One hundred thirty-six perennial rye-grass silages with wide variations in quality were evaluated for N and DM degradability in three beef steers offered grass silage of medium quality ad libitum. The silages were incubated in the rumen of each animal in triplicate for 0, 6, 12, 24, 48, and 72 h, respectively. The disappearance rates of N or DM were used to calculate the readily soluble fraction ("a" value), potentially degradable fraction ("b" value), and the fractional degradation rate of"b" ("c" value). The effective degradability (P) of N or DM was then estimated assuming a ruminal outflow rate of 0.02, 0.05, or 0.08/h (P0.02, P0.05, or P0.08). The objective was to use these data to develop prediction equations for N and DM degradability in grass silages. There were considerable variations in "a," "b," and "c" values and the P0.02, P0.05, or P0.08 of N and DM (e.g., the P0.02 of N ranged from 75.0 to 93.4% and the P0.02 of DM from 51.5 to 82.5%). The P0.02, P0.05, or P0.08 of N and DM were negatively related (P < 0.001) to ADF, NDF, and lignin concentrations but positively related (P < 0.001) to protein fractions (CP, soluble CP, and true protein concentrations) and digestibility of DM, OM, GE, CP, and NDF and digestible OM in the total DM (measured with sheep). The N and DM degradability data were also positively related to silage lactic acid concentration, but the relationships between DM degradability data and pH, ammonia N/total N, and VFA concentration in silages were negative (P < 0.05). Several sets of prediction equations (linear and multiple) were thus developed for N and DM degradability using CP or NDF concentration, "a" value or digestibility data as primary predictors, together with or without other nutrient concentration and silage fermentation variables. All these relationships were highly significant (P < 0.001), and each predictor had a significant effect on the relationship (P < 0.05). The R2 values in multiple regression for N and DM degradability were generally over 0.70 and higher than in linear regression equations. Four equations were also developed to convert N and DM degradability at a given ruminal outflow rate, predicted using the above-mentioned equations, to their counterparts at any ruminal outflow rate (0.02 to 0.10/h), respectively.

1. The objective of this study was to evaluate whether the oligosaccharide stachyose enhances gastrointestinal tract health by fermentation and proliferation of desirable bacteria species and thus affects growth performance and nutrient digestibility in broilers. 2. A total of 432 1-d-old male Arbor Acres (AA) broilers were randomly allocated to one of 6 treatments, with 12 replicate pens per treatment and 6 birds per pen. Chicks were fed a maize-hamlet protein 300 (HP300) basal diet with 0, 4.0, 8.0, 12.0 or 16.0 g/kg stachyose. A sixth diet contained no HP300 but soybean meal (SBM) and provided 8.7 g/kg stachyose and 3.1 g/kg raffinose. The duration of the study was 42 d. 3. Stachyose contents above 12.0 g/kg depressed group body weights, average daily gain and feed/gain but not feed intake during the whole experimental period. Broiler growth decreased linearly and quadratically with increasing stachyose content. No differences were detected between diets supplemented with 12.0 g/kg stachyose and SBM. 4. Nutrient digestibility tended to decrease but not significantly with increasing stachyose. 5. Stachyose content had no significant positive effects on caecal pH, microflora population and the resulting short-chain fatty acid (SCFA) metabolites during the 42 d experiment, with only butyrate differing significantly in the initial period.

This study was aimed to examine the characteristics of three isolated lactic acid bacteria (LAB) strains (HG24, M1, and TG1), and to evaluate their effects on fermentation characteristics, nutritive value, and in vitro digestibility in a variety of substrates. The experiments were conducted with Italian ryegrass ( Lolium multiflorum Lam.), tall fescue ( Festuca arundinacea Schred.), and oat ( Avena sativa L.) in laboratory silo (1 L) for ensiling 30 days. All the treated silages had significantly ( p < 0.05) higher in vitro dry matter digestibility (IVDMD), and lower undesirable microorganism populations compared with controls. In Italian ryegrass and oat silages, all the inoculants improved the fermentation quality, evidenced by distinctly ( p < 0.05) higher LA contents and ratios of LA to acetic acid, and lower pH, butyric acid and ammonia nitrogen contents. Strains HG24 and commercial inoculant G ( Lactobacillus plantarum MTD‐1) performed better than strains M1 and TG1, indicated by obviously ( p < 0.05) higher LA contents, LAB numbers, and IVDMD. In tall fescue silage, all the isolated LAB strains (HG24, M1 and TG1) markedly ( p < 0.05) increased IVDMD compared with strain G. Strain HG24 is recommended as a good LAB inoculant for ensiling in various forages.

