We investigated the effect of nursery inoculation techniques on mycorrhizal colonization and sporulation, growth responses, and nutrient (N and P) uptake to determine the suitable nursey inoculation method of wetland rice (Oryza sativa L.) under high-fertility soil conditions. Seedlings were produced in dry-nursery (DN, watered to 60% of –0.03 MPa) and wet-nursery (WN, 3–5 cm water from the soil surface) conditions with or without arbuscular mycorrhizal fungal (Glomus spp.) inoculation. Soil was γ-ray sterilized before use in this experiment. Mycorrhizal fungal colonization was 56% in DN and 23% in WN plants at 6 weeks of growth. The arbuscular mycorrhizal fungal colonization was significantly higher in plants of DN origin than in WN plants after transplantation to the pots, irrespective of growing stages. Mycorrhizal colonization was significantly decreased to 28% in DN plants and to 25% in WN plants at harvest. The grain yield was significantly influenced by nursery conditions. N and P acquisition of wetland rice plants inoculated with Glomus spp. was significantly greater than that of non-inoculated plants at maturity, especially in those originating from DN conditions. P translocation from shoots to grain was accelerated by mycorrhizas.

Mycorrhizal technique is a promising biotechnology in horticultural industry, benefiting plants exposed to diverse abiotic stresses. In this study, the effects of three arbuscular mycorrhizal fungi (AMF), Acaulospora laevis, Glomus mosseae, and Glomus caledonium on plant growth and nutrient uptake of loquat (Eriobotrya japonica Lindl.) seedlings under three water regimes (well watered, water stressed-slight, water stressed-heavy) were investigated. Results showed that inoculated seedlings had higher dry biomass, plant height, and total leaf areas than those un-inoculated ones. AMF effect was the greatest for water stressed-heavy seedlings, followed by water stressed-slight seedlings and well watered seedlings. All AMF species increased the uptake of nitrogen (N) potassium (K), phosphorus (P), calcium (Ca), magnesium (Mg), zinc (Zn), copper (Cu), and the mycorrhizal contributions to the nutrient uptake were positively related to that to the biomass. Data suggest that AMF inoculation increases the tolerance of loquat seedlings to drought stress, and the improved nutrient uptake by AMF contributes greatly to the tolerance.

Effects of arbuscular mycorrhizal fungi (AMF) on the growth, nutrient absorption, and inoculation effectiveness of AMF on pioneer plants Pharagmites japonica (C4) and Polygonum cuspidatum (C3) were evaluated by performing a pot experiment in a greenhouse at Saitama University, Japan. AMF spores were collected from the commercial product, Serakinkon. The average colonization levels of P. japonica and P. cuspidatum were 24–33% and 0.2–0.5% respectively and no colonization was found in sterilized soil treatment. AMF colonization increased the plant dry mass, phosphorus (P), and nitrogen (N) concentrations of P. japonica’s roots, stems, and leaves when AMF applied with natural and sterilized soil compared with only sterilized and natural soil. This was a significant effect for N-loss minimization from soil. Maximum value showed when P. japonica was grown with natural soil in combination with AMF whereas P. cuspidatum showed very less or a negative response to AMF colonization in all cases.

The effect of the application of acidified porous hydrate calcium silicate (APS) in nursery bed soil and porous hydrate calcium silicate (PS) in paddy fields on the growth of rice plants ( Oryza sativa L. cv. Hitomebore) was examined in 2002 and 2003. The results revealed the following: 1) Shoot dry weight of rice seedlings increased by APS treatment in nursery bed soil. The tiller number of rice plants after transplanting in both years also increased by APS treatment in nursery bed soil, and in 2003, the tiller number in the treatment with a combination of APS in nursery bed soil and PS in paddy fields was significantly higher than that in the other treatments until the maximum tiller number stage. Furthermore, the root length of rice plants 14 d after transplanting increased by APS treatment in nursery bed soil. 2) Silicon concentration in the soil solution significantly increased by PS treatment in paddy fields, and the concentration of dissolved carbon oxide increased by APS treatment in nursery bed soil. 3) Only in the APS treatment the rice yield was 341 g m−2, while 400 and 450 g m−2 in the PS and both APS and PS treatments, respectively, in 2003. Percentages of ripened grains in the plots without PS treatment ranged from 57 to 63%, respectively, while, those in the PS treated plots were 82%. The numbers of panicles and ripened grains in both APS and PS treatments were the highest among the treatments. Based on the above results, we concluded that both APS in nursery bed soil and PS in paddy field treatments were effective in improving the silicon nutrition and growth of rice plants, and that this effect was enhanced by a combination of treatments with the two.

