The lipoprotein and lipid composition of the serum in postabsorptive haddock was characterized. A low level of very low density lipoproteins (VLDL; <50 mg dL?1) was observed in serum. High density lipoprotein (HDL) was the predominant lipoprotein class followed by low density lipoproteins (LDL). Of the lipoprotein classes, the highest proportions of triacylglycerol (28%) and phospholipid (64%) were observed in the lipid of VLDL and HDL, respectively. Serum lipid (48%), phospholipid (50%) cholesterol ester (65%) and HDL (51%) contained a high level of polyunsaturated FA, particularly eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). Serum triacylglycerol (38%) and VLDL (36%) contained a lower proportion of polyunsaturated fatty acid. The phospholipid, triacylglycerol, cholesterol ester and cholesterol contents of the whole serum were 739, 214, 201 and 141 mg dL?1 respectively. On the basis of total acyl carbon number, the 38C (36%) and 36C (24%) molecules were the predominant phospholipids. The 50C (11%), 52C (17%), 54C (21%), 56C (21%) and 58C (15%) molecules were the major triacylglycerols. High molecular weight apo B‐like proteins were observed in the VLDL, LDL and intermediate density lipoprotein (IDL) fractions. An apo AI‐like protein was predominant in the HDL fraction. The prevalence of enlarged fatty livers in cultured gadoid fish (i.e. haddock and cod) is a major constraint to their commercialization and has been associated with the level of dietary lipid. Information on lipid (triacylglycerol) transport out of the liver through the serum as lipoproteins (VLDL) has implications for our understanding of fatty liver development in cultured gadoid fish. The low level of VLDL triacylglycerol (<12 mg dL?1) circulating in the serum of postabsorptive haddock suggests either a low level of lipid transport out of the liver as triacylglycerol or relatively rapid turn over of VLDL.  相似文献

The digestible energy (DE) content and the apparent digestibility coefficients (ADCs) of nutrients in common feed ingredients available in Atlantic Canada for haddock, Melanogrammus aeglefinus, feed formulations were determined. Juvenile haddock (mean weight, 93.9±2.1 g) were held in tanks equipped with fecal collection columns and fed practical fish meal‐based diets for 5 weeks. The experimental diets consisted of a reference diet and six test diets, each containing 30% test ingredient, with all diets being supplemented with chromic oxide (Cr2O3, 5 g kg?1) as the inert digestion indicator. Three marine fish by‐products, herring meal (HM), shrimp meal (SM) and crab meal (CRM) and three plant protein supplements, dehulled soybean meal (SBM), canola meal (CAM) and corn gluten meal (CGM) were the test ingredients. The DE content of HM, SBM, CGM, CRM, CAM and SM were 18.3, 18.0, 17.8, 12.4, 10.9 and 8.8 MJ kg?1 respectively. Protein ADCs were 95.9%, 92.2%, 92.3%, 82.0%, 83.0% and 73.5% respectively. Organic matter ADCs were 95.9%, 89.0%, 72.6%, 68.9%, 58.9% and 54.9% respectively. Lipid ADCs were 97.9%, 83.0%, 57.4%, 62.0%, 87.2% and 55.8% respectively. Based upon its high crude protein content and nutrient ADC and DE content, properly processed dehulled SBM was found to be a good plant protein supplement to partially replace HM in haddock feeds.  相似文献

Urine collection from fish is an integral part of metabolic studies designed to measure the excretion of various biochemical compounds. The techniques developed for urine collection in salmonids cannot be applied to gadoid fish due to the anatomical differences in their urinary system. The anterior ureter of haddock (Melanogrammus aeglefinus L.) is an elongated duct that originates from the anterior part of the trunk kidney and courses dorsal‐posterior to the region of the uropore. The posterior ureter originates from the middle of the caudal kidney, and eventually fuses with the anterior ureter before forming a small urinary bladder. After the two ureters join together, a short common duct empties into the urinary bladder and terminates at the uropore behind the anus. Just before the end, the terminal posterior ureter takes a sharp U‐shaped turn, which makes cannulation difficult. We investigated the development of a cannulation technique for urine collection in three different‐sized groups of juvenile haddock. In anaesthetized fish, a catheter was inserted in the uropore in a posterior direction to follow the U‐shaped course of the ureter. After insertion of the catheter into the uropore, the externalized segment of tubing was connected to a needle attached to a syringe. Urine was then aspirated from each of the 20 fish at 15 μL min?1 for 5 min to measure urine volume and urinary phosphate concentration. The results were reproducible and this cannulation technique has potential for use in other gadoid metabolic studies.  相似文献

