In recent years several interspecific hybrids have been reported in the plant pathogenic oomycete genus Phytophthora. Due to the large genotypic and phenotypic changes, these hybrids might have broader or more limited host ranges compared with their parental species. It is crucial to understand the host range of Phytophthora hybrids to minimize the economic losses caused by their infection. The potential host range of four hybrids belonging to Clade 8a of the Phytophthora phylogenetic tree was investigated in this study. Thirty species of herbaceous plants as well as eight species of woody plants were inoculated and monitored for any symptom of infection. In addition, the detached twigs of 32 tree species, fruits of six plant species, tubers of potato, and roots of carrot and sugar beet were investigated for susceptibility to these hybrids. Almost all hybrids caused severe rot on all tested fruits, tubers, and roots, although different isolates showed different pathogenicity on detached tree twigs. All hybrids tested had a different host range compared with their parental species: they were able to infect plants outside the host range of their parents, infect hosts of both parental species, although these parents did not have overlapping hosts, or, in some cases, they were not able to infect hosts infected by the parents.  相似文献

The host ranges of 33 isolates of the downy mildew fungus Peronospora parasitica from different Brassica hosts and diverse geographic origins were assessed on a standard set of Brassica accessions Isolates from each host species had a distinct host range, and in some cases isolates from the same host species but different geographic origins differed in host range. Most isolates were capable of infecting hosts other than their species of origin. These results are discussed in relation to species specificity and the conservation of certain virulence gene combinations. The epidemiological implications for cross-infection of Brassica hosts by P. parasitica isolates in the field are also considered.  相似文献

The host range of Polymyxa betae on common arable weed species in Britain was determined by growing plants in naturally infested soil and examining their root systems for the presence of resting spores (cystosori). Of the 24 species tested, only Atriplex patula and Chenopodium album of the Chenopodiaceae, and Silene alba of the Caryophyllaceae, were found to be heavily infected. S. alba is a newly recorded host species for Polymyxa. The host specificity of isolates of P. betae from Beta vulgaris, C. album and A. patula was investigated by observing which of 11 test plants could be infected by the isolates obtained from this soil. Three main biotypes of P. betae appeared to be distinguishable: one which was able to infect all chenopodiaceous species; one which had a narrower host range; and one which was able to infect S. alba. The role of weed species in the epidemiology of rhizomania is discussed.  相似文献

The abilities of different isolates of tomato spotted wilt tospovirus (TSWV) collected from northeastern and eastern Spain to infect 10 host species, and to be acquired and transmitted by the western flower thrips (WFT), Frankliniella occidentalis were compared. Two isolates of TSWV from a single source plant could be separated according to the different type of local lesions they induced in Nicotiana glutinosa. Host ranges of the studied TSWV isolates were very similar, but differences were found in the symptoms induced and in their capacity to infect specific hosts systemically. Lycopersicon esculentumDatura stramonium were evaluated for their potential as virus acquisition host species. The proportion of transmitter adult thrips obtained from WFT larvae fed on L. esculentum was greater than from D. stramonium. No differences were detected between TSWV isolates in their ability to be acquired and transmitted by WFT. No evidence was obtained of alterations in TSWV particles which could affect WFT transmissibility due to the repeated mechanical transfers used to clone the isolates. Our findings do not support the existence of pathological effects of TSWV on WFT.  相似文献

Plant–fungal specificity between cucurbitaceous crops and Diaporthe sclerotioides, the causal agent of black root rot, was studied using cucumbers (Cucumis sativa), melons (Cucumis melo), pumpkins (Cucurbita maxima), watermelons (Citrullus lanatus) and bottlegourd (Lagenaria siceraria var. gourda). Twelve D. sclerotioides isolates from these cucurbit species were cross‐inoculated. The virulence of the isolates was evaluated as the area under the disease progress curve (AUDPC). All cucurbit species were susceptible to each isolate, but AUDPCs were significantly different among the hosts, with the order of greatest to least being melon, cucumber, watermelon, bottlegourd and finally, pumpkin. The infectiveness of isolates was assessed as the quantity of D. sclerotioides DNA detected in the hypocotyls of seedlings 2 weeks after inoculation using a real‐time PCR protocol. The fungal DNA quantities varied among the species in the same order as the AUDPCs. Whilst there were statistically significant correlations between the virulence and infectiveness of D. sclerotioides isolates in cucumbers, melons and bottlegourds, their coefficients of determination were not high (r2 < 0·6). Orthogonal contrasts indicated no specificity in either the fungal virulence or infectiveness between D. sclerotioides isolates and the cucurbit hosts from which these isolates originated. Thus, although the degree of host susceptibility to D. sclerotioides varies among cucurbit species, the absence of specificity to the host species in either virulence or infectiveness suggests the pathogen may spread via various cucurbit crops, irrespective of their original host species.  相似文献

