Abstract

This study was conducted to ascertain the percent of available K, Cl, and SO4‐S recovered by alfalfa (Medicago sativa L. cv. ‘Vernal') herbage when various rates of K as KCl and K2SO4 were topdressed and also to determine where residual K, Cl, and SO4‐S accumulated in the soil profile. An established stand of alfalfa growing on low fertility silt loam soil was topdressed in the spring of each of two harvest years with 0, 448, 896, 1344, and 1792 kg/ha of K as KCl or K2SO4. Four harvests were taken during each harvest year (1972 and 1973). Soil samples were taken during the autumn of 1973 to a depth of 91.4 cm in KCl‐fertilized plots, and to a depth of 76.2 cm in K2SO4‐fertilized and control plots.

Potassium recovery by alfalfa during two harvest years where K as KCl was applied at 448, 896, 1344, and 1792 kg/ha/yr was 56, 33, 20, and 17%, respectively. Recovery of available Cl from those same treatments was 30, 17, 12, and 10%, respectively. Where K as K2SO4 was applied at 448, 896, 1344, and 1792 kg/ha/yr, 55, 35, 27, and 22%, respectively, of available K was recovered. Recovery of available SO4‐S from those same treatments was 16, 9, 7, and 5%, respectively. At the end of two years, a majority of the residual K was in the top 15.2 cm of soil. Residual Cl and SO4‐S were concentrated at a depth of 30.5 to 76.2 cm in the soil profile.  相似文献

Abstract

Ca, Mg, Na, K and P contents of 3 plant tissues obtained by H2SO4‐H2O2‐Li2SO4‐Se peroxidation digestion were compared with those determined by mixed acid (HNO3‐H2S04‐HC104) and dry ashing procedures. Differences between methods were not consistent between elements or tissue types but the mixed acid digestion generally yielded higher Ca, Mg and P values than did dry ashing and peroxidation. Tissue K contents estimated by both wet digestion techniques were not significantly (P ≤0.05) different. Differences in Na values between digestion methods were highly inconsistent between tissue types and the precision of the Ma data was poor. N contents determined by peroxidation digestion and a conventional Kjeldahl method agreed closely.  相似文献

为预测木瓜蛋白酶在PEG/PEG-IDA-Fe3+/(NH4)2SO4亲和双水相中的分配行为,在298.15K条件下利用浊点法测定并比较了PEG/(NH4)2SO4和PEG/PEG-IDA-Fe3+/(NH4)2SO4的三角相图,建立了木瓜蛋白酶在无亲和配基双水相和含亲和配基双水相中分配模型。考察双水相各组分浓度与木瓜蛋白酶在该体系中分配系数的相关度,基于酶分配系数与双水相中上下相组分浓度差之间较高相关度,提出了木瓜蛋白酶在PEG/(NH4)2SO4双水相中分配系数与上下相组分浓度差的关联模型,模型相对偏差为7.02%。引入亲和配基浓度对酶分配系数的影响因子η,提出了木瓜蛋白酶在PEG/PEG-IDA-Fe3+/(NH4)2SO4亲和双水相中分配系数模型,试验验证,预测值和试验值之间相对误差均在15%以内,能实现亲和双水相中酶的准确预测。研究结果为木瓜蛋白酶在双水相中分配系数的工程计算提供参考。  相似文献

采用室内培养试验研究添加玉米秸秆和Al2(SO4)3对苏打盐碱土pH值、可溶性盐含量和阳离子交换量的影响,旨在为盐碱土合理改良利用提供理论依据。研究发现,加入玉米秸秆和Al2(SO4)3后,苏打盐碱土pH值有不同程度的降低。当Al2(SO4)3加入量为0%~0.8%时,随着Al2(SO4)3加入量的增加,苏打盐碱土pH值急剧下降,而当Al2(SO4)3加入量继续增加时,则pH值变化较缓慢。Al2(SO4)3加入量相同时,玉米秸秆的添加对降低苏打盐碱土的pH值具有略微促进的作用。在玉米秸秆和Al2(SO4)3的共同作用下,苏打盐碱土Na+、K+、Ca2+和Mg2+的含量均升高,而CO32-和HCO3-的含量则随着玉米秸秆和Al2(SO4)3加入量的增加而逐渐降低。玉米秸秆和Al2(SO4)3的加入使苏打盐碱土的阳离子交换量也有所增加,阳离子与土壤胶体上的Na+进行置换,最终使苏打盐碱土碱化度下降。综合而言,试验条件下,15%玉米秸秆和0.8%Al2(SO4)3对苏打盐碱土的改良效果最佳。  相似文献