Two nonlactating cows and two wether sheep, all fitted with a permanent cannula into the rumen, were fed either hay plus concentrate, grass silage or corn silage to study the effect of the donor animal and its diet on in vitro fermentation and microbial protein synthesis. Rumen inoculum was obtained before the morning feeding. Grass silage or corn silage was incubated in a semi‐continuous rumen simulation system for 14 days. Four replicated vessels were used per treatment. Degradation of crude nutrients and detergent fibre fractions as well as microbial protein synthesis and the production of volatile fatty acids were studied. Additionally, total gas and methane production was measured with a standard in vitro gas test. Gas production and methane concentration was higher when the inoculum used was from sheep than that from cows. The donor animal also affected the degradation of organic matter and ether extract as well as the amount of propionate and butyrate, and the acetate‐to‐propionate ratio. The effect of the diet fed to the donor animal on fermentation was much greater than the effect of the donor animal itself. Feeding hay plus concentrate resulted in higher gas production and degradation of acid detergent fibre, but in lower degradation of ether extract and reduced microbial protein synthesis. Additionally, the pattern of volatile fatty acids changed significantly when the diet of the donor animals was hay plus concentrate or one of the silages. These results show that in vitro fermentation and microbial protein synthesis is different when based on inoculum from either cattle or sheep. The diet fed to the donor animal is more important than the animal species and is probably mediated by an adjusted microbial activity. With regard to standardized feed evaluations, these results further support the need to harmonize in vitro approaches used in different laboratories.

BackgroundCorn silage is the main dietary component used for ruminant breeding in China and is an important dietary source of fatty acids for these animals. However, little is known regarding effective means to protect the fatty acid (FA) contents in silages. In this study, we examined the changes in FA contents and compositions during corn ensiling and screened several antioxidants for their inhibition of lipid oxidation during corn ensiling. MethodsWe conducted two different experiments. In Experiment 1, corn was ensiled in 30 polyethylene bottles (bottle volume: 1 L, silage density: 600 g/dm 3) and three bottles were opened at 0.5 d, 1 d, 1.5 d, 2 d, 2.5 d, 3 d, 5 d, 7 d, 14 d, and 28 d after ensiling. In Experiment 2, corn was treated with various antioxidants: (1) No additives (CK); (2) BHA (Butylated hydroxyanisole); (3) TBHQ (Tertiary butyl hydroquinone); (4) TPP (Tea polyphenols); and (5) VE (Vitamin E). These treatments were applied at 50 mg/kg and 100 mg/kg of fresh weight with each treatment replicated 3 times. ResultsDuring ensiling in Experiment 1, saturated fatty acids (SFA; C16:0 and C18:0) and malondialdehyde (MDA) contents tended to increase, whereas unsaturated fatty acids (UFA; C18:1, C18:2 and C18:3) tended to decrease. However, these changes were only significant on the first 2 days of ensiling. In Experiment 2, all of the antioxidants tested affected the total FA contents and those of unsaturated fatty acids (C18:1, C18:2 and C18:3) and MDA. The effects of TBHQ and TPP were greater than those of the other antioxidants. ConclusionsThe reduced total FA contents in corn silages were due to unsaturated fatty acids’ oxidation during the early stages of ensiling. Adding an antioxidant could prevent fatty acids’ oxidation in corn silages.

Conclusions; The reduced total FA contents in corn silages were due to unsaturated fatty acids' oxidation during the early stages of ensiling. Adding an antioxidant could prevent fattv acids' oxidation in corn silages.