Arbuscular mycorrhizal fungi (AMF) have the capability to improve crop yields by increasing plant nutrient supply. A pot experiment was conducted under natural conditions to determine the response of AMF inoculation on the growth of maize (Zea mays L.), sorghum (Sorghum bicolor L.), millet (Pennisetum glaucum L.), mash bean (Vigna mungo L.), and mung bean (Vigna radiata. L.) crops during 2008. The experiment was conducted as a completely randomized design in three replications using phosphorus (P)–deficient soil. Three plants were grown in 10 kg soil up to the stage of maximum growth for 70 days. Spores of AMF were isolated from rhizosphere of freshly growing wheat and berseem crops and mixed with sterilized soil with fine particles. Crops were inoculated in the presence of indigenous mycorrhiza with the inoculum containing 20 g sterilized soil mixed with 40–50 AMF spores. Inoculation with AMF improved yield and nutrient uptake by different crops significantly over uninoculated crops. Inoculated millet crop showed 20% increase in shoot dry matter and 21% in root dry matter when compared with other inoculated crops. Increases of 67% in plant nitrogen (N) and iron (Fe) were observed in millet, 166% in plant P uptake was observed in mash beans, 186% in zinc (Zn) was measured in maize, and 208% in copper (Cu) and 48% in manganese (Mn) were noted in sorghum crops. Maximum root infection intensity of 35% by AMF and their soil spore density were observed in millet crop followed by 32% in mash beans. Results suggest that inoculation of AMF may play a role in improving crop production and the varied response of different crops to fungi signifies the importance of evaluating the compatibility of the fungi and plant host species.

Nutrient-poor, sandy soils form the prevailing substrate at post-mining sites of the Lusatian region(Brandenburg, Germany) and present a challenge for vegetation development. We studied the organic acid quantity and composition of three commonly occurring pioneer plant species, the legumes Lotus corniculatus L. and Trifolium arvense L. and the grass Calamagrostis epigeios(L.) Roth, to determine if plant growth and exudation differed with(non-sterilized soil) and without(sterilized soil) an indigenous soil microbial community. We investigated whether organic acids were found in the rhizosphere and surrounding soil and whether this influenced nutrient mobilization. This study consists of linked field investigations and a greenhouse experiment. Plants were grown in the greenhouse in either sterilized or non-sterilized sandy soil from a reclamation site in the Lusatian mining landscape(Welzow Su¨d, East Germany). After seven months, the plant biomass, root morphology, organic acids, and water-soluble nutrients and root colonization with arbuscular mycorrhizal fungi(AMF) and dark septate endophytes(DSE) were analyzed. Roots of all three plants in the field and greenhouse experiments were highly colonized with AMF. Calamagrostis epigeios and T. arvense had a significantly higher colonization frequency with DSE than L. corniculatus. The quantity and composition of organic acids strongly differed among plant species, with the highest number of organic acids found for L. corniculatus and lowest for C. epigeios. The quantity of organic acids was greatly reduced in all plants under sterilized soil conditions. However, the composition of organic acids and plant growth in sterilized soil were reduced for both legumes, but not for C. epigeios, which had a higher biomass under sterilized conditions. Changes in nutrient concentrations in the field rhizosphere soil relative to those in the control were measurable after seven months. While the spectrum of organic acids and the growth of legumes seemed to be dependent on a highly diverse soil microbial community and a symbiotic partner, the grass C. epigeios appeared capable of mobilizing enough nutrients without an indigenous microbial community, and might be more competitive on sites where soil microbial diversity and activity are low.

