Although most of the species in Eriobotrya originated in China there are few studies on the genetics and genetic relationship of this genus. The aim of the present study was to clarify genetic relationships among 18 Eriobotrya accessions from China using amplified fragment length polymorphism (AFLP). Two close relatives Raphiolepisindica (L.) Lindl. and Photiniaserrulata Lindl. were used as outgroups. The results showed that 5 selected AFLP primer combinations amplified 297 scorable bands, of which 282 were polymorphic, yielding a polymorphism rate of 95%. Genetic similarity coefficient among Eriobotrya accessions ranged from 0.82 (between E. bengalensis Hook.f. and E. bengalensis forma angustifolia Vidal) to 0.41 (between E. serrate Vidal and E. deflexa Nakai) indicating substantial genetic variations in the Eriobotrya accessions evaluated. The unweighted pair-group method of arithmetic averages (UPGMA) cluster analysis, based on genetic similarity coefficient, produced a dendrogram which grouped 20 accessions into three main clusters. The result of cluster analysis was generally in agreement with the known taxonomic grouping. Furthermore, the clustering of the 18 accessions of Eriobotrya was consistent with systematic classification based on morphological characteristics.  相似文献

Inter-Simple Sequence Repeat (ISSR) markers were used for the assessment of genetic diversity among walnut (Juglans regia L.) selections from Greek native populations in comparison to internationally cultivated walnut genotypes. Similarity coefficient values from 0.13 to 0.93 (with an average of 0.48) were found among the 56 accessions examined, which indicated the presence of a high degree of genetic variability. Most international cultivars were grouped together while most Greek native populations could not be placed into a distinct group. The Greek native population genotypes were found more diverse than the international cultivars. The mean similarity coefficient values for the former and latter were 0.44 and 0.56, respectively. In the cultivar group, two subgroups were distinguished; one consisted of genotypes involving ‘Payne’ and the other ‘Franquette’ in their pedigrees. Some cultivars and populations could not be grouped according to their pedigrees or collection area. Analysis of molecular variance (AMOVA) revealed that a larger part of the genetic variation exists among Greek walnut populations within a collection region (89%) than among the regions (11%). The pairwise regional PhiPT values indicated that the most geographically distant regions are the most genetically differentiated. The high variability existing in the Greek germplasm in combination with their valuable agro-morphological traits suggested that it would be beneficial to utilize this native germplasm pool in walnut breeding programs and germplasm management activities to maximize genetic diversity in cultivated walnut.  相似文献

SRAP analytic system was used to assess genetic diversity of Rehmnnia glutinosa. Twenty-three Rehmnnia glutinosa cultivars were screened with 288 primer combinations, of which 13 produced stable and reproducible amplification patterns in three repetitive experiments. Among a total of 338 amplified fragments, 306 (90.5%) were polymorphic, with an average of 23.5 fragments for each primer combination. The percentage of polymorphic bands for each primer combination varied from 58.3 to 100%. The cultivars had a similarity ranging from 0.335 to 0.713 with a mean of 0.518. Shannon's diversity index and expected heterozygosity were 0.3217 and 0.2008, respectively. Based on the cluster, which were conducted on the similarity matrix of SRAP marker data, the cultivars were divided into four groups at the 20 rescaled distance cluster combine. The results demonstrated that SRAP is a stable marker technique for the assessment of genetic diversity of Rehmannia glutinosa cultivars, and that the level of genetic diversity among them from different production areas was relatively high.  相似文献

A global collection of ginger germplasm consisting of 46 accessions were characterized using two types of molecular markers, RAPD (Random Amplified Polymorphic DNA) and ISSR (Inter Simple Sequence Repeats). UPGMA dendrograms constructed based on three similarity coefficients, i.e., Jaccard's, Sorensen–Dice and Simple Matching using the combined RAPD and ISSR markers placed the accessions in four similar clusters in all the three dendrograms revealing the congruence of clustering patterns among the similarity coefficients and a rather less genetic distance among the accessions. Improved varieties/cultivars are grouped together with primitive types. Moreover, in the clustering pattern of the accessions, a geographical bias was also evident implying that germplasm collected from nearby locations especially with vernacular identity may not be genetically distinct. The clustering of the accessions was largely independent of its agronomic features.  相似文献

