The objectives of this study were to determine 1) the ability of charcoal-extracted bovine follicular fluid (bFF) to suppress endogenous follicle stimulating hormone (FSH) at various stages of the estrous cycle and 2) the effects of suppression of FSH on luteal function and lengths of the current and subsequent estrous cycles. Twenty-six mature ewes were assigned randomly to receive 5 ml of either bFF or saline, subcutaneously, at 8-h intervals on d 1 through 5 (bFF n = 6; saline n = 3), d 6 through 10 (bFF n = 6; saline n = 3) or d 11 through 15 (bFF n = 6; saline n = 2) of the estrous cycle (d 0 = estrus). Blood was collected daily beginning at estrus and continued until the third estrus (two estrous cycles) or 40 d; more frequent samples were collected 2 h prior to initiation of treatment (0600), hourly for the first 8 h of treatment, then every 4 h until 0800 on the first day after treatment, and finally at 1600 and 2400 on that day. Plasma concentrations of FSH were lower (P less than .001) in bFF-treated than in saline-treated ewes. Treatment with bFF reduced (P less than .05) plasma concentrations of progesterone during the current but not during the subsequent estrous cycle. Treatment with bFF did not affect plasma concentrations of estradiol-17 beta. Administration of bFF on d 11 through 15 of the estrous cycle lengthened the interval from the decline in progesterone to estrus and the inter-estrous interval by approximately 3 and 4 d, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Two experiments were designed to examine whether hormonal profiles were related to luteal life span in pluriparous postpartum anestrous beef cows. Cows (Exp. 1, n = 34; Exp. 2, n = 23) received norgestomet (N) for 9 d or served as controls (C). Each cow received 1,000 IU human chorionic gonadotropin (hCG) 48 h after removal of N (d 0). Blood samples collected every 15 min for 8 h on d -5, 3 and 5 (Exp. 1) or on d -10 and -1 (Exp. 2) were assayed for luteinizing hormone (LH) and follicle stimulating hormone (FSH). Cortisol was determined in hourly samples collected on d -5 and in samples collected every 2 min during suckling on the same day (Exp. 1). Concentrations of 15-keto-13,14-dihydro-PGF2 alpha (PGFM) were determined in samples collected at 15-min intervals for 2 h on d -5, 3, 5 and 10 (Exp. 1). Estradiol-17 beta was measured in samples collected on d -5 (Exp. 1) or on d -10 and -1 (Exp. 2). Life span of induced corpora lutea was longer (P less than .05) in N than C cows. Percentages of N cows in which corpora lutea, formed in response to hCG, exhibited a normal life span were 83% on farm 1 and 25% on farm 2 (Exp. 1), and 90% (Exp. 2), compared with 0% in C cows. Concentrations of FSH were not affected by N but were lower (P less than .05) on d -5 in cows on farm 2 (.6 +/- .1 ng/ml) than in cows on farm 1 (.8 +/- .1 ng/ml). On d -5, a treatment X farm interaction (P less than .05) for mean LH was observed and frequency of pulses of LH was higher (P less than .01) in N than C cows (2.7 +/- .4 vs. .8 +/- .8 pulses/8 h). Neither cortisol nor PGFM was affected by N. Estradiol was increased in d -1 (6.1 +/- .5 vs 2.6 +/- .8 pg/ml; P less than .01) by N. It is suggested that pre-treatment with N enhanced life span of induced corpora lutea, in part, by influencing secretion of LH and development of follicles, but a threshold concentration of FSH was required for N to exert this effect.  相似文献

