Impacts of chlorpyriphos (2, 4, 6 ppm) on the profiles of antioxidant (catalase) and anaerobic (lactate dehydrogenase) enzymes and other macromolecular contents (DNA, RNA, protein) of various tissues of the freshwater fish, Heteropneustes fossilis were studied. Chlorpyriphos significantly decreased the specific activity of catalase (CAT) and lactate dehydrogenase (LDH) in the brain, liver, gill and skeletal muscle of the fish. The reduction in specific activity might be due to the binding of chlorpyriphos or its metabolites with the enzyme molecules or affecting the synthesis and/or degradation of the enzymes. Like enzymes, the DNA, RNA and protein contents decreased in the brain, liver, gill and skeletal muscle of the fish as a function of increase in chlorpyriphos concentrations (2-6 ppm). The chlorpyriphos-induced reduction in these biochemical constituents might be because of alteration in their turnover (synthesis/degradation) in different tissues. The maximum effects on CAT, LDH, DNA, RNA and protein were obtained in response to 6 ppm chlorpyriphos. The present findings suggest chlorpyriphos concentration related impairment in antioxidative, anaerobic and protein synthesizing capacity of the fish. Therefore, the use of higher concentrations of chlorpyriphos should be avoided to protect the health of economically important freshwater food fish.  相似文献

Pesticides, such as herbicides can affect the metabolic and toxicological parameters on fish. For this reason, an experiment was carried out with the objective of to evaluate the effects of commercial formulations of clomazone and propanil herbicides on acetylcholinesterase (AChE), thiobarbituric acid-reactive substances (TBARS), catalase (CAT) and metabolic parameters in teleost fish (Leporinus obtusidens). Fish were exposed during 90 days to field measured concentration of the herbicides clomazone and propanil (376 and 1644 μg/L, respectively) on rice paddy water. Specific AChE activity in the brain and muscle decreased and TBARS levels decreased in brain, muscle and liver tissues. Liver catalase decreased after exposure to both herbicides. Metabolic parameters in the liver and white muscle showed different changes after exposure to both herbicides. In summary, the results showed that clomazone and propanil affects toxicological and metabolic parameters of piavas. These results suggest that environmentally relevant herbicides concentrations are toxic to Leporinus obtusidens.  相似文献

The Cyprinus carpio fingerlings on exposure to lethal (1 mg/L) and sub lethal concentrations (0.066 mg/L) of sodium cyanide showed inhibition in the activity of catalase. The disruption of catalase activity in freshwater fish, C. carpio is demonstrated in the present study using UV–visible spectrophotometer at 240 nm using hydrogen peroxide as a substrate. It suggests toxic effects of sodium cyanide and consequent accumulation of hydrogen peroxide in the functionally different tissues namely, liver, gill, muscle and brain. This might lead to cellular damages, and create widespread physiological disturbance. The results suggest that catalase activity can be a good diagnostic tool for sodium cyanide toxicity in biomonitoring programme.  相似文献

The effect of carbofuran, an organo-carbamate pesticide, upon the level of protein as well as the activity of lactate dehydrogenase (EC 1.1.1.28, LDH) was studied by exposing the teleost fish, Clarias batrachus, to different subacute con-centrations (0·01 and 0·02 mg litre-1) for 96 h and 15 days. The results showed a drastic decrease in the protein content in different body organs of the fish. The pesticide also caused a significant decrease in the level of activity of LDH in different body tissues of the fish, the effect being more pronounced in the gills, muscle, brain and liver than in kidney and heart. The decrease in protein content and the activity of LDH in fish tissues was more marked at the higher concentration of the pesticide for the longer duration of treatment. The results suggested that carbofuran has an effect at very low concentration (compared to its LC50 value) possibly at the level of protein metabolism, and also inhibits the activity of LDH, the terminal glycolytic enzyme. © 1998 SCI  相似文献