A total of 100 Pyrus L. accessions native mainly to East Asia were subjected to amplified fragment length polymorphism (AFLP) analysis to evaluate genetic variability and relationships among the accessions. Six AFLP primer combinations produced a total of 459 fragments, of which 410 were polymorphic with a polymorphism percentage of 89%. The Dice's similarity coefficient among pear accessions ranged from 0.671 (P. betulaefolia Bge and P. elaeagrifolia Pall.) to 0.947 (‘Umajirou’ and ‘Immuraaki’). Occidental pears generally had low similarities to Asian pears. The dendrogram generated from all the accessions by unweighted pair-group method of arithmetic analysis (UPGMA) cluster analysis clearly distinguished Occidental pears from accessions of East Asia. P. ussuriensis Maxim., P. betulaefolia and P. communis L. clustered separately into independent groups in accordance with their morphological classification. Japanese pear cultivars formed two groups with some Chinese white pears and Chinese sand pears. Chinese white pears and Chinese sand pears independently formed their own groups and also mingled into mixed groups in the dendrogram. Therefore, Chinese white pears were treated as a cultivated group or an ecotype of P. pyrifolia: P. pyrifolia White Pear Group. The information obtained from this study will be of great help for understanding the origin and evolution of Asian pear cultivars.  相似文献

Genetic diversity and genetic relationships of lotus (Nelumbo Adanson) cultivars were evaluated using allozyme and ISSR markers. The samples used covered 11 accessions of possible hybrids between Nelumbo nucifera and Nelumbo lutea and 92 accessions of N. nucifera including 69 flower lotus, 13 rhizome lotus, 5 seed lotus and 5 wild lotus. For allozyme studies, a total of 31 alleles at 23 loci of 18 enzyme systems were detected of which 5 (21.7%) loci Aat, Idh, Mdh-2, Pgd, Sod were polymorphic. The loci of Aat and Idh included two alleles, Mdh-2, Pgd and Sod included three alleles. Eighteen genotypes were detected with the 13 alleles of the 5 polymorphic loci. The parameters of average allele number, observed heterozygosity, expected heterozygosity and Shannon information index of 92 N. nucifera samples were 1.35 ± 0.71, 0.06 ± 0.21, 0.05 ± 0.14, 0.10 ± 023, respectively. Thirteen ISSR primers generated 93 loci, of which 37.63% were polymorphic across all samples. The percentage of polymorphic loci, average allele number, expected heterozygosity and Shannon information index of 92 N. nucifera samples were 26.67%, 1.30 ± 0.46, 0.10 ± 0.18 and 0.15 ± 0.25, respectively for the ISSR data. The ‘Bottleneck effect’ and rapid propagation of clones after the ice ages may explain the low genetic diversity of lotus. The dendrograms based on ISSR and allozymes were not congruent. Based on the ISSR data, the 103 samples were divided into the N. nucifera group (Group I), and the group containing inter-specific hybrids between N. nucifera and N. lutea (Group II). The flower lotus, rhizome lotus, and seed lotus each has multiple sources of origin. Plant size, a criterion commonly used in the classification of cultivars of lotus, is not correlated with genetic variation. Flower color is correlated with the cultivar classification to some degree, but its variation is complex in the hybrids.  相似文献