Isolates of the take-all fungus, Gaeumannomyces graminis var. avenae , which affects oats, wheat and other grasses, and of G.g. var. tritici , which preferentially affects wheat, rye and barley, contain a high proportion of repeated sequences. Total DNA from 57 fungal isolates collected from many locations and different cereal hosts, and scored for virulence on wheat, rye and oats, revealed many restriction fragment length polymorphisms. These RFLP s were observed either by staining the DNA directly, by hybridization to radioactively labelled total fungal DNA , or by hybridization with labelled wheat ribosomal DNA . With only a few exceptions, the isolates with the same preferred cereal hosts showed more similar patterns of restriction fragments than isolates that had different pathogenicity properties on cereal hosts, irrespective of the geographical origins of the isolates. This was even the case for R isolates of G.g. var. tritici that were virulent on wheat and rye compared with N isolates that were virulent only on wheat. Isolates were identified by hybridizing DNA from infected root samples with 32P-labelled total fungal DNA . The restriction fragment polymorphisms involving families of repeated sequence can therefore be used as a predictive assay for host preference of an isolate, and have probably arisen by host selection of fungal lineages. The variation between isolates in different pathogenicity groups suggests that there is little gene flow between isolates that can infect different hosts, even though they can coexist in the same field.  相似文献

ABSTRACT To investigate host specialization in Macrophomina phaseolina, the fungus was isolated from soybean, corn, sorghum, and cotton root tissue and soil from fields cropped continuously to these species for 15 years in St. Joseph, LA. Chlorate phenotype of each isolate was determined after growing on a minimal medium containing 120 mM potassium chlorate. Consistent differences in chlorate sensitivity were detected among isolates from different hosts and from soil versus root. To further explore genetic differentiation among fungal isolates from each host, these isolates were examined by restriction fragment length polymorphism and random amplified polymorphic DNA (RAPD) analysis. No variations were observed among isolates in restriction patterns of DNA fragments amplified by polymerase chain reaction covering the internal transcribed spacer region, 5.8S rRNA and part of 25S rRNA, suggesting that M. phaseolina constitutes a single species. Ten random primers were used to amplify the total DNA of 45 isolates, and banding patterns resulting from RAPD analysis were compared with the neighbor-joining method. Isolates from a given host were genetically similar to each other but distinctly different from those from other hosts. Chlorate-sensitive isolates were distinct from chlorate-resistant isolates within a given host. In greenhouse tests, soybean, sorghum, corn, and cotton were grown separately in soil infested with individual isolates of M. phaseolina that were chosen based on their host of origin and chlorate phenotype. Root colonization and plant weight were measured after harvesting. More colonization of corn roots occurred when corn was grown in soil containing corn isolates compared with isolates from other hosts. However, there was no host specialization in isolates from soybean, sorghum, or cotton. More root colonization in soybean occurred with chlorate-sensitive than with chlorate-resistant isolates.  相似文献

