北京地区发现番茄斑萎病毒   总被引:6,自引:0,他引:6  
通过采用特异快速试纸条以及进一步的RT-PCR和序列测定证实,在北京局部地区的辣椒和茄子上已发生番茄斑萎病毒,这两种作物上同时发生的蓟马种类主要为西花蓟马,并且携带该病毒。番茄斑萎病毒病是极其危险的一种病害,可对作物造成毁灭性灾害。建议有关部门立即采取相应的预防与管理措施,防止其扩散蔓延。  相似文献

辣椒是一类重要的经济作物,在世界各地广泛种植.辣椒感染病毒后,对其产量和品质都有较大的影响.目前可侵染辣椒的病毒至少有45种,其中包括番茄斑萎病毒属Tospovirus的番茄斑萎病毒Tomato spotted wilt virus(TSWV)、凤仙花坏死病毒Impatiens necrotic spot virus(INSV)、番茄褪绿斑病毒Tomato chlorosis spot virus(TCSV)以及辣椒褪绿病毒Capsicum chlorosis virus(CaCV).  相似文献

从云南蝴蝶兰上检测到番茄斑萎病毒属病毒   总被引:4,自引:0,他引:4  
 应用电镜观察、DAS-ELISA以及RT-PCR检测,从症状表现黄化、环斑的云南蝴蝶兰(Phalaenopsis amabilis)病样中分离得到的一个病毒分离物Tospo-Pha。该分离物粒子近球形、具包膜、直径约90nm,与番茄斑萎病毒属(Tospovirus)的西瓜银色斑驳病毒(Watermelon silver mosaic virus,WSMoV)/花生芽坏死病毒(Groundnut bud necrosis virus,GBNV)复合抗血清呈强阳性反应,分子检测发现该分离物SRNA5'末端序列与CaCV大岩桐分离物(CaCV-Gloxinia)同源性最高(91.0%),在系统进化树中与CaCV聚于同一分支。上述结果表明,从云南蝴蝶兰中分离到的Tospo-Pha属于Tospovirus病毒。  相似文献

根据番茄斑萎病毒属(Tospovirus)中6种病毒S RNA上的N基因序列设计特异性引物,建立可同时检测这6种Tospovirus病毒的多重PCR体系。多重PCR扩增结果显示,番茄环纹斑点病毒(776 bp)、甜瓜黄斑病毒(505 bp)、鸢尾黄斑病毒(296 bp)、凤仙花坏死斑病毒(221 bp)、番茄斑萎病毒(175 bp)和番茄褪绿斑病毒(110 bp)均出现清晰的目标条带,各病毒的特异性引物不会对其他病毒产生非特异性扩增。本研究建立的6种Tospovirus病毒的多重PCR检测方法,有助于提高目标病毒的检测效率。  相似文献

辣椒是云南省主要经济作物之一,近年来病毒病尤其是正番茄斑萎病毒属病毒发病严重,影响了辣椒产量和品质。利用RT-PCR技术对从云南辣椒主产区采集的疑似感染正番茄斑萎病毒属病毒的25份辣椒样品进行分子鉴定,结果显示,12份样品检测出正番茄斑萎病毒属病毒,检出率为48.0%,其中6份是番茄斑萎病毒tomato spotted wilt orthotospovirus (TSWV),检出率为24.0%;5份是番茄环纹斑点病毒tomato zonate spot orthotospovirus (TZSV),检出率为20.0%;有1份是TSWV和TZSV复合侵染,检出率为4.0%,这是在云南辣椒生产上首次发现TSWV和TZSV的复合侵染。通过鉴定,初步了解正番茄斑萎病毒属病毒在云南辣椒生产中的发生情况和种类,为制定云南地区该属病毒的防治策略提供理论依据。  相似文献

番茄斑萎病毒Tomato spotted wilt tospovirus(TSWV)是严重危害世界经济作物的一种病毒,寄主范围广泛。我们在研究中发现番茄斑萎病毒能侵染我国大蒜Allium sativum L.。利用摩擦接种法将TSWV接种到健康大蒜上,结果显示:接种14 d后大蒜新生叶片出现褪绿和白色斑点症状。ELISA检测显示大蒜叶片汁液与TSWV的单克隆抗体产生血清学反应,采用TSWV N基因的引物对大蒜叶片总RNA进行RT-PCR,结果扩增出约800 bp的条带,在NCBI上BLAST显示与TSWV YN5576的同源性最高,为98.52%。这些数据表明番茄斑萎病毒系统侵染我国大蒜。  相似文献

西花蓟马Frankliniella occidentalis是世界性重要检疫性害虫之一,不仅直接取食危害作物而且传播病毒,从而造成极为严重的经济损失。由于西花蓟马在我国具有广泛的适生范围,随其入侵我国并随之传播的番茄斑萎病毒(Tomato spotted wilt virus)已在我国不同地域发现,对经济作物已形成严重威胁。本文综述了西花蓟马对番茄斑萎病毒的获取、携带和传播扩散过程及其病毒在蓟马体内的循环过程和机理,总结了影响西花蓟马传播番茄斑萎病毒效率的因素,并评述了西花蓟马-病毒-植物这一互作系统及其对西花蓟马生长发育适合度的影响,以期为我国西花蓟马传播番茄斑萎病毒的基础研究和防控提供理论依据与指导。  相似文献

