Methanogenic archaeal communities inhabiting the paddy field soils in the Kojima Bay polder were investigated using polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE), real-time PCR and sequencing analyses. Soil samples of the plow and subsoil layers were collected in 2006 from four paddy fields that were reclaimed between 1692 and 1954. The DGGE band patterns of the targeted 16S rRNA genes amplified from the extracted DNA from the samples were different from the patterns from the paddy field soils in diluvial and alluvial areas. The numbers of targeted 16S rRNA genes, which were involved with methanogenic archaeal and other archaeal sequences, were approximately 107–108 and 106 g−1 dry soil in the plow and subsoil layers, respectively. Sequences of methanogenic archaeal 16S rRNA genes belonging to Methanocellales (Rice cluster I), Methanosarcinales and Methanobacteriales were obtained from the major DGGE bands. Whereas sequences in Methanomicrobiales, which were predominant methanogens in the diluvial and alluvial paddy fields, were not recovered. Known halophilic and methylotrophic methanogens, which are characteristic of saline and marine environments, were not detected. These results indicate that distinctive methanogenic archaeal communities have developed in the paddy field soils in the Kojima Bay polder.  相似文献

珠江三角洲是华南最大的三角洲平原,全国著名的水稻生产基地,其土壤的形成发育是与.三角洲的形成发展密切相关,并随其演变而不断演化。即泥滩生草或长红树林后,围垦种稻初期,形成沼泽型或酸性盐渍型、盐渍型水稻土,随着种稻时间的延伸,脱沼泽化、脱酸化、脱盐化作用的不断进行,发育成潜育型和潴育型水稻土。在此发育过程,肥力自然趋于下降,但人为的耕作、施肥水平是影响其肥力和发育方向的关键因素。  相似文献

The physical analysis of soil tilth has long been one of the important problems in relationship to the solution of soil productivity. Various methods have been proposed to measure soil tilth and most of them are based upon granulation, porosity, penetration and compressibility of soils. In our laboratory here studies3,4) for the physical improvement of soil are being continued to find a suitable method of reclamation for heavy clayey polder soils of extremely bad structure in Kojima Bay. In this paper several methods for measuring the consistency limits, and consistency of dry and wet soil are reported, and the value of these methods as a measure of soil tilth is discussed.  相似文献

旱改水对水稻幼苗生长的影响及秸秆的改良作用   总被引:3,自引:0,他引:3  
本研究以江汉平原旱改水为研究背景,采用土壤盆栽试验和室内淹水培养相结合的方法,以多年水稻土为对照,研究了多年棉田土旱改水及添加秸秆(9 g·kg-1)对水稻幼苗生长和矿质元素吸收的影响以及土壤氧化还原电位和有效态铁、锰、铜、锌含量变化,为旱改水水稻的种植提供参考。结果表明,棉田土旱改水后,水稻幼苗生长缓慢并出现失绿黄化症状,其地上部干重和叶绿素含量仅分别约为水稻土处理的30%和20%。旱改水处理水稻植株Fe含量显著低于、而Cu和Zn含量则显著高于水稻土处理。棉田土旱改水土壤氧化还原电位(Eh)显著高于水稻土;淹水处理10 d,土壤DTPA-Fe含量仅为水稻土的7%左右,而DTPA-Cu和DTPA-Zn含量则分别是水稻土的1.4~2.5倍和1.6~1.8倍。随着淹水时间的延长,棉田土旱改水土壤有效态铁含量逐渐增加,有效态锰、铜和锌含量呈先升高后降低趋势;到淹水处理的第28 d,棉田土旱改水土壤有效态铁、锰、铜和锌含量与水稻土之间的差异逐渐缩小。Fe不足及Cu过量可能是导致旱改水水稻幼苗生长缓慢、失绿黄化的主要原因。旱改水条件下添加秸秆可以降低土壤的Eh值,提高土壤DTPA-Fe含量及降低土壤DTPA-Cu和DTPA-Zn含量,显著提高旱改水初期水稻幼苗叶绿素含量,但对水稻生物量无显著影响。添加秸秆并不能完全消除旱改水对水稻幼苗生长的抑制作用。  相似文献

