为明确不同发育阶段小麦赤霉病菌对甾醇生物合成抑制剂类(SBIs)杀菌剂的敏感性差异及杀菌剂的作用方式,对4种SBIs类杀菌剂进行了室内毒力测定及田间药效试验。室内毒力测定结果表明:不同发育阶段小麦赤霉病菌对4种SBIs类药剂的敏感性存在较大差异,其敏感性由高到低依次为菌丝生长阶段和产孢阶段芽管伸长阶段孢子萌发阶段;其中,戊唑醇对菌丝生长、产孢、芽管伸长和孢子萌发阶段的EC50值分别为0.686、0.191、3.532和3.825μg/m L,咪鲜胺分别为0.063、0.305、0.827和36.581μg/m L,苯醚甲环唑分别为0.873、3.659、5.687和79.465μg/m L,烯唑醇分别为1.961、33.658、41.881和54.986μg/m L。显微观察4种SBIs类杀菌剂对小麦赤霉病菌菌丝生长和孢子萌发形态的影响,发现杀菌剂处理后菌丝末端分枝增多,新生菌丝伸长受阻,菌丝体局部膨大,有时可见菌丝及芽管的消解。田间试验结果表明:用量为有效成分100 g/hm2的戊唑醇、200 g/hm2的咪鲜胺和200 g/hm2的苯醚甲环唑对小麦赤霉病的保护效果为77.15%~79.26%,治疗效果为70.64%~72.77%,于发病前后使用均可有效防治该病害。  相似文献

腔孢叶斑病是牡丹生产上出现的一种新病害,近几年在洛阳及菏泽牡丹种植园危害越来越重。为了筛选防治该病的化学药剂,本文采用菌丝生长速率法测定了3类8种杀菌剂对病菌菌丝生长的抑制活性,采用涂布平板法测定了多菌灵、戊唑醇及嘧菌酯对病菌孢子萌发和芽管伸长的影响,并在含药平板上测定了3种药剂对病菌产孢量的作用。结果表明:三唑类药剂对病菌菌丝生长的毒力最强,EC50为0.020~0.096μg/mL,且戊唑醇对孢子萌发和产孢也有很强的抑制活性,在0.1μg/mL时孢子即不能萌发;甲氧基丙烯酸酯类药剂对菌丝生长的毒力较强,EC50为0.408~0.939μg/mL,嘧菌酯对芽管伸长和产孢量均表现出很强的抑制活性,10μg/mL时,芽管几乎不再伸长,对产孢的抑制率可达到99%以上;而苯并咪唑类药剂对菌丝生长的毒力较弱,EC50为12.167~30.104μg/mL,多菌灵对孢子萌发的抑制作用弱,100μg/mL时孢子萌发率仍达到90%以上,对产孢的抑制作用也较弱,为63.02%。根据药剂对病菌生长和发育各阶段的影响,生产中应采用适宜的施用方法:戊唑醇和嘧菌酯可作为保护剂在病害发生前期喷施,也可作为治疗剂在病害发生流行期应用,但为防止抗药性菌株的出现,两者应当轮换或复配使用。  相似文献

