Maximum limits of 0.02% are recommended for precision of IR analysis for fat, protein, and lactose, and 0.04% for total solids. Recommended maximum systematic errors are 0.06% for fat, protein, and lactose, and 0.12% for total solids, when instrument results are compared with results of specified AOAC methods. Recommended maximum mean differences between instrument and standard results are based on 95% confidence limits for the mean of 8 samples; precision and systematic errors are calculated as the standard deviation of difference between duplicates for 8 samples, and systematic errors are calculated as the standard deviation of difference between instrument and reference results for 8 samples. Automated sections for milk of Official Methods of Analysis were rewritten to eliminate references to individual instruments, to eliminate possible differences between calibration procedures for different instruments, and to stress important instrument operational characteristics which can have significant effects on instrument performance.

Near-infrared (NIR) spectroscopy calibrations that will allow prediction of the solid fat content (SFC) of milk fat extracted from butter by one measurement during manufacture were developed. SFC is a measure of the amount of the solid fraction of fat crystallized at a temperature expressed as a percentage (w/w). At-line SFC determinations are currently performed by nuclear magnetic resonance (NMR) spectroscopy, which involves a 16 h delay period for tempering of the milk fat at 0 degrees C prior to the SFC measurements, from 0 to 35 degrees C in a series of 5 degrees C increments. The NIR spectra (400-2500 nm) were obtained using a sample holder maintained at 60 degrees C. Accurate predictions for the SFC (%) were developed by principal component analysis (PCA) and partial least-squares (PLS) regression models to relate the NIR spectra to the corresponding NMR values. The independent validation samples (N = 22) had a standard error of prediction (SEP) of 0.385-0.762% for SFC between 0 and 25 degrees C, with SFC reference values ranging between 70.42 and 8.96% with a standard deviation range of 3.36-1.47. The low bias (from -0.351 to -0.025), the slopes (0.935-1.077), and the excellent predictive ability (R2; 0.923-0.978) supported the validity of these calibrations.

A collaborative study was conducted to determine the standard error of difference among laboratories for near-infrared reflectance spectroscopic (NIRS) determination of acid-detergent fiber (ADF) and crude protein in forages. The 6 participating laboratories were members of the USDA/ARS National Near-Infrared Reflectance Spectroscopy Forage Research Project. The NIRS calibration equations were developed in the Associate Referee's laboratory for crude protein and ADF and were transferred to the instrument in each of the other collaborating laboratories. The calibration set included over 650 diverse forage samples with crude protein and ADF calibration data; the validation set included 94 samples of bermudagrass. Among-laboratory reproducibility for the NIRS method, calculated as the relative standard deviation for reproducibility (RSDR), was 1.14% for ADF and 0.42% for crude protein. The variance component for among-laboratory variation (coefficient of variation) was 2.54% for ADF and 2.89% for crude protein. These results confirm that it is possible to calibrate, validate, and transfer (NIRS) equations and data among laboratories for the accurate determination of ADF and crude protein, and thereby demonstrate that NIRS can be used as a standard method for the analysis of forages. The method has been adopted official first action.

This study evaluates nutritive, morphological and agronomic characteristics of forage maize predicted by using a high-quality near-infrared (NIR) spectrometer and an NIR hyperspectral-imaging technique using partial least squares (PLS) regression models. The study includes 132 samples of dried milled whole-plant homogenates of forage maize with variation in maturity, representing two growing seasons, three locations in Sweden and three commercial maize hybrids. The samples were measured by a classical sample cup NIR spectrometer and by a pushbroom hyperspectral-imaging instrument. The spectra and a number of variables (crude protein, CP, neutral detergent fibre, starch, water soluble carbohydrates (WSC) and organic matter digestibility), morphological variables (leaves, stems & ears) and crop yield were used to make PLS calibration models. Using PLS modelling allowed the determination of how well maize variables can be predicted from NIR spectra and a comparison of the two types of instruments. Most examined variables could be determined equally well, by both instruments, but the pushbroom technique gave slightly better predictions and had higher analytical capacity. Predictions of CP, starch, WSC and the proportions of ears in the maize gave robust. The findings open new possibilities to further utilise the technology in plant breeding, crop management, modelling and forage evaluation.