It is well known that arbuscular mycorrhizal fungi (AMF) effects on plant growth largely depend on fungus identity. The objective of this study was to test whether three individual AMF isolates and their mixture mitigate drought stress (DS) differentially in soybean (Glycine max) genotype, predicting that under DS, the mixture of the AMF isolates would provide greater benefits to soybean plants than individual ones. In a greenhouse experiment, a drought-susceptible soybean genotype was inoculated with Septoglomus constrictum, Glomus sp., and Glomus aggregatum, known to be among the most abundant in agricultural and natural soils from central Argentina, and their mixture (Mx). Whereas under well-watered (WW) conditions, individual isolates and Mx treatment were similarly infective; under DS conditions, the Mx treatment showed lower rates of root colonization. Between WW and DS conditions, biomass was decreased in all treatments, although this effect was more marked in non-AM plants. Moreover, AMF strains improved water content and P and N concentrations. Under DS, the Mx treatment was unable to exceed the highest contents that were recorded by AMF isolates. However, under WW conditions, the Mx treatment showed a higher N content than individual isolates. Under both watering conditions, AM plants reduced oxidative damage evaluated as malondiadehyde and chlorophyll content and keep constant osmotic metabolites such as soluble sugars and proline content, without significant differences between AMF isolates and the Mx treatment. These results show that AMF play an important role in mitigating drought impacts on soybean, but that mixtures of AMF isolates did not perform as well as the best single strain inoculum, excluding complementarity effects and suggesting selection effect of AMF on DS alleviation in soybean.

Cowpea is an important crop that serves as a legume and vegetable source to many smallholder farmers in sub-Saharan Africa. Soil fertility is a significant limitation to its production thus; inoculation with beneficial soil biota such as arbuscular mycorrhizal fungi (AMF) could improve its performance. However, plant–AMF interaction could vary based on crop cultivar hence affecting overall crop production. The present study aimed at determining the effect of AMF inoculation and soil sterilization on root colonization and growth of a wild-type and three modern cowpea cultivars grown by smallholder farmers in Kenya. Potted cowpea plants were inoculated with a commercial AMF inoculum comprising of Rhizophagus irregularis, Funneliformis mosseae, Glomus aggregatum and Glomus etunicatum and maintained in a greenhouse for 40 days. After harvesting, mycorrhizal colonization, nodule number and dry weight, root and shoot dry weights, nitrogen (N,) phosphorus (P) and potassium (K) content were determined. Interestingly, the modern cultivars showed significantly (p < 0.001) higher root colonization, nodulation, shoot P and N compared to the wild-type cultivar. Moreover, a strong positive correlation between AMF root colonization and shoot P (r2 = 0.73, 0.90, p < 0.001), AMF root colonization and shoot N (r2 = 0.78; 0.89, p < 0.001) was observed in both sterilized and non-sterilized soil, respectively. Soil sterilization affected root colonization and growth parameters with plants grown in non-sterilized soil performing better than those grown in sterilized soil. This study provides major evidence that modern cowpea cultivars are still responsive to mycorrhizal inoculation suggesting that modern breeding programs are not deleterious AMF symbiosis.

Arbuscular mycorrhizal inoculation and composted residue application are being assayed to aid restoration of desertified areas under Mediterranean climate. The particular objective of the present study was to assess the short-term (8 months) effects on the initial stages of plant performance and on mycorrhizal propagule release, key factors to decide further developments in the restoration process. Mycorrhizal inoculation, with Glomus intraradices, was practised during nursery production of representative shrub species from Mediterranean ecosystems and composted residues were added to soil before transplanting to a desertified area in southern Spain. Pistacia lentiscus, Rhamnus lycioides, Olea europaea subsp. sylvestris and Retama sphaerocarpa, key species from the natural succession in the target area, were the test plants. Mycorrhizal inoculation, and in some cases compost addition, improved the ability for nutrient acquisition by plants upon transplanting in the field. The number of "infective" mycorrhizal propagules was higher in soil around mycorrhiza-inoculated shrubs than that around the corresponding non-inoculated controls. The organic amendment significantly increased propagule production in the rhizosphere of mycorrhiza-inoculated plants. The number of mycorrhizal spores was relatively low in soil around transplants, being hardly affected by treatments. Only three distinguishable glomalean spore morphotypes were found, belonging to the species Glomus geosporum, G. contrictum and Scutellospora calospora, with very few unidentified spores, corroborating the low diversity in degraded ecosystems. An increased development of the extramatrical AM mycelium was found in soil around the roots of the four mycorrhiza-inoculated test plants, probably the main source of AM fungal propagules in the ecosystem at this stage of plant development. In conclusion, the tailored AM inoculation assayed was functioning under field conditions to enhance nutrient acquisition by the target indigenous shrubs and, in interaction with organic amendments, promoted mycorrhizal propagule production in soil, critical factors to benefit further stages of the revegetation process.

