广东稻瘟病菌的遗传宗谱与致病性的关系   总被引:13,自引:2,他引:11  
不同病圃同一套寄虫主的田间表现及其穗颈瘟分离菌株的遗传宗谱和致病性分析试验结果表明,不同生态稻区稻瘟病菌的小种分布类型不同,对品种的致病性也存在差异。将已经过RFLP和PCR技术进行DNA指纹分析,并划分为不同遗传宗谱的稻瘟病菌株进行致病性测定。结果表明,菌株的致病性与其遗传宗谱类型、生理小种类型、寄主及采集地点存在密切的关系:(1)同一遗传宗谱、分离自相同寄主、鉴定出来的生理小种类型又较接近的菌  相似文献

Quantitative trait loci (QTLs) conferring resistance to rice blast, caused by Magnaporthe oryzae, have been under-explored. In the present study, composite interval mapping was used to identify the QTLs that condition resistance to the 6 out of the 12 common races (IB1, IB45, IB49, IB54, IC17, and ID1) of M. oryzae using a recombinant inbred line (RIL) population derived from a cross of the moderately susceptible japonica cultivar Lemont with the moderately resistant indica cultivar Jasmine 85. Disease reactions of 227 F(7) RILs were determined using a category scale of ratings from 0, representing the most resistant, to 5, representing the most susceptible. A total of nine QTLs responsive to different degrees of phenotypic variation ranging from 5.17 to 26.53% were mapped on chromosomes 3, 8, 9, 11, and 12: qBLAST3 at 1.9 centimorgans (cM) to simple sequence repeat (SSR) marker RM282 on chromosome 3 to IB45 accounting for 5.17%; qBLAST8.1 co-segregated with SSR marker RM1148 to IB49 accounting for 6.69%, qBLAST8.2 at 0.1 cM to SSR marker RM72 to IC17 on chromosome 8 accounting for 7.22%; qBLAST9.1 at 0.1 cM to SSR marker RM257 to IB54, qBLAST9.2 at 2.1 cM to SSR marker RM108, and qBLAST9.3 at 0.1 cM to SSR marker RM215 to IC17 on chromosome 9 accounting for 4.64, 7.62, and 4.49%; qBLAST11 at 2.2 cM to SSR marker RM244 to IB45 and IB54 on chromosome 11 accounting for 26.53 and 19.60%; qBLAST12.1 at 0.3 cM to SSR marker OSM89 to IB1 on chromosome 12 accounting for 5.44%; and qBLAST12.2 at 0.3 and 0.1 cM to SSR marker OSM89 to IB49 and ID1 on chromosome 12 accounting for 9.7 and 10.18% of phenotypic variation, respectively. Rice blast is the most serious disease threat to rice production worldwide. It is difficult to control due to the complex diversity and wide geographic distribution of the causal pathogen Magnaporthe oryzae. In Australia, rice blast occurs in northern Australia but remains exotic to the main south-eastern rice growing area; however, there is the potential for rice blast to threaten this area; in addition, rice production is currently expanding from south-eastern Australia into northern Australia, which makes rice blast a major concern and challenge to rice industry in Australia. Prior to this study, there was lack of information on the race status of M. oryzae present in Australia and on how to manage the disease through host resistance. The races of rice blast isolates collected in northern Australia was characterised based on the disease reactions of eight standard rice differentials used in an international race differential system. The following studies revealed genes conferring resistance to these races through investigating the responses of 25 monogenic rice lines with targeted resistance gene against different races. The rice blast isolates were characterised into five races: IA-1, IA-3, IA-63, IB-3 and IB-59. Genes Pi40, Piz-t, Pi9, Pi5(t) and Pi12(t) exhibited resistance to all the isolates belonging to five races. In addition, two genes showed complete resistance to multiple races, viz. Pi9 that showed complete resistance to races IA-1, IA-3, IA-63 and IB-3 and Pita2 that had complete resistance to races IA-3, IB-3 and IB-59. This study provides information about the races of M. oryzae in Australia. Genes identified conferring resistance to multiple races will not only streamline the identification via molecular markers of imported rice varieties with resistance to rice blast in Australia, but will also allow the Australian rice breeding program to develop new varieties with broad-spectrum resistance to rice blast and pyramid multi-gene resistance into Australian rice varieties.  相似文献

