While developing a low-sulphate system combining indirect chromate-reduction by biologically-produced hydrogen sulphide and direct biological chromate-reduction to treat chromate-bearing waters, the aim of the present work was to evaluate the influence of sulphate and H2 starvation on chromate reduction. Chromate-reduction was performed under continuous-feed conditions in a fixed-film column bioreactor originally inoculated with a bacterial consortium containing Desulfomicrobium norvegicum, and fed with H2. With 500 mg l?1 of sulphate in the feed solution, total chromate-reduction was observed in the effluent whereas sulphate-reduction was strongly decreased, as also confirmed by measurements of isotopic ratios for sulphur. In the absence of sulphate, a chromate-reduction activity was still observed but was lower than in the presence of sulphate, and chromate-reduction was H2-dependent. Molecular biology techniques revealed the composition of the bacterial population in the effluent. D. norvegicum together with other micro-organisms of the Bacteria domain were detected. They include members related to the genera Acinetobacter, Acetobacterium and Rhodocyclus. Even when sulphate-reduction was strongly decreased, the presence of sulphate enhances the efficiency of the H2-dependent chromate-reduction. A H2- and CO2-consuming bacterial population may be used in a globally autotrophic process to reduce chromate at low sulphate concentration, thus avoiding excess sulphide production.  相似文献

The main purpose of this work was to conduct a kinetic study on cell growth and hexavalent chromium [Cr(VI)] removal by Candida sp. FGSFEP in a concentric draft-tube airlift bioreactor. The yeast was batch-cultivated in a 5.2-l airlift bioreactor containing culture medium with an initial Cr(VI) concentration of 1.5 mM. The maximum specific growth rate of Candida sp. FGSFEP in the airlift bioreactor was 0.0244 h?1, which was 71.83% higher than that obtained in flasks. The yeast strain was capable of reducing 1.5 mM Cr(VI) completely and exhibited a high volumetric rate [1.64 mg Cr(VI) l?1 h?1], specific rate [0.95 mg Cr(VI) g?1 biomass h?1] and capacity [44.38 mg Cr(VI) g?1 biomass] of Cr(VI) reduction in the airlift bioreactor, with values higher than those obtained in flasks. Therefore, culture of Candida sp. FGSFEP in a concentric draft-tube airlift bioreactor could be a promising technological alternative for the aerobic treatment of Cr(VI)-contaminated industrial effluents.  相似文献

Abstract

To evaluate the hypothesis that plant-mediated oxygen supplies decrease methane (CH4) production and total global warming potential (GWP) in a tropical peatland, the authors compared the fluxes and dissolved concentrations of greenhouse gases [GHGs; CH4, carbon dioxide (CO2) and nitrous oxide (N2O)] and dissolved oxygen (DO) at multiple peatland ecosystems in Central Kalimantan, Indonesia. Study ecosystems included tropical peat swamp forest and degraded peatland areas that were burned and/or drained during the rainy season. CH4 fluxes were significantly influenced by land use and drainage, which were highest in the flooded burnt sites (5.75 ± 6.66 mg C m?2 h?1) followed by the flooded forest sites (1.37 ± 2.03 mg C m?2 h?1), the drained burnt site (0.220 ± 0.143 mg C m?2 h?1), and the drained forest site (0.0084 ± 0.0321 mg C m?2 h?1). Dissolved CH4 concentrations were also significantly affected by land use and drainage, which were highest in the flooded burnt sites (124 ± 84 μmol L?1) followed by the drained burnt site (45.2 ± 29.8 μmol L?1), the flooded forest sites (1.15 ± 1.38 μmol L?1) and the drained forest site (0.860 ± 0.819 μmol L?1). DO concentrations were influenced by land use only, which were significantly higher in the forest sites (6.9 ± 5.6 μmol L?1) compared to the burnt sites (4.0 ± 2.9 μmol L?1). These results suggest that CH4 produced in the peat might be oxidized by plant-mediated oxygen supply in the forest sites. CO2 fluxes were significantly higher in the drained forest site (340 ± 250 mg C m?2 h?1 with a water table level of ?20 to ?60 cm) than in the drained burnt site (108 ± 115 mg C m?2 h?1 with a water table level of ?15 to +10 cm). Dissolved CO2 concentrations were 0.6–3.5 mmol L?1, also highest in the drained forest site. These results suggested enhanced CO2 emission by aerobic peat decomposition and plant respiration in the drained forest site. N2O fluxes ranged from ?2.4 to ?8.7 μg N m?2 h?1 in the flooded sites and from 3.4 to 8.1 μg N m?2 h?1 in the drained sites. The negative N2O fluxes might be caused by N2O consumption by denitrification under flooded conditions. Dissolved N2O concentrations were 0.005–0.22 μmol L?1 but occurred at < 0.01 μmol L?1 in most cases. GWP was mainly determined by CO2 flux, with the highest levels in the drained forest site. Despite having almost the same CO2 flux, GWP in the flooded burnt sites was 20% higher than that in the flooded forest sites due to the large CH4 emission (not significant). Abstract