不同杀菌剂对黄瓜靶斑病菌的毒力作用特性比较   总被引:2,自引:2,他引:2  
为筛选防治黄瓜靶斑病的有效药剂,采用菌丝生长速率法、孢子萌发法和芽管伸长法测定了19种杀菌剂对黄瓜靶斑病菌的毒力,探究了4种毒力较高杀菌剂对病原菌菌丝形态的影响。结果表明:氟啶胺、吡唑萘菌胺、咯菌腈和四霉素对黄瓜靶斑病菌菌丝生长、孢子萌发和芽管伸长各生长发育阶段均有较强的抑制活性,其中,4种药剂抑制菌丝生长的平均EC50值分别为0.516 3、1.538、1.605和0.648 2 mg/L,抑制孢子萌发的平均EC50值分别为0.164 4、1.359、1.127和0.002 5 mg/L,抑制芽管伸长的平均EC50值分别为0.189 6、0.144 7、0.060 4和0.001 5 mg/L;啶酰菌胺、百菌清和肟菌酯3种不同作用机制杀菌剂对黄瓜靶斑病菌孢子萌发和芽管伸长的抑制活性较强,3种药剂抑制孢子萌发的平均EC50值分别为1.564、0.373 0和0.021 3 mg/L,抑制芽管伸长的平均EC50值分别为0.629 5、0.233 4和0.405 0 mg/L,但对菌丝生长的抑制作用较弱,平均EC50大于16 mg/L;氟啶胺、吡唑萘菌胺、咯菌腈和四霉素可引起黄瓜靶斑病菌菌丝不同程度的异常生长,主要表现为菌丝隔膜间距变小、菌丝节间肿大、主菌丝上二次分枝发生频繁、菌丝变形黏连、菌丝新分枝处生长受抑制或分生孢子生长变形。研究表明,氟啶胺、吡唑萘菌胺、咯菌腈和四霉素对黄瓜靶斑病菌具有较高的毒力,在黄瓜靶斑病的田间防治中具有较好的开发应用潜力。  相似文献

月季黑斑病病原菌鉴定及室内药剂初步筛选   总被引:2,自引:0,他引:2  
为有效防控月季黑斑病,于2017年6—7月自运城学院花圃采集具有典型黑斑症状的月季病叶,分离纯化获得病原菌,利用形态学特征、分子生物学特征及致病性测定对其进行鉴定;同时室内测定5种常用药剂对该病原菌的防治效果,筛选能有效抑制该病原菌菌丝生长及孢子萌发的药剂。本试验共分离纯化得到12株菌株,其病原菌形态特征一致、rDNA-ITS、GPD、EF-1α基因扩增序列均相同。结合形态学特征观察以及rDNA-ITS、GPD、EF-1α基因联合构建的系统发育树将运城月季黑斑病病原菌鉴定为链格孢Alternaria alternata。在5种药剂中,50%咯菌腈和50%异菌脲各浓度对月季黑斑病病原菌的抑菌效果均较好,对菌丝生长抑制率和孢子萌发抑制率分别达到95.2%和96.6%以上;其次是70%代森锰锌,3个浓度的菌丝生长抑制率在84.1%~87.2%之间,3个浓度的孢子萌发抑制率均高于93.5%,其中600倍液的孢子萌发抑制率最高,达98.1%;80%多菌灵和12.5%腈菌唑对月季黑斑病病原菌的抑制效果最差,菌丝生长抑制率和孢子萌发抑制率分别低于42.4%和46.3%。毒力测定结果显示,50%咯菌腈毒力最强,抑制中浓度EC_(50)为25.8 mg/L;12.5%腈菌唑毒力较弱,EC_(50)为160.3 mg/L,其它3种药剂的EC_(50)介于30.4~60.7 mg/L之间。因此,50%咯菌腈可湿性粉剂和50%异菌脲悬浮剂可作为防治由链格孢引起的月季黑斑病的优选药剂。  相似文献

用平板菌落法测定了杀菌剂对玉米顶腐病菌菌丝生长、产孢、孢子萌发及芽管伸长的抑制作用。结果表明:浓度为1000μg/mL的60%氟吗·锰锌可湿性粉剂、15%三唑酮可湿性粉剂、47%腈菌·锰锌可湿性粉剂3种杀菌剂对菌丝生长抑制率为100%。1000μg/mL和500μg/mL供试各杀菌剂能抑制分生孢子萌发,50%多菌灵可湿性粉剂和77%氢氧化铜可湿性粉剂抑制孢子萌发EC50分别为0.2719μg/mL和762.8854μg/mL。供试杀菌剂抑制芽管伸长的EC50差异显著,47%腈菌·锰锌可湿性粉剂EC50为0.1268μg/mL,70%甲基硫菌灵可湿性粉剂EC50为383.9653μg/mL。杀菌剂对分生孢子产生抑制作用显著,产孢量显著小于对照,60%氟吗·锰锌可湿性粉剂能完全抑制孢子形成。  相似文献