Haddock (Melanogrammus aeglefinus L.) were maintained on either simulated natural (SNP) or advanced, phase‐shifted photoperiods (ADV) and reproductive characteristics of female fish were measured throughout two reproductive cycles. By advancing the seasonal photoperiod 8 weeks, relative to ambient conditions, follicle development, peak levels of estradiol and initiation of spawning were all accelerated compared with fish maintained on an SNP. Profiles of steroid synthesis and follicular development were similar between the two groups but the spawning period was protracted by 40 days in the fish maintained on ADV. Ultrasound imaging proved to be an effective and non‐invasive tool for accurately assessing ovarian development throughout the reproductive cycle. These results suggest that photoperiod manipulation has significant potential for extending the availability of eggs and larvae for haddock aquaculture.  相似文献

We estimated recent growth of Atlantic cod (Gadus morhua) and haddock (Melanogrammus aeglefinus) larvae collected on the southern flank of Georges Bank in May 1992–94 from the ratio of RNA to DNA (R/D) and water temperature. Growth of both species increased with water temperature to about 7°C and then decreased. The highest growth rates were observed in May 1993 at water temperatures around 7°C. These data confirm an earlier observation of comparable temperature optima for growth of Atlantic cod and haddock larvae in the north‐west Atlantic. Comparisons of field growth rates and temperature optima with data for larvae cultured at high temperatures and prey densities in the laboratory suggest that growth may have been food‐limited at higher temperatures on Georges Bank. Given that 7°C is the long‐term mean water temperature on the southern flank in May and that climate models predict a possible 2–4°C rise in water temperatures for the western North Atlantic, our findings point to a possible adverse effect of global warming on Atlantic cod and haddock.  相似文献

Juvenile Atlantic cod (Gadus morhua L.) and haddock (Melanogrammus aeglefinus L.) were subjected to 30 s air exposure stressors following acclimation to 4, 10 and 14 and 4, 8 and 14°C respectively. Both species responded to the stressor with increases in plasma cortisol at all temperatures tested. At 14°C cortisol levels peaked within 1 h post‐stressor, and returned to pre‐stressor levels within 24 h. In contrast, at 4°C, peak cortisol levels were not attained until 6 h post‐stressor in haddock and remained elevated beyond 24 h in both species. The rate of plasma glucose accumulation was greater at higher temperatures in both species and no increase was seen at 4°C. Lysozyme activity in cod, in response to the stressor, was lower than the values reported for some other species and increased slightly at 14°C. The results show that cod and haddock acclimated to different temperatures respond to common, acute stressors in a manner similar to other teleosts.  相似文献

In two separate experiments, haddock (Melanogrammus aeglefinus) larvae were raised under different photoperiods (24L : 0D or 15L : 9D), or different combinations of tank colour (black or white) and light intensity (1.1 mol s–1 m–2 or 18 mol s–1 m–2). Growth (0.8% day–1 in standard length; 2.9% day–1 in body area) and survival (2%) were not significantly different between photoperiod treatments after 35 days. Larval survival was greater in white versus black tanks after 41 days (2% versus l%, respectively). Growth of larvae was impaired in black tanks at low (1.1 mol s–1 m–2) light intensity (0.8% day–1 in standard length and 2.2% day–1 in body area versus 1.1% day 21 in standard length and 3.1% day–1 in body area, for all other treatments). Transmission and reflection of light was low in black tanks at low incident light, and there was very little upwelling light. The resultant poor prey to background contrast probably resulted in larvae being unable to consume sufficient food to sustain a level of growth comparable to that in other treatments.  相似文献