A study was done to test whether ovulatory follicles destined to form subfunctional corpora lutea differed from normal ovulatory follicles in steroidogenic function. Twenty-five ewes were treated with prostaglandin F2 alpha on d 11 of the estrous cycle, then unilaterally ovariectomized before (n = 13) or after (n = 12) the surge of luteinizing hormone (LH) at the induced estrus to collect "control" follicles, which would have produced normal corpora lutea. In 15 ewes, the second ovary was removed 63 to 84 h later to collect "treated" follicles before (n = 7) or after (n = 8) the second expected surge of LH. Five ewes (control) were allowed to ovulate from the remaining ovary at first estrus and another five (treated) at the second estrus (3 to 4 d later). Treated ewes had lower serum progesterone than control ewes during the ensuing cycle (P less than .05). Treated follicles contained less estradiol in the theca (4.4 +/- .6 vs 10.0 +/- 2.5 ng; P less than .05), less androstenedione (.1 +/- .1 vs 1.0 +/- .2 ng) and estradiol (.5 +/- .1 vs 2.9 +/- 2.2 ng) in the granulosa (P less than .05) and less progesterone in the follicular fluid (.8 +/- .4 vs 3.3 +/- .8 ng; P less than .05) than control follicles, when removed before the surge of LH. Follicles removed after the surge of LH did not differ. In conclusion, ovulatory follicles with low steroidogenic function became corpora lutea that secreted lower-than-normal quantities of progesterone.  相似文献

Morphine (M), an opioid agonist, was administered to postpartum (PP) Angus cows to investigate opioid modulation of gonadotropin secretion. In Exp. 1, eight PP cows (36.9 +/- 2.3 d) received either M (1 mg/kg; n = 4) or saline solution (S) (n = 4) via i.v. injection 36 h after calf removal. Morphine decreased (P less than .01) the number of serum LH pulses (3.0 +/- 1.1 pre- vs .3 +/- .3 post-pulses/h) and, compared with pretreatment values (3.3 mg/ml), decreased (P less than .05) mean LH at 105 min (2.1 ng/ml) through 270 min 1.9 ng/ml +/- .4). Serum prolactin (PRL) increased (P less than .01) following M from 16.4 ng/ml to a peak of 59.3 ng/ml (+/- 3.9). Serum FSH concentrations were unaffected. In Exp. 2, M (.31 mg/kg i.v. injection followed by .15 mg/(kg.h) infusion; n = 6) or S (n = 6) treatments were given for 7 h beginning 36 h after calf removal. Serum LH was similar between groups during the pretreatment and the first 6 h of infusion, but M decreased (P less than .001) the number of serum LH pulses (.44 +/- .09 vs .06 +/- .04 pulses/h). Morphine increased (P less than .05) serum PRL. It is concluded that M differentially modulated gonadotropin secretion in the cow such that PRL increased, LH decreased and FSH was unchanged.  相似文献

Adult Suffolk ewes (n = 14) were treated on d 10 of the estrous cycle with anti-bovine luteinizing hormone (LH) antiserum. Control ewes (n = 10) were treated with normal horse serum. Estrous behavior and the number of corpora lutea and ovarian follicles were examined at the subsequent estrous cycle. Daily plasma concentrations of progesterone (P4), follicle stimulating hormone (FSH) and estradiol were determined before and after treatment. Ewes treated with antiserum had a higher (P less than .05) ovulation rate (2.7 +/- .2) than did controls (2.1 +/- .1). No differences were found in the numbers of large (greater than 5 mm) or small (less than 5 mm) follicles between treatment groups. Estrus was delayed (P less than .025) approximately .6 d/in ewes treated with antisera. Immunoreactive FSH increased (P less than .05) within 1 d after treatment and remained higher than the controls for 5 d. Peak estradiol concentrations occurred on d 17 for treated ewes compared with peak concentrations on d 15 or 16 for control ewes. The P4 concentrations were generally less (P less than .025) in treated ewes throughout the luteal phase of the treatment cycle. These data demonstrate that ovulation rate is increased in ewes treated with LH antiserum. The marked increase in plasma FSH suggests a possible mechanism whereby ovulation rate is enhanced.  相似文献

