The effect of four growing environments (two at Ames, IA; one at Clinton, IL; and one at Columbia, MO) on the thermal properties of starch from five exotic‐by‐adapted corn inbred lines (Chis37, Cuba34, Cuba38, Dk8, Dk10) and two control lines (B73 and Mo17) were studied using differential scanning calorimetry (DSC). The variations in thermal properties within environments were similar for the exotic‐by‐adapted lines and control lines. Missouri was the warmest environment and generally produced starch with the greatest gelatinization onset temperature (ToG), the narrowest range of gelatinization (RG), and the greatest enthalpy of gelatinization (ΔHG). Illinois was the coolest environment and generally resulted in starch with the lowest ToG, widest RG, and lowest ΔHG. These differences were attributed to higher temperatures in Missouri during grain‐filling months either increasing the amount of longer branches of amylopectin or perfecting amylopectin crystalline structure. The Ames 1 environment produced starch with thermal properties most similar to those of Illinois, whereas the Ames 2 environment produced starch with thermal properties most similar to those of Missouri. Ames 2, located near a river bottom where temperatures tend to be warmer, likely had temperatures most similar to those found in Missouri during grain filling.  相似文献

The changes in thermal properties of maize starches during five stages of kernel maturity, (12, 18, 24, 30, and 36 days after pollination [DAP]), from three mutant genotypes, amylose extender (ae), sugary-2 (su2), and waxy (wx) in an OH43 background, and the OH43 genotype were studied using differential scanning calorimetry (DSC). Within a genotype, DSC values of starches at 24, 30, and 36 DAP were similar to each other and often were significantly different (P < 0.05) from the values at 12 DAP, indicating possible differences in the fine structure of starch during endosperm development. For su2 starches, the gelatinization onset temperature (ToG) significantly decreased after 12 DAP and remained low throughout the study. The gelatinization range (RG) had a similar pattern. For wx starches, ToG at 18 DAP was significantly lower than at 12 DAP but tended to increase after 18 DAP. The RG increased significantly after 12 DAP and significantly decreased after 30 DAP. Thus, thermal properties of starches during early development were different from those of their mature counterparts, and differences among the mutant genotypes and the normal starch originated from the earliest endosperm development stage studied (12 DAP).  相似文献

The objectives of this research were to evaluate the intra‐ and interpopulation variability in gelatinization properties of starches from exotic corn lines and their derivatives when grown 1) during two successive years in the same location; and 2) in both temperate and tropical environments. Six novel exotic corn lines (two 100% exotic and four 25% exotic derived from a breeding cross developed by crossing an exotic hybrid with Corn Belt lines) were selected for this research because their starches have significantly different (and potentially useful) thermal properties from those found in starch from normal Corn Belt corn. The Sn (n = 3 for 25% exotic lines and n = 1 for 100% exotic lines) generations of the six exotic lines were self‐pollinated and grown in the winter nursery in Puerto Rico. Two successive generations (Sn+1 and Sn+2) of lines selected for low onset of gelatinization temperature were self‐pollinated and grown in the same environment near Ames, IA. To evaluate the effect of environment, the Sn+2 generation also was self‐pollinated and grown in the winter nursery in Puerto Rico. Thermal properties of starches from 10 single kernels from each line were analyzed by using differential scanning calorimetry (DSC) at a ratio of 4 mg of dry starch to 8 mg of distilled water. After subsequent generations, the differences in DSC gelatinization properties between selected kernels within each progeny line narrowed, suggesting increased homogeneity of starch structural properties within each line. Unusual thermal properties were fixed in some progeny lines. Environmental factors also affected the thermal properties of starch and a significant interaction between environment and genotype was observed. These results suggest that introgression of adapted germplasm with useful genes from exotic corn would increase the available genetic variability for starch functionality and allow the development of hybrids with important value‐added traits.  相似文献

以苏玉糯1号、苏玉糯5号和渝糯7号为材料,研究了拔节期追氮量(N 0、150和300 kg/hm2)对鲜食糯玉米粉糊化和热力学特性的影响。结果表明,随着拔节期追氮量的增加,峰值黏度和崩解值下降,糊化温度升高,而谷值黏度、终值黏度和回复值呈先降后升趋势; 热力学特征参数中,回生值、终值温度、糊化范围和峰值指数受拔节期追氮量影响较小。原样品热焓值和回生后样品热焓值均表现为随着拔节期追氮量的增加呈先降后升趋势。糊化和热力学特征值对拔节期追氮量的响应不同品种间存在差异。鲜食糯玉米粉的理化特性存在显著的基因型差异,峰值黏度以苏玉糯5号最高,苏玉糯1号最低; 峰值温度苏玉糯1号和苏玉糯5号无显著差异,但均高于渝糯7号; 热焓值、回生值、糊化范围和峰值指数不同品种间相对稳定。相关分析表明,峰值黏度与原样品热焓值和峰值指数呈显著正相关; 崩解值与糊化温度及转变温度,回生值与淀粉的终值黏度和回复值呈显著负相关。在本试验条件下,拔节期追氮总体上使糯玉米食用品质降低,其中以苏玉糯5号在不追氮处理下的糊化和热力学特性较优,即峰值黏度、崩解值和热焓值较高,回生值较低。  相似文献

