Twenty ten-week-old newly weaned male Batinah goats were randomly assigned to a control (n = 10) and a treated (n = 10) group and were fed a diet containing 0.1 mg/kg DM cobalt (Co). Goats in the treated group received bi-monthly subcutaneous injections of 2000 μg of hydroxycobalamin. The phagocytic function of the polymorphonuclear leukocytes (PMN) were tested using a luminol-dependent chemiluminescence assay with opsonized zymosan as the phagocytic target. One month after the onset of the experiment PMN from the control group exhibited a significantly (p < 0.05) lower CL response, which continued for the second month. The results of the present study demonstrated that low levels of dietary cobalt leads to an early impairment of phagocytic function. This may at least in part, be an explanation as to why at the field level in Oman young goats fed diets containing low levels of Co appear to be more susceptible to infections.  相似文献

Dietary cobalt (265 ppm Co) induced polycythemia and consistent degenerative and necrotic lesions in the seminiferous tubules of rats. Cyanosis and engorgement of testicular vasculature on day 35 and thereafter was followed on day 70 by degenerative and necrotic changes in the germinal epithelium and Sertoli cells. Spermatogonia, primary spermatocytes and round spermatids were markedly affected, while elongated spermatids, spermatozoa, and sertoli cells were more resistant. Damaged tubules, often present side by side with normal tubules, contained multinucleated giant cells composed of degenerated and necrotic spermatocytes and/or spermatids, sloughed germinal and Sertoli cells, and calcified necrotic debris. Necrotic tubules were frequently collapsed and devoid of epithelium except for occasional spermatogonia and surviving Sertoli cells. Lesions were not observed in the Leydig cells, cauda epididymis or seminal vesicles.  相似文献

BACKGROUND: Fatty change of the liver (FCL) is very common in dairy cattle periparturiently. Many laboratory methods have been implicated in order to assist the diagnosis. HYPOTHESIS: To investigate whether FCL in dairy cattle could be evaluated by assessment of ornithine carbamoyl transferase (OCT) by means of an assay modified for bovine serum, other enzyme activity, serum bile acids (SBA) concentration, or other biochemical constituents. ANIMALS: A total of 187 dairy cattle were included: 106 were suspected to have liver dysfunction and were examined after referral by veterinarians; 70 were clinically healthy with mild FCL; and 11 were clinically healthy without FCL. METHODS: Blood and liver biopsy samples were obtained after clinical examination. Histologic examination by light microscopy and classification of samples according to the severity of FCL was done, and total lipid and triglyceride concentration was measured. In serum, OCT, aspartate aminotransferase (AST), alanine aminotransferase (ALT), sorbitol dehydrogenase (SDH), glutamate dehydrogenase (GDH), alkaline phosphatase (ALP), and gamma-glutamyltransferase (gamma-GT) activity as well as SBA, glucose, ketones, total bilirubin (tBIL), and nonesterified fatty acids (NEFA) concentration were measured. RESULTS: OCT and AST activity and tBIL concentration correlate well with the degree of FCL. SBA concentration does not contribute well to FCL diagnosis. The majority of FCL cases appeared within the first 21 days-in-milk (DIM). The majority of moderate-to-severe and severe FCL cases arose in the first 7 DIM. CONCLUSIONS AND CLINICAL IMPORTANCE: Except for OCT, AST, and tBIL, none of the biochemical tests used, including SBA, had sufficient discriminatory power to differentiate reliably between mild and severe FCL because of poor sensitivity. A weak correlation between clinical signs and the extent of FCL was evident.  相似文献

The usefulness of computed tomography (CT) for the diagnosis of feline hepatic lipidosis (FHL) was evaluated. Liver CT number was 54.7+/-5.6 HU (mean+/-SD) in 26 healthy cats. We fast 6 healthy cats for 72 hr to induced FHL experimentally and the cats were assessed by CT and serum biochemical analysis. Liver CT number of the six cats was 53.8+/-3.0 HU before fasting, 46.8+/-2.4 HU after fasting, and 50.2+/-3.6 HU two weeks after restarted feeding. The decreased CT number was associated with the elevation of serum non-esterified fatty acid (NEFA) and beta-hydroxybutyrate levels. These results indicate that measurement of CT number of the liver is an effective procedure for the diagnosis of FHL.  相似文献