A study was conducted to determine the effect of charcoal-extracted, bovine follicular fluid (CFF) on plasma follicle stimulating hormone (FSH) and luteinizing hormone (LH) concentrations, the interval from luteolysis to estrus, and subsequent luteal function in heifers. Fifteen Angus, Simmental and Hereford heifers were allotted by age, weight and breed to a control (C, n = 8) or a CFF (n = 7) group. Heifers received injections of saline or CFF (iv, 8 ml/injection) every 12 h from d 1 (d 0 estrus) through d 5 of the estrous cycle. On d 6, each heifer was injected (im) with 25 mg of prostaglandin F2 alpha (PGF2 alpha). Blood samples were collected every 12 h by venipuncture starting just before the first saline or CFF injection and continuing until estrus. Thereafter, blood samples were collected every other day during the subsequent estrous cycle and assayed for FSH, LH, estradiol-17 beta and progesterone by radioimmunoassay. Injections of CFF had no effect (P greater than .05) on circulating FSH or LH concentrations from d 1 to 5 relative to the C group; however, there was a transient rise (P less than .05) in FSH concentrations 24 h following cessation of CFF injections. This transient rise in FSH was not immediately followed by an increase in plasma estradiol-17 beta concentrations. Luteinizing hormone (LH) and ACTH concentrations were measured in plasma from 7 cows to determine whether ACTH secretion changes with the phase of the estrous cycle, and to determine whether any ACTH peaks are associated with LH peaks. Blood was collected every 5 minutes for 190 minutes during the luteal and follicular phases of the estrous cycle. Radioimmunoassays were used to measure ACTH and LH in plasma. Mean concentration of ACTH in all cows did not differ significantly between luteal (35.1 +/- 8.0 pg/ml) and follicular (37.5 +/- 9.4 pg/ml) phases of the estrous cycle. Mean concentration of luteal-phase LH of all cows (2.0 +/- 1.1 ng/ml) was significantly (P less than 0.01) lower than mean concentration of follicular-phase LH (5.4 +/- 1.6 ng/ml). Frequency of peaks in ACTH concentration was low during the sampling period. Mean number of luteal-phase ACTH peaks (0.29 +/- 0.49) was not significantly different from that of follicular-phase samples (0.43 +/- 0.530). Unlike ACTH, mean frequency of LH peaks was significantly (P less than 0.05) higher in plasma from cows in the follicular phase of the estrous cycle (2.9 +/- 0.7), compared with that from cows in the luteal phase (0.29 +/- 0.49).  相似文献

Twenty mature, lactating Hereford-cross cows were used to determine the effect of phlorizin-induced hypoglycemia on gonadotropin secretion following prostaglandin-induced luteolysis. Cows were 43 to 108 d postpartum and had a functional corpus luteum (CL) at the start of infusion treatment (d 1). Infusions consisted of either saline (control) or 3 g/d of phlorizin infused continuously from the time of prostaglandin injection at 1000 on d 1 until 0800 on d 5. Blood samples were collected for determination of plasma concentrations of insulin, glucose and free fatty acids (FFA) and for serum concentrations of luteinizing hormone (LH), follicle-stimulating hormone (FSH) and progesterone. Plasma concentrations of insulin (P less than .05) and glucose (P less than .05) were lower, whereas FFA concentrations increased (day X treatment, P less than .05) over the infusion period in phlorizin-treated cows compared with saline-infused controls. Mean serum concentrations of LH (1.17 +/- .10 vs 1.53 +/- .20 ng/ml; P less than .05) and LH pulse amplitude (1.69 +/- .14 vs 2.47 +/- .37 ng/ml; P less than .10) were lower in phlorizin-infused compared with saline-infused cows during the 0 to 24-h period immediately preceding the ovulatory gonadotropin surge. The FSH pulse frequency increased (.33 +/- .11 to .55 +/- .12 pulses/h) in saline-infused cows, but decreased (.61 +/- .10 to .41 +/- .11 pulses/h) in phlorizin-infused cows before the gonadotropin surge. Other characteristics of gonadotropin secretion were similar among phlorizin-infused and saline-infused cows. Two experiments were conducted to determine whether oral administration of melatonin alters the onset of puberty in gilts during naturally increasing or decreasing daylength. In Exp. 1, 20 crossbred prepubertal gilts weighing 77.5 +/- .5 kg at 171.8 +/- 1.0 d