Alterations in the levels of hemoglobin, total plasma proteins, glucose, and lactic acid in the blood; glycogen and lactic acid content of liver and white skeletal muscle; and the activities of lactate dehydrogenase, pyruvate dehydrogenase, and succinate dehydrogenase in liver, kidney, intestine, brain, gills, and muscles were examined in the fresh-water snake-head fish, Channa punctatus, after exposure to a sublethal concentration (25 μg/liter) of quinalphos for 60 and 120 days. Hemoglobin, plasma protein, glucose, and lactic acid decreased in pesticide-exposed fish. The glycogen content of the liver and muscles increased but lactic acid decreased. Lactate dehydrogenase activity decreased in all six tissues. Pyruvate dehydrogenase activity of liver, kidney, gill, and muscle decreased, but the enzyme activity was elevated in intestine and brain. In intestine, succinate dehydrogenase activity was elevated, and in the remaining five tissues the enzyme activity was significantly reduced. The present study showed that formation of glycogen and its breakdown was impaired in the liver, and aerobic oxidation of nutrients was adversely affected in quinalphos-exposed fish.  相似文献

This study describes the subacute and subchronic effects of two plant growth regulators (PGRs) [abcisic acid (ABA) and gibberellic acid (GA3)] on serum marker enzymes [aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatine phosphokinase (CPK) and lactate dehydrogenase (LDH), γ-glutamil transpeptidase (GGT)], antioxidant defense systems [reduced glutathione (GSH), glutathione reductase (GR), glutathione peroxidase (GPx), superoxide dismutase (SOD), glutathione-S-transferase (GST) and catalase (CAT)] and lipid peroxidation level (Malondialdehyde = MDA) in various tissues of rats. Rats (Sprague-Dawley albino) were exposed to 75 ppm (parts per million) of ABA and GA3. Seventy-five parts per million of PGRs as drinking water was administered orally ad libitum for 25 and 50 days continuously. The PGRs treatments caused different effect on the serum marker enzymes, antioxidant defense systems and the content of MDA in comparison to those of control rats. Results show that ABA caused a significant decrease in serum LDH and CPK activity with both periods. Also, GA3 significantly decreased serum AST, CPK, and LDH activity with subacute and decreased serum ALT, CPK, LDH, and GGT treated with subchronic periods. The lipid peroxidation end product MDA significantly increased in the erythrocyte, liver, brain, and muscle of rats treated with both the period of GA3 without significantly change in the erythrocyte and muscle of rats treated with the subacute period of ABA. The GSH levels were significantly depleted in the erythrocyte and brain of rats treated with both the period of GA3 without any change in the erythrocyte, liver, brain, and muscle of rats treated with both the period of ABA. Also GSH levels in the muscle significantly depleted with the subchronic period of GA3. Antioxidant enzyme activities such as SOD significantly decreased in the erythrocyte, liver and brain tissues but increased in the muscle tissue of rats treated with both the periods of GA3. Meanwhile, SOD significantly decreased in liver and brain, and increased in muscle of rats treated with both the period of ABA. While CAT significantly decreased in the all tissues of rats treated with both the period of GA3, decreased in the liver and muscle of rats treated with both the periods of ABA too. On the other hand, the ancillary enzyme GPx and GR activity in the erythrocytes, liver, brain and muscle were either significantly depleted or not changed with two periods of PGRs. The drug metabolizing enzyme GST activity significantly decreased in the brain of rats treated with subacute period of PGRs but increased in the erythrocytes of rats treated with subacute period of GA3. As a conclusion, ABA and GA3 had significantly increased the activity of hepatic damage enzymes. Also the rats resisted to oxidative stress via antioxidant mechanism. However, the antioxidant mechanism could not prevent the increases in lipid peroxidation in rat’s tissues. These data, along with changes, suggest that PGRs produced substantial systemic organ toxicity in the erythrocyte, liver, brain, and muscle during the period of a 25-day subacute and 50-day subchronic exposure.  相似文献