To determine the means and variations in CH4 uptake and N2O emission in the dominant soil and vegetation types to enable estimation of annual gases fluxes in the forest land of Japan, we measured monthly fluxes of both gases using a closed-chamber technique at 26 sites throughout Japan over 2 years. No clear seasonal changes in CH4 uptake rates were observed at most sites. N2O emission was mostly low throughout the year, but was higher in summer at most sites. The annual mean rates of CH4 uptake and N2O emission (all sites combined) were 66 (2.9–175) µg CH4-C m?2 h?1 and 1.88 (0.17–12.5) µg N2O-N m?2 h?1, respectively. Annual changes in these fluxes over the 2 years were small. Significant differences in CH4 uptake were found among soil types (P < 0.05). The mean CH4 uptake rates (µg CH4-C m?2 h?1) were as follows: Black soil (95 ± 39, mean ± standard deviation [SD]) > Brown forest soil (60 ± 27) ≥ other soils (20 ± 24). N2O emission rates differed significantly among vegetation types (P < 0.05). The mean N2O emission rates (µg N2O-N m?2 h?1) were as follows: Japanese cedar (4.0 ± 2.3) ≥ Japanese cypress (2.6 ± 3.4) > hardwoods (0.8 ± 2.2) = other conifers (0.7 ± 1.4). The CH4 uptake rates in Japanese temperate forests were relatively higher than those in Europe and the USA (11–43 µg CH4-C m?2 h?1), and the N2O emission rates in Japan were lower than those reported for temperate forests (0.23–252 µg N2O-N m?2 h?1). Using land area data of vegetation cover and soil distribution, the amount of annual CH4 uptake and N2O emission in the Japanese forest land was estimated to be 124 Gg CH4-C year?1 with 39% uncertainty and 3.3 Gg N2O-N year?1 with 76% uncertainty, respectively.  相似文献

The effect of medium-term (5 years) application of organic and inorganic sources of nutrients (as mineral or inorganic fertilizers) on soil organic carbon (SOC), SOC stock, carbon (C) build-up rate, microbial and enzyme activities in flooded rice soils was tested in west coast of India. Compared to the application of vermicompost, glyricidia (Glyricidia maculate) (fresh) and eupatorium (Chromolaena adenophorum) (fresh) and dhaincha (Sesbania rostrata) (fresh), the application of farmyard manure (FYM) and combined application of paddy straw (dry) and water hyacinth (PsWh) (fresh) improved the SOC content significantly (p < 0.05). The lowest (p < 0.05) SOC content (0.81%) was observed in untreated control. The highest (p < 0.05) SOC stock (23.7 Mg C ha?1) was observed in FYM-treated plots followed by recommended dose of mineral fertilizer (RDF) (23.2 Mg C ha?1) and it was lowest (16.5 Mg C ha?1) in untreated control. Soil microbial biomass carbon (Cmb) (246 µg g?1 soil) and Cmb/SOC (1.92%) were highest (p < 0.05) in FYM-treated plot. The highest (p < 0.05) value of metabolic quotient (qCO2) was recorded under RDF (19.7 µg CO2-C g?1 Cmb h?1) and untreated control (19.6 µg CO2-C g?1 Cmb h?1). Application of organic and inorganic sources of nutrients impacted soil enzyme activities significantly (p < 0.05) with FYM causing highest dehydrogenase (20.5 µg TPF g?1 day?1), phosphatase (659 µg PNP g?1 h?1) and urease (0.29 µg urea g?1 h?1) activities. Application of organic source of nutrients especially FYM improved the microbial and enzyme activities in flooded and transplanted rice soils. Although the grain yield was higher with the application of RDF, but the use of FYM as an organic agricultural practice is more useful when efforts are intended to conserve more SOC and improved microbial activity.  相似文献

