The fungus Pyrenophora tritici-repentis, causal agent of tan spot of wheat, produces two phenotypically distinct symptoms, tan necrosis and extensive chlorosis. The inheritance of resistance to chlorosis induced by P. tritici-repentis races 1 and 3 was studied in crosses between common wheat resistant genotypes Erik, Hadden, Red Chief, Glenlea, and 86ISMN 2137 and susceptible genotype 6B-365. Plants were inoculated under controlled environmental conditions at the two-leaf stage and disease rating was based on presence or absence of chlorosis. In all the resistant × susceptible crosses, F1 plants were resistant and the segregation of the F2 generation and F3 families indicated that a single dominant gene controlled resistance. Lack of segregation in a partial diallel series of crosses among the resistant genotypes tested with race 3␣indicated that the resistant genotypes possessed␣the same resistance gene. This resistance gene was effective against chlorosis induced by P.␣tritici-repentis races 1 and 3.  相似文献

Summary Inheritance of black leaf mold (BLM) (caused by Pseudocercospora fuligena) resistance was studied in four crosses involving two resistant Lycopersicon accessions (PI134417, L. hirsutum and PI254655, L. esculentum) and four susceptible Asian Vegetable Research and Development Center tomato lines (CLN657BC1F2-267-0-3-12-7, CL143-0-10-3-0-1-10, CLN698BC1F2-358-4-13 and CL5915-93D4-1-0-3). For each cross, six generations, i.e. P1, P2, F1, F2, BC1F1 and BC1F2 were evaluated following inoculations with isolate Pf-2 of P. fuligena. Chi-square analyses of the data based on the ratio of resistant to susceptible plants in the F2 in three of four crosses gave a good fit to a segregation ratio of 1 R : 15 S, and BC1F2 data in three of four crosses gave an acceptable fit to the segregation ratio of 1 R : 63 S. The results indicate that resistance to BLM may be conditioned by two recessive genes acting epistatically in both PI134417 and PI254655.  相似文献

Genetic control of somatic embryogenesis in cotton petiole callus cultures   总被引:8,自引:0,他引:8  
Summary Three commercial varieties (Acala SJ-5, Coker 312 and Paymaster 303) and three exotic accessions (T1, T25 and T169) of cotton (Gossypium hirsutum L.) were tested for ability to undergo somatic embryogenesis. Sections of split petiole were cultured on 3 media and evaluated for embryogenesis after 180 days. Embryogenic T25 and Coker 312 plants were selected and crossed in a diallel with non-embryogenic Acala SJ-5, Paymaster 303, T1 and T169 plants. F1, F2 and BC1 populations were generated and tested for embryogenesis on a medium of MS salts and vitamins (1962) plus (per liter) 4.0 mg NAA, 1.0 mg Kn, 30 g glucose, 100 mg myo-inositol, 2.0 g Gelrite and 0.75 g MgCl2. Segregation for both occurrence and magnitude of embryogenesis was observed, suggesting the action of more than one gene.  相似文献