The aim of the present study was to determine equations that predict ME in total mixed rations (TMR) based on routine methods. The ME content of 30 TMR for dairy cows was determined based on digestible crude nutrients obtained with wether sheep. Concentrations in the TMR (in g/kg DM) varied between 118 and 234 for crude protein, 26 and 48 for crude lipid, 131 and 250 for crude fibre, 281 and 488 for NDF, and 173 and 304 for ADF. Gas production ranged from 40.7 to 54.1 ml/200 mg DM, and enzymatically degraded organic matter from 652 to 800 g/kg DM. Digestibility [%] ranged from 68.6 to 84.0 for organic matter, from 55.6 to 84.3 for crude lipid, from 55.0 to 77.8 for crude fibre, from 57.6 to 77.0 for NDF and from 53.1 to 79.6 for ADF. ME ranged from 9.6 to 11.9 MJ/kg DM, and NEL from 5.7 to 7.4 MJ/kg DM. ME content was highly correlated with the concentration of both crude fibre and enzymatically degradable organic matter as well as with organic matter digestibility. A multiple regression equation based on crude fibre and crude lipid predicted ME with a reasonable goodness of fit (r2 = 0.81; s(y.x) = 2.4%). The inclusion of other nutrients, of neutral and acid detergent fibre, neither of gas production did improve the goodness of fit. The best prediction was achieved with inclusion of enzymatically degraded organic matter (r2 = 0.90; s(y.x) = 1.7%).

The DE content within cereal grains can vary 25% mainly due to changes in apparent total tract digestibility (ATTD) of energy. In vitro digestibility techniques have been developed to predict the DE value among feedstuffs. However, these techniques have not been tested properly for their suitability to predict the variation in energy digestibility and DE content within a cereal grain. The objective of the present study was to establish and evaluate an in vitro digestibility technique to predict in vivo ATTD of energy of barley in grower pigs. Barley grain samples (hulled, n = 21) with a large range in quality were collected; the ADF and CP content ranged from 4.5 to 11.4% and 10.0 to 16.4% (DM basis), respectively. The ATTD of energy was determined using barrows (n = 63, 33 +/- 2.1 kg of initial BW) in 2 periods with 6 observations per sample and ranged from 51.9 to 78.5%, with relative errors between 0.4 and 5.0%. A preliminary study, comparing a 2- and a 3-step in vitro digestibility technique using 3 barley samples, indicated that R(2) between in vivo and in vitro energy digestibility was greater using the 3- than the 2-step technique (0.92 vs. 0.76). Therefore, the 3-step in vitro digestibility technique was used solely in subsequent analyses. Briefly, ground barley was subsequently incubated with pepsin for 6 h, pancreatin for 18 h, and cellulase for 24 h. The DM and GE content of samples and residues were measured to calculate digestibility. The in vitro energy digestibility of the 21 barley samples with duplicate measurements ranged from 63.7 to 82.2%, with relative errors between 0.1 and 2.6%. In vitro energy digestibility was strongly related (y = 1.25 x - 25.22; R(2) = 0.81) to in vivo energy digestibility. Finally, a subset of 7 barley samples was analyzed in quadruplicate using the 3-step in vitro technique. The relationship between in vitro and in vivo energy digestibility was very strong (y = 1.23 x - 25.33; R(2) = 0.97) with relative errors between 0.5 and 2.7%. In vitro DE and energy digestibility were perfectly related (R(2) = 1.00). In summary, the 3-step in vitro energy digestibility technique can accurately predict the ATTD of energy in barley in grower pigs. The 3-step in vitro digestibility technique, thus, might be useful as the reference laboratory procedure to calibrate analytical equipment to rapidly predict the ATTD of energy in barley.

1. The aim of this study was to assess regression equations able to predict the digestible energy (DE) and gross energy digestibility (GEd) of feed ingredients and diets for ostriches. 2. Results of chemical-nutritional characteristics from 17 ingredients (two varieties of maize, two barleys, oat, triticale, wheat bran, soybean meal, sunflower meal, beet pulp, maize silage, alfalfa hay, 4 alfalfa meals and lupin) and 12 experimental diets were used in a stepwise procedure. 3. Acid detergent lignin (ADL) was the first independent variable included in the model to predict the DE of all the samples (R2 = 0.65 and Residual Standard Deviation (RSD) 1.02). When the concentration of ash, acid detergent fibre (ADF) and crude fibre were included in the model, the R2 value of the regression equation increased (from 0.65 to 0.85) and RSD decreased (from 1.02 to 0.48). 4. The ADL concentration was also the first independent variable chosen by the stepwise regression analysis for the estimation of GEd from chemical-nutritional characteristics of feeds, explaining 57% of the total GEd variation. The concentrations of crude protein and ADF included at the second and third steps in the model increased the R2 (up to 0.70 and 0.73, respectively) and decreased the RSD values (from 0.29 to 0.21 and 0.20, respectively). When other variables as crude fibre, ash and gross energy were included in the model, the coefficient of determination and the RSD strongly improved (0.85 and 0.12, respectively).