The present study compared the capsid gene ( g23 ) of T4-type bacteriophages (phages) in Mn nodules with those in the plow layer soil and subsoils of two Japanese paddy fields by applying the primers MZIA1bis and MZIA6 to DNA extracts from the nodules and soils. The deduced amino acid sequences of the g23 genes in the Mn nodules were similar to those in the plow layer soil and in the subsoils. This result indicated that similar T4-type phage communities developed at these sites and that the diversity of T4-type phage communities was wide enough to cover those in the plow layer soil and in the subsoils. The majority of g23 clones formed several clusters with the clones and phages obtained from far-apart paddy fields, and the sequences of two clones were completely identical to a phage and a clone from other paddy fields at the nucleotide or amino acid level, indicating horizontal transfer of g23 genes between those paddy fields. A clone with a long nucleotide residue (686 bp) and a distribution remote from the other clones in the phylogenetic tree indicated that there were many uncharacterized, novel g23 genes in the paddy fields.  相似文献

时间序列方法是研究土壤发生特性演变的重要途径,而比较土壤变化的重要前提之一是序列中的土壤具有相同的起源,即具有母质的相对均一性。本研究根据史料记载中浙江慈溪海塘修筑年代估计出水稻土的耕作年龄,选择了植稻年龄约为50、300、500、700、1000a以及一个未垦滩涂剖面组成的一个时间序列作为研究对象。利用各种土壤属性参数对该时间序列的母质不连续性(或母质均一性)以及水稻土相对年龄进行了判定和验证。结果表明,时间序列的6个剖面虽然具有微小的差异,但其剖面内与剖面间母质来源相同。在水稻土母质不连续性判定中,去除黏粒的粉粒与粉粒中稳定元素Ti/Zr比值具有较好的指示作用。相对易变的土壤属性参数如碳酸钙、磁化率以及游离铁的剖面分异程度在水稻土相对年龄的判定中具有较好的指示作用。综合这些参数在时间序列中的演化趋势,发现500a剖面与整个序列的变化趋势不相符合,可能是利用历史的差异所致,在相关的性质演变研究中应该从序列中剔除。  相似文献

通过连续4 a多的研究,科学地证明了位于江苏昆山绰墩遗址的稻田群是迄今发现的地球上最早的灌溉稻田群(6 280 a BP),提出了一套诊断古水稻田和古水稻土的技术指标;获得了新石器时期"火耕水溽"——原始灌溉稻作技术的科学证据。阐明了史前水稻土和现代水稻土表层有机质、全氮和全钾含量接近,而全磷、有效磷则是前者大大高于后者的事实,是先民长期以动物残余物肥田的结果。古、今水稻土有机质含量均高于同时期旱地土,揭示了稻田生态系统和水稻土固持大气二氧化碳的巨大潜力。新石器时期水稻土也含有较高的多环芳烃(PAHs)等有机污染物,是源于田间秸秆的焚烧。微生物学的研究表明,史前水稻土产甲烷潜势很低,硝化功能完全丢失。提出了今后应进一步研究的问题。  相似文献

The geochemical behavior of metals, including Fe, Mn, Pb and Zn, in contaminated paddy soils was investigated during the cultivation of rice crops through laboratory microcosm experiments. From the two paddy fields contaminated by mine tailings, Siheung and Deokeum in Korea, paddy soils were collected and analyzed for their geochemical characteristics. The Siheung paddy soil showed higher levels of heavy metals, whereas the higher potential for the release of metals was anticipated due to the extremely acidic conditions at Deokeum. In microcosm experiments of flooded paddy soils over 18 weeks, Fe and Mn were released in subsurface pore waters by reductive dissolution, and Pb and Zn were dissolved in high amounts at the surface by oxidation of sulfides. Although amorphous Fe oxide-rich layers were formed at the surface of both paddy soils, the release of Pb and Zn were controlled at the surface by these layers only under slightly alkaline conditions at Siheung. Lead and Zn were associated with the reducible and carbonate fractions at the surface paddy soil of Siheung from the sequential extraction on core samples collected during the flooded period. In the acidic conditions at Deokeum, Pb and Zn were continuously released until the late stage of flooding. A great increase in the exchangeable fraction of metals was observed after the soils had drained. The bioavailability of metals for rice crops would be high under acidic conditions at Deokeum, despite the lower levels of heavy metal contamination.  相似文献