The sesquiterpene zingiberene, present in leaf glandular trichomes, is reportedly responsible for the high level of arthropod resistance found in Lycopersicon hirsutum var. hirsutum. This paper reports on the inheritance of zingiberene contents and of the various types of glandular trichomes in the interspecific cross L. esculentum × Lycopersicon hirsutum var. hirsutum. Plants of L. esculentum ‘TOM-556’ (= P1), L. hirsutum var. hirsutum ‘PI-127826’(= P2), F1 (P1 × P2) and F2 (P1 ×P2) were evaluated for zingiberene contents and densitities of and glandular (types I, IV, VI and VII) trichomes. Broad sense heritabilities were high for all traits studied (0.678, 0.831, 0.996, 0.799 and 0.717 respectively for zingiberene and trichome types I, IV, VI, VII). There were significant positive genetic correlations between zingiberene contents and densities of trichomes types IV, VI and VII. Inheritance of zingiberenecontents can be explained mostly by the action of a single major locus, inwhich the allele from L. hirsutum that conditions high content is incompletely recessive over the allele from L. esculentum. Action of an incompleteley recessive allele in one major locus appears to be evident for densities of trichome types IV, VI and VII, but there is also evidence of the action of other epistatic loci for types IV and VI. F2 genotypes selected for high zingiberene levels showed higher levels of resistance to the silverleaf whitefly Bemisia argentifolii than L. esculentum ‘TOM-556’, levels that were comparable those found in L. hirsutum var. hirsutum ‘PI-127826’ and other whitefly resistant accessions. Summary Molecular mapping is a promising strategy for studying and understanding traits with complex genetic control, such as partial resistance to oat crown rust. The objectives of this research were to develop molecular maps from the progenies of the cross UFRGS7 (susceptible) × UFRGS910906 (partially resistant) and to identify QTLs (quantitative trait loci) associated to partial resistance to oat crown rust in two generations of that population.DNA of 86 genotypes of the F2 and 90 genotypes of the F6 UFRGS7 × UFRGS910906 population were used to generate AFLP markers. Molecular maps were constructed using Mapmaker Exp. 3.0 and QTLs for partial resistance to oat crown rust were identified with Mapmaker/QTL software. Five hundred and fifty seven markers in the F2 and 243 markers in the F6 generations were identified. The F2 map integrated 250 markers in 37 linkage groups. The F6 map integrated 86 markers in 17 linkage groups.Five QTLs were identified for partial resistance to oat crown rust in the F2 generation and three QTLs in the F6. The QTL identified on F6 through the PaaaMctt340 AFLP marker showed consistency across two environments and two generations (F4 and F6), and appear to have potential for marker-assisted selection in oat.  相似文献

The improvement of cotton fiber quality has become more important because of changes in spinning technology. Stable quantitative trait loci (QTLs) for fiber quality will enable molecular marker-assisted selection to improve fiber quality of future cotton cultivars. A simple sequence repeat (SSR) genetic linkage map consisting of 156 loci covering 1,024.4 cM was constructed using a series of recombinant inbred lines (RIL) developed from an F2 population of an Upland cotton (Gossypium hirsutum L.) cross 7235 × TM-1. Phenotypic data were collected at Nanjing and Guanyun County in 2002 and 2003 for 5 fiber quality and 6 yield traits. We found 25 major QTLs (LOD ≥ 3.0) and 28 putative QTLs (2.0 < LOD < 3.0) for fiber quality and yield components in two or four environments independently. Among the 25 QTLs with LOD ≥ 3, we found 4 QTLs with large effects on fiber quality and 7 QTLs with large effects on yield components. The most important chromosome D8 in the present study was densely populated with markers and QTLs, in which 36 SSR loci within a chromosomal region of 72.7 cM and 9 QTLs for 8 traits were detected.  相似文献

The Lycopersicon hirsutum var. hirsutum accession PI 127826 is recognized as a good source of resistance to arthropod pests due to the action of the allelochemical zimgiberene, a sesquiterpene present in its glandular trichomes. Five genotypes were selected from the F2 generation of the interspecific cross Lycopersicon esculentum ‘TOM-556’ × Lycopersicon hirsutum var. hirsutum ‘PI 127826’, based on their low levels (BPX-368-clone#56) or high levels(BPX-368-clone#92, BPX-368-clone#105,BPX-368-clone#179, BPX-368-clone#250) of zingiberene. The five F2 genotypes were tested for resistance to the South American tomato pinworm Tuta absolutaalong with accession L. esculentum ‘TOM-556’ (pinworm susceptible), and the accessions L. hirsutum var. hirsutum ‘PI 127826’ and L. pennellii ‘LA716’ (resistant). The F2 clones selected for high foliar zingiberene levels showed lower scores for leaflet lesion type(LLT), percent leaflets attacked (PLA) and overall plant damage (OPD) than the low zingiberene genotypes. The results indicated that zingiberene mediates resistance to the South American pinworm, based on feeding and on ovipositing deterrence, in populations derived from the interspecific cross between Lycopersicon. esculentum and Lycopersicon hirsutum var. hirsutum. Indirect selection for high foliar zingiberene content is suggested as an efficient technique for breeding tomatoes for resistance to the South American tomato pinworm. The objective of this study was to identify polymorphic molecular markers associated with partial resistance to coffee leaf rust, Hemileia vastarix. A segregating F 2 population derived from a cross between the susceptible Coffea arabica cv. Caturra and a C. canephora-introgressed Arabica line exhibiting high partial resistance was analyzed. Rust resistance measured as rust incidence (RI) and defoliation (DEF) was evaluated in field conditions in three consecutive years (2003–2005). During the 2003 season, which was characterized by favorable conditions for a rust epidemic, the F 2 plants exhibited different levels of resistance ranging from very susceptible (50.1% for DEF and 49.5% for RI) to highly partial resistance (9.1% for DEF and 3.7% for RI). Molecular analysis enabled identification of seven polymorphic markers (5 AFLP and 2 SSR) exhibiting significant association with partial resistance. Coexistence of resistance homozygous alleles (RR) at codominant SSR loci was correlated with high resistance. This study is the first attempt to develop PCR-based sequence specific markers linked to partial rust resistance in coffee.  相似文献