The euryhaline fish, Oreochromis mossambicus was exposed to sub-lethal concentration (0.017 mg L−1) of a novel phosphorothionate, 2-butenoic acid-3-(diethoxy phosphinothionyl) ethyl ester (RPR-V) for 30 days and allowed to recover for 7 days. Important biomarker enzymes were assayed in plasma, brain, gill, liver, kidney, and muscle during exposure tenures of day-3, -7, -15, -30, and also at 7 days (withdrawal) after stopping treatment. Acetylcholinesterase (AChE) activities of brain, gill, and muscle were strongly inhibited by 67, 75, and 66%, respectively, on day-30. Exposure (time) dependent increases in alanine aminotransferase (ALAT), and aspartate aminotransferase (ASAT), acid phosphatase (AcP), and alkaline phosphatase (AkP), activities in plasma and kidney; AcP and AkP activities in gill were noticed. However, significant decrease in ALAT, ASAT, AcP, and AkP activities in liver was observed. The depletion of glycogen was observed in liver, brain, and gill tissues, an indication of typical stress related response of the fish with pesticide. A significant increase in lactate dehydrogenase (LDH) activity in gill and brain was observed and decreased in liver and muscle, indicating tissue damage and muscular harm. Depletion of glutathione (GSH) was observed in the above tissues, there by enhancing the lipid peroxidation resulting in cell damage. The induction in hepatic glutathione-S-transferase (GST) levels indicates the protection against the toxicity of xenobiotic-induced lipid peroxidation. There was a significant recovery in all the above biochemical parameters, in all the tissues of fish after a recovery period of 7 days. These results revealed that RPR-V affects the intermediary metabolism of O. mossambicus and the increase of biomarker enzymes in plasma, might be due to the necrosis of liver.  相似文献

The purpose of this study was to evaluate oxidative stress and neurotoxic potential of organophosphorus (OP) insecticide diazinon in the sentinel freshwater fish, Oreochromis niloticus. Antioxidant and acetylcholinesterase (AChE) enzyme activities and malondialdehyde (MDA) and protein levels were measured spectrophotometrically in gill, kidney, alimentary tract, and muscle tissues of fish treated with sub-lethal diazinon concentrations for 1, 7, 15, and 30 days. Dose-dependent inhibitions of AChE were observed in all the experimental fish. On the contrary of alimentary tract, MDA levels were elevated in kidney and muscle and gill was not affected. AChE and MDA levels intercorrelated in kidney and muscle tissues. Diazinon had increased superoxide dismutase (SOD) activities in all the tissues, while kidney was the most affected tissue. Tissue-specific alterations were observed on catalase (CAT) and glutathione peroxidase (GPx) activities; however, the activities were not changed in gill and muscle tissues for GPx and in gill, muscle, and kidney tissues for CAT. Protein levels decreased in kidney, muscle, and alimentary tract, while increased in gill and alimentary tract in 15 days. With respect to these results, diazinon has oxidative and neurotoxic potentials in O. niloticus. Observed changes with diazinon treatment were generally tissue-specific and dose-dependent.  相似文献

This study aims to investigate the effects of the trichloroacetic acid (TCA) on serum marker enzymes [aspartate aminotransferase (AST), alanin aminotransferase (ALT), creatine phosphokinase (CPK), acid phosphatase (ACP), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH)], antioxidant defense systems [Reduced glutathione (GSH), glutathione reductase (GR), superoxide dismutase (SOD), glutathione-S-transferase (GST) and catalase (CAT)] and lipid peroxidation content (Malondialdehyde, MDA) in various tissues of rats. TCA (2000 ppm) as drinking water was administered orally to rats (Sprague-Dawley albino) ad libitum for 50 days continuously. TCA treatments caused different effects on the serum marker enzymes, antioxidant defense systems and the MDA content in experimented rats compared to controls. Results showed that TCA caused a significant increase in serum AST, ALT, CPK and ACP activity. The lipid peroxidation end product MDA slightly increased in the erythrocytes, liver and kidney of rats treated with TCA, whereas did not change in the brain. In addition, antioxidant enzyme activity such as CAT and SOD significantly increased in the brain, liver and kidney tissues of TCA induced group whereas the ancillary enzyme GR and the drug metabolizing enzyme GST activity did not significantly change in the all tissues. The observations presented led us to conclude that the administration of subchronic TCA promotes lipid peroxidation content, elevates tissue damage serum marker enzymes and fluctuates in the antioxidative systems in rats. Also the rats resisted to oxidative stress via antioxidant mechanism but the antioxidant mechanism could not prevent the increases in lipid peroxidation in rat’s tissues. These data, along with the determined changes suggest that TCA produced substantial systemic organ toxicity in the erythrocyte, liver, brain and kidney during the period of a 50-day subchronic exposure.  相似文献