以苗期16个稻瘟病生理小种的抗谱测定以及成株期叶瘟、穗颈瘟鉴定结果为依据,分析了75份中国栽培稻稻瘟病抗性资源的抗瘟性。结果显示,不同抗性品种的抗谱以及对成株期叶瘟和穗颈瘟的抗性反应差异较大。相关分析表明,苗期抗谱测定结果与成株期叶瘟抗性显著相关,而与穗颈瘟,以及叶瘟与穗颈瘟间相关不显著。根据聚类分析结果,抗性资源可分成9个类群,其中以第6类群包含品种最多,抗谱最广(81.3%~100%),并高抗A、B、E、G群生理小种和抗穗颈瘟,可作为水稻稻瘟病抗性育种优先利用的种质资源。  相似文献

云南元阳哈尼梯田稻瘟病菌遗传多样性分析   总被引:7,自引:0,他引:7  
 利用12对SSR引物,对来自云南元阳哈尼梯田不同海拔高度、不同水稻品种上的稻瘟病菌菌株进行遗传多样性分析,并运用日本清泽建立的鉴别品种对其中62个菌株进行生理小种鉴定。结果显示:稻瘟病菌总体遗传多样性水平较高(He=0.57±0.27,I=1.17±0.60)。聚类分析表明,在80%相似水平下菌株可划分为9个多样性群体。供试菌株划分为16个生理小种,其中002、000和006为优势小种。分离的不同海拔稻瘟病菌生理小种和遗传多样性存在差异。海拔1600~1700m生理小种最丰富,达9个生理小种;海拔1500~1600m、1400~1500m和1700~1800m,生理小种分别为6、5和4个;遗传多样性分析表明,从海拔1400~1500m分离的菌株遗传多样性水平最高(He=0.57±0.24),并划分为6个群体。综合分析表明,在元阳哈尼梯田系统中,栽培传统品种遗传多样性越丰富的区域,其稻瘟病菌群体多样性和生理小种也越丰富。  相似文献

DNA markers in chickpea, targetting resistance genes for different races of Fusarium oxysporum f.sp. ciceris (Foc), have been identified in chickpea, but validation of these markers is essential for effective use in resistance breeding. In view of this, different simple sequence repeats (SSR) markers were analysed in Pakistani germplasm including induced mutants and some local lines. Most of the SSR markers showed good correlation with phenotypic evaluation of genotypes to different races of Foc and may be used effectively in resistance breeding, except those markers for race 3. Markers for race 3 showed deviations from phenotypic data and the reason might be that race 3 is actually Fusarium proliferatum as reported recently and resistance to this race might involve some other major resistance genes. Poor correlation of markers with foc-3 on LG2 in our study and a recent report of independent segregation of foc-2 and foc-3 in near isogenic lines suggested that linkage distances among different resistance genes need further investigation. Moreover three Pakistani mutant lines (97477, CM444/92 and CM368/93) depicted high levels of resistance to Foc races and can be deployed as a valuable source in resistance breeding programmes.  相似文献

Sunflower rust, caused by Puccinia helianthi, is a prevalent disease in many countries throughout the world. The U.S. Department of Agriculture (USDA)-Agricultural Research Service, Sunflower Research Unit has released rust resistant breeding materials for several decades. However, constantly coevolving rust populations have formed new virulent races to which current hybrids have little resistance. The objectives of this study were to identify resistance to race 336, the predominant race in North America, and to race 777, the most virulent race currently known, and to validate molecular markers known to be linked to rust resistance genes in the sunflower gene pool. A total of 104 entries, including 66 released USDA inbred lines, 14 USDA interspecific germplasm lines, and 24 foreign germplasms, all developed specifically for rust resistance, were tested for their reaction to races 336 and 777. Only 13 of the 104 entries tested were resistant to both races, whereas another six were resistant only to race 336. The interspecific germplasm line, Rf ANN-1742, was resistant to both races and was identified as a new rust resistance source. A selection of 24 lines including 19 lines resistant to races 777 and/or 336 was screened with DNA markers linked to rust resistance genes R(1), R(2), R(4u), and R(5). The results indicated that the existing resistant lines are diverse in rust resistance genes. Durable genetic resistance through gene pyramiding will be effective for the control of rust.  相似文献

Rice blast disease, caused by the filamentous fungus Pyricularia oryzae, is one of the most destructive diseases in rice worldwide. Breeding of resistant rice cultivars remains a cost-effective and environment-friendly means for controlling blast disease, but the resistance tends to break down over time because of the pathogen's rapid adaptation. In this study, AVRPiz-t gene sequences of 46 rice blast isolates were evaluated using a Southern blot analysis. The AVRPiz-t gene was present in 24 of 46 (52.2%) rice blast isolates. The pathogenicity assay showed that all blast isolates were avirulent against Japanese rice cv. Toride 1, which carries several rice blast resistance genes including Piz-t, Pii, Pi37, and Pi-ta. Screening for the Piz-t gene in Thai rice germplasm revealed that less than 20% of rice varieties harbour the Piz-t gene. Therefore, the Toride 1 rice variety could serve as an effective donor of rice blast resistance to be used in rice breeding programmes in Thailand. This study provides evidence for co-evolution between the rice blast resistance gene Piz-t and the rice blast fungal avirulence gene AVRPiz-t. Understanding this relationship will facilitate the sustainable development of breeding for rice blast resistance in the future.  相似文献