Anthocyanins from blackberries are natural dietary pigments. The aim of this study was to investigate the occurrence of anthocyanins in fruits of wild Norwegian blackberries and three blackberry ( Rubus fruticosus L.) cultivars and to report the complete identification of cyanidin 3-O-β-(6″-(3-hydroxy-3-methylglutaroyl)glucopyranoside), 5. This new pigment is most probably the same pigment that has previously been reported to occur in various blackberry samples as cyanidin 3-dioxalylglucoside. All of the examined blackberry samples contained in similar relative proportions the 3-glucoside (1), 3-rutinoside (2), 3-xyloside (3), and 3-O-β-(6″-malonylglucoside) (4) of cyanidin and 5. The absolute amounts of 1-5 in the wild Norwegian blackberries were 249, 18, 10, 24, and 22 mg of cyanidin 3-glucoside equivalents/100 g of fresh weight, respectively.  相似文献

Milk proteins contain numerous potential bioactive peptides, which may be released by digestive proteases or by the proteolytic system of lactic acid bacteria during food processing. The capacity of Streptococcus thermophilus to generate peptides, especially bioactive peptides, from bovine caseins was investigated. Strains expressing various levels of the cell envelope proteinase, PrtS, were incubated with α(s1)-, α(s2)-, or β-casein. Analysis of the supernatants by LC-ESI-MS/MS showed that the β-casein was preferentially hydrolyzed, followed by α(s2)-casein and then α(s1)-casein. Numbers and types of peptides released were strain-dependent. Hydrolysis appeared to be linked with the accessibility of different casein regions by protease. Analysis of bonds hydrolyzed in the region 1-23 of α(s1)-casein suggests that PrtS is at least in part responsible for the peptide production. Finally, among the generated peptides, 13 peptides from β-casein, 5 from α(s2)-casein, and 2 from α(s1)-casein have been reported as bioactive, 15 of them being angiotensin-converting enzyme inhibitors.  相似文献

测定了2羟-基-4-甲氧基苯甲醛和2羟-基-4-甲氧基苯甲酸两种效应物对菜粉蝶多酚氧化酶(po lypheno lox idase,简称PPO,EC.1.14.18.1)催化L-多巴(L-DOPA)氧化活力的抑制作用。结果表明,这两种效应物对该酶的活性均有明显的抑制作用,抑制中浓度(IC50)分别为2.71和5.66 mm o l/L,抑制常数分别为2.82和3.35 mm o l/L;2羟-基-4-甲氧基苯甲醛对该酶的抑制作用显著高于2羟-基-4-甲氧基苯甲酸的;这两种抑制剂对酶的抑制作用机理完全不同,2羟-基-4-甲氧基苯甲醛对酶的作用表现为混合型抑制,而2羟-基-4-甲氧基苯甲酸对酶的作用表现为竞争性抑制。研究结果为设计以该酶为靶标的杀虫剂提供理论依据。  相似文献

Abstract

The effects of ambient acidity on NO3 ? and Nh4 + absorption by 26‐day‐old tomato plants (Lycopersicon esculentum Mill.) were examined in solution culture. The absorption rate per unit root mass was measured for 6 hr. The NO3 ? absorption rate from 0.4 mM NaNO, was 36% greater at pH 4.5 than at pH 6.5. In contrast, the NhY absorption rate was approximately 42% greater at pH 5.5 or 6.5 than at pH 4.5. The presence of equimolar NHr from 0.4 mM NH^NO, decreased the NO, absorption rate at pH 5.5 or 6.5 but did not reduce the rate at pH 4.5. The NO, absorption rate was inhibited less at pH 5.5 when equimolar NHr was supplied from 0.2 mM (NH4)2S04 as opposed to NH4NO3. At pH 5.5, the N03 ? absorption rate increased with increased #OPNH4#CP2SO4 concentration. The presence of equimolar NO3 ? supplied as either NaNO3 or NH4NO3 had no effect on the NH4 + absorption rate at pH 5.5 or 6.5. However, at pH 4.5, the NH4 + absorption rate was slightly reduced from NH4NO3 solutions relative to that from a (NH4)2S04 solution.  相似文献

