During the period 1982-1983, just under 800 samples of agricultural commodities, comprising cereals, compound feeds, hay, and silage, were examined for molds and mycotoxins. Aflatoxin B1 showed the highest incidence rate; it occurred in over 27% of all samples analyzed, the highest levels being found in peanut meal at 1500 ppb. Other mycotoxins detected were patulin and a number of trichothecene toxins at incidence rates in all commodities of 5.6 and 3.1%, respectively. The commodities at highest risk were oil seeds, excluding soya bean; the latter was found to be fairly free from contamination with mycotoxins. The most prevalent fungi were Aspergillus flavus and parasiticus, which were found in over 22% of all samples, whereas Penicillium spp. showed the lowest incidence of genera, specifically identified in 8.3% of all samples examined. This latter finding explains in part the low incidence of Penicillium mycotoxins.  相似文献

Maize (Zea mays) and wheat (Triticum aestivum) collected in the foothills of the Nepal Himalaya Mountains were analyzed for Fusarium species and mycotoxins: fumonisins, nivalenol (NIV), and deoxynivalenol (DON). Predominant species were Gibberella fujikuroi mating population A (F. moniliforme) in maize and F. graminearum in maize and wheat; G. fujikuroi mating population D (F. proliferatum), F. acuminatum, F. avenaceum, F. chlamydosporum, F. equiseti, F. oxysporum, F. semitectum, and F. torulosum were also present. Strains of G. fujikuroi mating population A produced fumonisins, and strains of F. graminearum produced NIV or DON. By immunoassay or high-performance liquid chromatography, fumonisins were >1000 ng/g in 22% of 74 maize samples. By immunoassay or fluorometry, NIV and DON were >1000 ng/g in 16% of maize samples but were not detected in wheat. Fumonisins and DON were not eliminated by traditional fermentation for producing maize beer, but Nepalese rural and urban women were able to detoxify contaminated maize by hand-sorting visibly diseased kernels.  相似文献

Near-infrared (NIR) spectroscopy is a practical spectroscopic procedure for the detection of organic compounds in matter. It is particularly useful because of its nondestructiveness, accuracy, rapid response, and easy operation. This work assesses the applicability of NIR for the rapid identification of micotoxigenic fungi and their toxic metabolites produced in naturally and artificially contaminated products. Two hundred and eighty maize samples were collected both from naturally contaminated maize crops grown in 16 areas in north-central Italy and from ears artificially inoculated with Fusarium verticillioides. All samples were analyzed for fungi infection, ergosterol, and fumonisin B1 content. The results obtained indicated that NIR could accurately predict the incidence of kernels infected by fungi, and by F. verticillioides in particular, as well as the quantity of ergosterol and fumonisin B1 in the meal. The statistics of the calibration and of the cross-validation for mold infection and for ergosterol and fumonisin B1 contents were significant. The best predictive ability for the percentage of global fungal infection and F. verticillioides was obtained using a calibration model utilizing maize kernels (r2 = 0.75 and SECV = 7.43) and maize meals (r2 = 0.79 and SECV = 10.95), respectively. This predictive performance was confirmed by the scatter plot of measured F. verticillioides infection versus NIR-predicted values in maize kernel samples (r2 = 0.80). The NIR methodology can be applied for monitoring mold contamination in postharvest maize, in particular F. verticilliodes and fumonisin presence, to distinguish contaminated lots from clean ones, and to avoid cross-contamination with other material during storage and may become a powerful tool for monitoring the safety of the food supply.  相似文献

Fumonisins are polyketide-derived mycotoxins produced by the maize pathogen Fusarium verticillioides. Previous analyses identified naturally occurring variants of the fungus that are deficient in fumonisin C-10 hydroxylation or that do not produce any fumonisins. In the current study, gene deletion and genetic complementation analyses localized the C-10 hydroxylation deficiency to a cytochrome P450 monooxygenase gene in the fumonisin biosynthetic gene (FUM) cluster. Sequence analysis indicated that the hydroxylation deficiency resulted from a single nucleotide insertion that caused a frame shift in the coding region of the gene. Genetic complementation localized the fumonisin-nonproduction phenotype to the polyketide synthase gene in the FUM cluster, and sequence analysis indicated that the nonproduction phenotype resulted from a nucleotide substitution, which introduced a premature stop codon in the coding region. These results provide the first direct evidence that altered fumonisin production phenotypes of naturally occurring F. verticillioides variants can result from single point mutations in the FUM cluster.  相似文献