of age were assigned randomly to receive either a daily oral dose of 3 mg of melatonin (MEL) or ethanol vehicle (ETH) at 1530 from August 31 to December 1, 1987 (decreasing daylength). Gilts were exposed to mature boars for 20 min thrice weekly and blood samples were collected twice weekly. Serum concentrations of progesterone were used to establish age at puberty and length of estrous cycle. In Exp. 2, 20 crossbred prepubertal gilts weighing 67.7 +/- .7 kg at 143.8 +/- 1.1 d of age received either MEL or ETH treatment from February 1 to May 15, 1988 (increasing daylength). Age of puberty was less in gilts that received MEL than in gilts that received ETH in both Exp. 1 (198 +/- 3 vs 228 +/- 7 d; P less than .01) and Exp. 2 (183.8 +/- 2.7 d vs 194.3 +/- 3.3 d; P less than .05). Gilts that received MEL reached puberty at a lighter weight than gilts that received ETH in Exp. 1 (95.6 +/- 2.1 vs 112.4 +/- 3.9 kg; P less than .01) and Exp. 2 (88.1 +/- 1.5 vs 96.0 +/- 1.8 kg; P less than .01). To determine influences of insulin and body condition on follicular growth, prepuberal gilts (n = 16) treated with pregnant mare's serum gonadotropin (PMSG) were used in a 2 X 2 factorial experiment with main effects of insulin (0 or .4 IU/kg every 12 h beginning at 1800 on the day before PMSG) and backfat depth (moderate, 25 +/- .8; high, 32 +/- .7 mm; P less than .0001). Body weights were similar. Blood sampling was at 6-h intervals for analyses of LH, FSH, growth hormone (GH), glucagon, cortisol, insulin, insulin-like growth factor-I (IGF-I), plasma urea nitrogen (PUN), nonesterified fatty acids (NEFA), testosterone, estradiol-17 beta, and progesterone. Ovaries were removed 75 h after PMSG treatment, and visible small (less than or equal to 3 mm), medium (4 to 6 mm), large (greater than or equal to 7 mm), and macroscopically atretic follicles were counted. Administration of insulin increased IGF-I in fluid of medium follicles (108.8 vs 60.7 ng/ml; SEM = 13.3; P less than .05). Neither insulin nor fatness affected hCG binding by granulosa cells (12.5 +/- 1.6 ng/10(6) cells) or numbers of large (16.7 +/- 2.6) and medium (10.4 +/- 2.3) follicles. However, insulin increased the number of small follicles (58.9 vs 29.9; SEM = 9.7; P less than .05) and reduced the number of atretic follicles (3.8 vs 11.3; SEM = 1.1; P less than .05). The predominant effect of insulin on reducing number of atretic follicles was in the small size class (.6 vs 6.9; SEM = .6, P less than .01). Follicular fluid estradiol and progesterone were not affected by treatments; however, testosterone concentrations in large follicles were lower in gilts with higher backfat (32.5 vs 59.9 ng/ml; SEM = 4.0; P less than .05). Systemic LH, FSH, glucagon, cortisol, PUN, NEFA, estradiol, and testosterone were not affected by insulin or level of feeding. However, GH was lower in gilts that had higher backfat (overall average of 3.2 vs 2.8 ng/ml; SEM = .1; P less than .05). Insulin reduced atresia and altered intrafollicular IGF-I independently of body condition and without sustained effects on other hormones.  相似文献

In three experiments, we examined the effects of suckling, progestogen treatment, hysterectomy or exogenous gonadotropin releasing hormone (GnRH) on ovarian function in autumn-lambing, postpartum ewes. In each experiment, GnRH was injected on approximately d 25 postpartum. Suckling reduced (P less than .01) GnRH-induced release of luteinizing hormone (LH) but not of follicle stimulating hormone (FSH), and reduced (P less than .05) the proportion of ewes that developed corpora lutea in response to GnRH. Suckling had no effect on duration (8.8 d) of GnRH-induced luteal phases. Progestogen prior to GnRH increased (P less than .01) the duration of the first luteal phase (10.1 vs 7.6 d; progestogen-treated ewes vs control ewes), but progestogen did not affect the release of LH or FSH. Progestogen treatment did not alter the interval from parturition