Anaerobic ammonium oxidation (anammox process) widely occurs in paddy soil and may substantially contribute to permanent N removal; however, little is known about the factors controlling this process. Here, effects of temperature, pH, organic C, and substrates on potential rate of anammox and the relative contribution of anammox to total N2 production in a paddy soil were investigated via slurry incubation combined with 15N tracer technique. Anammox occurred over a temperature range from 5 to 35 °C with an optimum rate at 25 °C (1.7 nmol N g?1 h?1) and a pH range from 4.8 to 10.1 with an optimum rate at pH 7.3 (1.7 nmol N g?1 h?1). The presence of glucose and acetate (5–100 mg C L?1) significantly inhibited anammox activities and the ratio of anammox to total N2 production. The response of potential rates of anammox to ammonium concentrations fitted well with Michaelis-Menten relationship showing a maximum rate (Vmax) of 4.4 nmol N g?1 h?1 and an affinity constant (Km) of 6.3 mg NH4+-N L?1. Whereas, nitrate addition (5–15 mg 15NO3?-N L?1) significantly inhibited anammox activities and the ratio of anammox to total N2 production. Our results provide useful information on factors controlling anammox process and its contribution to N loss in the paddy soil.  相似文献

Abstract

Gypsum (CaSO4 · 2H2O) is used in agriculture both as a source of calcium (Ca) and sulphate (SO4 2?) and as an amendment to improve soil structure. The effect of gypsum on the adsorption of SO4 2? in irrigated and nonirrigated soils was examined. Almost all of the indigenous sulphate (SO4) in a range of Golesthan and North Khorasan soils with moderate pH values (>6) was found to be present in the soil solution and, as a consequence, was highly susceptible to leaching. The adsorption of sulphate to the soils receiving no gypsum was greater with correlation coefficient of r=0.91 at 0 kg S ha?1 as compared to the soils received 40 kg ha?1 of gypsum as fertilizer with the value of r=0.88 in Golesthan Province. The same trend was observed in Khorasan Province with r=0.79 and r=0.75 with soils receiving 0 and 40 kg S ha?1, respectively. The results were more pronounced in irrigated fields for both provinces. The amount of sulphate adsorption in Golesthan Province soils was comparatively greater than soils of Khorasan Province. The results raise questions regarding the efficiency of SO4‐containing fertilizers in correcting and preventing S deficiency in situations where leaching is a concern.  相似文献

Photocatalytic reduction of CO2 in seawater into chemical fuel, methanol (CH3OH), was achieved over Cu/C-co-doped TiO2 nanoparticles under UV and natural sunlight. Photocatalysts with different Cu loadings (0, 0.5, 1, 3, 5, and 7 wt%) were synthesized by the sol–gel method and were characterized by XRD, SEM, UV–Vis, FTIR, and XPS. Co-doping with C and Cu into TiO2 remarkably promoted the photocatalytic production of CH3OH. This improvement was attributed to lowering of bandgap energy, specific catalytic effect of Cu for CH3OH formation, and the minimization of photo-generated carrier recombination. Co-doped TiO2 with 3.0 wt% Cu was found to be the most active catalyst, giving a maximum methanol yield rate of 577 μmol g-cat?1 h?1 under illumination of UV light, which is 5.3-fold higher than the production rate over C-TiO2 and 7.4 times the amount produced using Degussa P25 TiO2. Under natural sunlight, the maximum rate of the photocatalytic production of CH3OH using 3.0 wt% Cu/C-TiO2 was found to be 188 μmol g-cat?1 h?1, which is 2.24 times higher than that of C-TiO2, whereas, no CH3OH was observed for P25.  相似文献