Random amplified microsatellite polymorphisms (RAMPOs) were used to assess genetic diversity among 30 date-palm cultivars and 10 male trees. Using 18 primers combinations, 197 bands were scored and 186 were polymorphic suggesting the high level of polymorphism among studied cultivars. Moreover, taking into account the high percentage of polymorphic bands (ppb), the resolving power (Rp) together with the polymorphism information content (PIC) scored values, all the tested primer sets contribute strongly in the discrimination of date-palm genotypes. In addition, the topology of the derived UPGMA dendrogram exhibited cultivars’ clustering made independently both from the geographical origin and/or from the sex of trees. The present data support the Mesopotamian origin of the date-palm domestication. Thus we assume that the used method is efficient to assess genetic diversity within date-palm cultivars. Data are discussed in relation with the opportunity of the RAMPO method to provide additional molecular markers suitable in the improvement of the date-palms germplasm characterisation.  相似文献

Italian lentil landraces are principally cultivated for self or local consumption. Most of them are disappearing, particularly macrosperma types by being less required by the market. A pre-requisite for the conservation and the efficient use of genetic resources is the better understanding of the extent and the distribution of the existing genetic variation, useful for future breeding programmes. Our study was undertaken to analyse and quantify the genetic diversity within and among three macrosperma Italian lentil landraces (Onano, Altamura and Villalba), using fluorescent AFLP markers. AFLP markers generated information to differentiate among closely related genotypes and group within the same cluster individuals belonging to the same landrace. The total genetic diversity (HT), the genetic diversity within population (HS) and the extent of differentiation between populations (DST) were 0.198, 0.155 and 0.043, respectively. The fixation index (GST = 0.219) showed that about 78% of the observed total genetic variation can be attributed to within population differences and around 22% is due to differences among populations. The gene flow estimate (Nm = 1.774) and the mean genetic distance value (0.077) suggested narrow genetic base among the analysed populations, confirming the tendency of Italian lentil landraces to group together. The present study showed that fluorescence-based AFLP technique is a biotechnological tool that can provide significant insights for research in genetic diversity of lentil landraces and their subsequent conservation and utilization in breeding programs.  相似文献

Collection and characterization of all sorts of germplasm resources are required for the development of new cultivars. Molecular characterization is more reliable than morphological characterization. Here, we employed sequence-related amplified polymorphism (SRAP) markers to evaluate genetic variation in a diverse collection of 56 Solanum accessions. Fifty-five SRAP primer combinations were used and a total of 635 polymorphic bands were observed. Cluster analysis by the unweighted pair-group method with arithmetic averages based on similarity matrices indicated that there were three clusters: (i) S. melongena; (ii) S. aethiopicum; (iii) S. surattense. The coefficients of genetic similarity among all the accessions ranged from 0.04 to 0.96 with an average of 0.73, and averaged 0.78 among S. melongena accessions originated from China, indicating extensive genetic variation. These results demonstrated that SRAP can be efficiently used to estimate genetic diversity and analyze phylogenetic relationship.  相似文献

Eighteen cultivars of cocoyam (Xanthosoma spp.) and two cultivars of taro (Colocasia esculenta (L.) Schott) from the USDA/ARS germplasm collection were evaluated for genetic relatedness using RAPD data. Seven random primers generated 40 RAPD loci. Of the 18 cultivars screened, 11 (61%) were identical at all RAPD loci evaluated. A similarity matrix was constructed on the basis of the presence or absence of bands. Among cocoyam cultivars the genetic similarity ranged from 0.86 to 0.97 with a mean of 0.91. Cluster analysis identified two main clusters with some unexpected groupings. These data indicate that very little genetic variation exists within the accessions used in this study and that this Xanthosoma spp. collection is of limited value as a genetic resource.  相似文献