Larvae (metacestodes) of tapeworms of the cyclophyllidean family Gryporhynchidae (previously included in the Dilepididae) occur in different internal organs of fresh- and brackish water fish (110 fish species of 27 families in 12 orders reported), which serve as the second intermediate hosts. The species composition, spectrum of fish hosts, sites of infection, and geographical distribution of gryporhynchids recorded from fish are reviewed here on the basis of literary data and examination of extensive material from helminthological collections. Metacestodes of the following genera have been found in fish: Amirthalingamia Bray, 1974 (1 species), Ascodilepis Guildal, 1960 (1), Cyclustera Fuhrmann, 1901 (4), Dendrouterina Fuhrmann, 1912 (1), Glossocercus Chandler, 1935 (3), Neogryporhynchus Baer et Bona, 1960 (1), Paradilepis Hsü, 1935 (5), Parvitaenia Burt, 1940 (2), and Valipora Linton, 1927 (3). However, most published records concern only three species, namely Neogryporhynchus cheilancristrotus (Wedl, 1855) from the intestinal lumen, Paradilepis scolecina (Rudolphi, 1819) from the liver and mesenteries, and Valipora campylancristrota (Wedl, 1855) from the gall bladder of cyprinids and other fish in the Palaearctic Region. Data on other species as well as reports from other regions are very scarce and almost no information is available from Australia, tropical Asia and South America. A recent study of gryporhynchid metacestodes from Mexico (Scholz and Salgado-Maldonado 2001), which reported 13 species, suggested that they may be more common than indicated by records in the literature. Although only a few cases of pathogenic influence of larvae on fish hosts have been reported, the veterinary importance of gryporhynchids remains to be assessed on the basis of more detailed studies. The data available indicate a strict host and site specificity of some species whereas others occur in a wide spectrum of fish hosts and are not strictly site-specific. Evaluation of Paradilepis larvae from the liver of salmonid fish from British Columbia, Canada, identified as P. simoni Rausch, 1949 by Ching (1982), has shown that they probably belong to two species, P. simoni and P. rugovaginosus Freeman, 1954. Metacestodes of the latter species and those of Cyclustera magna (Baer, 1959) from the intestinal wall of Tilapia zillii (Gervais) from Kenya are reported from fish for the first time.  相似文献

Colletotrichum truncatum (syn. C. capsici) has been identified as the causal agent of anthracnose on various hosts, predominantly pepper (Capsicum spp.) plants. The aim of this study was to determine whether C. truncatum isolates infecting papaya, pepper and physic nut in southeastern Mexico are morphologically, genetically and pathogenically different, in order to improve disease management strategies. A total of 113 C. truncatum isolates collected from five producer states were subjected to phenotypic characterization and divided into six different morphological groups. These morphological traits and the location of the isolates were used to select a subset of 20 isolates for further studies. Differences in the pathogenicity of the isolates were tested with a cross‐inoculation assay using pepper, papaya and physic nut. The pathogenicity tests revealed that all isolates could infect the three hosts and produce typical anthracnose symptoms, indicating a lack of host specificity for this species and therefore its pathogenic potential on other plants. Phylogenetic analysis using internal transcribed spacer (ITS) and glyceraldehyde 3‐phosphate dehydrogenase (GAPDH) sequences of the C.   truncatum isolates from this study and reference strains was performed, grouping the isolates into a monophyletic clade. This study reports for the first time the characterization of C. truncatum causing anthracnose disease on three different hosts in Mexico.  相似文献

ABSTRACT Genetic diversity was investigated among California populations of Botryosphaeria dothidea, causal agent of Botryosphaeria panicle and shoot blight of pistachio, with random amplified polymorphic DNA (RAPD) and microsatellite-primed polymerase chain reaction (MP-PCR). We surveyed 120 isolates, 112 of which originated from the California Central Valley and included pistachio isolates (n = 52) and isolates from other plant species (n = 60). Out-group isolates (n = 8) were obtained from pistachio in Greece. There was a strong correlation (r = 0.99; P < 0.0001) between the RAPD- and MP-PCR dissimilarity data sets. Little genetic variation (haplotypic diversity [Hs] < 0.002) was detected among B. dothidea isolates collected from central and southern California pistachio plantings. We observed relatively high diversity for isolates from a northern California pistachio orchard (Hs = 0.0146), where the disease was first diagnosed, and from the Chico U.S. Department of Agriculture Germ Plasm Repository (Hs = 0.0726), where the first pistachio trees were planted in California in 1929. Isolates obtained from other hosts, especially those associated with the rare occurrence of the sexual stage of this fungus, showed the highest levels of diversity (Hs = 0.1689). Thirty-eight pistachio isolates (73.1%) had DNA fingerprints identical to 28 pycnidiospore-derived isolates (56.0%) obtained from other host species. Greenhouse inoculations demonstrated that all isolates obtained from other hosts were capable of infecting pistachio and produced characteristic disease symptomology. Thus, California populations of B. dothidea from pistachio are, for the most part, genetically uniform, with the sexual stage rare to absent. However, the rare occurrence of the sexual stage of B. dothidea on other hosts, and more importantly, the capacity of these isolates to infect pistachio, indicate that other host species may serve as sources of inoculum and genetic variation.  相似文献