二氧化碳(CO2)和甲烷(CH4)是重要的温室气体,研究免耕稻田CO2和CH4排放有助于评价稻田免耕技术对全球气候变化及碳循环的影响。本文通过运用静态箱技术和田间原位碱液吸收法研究了免耕稻田土壤CO2和CH4的排放规律和排放量,及其稻田碳(C)的收支状况。研究表明,施肥提高了CH4排放,而不影响CO2的排放;免耕显著影响稻田CH4排放,而CO2的排放不受耕作影响。对稻田C收支及平衡的分析表明,施肥提高了稻田系统C的输入,同时,相对于翻耕处理,免耕处理表现为大气C的“汇”,表明了稻田免耕能将更多的碳累积于农田土壤碳库中,有利于提高稻田生态系统在减缓气温上升过程中所发挥的作用。  相似文献

Paddy fields are one of the largest anthropogenic sources of global CH4 emission. A decrease in paddy CH4 emission can contribute significantly towards the control of global warming. Recent studies have demonstrated that the application of biochar in paddy soils has such a capability, but its underlying mechanism has yet to be elucidated. In this investigation, we studied CH4 emission, methanogenic archaeal, as well as methanotrophic proteobacterial communities, from microcosms derived from two paddy soils, Inceptisol and Ultisol. Both soils were amended with biochar at different pyrolysis temperatures (300 °C, 400 °C and 500 °C) at field condition. The soil CH4 flux was monitored across whole rice season in 2010; the functional guilds communities were analyzed by PCR–DGGE and real-time quantitative PCR (qPCR). It is found that paddy CH4 emissions significantly decreased under biochar amendments, which, interestingly, didn't result from the inhibition of methanogenic archaeal growth. qPCR further revealed that biochar amendments (1) increased methanotrophic proteobacterial abundances significantly, and (2) decreased the ratios of methanogenic to methanotrophic abundances greatly. These results shed insight on the underlying mechanism of how biochar decreases paddy CH4 emission. This knowledge can be applied to develop a more effective greenhouse gas mitigation process for paddy fields.  相似文献

采用15N成对标记技术结合数值模型,测定太湖地区两种土地利用方式(稻麦轮作农田和葡萄园)下的土壤氮素初级转化速率,探讨了土地利用方式改变对土壤供氮和保氮能力的影响。结果表明,葡萄园土壤初级矿化速率高于稻麦轮作农田土壤,但是其NH4+-N同化速率几乎可以忽略不计(0.02 mg kg-1 d-1),自养硝化成为培养条件下葡萄园土壤NH4+-N的唯一去向。葡萄园土壤初级自养硝化速率(15.85 mg kg-1 d-1)显著高于稻麦轮作农田土壤(13.65 mg kg-1 d-1),但两者初级异养硝化速率和NO3--N同化速率均接近零值。可见,太湖地区稻麦轮作农田改种为葡萄园后,土壤NH4+-N同化速率显著降低而自养硝化速率增加,由此导致更多的NO3--N在土壤中累积,进而可能增加土壤中N的淋溶和径流损失风险。  相似文献

湖南几种耕地土壤固定添加铵的动力学研究   总被引:2,自引:0,他引:2  
通过野外调查取样、室内培养试验和分析测定,研究了湖南省几种成土母质发育的旱地土壤和稻田土壤固定添加铵的动力学特性。结果表明,供试土壤对添加铵的固定速度很快,尤其在反应的前8~12h内速度更快,12h后速度逐渐变慢,24h以后,土壤对外源铵的固定已基本达到平衡。数学拟合表明,一级动力学方程和Elovich方程两种动力学模型能较好地拟合供试土壤固定添加铵的动力学特性,经统计均达极显著水平,抛物扩散方程也有较好的拟合效果,零级方程较差。由一级动力学方程求得的不同土壤固铵动力学参数:理论最大固铵量(A)、反应速率常数(b)以及反应半时值明显不同。耕型石灰性紫色土、耕型酸性紫色土、耕型棕色石灰土和耕型石灰岩红壤的理论最大固铵量和反应半时值分别为212.3mg kg^-1、179.0mg kg^-1、142.9mg kg^-1.13.7mg kg^-1和29.75h、25.96h、27.18h、23.49h;紫泥田、河沙泥、灰泥田和红黄泥的理论最大固铵量和反应半时值分别为86.2mg kg^-1、68.7mg kg^-1,31.8mg kg^-1、19.1mg kg^-1和14.50h、15.10h、15.51h、18.43h。耕型石灰性紫色土、耕型酸性紫色土、耕型棕色石灰土和耕型石灰岩红壤的反应速率常数分别为0.0233 h^-1、0.0267h^-1、0.0255h^-1、0.0295h^-1;紫泥田、河沙泥、灰泥田和红黄泥的反应速率常数分别为0.0478h^-1、0.0459h^-1、0.0447h^-1、0.0376h^-1.除耕型石灰岩红壤以外,旱地土壤的理论最大固铵量和反应半时值均明显大于水田土壤,而反应速率常数明显小于水田土壤。  相似文献