 采用我国当前流行的小麦条锈菌小种和重要致病类型, 在常温条件下对普通小麦-华山新麦草易位系H9015-17进行苗期抗条锈性鉴定, 并用当前主要流行小种CYR32对H9015-17与铭贤169的杂交后代及其双亲进行抗条锈性遗传分析, 以揭示H9015-17抗条锈性遗传基础。结果显示, H9015-17对小麦条锈菌小种CYR31、CYR32、CYR33和致病类型Su11-4、Su11-7、V26、Su11-11均有良好的抗病性, 对当前主要流行小种CYR32的抗病性由1对显性基因控制, 暂命名为YrHua1。 采用分子标记定位技术,筛选到5个与抗病基因YrHua1连锁的RGAP标记(M1M2M3M4M5)和1个SSR标记(Xgwm292),这些标记与抗病基因YrHua1的遗传距离分别为17.3、15.7、13.1、3.3、2.9和11.2,并将基因YrHua1定位在小麦染色体5DL上。研究结果将为分子标记辅助选择改良小麦抗条锈性提供宝贵的种质材料,建议在抗病育种加以利用。  相似文献

用4个稻瘟病菌小种对浙江省280份水稻新品种(系)进行稻瘟病抗性分析,结果表明:浙江省水稻新品种(系)对稻瘟病菌具有较好的抗性,籼稻和粳稻对参试菌株的全抗率分别为21.62%和39.05%,说明粳稻具有较籼稻更广的抗谱;浙江省新品种(系)对ZB13小种具有较高的感病率,籼稻和粳稻的感病率分别为69.37%和52.07%。  相似文献

The rice blast resistance gene Pi54 (formerly Pi-k h ) isolated from indica rice line Tetep confers broad spectrum resistance to different strains of Magnaporthe oryzae in India. In this study, we performed PCR based allele mining for blast resistance gene Pi54 from six cultivated rice lines and eight wild rice species to understand its structural variation and its impact on the phenotypes. Sequence analysis indicates presence of more variation between cultivated and wild species (35–90 %) than variation found among cultivated species (1–20 %). Structural analysis of alleles showed presence of variable number of Open Reading Frames (0–2) principally having point mutations in the leucine rich repeats (LRR) regions. The Ka/Ks ratio of LRR region was >1, which shows the effect of selection pressure at this domain. The Pi54 alleles have 142 polymorphic sites with average nucleotide diversity of 0.04522. The Ka/Ks ratio of coding region ranged from 0 to >1 and Tajima’s D test showed negative as well as Darwinian selection within the alleles, which corresponded well with their phenotypic reaction to M. oryzae. The results obtained in this study shows divergent nature of Pi54 allele in wild species and land races of rice. The resistance alleles identified in this study can be used in effective management of rice blast disease through gene pyramiding.  相似文献

抗稻瘟病单基因系对籼稻稻瘟病菌小种鉴别力分析   总被引:16,自引:3,他引:16  
 利用30个抗稻瘟病单基因系对146个来自广东的稻瘟病菌单孢分离菌株进行了致病性测定,其中17个单基因系对测试菌株的致病性有较好的鉴别力。基于主成分因子分析法,对17个单基因系和146个菌株组成的互作变量矩阵进行了因子提取,结果表明其中9个单基因系为代表的抽提因子可以解释变量总方差的80.437%,初步组建了这9个单基因系构成的稻瘟病菌生理小种单基因系鉴别寄主。按其对变量方差贡献大小排序,它们分别是:IRBLkp-K 60(Pi-kp)、IRBLi-F5(Pi-i)、F128-1(Pi-ta2)、IRBL9-W (Pi-9(t))、IRBLsh-S (Pi-sh(1))、IRBLz-Fu (Pi-z)、IRBLkh-K3(Pi-kh)、IRBL1-CL (Pi-1)、IRBLz5-CA (Pi-z5)。用这9个单基因系可将146个菌株划分为83个小种,将来自广西、福建、湖南、湖北、浙江、四川等籼稻区的54个菌株划分为26个小种。筛选的9个单基因系对我国籼稻区稻瘟病菌致病性有较强的鉴别力。在抗病性鉴定中,基于单基因系的代表菌株选择,可考虑应用聚类和变量分析法进行。  相似文献