Arbuscular mycorrhizal fungi (AMF) alter heavy metal acquisition by higher plants and may alter plant response to soil-contaminating heavy metals. Two communities comprised of Glomus intraradices and G. spurcum were investigated for their influence on copper (Cu) and zinc (Zn) resistance of Sorghum bicolor. One community was isolated from a Cu- and Zn-contaminated soil (AMF-C) and one consisted of isolates from non-contaminated soil (AMF-NC). Non-mycorrhizal (NM) sorghum plants were also included. The two community ecotypes differed in their capacity to protect sorghum from Cu and Zn toxicity and exhibited differential metal uptake into hyphae and altered heavy metal uptake by roots and translocation to plant shoots. AMF-C reduced Cu acquisition under elevated Cu conditions, but increased Cu uptake and translocation by sorghum under normal Cu conditions, patterns not exhibited by AMF-NC or NM plants. Hyphae of both fungal ecotypes accumulated high concentrations of Cu under Cu exposure. AMF-C exhibited elevated hyphal Zn accumulation and stimulated Zn uptake and translocation in sorghum plants compared to AMF-NC and NM plants. Differences in metal resistance between fungal treatments and between mycorrhizal and non-mycorrhizal plants were not related to differences in nutrient relations. The enhanced Cu resistance of sorghum and altered patterns of Cu and Zn translocation to shoots facilitated by AMF isolated from the metal-contaminated soil highlight the potential for metal-adapted AMF to increase the phytoremediation potential of mycotrophic plants on metal-contaminated environments.

Arbuscular Mycorrhizae Fungi (AMF) inoculations may improve growth and nutrient uptake of cotton (Gossypium hirsutum L.) plant. Although the importance of mycorrhizal symbioses for growth and nutrient acquisition of cotton plant is known, less is known about mycorrhizal dependency on P and Zn nutrition under low Zn fertile soil conditions. A greenhouse experiment was conducted to investigate the effect of different of P and Zn fertilizer addition on cotton plant growth as well as Zn and P uptake. Sterilized and non-sterilized low Zn fertile Konya series soil was treated with different levels of P and Zn. Soils were inoculated with two mycorrhizae species like Funneliformis mosseae and Claroideoglomus etunicatum after sterilization. Results showed that mycorrhizal inoculation on plant growth and nutrient uptake has significant effect when soil was sterilized. Cl. etunicatum mycorrhizae species has greater effect than Fu. mosseae mycorrhizae species. Root colonization increased 23–65% due to mycorrhizal amendment. The shoot: root ratio increased by 13 and 22% for non-sterile and sterile condition respectively in mycorrhiza amended soil. Mycorrhizal dependency varies 1–55% and 3–64% for non-sterile and sterile soil respectively on mycorrhizae, P and Zn amended soil. Mycorrhizal dependency analysis showed that cotton plant in both sterile and non-sterile soil conditions depends on mycorrhizae species, P nutrition, however is less depend on Zn nutrition. This study concluded that the inoculation of cotton plant with selected mycorrhizae is necessary under both sterile and non-sterile soil conditions.

