibiting LP levels in post-thaw canine spermatozoa, without any adverse impacts on full-term development after transcervical insemination. 相似文献

Kisspeptin is a key molecule that stimulates gonadotropin secretion via release of
gonadotropin-releasing hormone (GnRH). In the present study, our aim was to
investigate whether kisspeptin has stimulatory effects on follicular development via
GnRH/gonadotropin secretion in cows. Japanese Black beef cows were intravenously
injected with full-length bovine kisspeptin [Kp-53 (0.2 or 2 nmol/kg)] or vehicle 5
days after they exhibited standing estrus (Day 0). In cows injected with Kp-53 at 2
nmol/kg, the follicular sizes of the first dominant follicles increased on Day 6 and
thereafter. Ovulation of the first dominant follicle occurred in 1 out of 4 cows
treated with Kp-53 at 2 nmol/kg. Injection of Kp-53 at 2 nmol/kg increased the
concentration of plasma luteinizing hormone (LH) but not follicle-stimulating
hormone, over a 4-h period following injection in all cows. The present study
suggests that administration of full-length kisspeptin causes LH secretion, which is
sustained for a few hours, and it is capable of stimulating follicular development

This study assessed the feasibility of using camera image analysis to detect behavioral changes as an indicator of the onset of calving in Japanese Black cattle. Thirty-five pregnant cattle individually housed in pens were used and were continuously monitored using a digital camera system. For the automatic determination of the x and y coordinates of a cow, trajectory analysis was conducted using thermal image and analysis software, and the distances moved were calculated using coordinate data. Further, the frequency of postural changes and the time spent tail raising per hour were measured for 14 cows using visible images. The measurement data were used to calculate hourly data for 12 h prior to amniorrhexis (first rupture of the allantoic sac). The hourly distances moved tended to increase at the time of amniorrhexis, with significantly longer distances measured 3–0 h before amniorrhexis than those at 12–8 h before amniorrhexis (P < 0.05). In all cows, amniorrhexis occurred within 11 h of hourly distances moved by more than 50% compared with distance moved the previous hour. The overall average elapsed time before amniorrhexis was 9 h 30 min (range: 5–11 h). Tail raising time and the frequency of postural changes significantly increased at 1–0 h and 2–0 h before amniorrhexis, respectively. This suggests that predicting the time of calving is possible by measuring the activity of Japanese Black cows during late pregnancy using camera image analysis as a non-invasive technique. 相似文献

This study aimed to determine the effects of feeding an increased volume of high‐fat milk during the early post‐natal life on metabolite concentrations in the blood, the expression of key genes regulating intermediary metabolism in the skeletal muscles, and the rate of growth of Japanese Black cattle. All calves were fed a high‐fat milk replacer (crude protein, 26%; crude fat, 25.5%; total dissolved nitrogen, 116%). Control calves (n = 4) were nursed with 500 g milk replacer until 3 months of age, whereas calves in the experimental group (n = 4) were nursed with 1800 g milk replacer until 3 months, and then the volume was gradually reduced until 5 months. Body weight was significantly higher in the experimental group than in the control group at 7 months. Plasma glucose concentrations were significantly lower in the experimental group. Expression of glucose‐transporter‐4 messenger RNA (mRNA) was lower, whereas that of glucose transporter 1, cluster of differentiation 36, and carnitine palmitoyltransferase‐1b mRNA was significantly higher in the Longissimus thoracis of the experimental group. Nutritional status during early post‐natal life appears to strongly influence the growth rate and glucose and lipid metabolism in Japanese Black cattle. 相似文献

This study was conducted to examine the effects of adding glutathione (1 mM) to media used for sperm washing and in vitro fertilization (IVF) on the improvement of early development of embryos produced using cryopreserved spermatozoa of the less IVF-competent bull (the one considered unqualified as spermatozoa supplier for the production of bovine blastocysts using IVF). The cryopreserved spermatozoa of this bull were characterized by normal motility and lower ATP content and blastocyst productivity than those of IVF-competent bulls. The addition of glutathione to the sperm washing medium was more effective in improving the productivity of blastocysts and ATP content than the addition of glutathione to the IVF medium or no glutathione addition at all (control). These results suggest that this simple method may be used to improve the potential of cryopreserved spermatozoa of less IVF-competent bulls to fertilize oocytes in vitro and to induce normal embryonic development after fertilization. 相似文献