The increasing demand for triticale as food, feed, and fuel has resulted in the availability of cultivars with different grain quality characteristics. Analyses of triticale composition can ensure that the most appropriate cultivars are obtained and used for the most suitable applications. Near‐infrared (NIR) spectroscopy is often used for rapid measurements during quality control and has consequently been investigated as a method for the measurement of protein, moisture, and ash contents, as well as kernel hardness (particle size index [PSI]) and sodium dodecyl sulfate (SDS) sedimentation from both whole grain and ground triticale samples. NIR spectroscopy prediction models calculated using ground samples were generally superior to whole grain models. Protein content was the most effectively modeled quality property; the best ground grain calibration had a ratio of the standard error of test set validation to the standard deviation of the reference data of the test set (RPDtest) of 4.81, standard error of prediction (SEP) of 0.52% (w/w), and r2 of 0.95. Whole grain protein calibrations were less accurate, with optimum RPDtest of 3.54, SEP of 0.67% (w/w), and r2 of 0.92. NIR spectroscopy calibrations based on direct chemical reference measurements (protein and moisture contents) were better than those based on indirect measurements (PSI, ash content, and SDS sedimentation). Calibrations based on indirect measurements would, however, still be useful to identify extreme samples.

Visible and near infrared (VIS/NIR) transmission spectroscopy and chemometric methods were utilized for the fast determination of soluble solids content (SSC) and pH of cola beverage. A total of 180 samples were used for the calibration set, whereas 60 samples were used for the validation set. Some preprocessing methods were applied before developing the calibration models. Several PLS factors, extracted by partial least squares (PLS) analysis, were used as the inputs of least squares-support vector machine (LS-SVM) model according to their accumulative reliabilities. The correlation coefficient (r), root mean square error of prediction (rmsEP), bias, and RPD were 0.959, 1.136, -0.185, and 3.5 for SSC, whereas 0.973, 0.053, 0.017, and 4.1 for pH, respectively. An excellent prediction precision was achieved by LS-SVM compared with PLS. The results indicated that VIS/NIR spectroscopy combined with LS-SVM could be applied as a rapid and alternative way for the fast determination of SSC and pH of cola beverage.

Samples (n = 620) of homogenized red grape berries were analyzed using a visible and near-infrared (NIR) spectrophotometer (400-2500 nm) in reflectance. The spectra and the analytical data were used to develop partial least-squares calibrations to predict dry matter (DM) content and condensed tannins (CT) concentrations. The coefficient of determination in cross-validation and the standard error of cross-validation were 0.92 and 0.83% w/w for DM and 0.86 and 0.46 mg/g epicatechin equivalents for CT, respectively. The standard error in prediction was 1.34% w/w for DM and 0.89 mg/g epicatechin equivalents for CT, respectively. By implementing a NIR spectroscopy method to measure DM and CT in red grape homogenates, we have developed an approach that is suited to large-scale compositional analysis in commercial wine production facilities, as it enables the analysis of large numbers of samples needed to stream batches of fruit. From an economical point of view, the calibration models could be achieved with relatively small data sets. Thus, NIR offers a suitable and efficient tool for the simultaneous measurement of DM and CT in addition to other important parameters in red grape homogenates such as total anthocyanins, total soluble solids, and pH, with minimal sample preparation and low cost.