Drought stress greatly affects the growth and development of plants in coal mine spoils located in the Inner Mongolia grassland ecosystem. Arbuscular mycorrhizal fungi (AMF) can increase plant tolerance to drought. However, little is known regarding the contribution of AMF to plants that are grown in different types of coal mine spoils under drought stress. To evaluate the mycorrhizal effects on the drought tolerance of maize (Zea mays L.) grown in weathered (S1) and spontaneously combusted (S2) coal mine spoils, a greenhouse pot experiment was conducted to investigate the effects of inoculation with Rhizophagus intraradices on the growth, nutrient uptake, carbon:nitrogen:phosphorus (C:N:P) stoichiometry and water status of maize under well-watered, moderate and severe drought stress conditions. The results indicated that drought stress increased mycorrhizal colonization and decreased plant dry weights, nutrient contents, leaf moisture percentage of fresh weight (LMP), water use efficiency (WUE) and rehydration rate. A high level of AMF colonization ranging from 65 to 90% was observed, and the mean root colonization rates in S1 were lower than those in S2. In both substrates, inoculation with R. intraradices significantly improved the plant growth, P contents, LMP and WUE and decreased the C:P and N:P ratios of plants under drought stress. In addition, maize grown in S1 and S2 exhibited different wilting properties in response to AMF inoculation, and plant rehydration after drought stress occurred faster in mycorrhizal plants. The results suggested that inoculation with R. intraradices played a more positive role in improving the drought stress resistance of plants grown in S2 than those grown in S1. AMF inoculation has a beneficial effect on plant tolerance to drought and effectively facilitates the development of plants in different coal mine spoils.

The impact of arbuscular mycorrhizal fungi (AMF), inorganic phosphorus (P), and irrigation regimes was studied in an okra (Abelmoschus esculentus)–pea (Pisum sativum) cropping system in an acidic Alfisol. Experimentation was carried out at Palampur, India, in a randomized bock design (RBD), replicated three times with fourteen treatments comprising AM fungi (Glomus mosseae), inorganic phosphorus (50, 75, and 100% soil-test-based recommended P dose), irrigation regimes (at 40 and 80% available water capacity), generalized recommended NPK and irrigations (GRD), and farmers’ practice. Effects of AM fungi on plant height, leaf area index (LAI), and dry-matter accumulation (DMA) were nominal at early crop growth stage, i.e., 50 DAS (days after sowing). However, at 100 DAS, AMF imbedded treatments led to higher plant height (4%), LAI (3%), and DMA (6%) in okra, whereas in pea the magnitude of increase in these parameters following AMF inoculation was 6, 5, and 8%, respectively, over non-AMF counterparts. AMF + 75% soil-test-based P dose at either of these irrigation regimes gave statistically similar yields in both okra and pea as that obtained under 100% soil-test-based P dose at either of two irrigation regimes, thus indicating an economy of about 25% in soil-test-based P dose. Regarding nutrient harvest index in okra and pea, statistically similar values were registered with most nutrients under both AMF inoculated and non-AMF counterparts. In the case of okra, P harvest index was registered less by 3% with AMF inoculation; however, its magnitude increased by 3% in pea following AMF inoculation compared to non-AMF counterparts at similar levels of P and irrigation. At completion of two cycles of okra-pea system, AMF imbedded treatments did not alter available soil nutrient status significantly in comparison to non-AMF counterparts. Overall, current study suggests that practice of AMF inoculation has great potential in enhancing growth parameters for better productivity, fertilizer-P economy, and nutrient harvest efficiency in okra-pea production system in Himalayan acidic Alfisol.

A pot experiment was used to evaluate the effects of an arbuscular mycorrhizal fungus (AMF) Funneliformis mosseae on plant growth performance, root-hair growth, and root hormone levels in trifoliate orange (Poncirus trifoliata) seedlings under well-watered (WW) and drought stress (DS). A 9-week mild DS treatment significantly reduced mycorrhizal colonization of 2nd- and 3rd-order lateral roots. Root mycorrhizal colonization was relatively higher in the 2nd- and 3rd-order lateral roots than in the taproot and the 1st-order lateral root under WW and DS. AMF seedlings exhibited significantly higher root-hair density, length (except for the taproot) and diameter in taproot and 1st-, 2nd-, and 3rd-order lateral roots under WW, and considerably higher root-hair density (except for 1st-order lateral root), length (except for 2nd-order lateral root) and diameter under DS. Mycorrhizal inoculation remarkably increased root abscisic acid (ABA), indole-3-acetic acid (IAA), methyl jasmonate, and brassinosteroids (BRs) concentrations under DS, in company with the decrease in root zeatin riboside and gibberellins levels and root IAA effluxes. Root-hair traits were significantly positively correlated with root colonization and root ABA and BRs levels. It is concluded that mycorrhizal plants possessed better root-hair growth to adapt mild DS, which is associated with mycorrhizal colonization and endogenous hormone changes.