to the first detected estrus (42.6 d). The concentration of 13,14-dihydro-15-keto-PGF2 alpha (PGFM) just after lambing was greater than 400 pg/ml of jugular plasma, but concentrations of PGFM declined thereafter. Hysterectomy the day after lambing hastened (P less than .001) the decline in concentrations of PGFM, indicating that prostaglandins from the postpartum uterus probably caused the high concentrations of PGFM in jugular plasma. Hysterectomy reduced (P less than .05) the interval from parturition to detectable luteal function (19.6 vs 25.3 d) and enhanced (P less than .001) luteal production of progesterone. This study of autumn-lambing ewes indicates that the uterus has a negative effect on ovarian function and that suckling and progestogen affect ovarian response to GnRH.  相似文献

In Exp. 1, 45 fine-wool ewes received (sc) either 0, 50 or 100 mg epostane (3-beta-hydroxysteroid dehydrogenase inhibitor) to examine effects on return to estrus and conception rates. Treatments were imposed on d 10 of an estrous cycle (estrus = d 0) and jugular blood samples were collected once daily on d 8 and 9, twice daily on d 10 through 13 and once daily from d 14 until first post-treatment estrus or d 20 (15 ewes/treatment). Intensive samples were obtained hourly for 6 h after treatment (d 10; five ewes/treatment). Serum progesterone (P4) before treatment was similar among groups; but by 2 h after treatment, epostane-treated ewes had lower (P less than .10) values than controls. By 6 h post-treatment, serum P4 in ewes receiving 50 (1.0 ng/ml) and 100 (.9 ng/ml) mg epostane was well below control values (3.2 ng/ml). By 7 d after treatment, 93.3% of ewes treated with 50 mg epostane had recycled compared with 66.7 and 33.3% of those receiving 100 and 0 mg, respectively (P less than .10). Similarly, 93.3, 53.3 and 26.7% of ewes receiving 50, 100 or 0 mg epostane, respectively, lambed to breeding within 7 d of treatment (P less than .05). Overall conception rates during a 34-d breeding season were similar among groups. In order to determine the role of follicle-stimulating hormone (FSH) on the resumption of ovarian function in cows early postpartum (PP), bovine follicular fluid (FF) was used to selectively suppress concentrations of FSH. Calves were removed from all cows within 24 hr of birth. Follicular fluid that was treated with charcoal to remove steroids (15 ml; n = 14) or serum (S) from an ovariectomized cow (15 ml, n = 14) was injected i.m. twice daily from days 1 to 10 PP. Blood samples were collected before each injection and frequent samples (every 15 min for 6 hr) taken on days 5 and 10 PP. Eight cows from each group (FF and S) were slaughtered on the morning of day 11 PP and pituitaries and ovaries collected. The remaining cows (n = 6) were observed for estrus. Treatment with FF delayed follicular growth (P less than 0.01), as evidenced by the largest follicle per cow observed at time of slaughter (3.6 +/- 0.42 vs 11.5 +/- 1.77 mm dia; FF vs S). The intervals from parturition to first estrus (P less than 0.11) and to first progesterone rise (25.3 +/- 1.97 vs 18.0 +/- 3.62 d; P less than 0.06) tended to be delayed by treatment with FF vs S. Many of the cows treated with S ovulated by day 10 PP, they were divided retrospectively into those that had ovulated by (n = 9) or after (n = 5) day 10 PP for analysis. Cows treated with FF had lower (P less than 0.05) and less variable (P less than 0.01) serum FSH concentrations while levels of luteinizing hormone (LH) tended (P less than 0.08) to be greater on days 5 and 10 PP. Follicular fluid decreased levels of FSH (P less than 0.001), but not LH (P less than 0.15), in the samples obtained twice daily compared to S-treated cows that did not ovulate by day 10 PP. Anterior pituitaries were dissociated, and cells from each cow were cultured in order to ascertain whether treatment with FF in vivo would affect gonadotropin secretion in vitro. Estradiol-17 