Soybean Cyst nematode (SCN) Heterodera glycines Ichinohe is the most serious pest of soybean [Glycine max (L.) Merr.] in the world and genetic resistance in soybean cultivars have been the most effective means of control. Nematode populations, however, are variable and have adapted to reproduce on resistant cultivars over time due mainly to the narrow genetic base of SCN resistance in G. max. The majority of the resistant cultivars trace to two soybean accessions. It is hoped that new sources of resistance might provide durable resistance. Soybean plant introductions PI 467312 and PI 507354, are unique because they provide resistance to several nematode populations, i.e. SCN HG types 0, 2.7, and 1.3.6.7 (corresponding to races 3, 5, and 14) and HG types 2.5.7, 0, and 2.7 (corresponding to races 1, 3, and 5), respectively. The genetic basis of SCN resistance in these PIs is not yet known. We have investigated the inheritance of resistance to SCN HG types 0, 2.7, and 1.3.6.7 (races 3, 5, and14) in PI467312 and the SCN resistance to SCN HG types 2.5.7 and 2.7 (races 1 and 5) in PI 507354. PI 467312 was crossed to ‘Marcus’, a susceptible cultivar to generate F1 hybrids, 196 random F2 individuals, and 196 F2:3 families (designated as Pop 467). PI 507354 and the cultivar Hutcheson, susceptible to all known SCN races, were crossed to generate F1 hybrids, 225 random F2 individuals and 225 F2:3 families (designated as Pop 507). The F2:3 families from each cross were evaluated for responses to the specific SCN HG types in the greenhouse. Chi-square (χ2) analyses showed resistance from PI 467312 to HG types 2.7, and 1.3.6.7 (races 5 and 14) in Pop 467 were conditioned by one dominant and two recessive genes (Rhg rhg rhg) and resistance to HG type 0 (race 3) was controlled by three recessive genes (rhg rhg rhg). The 225 F2:3 progenies in Pop 507 showed a segregation of 2:223 (R:S) for response to both HG types 2.5.7 and 2.7 (corresponding to races 1 and 5). The Chi-square analysis showed SCN resistance from PI 507354 fit a one dominant and 3 recessive gene model (Rhg rhg rhg rhg). This information will be useful to soybean breeders who use these sources to develop SCN resistant cultivars. The complex inheritance patterns determined for the two PIs are similar to the three and four gene models for other SCN resistance sources known to date.  相似文献

To identify DNA markers linked to a fertility restorer (Rf) genefor Ogura cytoplasmic male sterility in radish (Raphanus sativus L.),a non-radioactive, amplified fragment length polymorphism (AFLP) analysiswas performed on bulked DNA samples from male-sterile and male-fertileradishes. Ten male-fertile and 10 male-sterile plants selected arbitrarilyfrom an F2 population made by selfing of F1 plant from a crossbetween a male-sterile (`MS-Gensuke') plant and a restorer (`Comet') plantwere used as material. Using 32 AFLP primer pairs, one AFLP fragment(AFLP190) which is specific to the bulked DNA samples from male-fertileF2 plants was identified. AFLP190 was characterized by molecularcloning and nucleotide sequencing, and was converted to a sequence-taggedsite (STS) marker, STS190. A linkage analysis performed in 126individuals of two independent F2 populations showed tight linkageof STS190 to the Rf gene. The rate of recombination between themarker and Rf was estimated to be less than 1%, making STS1901.2 cM from the gene.  相似文献