Near-infrared reflectance spectroscopy (NIRS) calibrations were developed to enable the accurate and fast prediction of the total contents of methionine, cystine, lysine, threonine, tryptophan, and other essential amino acids, protein, and moisture in the most important protein-rich feed ingredients. More than 1000 samples of global origin collected over four years were analyzed on amino acids following the official methods of the United States and the European Union. Detailed data and graphics are given to characterize the obtained calibration equations. NIRS was validated with independent samples for soy and meat meal products and compared to the amino acid predictions using linear crude protein regressions. With a few exceptions, validation showed that 85-98% of the amino acid variance in the samples could be explained using NIRS. NIRS predictions compared to reference results agree excellently, with relative mean deviations below 5%. Especially for meat and poultry meals, NIRS can predict amino acids much better than crude protein regressions. By enabling the amino acid analysis of many samples to be completed in a short time, NIRS can improve the accuracy of feed formulation and thus the quality and production costs of mixed feeds.

Further NIRS calibrations were developed for the accurate and fast prediction of the total contents of methionine, cystine, lysine, threonine, tryptophan, and other essential amino acids, protein, and moisture in the most important cereals and brans or middlings for animal feed production. More than 1100 samples of global origin collected over five years were analyzed for amino acids following the Official Methods of the United States and European Union. Detailed data and graphics are given to characterize the obtained calibration equations. NIRS was validated with 98 independent samples for wheat and 78 samples for corn and compared to amino acid predictions using linear crude protein regression equations. With a few exceptions, validation showed that 70-98% of the amino acid variance in the samples could be explained using NIRS. Especially for lysine and methionine, the most limiting amino acids for farm animals, NIRS can predict contents in cereals much better than crude protein regressions. Through low cost and high speed of analysis NIRS enables the amino acid analysis of many samples in order to improve the accuracy of feed formulation and obtain better quality and lower production costs.

The development of new wheat cultivars that target specific end‐uses, such as low or zero amylose contents of partially waxy and waxy wheats, has become a modern focus of wheat breeding. But for efficient and cost‐effective breeding, inexpensive and high‐throughput quality testing procedures, such as near infrared (NIR) spectroscopy, are required. The genetic nature of a set of wheat lines, which included waxy to nonwaxy cultivars, results in a bimodal distribution of amylose contents that presents some special challenges for the formulation of stable NIR calibrations for this property. The obvious and intuitive solution lies in the use of some form of localization procedure and we explored three localization algorithms in comparison with the default partial least squares. Localization with respect to the waxy (zero amylose) cultivars resulted in a modified partial least squares calibration with a standard error of prediction of 0.16%. The results establish unambiguously that there are advantages in performing a suitable localization to achieve a reliable NIR calibration and prediction. The accuracy of the method can also be enhanced by application of an appropriate resampling strategy. In addition, there are advantages in performing a suitable localization to achieve a reliable NIR calibration‐prediction. It resolves the issue of how to utilize the bimodal distribution of apparent amylose values. The best results are obtained when the localization is performed simultaneously with respect to the sample property under investigation and the NIR spectra. The key problem with the measurement of amylose is the laboratory reference method which, in reality, only measures the apparent amylose content of the wheat. As a direct consequence, the measurements of amylose have such a large error that traditional calibration‐prediction procedures generate unacceptable results. To resolve this difficulty, a statistically based resampling strategy is proposed as a method of identifying samples where there is a large error in the reference measurement.

The identification of Melaleuca cajuputi leaf samples (trees) that demonstrate enhanced oil characteristics using near infrared (NIR) spectroscopy is described. Leaf samples from an unthinned M. cajuputi seedling seed orchard in Indonesia were collected and air-dried, and their 1,8-cineole content and oil concentrations were determined. NIR spectra of the leaves were obtained, and calibrations for 1,8-cineole content and oil concentration were developed using spectra that had been selected using spectral features; that is, no knowledge of 1,8-cineole content or oil concentration was used to select the calibration samples. The calibrations were used to predict the 1,8-cineole content and oil concentration of the remaining samples. It was demonstrated that NIR spectroscopy could be used to identify leaf samples that had high 1,8-cineole contents and oil concentrations. The technique has the potential to greatly reduce the time involved in ranking large numbers of samples for these attributes, as is a requirement in tree breeding programs to enhance oil production.