beta (E) was incubated with pituitary cells to determine the effect of E on gonadotropin secretion from cells of PP cows, and to ascertain whether treatment with FF in vivo and with E in vitro would interact to affect secretion of FSH and LH in culture. Fine-wool ewes received for 2 yr a complete pelleted basal diet (11% protein) or the basal diet fortified with 3.5% cottonseed meal (CSM; 12% protein) or gamma-irradiated (1 megarad) dried solids (SS; 12% protein) from primary (undigested) sewage (Las Cruces, New Mexico, municipal sewage). Five ewes fed each diet were sampled to determine Ag, Ca, Cd, Co, Cr, Cu, Fe, K, Mg, Mn, Na, Ni, P, Pb and Zn in blood, milk and tissues. Tissues and blood were sampled at slaughter 40 d after weaning of lambs. Mean whole blood mineral concentrations were similar (P less than .05) among treatments 3 d postpartum; however, at 42 d after lambing (mean +/- SE) both basal- (54 +/- 2 micrograms/ml) and sewage-fed (54 +/- 2 micrograms/ml) ewes had elevated (P less than .05) blood Ca compared with ewes fed CSM (46 +/- 2 micrograms/ml). No biologically important differences were detected in the concentrations of elements in milk. Ewes fed SS had lower (P less than .05) blood Fe than animals in the other groups. Sewage-fed ewes also had higher (P less than .05) liver Fe (1,092 +/- 100 micrograms/g) than basal-fed ewes (626 +/- 100 micrograms/g); whereas Fe in CSM-fed ewes (873 +/- 100 micrograms/g) was similar to both. Basal-fed animals had 1.1- to 1.3 times more (P less than .05) liver Mg and two- to threefold higher liver Na than CSM or SS. The hypothesis that ovulation in response to short-term (48 h) calf removal (CR) is dependent on the developmental stage of the dominant follicle was tested in two studies. The objective of Exp. 1 was to characterize the fate of a dominant follicle following 48-h CR on d 2, 4, or 8 of a postpartum follicular wave. Ovaries of 61 beef cows were examined daily by transrectal ultrasonography starting at d 20 to 21 postpartum. Treatments were no CR (n = 14) and CR on d 2 (n = 12), 4 (n = 16), or 8 (n = 10) of first detected follicular wave. Percentage of cows that ovulated a dominant follicle following treatment was not different among groups (P = 0.62). Maximum size of dominant follicles was larger in cows that ovulated (P = 0.002) than in cows that did not ovulate. The objectives of Exp. 2 were 1) to determine whether a follicular wave could be synchronized in anestrous cows following injection of 1 mg of estradiol benzoate (EB) and 200 mg of progesterone (P4; EB + P4); 2) to characterize the fate of dominant follicles following 48-h CR at three stages of a synchronized follicular wave; and 3) to determine whether estrous cycles of normal length followed ovulation in cows pretreated with EB + P4. Ovaries of 50 anestrous beef cows were examined daily as in Exp. 1. Treatments were sesame oil (SO) injected (i.m.) on d 25 postpartum and no CR (n = 9); EB + P4 and no CR (n = 9); EB + P4 and CR on 6 (n = 12), 8 (n = 9), or 12 (n = 11) d after injection. The EB and P4 injections were given on d 25 postpartum. Variability in day of emergence of subsequent follicular waves was lower in cows receiving EB + P4 than in SO-injected cows (P < 0.05). The percentage of cows that ovulated was not different (P = 0.16), but CR increased the percentage of cows that ovulated when groups that received EB + P4 were compared to the EB + P4 group that did not have CR (53.1 vs 11.1%, respectively; P < 0.05). Maximum diameter of dominant follicles was larger (P = 0.05) in ovulatory follicles. The luteal phase was longer (P < 0.03) in cows receiving EB + P4 injection (10.6 +/- 1.2 d) than in cows receiving SO (4.4 +/- 2.2 d). Cyclic gilts from Control (C, randomly selected, n = 11) and Relax Select (RS, nine generations of selection for increased ovulation rate followed by seven generations of relaxed or random selection, n = 9) lines of the University of