We evaluated the effects of enriched rotifers on growth, survival and on the lipid composition of haddock larvae. The treatments tested were (1) AlgaMac 2000®, (2) AquaGrow® Advantage and (3) Pavlova sp. paste and AlgaMac 2000®. The treatments did not influence larval growth rate throughout the experimental period (P = 0.70). Larvae from all treatments grew approximately 8% of their dry weight per day between 1 and 29 days post hatch (dph). Treatment 3 resulted in the best survival, estimated to be 3 on a scale from 0 to 5, whereas for the two other groups the survival estimates were 0 and 2. Rotifers from treatment 1 had low sterol concentrations, high eicosapentaenoic acid/arachidonic acid ratio and their feeding resulted in high larval mortality. Rotifers enriched with Pavlova sp. had the lowest proportions of the sum of saturated fatty acids, docosahexaenoic acid and sum of ω3 and the highest proportions of the sum of monounsaturated fatty acids (ΣMUFA). This was partially reflected in larvae from treatment 3 in that they had the highest proportions of ΣMUFA and the lowest proportions of Σω3 (P < 0.0001 for both analyses). In addition, these larvae had the highest and lowest ΣC20 and ΣC22 polyunsaturated fatty acids (PUFA) respectively (P < 0.0001 for both analyses). We suggest that more research with ω3 and ω6 PUFA can lead to improvements in the rearing of haddock larvae produced in hatcheries.  相似文献

In an effort to optimize the efficiency of high‐density incubation of Atlantic cod Gadus morhua and haddock Melanogrammus aeglefinus eggs, the per cent hatch of eggs treated with four disinfectants (3% hydrogen peroxide, 1% polyvinylpyrrolidone iodine, 0.1% sodium hypochlorite and a 0.005% antibiotic solution – penicillin/streptomycin) was compared in both species. The per cent hatch of eggs of each species was greatest after a 24 h treatment with the antibiotic solution. The hatching success of eggs treated within the different disinfectant treatments depended upon the embryonic developmental stage in both species. The sodium hypochlorite treatment had the lowest % coverage of colony growth after disinfected haddock eggs were plated onto sterile agar media, highest survival rates to the end of the embryonic period, but the lowest per cent hatch.  相似文献

An individual-based modelling approach was developed to investigate the spatial and temporal patterns in the recruitment processes of North Sea haddock, Melanogrammus aeglefinus . The approach was based on the realization that the survivors to recruitment of an annual cohort are most probably not drawn at random from the initial population of eggs, but represent the fastest-growing individuals. Individual growth rates reflect the unique exposure of each larva to the environment along its drift trajectory. In this context, the environment refers to a wide range of factors affecting growth such as food, turbulence and temperature. A combination of a model of egg production by the adult stock, a particle-tracking scheme, and a model of larval growth and mortality rate was used to simulate the dispersal trajectories, and the survival of haddock larvae spawned at different times and locations on the continental shelf. The particle tracking was driven by flowfields from a climatological implementation of the Hamburg Shelf–Ocean Model (HAMSOM) for the North Sea and NE Atlantic. The system was able to resolve spatial and temporal patterns in the recruitment process and indicated that the surviving population of larvae was drawn from a restricted part of the spawning distribution. The results have the potential to guide the development of future conservation measures in fisheries management.  相似文献

Juvenile haddock, Melanogrammus aeglefinus L. (initial weight, 13.5 ± 0.1 g) were fed practical diets containing digestible protein to digestible energy (DP DE?1) ratios of 25–30 g DP MJ DE?1as‐fed using three protein levels (450, 500 and 550 g kg?1) each at two lipid levels (110 and 160 g kg?1) for 63 days. The results showed mean weight gain and feed conversion ratio were highest for diets containing 28.5 and 30.2 g DP MJ DE?1. DP DE?1 ratio had no significant effect on protein efficiency ratio except at the lowest level (24.7 g DP MJ DE?1) indicating a protein sparing effect of higher lipid when dietary protein is below the requirement. Haddock appears to preferentially use protein as the prime source of DE. DP DE?1 ratio had little effect on apparent digestibility (AD) of protein while AD of lipid was significantly affected. Significant differences in AD of energy and organic matter were found to be inversely related to the carbohydrate level of the diet. DP DE?1 ratios of 28.5 g DP MJ DE?1 or lower resulted in significantly higher hepatosomatic indexes. The highest whole‐body nitrogen gains and energy retention efficiencies were achieved at 28.5 and 30.2 g DP MJ DE?1, whereas only slight differences in nitrogen retention efficiencies were observed. The highest levels of energy retained in the form of protein were achieved at 28.5 and 30.2 g DP MJ DE?1. The diet that provided the best growth, feed utilization and digestibility with minimal HSI contained 546 g kg?1 protein (513 g kg?1 DP), 114 g kg?1 lipid, 164 g kg?1 carbohydrate, 17.0 MJ kg DE?1 and a DP DE?1 ratio of 30.2 g DP MJ DE?1.  相似文献