Most studies on dissimilatory nitrate reduction to ammonium (DNRA) in paddy soils were conducted in the laboratory and in situ studies are in need for better understanding of the DNRA process.In this study,in situ incubations of soil DNRA using 15 N tracer were carried out in paddy fields under conventional water (CW) and low water (LW) managements to explore the potential of soil DNRA after liquid cattle waste (LCW) application and to investigate the impacts of soil redox potential (Eh) and labile carbon on DNRA.DNRA rates ranged from 3.06 to 10.40 mg N kg 1 dry soil d 1,which accounted for 8.55%-12.36% and 3.88%-25.44% of consumption of added NO 3-15 N when Eh at 5 cm soil depth ranged from 230 to 414 mV and 225 to 65 mV,respectively.DNRA rates showed no significant difference in paddy soils under two water managements although soil Eh and/or dissolved organic carbon (DOC) were more favorable for DNRA in the paddy soil under CW management 1 d before,or 5 and 7 d after LCW application.Soil DNRA rates were negatively correlated with soil Eh (P < 0.05,n=5) but positively correlated with soil DOC (P < 0.05,n=5) in the paddy soil under LW management,while no significant correlations were shown in the paddy soil under CW management.The potential of DNRA measured in situ was consistent with previous laboratory studies;and the controlling factors of DNRA in paddy soils might be different under different water managements,probably due to the presence of different microfloras of DNRA.  相似文献

Paddy fields in the southeastern basin of Dianchi Lake have rapidly changed to greenhouses since 1999. A total of 61 surface soil samples, including 43 greenhouse soils, 12 upland soils, and 6 paddy soils, were collected from a flat lowland area mainly used for agricultural production fields in the southeastern basin of Dianchi Lake. Analyses of the soil samples indicated that the greenhouse soils were characterized by a lower organic matter content, lower pH, and higher soluble nutrients than the paddy soils in the area. The lower organic matter content of the greenhouse soils was ascribed to environmental or management factors rather than the clay content of the soil. Accumulation of soluble nutrients, especially inorganic N, was due to over-application of fertilizers, which also caused soil acidification. The average amount of readily available N, P, and K accumulated in the greenhouse soils was estimated to be equal to or higher than the annual input of these nutrients as a fertilizer, indicating that a reduction in fertilizer application was possible and recommended. In contrast, a very low available Si content was observed in the paddy soils, suggesting the need for Si application for rice production.  相似文献

Experiments were conducted with two typical paddy soils from China and a vermiculite to study the influence of iron oxides on the fixation and release of ammonium. Removing iron oxides, especially amorphous iron oxides, from the soils favoured the release of non-exchangeable NH4-N and stimulated the fixation of NH4-N in the presence of added (NH4)2SO4. Addition of artificial goethite and hematite to the original soils or to the soils free of iron oxides reduced the fixation of NH4+-ions. This effect was also observed with vermiculite. We conclude that the coating of clay minerals with iron oxides has an impact on the diffusion of NH4+-ions into and out of the interlayers of the clay minerals. The reduction and dissolution of iron oxides induced by low redox potential (Eh) after flooding of paddy soils is assumed to be an important mechanism controlling NH4+-fixation in paddy fields.  相似文献

This study attempted to determine the characteristics of the communities, the ecological factors, and the denitrifying enzyme activity for denitrifying bacteria found in the paddy soils of the Taihu Lake Basin, China. Samples of the six main soil types of the basin were taken from paddy fields with different fertilities. The total numbers of bacteria and denitrifying bacteria in the high fertility soils were much more than those in low fertility soils, and the number of denitrifying bacteria accounted for 49% to 80% of the total number of bacteria. The O2 content was an important ecological factor that affected denitrification. Of test the strains isolated from the paddy soils in the Taihu Lake Basin, some (e.g., Pseudomonas spp.) grew well under low oxygen partial pressure, while others (e.g., Bacillus spp.) had no strict predilection with O2 content. Another critical ecological factor was the nitrogen concentration. Three selected denitrifying bacteria grew better in aculture medium with 135 instead of 276mg L^-1 nitrogen. At the same time 67% of the test strains were able to reduce NO3^- to NO2^- and 56% had N2O reductase.  相似文献