This study was done to determine the effectiveness of supplementary enzymes at increasing the fiber digestion by ruminal microorganisms and to assess whether enzyme activity limits the rate of fiber digestion in ruminal digesta. In vitro comparisons of enzyme activities in two feed enzyme preparations (A and B) with enzyme activities extracted from ruminal fluid indicated that the addition of fibrolytic enzymes at the application rates recommended by the manufacturers would not be expected to increase significantly glycanase and polysaccharidase activities in ruminal fluid. Preparations A and B both increased (P < 0.001) the rate of gas production from freeze-dried corn and grass silages in in vitro incubations with ruminal fluid, but only at concentrations much higher than recommended application rates. Autoclaved controls had little or no effect. Ultrafiltration of enzyme B indicated that most stimulation was due to components >100 kDa, which is consistent with the cause of the stimulation being enzyme activity. Fibrolytic enzymes from other sources were also able to stimulate gas production: increased rates of gas production were observed in seven out of eight combinations of "cellulase" and corn or grass silage (P < 0.05). The comparison of glycanase and polysaccharidase activities with gas-stimulatory activity in the different enzyme preparations indicated that the highest correlation was between increased gas production and enzyme activity against microgranular cellulose (P < 0.05). In a wider range of fibrolytic enzyme preparations, those with endo-(beta-1,4)- or exo-(beta-1,4)-xylanase activity equal to that of preparation A did not produce similar increased rates of fermentation of corn silage when glucanase activity was low (P > 0.05). In contrast, preparations with glucanase activity similar to enzyme A gave at least as great (P < 0.05) an improvement in gas production than enzyme A, irrespective of xylanase activity. It was concluded that enzyme activity, probably a type of endo-(beta-1,4)-glucanase activity, limits the rate of fermentation of corn and grass silage in the rumen. Enzyme supplements of the type used in these experiments are unlikely to possess sufficient activity to overcome this limitation by direct application to ruminal digesta, implying that treatment of the ration prefeeding will be key to harnessing the potential of exogenous fibrolytic enzymes in ruminant nutrition.

Two experiments were conducted to determine the effects of wet distillers grain plus solubles (WDG; <15% sorghum grain) concentration in steam-flaked corn (SFC) diets on feedlot performance, carcass characteristics, ruminal fermentation, and diet digestibility. In Exp. 1, six hundred crossbred steers (364 ± 35 kg of BW) were used in a randomized complete block design with 8 replications/treatment. Dietary treatments consisted of a dry-rolled corn (DRC) control diet without WDG, a SFC control without WDG, and SFC with 4 WDG concentrations (15, 30, 45, 60% DM basis) replacing SFC, cottonseed meal, urea, and yellow grease. Final BW, ADG, G:F, HCW, and 12th-rib fat depth were greater (P ≤ 0.05) for SFC compared with DRC. Dry matter intake tended (P = 0.06) to be greater for DRC compared with SFC. Final BW, ADG, G:F, HCW, 12th-rib fat depth, and marbling score decreased linearly (P < 0.01) with increasing WDG concentration. In Exp. 2, six ruminally and duodenally cannulated crossbred steers (481 ± 18 kg of BW) were used in a 6 × 6 Latin square design using the same diets as Exp. 1. Ruminal, postruminal, and total tract OM and NDF digestibility were not different (P > 0.14) for DRC compared with SFC. Ruminal and total tract starch digestibility were greater (P < 0.01) for SFC compared with DRC. Dry matter and OM intake were not different (P ≥ 0.43) among WDG treatments. Ruminal and total tract OM digestibility decreased linearly (P < 0.01) with increasing WDG concentration. Intake, ruminal digestibility, and total tract digestibility of NDF increased linearly (P < 0.01) with increasing WDG concentration. Starch intake decreased linearly (P < 0.01) with increasing WDG concentration. Ruminal starch digestibility increased (P = 0.01) with increasing concentration of WDG. Total tract starch digestibility decreased quadratically (P < 0.01) with increasing concentration of WDG. Feeding SFC improved steer performance compared with DRC. The concentration of WDG and corn processing method influences nutrient digestibility and ruminal fermentation. The addition of WDG in SFC-based diets appears to negatively affect animal performance by diluting the energy density of the diet.