Abstract

Forest fires can change the greenhouse gase (GHG) flux of borea forest soils. We measured carbon dioxide (CO2), methane (CH4) and nitrous oxide (N2O) fluxes with different burn histories in black spruce (Picea mariana) stands in interior Alaska. The control forest (CF) burned in 1920; partially burned (PB) in 1999; and severely burned (SB1 and SB2) in 2004. The thickness of the organic layer was 22 ± 6 cm at CF, 28 ± 10 cm at PB, 12 ± 6 cm at SB1 and 4 ± 2 cm at SB2. The mean soil temperature during CO2 flux measurement was 8.9 ± 3.1, 6.4 ± 2.1, 5.9 ± 3.4 and 5.0 ± 2.4°C at SB2, SB1, PB and CF, respectively, and differed significantly among the sites (P < 0.01). The mean CO2 flux was highest at PB (128 ± 85 mg CO2-C m?2 h?1) and lowest at SB1 (47 ± 19 mg CO2-C m?2 h?1) (P < 0.01), and within each site it was positively correlated with soil temperature (P < 0.01). The CO2 flux at SB2 was lower than that at CF when the soil temperature was high. We attributed the low CO2 flux at SB1 and SB2 to low root respiration and organic matter decomposition rates due to the 2004 fire. The CH4 uptake rate was highest at SB1 [–91 ± 21 μg CH4-C m?2 h?1] (P < 0.01) and positively correlated with soil temperature (P < 0.01) but not soil moisture. The CH4 uptake rate increased with increasing soil temperature because methanotroph activity increased. The N2O flux was highest [3.6 ± 4.7 μg N2O-N m?2 h?1] at PB (P < 0.01). Our findings suggest that the soil temperature and moisture are important factors of GHG dynamics in forest soils with different fire history.  相似文献

Abstract

Nitrous oxide (N2O) emissions were measured monthly over 1 year in three ecosystems on tropical peatland of Sarawak, Malaysia, using a closed-chamber technique. The three ecosystems investigated were mixed peat swamp forest, sago (Metroxylon sagu) and oil palm (Elaeis guineensis) plantations. The highest annual N2O emissions were observed in the sago ecosystem with a production rate of 3.3 kg N ha?1 year?1, followed by the oil palm ecosystem at 1.2 kg N ha?1 year?1 and the forest ecosystem at 0.7 kg N ha?1 year?1. The N2O emissions ranged from –3.4 to 19.7 µg N m?2 h?1 for the forest ecosystem, from 1.0 to 176.3 µg N m?2 h?1 for the sago ecosystem and from 0.9 to 58.4 µg N m?2 h?1 for the oil palm ecosystem. Multiple regression analysis showed that N2O production in each ecosystem was regulated by different variables. The key factors influencing N2O emissions in the forest ecosystem were the water table and the NH+ 4 concentration at 25–50 cm, soil temperature at 5 cm and nitrate concentration at 0–25 cm in the sago ecosystem, and water-filled pore space, soil temperature at 5 cm and NH+ 4 concentrations at 0–25 cm in the oil palm ecosystem. R2 values for the above regression equations were 0.57, 0.63 and 0.48 for forest, sago and oil palm, respectively. The results suggest that the conversion of tropical peat swamp forest to agricultural crops, which causes substantial changes to the environment and soil properties, will significantly affect the exchange of N2O between the tropical peatland and the atmosphere. Thus, the estimation of net N2O production from tropical peatland for the global N2O budget should take into consideration ecosystem type.  相似文献