Upgma analysis, principal component analysis, genetic diversity analysis and genetic distance analysis of RAPD data were used to assess the extent of host specialization in 50 isolates of S. homoeocarpa from five turfgrass hosts. In upgma analysis and principal component analysis, the occurrence of host specialization was not readily apparent based on visual inspection. Genetic diversity analysis showed significant differentiation among isolates from different host species ( G ST = 0.34, P  < 0.001). The strongest evidence for some degree of host specialization came from the statistical analysis of genetic distances among isolates. By grouping pairwise genetic distances between isolates based on their host species, and analysing for average distance within the same host species and among different host species, it was found that the average distance within species was less than among species ( P  < 0.0001). An analysis of molecular variance of the genetic distances among isolates found that 32.3% of the total variation was attributable to host species. It is concluded that these isolates of S. homoeocarpa showed a weak level of host specialization, which was not readily apparent by upgma or principal component analyses, but was revealed by genetic diversity analysis and statistical analysis of genetic distances among isolates. Inoculation tests on different host species and tests using a greater number of isolates are required to confirm the extent of specialization.  相似文献

Thirty-nine isolates of Phytophthora infestans were collected from the wild host Solanum ochranthum in the highland tropics of Ecuador and characterized with a set of phenotypic and molecular markers (mating type, metalaxyl sensitivity, the allozyme loci Gpi, and Pep, mitochondrial DNA haplotype, RFLP, and SSR), as well as for pathogenicity on various hosts. Three groups of isolates (A, B, and C) were identified based on their multilocus genotypes and variable abilities to cause disease on different hosts. Group A had a combination of alleles for the Gpi (86/100), Pep (96/100) and mtDNA (Ia) loci, as well as an RFLP fingerprint, that have not been reported for P. infestans in Ecuador, or elsewhere. Group B shares many marker characteristics with the US-1 lineage described in Ecuador on tomato, pear melon (S. muricatum), and S. caripense, but has SSR alleles not present in typical US-1 isolates. Group C for all markers tested is identical to the EC-1 lineage described on cultivated and wild potatoes in Ecuador. All isolates from S. ochranthum were able to re-infect their host of origin in the detached leaf assay; however, we did not draw clear conclusions as to the relative aggressiveness of the three groups on this host. Isolates of group A were the most specialized and were generally non-pathogenic or weakly pathogenic on all hosts other than S. ochranthum. Groups B and C infected tuber-bearing hosts, including the cultivated potato but were generally non-pathogenic on other non-tuber bearing hosts. Solanum ochranthum was infected by isolates coming from tuber-bearing Solanum hosts (i.e., the EC-1 lineage of P. infestans) and some US-1 isolates from non-tuber bearing hosts. Thus, in nature this species might be a potential reservoir of inoculum of different pathogen populations able to infect the cultivated hosts potato, tomato and pear melon (S.␣muricatum). Unlike potato and tomato in Ecuador, each of which is primarily attacked by a highly specialized pathogen population, S. ochranthum appears to harbour at least three pathogen groups of␣different genetic make-up. The unresolved issue of potential host specificity in isolates found on S.␣ochranthum could complicate efforts to use this species in tomato improvement.  相似文献