Dried whole whey, lard or dried skim milk was added to a corn-soybean meal basal diet to determine the effect on apparent nitrogen (ND), energy (ED) and dry matter digestibility (DMD) in pigs weaned at 28 d. Four groups of four littermate barrows were allotted to the experimental diets and placed in individual metabolism pens for 12 d (3 d adjustment and three collection periods of 3 d each) after weaning. The study was a 4 X 3 factorial with diet and period as main effects. Addition of dried skim milk or dried whey to the diet resulted in higher values (P less than .05) for DMD and ED as compared with the basal or corn-soy and lard diet. Pigs fed added dried skim milk had improved (P less than .05) ND over pigs on other dietary treatments. All nutrient digestibility coefficients increased (P less than .05) from period 1 to period 2, and ND increased (P less than .05) from period 2 to 3. It was concluded that the young pig requires at least 6 to 9 d after weaning at 28 d to adjust to typical corn-soybean meal starter diets.

To gain further insights into temperate and tropical grass ensiling, fermentation products and bacterial communities were examined at both the initial and late stages of ensiling of wilted Italian ryegrass and wilted guinea grass silages. 2,3‐Butanediol and ethanol fermentation were observed in wilted Italian ryegrass silage. Enterobacteria such as Rahnella sp. and Enterobacter sp. may have been involved in fermentation; however, alcohol production was intensified after the silage enterobacterial community overwhelmed the pre‐ensiled enterobacterial community. Pediococcus spp. appeared in silage stored for 4 months, when a significant increase in lactic acid content was seen compared with that at 2 months. Prolonged storage enhanced acetic acid fermentation in wilted guinea grass silage. The disappearance of Enterococcus sulfureus and appearance of Lactobacillus plantarum may have been associated with the increased acetic acid content. Although many species of enterobacteria were found in common between the pre‐ensiled crop and silages of Italian ryegrass and guinea grass, marked differences were seen in the type of fermentation from the initial stages. These results indicate that the bacterial community of pre‐ensiled crops may be immediately replaced by one that is adapted to ensiling environments, although metabolic changes may continue over the course of ensiling.

The purpose of this study was to investigate the effects of feeding Bacillus subtilis on rumen fermentation, blood metabolites, nutrient digestibility, and energy and nitrogen balances in non-lactating crossbred (Holstein-Friesian × Bos indicus) cows. Four cows were assigned to the control and B. subtilis diets in a crossover design, and respiratory and metabolic experiments were conducted. For the B. subtilis diet, B. subtilis DSM15544 spores were added at the rate of 1.0 × 10 10 CFU/head/day to the control diet. At 4 hr after feeding, cows fed the B. subtilis diet had increased levels of i-butyric acid in the rumen fluid and tended to have lower concentrations of plasma non-esterified fatty acids when compared with cows fed the control diet. This suggests that feeding B. subtilis could improve energy efficiency. However, there was no effect on energy retention in this study. Although there were no effects on nutrient digestibility, nitrogen balance, or methane production, heat production was significantly higher in cows fed the B. subtilis diet than in those fed the control diet.

Two comparative slaughter experiments conducted with growing beef steers (300 kg to 480 kg BW) fed alfalfa cubes demonstrated that animal performance diminished as alfalfa matured. In each trial, ADG (empty BW basis) was 10 to 20% greater from pre-bloom than from early-bloom alfalfa. Net energy for maintenance (Mcal/kg) followed a pattern similar to that of ADG, but NEg (Mcal/kg) did not decrease (P greater than .05) as alfalfa maturity increased. Digestion trials with steers and wethers clearly indicated significant reductions in apparent digestibilities of DM, energy, and cell wall fractions as alfalfa matured. Steers fed at 110% of maintenance in digestion Trails 1, 2, and 3 consistently digested cubed alfalfa to a greater extent than wethers fed the same alfalfa ground and pelleted. Digestion coefficients (percentages) for ADF, cellulose, NDF, and crude fiber were 5 to 14% greater for steers fed cubes than for wethers fed pellets. Regression equations calculated from results of three digestion trials indicate that digestible DM % and DE (Mcal/kg) could be predicted from ADF %, but they were 4 to 5% lower for wethers fed pelleted alfalfa than for steers fed cubes.