用Calcofluor荧光染色和显微观察的方法研究了6种常用杀菌剂对黄瓜霜霉病菌Pseudoperonospora cubensis不同发育阶段的影响。结果表明:烯酰吗啉和氟吗啉对黄瓜霜霉病菌孢子萌发、芽管伸长、附着胞形成、菌丝扩展、孢囊梗和孢子囊的形成具有明显的抑制作用,其EC50值分别在0.07~5.98 μg/mL和0.10~3.50 μg/mL之间,而对游动孢子释放、游动孢子游动和休止孢形成没有抑制作用;霜脲氰对休止孢萌发和芽管伸长的EC50值分别为1.10和0.002 μg/mL,对其他阶段无明显的抑制效果;腈嘧菌酯对休止孢萌发、菌丝扩展和孢囊梗产生无影响,对游动孢子释放、芽管伸长和附着胞形成的EC50值分别为0.018、0.008和0.010 μg/mL;代森锰锌对游动孢子释放、休止孢萌发和芽管伸长的EC50值分别为2.40、2.88和1.53 μg/mL;甲霜灵对该菌的整个发育期均没有影响,这可能是因为供试菌株对甲霜灵已经产生了抗药性。  相似文献

为探索甘薯长喙壳菌Ceratocystis fimbriata对咯菌腈的敏感性,明确咯菌腈在苗期和储藏期对甘薯黑斑病的防治效果,分别从中国河南、四川、河北、山东采集并分离得到64株甘薯长喙壳菌;采用凹玻片法观察了咯菌腈对病原菌分生孢子萌发和芽管伸长的影响;采用孢子萌发法和菌丝生长速率法测定了64个菌株对咯菌腈的敏感性,并对50%咯菌腈可湿性粉剂(WP)防治苗期和储藏期甘薯黑斑病的效果进行了评价。结果表明:随咯菌腈处理浓度增加,甘薯长喙壳菌分生孢子的萌发率逐渐下降,芽管扭曲程度逐渐加大。咯菌腈还可造成菌株芽管过早出现分支。咯菌腈对供试菌株分生孢子萌发的EC50值在0.20~0.99μg/mL,平均值为0.52μg/mL;对菌丝生长的EC50值在0.17~0.31μg/mL,平均值为0.24μg/mL,不同敏感性菌株所占频率呈正态分布,可作为甘薯长喙壳菌对咯菌腈的敏感基线。50%咯菌腈WP在有效成分500 mg/L下浸苗处理,对薯苗黑斑病的防治效果可达90.24%,显著高于500 g/L甲基硫菌灵悬浮剂(SC)同浓度下的处理。采用浸薯块法防治储藏...  相似文献

为筛选防治牡丹红斑病的杀菌剂,采用涂布平板法测定了多菌灵、戊唑醇、嘧菌酯对病菌分生孢子形成和萌发的影响。结果表明:3种杀菌剂对病菌孢子萌发均有很强的抑制作用,浓度越高,抑制作用越强,以嘧菌酯最强,1μg/mL浓度处理其孢子萌发率低于10%,5μg/mL处理孢子完全不能萌发;3种杀菌剂均可显著抑制病菌芽管的伸长生长,1μg/mL处理下孢子萌发24 h后芽管长度均不足10μm,5μg/mL处理对芽管伸长的抑制率达到90%;3种杀菌剂可使分生孢子及芽管发生畸形,多菌灵的致畸作用最强,0.1μg/mL处理即表现出明显的致畸作用,而戊唑醇致畸作用较弱;嘧菌酯和戊唑醇对病菌产孢结构形成和分生孢子产生数量有强烈的抑制作用,0.05μg/mL浓度下即完全不产孢,但多菌灵在0.05μg/mL浓度下则促进病菌产孢结构形成及产孢量增加。表明3种杀菌剂均可作为保护剂在病害发生前期喷施,但在流行期应尽量使用嘧菌酯和戊唑醇,以减少病菌再侵染数量。  相似文献