We have constructed a linkage map of the rice brown planthopper (BPH)resistance gene, Bph1. RFLP and AFLP markers were selected by thebulked segregant analysis and used in the mapping study of 262 F2sthat were derived from a cross of `Tsukushibare', a susceptible japonica cultivar, and `Norin-PL3', an authentic japonicaBph1-introgression line. Twenty markers were mapped within a 28.9-cMregion containing the Bph1 locus on the long arm of rice chromosome12. Combining the result of segregation analysis of BPH resistance by themass seedling test and that of the markers, the Bph1 locus wasmapped within a 5.8-cM region between two flanking markers. The closestAFLP markers, em5814N and em2802N, was at 2.7 cM proximal to theBph1 locus. Together with the previously constructed high-resolutionmap of bph2 locating the locus at ca. 10 cM proximal to the Bph1 locus, this improved version of the linkage map would facilitatepyramiding these two important BPH resistance genes.  相似文献

Resistance to the southern root-knot nematode Meloidogyne incognita Chitwood would be an important attribute of lettuce Lactuca sativa L. cultivars adapted to both protected and field cultivation in tropical regions. `Grand Rapids' and a few other cultivars are reported to be resistant to this nematode. In this paper, we studied the inheritance of the resistant reaction of `Grand Rapids' (P2) in a cross with a standard nematode-susceptible cultivar Regma-71 (P1). F1(Regina-71 × Grand Rapids) and F2 seed were obtained, and inoculated along with the parental cultivars with different races of M. incognita to evaluate nematode resistance. Broad sense heritability estimates for the number of galls and of egg masses per root system, gall size and gall index were generally in the order of 0.5 or higher. Class distributions of these variables over generations P1, P2, F1 and F2 were in agreement with simulated theoretical distributions based on monogenic inheritance models. F3 families were obtained from randomly sampled F2 plants and tested for reaction to the nematode. The frequency ratio of homozygous resistant, segregating and homozygous susceptible F3 families did not differ from the 1:2:1 ratio expected from monogenic inheritance. M. incognita resistance appears to be under control of a single gene locus. The Grand Rapids allele (for which the symbol Me is proposed) is responsible for the resistant reaction, and shows high (though incomplete) penetrance, variable expressivity and predominantly additive gene action. To determine and utilize RAPD markers linked to resistance to downymildew incited by Peronospora manshurica in soybean, a resistantcultivar `AGS129' was crossed to a susceptible cultivar `Nakhon Sawan 1'(NS1). F2 and BC1 populations were advanced from the F1 and evaluatedfor resistance to the disease. 2-test demonstrated that the resistancewas controlled by a single dominant gene (Rpmx). Near-isogenic lines(NILs) and bulked segregant analysis (BSA) were used to identify RAPDmarkers linked to the gene. Six DNA bulks namely F5(R), F5(S),BC6F3(R), BC6F3(S), F2(R) and F2(S) were set up by pooling equalamount of DNA from 8 randomly selected plants of each disease responsetype. A total of 180 random sequence decamer oligonucleotide primerswere used for RAPD analysis. Primer OPH-02 (5 TCGGACGTGA 3 andOPP-10 (5 TCCCGCCTAC 3) generated OPH-021250 and OPP-10831fragments in donor parent and resistant bulks, but not in the recurrentparent and susceptible ones. Co-segregation analysis using 102 segregatingF2 progenies confirmed that both markers were linked to the Rpmxgene controlling downy mildew disease resistance with a genetic distance of4.9 cm and 23.1 cm, respectively. Marker OPH-021250 was presentin 13 of 16 resistant soybean cultivars and absent in susceptible cultivars,thus confirming a potential for MAS outside the mapping population.  相似文献