The physiological significance of 5-(3',4'-dihydroxyphenyl)-γ-valerolactone, an important metabolite derived from the catabolism of flavan-3-ols by gut microbiota, has been often overlooked due to the lack of the commercial standard. In the present work, this metabolite has been chemically synthesized, and its analytical parameters and antioxidant capacity have been determined in comparison to other chemical analogues [isomer 3-(3',4'-dihydroxyphenyl)-δ-valerolactone and γ-valerolactone] and other structurally related compounds [(+)-catechin, (-)-epicatechin, and 3-(3,4-dihydroxyphenyl)-propionic acid]. The synthesized compound was also used to perform a targeted analysis in samples collected during the in vitro fermentation of a grape seed flavan-3-ol extract with human fecal microbiota from three healthy volunteers. The time-course formation of 5-(3',4'-dihydroxyphenyl)-γ-valerolactone revealed large interindividual differences among volunteers, with concentrations ranging from 3.31 to 77.54 μM at 10 h of fermentation. These results are further discussed in view of the scarce reports quantifying 5-(3',4'-dihydroxyphenyl)-γ-valerolactone in in vitro fermentation studies, and pharmacokinetic and intervention studies.  相似文献

This study addressed some complementary aspects related to plant Fe nutrition. A field and a greenhouse experiment were conducted to monitor changes in chlorophyll, Fe3+, Fe2+, Ca2+ and K+ along with the progressive evolution of lime‐induced chlorosis, and following soil (Fe‐EDDHA, Fe‐EDTA, Fe‐DTPA, DTPA) and foliar (Fe‐EDDHA, FeSO4, “Fe‐Metalosate") treatments, in a chlorosis‐susceptible ornamental plant, Hydrangea macrophylla, over a year's growing period. Though soil Fe‐EDDHA was the most effective compound in alleviating chlorosis symptoms, it became less so with time and was only partly effective as a foliar spray. Leaf analysis showed that as chlorosis intensified and chlorophyll content decreased, phenanthroline ‐ Fe (Fe2+) decreased with corresponding increases in total iron (Fe3+) and K+ concentrations. The reliability of these chlorosis‐indicators was confirmed as the reverse changes occurred upon chlorosis plant recovery.  相似文献

Growth and N‐P‐K uptake in pumpkin (Curcubita moschata Poir.) cv ‘Libby‐Select’ were studied in dryland and irrigated culture. In both moisture regimes, maximum rates of dry matter accumulation occurred between the early and mid‐fruiting developmental stages. Higher total dry matter production with irrigated than dryland culture was primarily associated with increased shoot growth. Concentrations of N, P, and K in foliage generally decreased as pumpkin age increased. Irrigated pumpkins in conjunction with higher total vegetative dry matter accumulated more N, P, and K than dryland pumpkins. Up through early fruit development, N, P, and K accumulation was primarily in leaves and vines and by the late growth stages was almost entirely in the fruit. Total N, P, and K uptake at late fruiting was estimated at 219, 32, and 228 kg/ha in irrigated pumpkins and 180, 21, and 177 kg/ha in dryland pumpkins. Approximately 58% of the N, 52% of the K, and 68% of the P accumulated by late‐fruiting was absorbed by the plant after the early‐fruiting stage in both moisture regimes. Potassium redistribution from vegetative tissues during late fruit development decreased foliar K contents 32% in dryland pumpkins and 21% in irrigated pumpkins.  相似文献

The area of planting of Vitis vinifera L. cv. “Dattier de Beiruth” (DB) grafted on the SO4 (Selection OPPENHEIM 4) rootstock is significant and constantly growing in Northern Algeria. A hydroponic culture was carried out with DB to investigate the effects of potassium-magnesium (K-Mg) ratio on plant growth and mineral nutrition. Four nutrient solutions were tested on which the K-Mg ratio is variable (nutritive solution (NS)1 = 0.3 K: 3.8 Mg, NS2 = 2.1 K: 2 Mg, NS3 = 2.6 K: 1.5 Mg and NS4 = 3.8 K: 0.3 Mg). The results showed that DB was characterized by a high capacity for K uptake. No significant differences among the treatments were detected in the leaf concentrations of calcium (Ca), however Mg uptake was inhibited by increase of K: Mg in the nutrient solution. Results showed that DB is sensitive to the variation of the cation composition in the nutrient solution. Antagonism between K-Mg was apparent.  相似文献

Abstract

This study was carried out to determine if ammonium bicarbonate‐DTPA soil test (AB‐DTPA) of Soltanpour and Schwab for simultaneous extraction of P, K, Zn, Fe, Cu and Mn can be used to determine the availability index for Se. Five Mollisols from North Dakota were treated with sodium selenate and were subjected to several wetting and drying cycles. These soils were extracted with hot water and with ammonium bicarbonate‐DTPA (AB‐DTPA) solution for Se analysis. Alfalfa plants were grown in these soils in a growth chamber to determine plant uptake of Se. In addition to the above experiment, coal mine soil and overburden materials from Western Colorado were extracted and analyzed as mentioned above.