ABSTRACT Verticillium dahliae isolates from artichoke, bell pepper, cabbage, cauliflower, chili pepper, cotton, eggplant, lettuce, mint, potato, strawberry, tomato, and watermelon and V. albo-atrum from alfalfa were evaluated for their pathogenicity on all 14 hosts. One-month-old seedlings were inoculated with a spore suspension of about 10(7) conidia per ml using a root-dip technique and incubated in the greenhouse. Disease incidence and severity, plant height, and root and shoot dry weights were recorded 6 weeks after inoculation. Bell pepper, cabbage, cauliflower, cotton, eggplant, and mint isolates exhibited host specificity and differential pathogenicity on other hosts, whereas isolates from artichoke, lettuce, potato, strawberry, tomato, and watermelon did not. Bell pepper was resistant to all Verticillium isolates except isolates from bell pepper and eggplant. Thus, host specificity exists in some isolates of V. dahliae. The same isolates were characterized for vegetative compatibility groups (VCGs) through complementation of nitrate nonutilizing (nit) mutants. Cabbage and cauliflower isolates did not produce nit mutants. The isolate from cotton belonged to VCG 1; isolates from bell pepper, eggplant, potato, and tomato, to VCG 4; and the remaining isolates, to VCG 2. These isolates were also analyzed using the random amplified polymorphic DNA (RAPD) method. Forty random primers were screened, and eighteen of them amplified DNA from Verticillium. Based on RAPD banding patterns, cabbage and cauliflower isolates formed a unique group, distinct from other V. dahliae and V. albo-atrum groups. Minor genetic variations were observed among V. dahliae isolates from other hosts, regardless of whether they were host specific or not. There was no correlation among pathogenicity, VCGs, and RAPD banding patterns. Even though the isolates belonged to different VCGs, they shared similar RAPD profiles. These results suggest that management of Verticillium wilt in some crops through crop rotation is a distinct possibility.  相似文献

ABSTRACT The Ceratocystis fimbriata complex includes many undescribed species that cause wilt and canker diseases of many economically important plants. Phylogenetic analyses of DNA sequences have delineated three geographic clades within Ceratocystis fimbriata. This study examined host specialization in the Latin American clade, in which a number of lineages were identified using sequences of the internal transcribed spacer (ITS) region of the rDNA. Three host-associated lineages were identified from cacao (Theobroma cacao), sweet potato (Ipomoea batatas), and sycamore (Platanus spp.), respectively. Isolates from these three lineages showed strong host specialization in reciprocal inoculation experiments on these three hosts. Six cacao isolates from Ecuador, Trinidad, and Columbia differed genetically from other cacao isolates and were not pathogenic to cacao in inoculation tests. Further evidence of host specialization within the Latin American clade of Ceratocystis fimbriata was demonstrated in inoculation experiments in growth chambers using sweet potato, sycamore, Colocasia esculenta, coffee (Coffea arabica), and mango (Mangifera indica) plants; inoculation experiments in Brazil using Brazilian isolates from cacao, Eucalyptus spp., mango, and Gmelina arborea; and inoculation experiments in Costa Rica using Costa Rican isolates from cacao, coffee, and Xantho-soma sp. Hosts native to the Americas appeared to be colonized by only select pathogen genotypes, whereas nonnative hosts were colonized by several genotypes. We hypothesize that local populations of Ceratocystis fimbriata have specialized to different hosts; some of these populations are nascent species, and some host-specialized genotypes have been moved to new areas by humans.  相似文献

Published data on the susceptibility of plant species to 18 tymoviruses were collated. These data showed that tymoviruses infected more species from the family of their natural field host than species from other families. The data also indicated that tymoviruses infected a greater proportion of species from the same primary division of the dicotyledons (crassinucellate or tenuinucelate) as their natural host than other species, but the correlation was not statistically significant.
The susceptibility of 18 crassinucellate and 18 tenuinucellate test plant species to 10 tymoviruses was determined. The viruses infected about three times more plant species from the primary taxonomic division that contained the family of their field host than species from the other division; this difference was statistically significant.
The separation of tymoviruses into two groups based on the taxonomic division containing their natural hosts and most of their experimental hosts correlates broadly with a classification of the tymoviruses based on the serological relationships of their particle coat proteins.  相似文献