Deoxynivalenol (DON) levels in harvested grain samples are used to evaluate the Fusarium head blight (FHB) resistance of wheat cultivars and breeding lines. Fourier transform near‐infrared (FT‐NIR) calibrations were developed to estimate the DON level and moisture content (MC) of bulk wheat grain samples harvested from FHB screening trials. Grains in a rotating glass petri dish were scanned in the 10,000–4,000 cm−1 (1,000–2,500 nm) spectral range using a Perkin Elmer Spectrum 400 FT‐IR/FT‐NIR spectrometer. The DON calibration predicted the DON levels in test samples with R 2 = 0.62 and root mean square error of prediction (RMSEP) = 8.01 ppm. When 5–25 ppm of DON was used as the cut‐off to classify samples into low‐ and high‐DON groups, 60.8–82.3% of the low‐DON samples were correctly classified, whereas the classification accuracy of the high‐DON group was 82.3–94.0%. The MC calibration predicted the MC in grain samples with R 2 = 0.98 and RMSEP = 0.19%. Therefore, these FT‐NIR calibrations can be used to rapidly prescreen wheat lines to identify low‐DON lines for further evaluation using standard laboratory methods, thereby reducing the time and costs of analyzing samples from FHB screening trials.

The feasibility of rapid analysis for oligosaccharides, including isomaltose, isomaltotriose, maltose, and panose, in Chinese rice wine by Fourier transform near-infrared (FT-NIR) spectroscopy together with partial least-squares regression (PLSR) was studied in this work. Forty samples of five brewing years (1996, 1998, 2001, 2003, and 2005) were analyzed by NIR transmission spectroscopy with seven optical path lengths (0.5, 1, 1.5, 2, 2.5, 3, and 5 mm) between 800 and 2500 nm. Calibration models were established by PLSR with full cross-validation and using high-performance anion-exchange chromatography coupled with pulsed amperometric detection as a reference method. The optimal models were obtained through wavelength selection, in which the correlation coefficients of calibration (r(cal)) for the four sugars were 0.911, 0.938, 0.925, and 0.966, and the root-mean-square errors of calibrations were 0.157, 0.147, 0.358, and 0.355 g/L, respectively. The validation accuracy of the four models, with correlation coefficients of cross-validation (r(cv)) being 0.718, 0.793, 0.681, and 0.873, were not very satisfactory. This might be due to the low concentrations of the four sugars in Chinese rice wine and the influence of some components having structures similar to those of the four sugars. The results obtained in this study indicated that the NIR spectroscopy technique offers screening capability for isomaltose, isomaltotriose, maltose, and panose in Chinese rice wine. Further studies with a larger Chinese rice wine sample should be done to improve the specificity, prediction accuracy, and robustness of the models.

The development of accurate calibration models for selected soil properties is a crucial prerequisite for successful implementation of visible and near infrared (Vis‐NIR) spectroscopy for soil analysis. This paper compares the performance of calibration models developed for individual farms with that of general models valid for three farms in three European countries. Fresh soil samples collected from farms in the Czech Republic, Germany and Denmark were scanned with a fibre‐type Vis‐NIR spectrophotometer. After dividing spectra into calibration (70%) and validation (30%) sets, spectra in the calibration set were subjected to partial least squares regression (PLSR) with leave‐one‐out cross‐validation to establish calibration models of soil properties. Except for the Czech Republic farm, individual farm models provided successful calibration for total carbon (TC), total nitrogen (TN) and organic carbon (OC), with coefficients of determination (R2) of 0.85–0.93 and 0.74–0.96 and residual prediction deviations (RPD) of 2.61–3.96 and 2.00–4.95 for the cross‐validation and independent validation respectively. General calibration models gave improved prediction accuracies compared with models of farms in the Czech Republic and Germany, which was attributed to larger ranges in the variation of soil properties in general models compared with those in individual farm models. The results revealed that larger standard deviations (SDs) and wider variation ranges have resulted in larger R2 and RPD, but also larger root mean square errors of prediction (RMSEP). Therefore, a compromise solution, which also results in small RMSEP values, should be found when selecting soil samples for Vis‐NIR calibration to cover a wide variation range.