ABSTRACT Stripe rust, caused by Puccinia striiformis f. sp. hordei, is one of the most important diseases of barley in the south-central and western United States. Growing resistant cultivars is the best approach for controlling the disease. The barley genotype BBA 2890 has all-stage resistance against all races of P. striiformis f. sp. hordei (PSH) identified thus far in the United States. The resistance in BBA 2890 is controlled by a single recessive gene, rps1.a. The objectives of this study were to identify resistance gene analog polymorphism (RGAP) markers for the all-stage resistance gene rps1.a, to map the gene on a barley chromosome using chromosome-specific simple sequence repeat (SSR) markers, and to determine the presence or absence of the flanking RGAP markers for the gene in 24 barley genotypes. Seedlings of the parents and 200 F(8) recombinant inbred lines (RILs) were tested for resistance to pathogen races PSH-14, PSH-48, and PSH-54 in the greenhouse in 2005. Genomic DNA was extracted from the parents and 150 F(8) RILs. The RGAP technique was used to identify molecular markers for the rps1.a gene. Twelve primer pairs generating repeatable polymorphic bands were selected for genotyping the 150 F(8) RILs. A genetic linkage group was constructed for the resistance gene with 13 RGAP markers and four chromosome-specific SSR markers. The four SSR markers mapped the gene on the long arm of barley chromosome 3H. The closest RGAP marker for the resistant allele was within a genetic distance of 2.1 centimorgans (cM). The closest marker for the susceptible allele was 6.8 cM away from the locus. The two closest RGAP markers for the resistant allele detected polymorphisms in 67 and 71% of the 24 barley genotypes when used individually, and detected polymorphism in 88% of the genotypes when used in combination. This information should be useful in incorporating the resistance gene into barley cultivars and in pyramiding the gene with other resistance genes for superior stripe rust resistance.  相似文献

 西藏半野生小麦(Triticum aestivum ssp. tibetanum Shao)、云南铁壳麦(T. aestivum ssp. yunnanense King)和新疆稻麦(T. petropavloski Udats et Migusch)是我国特有的3个普通小麦亚种,它们具有特定的地理分布、明显不同于普通小麦的形态学特征,具有抗病、耐逆等优良性状,是可用于现代小麦品种生物和非生物胁迫改良的优异种质资源。本研究利用当前在我国流行频率高、毒性强的条锈菌生理小种和致病类群组成的混合菌对213份中国特有小麦种质(包括117份西藏半野生小麦、78份云南铁壳麦和18份新疆稻麦)进行苗期和成株期抗性表型鉴定,并利用与12个已知条锈病抗性基因(Yr)相关的分子标记进行分子检测。结果表明,共有18份中国特有小麦种质对混合生理小种表现苗期抗性,89份表现成株期抗性。进一步分析发现,成株期抗性种质主要来源于云南铁壳麦。结合系谱分析、抗性表型及条锈病抗病基因分子标记检测结果发现,2份中国特有小麦种质携带Yr18,所有种质均不携带Yr5、Yr9、Yr10、Yr15、Yr17、Yr24/Yr26、Yr30、Yr41、Yr48、Yr65和Yr67等已知抗病基因。未检测到供试基因的抗性种质可能携带其他已知或新的条锈病抗性基因。该研究为进一步有效利用中国特有小麦亚种抗条锈病种质和发掘其抗性基因提供依据。  相似文献

 为明确吉林省主栽水稻品种的抗瘟性和抗瘟基因型,选用了一套已明确其无毒基因组成的稻瘟病菌标准菌株对吉林省68个主栽水稻品种进行喷雾和离体划伤接种试验。结果表明,68个品种的抗瘟能力存在较大差异,中抗以上抗病品种达到44个,占供试品种的64.7%;抗瘟基因推导结果结合特异性分子标记检测表明,Pita、Pia、Pish、Pita2、PibPi9等6个基因为吉林省主栽水稻品种中主要的抗瘟基因,并且品种中含有的抗瘟基因数量与抗瘟能力呈正相关;根据结果推测,测试的所有品种中可能不含有Pi2、Pikh、Pikm、Pi11和Pi12基因。本研究揭示了吉林省近年主栽水稻品种的抗瘟能力和抗瘟基因组成,明确了吉林稻区主效抗瘟基因和基因聚合对稻瘟病的抗性贡献,为进一步培育广谱持久抗瘟品种和合理布局抗病品种提供了重要参考。  相似文献