Four small tortilla plants were visited in Cameron County, Texas, where observations were made on their production methods. Samples of liquids and solids were collected at each stage of the nixtamalization process, and the pH was recorded. Samples were analyzed for fumonisin B(1) (FB(1)) using an immunoaffinity column/HPLC method chosen for its sensitivity for FB(1). It was found that production methods were highly variable among the producers visited, with major differences particularly in the amount of lime added and boiling times. As reported by others working in Mexico and Central America, FB(1) was found in some tortillas. This led to studies of the effects of the various recipes and across a greater range of initial FB(1) concentration/damaged corn than has typically been reported. Five initial concentrations of FB(1) were tested using irradiated corn kernels inoculated with Fusarium verticillioides MRC 826 as the source of FB(1). The amount of FB(1) detected in the masa and tortillas decreased as the concentration of Ca(OH)(2) increased, and boiling time had no apparent effect. Unexpectedly, as the initial concentrations were increased in the corn prior to nixtamalization, greater percentage reductions in FB(1) were observed.  相似文献

A multianalyte method was developed to identify and quantitate 26 mycotoxins simultaneously in maize silage by means of ultrahigh-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). The extraction and cleanup procedure consists of two extraction steps followed by purification on a Waters Oasis HLB column. The method developed was validated with the requirements of Commission Decision 2002/657/EC taken into account. The limit of detection and quantitation ranges were 5-348 and 11-695 ng/g, respectively. Apparent recovery varied between 61 and 116%, whereas repeatability and reproducibility were within the ranges of 3-45 and 5-49%, respectively. The method developed was successfully applied for maize silage samples taken at the cutting surface and 1 m behind that surface. Mainly Fusarium toxins (beauvericin, deoxynivalenol, enniatins, fumonisins, fusaric acid, and zearalenone) were detected, but postharvest toxins such as mycophenolic acid and roquefortine C were identified as well.  相似文献

The paper summaries information obtained through an initial survey undertaken to identify those avian populations which qualify for inclusion in a South African Red Data Book, and whose conservation status is in need of special attention. One hundred and one species' populations are included in the preliminary list. Nine of the 101 species are endemic to South Africa. There is a predominance of forest and wetland species, of birds of prey, and of mainly tropical species occurring as intrusions in the north east of South Africa. Many of the species listed are peripheral to South Africa and are widespread and abundant extra-limitally. Twenty species' populations which require urgent conservation attention are listed. As far as is known, no avian species has become extinct during historical times in South Africa, but two are at present endangered, i.e. threatened with immediate extinction.  相似文献

Unintended effects of genetic modification on chemical composition of Bt maize leaf litter may have impacts on its decomposition. In most agricultural systems in South Africa, maize litter is either left on the soil surface or incorporated into the soil during tillage. A litterbag experiment, using leaf litter of three maize hybrids (DKC80-12B, DKC80-10 and DKC6-125), was carried out at the University of Fort Hare Research Farm, South Africa, to determine the effects of genetic modification on decomposition of maize leaf litter when left on the soil surface under field conditions between July and November, the normal fallow period, in 2008. Another litterbag experiment was conducted at the University of Fort Hare Research Farm and Zanyokwe Irrigation Scheme, South Africa, using leaf ~itter of two maize hybrids genetically modified with the erylAb gene (MONS10), DKC75-15B and PAN6Q-3OSB, and their corresponding near isolines, CRN3505 and PAN6Q-121. The degradation of CrylAb protein in the litter, both surface-applied and soil-incorporated, was also investigated. Decomposition of Bt maize litter was similar to that of non-Bt maize litter both when applied on the surface and when incorporated into soil. Soil-incorporated litter, as well as its CrylAb protein, decomposed faster than that applied on the surface. The leaf litter C:N ratios of PAN6Q-308B and PAN6Q-121 were similar throughout the study, whereas those of DKC75-15B and CRN3505 declined by similar amounts during a 12-week period. These findings suggested that decomposition of leaf litter of Bt maize, with the MON810 event, was not affected by maize genetic modification, and that the CrylAb protein broke down together with plant leaf litter during the winter fallow regardless of whether the litter was applied on the soil surface or incorporated into soil.  相似文献