In this study, fish Rhamdia quelen, were exposed to different concentrations of herbicide clomazone: 0.0 (control), 0.45 and 0.91 mg L−1. After exposure for 8 days to herbicide, fish were transferred to clean water for a recovery period (8 days). Oxidative stress indicators such as thiobarbituric acid reactive substances (TBARS) levels and protein carbonyl content, as well as antioxidant defenses, such as catalase (CAT), superoxide dismutase (SOD), glutathione S-transferase (GST), ascorbic acid and non-protein thiols levels were studied, using the liver, brain and muscle tissues. Herbicide exposure increased TBARS in muscle and in liver at higher concentration. In liver protein carbonylation increased and catalase activity did not change in fish exposed to herbicide. SOD enhanced in liver at concentration of 0.91 mg L−1. GST, ascorbic acid and non-protein thiols levels increase at both concentrations. At the end of the recovery period the most of the parameters recovered whereas GST and ascorbic acid remain elevated. The present study demonstrates the occurrence of disorders in antioxidant parameters and importance in the assessment of the potential risk of herbicides as clomazone on fish species.  相似文献

Present study aimed mainly to assess oxidative stress pesticides such as methyl parathion (MP) and diazinon, which are widely used insecticides and contaminate aquatic ecosystems, on certain biomarkers in various tissues of rainbowtrout (Oncorhynchcus mykiss). Biomarkers selected for stress monitoring were malondialdehyde (MDA) and antioxidant defense system (ADS) mainly reduced glutathione (GSH), glutathione reductase (GR), peroxidase (GSH-PX), transferase (GST) and superoxidedismutase (SOD) activities in the liver, gills and muscle of fishes exposed to 0.5 and 1 ppm dosages of MP and diazinon for 24, 48 and 72 h. According to these results, after the administrations of MP and diazinon promote MDA content in some of the tissues of fishes treated with both dosages of MP and diazinon. With regard to the ADS, GSH-Px, GST, SOD, GR activities and GSH levels fluctuated after 24, 48 and 72 h in all the treatment groups compared with controls. Collective results demonstrated that exposure of fish to pesticides induced an increase in MDA joined with fluctuated ADS. This may reflect the potential role of these parameters as useful biomarkers for assessment of water pollution.  相似文献

In May 1972, 0.309 ppm methoxychlor black fly larvicide was applied in a single test on the North Saskatchewan River. Eight to nine days later residues of 0.05-0.10 ppm methoxychlor occurred in sand 21-22 km downstream from the point of injection. Methoxychlor was not detected in water, insect larvae, shellfish, or muscle tissues of three fish species on the same sampling date. Perhaps because of relatively high oil content in goldeye fish, methoxychlor residues in muscle tissues were 1.0-1.5 ppm in 8 percent of those sampled, 0.21-0.99 in 21 percent, and 0.02-0.20 in 37 percent. In 34 percent of the goldeye fish no residues were detected. Goldeye and other fish collected before or 17 weeks after this injection did not contain detectable levels of methoxychlor. River water in two samples of the injected slug of water collected 6.5 km downstream from the point of injection contained 0.14 and 0.16 ppm methoxychlor. The suspended solids filtered from these sample contained 40 and 47 percent of this methoxychlor (437 and 892 ppm, respectively). Thus methoxychlor may act selectively against filter-feeding species, especially black fly larvae.  相似文献