Recovery of valuable metals from acidic mine drainage (AMD) during bioremediation using straw bioremediation system was investigated, with observation of efficient metal recovery. The recovery loading rates of Cu and Zn were 46.19?±?6.13 and 43.86?±?6.76?mg?m?3?h?1, respectively. More than 97.0% of Cu and more than 87.0% of Zn were recovered from AMD during bioremediation. The recovery loading rate of Cu increased by 4.54?mg?m?3?h?1 for each 1?mg?L?1 increase in influent concentration while that of Zn increased by 4.08?mg?m?3?h?1. Heavy metal toxic effect on the metal recovery in the straw bioremediation system could be neglected in most cases. Low pH could severely decrease recovery rate of Zn, while it had no influence on that of Cu. The recovery loading rate of Zn decreased by almost 70% when the influent pH decreased from 7.0 to 3.0. Cu could be recovered efficiently even at a short hydraulic residence time (HRT) of 18?h, while Zn could only be efficiently recovered at a relatively long HRT of 80?h, indicating that selective metal recovery might be achieved with relatively short HRT and bioreactor process optimization. These findings suggested the feasibility of using sulfate-reducing straw bioremediation system to recover valuable metals during bioremediation of AMD.  相似文献

Biological treatment systems such as biofilters offer a potential alternative to the existing physicochemical techniques for the removal of volatile organic compounds from gaseous emissions. In this experimental work, continuous phase biofiltration of xylene vapors were performed in a laboratory scale compost biofilter that was inoculated with a xylene-acclimatized consortium. The performance was assessed by continuously monitoring the removal efficiency (RE) and elimination capacity (EC) of the biofilter at loading rates varying between 2–220 g?m?3?h?1. The steady-state removal efficiencies were maintained between 60% and 90% up to a loading rate of 80 g?m?3?h?1. The removal efficiency decreased significantly at loading rates higher than 100 g?m?3?h?1. The pressure drop values were consistently less and insignificant in affecting the performance of the system. The present study also focuses in evaluating the stability of biofilter during shut down, restart, and shock-loading operations. An immediate restoration of biological activity after few days of starvation indicated their capability to handle discontinuous treatment situations which is more common to industrial biofilters. The sensitiveness of the biofilm to withstand shock loads was tested by abruptly increasing/decreasing the loading rates between 9–55 g?m?3?h?1, where, removal efficiencies between 60–90% were achieved. These results prove the resilience of the biomass and the stability of the compost biofilter. Anew, results from kinetic analysis reveal that, steady-state xylene removal in the biofilter can be adequately represented by Michaelis–Menten type kinetics, and the kinetic constants namely, ECmax (120.4 g?m?3?h?1) and K s (2.21 g?m?3) were obtained.  相似文献

The sampling and analysis properties of 1-stage and 2-stage filter-pack methods were studied in detail in monitoring of sulphur and nitrogen containing inorganic gases and particles (sulphur dioxide, sulphate, sum of nitric acid and nitrate and total ammonium). The limit of detection and the limit of quantitation for 24-h samples were estimated using the results of a short-term field experiment completed with available data from long-term monitoring and internal quality assurance. Furthermore, the combined expanded measurement uncertainty including sampling and analysis (Utot) was estimated for filter-pack methods in order to give a tool for distinguishing long-term trends in air quality from the measurement variability. Utot was found to be very near the analytical uncertainty when measuring higher air concentration levels, being ± 4.0% for sulphur concentrations?>?1.0 μg m?3, ± 3.0% for sulphate concentrations?>?0.5 μg m?3, ± 3.5% for the sum of nitrate and nitric acid concentrations?>?0.3 μg m?3 and ± 4.5% for total ammonium concentrations?>?0.8 μg m?3. At the lower air concentration range Utot increases significantly due to the field blank values. The precision of the 24-h filter-pack sample results expressed by means of modified median absolute difference (M.MAD) and coefficient of variance (CoV) gave 8.3% for sulphur dioxide and 5.4% for particulate sulphate. For the sum of gaseous nitric acid and particulate nitrate the CoV was 5.5% and for total ammonium 4.3%. In addition the suitability of the 24-h filter-pack methods in weekly sampling was proved.  相似文献

Purpose

Biochar can be used to reduce the bioavailability and leachability of heavy metals, as well as organic pollutants in soils through adsorption and other physicochemical reactions. The objective of the study was to determine the response of microbial communities to biochar amendment and its influence on heavy metal mobility and PCBs (PCB52, 44, 101, 149, 118, 153, 138, 180, 170, and 194) concentration in application of biochar as soil amendment.