The fungus Fusarium verticillioides infects maize and produces fumonisins, inhibitors of ceramide synthase. Seeds of the cultivar Silver Queen were inoculated with fumonisin-producing or non-fumonisin-producing strains of F. verticillioides. Leaf lesion incidence and severity of effects on root and stalk growth were significantly correlated with fumonisin in roots and disruption of sphingolipid metabolism in roots. Uninoculated seeds grown in soil watered with solutions of fumonisin B1 exhibited above-ground symptoms indicative of F. verticillioides-induced seedling disease and dose-dependent reduction in root mass that was inversely correlated with fumonisin B1, sphingoid bases, and sphingoid base 1-phosphates in roots. There was also evidence of an adaptive response to disrupted sphingolipid metabolism in both the virulence and watering assays, suggesting induction of pathways responsible for metabolism of sphingoid base 1-phosphates after prolonged exposure. The results suggest that fumonisin, and its effects on sphingolipids, could contribute to all aspects of F. verticillioides maize seedling disease.  相似文献

Most conservation practitioners rely on experience-based information for management decisions. In South Africa, government policy thus directs managers to base decisions for elephant management on the best available scientific information. The reliance of their management decisions on science, however, is unknown. We interviewed 30 managers from small to large protected areas in South Africa to evaluate whether science underpinned strategic and applied management decisions regarding elephants. We also evaluated their perspectives on limitations to, and opportunities for, greater reliance on science. Twenty-nine managers valued science as a platform for their decisions. However, most managers based decisions on experience-based information. Only 28%, 30% and 8% of managers respectively developed objectives, identified issues and selected management methods on science-based information. Furthermore, only 30% selected a desired number of elephants, and 5% selected a population control method, according to science-based information. The probability of managers basing decisions on science-based information differed with management area size, tenure, planning timeframes and manager experience. However, except for experience, these relationships were weak, and overall an implementation gap between science and elephant management existed. Limitations for science to inform decisions included lack of relevance and application of science findings to respective management areas, and scientists’ attitudes and credibility. Opportunities included more site specific studies, better communication of, and easier access to scientific material. These opportunities could enable scientists to contribute more effectively to elephant management decisions and improve the reliance on science among the wider community of conservation practitioners in Africa.  相似文献

The Western Cape of South Africa is characterized by the hyperdiverse vegetation of the Fynbos biome. Typical fynbos vegetation is a fire-adapted sclerophyllous Mediterranean-type ecosystem on poor, sandy or stony soils. It is characterized by plants with low nutrient content producing slowly decomposing litter. Fire is recognized as a major factor for carbon and nutrient cycling in this vegetation type. However, knowledge of biological decomposition processes in this biome is limited. We used litter-bags to measure mass loss and changes in chemical composition in litter from three species representing characteristic taxa in fynbos, a Protea exima hybrid, Erica multumbellifera, and Restio multiflorus, during approximately 180 days. In addition, we used a standard litter of a species with high nutrient content, Galenia africana, and a mixture of Protea and Erica. We compare our results with a previous study from renosterveld including the geophyte Watsonia borbonica, which occurs in both vegetation types and occurs commonly in the study area. We found that decomposition rate among the true fynbos plant species P. exima, E. multumbellifera, R. multiflorus and W. borbonica varied almost eight-fold. Litter decomposition was strongly correlated to litter stoichiometry, i.e. C/N and C/P-ratios. Most litters accumulated one or several nutrients during the study period. The mixture of litters decomposed faster than expected from the results of each litter separately. Our study indicates that biological decomposition may be more important for carbon and nutrient cycling in fynbos than previously thought. These results are in accordance with recent studies showing large variation in plant litter quality within vegetation types and biomes. Such large variation in litter quality and decomposition rate suggests that some generalisations about ecosystem processes in the fynbos may need reevaluation.  相似文献

The fungus Fusarium verticillioides infects maize and produces fumonisins. The purpose of this study was to determine the ability of F. verticillioides to produce fumonisins in synthetic and natural soils and their biological availability to maize roots. Maize seeds were inoculated with a pathogenic strain of F. verticillioides (MRC826) and planted in synthetic and three different natural soils. There were statistically significant reductions in stalk weight and root mass and increased leaf lesions in the MRC826-treated seedlings in all soil types. Fumonisins were detected in all of the soils of seedlings grown from MRC826-inoculated seeds. The fumonisin produced in the soils was biologically available to seedlings as demonstrated by the statistically significant elevation of free sphingoid bases and sphingoid base 1-phosphates in their roots. These results indicate that F. verticillioides produced fumonisins in the autoclaved synthetic and natural soils and that the fumonisin produced is biologically available on the basis of evidence of inhibition of ceramide synthase.  相似文献