为明确河南省小麦赤霉病菌Fusarium graminearum对咯菌腈的敏感性,采用菌丝生长速率法测定了咯菌腈对从该省11个地市分离的95株菌株的毒力,通过方差分析法及聚类分析法对测定结果进行了分析,并研究了咯菌腈与多菌灵、戊唑醇对病菌毒力的相关性。结果显示:咯菌腈对小麦赤霉病菌菌丝生长的最低抑制浓度为0.1 μg/mL;咯菌腈对供试95株菌株的EC50范围在0.003~0.088 μg/mL之间,平均EC50为0.011 μg/mL;敏感性频率分布图显示,病菌群体中存在着对咯菌腈敏感性较低的亚群体,但67.4%供试菌株敏感性频率呈正态分布,将此部分菌株的EC50平均值0.007 μg/mL作为小麦赤霉病菌对咯菌腈的敏感性基线。方差分析及聚类结果均显示,同一县市内的菌株对咯菌腈EC50的最大值和最小值之比为1.1~8.3;除周口市沈丘县的菌株外,其余县市的菌株对咯菌腈敏感性差异不明显,咯菌腈EC50平均值变化范围在0.005~0.028 g/mL之间,最大值是最小值的5.6倍;小麦赤霉病菌对咯菌腈的敏感性与其对多菌灵、戊唑醇的敏感性之间无明显相关性。表明河南省小麦赤霉病菌群体中尽管存在着敏感性较低的亚群体,但可通过药剂复配进行防控。  相似文献

为筛选出有效防治番茄叶霉病的药剂,采用生长速率法及平板涂布法测定6种三唑类杀菌剂对番茄叶霉病菌菌丝生长及分生孢子萌发和芽管伸长的毒力,评价其对番茄植株的安全性和对叶霉病的田间防效。结果表明,己唑醇、苯醚甲环唑、戊唑醇和氟硅唑对番茄叶霉病菌菌丝生长的毒力均较高,EC_(50)分别为0.50、0.55、0.80、2.42 mg/L,其次为腈菌唑和四氟醚唑,EC50分别为6.92、15.08 mg/L。6种杀菌剂抑制病菌分生孢子萌发及芽管伸长的作用均较弱,对芽管伸长的抑制活性高于对孢子萌发的抑制活性;戊唑醇和四氟醚唑抑制孢子萌发的作用相对较强,100 mg/L处理的抑制率为60%~70%,戊唑醇、四氟醚唑和己唑醇抑制芽管伸长的作用相对较强,100 mg/L处理的抑制率均在70%以上。己唑醇和戊唑醇200 mg/L处理番茄植株,显著抑制其株高,苯醚甲环唑和腈菌唑对其影响相对较小,这4种杀菌剂对番茄植株的叶色及形态均无明显影响;且这4种杀菌剂对番茄叶霉病的田间预防效果均高于治疗效果,其中150 mg/L己唑醇的预防效果和治疗效果均最高,分别为90.67%和85.58%;苯醚甲环唑的最低,300 mg/L时预防效果为80.16%,治疗效果为71.68%。  相似文献

The epidemiology ofSitobion avenae and its natural enemies in winter wheat was studied in 1975, 1976 and 1977. Immigration was important until the end of flowering. The alate immigrants had apterous offspring. These became the driving force in population growth. Their offspring were mostly alatae which usually left the field. A model of the epidemic was developed. Quantitative relations between the aphids and their environment were obtained from literature or established in laboratory trials. The model simulated population development and population composition from the beginning of June till the population peak at the end of June or early in July. Because quantitative data on relations between aphids and their natural enemies and pathogens are scarce, and since the knowledge on wing formation is still limited, the population collapse could not be predicted. In the future, prognosis over a period of three weeks seems possible.Samenvatting De toenemende betekenis van graanbladluizen (vooralSitobion avenae) gepaard gaande met een sterke toename van het gebruik van insecticiden op granen maakte verbetering van de prognose over het schadelijk optreden wenselijk. Door gedetailleerde tellingen in het veld (Fig. 1–7) werden gegevens verkregen over het verloop van de epidemie en het optreden van natuurlijke vijanden in 1975, 1976 en 1977.Een immigratieperiode tot in de bloei kon worden vastgesteld. Daarna lijkt de aantrekkelijkheid van het gewas voor alate luizen te verminderen. De alate immigranten krijgen aptere nakomelingen. Deze vormen de stuwende kracht van de populatiegroei. De nakomelingen van apteren zijn merendeels alaat. Zij verlaten het gewas.Een model van de populatieontwikkeling gedurende de epidemie werd opgesteld. De relatiediagrammen Fig. 9 en 10 laten groei en ontwikkeling vanS. avenae en een predator (Syrphus corollae) zien. Kwantificering van de betrekkingen werd mogelijk door literatuurgegevens en laboratoriumexperimenten.Met het model kon de populatieontwikkeling vanS. avenae vanaf begin juni tot aan de populatiepiek in 1975, 1976 en 1977 vrij goed worden gesimuleerd (Fig. 12). Ook de populatieopbouw kon worden gesimuleerd (Fig. 14). De teruggang van de populatiedichtheid blijkt moeilijker te voorspellen door het ontbreken van gegevens over natuurlijke vijanden.Het lijkt waarschijnlijk dat in de toekomst met het model een prognose over de piek van de bladluispopulatie circa 3 weken tevoren mogelijk zal zijn.  相似文献