Materials and methods

A pot (macrocosm) incubation experiment was carried out with different biochar amendment (0, 3, and 6 % w/w) for 112 days. The CaCl2-extractable concentration of metals, microbial activities, and bacterial community were evaluated during the incubation period.

Results and discussion

The concentrations of 0.01 M CaCl2-extractable metals decreased (p?>?0.05) by 12.7 and 20.5 % for Cu, 5.0 and 15.6 % for Zn, 0.2 and 0.5 % for Pb, and 1.1 and 8.9 % for Cd, in the presence of 3 and 6 % of biochar, respectively, following 1 day of incubation. Meanwhile, the total PCB concentrations decreased from 1.23 mg kg?1 at 1 day to 0.24 mg kg?1 at 112 days after 6 % biochar addition, representing a more than 60 % decrease relative to untreated soil. It was also found out that biochar addition increased the biological activities of catalase, phosphatase, and urease activity as compared with the controls at the same time point. Importantly, the Shannon diversity index of bacteria in control soils was 3.41, whereas it was 3.69 and 3.88 in soils treated with 3 and 6 % biochar soil. In particular, an increase in the number of populations with the putative ability to absorb PCB was noted in the biochar-amended soils.

Conclusions

The application of biochar to contaminated soils decreased the concentrations of heavy metals and PCBs. Application of biochar stimulated Proteobacteria and Bacteroides, which may function to absorb soil PCB and alleviate their toxicity.

Purpose

The nitrification inhibitor dicyandiamide (DCD) has been shown to be highly effective in reducing nitrate (NO3 ?) leaching and nitrous oxide (N2O) emissions when used to treat grazed pasture soils. However, there have been few studies on the possible effects of long-term DCD use on other soil enzyme activities or the abundance of the general soil microbial communities. The objective of this study was to determine possible effects of long-term DCD use on key soil enzyme activities involved in the nitrogen (N) cycle and the abundance of bacteria and archaea in grazed pasture soils.

Materials and methods

Three field sites used for this study had been treated with DCD for 7 years in field plot experiments. The three pasture soils from three different regions across New Zealand were Pukemutu silt loam in Southland in the southern South Island, Horotiu silt loam in the Waikato in the central North Island and Templeton silt loam in Canterbury in the central South Island. Control and DCD-treated plots were sampled to analyse soil pH, microbial biomass C and N, protease and deaminase activity, and the abundance of bacteria and archaea.

Results and discussion

The three soils varied significantly in the microbial biomass C (858 to 542 μg C g?1 soil) and biomass N (63 to 28 μg N g?1), protease (361 to 694 μg tyrosine g?1 soil h?1) and deaminase (4.3 to 5.6 μg NH4 + g?1 soil h?1) activity, and bacteria (bacterial 16S rRNA gene copy number: 1.64?×?109 to 2.77?×?109 g?1 soil) and archaea (archaeal 16S rRNA gene copy number: 2.67?×?107 to 3.01?×?108 g?1 soil) abundance. However, 7 years of DCD use did not significantly affect these microbial population abundance and enzymatic activities. Soil pH values were also not significantly affected by the long-term DCD use.