A 3-year-old female llama that was 3 months into her first lactation and 10 weeks pregnant was evaluated for anorexia of 24 hours' duration. On physical examination, the llama was in lateral recumbency, bradycardic, tachypneic, and hyperthermic. Palpation per rectum confirmed the presence of a possible dry fecal mass in the spiral colon. A tissue biopsy specimen of the liver was obtained, and histologic examination revealed moderate diffuse lipid accumulation within the hepatocytes. Lactated Ringer's solution was administered for rehydration, and partial parenteral nutrition was then initiated. Hepatic lipidosis is a disease characterized by abnormal accumulation of lipid in the liver and is associated with high mortality in camelids. Anorexia associated with hepatic lipidosis promotes further lipid mobilization and fatty infiltration of the liver. Partial parenteral nutrition with enteral supplementation may be used to maintain adequate energy intake and minimize further lipid mobilization. The distinctive metabolism of camelids may require higher amino acid supplementation relative to nonprotein calories in parenteral solutions than those traditionally provided to other species. The accuracy of ultrasonography in detection of feline hepatic lipidosis was studied retrospectively. The following ultrasonographic criteria were associated positively with severe hepatic lipidosis: the liver hyperechoic, compared with falciform fat; the liver isoechoic or hyperechoic, compared with omental fat; poor visualization of intrahepatic vessel borders; and increased attenuation of sound by the liver. In a group of 36 cats with clinically apparent hepatobiliary disease and in which liver biopsy was done, liver hyperechoic, compared with falciform fat, was the best criterion for diagnosis of severe hepatic lipidosis with 91% sensitivity, 100% specificity, and 100% positive predictive value.  相似文献

Experiments were conducted to investigate the basis for the change in hair colour of black cats to reddish-brown. Black cats were given purified diets based on gelatin, casein plus lactalbumin, or crystalline amino acids as protein sources. Diets that caused the colour of hair to change to reddish-brown were associated with a reduction in melanin in hair (observed by direct microscopic examination), a decreased total melanin concentration and low concentrations of tyrosine in plasma. Reddish hair coat was induced in black kittens born to queens given a tyrosine-deficient diet during pregnancy. Black hair colour was maintained or restored by diets containing a high concentration of tyrosine or phenylalanine. Current dietary recommendations for dietary tyrosine and phenylalanine for cats are below those required to support maximal melanin synthesis in black cats. The requirement appears to be greater than a combination of 4.5 g tyrosine plus 12 g phenylalanine/kg diet but less than 24 g phenylalanine alone/kg diet.  相似文献

The purpose of this study was to evaluate changes of very low-density lipoprotein (VLDL) components in hepatic blood (HB) from 5 nonlactating nonpregnant cows fasted from days 0 to 3 and subsequently refed to day 10 and, in addition, to assess those of other lipoproteins. Increased phospholipid concentrations in each lipoprotein after the start of fasting suggested their availability for the surface lipids of lipoproteins. Although the VLDL-triglyceride (TG) concentration in HB from all cows increased on day 1, the value on day 4 became similar to that on day 0. However, the concentration on day 10 was significantly increased. In all cows, the decreased ratio of the VLDL-TG concentration in HB to the non-esterified fatty acids (NEFA) concentration in portal blood (PB) on day 4 appeared to reflect relatively decreased secretion of TG as VLDL by NEFA excessively mobilized to the liver via PB. The markedly increased ratio on day 10 was considered to contribute to the improvement of hepatic lipidosis.  相似文献

A study was conducted on 20 "Rahmani" male lambs of 16 kg body weight fed on normal feed level and high vitamin A level (NF-HV); normal feed level and normal vitamin A level (NF-NV); low feed level and high vitamin A level (LF-HV) or low feed level and normal vitamin A level (LF-NV) till slaughter weight of 35-40 kg. Average daily gains of lambs were 211, 148, 117 and 87 g for the NF-HV, NF-NV, LF-HV and LF-NV groups, respectively. The corresponding feed conversions were 4.3, 5.9, 5.0 and 6.4 kg DM/kg gain. Raising feed intake and vitamin A level enhanced nutrients digestibility and nitrogen balance. Rumen liquor reflected higher pH value in animals fed higher intake (NF) in the first two diets. Both feed intake and vitamin A levels enhanced volatile fatty acids and ammonia concentrations in the rumen liquor. Feed intake and vitamin A levels positively affected haemoglobin content, packed cell volume, glucose, urea, total protein, vitamin A, insulin, cholesterol, phospholipids, glutamate oxaloacetate and glutamate pyruvate transaminases and alkaline phosphatase in blood Vitamin A, insulin, cholesterol and phospholipids in blood increased gradually over the experimental period. Dressing percentages were 56.2, 49.6, 49.1 and 44.6% for the experimental groups, respectively. Liver content of glycogen and vitamin A increased with increasing feed intake and vitamin A levels. It is concluded that higher levels of vitamin A supplementation (than usually recommended) is required during fattening of lambs. Low energy and normal vitamin A levels are not recommended in lambs feeding.  相似文献