ABSTRACT Two hundred and seventy-six accessions of mainly Brassica spp. were screened for resistance to Xanthomonas campestris pv. campestris races. In Brassica oleracea (C genome), the majority of accessions were susceptible to all races, but 43% showed resistance to one or more of the rare races (2, 3, 5, and 6) and a single accession showed partial resistance to races 1, 3, 5, and 6. Further searches for resistance to races 1 and 4, currently the most important races worldwide, and race 6, the race with the widest host range, were made in accessions representing the A and B genomes. Strong resistance to race 4 was frequent in B. rapa (A genome) and B. napus (AC genome), indicating an A genome origin. Resistance to races 1 and 4 was present in a high proportion of B. nigra (B genome) and B. carinata (BC genome) accessions, indicating a B genome origin. B. juncea (AB genome) was the most resistant species, showing either strong resistance to races 1 and 4 or quantitative resistance to all races. Potentially race-nonspecific resistance was also found, but at a lower frequency, in B. rapa, B. nigra, and B. carinata. The combination of race-specific and race-nonspecific resistance could provide durable control of black rot of crucifers.  相似文献

1991 ̄1995年对广东稻瘟病菌生理小种及部分主栽品种抗性的研究结果表明,广东稻瘟病菌小种分布及发生动态相对稳定,优势种群为ZB群,ZC群;优势小种为ZB13、ZB5、ZC13和ZB1,不同生态稻区优势小种有差异。品种间具有不同的抗性水平,本省育成推广的主栽品种对稻瘟病属中等的抗性水平,个别品种抗性偏低,属于中感至感。粳籼89等的抗病性逐年下降,这些主栽品种抗性丧失的主要原因,一方面是原有小种的  相似文献

Blast caused by Pyricularia grisea [teleomorph: Magnaporthe grisea] is an economically important and widespread disease of finger millet in the world. Host resistance is the most economical and effective means of combating this disease as finger millet is predominantly grown by resource-poor and marginal farmers. At the International Crops Research Institute for the Semi-Arid Tropics (ICRISAT), we evaluated a finger millet mini-core collection of 80 germplasm accessions (about 1 % of the total germplasm collection representing major trait variability) for blast resistance both in the field and greenhouse. Field evaluation was done using a refined screening technique that included new improved rating scales for leaf, neck and finger infection. Sixty six of the 80 accessions showed combined resistance to leaf, neck and finger blast in two seasons (2009 and 2010) of field screening. A highly significant and positive correlation was found between neck and finger blast ratings (r?=?0.92), whereas small but significant correlations were found between leaf blast and neck blast (r?=?0.25) and between leaf blast and finger blast (r?=?0.30). These accessions were also screened for leaf blast resistance in the greenhouse by artificial inoculation of seedlings to confirm field observations. Fifty-eight of the 80 accessions were resistant to leaf blast in the greenhouse screen as well. These resistant accessions represented one wild (africana) and four cultivated races (vulgaris, plana, elongate and compacta) of finger millet that originated from 13 countries in Asia and Africa and exhibited considerable diversity for agronomic traits, such as maturity period, plant height and panicle type. These blast resistant accessions from the mini-core collection would be useful in finger millet disease resistance breeding programs.  相似文献

用中国7个统一鉴别品种对2002年在黑龙江省部分地区采集的57个单孢分离菌株进行了生理小种鉴定。共鉴定出6群10个生理小种,即ZA17、ZA49、ZB1、ZB19、ZB25、ZC9、ZD1、ZD5、ZE1和ZF1生理小种,其中ZD1为优势小种,出现频率为36.8%。生理小种的组成由1996年商世吉报导的4群7个生理小种发展到6群10个生理小种,优势小种也由ZE1转变为ZD1。其中ZA17、ZA49强致病型的生理小种开始出现。所鉴定出的10个生理小种对4个粳稻鉴别品种有较强的致病力。地域较近的地区稻瘟病菌的生理小种群体组成也有较大差异。  相似文献

稻瘟病是四川省水稻的主要病害,实践证明,选育种植抗病品种是控制该病害的最经济有效的措施。1981年开始,植物病理工作者与遗传育种工作者经过20年的协作攻关,研究制定了一套适合四川生态特点的有关病菌生理小种、水稻品种抗瘟性鉴定的规范化技术和评价体系,探明稻瘟病菌致病性变异和水稻品种抗瘟性丧失规律,筛选出200多份稻瘟病抗源材料,育成穗颈瘟1-5级的抗病品种(组合)27个,累计推广面积达2750万hm^2,有效地控制了稻瘟病的流行。在此基础上,提出今后重点研究领域和抗病育种策略。  相似文献