An important constraint for crop production in Colombia is the high incidence of anthracnose caused by Colletotrichum species. Although several studies have focused on these fungi, the relationship between the different fungal species within the genus and their hosts and whether they display any host preference or host specificity has yet to be examined. In Colombia, diseases caused by Colletotrichum species are particularly severe in mango (Mangifera indica) and tree tomato (Solanum betaceum) crops. In a previous investigation, the Colletotrichum phylogenetic species attacking these crops were identified. The present study aimed to determine whether isolates collected from tree tomato and mango showed host preference or host specificity by assessing aggressiveness, spore density, latent period, and fitness of each strain on the two hosts. In the departments of Cundinamarca and Tolima, Colombia, isolates were collected from plants that presented typical anthracnose symptoms and were identified as C. acutatum, C. asianum, C. boninense, C. gloeosporioides, C. tamarilloi and C. theobromicola. Inoculation of conidia of each isolate onto both hosts showed isolates had no host preference and only the C. gloeosporioides isolate showed host specificity. However, in general, isolates produced a higher spore density when inoculated on the alternate host, which may indicate a difference in the degree of adaptation to each host. Statistical analyses of the assessed parameter values revealed that isolates use different infection strategies when infecting each host. In light of these results, the implications of using quantitative estimations of fitness when studying fungal pathogens are discussed.  相似文献

The aggressiveness of Alternaria dauci isolates was investigated in greenhouse conditions. Twenty‐seven isolates were pre‐selected from a large collection to represent high diversity according to geographic or host origins and intergenic spacer (IGS) polymorphism. IGS sequence analysis revealed that isolates were grouped within three different clusters. Eleven isolates were selected and inoculated on a susceptible carrot cultivar. Three criteria (mean lesion number, mean necrotic leaf area and mean disease index) were used to assess the aggressiveness of isolates. Continuous variation in aggressiveness was shown and no clear division into isolate classes was evident. For the host range study, two isolates were inoculated under greenhouse conditions onto nine cultivated Apiaceae species, two wild Daucus species and six cultivated non‐Apiaceae species representing six botanical families. Lesions varying in severity were observed on all dicot species (Apiaceae and non‐Apiaceae), but no symptoms developed on the two monocots studied (leek and sweetcorn). Plant species were also differentiated on the basis of expanding lesions (cultivated and wild carrot, dill and fennel) or non‐expanding lesions (other dicot species). Typical A. dauci conidia were observed after in vitro incubation of leaves with symptoms. Fungal structures were isolated from lesions and A. dauci was confirmed on the basis of conidial morphology and specific conventional PCR results. Genotyping of individual isolates performed with microsatellite markers confirmed the presence of the inoculated isolate. The results clearly showed that, in controlled conditions, the host range of A. dauci is not restricted to carrot.  相似文献

The aggressiveness of four Phytophthora infestans isolates collected from wild and cultivated potato species (sect. Petota ) and the level of resistance of nine Petota species were assessed in the highland tropics of Ecuador. For this, isolates of P. infestans were inoculated on whole plants of Petota species in the field and net house and six epidemiological components – infection frequency (IF), incubation period (IP), latent period (LP), lesion size (LS), lesion growth rate (LGR), and relative area under the lesion expansion curve (RAULEC) – were measured during a single infection cycle. Additionally, host specificity was determined by testing for a significant host by pathogen interaction using the same components. The results showed significant differences among isolates of the EC-1 clonal lineage for IP, IF, and RAULEC. Significant differences among isolates were not found for the other components measured. There were significant differences in resistance among the accessions of Petota hosts tested. RAULEC, LGR, LP, and LS were in general more adequate in differentiating among the more resistant and more susceptible accessions but the importance of each component varied with host species. There was slight and inconsistent evidence for the existence of host specificity in some isolates of Petota hosts. ABSTRACT Numerous morphological species of pathogenic fungi have been shown to actually encompass several genetically isolated lineages, often specialized on different hosts and, thus, constituting host races or sibling species. In this article, we explore theoretically the importance of some aspects of the life cycle on the conditions of sympatric divergence of host races, particularly in fungal plant pathogens. Because the life cycles classically modeled by theoreticians of sympatric speciation correspond to those of free-living animals, sympatric divergence of host races requires the evolution of active assortative mating or of active host preference if mating takes place on the hosts. With some particular life cycles with restricted dispersal between selection on the host and mating, we show that divergence can occur in sympatry and lead to host race formation, or even speciation, by a mere process of specialization, with strong divergent adaptive selection. Neither active assortative mating nor active habitat choice is required in these cases, and this may explain why the phylo-genetic species concept seems more appropriate than the biological species concept in these organisms.  相似文献