Relationship of hepatic lipidosis to health and performance in dairy cattle   总被引:2,自引:0,他引:2  
In a field study of 80 cows in 9 dairy herds, serial liver biopsies were performed over the peripartum period to determine degree of hepatic lipidosis. Cattle were separated into categories of mild, moderate, and severe hepatic lipidosis on the basis of maximal amounts of hepatic triglyceride that accumulated during this period. Number of cattle with mild, moderate, and severe hepatic lipidosis were 52, 16, and 12, respectively. Cattle with severe hepatic lipidosis had greater concentrations of hepatic triglyceride before calving and after parturition, and greater serum nonesterified fatty acid concentrations and body condition loss after parturition than cattle with mild hepatic lipidosis. Rate of disease and culling and death rate because of disease were greater in cattle with severe hepatic lipidosis. Cattle with severe hepatic lipidosis had reproductive performance equal to clinically normal cattle; however, cattle with moderate hepatic lipidosis had increased days to conception, possibly related to greater milk production.  相似文献

The hepatic and serum lipid concentrations in 49 dairy cows with displaced abomasum, 7 postpartum cows fasted for 6 days, and 14 healthy postpartum cows were studied. The cows with displaced abomasums were retrospectively allotted to 2 groups: those with greater than 15% liver fat (DAHF) and those with less than 15% liver fat (DALF). Liver total lipid concentrations were high in the DAHF group, exceeding these values in the fasted cows by 30% and in the healthy and DALF cows by 63% on the average. In contrast, the liver phospholipid concentrations were low in the DAHF group, intermediate in the fasted and DALF groups and high in the healthy group. On a group basis, an inverse relationship was observed between serum and liver lipid concentrations. The serum concentrations of both total and dextran-sulfate-precipitable (DSP) lipids were high in the fasted cows and were less in the DALF and healthy cows and in the DAHF cows (lowest). The between-group differences in serum total and serum DSP concentrations of triacylglycerol, cholesterol, and phospholipid followed the same quantitative pattern as the total lipids. However, the relative difference between groups was greater for each of the DSP lipid fractions. These results support the hypothesis that severe hepatic lipidosis in cattle occurs due to impaired hepatic lipoprotein synthesis and secretion.  相似文献

Two trials involving housed cattle examined the effect of Co depletion and supplementation on immune status as assessed by the neutrophil function test which measures the ability of isolated neutrophils to kill the yeast Candida albicans. A third trial investigated the extent to which Co status influenced the severity of Ostertagia ostertagi infection. In the first two trials liveweight gains were unaffected until some 40-60 weeks on the low dietary Co intake despite very low serum vitamin B12 values being recorded after 10 weeks. However, the immune status as measured by the neutrophil function test was reduced within 10 weeks of commencing the low Co diet. On administration of Ostertagia ostertagi larvae, Co-depleted cattle showed a greater weight loss than Co-supplemented cattle but showed no difference in the length of the prepatent period, worm egg production or serum gastrin and pepsinogen concentrations. After anthelmintic treatment both groups showed a similar response. It is postulated that the lowered immune response of Co-depleted cattle resulted in the greater severity of the Ostertagia ostertagi infection.  相似文献

Blood samples were taken from eight multiparous cows at a dairy farm on eight occasions between the prepartum period and peak lactation to study the serum concentrations of amino acids and biochemical constituents. The cows were classified as having either severe hepatic lipidosis (HL) or non-hepatic lipidosis (non-HL) according to their clinical condition after calving and changes in serum biochemical parameters. The serum concentrations of beta-hydroxybutyric acid were higher in the HL group than in the non-HL group (ANOVA: p<0.01). The serum concentrations of methionine (Met), phenylalanine, and arginine were significantly different between the two groups (ANOVA: p<0.05). In particular, the Met levels were significantly low for 14 days after calving in the HL group (p<0.05), although Met levels in the HL group tended to be lower than the values in the non-HL group until 30 days after calving, starting 14 days before calving. The results suggest that an insufficiency of Met during the periparturient period is related to the development of hepatic lipidosis.  相似文献

The present study was conducted to evaluate the adverse effects of an interaction between low levels of dietary aflatoxins (AF) and Eimeria tenella infection on broiler chicks. A set of 1-day-old chicks were raised for 35 days in the following groups: a control group, a group fed AF, a group fed AF and inoculated with E. tenella (AF + E.ten), and a group inoculated with E. tenella alone. AF in the contaminated diet were given at 200 ppb starting from the seventh day after hatching while E. tenella was inoculated at a dose of 5 × 104 sporulated oocysts per chick at the 14th day after hatching. Worsened performance traits and high mortality were all observed in the treated birds, particularly the AF + E.ten group. Lesion scores and oocyst outputs were not different within groups. Chickens fed with AF had significantly increased serum ALT and ALP activities as well as decreased albumin content. They also showed hepatomegaly, hepatocytic vacuolation and necrosis, an atrophied bursa of Fabricius, and a thymus with tissue depletion. E. tenella-infected broilers displayed a significant reduction in packed cell volume, hemoglobin content and lymphocyte percentage, and showed hemorrhagic typhlitis. The deficits in hepatic function and hematologic parameters as well as the gross pathological, and histopathological changes, were more common and more severe in the group that was exposed to both aflatoxicosis and coccidiosis than in the groups exposed to either treatment alone. Thus, the combination of aflatoxicosis and E. tenella infection may influence the course of coccidial infection due to additive effects.  相似文献