Hydrogen cyanide (HCN) is a toxic chemical that can potentially cause mild to severe reactions in animals when grazing forage sorghum. Developing technologies to monitor the level of HCN in the growing crop would benefit graziers, so that they can move cattle into paddocks with acceptable levels of HCN. In this study, we developed near-infrared spectroscopy (NIRS) calibrations to estimate HCN in forage sorghum and hay. The full spectral NIRS range (400-2498 nm) was used as well as specific spectral ranges within the full spectral range, i.e., visible (400-750 nm), shortwave (800-1100 nm) and near-infrared (NIR) (1100-2498 nm). Using the full spectrum approach and partial least-squares (PLS), the calibration produced a coefficient of determination (R(2)) = 0.838 and standard error of cross-validation (SECV) = 0.040%, while the validation set had a R(2) = 0.824 with a low standard error of prediction (SEP = 0.047%). When using a multiple linear regression (MLR) approach, the best model (NIR spectra) produced a R(2) = 0.847 and standard error of calibration (SEC) = 0.050% and a R(2) = 0.829 and SEP = 0.057% for the validation set. The MLR models built from these spectral regions all used nine wavelengths. Two specific wavelengths 2034 and 2458 nm were of interest, with the former associated with C═O carbonyl stretch and the latter associated with C-N-C stretching. The most accurate PLS and MLR models produced a ratio of standard error of prediction to standard deviation of 3.4 and 3.0, respectively, suggesting that the calibrations could be used for screening breeding material. The results indicated that it should be feasible to develop calibrations using PLS or MLR models for a number of users, including breeding programs to screen for genotypes with low HCN, as well as graziers to monitor crop status to help with grazing efficiency.

AOAC method 996.01, used in cereal foods to determine total fat as defined by the U.S. Nutrition Labeling and Education Act (NLEA), is laborious and time-consuming and utilizes hazardous chemicals. Near-infrared (NIR) reflectance spectroscopy, a rapid and environmentally benign technique, was investigated as a potential method for the prediction of total fat using AOAC method 996.01 as the reference method. Near-infrared reflectance spectra (1104-2494 nm) of ground cereal products (n = 72) were obtained using a dispersive spectrometer, and total fat was determined according to AOAC method 996.01. Using multivariate analysis, a modified partial least-squares model was developed for total fat prediction. The model had a SECV of 1.12% (range = 0.5-43.2%) and a multiple coefficient of determination of 0.99. The model was tested with independent validation samples (n = 36); all samples were predicted within NLEA accuracy guidelines. The results indicate that NIR reflectance spectroscopy is an accurate means of determining the total fat content of diverse cereal products for nutrition labeling.

Abstract

The use of ultraviolet (UV), visible (VIS), near infrared reflectance (NIR), and midinfrared (MIR) spectroscopy techniques have been found to be successful in determining the concentration of several chemical properties in soils. The aim of this study was to evaluate the effect of two reference methods, namely Bray and Resins, on the VIS and NIR calibrations to predict phosphorus in soil samples. Two hundred (n=200) soil samples were taken in different years from different locations across Uruguay with different physical and chemical characteristics due to different soil types and management. Soil samples were analyzed by two reference methods (Bray and Resins) and scanned using an NIR spectrophotometer (NIRSystems 6500). Partial least square (PLS) calibration models between reference data and NIR data were developed using cross‐validation. The coefficient of determination in calibration (R2) and the root mean square of the cross validation (RMSECV) were 0.58 (RMSECV: 3.78 mg kg?1) and 0.61 (RMSECV: 2.01 mg kg?1) for phosphorus (P) analyzed by Bray and Resins methods, respectively, using the VIS and NIR regions. The R2 and RMSECV for P using the NIR region were 0.50 (RMSECV: 3.78 mg kg?1) and 0.58 (RMSECV: 2.01 mg kg?1). This study suggested that differences in accuracy and prediction depend on the method of reference used to develop an NIR calibration for the measurement of P in soil.