建立了采用高效液相色谱-二极管阵列检测器(HPLC-DAD)测定水、土壤和黄瓜中噻唑锌残留的方法。在碱性条件下先将噻唑锌转化为噻二唑(AMT),采用外标法通过测定噻二唑的量来进行噻唑锌的定量分析。样品在40℃恒温振荡条件下,依次经Na2S转化及乙腈提取;过滤后调节混合液pH值至3,经乙酸乙酯液-液分配后,用HPLC-DAD及BDS Hypersil-C18色谱柱,以V(乙腈):V(0.1%乙酸)=10:90为流动相,在313 nm波长下测定样品中的噻唑锌残留。结果表明:噻二唑在0.10~10 mg/L、噻唑锌在0.20~5.0 mg/L的质量浓度范围内线性关系良好(R2 > 0.999 5),噻二唑的检出限(LOD)为0.05 mg/L。在0.2、1和5 mg/L添加水平下,噻唑锌在水中的平均回收率为100%~110%,相对标准偏差(RSD)为0.90%~6.4%;在0.05、0.5和5 mg/kg添加水平下,噻唑锌在土壤中的平均回收率为81%~98%,RSD为0.70%~2.8%;在0.05、0.5和2 mg/kg添加水平下,噻唑锌在黄瓜中的平均回收率为95%~102%,RSD为1.3%~4.2%。噻唑锌在水、黄瓜和土壤中的定量限(LOQ)分别为0.03 mg/L、0.05 mg/kg和0.05 mg/kg。本方法简单、准确、可靠,能满足农药残留分析的要求。  相似文献

To study the behavior and mutation of Ralstonia solanacearum in Solanum toxicarium, which is resistant to bacterial wilt, S. toxicarium was grown in aseptic culture and inoculated with R. solanacearum. Although 60%–80% of the inoculated plants were wilting after 2 to 3 days, most wilted plants had recovered by 20 days after inoculation. The pathogen was reisolated from over 98% of inoculated plant stems, but the percentage of recovery decreased the closer the isolation sites were toward the upper stem sections. Three colony types, characterized as fluidal white, nonfluidal red, and a mixture of fluidal white and nonfluidal red, were reisolated from the stems. Nonfluidal red colonies were less virulent on tomato plants than fluidal white colonies.  相似文献

采用不同浓度NaCl溶液(100 mmol/L、200 mmol/L)胁迫处理甜高粱幼苗,测定了叶片中叶绿素含量、脯氨酸含量及三种保护酶活性等生理指标。结果表明:100 mmol/L和200 mmol/L处理的甜高粱幼苗质膜相对透性、脯氨酸和丙二醛含量升高;可溶性蛋白和叶绿素含量降低;保护酶系统中叶片超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性均升高,且在100 mmol/L浓度处理下达到最大,而过氧化物酶(POD)活性呈先升高再降低趋势。NaCl胁迫对甜高粱幼苗的一些生理特性产生一定影响。  相似文献

The impact of acute and sublethal toxicity of a synthetic organochlorine pesticide lindane on some haematological and biochemical parameters of a freshwater fish Cyprinus carpio was estimated. The median lethal concentration of lindane for 24 h was 0.38 ppm. During acute treatment (24 h), hemoglobin (Hb), hematocrit (Hct) and erythrocyte (RBC) values were decreased, whereas leucocyte (WBC) count increased in the pesticide treated fish. The hematological indices like mean cellular volume (MCV), mean cellular hemoglobin (MCH) and mean cellular hemoglobin concentration (MCHC) were decreased when compared to control group. Biochemical profiles like plasma glucose and protein levels were increased in lindane exposed fish showing a percent increase of 50.36 and 3.98 at the end of 24 h treatment. However, glycogen content in liver and muscle were decreased showing a percent decrease of 21.70 and 1.74. In sublethal treatment (1/10th of LC50 24 h value, 0.038 ppm), RBC count was decreased whereas WBC count increased in the pesticide treated fish throughout the study period (25 days). Hb and Hct values were decreased up to 10th day and after that recovered showing a significant increase in the rest of the study period. Similarly, a biphasic response was observed in the value of MCV, MCH and MCHC. Plasma glucose level was significantly increased while plasma protein level decreased throughout the study period. Further, glycogen level in muscle and liver were showed a mixed trend. In the present study, the pesticide lindane caused alterations on haematological and biochemical parameters of C. carpio and these alterations can be used as non specific biomarkers in pesticide contaminated aquatic ecosystem.  相似文献