OBJECTIVES: To determine whether feed restriction induces hepatic lipidosis (HL) in llamas and to evaluate the metabolic changes that develop during feed restriction. ANIMALS: 8 healthy adult female llamas. PROCEDURE: Llamas were fed grass hay at a rate of 0.25% of their body weight per day for 13 to 28 days. Llamas were monitored by use of clinical observation, serum biochemical analyses, and ultrasound-guided liver biopsies. RESULTS: All 8 llamas lost weight and mobilized fat. Five llamas developed HL, including 4 that were nursing crias. During the period of feed restriction, mean serum concentration of bile acids and activities of aspartate aminotransferase (AST), sorbitol dehydrogenase (SDH), and gamma-glutamyl transferase (GGT) were significantly higher in llamas that developed HL, compared with llamas that did not. Mean insulin-to-cortisol concentration ratios were lower in llamas with HL before and up to 7 days of feed restriction, compared with those that did not develop HL. CONCLUSIONS AND CLINICAL RELEVANCE: HL in llamas may be induced by severe feed restriction, particularly in the face of increased energy demand. Llamas with weight loss attributable to inadequate dietary intake may develop biochemical evidence of hepatopathy and HL. Increases in serum concentration of bile acids and activities of GGT, AST, and SDH may indicate the development of HL in llamas and identify affected animals for aggressive therapeutic intervention.  相似文献

The present study evaluated the effects of energetic undernutrition on liquid and solid digesta passage and on nutrient digestibility as well as their interdependencies. Using a 4 x 4 Latin square design, 12 growing Boran steers (183 ± 15.2 kg live weight) were allocated to four levels of metabolizable energy (ME) supply fixed at 100, 80, 60 and 40% of individual maintenance energy requirements (MER) during four experimental periods. Each period comprised three weeks of adaptation, two weeks of data collection and two weeks of recovery. Diets MER80, MER60 and MER40 only consisted of Rhodes grass hay (RGH), whereas diet MER100 contained (as fed) 83% RGH, 8% cotton seed meal and 9% sugarcane molasses. Feed intake differed between treatments (p < .001) and ranged from 40 ± 0.6 g dry matter (DM) per kg of metabolic weight (kg0.75) in MER40 to 81 ± 1.3 g DM in MER100. Digestibility of neutral and acid detergent fibre (NDF, ADF) was highest at MER80, whereas rumen retention time of liquid and solid digesta was longest at MER40. The correlation of rumen retention time of liquid and solid digesta with the digestibility of proximate diet components was weak but positive, whereas the correlation of liquid and solid rumen retention time with quantitative feed and nutrient intake was strong (p < .01) and negative. Our results suggest that tropical cattle are able to buffer a moderate energy deficit by prolonging rumen retention time of digesta and hence improve diet digestibility. Conversely, a severe energy deficit cannot be buffered by digestive adaptation mechanisms and will inevitably lead to productivity losses.  相似文献

In a study using a total of 36 German Simmental beef bulls weighing between 236 kg and about 620 kg the question of what constitutes an adequate Co supply was investigated with reference to the performance criteria growth, feed intake, energy intake, nutrient intake and carcass criteria. The bulls received a diet of corn silage ad libitum and 2.5 kg concentrate for a period of 280 days. Ten rations (R) with graduated Co supplements were fed with mean concentrations of 0.07 (R1), 0.09 (R2), 0.11 (R3), 0.15 (R4), 0.18 (R5), 0.26 (R6), 0.33 (R7), 0.42 (R8), 0.59 (R9) and 0.69 (R10) mg Co per kg dry matter (DM) intake. The Co supplement was added as CoSO47H2O. The diets with no or low Co supplementation produced either distinctly lower daily gains of 1045g (R1) and 1130g (R2) or lower daily gains of about 1260 g (R3, R4) than rations R5–R10, where daily gains averaged 1340 g. The mean daily feed intake per animal was also significantly or marginally affected with values of 6.0 kg DM (R1) rising to 7.7 kg DM (R2–R4) versus 7.9 kg DM on average (R5–R10). Calculations using the broken line model and the quadratic model show that the optimal Co supply for maximum growth is 0.12 mg/kg dietary DM and for maximum feed intake, 0.16–0.18 mg Co/kg dietary DM. After taking further biochemical criteria into account (Stangl et al. 2000), a level of 0.20 mg/kg dietary DM is recommended as an adequate Co supply for growing cattle.  相似文献