A pot experiment was conducted to study the plant growth and fruit yields of cucumber (Cucumis sativus L.) on a greenhouse soil with or without inoculation of arbuscular mycorrhizal fungi (AMFs) and Fusarium oxysporum f. sp. cucumerinum under unsterilized conditions. Two AMF inocula were tested: only one AMF strain Glomus caledonium 90036 and an AMF consortium mainly consisting of Glomus spp. and Acaulospora spp. There were four treatments including no inoculation (control), inoculation with F. oxysporum but without mycorrhizae (FO), inoculation with F. oxysporum and G. caledonium (FO+M1), and inoculation with F. oxysporum and the AMF consortium (FO+M2). Cucumber plants were harvested at weeks 3 and 9 after transplanting. Compared with the control, the FO treatment without AMF inoculation had less biomass both at weeks 3 and 9 (P < 0.05) and had higher incidence of Fusarium wilt and produced no cucumber fruit at week 9. Both FO+M1 and FO+M2 treatments had higher mycorrhizal colonization than the treatments which received no AMF inoculation at week 3 (P < 0.05), but only the FO+M2 treatment elevated plant biomass, decreased the incidence of Fusarium wilt, and improved cucumber yields to the same level as the control at week 9. The results indicated that the AMF consortium could suppress Fusarium wilt of cucumber and, therefore, showed potential as a biological control agent in greenhouse agroecosystems.

Tomato (Lycopersicon esculentum Mill.) yields are known to decrease for plants grown in saline soils. This study was conducted to determine the effects of arbuscular mycorrhizal fungi (AMF) inoculation on fruit yield and mineral content of salt-tolerant and salt-sensitive tomato cultivars grown with varied levels of salt. NaCl and CaCl2were added to soil in the irrigation water in equal molar ratios to give ECe values of 1.4 (nonstressed) and 4.9 dS m?1 (salt stressed). Plants were grown in a greenhouse using unsterilized, low phosphorus (P) (silty clay) soil-sand mix. Mycorrhizal root colonization occurred whether cultivars were salt stressed or nonstressed, but the extent of AMF root colonization was higher in AMF inoculated than uninoculated plants. The salt tolerant cultivar ‘Pello’ generally had higher AMF root colonization than the salt sensitive cultivar ‘Marriha’. Shoot dry matter (DM) yield, fruit fresh yield, and AMF colonization were higher for plants grown under nonstressed than for plants grown under salt stressed conditions. Shoot DM and fruit fresh yields were higher in AMF inoculated than uninoculated plants grown with or without salt stress. Pello generally had higher fruit fresh yield per plant and fruit weight than Marriha, but these differences were only significant for fruit weight in unioculated plants grown under salt stressed conditions. The enhancement in fruit fresh yield due to AMF inoculation was 26 and 23% under nonstressed and 28 and 46% under salt stressed treatments for Pello and Marriha, respectively. For both cultivars, fruit contents of P, potassium (K), zinc (Zn), copper (Cu), and iron (Fe) were higher in AMF inoculated compared with uninoculated plants grown under nonstressed and salt stressed conditions. Fruit Na concentrations were lower in AMF inoculated than uninoculated plants grown under salt stressed conditions. The enhancement in P, K, Zn, Cu, and Fe acquisition due to AMF inoculation was more pronounced in Marriha than in Pello cultivar under salt stressed conditions. The results of this study indicated that AMF inoculated plants had greater tolerance to salt stress than unioculated plants.