Nebraska Gene Pool population (derived from 14 different breeds) were utilized to characterize differences in gonadotropic and ovarian steroid hormones during preovulatory and postovulatory phases of the estrous cycle. Blood samples were collected during four periods (0500, 1100, 1700 and 2300) daily beginning 2 d prior to anticipated estrus (d -2, d 18 of a 20-d estrous cycle), and continuing through d 4 postestrus (d 0 = 1st of standing estrus). Sampling within a period consisted of five blood samples at 15-min intervals. All plasma samples were analyzed for concentrations of follicle stimulating hormone (FSH) and luteinizing hormone (LH). Neither mean LH nor peak concentration of LH during the preovulatory surge differed between genetic lines (P greater than .10). Concentrations of FSH increased faster (line X period, P less than .05) and tended (P less than .1) to peak at a higher concentration in RS (.88 ng/ml) than in C (.54 ng/ml) gilts (P less than .05) during the 12 h preceding the FSH and LH preovulatory peaks. The second FSH surge began approximately 24 h after the preovulatory FSH peak. Peak FSH concentrations were observed at 42 h in both lines (1.46 vs 1.74 ng/ml for C and RS gilts, respectively). The higher FSH concentration in RS gilts established during the preovulatory surge was maintained through the second FSH surge (P less than .01). No line differences were detected in plasma concentrations of estradiol-17 beta and progesterone.  相似文献

Mature gilts classified by low (12 to 16 corpora lutea [CL], n = 6) or high (17 to 26 CL, n = 5) ovulation rate (OR) were compared for plasma follicle-stimulating hormone (FSH), luteinizing hormone (LH), progesterone, estradiol-17beta, and inhibin during an estrous cycle. Gilts were checked for estrus at 8-h intervals beginning on d 18. Blood samples were collected at 8-h intervals beginning on d 18 of the third estrous cycle and continued for one complete estrous cycle. Analysis for FSH and LH was performed on samples collected at 8-h intervals and for ovarian hormones on samples collected at 24-h intervals. The data were standardized to the peak of LH at fourth (d 0) and fifth estrus for the follicular phase and analyzed in discrete periods during the periovulatory (-1, 0, +1 d relative to LH peak), early-luteal (d 1 to 5), mid-luteal (d 6 to 10), late-luteal (11 to 15), periluteolytic (-1, 0, +1 d relative to progesterone decline), and follicular (5 d prior to fifth estrus) phases of the estrous cycle. The number of CL during the sampling estrous cycle was greater (P < 0.005) for the high vs low OR gilts (18.8 vs 14.3) and again (P < 0.001) in the cycle subsequent to hormone measurement (20.9 vs 14.7). For high-OR gilts, FSH was greater during the ovulatory period (P = 0.002), the mid- (P < 0.05) and late-luteal phases (P = 0.01), and tended to be elevated during the early-luteal (P = 0.06), but not the luteolytic or follicular periods. LH was greater in high-OR gilts during the ovulatory period (P < 0.005), but not at other periods during the cycle. In high-OR gilts, progesterone was greater in the mid, late, and ovulatory phases (P < 0.005), but not in the follicular, ovulatory, and early-luteal phases. Concentrations of estradiol-17beta were not different between OR groups during the cycle. Inhibin was greater for the high OR group (P < 0.005) during the early, mid, late, luteolytic, and follicular phases (P < 0.001). The duration of the follicular phase (from last baseline estrogen value to the LH peak) was 6.5 +/- 0.5 d and was not affected by OR group. These results indicate that elevated concentrations of both FSH and LH are associated with increased ovulation rate during the ovulatory phase, but that only elevated FSH during much of the luteal phase is associated with increased ovulation rate. Of the ovarian hormones, both inhibin and progesterone are highly related to greater ovulation rates. These findings could aid in understanding how ovulation rate is controlled in pigs.  相似文献