Potato seed tubers are imported to Israel from northern Europe and planted in spring; tubers harvested early from the spring crop are used as seed for the autumn crop. Although only seed lots registered as certified are imported, a previous survey (1984–1994) indicated that most imported lots were affected by latent or active infections caused byErwinia carotovora,Streptomyces scabies, Rhizoctonia solani, Fusarium spp. andSpongospora subterranae. The survey was extended until 1998, and included additional pathogens:Ralstonia solanacearum,Helminthosporium solani, Colletotrichum coccodes andVerticillium dahliae. Most of these pathogens were also monitored in domestic seed tubers, and are reported for the first time. Brown rot was not observed in any of the imported lots. Blackleg and soft rot caused byErwinia spp. were detected in most of the imported lots; however, less than 7% of the lots were contaminated at high levels, while approximately 65% were contaminated at moderate levels. Common scab was detected in most of the imported lots; 51% of the imported lots were contaminated at moderate or high levels, whereas only 6.5% of the domestic seed lots were contaminated at these levels. Black scurf was detected in most of the imported lots; on average, 47.3%, 44.2% and 1.4% of the lots were contaminated at low, moderate and high levels, respectively, and only 7.1% were disease-free. In contrast, most of the domestic lots were either disease-free (45.4%) or had a low disease incidence (37.3%). Only 16.7% of the lots were moderately infected and 0.2% were highly contaminated. Silver scurf was observed in most of the imported lots during all years of the survey, with no differences among the producing countries; on average, 22.7%, 66.1% and 7.5% of the lots were contaminated at low, moderate and high levels, respectively, and only 3.7% were disease-free. Most of the domestic lots (76%) were disease-free and only 6.6% were infected at moderate or high levels. Black dot was observed in a considerable portion of the shipments from Holland during all years of the survey, particularly in 1998, when 34% of the lots were infected. The shipments from France and Germany were infected at low levels, except in 1998, when 19% and 11% of the lots, respectively, arrived infected. In shipments from Scotland and Ireland low incidences of the disease were observed in 1994 and 1995. In the domestic lots, black dot incidence was low (<2.4%) except in 1996, when 11% of the lots were infected.V. dahliae was monitored only in domestic seed tubers. The incidence of disease-free lots was 56–64%, whereas in 20–30% of the lots the level of infection was <5%, and in 6–16% of the lots the level was >5%. Bean hypocotyls, pea pods and tomato fruits were tested for phaseollin, pisatin and rishitin production when challenged with the phytopathogenic bacteriaErwinia carotovora, Pseudomonas phaseolicola, P. pisi andP. solanacearum, and their isolated extracellular polysaccharides. All bacteria induced phytoalexin accumulation, whereas only phaseollin and pisatin, but not rishitin, were elicited by EPS. Molecular diagnostic techniques have been developed to differentiate the Ascochyta pathogens that infect cool season food and feed legumes, as well as to improve the sensitivity of detecting latent infection in plant tissues. A seed sampling technique was developed to detect a 1% level of infection by Ascochyta rabiei in commercial chickpea seed. The Ascochyta pathogens were shown to be genetically diverse in countries where the pathogen and host have coexisted for a long time. However, where the pathogen was recently introduced, such as A. rabiei to Australia, the level of diversity remained relatively low, even as the pathogen spread to all chickpea-growing areas. Pathogenic variability of A. rabiei and Ascochyta pinodes pathogens in chickpea and field pea respectively, appears to be quantitative, where measures of disease severity were based on aggressiveness (quantitative level of infection) rather than on true qualitative virulence. In contrast, qualitative differences in pathogenicity in lentil and faba bean genotypes indicated the existence of pathotypes of Ascochyta lentis and Ascochyta fabae. Therefore, reports of pathotype discrimination based on quantitative differences in pathogenicity in a set of specific genotypes is questionable for several of the ascochyta-legume pathosystems such as A. rabiei and A. pinodes. This is not surprising since host resistance to these pathogens has been reported to be mainly quantitative, making it difficult for the pathogen to overcome specific resistance genes and form pathotypes. For robust pathogenicity assessment, there needs to be consistency in selection of differential host genotypes, screening conditions and disease evaluation techniques for each of the Ascochyta sp. in legume-growing countries throughout the world. Nevertheless, knowledge of pathotype diversity and aggressiveness within populations is important in the selection of resistant genotypes.  相似文献