The freezing conditions for preparation of frozen canine semen by the plunging method were investigated with regard to the period of sensitization in liquid nitrogen (LN2) vapor and the height from LN2, and the semen qualities after thawing were compared with those of canine semen prepared by the simple freezer method previously reported by us. In the plunging method, 9 semen straws were prepared under the same conditions, horizontally kept at 5, 7, and 10 cm above the LN2 surface in a styrene foam box for 5, 10, and 15 min, and then plunged into LN2. The semen qualities immediately after thawing were high in the 7 cm/10 min (cooling rate: -4 to -22 degrees C/min) and 10 cm/15 min groups (cooling rate: -6 to -10 degrees C/min). On comparison of frozen semen prepared by the plunging method (7 cm/10 min) with frozen semen prepared by the simple freezer method, sperm motility and viability were significantly higher for the frozen semen prepared by the plunging method. The cooling rate in freezing was higher for the simple freezer method (cooling rate: -6 to -50.9 degrees C/min) than the plunging method. Based on these findings, horizontal placement of canine semen straws above LN2 to reduce the temperature at a slow cooling rate of about -10 degrees C/min, followed by plunging into LN2 after sensitization for 10-15 min, provides good semen qualities after thawing. 相似文献

The aim of this study was to evaluate fertility and sex ratios after artificial insemination in dogs under field conditions. Semen was cryopreserved as unsorted (control) or was separated into X‐ and Y‐chromosome‐bearing sperm using a cell sorter. Sixty female dogs were inseminated with frozen–thawed spermatozoa of 100 × 106 unsorted (a dose in practice) and 4 × 106 sorted (X and Y group, respectively). A total of 20 dogs became pregnant and 126 puppies were born from the three groups. The percentage of parturition was similar for the X (5/20; 25.0%) and Y (4/20; 20.0%) group (P > 0.05), but lower than controls (11/20; 55.0%) (P < 0.05). Ultimately 28 out of the 32 puppies produced from X group were female (87.5%) and 19/22 (86.4%) puppies of Y group were male. In contrast, sex ratio (51.4% to 48.6%) in the control was significantly different from the X, Y group (P < 0.05). However, male and female puppies in the control had similar birth weights and weaning weights to those from the X and Y groups. This preliminary information indicated that normal puppies of predicted sex can be produced with low numbers of sorted cryopreserved dog spermatozoa at a farm level, making sperm‐sexing technology potentially applicable for elite breeding units. 相似文献

在我国南方很多省份,肉牛主要以散养为主,导致母牛发情后常不能及时冷配,而冷配员却又闲着的现状,从而使母牛配种率低、存栏母牛有下降趋势。针对此情况,研发了肉牛冷配软件(软著登字2009SR11157,软件产品登记测试(2009-12-S2701)),对发情(乏情)预报、发情控制以及冷配员日常工作等方面进行计算机管理。冷配员通过规范地记录日常工作数据,逐步输入计算机,利用软件制定的日常工作计划实施冷配。结果表明:(1)对发情正常但漏配的母牛,通过软件准确预报有效黄体期,实施1次PG同期发情技术,同期发情率高于直肠检查组;(2)对乏情母牛,既实现了不通过直肠检查尽早发现、及时治疗缩短产犊间隔的目的,又获得了较高的受胎率;(3)规范了母牛的繁殖记录、提高了数据利用效率,并促进了牛繁殖数据记录的持续开展。 相似文献