Bermudagrass (Cynodon dactylon L.) is an important warm-season turfgrass. Although it is widely distributed in China, studies on the genetic variation and relationship among the large-scale indigenous bermudagrass were relatively insufficient, especially at molecular level. The purpose of this study was to assess the molecular variation and relationship among one commercial cultivar ‘Tift3’ and 95 wild bermudagrass accessions collected from 11 provinces in China by ISSR marker technique. The results indicated that 29 ISSR primers generated a total of 248 bands among which 242 (97.6%) were polymorphic bands. The genetic similarity coefficients among accessions ranged from 0.51 to 0.97 with an average of 0.74. All accessions could be clustered into 11 groups with UPGMA method. Accessions from the same or adjacent regions generally were clustered into the same group or subgroups. A few accessions, however, were greatly different from the majority of all accessions. The results suggested that ISSR marker is an effective tool for the study of genetic variation in Chinese natural bermudagrass.  相似文献

利用开发的11对EST-SSR引物分析了33份猕猴桃种质资源的遗传多样性及其遗传关系。结果表明,11对EST-SSR引物在所有供试材料中均可扩增出清晰条带,其中有8对引物呈现多态性,多态性扩增率为72.7%,对33份种质材料的区分率达100%。8对多态性引物共检测到61个等位基因,每对引物可检测到的等位基因数为2-17,平均为7.6个。利用NTSYS-pc软件,以不加权成对算术平均法(UPGMA)对扩增结果进行聚类,谱系图显示,33份种质材料之间的相似系数在0.60~0.97,表明猕猴桃种质之间遗传关系相对来说不是很远。在相似系数0.73的水平上可将供试猕猴桃材料分为7个类群,其结果与传统的形态分类大体一致。从分子角度揭示了猕猴桃种质资源的遗传多态性及其遗传关系。可为猕猴桃种质改良提供理论依据。EST-SSR是非常有效和可靠的分子标记,可为猕猴桃分子育种及遗传连锁图构建奠定基础。  相似文献

Genetic relationship and variation of 29 accessions of teasle gourd (Momordica dioica Roxb.) and 1 accession of Momordica cochinchinensis Spreng. (wild relatives of teasle gourd) were examined by RAPD analysis using 44 dodecamer oligonucleotide primers. A total of 496 fragments were produced by 44 primers of which 95% bands were polymorphic. Using presence or absence of specific RAPD markers or combination of primers, 23 out of 30 accessions were identified. The genetic relatedness or genetic distance based on Nei and Li's genetic similarity varied from 0.86 to 0.65 with an average of 0.74 among 29 M. dioica accessions (when M. cochinchinensis excluded). In the phenetic dendrogram developed from cluster analysis using UPGMA method, M. cochinchinensis was out grouped as single accession, while others showing relatively weak grouping formed four groups. Clustering pattern did not demonstrate any relationship between geographical origin and genetic diversity. A DNA extraction method has been standardized. This is the first report of using RAPD techniques in teasle gourd. It was concluded that RAPD analysis is a useful tool for genotypic identification and estimation of genetic similarity in teasle gourd.  相似文献

Myrtle is an important plant species of the Mediterranean maquis, and is widely exploited for its aromatic properties. It is used in Italy for the production of a typical liqueur, for cut foliage and as an ornamental pot plant. We report the use of amplified fragment length polymorphism (AFLP) profiling to estimate genetic similarities within myrtle germplasm collected from six Italian regions, and from the Botanical Gardens of six other Mediterranean countries (including the outgroup Myrtus communis subsp. tarentina). Five AFLP primer combinations identified 122 polymorphic fragments analysing 92 individual samples, most of them (56%) were informative in discriminating among the populations. The AFLP patterns indicated that the majority of the variation occurs among rather than within populations (GST = 0.61). A neighbour-joining (NJ) tree separated the populations into two main branches: the first one grouped some of the Italian populations with those from Spain and Portugal; the second one included a Southern Italian subcluster together with samples from Greece, Israel, France and Croatia. A principal coordinate analysis supported the two major branches identified in the NJ analysis and showed the separation of Western and Eastern Mediterranean populations along the first axis. The Italian populations did not cluster in a single clade, but rather form distinct regional groups. The present analysis suggests that Italy represents a botanical transition zone between the Western and Eastern Mediterranean region in Myrtus communis.  相似文献