Tospoviruses infecting vegetable crops in Israel   总被引:1,自引:0,他引:1  
Symptoms of vein clearing, stem necrosis, curling, necrotic spots and rings on the leaves associated with infection by tomato spotted wilt tospovirus (TSWV) were documented among vegetable crops growing in commercial glasshouses and open fields in Israel. Plants exhibiting symptoms were collected, from 1994-01 to 1998-12. Among cultivated vegetable crops analysed for TSWV by ELISA, the following plants were found to be infected: tomato, capsicum, aubergine, lettuce, cabbage and cucumber. These incidences of the virus were all correlated with the occurrence in high population of Frankliniella occidentalis. Transmission of the virus from infected Datura stramonium to Petunia leaf discs, by F. occidentalis , was up to 26%. TSWV antigens were readily detected by ELISA in seeds harvested from naturally infected vegetable crops. However, we failed to show virus transmission to the progeny plants. Iris yellow spot tospovirus (IYSV) was detected in onion. High incidence of the disease was associated with large populations of Thrips tabaci.  相似文献

Tospovirus serogroups I and III have recently been designated as species, tomato spotted wilt virus (TSWV) and impatiens necrotic spot virus (INSV), while the species status of serogroup II isolates remains undefined. Fifteen Tospovirus isolates from ornamental and vegetable crops in Liguria, Italy, were found to belong either to TSWV (seven isolates) or to INSV (eight isolates) on the basis of test-plant reactions, serological techniques using DAS ELISA kits raised against the nucleoproteins of the type members of the two species, and cytopathology. None of them could be assigned to serogroup II using DAS ELISA kits raised against nucleoproteins of this serogroup. Italian isolates representative of the two species reacted in indirect ELISA using a polyclonal antiserum against the entire particle of a TSWV isolate, but with higher intensity for our TSWV isolates than for the INSV isolates. Western blots and dot immunobinding assays confirmed that the nucleoproteins of the two species are unrelated whereas the glycoproteins are related. The cytopathology was similar for two isolates representative of TSWV and INSV, except that the type of filaments encountered was different, and appeared to be characteristic of the species.  相似文献

选用小鼠血清及脾脏组织中超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活力及丙二醛(MDA)含量为观察指标,从机体氧化和抗氧化系统是否失衡的角度研究了乙烯利的氧化应激作用。结果表明,乙烯利能明显降低小鼠血清及脾脏组织中SOD和GSH-Px的活性。其中:134,268 mg/kg bw剂量处理组血清SOD和GSH-Px活性分别降低了8.91% ,12.44%和4.18% ,8.54% ;268, 536 mg/kg bw剂量组脾脏SOD和GSH-Px活性分别降低了8.34% ,19.61%和9.72%,24.86% ;与对照组差异显著(PP<0.01)。结果显示,乙烯利能破坏小鼠机体氧化和抗氧化系统的平衡,引起氧化应激。  相似文献

对昆明市白车轴草上蓟马种类及其周年种群动态进行了调查。白车轴草上蓟马主要有西花蓟马(Fran-kliniella occidentalis)、丽花蓟马(F.intonsa)、八节黄蓟马(Thrips flavidulus)、端大蓟马(Megalurothrips distalis)、稻单管蓟马(Haplothrips aculeatus)和烟蓟马(T.tabaci),有时会出现黄胸蓟马(T.hawaiiensis)、玉米黄呆蓟马(Anaphothrips obscurus)、苏丹呆蓟马(A.sudanensis)和云南纹蓟马(Aeolothrips yunnanensis)。本文分析了前6种蓟马的周年种群动态,重点阐述了外来有害生物西花蓟马,该虫主要在3~6月发生,是此期间白车轴草上的蓟马优势种。  相似文献

ABSTRACT The NSs protein of Watermelon silver mottle virus (WSMoV) was expressed by a Zucchini yellow mosaic virus (ZYMV) vector in squash. The expressed NSs protein with a histidine tag and an additional NIa protease cleavage sequence was isolated by Ni(2+)-NTA resins as a free-form protein and further eluted after sodium dodecyl sulfate-polyacrylamide gel electrophoresis for production of rabbit antiserum and mouse monoclonal antibodies (MAbs). The rabbit antiserum strongly reacted with the NSs crude antigen of WSMoV and weakly reacted with that of a high-temperature-recovered gloxinia isolate (HT-1) of Capsicum chlorosis virus (CaCV), but not with that of Calla lily chlorotic spot virus (CCSV). In contrast, the MAbs reacted strongly with all crude NSs antigens of WSMoV, CaCV, and CCSV. Various deletions of the NSs open reading frame were constructed and expressed by ZYMV vector. Results indicate that all three MAbs target the 89- to 125-amino-acid (aa) region of WSMoV NSs protein. Two indispensable residues of cysteine and lysine were essential for MAbs recognition. Sequence comparison of the deduced MAbs-recognized region with the reported tospoviral NSs proteins revealed the presence of a consensus sequence VRKPGVKNTGCKFTMHNQIFNPN (denoted WNSscon), at the 98- to 120-aa position of NSs proteins, sharing 86 to 100% identities among those of WSMoV, CaCV, CCSV, and Peanut bud necrosis virus. A synthetic WNSscon peptide reacted with the MAbs and verified that the epitopes are present in the 98- to 120-aa region of WSMoV NSs protein. The WSMoV sero-group-specific NSs MAbs provide a means for reliable identification of tospoviruses in this large serogroup.  相似文献

近年来,昆明市榕树(Ficus)和鹅掌柴(Sche fflera)受到蓟马为害比较严重.经鉴定,为害榕树、造成叶片饺子状虫瘿的蓟马优势种类为母管蓟马属(Gynaikothrips)的榕母管蓟马(Gynaikothrips ficorum).为害鹅掌柴,造成叶片角状虫瘿的管蓟马属于滑管蓟马属(Liothrips)的鹅掌柴滑管蓟马(Liothrips sp.).5~6月和9~10月是虫瘿蓟马的发生高峰期,捕食性天敌黑纹透翅花蝽(Montandoniola moraguesi)在局部地区能起到自然控制蓟马种群的作用.  相似文献