Phosphorus (P) deficiency in the diet of teleost fishes decreases the mineral content of bone. The present study investigated the changes in the microanatomy and metabolism of acellular-boned juvenile haddock (Melanogrammus aeglefinus L.) in relation to the P content of the diet and to determine whether bone deformities develop in association with P deficiency. Fish were fed diets containing low (0.42% diet), optimum (1.02% diet) and excess (1.42% diet) levels of P for 8 and 20 weeks. At the end of two experimental periods, fish were subjected to gross examination, x-rayed, and the mineral content of the plasma and the vertebrae was measured. The vertebrae were also subjected to histological, histomorphometric and enzyme histochemical examinations to determine the level of bone formation and resorption. A P-deficient diet caused a delay in bone mineralization. In contrast, excess P in the diet increased matrix deposition and resulted in accelerated mineralization. Counting the numbers of osteoblasts and osteoclasts, and measuring the mineralized bone and the non-mineralized bone suggest that P deficiency in haddock initially increases bone resorption and subsequently decreases bone mineralization followed by a decrease in bone formation. The increase of the number of osteoclasts in P-deficient animals suggests that osteoclasts could be involved in P homeostasis. In the same fish group low bone mineral and deformed bones observed on x-rays, could be the result of a decrease in mineralization of the bone matrix.  相似文献

Further development of high‐quality feeds for hatchery‐reared haddock in the North Atlantic would benefit from a standard formulation that can be used as a reference for hatcheries and laboratory studies. A practical microparticulate diet (PMD) developed and evaluated with newly metamorphosed juvenile haddock, Melanogrammus aeglefinus L., postlarvae is proposed. Survival of fish fed PMD was just as high (88–89%; P > 0.05) as those fed a high‐quality imported feed (Biokyowa). Alternatively, fish fed PMD had higher (P < 0.05) final fork lengths (39.5 vs. 35.1 mm), wet weights (851.3 vs. 580.2 mg) and weight gains (1637.2 vs. 1115.7%). No differences (P > 0.05) in whole‐body moisture (846–857 g/kg), ash (17–18 g/kg), or protein (101 g/kg) contents were found. Lipid content of fish fed PMD (26 g/kg) was higher (P < 0.05) than those fed Biokyowa (21 g/kg) despite PMD containing 15 g/kg lower dietary lipid, suggesting higher intake and/or lipid retention. The PMD formulation proved to be a highly suitable weaning diet for haddock postlarvae based on high feed acceptance, survival, and fish growth. Given the economic and logistical difficulties associated with importing commercial weaning feeds, this easily produced practical weaning diet has good potential for use by laboratory researchers and farm managers for hatchery‐based nutrition research with haddock postlarvae.  相似文献

A feeding study was conducted in the winter 2001 to determine the effects of feeding rotifers (Brachionus plicatilis) enriched with various levels of essential fatty acids on the growth and survival of haddock larvae (Melanogrammus aeglefinus). Rotifer enrichment treatments were: 1) mixed algae, 2) high DHA (docosahexaenoic acid, 22:6n-3), 3) high DHA and EPA (eicosapentaenoic acid, 20:5n-3), and 4) DHA, EPA, and AA (arachidonic acid, 20:4n-6). Larvae were fed rotifers enriched with the different treatments from days 1 to 16 post-hatch. From day 17 until 25 all treatment groups were fed rotifers reared on mixed algae and then weaned onto the International Council for Exploration of the SEA (ICES) Standard Reference Weaning diet (http://allserv.rug.ac.t/aquaculture/rend/rend.htm) over a five day period. The experiment was terminated on day 41 post-hatch. The enrichment treatments affected the fatty acid composition of the rotifers and correlated with the accumulation of these fatty acids in the haddock larvae. However, no significant differences in larval growth or survival to 40 days post hatch were detected, suggesting that all treatments provided the minimal essential fatty acid requirements for haddock.  相似文献