The fungus Fusarium graminearum (sexual stage Gibberella zeae) causes ear rot of maize (Zea mays) and contamination with the 8-ketotrichothecenes nivalenol (1) or 4-deoxynivalenol (2), depending on diversity of the fungal population for the 4-oxygenase gene (TRI13). To determine the importance of 1 and 2 in maize ear rot, a survey of naturally contaminated maize in Nepal was combined with experiments in the field and in a plant growth room. In the survey, 1 contamination was 4-fold more frequent than 2 contamination and 1-producers (TRI13) were isolated more than twice as frequently as 2-producers (Psi TRI13). In maize ear rot experiments, genetically diverse 1-producers and 2-producers caused ear rot and trichothecene contamination. Among strains with the same genetic background, however, 1-producers caused less ear rot and trichothecene contamination than did 2-producers. The high frequency of 1 contamination and the high virulence of many 1-producers are of concern because maize is a staple food of rural populations in Nepal and because 1 has proven to be more toxic than 2 to animals.  相似文献

The relationship between soil water regime and soil profile morphology could contribute towards improving catchment water yield. This paper proposes refinements to the interpretation of orthic A horizons (ochric), defined in Soil Classification — A Taxonomic System for South Africa, to aid interpretation for hydrological purposes. Soil water contents have been measured for six years in the Weatherley catchment in South Africa and used to classify orthic A horizons in terms of wetness classes defined in the Soil Survey Manual. The well drained class is applicable to orthic A horizons overlying red apedal B, yellow-brown apedal B (agric or ferralic) or neocutanic B (cambic) horizons. The moderate poorly and poorly drained classes are applicable to orthic A horizons overlying E (albic), soft plinthic B (ferric) or G (gleyic) horizons. The occurrence of mottles and matrix colour can be used for differentiation between the latter two wetness classes. Interpretation of orthic A horizons can therefore be enhanced by wetness classification, utilizing the nature of the underlying horizon, the occurrence of mottles and matrix colour.  相似文献

South Africa is the most industrialised country in southern Africa and stands at some risk from negative pollution impacts. To the authors' knowledge, this paper presents the first attempt to apply the critical loads approach on the African continent; although sensitivity mapping has been performed for Africa and the rest of the world (Kuylerstierna et al, this conference). Actual sulphate and base cation deposition loads in Mpumalanga (formerly the Eastern Transvaal province of South Africa) were mapped from 16 monitoring sites. The region is characterised by long, dry periods with little rain, high evaporation (up to 8 mm per day) and low run-off (15% of MAP). Provisional critical load and exceedance maps were produced for the surface waters using the Steady-State Water Chemistry Model and the Diatom model. Maps of soil sensitivity to acid deposition, based on bedrock lithology, soil chemical characteristics and land cover, were produced. A weathering rate of 0.39–0.86 keq/ha/year was calculated for the most sensitive sites and taken as the critical load, based on the assumption that the weathering rate represents the buffering ability of the system. The critical loads were contrasted with measures of actual deposition to examine potential scenario's for critical load exceedances. Akey factor in refining the sensitivity maps, and allowing estimation of the critical loads, is the accurate calculation of weathering rates under the warmer and more arid environmental conditions prevalent in South Africa. In a developing country such as South Africa, where research resources are limited, the critical loads approach is a valuable means of assessing the risk of potential impacts of atmospheric deposition.  相似文献

Fungi of the genus Fusarium are common fungal contaminants of maize and are also known to produce mycotoxins. Maize that has been genetically modified to express a Bt endotoxin has been used to study the effect of insect resistance on fungal infection of maize grains by Fusarium species and their related mycotoxins. Maize grain from Bt hybrids and near-isogenic traditional hybrids was collected in France and Spain from the 1999 crop, which was grown under natural conditions. According to the ergosterol level, the fungal biomass formed on Bt maize grain was 4-18 times lower than that on isogenic maize. Fumonisin B(1) grain concentrations ranged from 0.05 to 0.3 ppm for Bt maize and from 0.4 to 9 ppm for isogenic maize. Moderate to low concentrations of trichothecenes and zearalenone were measured on transgenic as well as on non-transgenic maize. Nevertheless, significant differences were obtained in certain regions. The protection of maize plants against insect damage (European corn borer and pink stem borer) through the use of Bt technology seems to be a way to reduce the contamination of maize by Fusarium species and the resultant fumonisins in maize grain grown in France and Spain.  相似文献