In 1975 many tumours were observed in plants ofBegonia Schwabenland grown in Aalsmeer. Submersion of the roots ofNicotiana megalosiphon seedlings in a homogenate of tumorous tissue, induced tumours after two weeks. Short periods of submergence yielded results similar to those obtained after longer periods. Tumour homogenates lost their infectivity after ten min at 50°C. Aphids transmitted the infectious agent.Treatment with propylene oxide did not inhibit infectivity completely. Filtration through a 450 nm filter removed the infectious agent.Tobacco tumor virus or a viroid could not be isolated. Cultures ofCorynebacterium fascians, isolated from tumours ofN. megalosiphon were highly infectious and induced tumours in healthyN. megalosiphon andBegonia. Tumorous tissue homogenates ofPelargonium zonale, Dahlia sp.,Gladiolus sp., andLilium sp. also caused tumours inN. megalosiphon, from whichC. fascians was isolated. Infectieus sap verloor zijn infectievermogen na 10 min verhitting bij 50°C. Bladluizen brachten de smetstof over. Propyleenoxide verminderde de infectiositeit wel, doch onderdrukte deze niet totaal. Bij filtratie door een 450 nm filter bleef het infectieuse agens op het filter achter. The survival of two species of plant parasitic nematodes: the root-lesion nematode Pratylenchus brachyurus, and the root-knot nematode Meloidogyne javanica, was evaluated in saturated atmospheres of 12 natural chemical compounds. The infectivity of two isolates of arbuscular mycorrhizal fungi: Glomus mosseae and Glomus intraradices, under identical experimental conditions, was also determined. All the compounds tested exerted a highly significant control against M. javanica and among them, benzaldehyde, salicilaldehyde, borneol, p-anisaldehyde and cinnamaldehyde caused a mortality rate above 50% over P. brachyurus. The infectivity of G. intraradices was inhibited by cinnamaldehyde, salicilaldehyde, thymol, carvacrol, p-anisaldehyde, and benzaldehyde, while only cinnamaldehyde and thymol significantly inhibited mycorrhizal colonization by G. mosseae.  相似文献