Few records are available about local Tunisian pear cultivars characterized by low chilling requirements and adaptation to dry conditions. In this work, seven SSRs derived from apple were successfully transferred to 25 local Tunisian pear genotypes and 6 common varieties of Pyrus communis cultivated in Europe. The 7 SSRs used amplified a total of 36 fragments. All the microsatellites except one seem to amplify more than one locus in some of the genotypes studied. Only 12 different fingerprinting patterns could be distinguished among the 25 Tunisian cultivars studied indicating a high number of synonymies. The mean expected and observed heterozygosities in the 25 Tunisian cultivars analyzed averaged 0.71 indicating a high level of genetic diversity among the local Tunisian pear germplasm. These markers will be useful to optimize the conservation of this highly threatened germplasm.  相似文献

Random amplified polymorphic DNA (RAPD) marker was used to assess genetic diversity and inter-specific relationships among nine taxa of Calliandra (Leguminosae: Mimosoideae) grown in Indian gardens. DNA from leaf sample was isolated and RAPD analysis was performed using 22 primers. The genetic similarities were analyzed from the dendrogram constructed by the RAPD data using a similarity index which supported the segregation of the nine taxa of the genus into two groups; the sect. Androcallis with seven taxa, viz. C. haematocephala, C. haematocephala var. alba, C. surinamensis, C. tweedii, C. tergemina var. emarginata and C. selloi and sect. Calliandra having two species namely, C. inermis and C. calothyrsus. The intra-generic classification and phylogeny inferred from molecular markers supported the traditional classification of the genus based on morphological characters at the level of sections and series except in case of C. selloi (C. brevipes) which did not show much genetic similarity with C. tweedii and C. surinamensis; all the three species being members of the sect. Genetic diversity is essential to crop improvement. However, lack of molecular markers prevents the understanding of genetic diversity in celery and related species. In this study, SRAP and SSR markers was firstly used to evaluate genetic variation in 68 accessions of Apium graveolens and related species. A total of 888 bands were generated by 40 primer combinations of SRAP and 32 bands were produced by eight SSR markers. Of the 920 bands, 95.1% were polymorphic between A. graveolens and related species, and 49.2% were polymorphic within A. graveolens. Cluster and structure analysis could distinguish local celery, celery, and related species. These results suggested that both SRAP and SSR technologies can be efficiently used to characterize genetic variation and analyze genetic relationship in celery and related species.  相似文献

This work investigated the fingerprinting and phenotyping of Citrus germplasm; species selected were of historical importance belonging to Citrus limonimedica Lush. and its supposed ancestors, along with some other species of the Citrus genus. An integrated approach based on the exploitation of nuclear DNA content, morphological traits and molecular markers, such as RAPD fingerprints and ITS-based SNPs, was employed. We studied a core collection of 54 distinct accessions, including 43 genotypes of the Citrus species (18 species or supposed species) and 11 genotypes of the Poncirus genus, which was used as the reference outgroup. Morphological trait analysis and statistical analysis of DNA content and markers were useful for reconstructing a Citrus phylogeny. In particular, our experiments aimed at estimating the genetic variation within and the genetic relatedness among C limon (L.) Burm., C. limonimedica and C. medica L. to shed light on the hybrid origin hypothesis of C. limonimedica. The results of the multidisciplinary analyses allowed us to confirm a remarkable differentiation between Poncirus and Citrus genera and to highlight a close relationship among the three investigated Citrus species but a distinct difference between these three species and other species in the Citrus genus. RAPD fingerprints and ITS polymorphisms enabled us to point out a variation gradient between lemon and citron, with C. limonimedica as a possible intermediate species. Some accessions of C. medica and C. limonimedica that deviate from such a trend suggest recurrent introgression and/or hybridisation with other species of Citrus.  相似文献