Potassium (K) availability in soils is largely governed by their mineralogical composition. The extent of weathering of primary K‐bearing minerals, the chemical pathways through which weathering takes place, as well as the dynamic equilibrium between various K fractions in soils are factors which determine different soil types of varying K‐supplying capacity. The marked variability of K availability in soils in South Asia needs to be taken into account when formulating K‐management strategies in intensive cereal‐based systems in response to K application. Evidence from long‐term fertilizer experiments in rice–rice (R‐R) or rice–wheat (R‐W) systems strongly indicates significant yield responses to K application and negative K balances where K application is either omitted or applied suboptimally. However, K‐fertilizer recommendations in South Asia are generalized over large areas while farmers neglect K application to crops and remove crop residues from fields. These practices may strongly affect yield and soil K‐fertility status in the emerging rice–maize (R‐M) systems in different locations of South Asia. The dry‐matter yield of the R‐M system is usually much higher than that of the R‐R or R‐W system causing high withdrawal of nutrients from the soil. The current review assesses various K forms and K availability in diverse soil types of South Asia supporting rice‐based systems. Aspects considered include: long‐term crop yield and its response to added nutrients, K balance for intensive rice‐based systems, and the role of crop residues in supplying K to crops. Emerging data from either completed or on‐going experiments on the R‐M systems in India and Bangladesh have revealed very high system productivity and variable responses and agronomic K‐use efficiency of maize and rice. Potassium responses of maize are extremely high and variable for soils in Bangladesh. Finally, a plant‐based strategy for field‐specific nutrient management is presented and the need for models and decision support systems for developing efficient K management of the R‐M system is also discussed.  相似文献

The production of fusaproliferin (FUS), a recently described mycotoxin, and beauvericin (BEA), a mycotoxin recently reported to co-occur with FUS in Fusarium-infected corn, by South African isolates in the Fusarium section Liseola, was investigated. Five isolates each of F. verticillioides, F. proliferatum, F. subglutinans, and F. globosum were cultured on corn kernels. Four each of the five South African isolates of F. proliferatum and F. subglutinans produced FUS (10-1725 and 330-2630 mg/kg, respectively). BEA was produced by four of the F. proliferatum strains (310-1130 mg/kg) and three of the F. subglutinans strains (140-700 mg/kg). The isolates of F. verticillioides failed to produce significant levels of either of these secondary metabolites. F. globosum was a weak producer of both in that one isolate of five produced 25 mg/kg FUS and five out of five produced BEA at levels ranging between 10 and 110 mg/kg. To further characterize these strains, their production of fumonisins B(1), B(2), and B(3), as well as moniliformin, was investigated. Of the four species investigated, fumonisins were produced by all except F. subglutinans, which in turn was the only species whose isolates in this study produced moniliformin (four of five isolates, ranging from 155 to 2095 mg/kg). Analysis of visibly Fusarium-infected home-grown corn collected in the Transkei region of the Eastern Cape Province of South Africa showed that nine of the ten samples contained low levels of FUS (up to 62 microg/kg), whereas all ten samples showed BEA contamination ranging from 8 to 1734 microg/kg with a mean of 258 microg/kg.  相似文献

The existence of multiple Tylenchulus semipenetrans biotypes within a citrus-producing region complicates the selection of nematode-resistant rootstocks for managing the slow decline of citrus. The potential existence of multiple T. semipenetrans biotypes in South Africa was investigated under greenhouse conditions on differential host plants using nematode isolates from 18 citrus-producing districts with at least six diverse soil groups and three different climates. After three nematode generations, the regional effects had no influence on final nematode population densities, whereas the differential host plants significantly segregated the nematode reproductive potentials. On Olea europaea reproductive potentials (eggs and juveniles/g) were zero, whereas those on Citrus jambhiri and Poncirus trifoliata ranged from 79 to 482 and 86 to 433, respectively. All T. semipenetrans isolates, regardless of the soil group and climate type, behaved similarly to the Poncirus biotype, which therefore rules out the existence of multiple T. semipenetrans biotypes in South Africa.  相似文献