Conclusions

These results support the hypothesis that DCD is a specific enzyme inhibitor for ammonia oxidation and does not affect other non-target microbial and enzyme activities. The DCD nitrification inhibitor technology, therefore, appears to be an effective mitigation technology for nitrate leaching and nitrous oxide emissions in grazed pasture soils with no adverse impacts on the abundance of bacteria and archaea and key enzyme activities.  相似文献

Abstract

To develop an advanced method for estimating nitrous oxide (N2O) emission from an agricultural watershed, we used a closed-chamber technique to measure seasonal N2O and nitric oxide (NO) fluxes in cornfields, grassland, pastures and forests at the Shizunai Experimental Livestock Farm (467 ha) in southern Hokkaido, Japan. From 2000 to 2004, N2O and NO fluxes ranged from –137 to 8,920 µg N m?2 h?1 and from –12.1 to 185 µg N m?2 h?1, respectively. Most N2O/NO ratios calculated on the basis of these N2O and NO fluxes ranged between 1 and 100, and the log-normal N2O/NO ratio was positively correlated with the log-normal N2O fluxes (r 2 = 0.346, P < 0.01). These high N2O fluxes, therefore, resulted from increased denitrification activity. Annual N2O emission rates ranged from –1.0 to 81 kg N ha?1 year?1 (average = 6.6 kg N ha?1). As these emission values varied greatly and included extremely high values, we divided them into two groups: normal values (i.e. values lower than the overall average) and high values (i.e. values higher than average). The normal data were significantly positively correlated with N input (r 2 = 0.61, P < 0.01) and the “higher” data from ungrazed fields were significantly positively correlated with N surplus (r 2 = 0.96, P < 0.05). The calculated probability that a high N2O flux would occur was weakly and positively correlated with precipitation from May to August. Mixed microbial culture was isolated from heavy metal-contaminated ground soils located inside iron, vinyl and cement factory area. Isolated mixed microbial culture was used for the heavy metal ions (Fe2+, Cu2+, Ni2+ and Zn2+) removal process in horizontal rotating tubular bioreactor (HRTB). In this research, the effect of bioreactor process parameters on the bioprocess dynamics in the HRTB was studied. Results of this research have shown that profiles of heavy metals concentration were gradually reduced along HRTB at all combinations of bioreactor process parameters [inflow rates (0.5?C2.0 L?h-1) and rotation speed (5?C30 min-1)]. Hydrodynamic conditions and biomass sorption capacity have main impact on the metal ions removal efficiency that was varied in the range of 38.1% to 95.5%. Notable pH gradient (cca 0.7 pH unit) along the HRTB was only observed at the inflow rate of 2.0 L?h-1. On the basis of obtained results, it is clear that medium inflow rate (F) has higher impact on the heavy metal removal process than bioreactor rotation speed (n) due to the fact that increase of inflow rate was related to the reduction of equilibrium time for all examined metal ions. Furthermore, equilibrium times for all metal ions are significantly shorter than medium residence times at all examined combinations of bioreactor process parameters. The main impact on the biofilm sorption capacity has covalent index of metal ions and biofilm volumetric density. The sorption capacity of suspended microbial biomass is closely related to its concentration. Results of this research have also shown that the removal of heavy metals ion can be successfully conducted in an HRTB as a one-step process.  相似文献

Purpose

Rice-paddy-dominated watersheds in eastern China are intensively cultivated, and lands with two crops receive as much as 550–600 kg?ha–1?year–1 of nitrogen (N), mainly through the addition of N-based fertilizers. However, stream N concentrations have been found to be relatively low. Waterways in the watersheds are assumed to be effective “sinks” for N, minimizing its downstream movement. We directly measured net sediment denitrification rates in three types of waterways (ponds, streams/rivers, and a reservoir) and determined the key factors that control net sediment denitrification. Such information is essential for evaluating the impact of the agricultural N cycle on the quality of surface water.

Materials and methods

The pond–stream–reservoir continuum was sampled every 2 months at nine sites in an agricultural watershed between November 2010 and December 2011. Net sediment N2 fluxes/net sediment denitrification rates were determined by membrane inlet mass spectrometry and the N2/Ar technique. A suite of parameters known to influence denitrification were also measured.