This study sought to establish whether temperature gradients between the cervix, vagina, and rectum at and 7 days post-artificial insemination (AI) were associated with the incidence of pregnancy in lactating dairy cows (Experiment I; n = 90 ovulating cows) and to evaluate temperature gradient dynamics from the time of insemination to 7 days post-AI under heat stress conditions (Experiment II; n = 16 ovulating and 4 non-ovulating cows). In Experiment I, 39 cows (43.3%) became pregnant. The odds ratio for pregnancy was 2.5 for each one-tenth of a degree drop in cervical temperature with reference to the control rectal temperature at the time of AI (P = 0.01), whereas the same decrease in the cervix–rectum temperature differential 7 days post-AI resulted in an odds ratio of 0.44 (P = 0.02). In Experiment II, 5 of the ovulating cows (31.3%) became pregnant. The mean values of the vagina–rectum, vagina–cervix, and cervix–rectum temperature differentials at AI (day 0), 8 h, 24 h, and 7 days post-AI changed significantly from day 0 to day 7 (within-subject effect; P < 0.02) in ovulating cows but not in non-ovulating cows. Temperature differentials on days 0 and 7 were similar between ovulating cows and cows of Experiment I. Overall, our findings support the notion that a temperature differential between the caudal cervical canal and rectum at AI may be an indicator of the likelihood of pregnancy. Possible prospects of confirming estrus at the herd-level are also suggested. 相似文献

Eosinophilic enteritis (EOE) is a type of inflammatory bowel disease and is characterized
clinically by chronic obstinate diarrhea. Three Japanese Black (JB) fattening cattle (2
males and 1 female) on different cattle farms presented with chronic episodic diarrhea
without fever or dehydration. Soft reddish spherical carneous tissues (1−3 cm) were
occasionally excreted within the diarrheic feces. Administration of antibiotics,
antidiarrheal drugs and vermicides had no therapeutic effect, but dexamethasone improved
the fecal characteristics. The symptoms persisted until the animals were slaughtered at
27–30 months of age. Histopathological examination of the intestines revealed marked
eosinophilic infiltration in the lamina propria and submucosa. From these findings, we
diagnosed these cattle as the first cases of EOE in JB cattle. 相似文献

Monosaccharides such as glucose contribute to the development of meat flavor upon heating via the Maillard reaction; therefore, monosaccharide content is related to beef palatability. Here, we analyzed the effects of genetic and environmental factors on the content of glycogen, one of the precursors of monosaccharides, in the muscles of 958 fattened Japanese Black cattle from Yamagata Prefecture. Analysis of variance showed that muscle glycogen content was affected by the farm and postmortem periods, but not by sex, slaughter age, slaughter month or number of days detained at the slaughter yard. Additionally, consumption of digestible brown rice feed elevated muscle glycogen levels. Glycogen heritability was estimated to be 0.34, and genetic correlations between glycogen and carcass weight (CW) or beef marbling standard (BMS) were weak. The predicted breeding values varied among paternal lines. These results demonstrated that genetic factors might improve muscle glycogen content and therefore beef palatability, but do not influence CW or BMS. 相似文献

Variances caused by the differential expression of paternally and maternally imprinted genes controlling carcass traits in Japanese Black cattle were estimated in this study. Data on marbling score (BMS), carcass weight, rib thickness, rib‐eye area (REA) and subcutaneous fat thickness (SFT) were collected from a total of 13,115 feedlot steers and heifers in a commercial population. A sire–maternal grandsire model was used to analyse the data, and then, imprinting parameters were derived by replacing the genetic effect of the dam with the effect of the maternal grandsire in the imprinting model to calculate the genetic parameter estimates. The proportions of the total genetic variance attributable to imprinted genes ranged from 8.7% (SFT) to 35.2% (BMS). The remarkably large imprinting variance of BMS was mainly contributed by maternally expressed inheritance because the maternal contribution of the trait was much larger than that of the paternal trait. The parent‐of‐origin effect originating from maternal gene expression was also observed for REA. The results suggested the existence of genomic imprinting effects on the traits of the Japanese Black cattle. Hence, the parent‐of‐origin effect should be considered for the genetic evaluation of Japanese Black cattle in breeding programmes. 相似文献