The genetics of resistance to Phomopsis stem blight caused by Diaporthe toxica Will., Highet, Gams & Sivasith. in narrow-leafed lupin (Lupinus angustifolius L.) was studied in crosses between resistant cv. Merrit, very resistant breeding line 75A:258 and susceptible cv. Unicrop. A non-destructive glasshouse infection test was developed to assess resistance in the F1, F2, selected F2-derived F3 (F2:3) families, and in selfed parent plants. The F1 of Unicrop × 75A:258 (and reciprocal cross) was very resistant, and the F2 segregated in a ratio of 3:1 (resistant: susceptible), which suggested the presence of a single dominant allele for resistance in 75A:258. In Merrit × Unicrop (and reciprocal), the F1 was moderately resistant, and the F2 segregated in a ratio of 3:1 (resistant: susceptible). Thus Merrit appeared to carry an incompletely dominant resistance allele for resistance. The F1 of Merrit × 75A:258 (and reciprocal) was very resistant and the F2 segregated in a ratio of 15:1 (resistant: susceptible), which supported the existence of independently segregating resistance alleles for resistance in 75A:258 and Merrit. Alleles at loci for early flowering (Ku) and speckled seeds (for which we propose the symbol Spk) segregated normally and independently of the resistance alleles. Resistant F2 plants gave rise to uniformly resistant or segregating F2:3 families, whereas susceptible F2 plants gave rise only to susceptible F2:3 families. However, the variation in resistance in the F2 and some F2:3 families of crosses involving 75A:258, from moderately to extremely resistant, was greater than that expected by chance or environmental variation. We propose the symbols Phr1 to describe the dominant resistance allele in 75A:258, and Phr2 for the incompletely dominant resistance allele in Merrit. Phr1 appears to be epistatic to Phr2, and expression of Phr1 may be altered by independently segregating modifier allele(s). In this research, we used two sets of cotton aneuploid (G. hirsutum × G. tomentosum and G. hirsutum × G. barbadense) plants to locate AFLP markers to chromosomes using deletion analysis method. Thirty-eight primer combinations were used to generate 608 polymorphic AFLP markers. A total of 98 AFLP markers were assigned to 22 different cotton chromosomes or chromosome arms. Of those assigned markers, 63.3% were assigned to A genome and 36.7% were assigned to D genome. A low rate (14.3%) of common markers were found between those assigned AFLP markers with the AFLP markers from an intraspecific cross population developed previous in our lab. Based on the 16 common markers, we were able to associate the 13 linkage groups previously identified in our lab to eight chromosomes. Further research will be carried out by using SSR markers with known location to associate unassigned linkage groups to chromosomes.  相似文献

Sunflower downy mildew caused by Plasmopara halstedii is an important disease of sunflower capable of causing losses of more than 80% of production. Races 100, 300, 310, 330, 710, 703, 730 and770 of the fungus have been identified in Spain. Race 703, of high virulence, has been identified frequently in the northeast, while race 310 seems to occur over the south, the main sunflower growing region of the country. Oil sunflower lines RHA-274 and DM4 were studied for their resistance to races 310(RHA-274 and DM4) and 703 (DM4). In each cross, only one plant of the resistant parent was crossed to the inbred susceptible line HA-89 (or cmsHA-89).Plants from F2 and backcross(BC1F1 to susceptible parent)generations were evaluated for fungal sporulation on true leaves and/or cotyledons. The resistant-to-susceptible ratios obtained in the F2 and BC1F1 progenies from the crosses cmsHA-89 × RHA-274 and HA-89 × DM4suggested that one major gene in each line is responsible for resistance to race 703.The segregations of the progenies of the cross HA-89 × DM4 inoculated with race 703also fitted the ratios 1:1 and 3:1 (for BC1F1 and F2, respectively)corresponding to control of resistance by a single dominant gene. In RHA-274, the gene for resistance to race 310 was designated Pl 9, whereas Pl v is tentatively proposed to designate the gene in DM4 responsible for resistance to races310 and 703. Summary The levels of the naturally occurring insecticide 2-tridecadone (2TD) were measured in leaves of Lycopersicon hirsutum f. glabratum, L. esculentum, the interspecific F1 hybrid, the F2 and backeross generations. The wild species contains 50 times more 2TD than the cultivated tomato and the frequency distribution of the substance indicates that there is dominance for low levels of the compound. The genotype of F2 plants was determined with respect to 6 codominant isozyme markers and 4 dominant/recessive markers. Significant associations were detected between 5 of the marker genes and the level of 2TD. We interpret these results as implying linkage between marker genes and genes that control the level of 2TD. The behavior of the gene for the determinant growth habit of the plants suggests that it has a pleiotropic effect on the level of 2TD.  相似文献