Paddy soil management is generally thought to promote the accumulation of soil organic matter (SOM) and specifically lignin. Lignin is considered particularly susceptible to accumulation under these circumstances because of the recalcitrance of its aromatic structure to biodegradation under anaerobic conditions (i.e ., during inundation of paddy fields). The present study investigates the effect of paddy soil management on SOM composition in comparison to nearby agricultural soils that are not used for rice production (non‐paddy soils). Soil types typically used for rice cultivation were selected, including Alisol, Andosol and Vertisol sites in Indonesia (humid tropical climate of Java) and an Alisol site in China (humid subtropical climate, Jiangxi province). These soil types represent a range of soil properties to be expected in Asian paddy fields. All upper‐most A horizons were analysed for their SOM composition by solid‐state 13C nuclear magnetic resonance (NMR) spectroscopy and for lignin‐derived phenols by the CuO oxidation method. The SOM composition was similar for all of the above named parent soil types (non‐paddy soils) and was also not affected by paddy soil management. A substantial proportion (up to 23%) of the total aryl‐carbon in some paddy and non‐paddy soils was found to originate from condensed aromatic‐carbon (e.g ., charcoal). This may be attributed to the burning of crop residues. On average, the proportion of lignin was low and made up 20% of the total SOM, and showed no differences between straw, particulate organic matter (POM), and the bulk soil material. The results from CuO oxidation are consistent with the data obtained from solid‐state 13C NMR spectroscopy. The extraction of lignin‐derived phenols revealed low VSC (vanillyl, syringyl, cinnamyl) values for all investigated soils in a range (4 to 12 g kg−1 OC) that was typical for agricultural soils. In comparison to adjacent non‐paddy soils, the data do not provide evidence for a substantial accumulation of phenolic lignin‐derived structures in the paddy soils, even for those characterized by higher organic carbon (OC) contents (e.g ., Andosol‐ and Alisol (China)‐derived paddy soils). We conclude that the properties of the parent soil types are more important for the lignin content of the soils than the effect of paddy management itself.  相似文献

黄泥田为福建省主要中低产田类型之一,约占水稻土面积的30%。为解析关键限制因子及开展黄泥田肥力质量评价,进而实施针对性的改良措施,采用配对采样方法,采集福建省20对典型黄泥田与邻近同一微地貌单元内高产灰泥田表层土壤,分析了两种土壤类型28项属性因子指标差异及其原因,并采用主成分分析等方法构建福建省黄泥田肥力质量评价因子最小数据集,通过加权指数法分别计算最小数据集土壤肥力质量指数与差异显著因子构成的重要数据集土壤肥力质量指数。结果表明,与灰泥田相比,黄泥田的有机质含量低19.1%,全氮、全磷、全钾含量分别低14.8%、29.9%和25.4%,碱解氮、有效磷和速效钾含量分别低17.8%、56.7%和39.3%,CEC、交换性钙、交换性镁含量分别低12.9%、50.6%和30.8%,有效铁、有效硼和有效锌含量分别低25.6%、33.3%和44.1%。黄泥田的物理性黏粒、0.001 mm黏粒和容重分别较灰泥田高20.8%、25.6%和12.3%,而孔隙度低19.3%。黄泥田过氧化氢酶活性较灰泥田高20.4%,脲酶活性较灰泥田低40.4%。用主成分分析方法从上述19项有显著差异的因子构成的重要数据集中归纳出累计贡献率达76.22%并能反映黄泥田综合肥力特征的6个主成分,建立了由CEC、全钾、有效磷、有效硼和孔隙度5项因子组成的黄泥田肥力评价最小数据集,相应的黄泥田最小数据集土壤肥力质量指数仅相当于灰泥田的69.5%,通过与重要数据集的土壤肥力质量指数相关分析比较,最小数据集可代替重要数据集对福建省黄泥田土壤肥力质量进行正确评价。  相似文献