It was found that hot water and AB‐DTPA extracted approximately equal amounts of Se from Mollisols. A high degree of correlation (r =0.96) was found between Se uptake by plants and AB‐DTPA extractable Se. Extractable level of Se in treated soils was decreased with time due to change of selenate to less soluble Se forms and plant uptake of Se. An AB‐DTPA extractable Se level of over 100 ppb produced alfalfa plants containing 5 ppm or higher levels of Se that can be considered toxic to animals. Soils with about 2000 ppb of extractable Se were highly toxic to alfalfa plants and resulted in plant concentrations of over 1000 ppm of Se. The high rate of selenate (4ppm Se) was less toxic to alfalfa plants in soils of high organic matter content. This lower toxicity was accompanied with lower extractable levels of Se.

The AB‐DTPA solution extracted on the average about 31% more Se than hot water from the mine and overburden samples and was highly correlated with the latter (r =0.92). The results indicated the presence of bicarbonate‐exchangeable Se in these materials.  相似文献

The present work aims at evaluating the effect of cycloheximide at concentrations of between 0.5 and 5mgg–1 on N2O and NO3 production in two slightly alkaline soils, sampled from deciduous woodland and arable cultivation. In the first experiment, peptone was used as the “inducing substrate” for heterotrophic activity, and soil was incubated with cycloheximide (at different concentrations) and/or acetylene (1mll–1) to block induced eukaryotic protein synthesis and ammonia monooxygenase activity, respectively. Peptone addition stimulated N2O and NO3 production significantly in woodland soil, whereas arable soil showed no significant N2O emissions and low NO3 production. Low cycloheximide concentrations drastically reduced N2O emissions in woodland soil, suggesting a potential role of fungi in N2O emissions. However, acetylene was equally effective in blocking N2O emissions and part of NO3 production, so that a possible role of ammonia monooxygenase in an organic-inorganic pathway of N nitrification in fungal metabolism can be hypothesized. A second experiment was carried out on the woodland soil to check if low cycloheximide concentrations had non-target biocidal effects on soil microorganisms. Attention was focused on the range of concentrations which had reduced N2O emission in the woodland soil. The results suggested that at concentrations of cycloheximide between 0.5 and 2mgg–1 any biocidal effect on microbial biomass was negligible in the first 48h; therefore only selective inhibition of protein synthesis could be expected. Abstract

Direct fertilization of peanuts (Arachis hypogaeaL.) with P and K has generally shown few yield responses, resulting in only limited information concerning critical soil‐test levels of P and K. The purpose of the experiments in this report was to determine the critical soil‐test levels of P and K for runner peanuts using the double‐acid extraction procedure. Fertilizer experiments were conducted on farmers’ fields from 1973 to 1986. Site selection was based on soil test data that indicated “medium”; or “low”; levels of available P or K but “high”; in Ca and Mg. Phosphorus and potassium were applied together at all sites at rates of 20 and 74 kg/ha, respectively, as concentrated superphosphate and potassium chloride.

There were yield increases to fertilizer in 6 of the 39 experiments. Soil‐test P for these six ranged between 4 and 53 kg/ha; soil‐test K ranged between 10 and 31 kg/ha. Delineating the yield effect into their P and K components with the aid of multiple regressions of yield on soil test values showed that yield increases were due to the K component of the fertilizer. The critical soil‐test K value was calculated to be 37 kg/ha. Sound mature kernels (SMK) were generally unaffected by fertilizer.  相似文献

Amylose-like α-(1,4)-glucan known as the most favorable substrate for the enzymatic production of cycloamyloses (CAs) using 4-α-glucanotransferase has a solubility issue, which requires an additional solubilization process. In our study, two glucosyltransferases, Synechocystis 4-α-glucanotransferase and Neisseria amylosucrase, were adopted to develop an efficient biocatalytic production process of CAs directly from sucrose. From one-pot synthesis, the maximum CA yield (9.6%, w/w) with 0.3 M sucrose was achieved with 10 units/mL of amylosucrase and 0.1 unit/mL of 4-α-glucanotransferase at 40 °C for a 3 h reaction in a simultaneous dual enzyme reaction mode. The size of linear α-(1,4)-glucan was positively related to the CA productivity by 4-α-glucanotransferase in a hyperbolic manner. Using our innovative bioprocess, there was no practical limitation on the initial sucrose concentration and no use of insoluble linear α-(1,4)-glucan substrate. Consequently, the concomitant dual enzyme reaction converted sucrose directly to CAs via in situ transient linear α-(1,4)-glucan as an soluble intermediate.  相似文献