The interaction among exogenous estradiol-17 beta, naloxone and gonadotropin releasing hormone (GnRH) in the control of luteinizing hormone (LH) secretion was studied in intact postpartum ewes nursing their offspring. One-half of 30 fall-lambing ewes were implanted subcutaneously with an estradiol-17 beta containing Silastic capsule between postpartum d 1 and 12 which doubled their serum concentrations of estradiol (16.0 +/- .1 vs 8.4 +/- .1 pg/ml). Blood samples were collected from implanted and non-implanted ewes at 15-min intervals for 5 h on d 3, 8, 13, 20 and 28 postpartum. Pre-injection samples were collected for 1 h, and ewes were injected with saline, naloxone (NAL;1 mg/kg) or GnRH (100 micrograms/ewe). When averaged across all days and implant groups, serum LH in the three post-NAL samples was higher (P less than .05) than in the three pre-NAL samples (3.6 +/- 1.2 vs .6 +/- .2 ng/ml). Post-GnRH concentrations of serum LH were lower (P less than .05) in estradiol-implanted ewes than in non-implanted ewes on d 8 and 13, but there were no differences in any LH characteristics on d 20 and 28 after implant removal on d 12. In non-implanted ewes, serum LH responses to GnRH increased (P less than .05) eightfold from d 3 (3.8 +/- 1.4 ng/ml) to d 8 (31.6 +/- 1.4 ng/ml), remained elevated through d 20, but declined by d 28 (10.8 +/- 1.4 ng/ml).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Experiments were conducted to examine the effects of exogenous GnRH and LH on serum concentrations of progesterone (P4) in the ewe. Ewes in Exp. 1 and 2 were laparotomized on d 2 of an estrous cycle and ewes with corpora lutea (CL) in both ovaries were unilaterally ovariectomized. Ewes with CL in one ovary only were not ovariectomized. While they were anesthetized, ewes (n = 5) were injected with 25 micrograms GnRH (Exp. 1) or 50 ng GnRH (Exp. 2) into the artery supplying the ovary bearing the CL. Control ewes (n = 5 in each experiment) were injected similarly with saline. In Exp. 3, six ewes were injected i.v. (jugular) on d 2 with 100 micrograms oLH (t = 0) and 50 micrograms oLH at 15, 30 and 45 min; six control ewes were injected similarly with saline. Jugular blood was collected from all ewes at frequent intervals after treatment for LH analysis and on alternate days of the cycle through d 10 or 11 for P4 analysis. Treatment with 25 micrograms GnRH increased serum concentrations of LH at 15, 30, 45 and 60 min postinjection (P less than .001) and reduced serum concentrations of P4 on d 7 through 11 (treatment x day interaction; P less than .05). Four experiments assessed changes in serum IGF-I under various physiologic conditions in postpartum cows. In Exp. 1, anestrous suckled cows (n = 25) were infused for 6 d with either saline or glucose at two different infusion rates. In Exp. 2, anestrous cows (n = 29) received either a saline (weaned and suckled controls) or 3 g/d phlorizin (weaned phlorizin) infusion for 3 d. Calves from the weaned groups were removed from 15 h before and throughout infusions. In Exp. 3, cycling suckled cows (n = 20) received prostaglandin F2 alpha (PGF2 alpha) when the 5-d saline or phlorizin infusion began. In Exp. 4, suckled cows (n = 20) had ad libitum access to feed or received 50% of control feed consumption from 30 to 40 d postpartum. Increasing glucose availability (Exp. 1) increased (P less than .05) serum IGF-I by 30 to 35%. IGF-I remained stable after weaning (Exp. 2) in phlorizin-infused cows (128.8 +/- 12.7 ng/ml), but increased (P less than .05) by 3 d after calf removal in weaned control cows (152.2 +/- 7.5 ng/ml). IGF-I also remained stable in phlorizin-infused cows following PGF2 alpha injection (Exp. 3), but increased in control cows by 2 d after PGF2 alpha (156.8 +/- 18.3 on d 2 vs. 133.7 +/- 9.8 ng/ml pre-injection; P less than .05) and remained elevated (P less than .05) during the periovulatory period.