In carrot, two codominant sequence-tagged site (STS) markers, flanking in tight linkage the Meloidogyne javanica resistance (Mj-1) locus, were employed to investigate the association between expression of resistance and locus dosage. Phenotypic expression of homozygous resistant (R); heterozygous; and homozygous susceptible (S) individuals in an F2 population of 396 F2 plants from ‘Brasília-1252’ (R) × ‘B6274’ (S) was estimated for three resistance criteria: total egg production per plant (TEP), egg production per gram of fibrous root (EPG) and root gall index (RGI). The homozygous resistant class had average values of 403.9 for TEP; 147.5 for EPG and 0.8 for RGI. The heterozygous class had 1,673; 477.3; and 0.16 whereas the homozygous susceptible class had 68,604; 11,877; and 2.54, respectively. The dominance ratio (d/a) indicated that genomic region(s) derived from the resistant parent encompass genetic factor(s) with almost complete dominance for RGI (d/a = 0.93–0.94) and incomplete dominance for transformed (TEP)0.25 and (EPG)0.25 (d/a = 0.63–0.65). Broad sense heritabilities were high varying from 72.9% for (EPG)0.25 to 86.0% for RGI. Narrow sense heritability values ranged from 55.9% for RGI to 64.3%for (TEP)0.25. Highly significant orthogonal contrasts were observed between homozygous resistant vs. heterozygous for (TEP)0.25 and (EPG)0.25. Marker-assisted selection could greatly facilitate the incorporation of the Mj-1 allele in both male-fertile and male-sterile counterpart lines in order to obtain F1 hybrids with the most effective levels of resistance. Summary An intergeneric hybrid (2n=38) between Gossypium hirsutum L. (2n=52) × Hibiscus panduraeformis Burm. (2n=24) was obtained by pollinating about 2000 flower buds of G. hirsutum var. Gregg Male Sterile with pollen from H. panduraeformis. The F1 hybrid was intermediate in plant habit, but possessed gossypol glands and nectaries on the leaves, bolls containing seeds with fuzz and lint as dominant characters of G. hirsutum. Flowers with yellow corolla and anthers; purple petal spot, profuse growth of epidermal hairs on all plant parts including the boll sutures, and jassid tolerance were dominant characters of H. panduraeformis. The partial fertility of the F1 indicated the possibilities of combining jassid and drought tolerance of H. panduraeformis with the desired economic characters of G. hirsutum for rainfed cultivation.The F1 hybrid showed various meiotic irreguarities and about 40% pollen sterility. Formation of the normal bivalents occurred quite frequently suggesting a close relationship between the parental species. The sterility observed in the hybrid may be due to small structural differences between the chromosomes of the two genera and meiotic abnormalities.  相似文献

Summary Inheritance of resistance to tomato leaf curl virus (TLCV) was studied in the progenies derived from interspecific crosses between TLCV resistant Lycopersicon hirsutum f. glabratum line B 6013 and five susceptible cultivars (HS 101, HS 102, HS 110, Pusa Ruby and Punjab Chhuhara) of L. esculentum. P1, P2, F1, F2, B1 and B2 progenies of the five crosses were artificially inoculated with local strains of TLCV by means of the vector whitefly, Bemisia tabaci (Genn.). and the disease reaction was studied in all the crosses. Reaction of parents, F1, F2 and backcrosses suggests that resistance derived from L. hirsutum f. glabratum B 6013 is based on two epistatic genes, one from the wild parent and one from the cultivated one, resulting in a 13:3 segragation in the F2.  相似文献