Five Pythium species (Pythium irregulare, P. mamillatum, P. myriotylum, P. spinosum and P. ultimum var. ultimum) were isolated from the hypocotyls and roots of kidney bean plants with damping-off from a commercial field and from experimental plots that have undergone either continuous cropping with kidney bean or rotational cropping with arable crops. In inoculation tests, all five Pythium species were pathogenic to kidney bean. This is the first report of damping-off of kidney bean caused by Pythium species; we named this disease damping-off of kidney bean. Leaves ofPelargonium domesticum andRuscus hypoglossum infected byBotrytis cinerea Pers., produced 12 and 1.5 nl ethylene/h/g, respectively, 34 days after inoculation; wounded or healthy leaves and phyloclades of them produced much lower amounts. When the fungus was grown on dead leaves it produced negligible amounts of the gas even when supplemented with methionine. Exogenous ethylene enhanced gray mold development in both hosts. Silver thiosulfate, aminooxyacetic acid (AOA) and aminoethoxyvinylglycine significantly inhibited disease development in pelargonium, and the latter two compounds inhibited ethylene production. AOA inhibited disease development and ethylene production in cut rose flowers; calcium ions inhibited disease development whereas the chelator EGTA [ethylene glycol bis-(β-aminoethyl ether)N,N,N′,N′-tetraacetic acid] enhanced it. Disease suppression by an excess of Ca2+ was correlated with repression of ethylene production by the flowers, whereas deficiency in Ca2+ increased disease severity. A survey was made to identify the most important soilborne fungal pathogens of asparagus crops in the Netherlands. Ten plants were selected from each of five fields with a young (1–4 y) first planting, five fields with an old (6–13 y) first planting and five fields with a young replanting. The analysis included fungi present in the stem base and the roots of plants with symptoms of foot and root rot or showing growth decline without specific disease symptoms. Isolates of each species were tested for pathogenicity to asparagus on aseptically grown plantlets on Knop's agar. Symptoms were caused byFusarium oxysporum, F. culmorum, Botrytis cinerea, Penicillium verrucosum var.cyclopium, Cylindrocarpon didymum, Phialophora malorum, Phoma terrestris andAcremonium strictum. F. oxysporum was by far the most common species and was isolated from 80% of the plants. Not all of its isolates were pathogenic to asparagus. Symptoms were caused by 67%, 78% and 93% of the isolates obtained from young first plantings, old first plantings and replantings, respectively.F. culmorum was isolated from 31% of the plants. Two other notorious pathogens of asparagus,F. moniliforme andF. proliferatum, did not occur in our samples.Species causing symptoms in the vitro test that were found on more than 5% of the plants were additionally tested for their pathogenicity in pot experiments.F. oxysporum f.sp.asparagi caused severe foot and root rot, significantly reduced root weights and killed most of the plants.F. culmorum caused lesions on the stem base often resulting in death of the plant.P. terrestris, a fungus only once reported as a pathogen of asparagus, caused an extensive root rot, mainly of secondary roots that became reddish. The fungus was isolated in only a few samples and is not to be regarded as an important pathogen in Dutch asparagus crops.P. malorum caused many small brown lesions on the stem base and incidentally also on the upper part of small main roots. This is the first report of its pathogenicity to asparagus. The fungus is one of the organisms inciting spear rust and it reduced crop quality rather than crop yield.P. verrucosum var.cyclopium andC. didymum did not cause symptoms in pot experiments.Because of its predominance on plants with foot and root rot and its high virulence,F. oxysporum f.sp.asparagi was considered to be the main soilborne pathogen of asparagus in the Netherlands.  相似文献

Agave tequilana is the raw material for the production of the alcoholic beverage tequila. A bacterial disease has affected the A. tequilana crop in recent years. Previous reports based on colony and cell morphology, Gram stain and potato rot indicated that Erwinia sp. is the main pathogen. We isolated a several bacterial isolates capable of producing soft-rot symptoms in greenhouse pathogenicity assays. An extensive characterisation involving pathogenicity tests, fatty acid profile, metabolic and physiological properties, ribosomal DNA sequence and intergenic transcribed spacer amplification (ITS-PCR) and restriction banding pattern (ITS-RFLP) was made of each isolate. Three different species: Erwinia cacticida, Pantoea agglomerans and Pseudomonas sp. were identified. Fatty acid and metabolic profiles gave low similarity values of identification but 16S rDNA sequence, ITS-PCR and ITS-RFLP confirmed the identification of E. cacticida. In the phylogenetic tree, E. cacticida from blue agave was grouped neither with E. cacticida type strains nor with Erwinia carotovora. This is the first report that associates E. cacticida with A. tequilana soft-rot symptoms.  相似文献