In this study, the acute toxicity of the organophosphorous pesticide (OP) dichlorvos and both in vitro and in vivo effects of dichlorvos on cholinesterase (ChE) activity of the European sea bass (Dicentrarchus labrax) were investigated. The characterisation of ChE and the “normal” range of activity in brain and muscle of non-exposed fish were determined in a first phase of the study. Acetylthiocholine was the substrate preferred of both brain and muscle ChE. Eserine sulphate and BW284C51 significantly inhibited the brain and muscle enzyme activity at low concentrations (μM range). Iso-OMPA had a significant effect in muscle, but not in brain tissue. These results suggest that acetylcholinesterase (AChE) is the predominant ChE form in brain tissue. In contrast, both acetylcholinesterase and butyrylcholinesterase seem to exist in muscle. Using acetylthiocholine as substrate, the “normal” range of fingerling head and muscle ChE were 58.05±2.11 and 118.03±8.67 U/mg protein, respectively. Corresponding values for juveniles were 43.32±4.42 and 19.44±2.44 U/mg protein for brain and muscle, respectively. Dichlorvos significantly inhibited the activity of ChE in the selected tissues, both in vitro and in vivo conditions. Differences in ChE sensitivity were found in relation to the age of the fish and the tissue analysed. The present study also showed that fingerlings of the European sea bass are relatively resistant to in vivo acute (96 h) dichlorvos exposure to concentrations between 0.125 and 1 mg/L, being able to tolerate high percentages of head ChE inhibition (37% and 76%) without lethal effects.  相似文献

A case of heavy infestation on tubers of three potato (Solanum tuberosum L) cultivars (Nicola, Spunta and Sieglinde) by the cyst nematodeGlobodera pallida Stone, 1973 was observed in Southern Italy on early producing potato cultivars in spring. Tubers were covered by white females and cysts but no other external symptoms were detectible on their surfaces. Detailed observations were directed to study the response of phellem and secondary cortex tissues induced by the expansion of syncytia during nematode feeding activity. The micro details of histological changes observed on serially sectioned infected tissues are described and illustrated. The nematode's feeding activity was confined to the outer part of the tuber while the host-parasite relationships were similar to those induced by the same nematode in the feeder roots.  相似文献

The activity levels of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) enzymes were estimated in liver, muscle and brain tissues of the fish, Sarotherodon mossambicus (Peters), which had been exposed to sub-lethal (3 mg litre−1) and lethal (25 mg litre−1) concentrations of the carbamate insecticide carbaryl. Based on the results obtained, the changes in GOT and GPT levels in liver, muscle and brain following different periods of sub-lethal and lethal carbaryl exposures suggested that S mossambicus showed adaptive elevation in the activity levels of the two aminotransferase enzymes in the tissues, thereby probably aiding gluconeogenesis through transamination of glucogenic amino acids to meet the energy demand under carbaryl toxicity. © 1999 Society of Chemical Industry  相似文献

The effects of exposure of human erythrocytes to different concentrations of Roundup Ultra 360 SL and its active compound glyphosate were studied. We studied hemolysis after 1, 5, and 24 h incubation; lipid peroxidation, hemoglobin oxidation, the level of reduced glutathione, and the activity of catalase after 1 h. Human erythrocytes were incubated with 100-1500 ppm (100 μg/ml erythrocytes at 5% hematocrite) Roundup Ultra 360 SL and glyphosate. We have found that after 1 h of incubation only Roundup Ultra 360 SL increased the level of methemoglobin, products of lipid peroxidation at 500 ppm and hemolysis at 1500 ppm [Curr. Top. Biophys. 26 (2002) 245], while its active compound glyphosate increased the level of methemoglobin and the level of lipid peroxidation at much higher dose—1000 ppm. At the same time hemolysis was observed to only at the highest dose of glyphosate (1500 ppm) and the longest time of incubation (24 h). Both Roundup Ultra 360 SL and glyphosate did not cause statistically significant changes in the level of GSH, but increased the activity of catalase. Roundup Ultra 360 SL provokes more changes in the function of erythrocytes than its active substance glyphosate, which is probably a result of the properties of additives. Taking into account the limited accumulation of Roundup Ultra 360 SL and glyphosate in the organism as well as the fact that the threshold doses which caused changes in erythrocytes for Roundup Ultra 360 SL were only 500 and 1000 ppm for glyphosate, one may conclude that this pesticide is safe towards human erythrocytes.  相似文献