Results and discussion

Net denitrification rates ranged between 28.2?±?18.2 and 674.3?±?314.5 μmol N2–N?m–2?h–1 for the streams, 23.7?±?23.9 and 121.2?±?38.7 μmol N2–N?m–2?h–1 for the ponds, and 41.8?±?17.7 and 239.3?±?49.8 μmol N2–N?m–2?h–1 for the reservoir. The mean net denitrification rate of the stream sites (173.2?±?248.4 μmol N2–N?m–2?h–1) was significantly higher (p?<?0.001) than that of the pond sites (48.3?±?44.5 μmol N2–N?m–2?h–1), and the three types of waterways all had significantly higher (p?<?0.01) mean net denitrification rates in summer than in other seasons. Linear regression and linear mixed effect model analysis showed that nitrate (NO3 ?–N) concentration in surface water was the primary controlling factor for net sediment denitrification, followed by water temperature. Using monitoring data on NO3 ?–N concentrations and temperature of the surface water of waterways and an established linear mixed effect model, total N removed through net sediment denitrification in the pond–stream–reservoir continuum was estimated at 46.8?±?24.0 t?year–1 from July 2007 to June 2009, which was comparable with earlier estimates based on the mass balance method (34.3?±?12.7 t?year–1), and accounted for 83.4 % of the total aquatic N. However, the total aquatic N was only 4.4 % of the total N input to the watershed, and thus most of the surplus N in the watershed was likely to be either denitrified or stored in soil.

Conclusions

High doses of N in a rice-paddy-dominated watershed did not lead to high stream N concentrations due to limited input of N into waterways and the high efficiency of waterways in removing N through denitrification.  相似文献

Purpose

Evergreen broad-leaved forest ecosystems are common in east China, where they are both ecologically and economically important. However, nitrogen (N) addition over many years has had a detrimental effect on these ecosystems. The objective of this research was to evaluate the effect of 4 years of N addition on microbial communities in an evergreen broad-leaved forest in southern Anhui, China.

Materials and methods

Allochthonous N in the form of aqueous NH4NO3 and phosphorus (P) in the form of Ca(H2PO4)2·H2O were applied at three doses with a control (CK, stream water only without fertilizer): low-N (50 kg N ha?1 year?1), high-N (100 kg N ha?1 year?1) and high-N+P (100 kg N ha?1 year?1 + 50 kg P ha?1 year?1). Quantitative PCR analysis of microbial community size and Illumina platform-based sequencing analysis of the V3-V4 16S rRNA gene region were performed to characterize soil bacterial community abundance, structure, and diversity.

Results and discussion

Bacterial diversity was increased in low-N and high-N treatments and decreased in the high-N+P treatment, but α-diversity indices were not significantly affected by N additions. Proteobacteria, Acidobacteria, and Actinobacteria were the predominant phyla in all treatments, and the relative abundance of different genera varied among treatments. Only soil pH (P = 0.051) showed a weak correlation with the bacterial community in CK and low-N treatment.

Conclusions

The composition of the bacterial community and the abundance of different phyla were significantly altered by N addition. The results of the present study indicate that soil bacterial communities in subtropical evergreen broad-leaved forest are, to a certain extent, resilient to changes derived from N additions.

Corbicula fluminea (Asian clam) can assemble in high densities and dominate benthic communities. To evaluate the influence of this clam on sediment oxygen uptake and nutrient fluxes across the sediment–water interface, a microcosm study was conducted using a continuous-flow cultivation system with sediment, lake water, and C. fluminea specimens from Taihu Lake, China. C. fluminea destroyed the initial sediment surface, enhanced O2 penetration into the sediment partially, and increased the sediment water content, the volume of oxic sediment, and total microbial activity. Sediment O2 uptake was significantly stimulated by C. fluminea. Linear regression results for O2 uptake versus clam biomass ranged from 21.9 to 9.47 μmol h?1 g?1?DW (dry weight). The release of soluble reactive phosphorus, ammonium, and nitrate from the sediment was also increased by the clams. The increase in the amount of soluble reactive phosphorus and ammonium released into the overlying water was 0.042?~?0.091 and 2.77?~?3.03 μmol h?1 g?1?DW, respectively, and this increase was attributed to increased diffusion, enhanced advection between the pore water and the overlying water, and the excretions from C. fluminea. Enhanced nitrification was suggested as the reason for the increase in nitrate release (2.95?~?4.13 μmol h?1 g?1?DW) to the overlying water.  相似文献