以垦粘1号、苏玉糯1号和苏玉糯5号为材料,研究了拔节期追氮量(N 01、50和300 kg/hm2)对春播和秋播糯玉米淀粉胶凝和回生特性的影响,试验于扬州大学实验农牧场进行。结果看出,播期、品种和拔节期追氮量单因素及其互作对淀粉胶凝和回生主要特征值存在显著影响。糯玉米淀粉胶凝和回生特征值在拔节期追氮150 kg/hm2时和秋播条件下较优,即原淀粉具有较低的峰值温度、较高的热焓值,回生淀粉具有较低的回生值,且以垦粘1号表现较好。糯玉米淀粉胶凝和回生特征值之间存在一定的相关性。回生值分别与回生淀粉的热焓值、原淀粉的终值温度显著正相关,相关系数分别为0.82(P0.01)和0.47(P0.05);原淀粉的热焓值与峰值指数显著正相关,相关系数为0.53(P0.05),与原淀粉峰值温度、回生淀粉的终值温度显著负相关,相关系数分别为-0.53(P0.05)和-0.52(P0.05);回生淀粉的热焓值分别与回生淀粉起始温度、终值指数显著正相关,相关系数分别为0.46(P0.05)和0.66(P0.01)。综合考虑淀粉胶凝和回生特性在不同处理下的变化趋势,以秋播糯玉米淀粉在拔节期追N 150 kg/hm2处理下较优。  相似文献

Differential scanning calorimetry (DSC) was used to study the effect of sucrose on wheat starch glass transition, gelatinization, and retrogradation. As the ratio of sucrose to starch increased from 0.25:1 to 1:1, the glass transition temperature (Tg, Tg′) and ice melting enthalpy (ΔHice) of wheat starch‐sucrose mixtures (with total moistures of 40–60%) were decreased to a range of −7 to −20°C and increased to a range of 29.4 to 413.4 J/g of starch, respectively, in comparison with wheat starch with no sucrose. The Tg′ of the wheat starch‐sucrose mixtures was sensitive to the amount of added sucrose, and detection was possible only under conditions of excess total moisture of >40%. The peak temperature (Tm) and enthalpy value (ΔHG) for gelatinization of starch‐sucrose systems within the total moisture range of 40–60% were increased with increasing sucrose and were greater at lower total moisture levels. The Tg′ of the starch‐sucrose system increased during storage. In particular, the significant shift in Tg′ ranged between 15 and 18°C for a 1:1 starch‐sucrose system (total moisture 50%) after one week of storage at various temperatures (4, 32, and 40°C). At 40% total moisture, samples with sucrose stored at 4, 32, and 40°C for four weeks had higher retrogradation enthalpy (ΔH) values than a sample with no sucrose. At 50 and 60% total moisture, there were small increases in ΔH values at storage temperature of 4°C, whereas recrystallization of samples with sucrose stored at 32 and 40°C decreased. The peak temperature (Tp), peak width (δT), and enthalpy (ΔH) for the retrogradation endotherm of wheat starch‐sucrose systems (1:0.25, 1:0.5, and 1:1) at the same total moisture and storage temperature showed notable differences with the ratio of added sucrose. In addition, Tp increased at the higher storage temperature, while δT increased at the lower storage temperature. This suggests that the recrystallization of the wheat starch‐sucrose system at various storage temperatures can be interpreted in terms of δT and Tp.  相似文献

The starch from eight ethyl methanesulfonate (EMS) treated M4 families of the corn (Zea mays L.) inbred line B73 was analyzed using differential scanning calorimetry (DSC), a Rapid Visco Analyser (RVA), a texture analyzer (TA), and scanning electron microscopy (SEM) coupled with image analysis. The eight families were chosen from 144 families previously selected for having starch with unusual DSC parameters. Apparent amylose contents of the starch from the eight families generally were lower than that of the control. According to DSC, starches from mutagenized families tended to have lower onset temperature (T0) of gelatinization, enthalpy (ΔH) of gelatinization, and peak height index (PHI), but broader gelatinization range (R) than the B73 control. Their values for ΔH and percentage of retrograzdation (%R) were clustered around that of the control. Pasting properties from the RVA of the starches from the M4 families also were clustered around those of the control B73 starch, except for the setback values which were lower than B73 for M4 starches. Gel firmness values, as measured by TA, of all the M4 starches were generally lower than that of the B73 starch at storage treatments of one day at 25°C or seven days at 4°C. The stickiness of the gels of the M4 starches tended to be greater than that of B73 after seven days of storage at 4°C. These observations were consistent with the lower apparent amylose values for the M4 starches. SEM and image analysis data revealed no differences among the treatments in granule size and shape. Possibly, EMS treatment altered the genes, affecting internal structure of the starch granules. Starch from the mutagenized families likely had lower bonding forces among molecules and fewer long chains in the amylopectin molecules than did B73.  相似文献

A traditional waxy rice gel cake in Korea, Injulmi, was prepared with hydroxypropylated waxy rice and corn starches (molar substitutions 0.13 and 0.11, respectively), and the textural and retrogradation characteristics of the cake were compared with a conventional cake made of waxy rice flour. In the pasting viscogram, hydroxypropylated starches exhibited reduced pasting temperatures, but increased peak viscosities compared with the unmodified starches. Under differential scanning calorimetry, the Tg′ and ice melting enthalpy of the starch gel cakes were reduced by hydroxypropylation, which indicated that the modified starches had higher water‐holding capacity than the unmodified starches. The degree of retrogradation, as measured by the hardness of the gel cake and the melting enthalpy, was significantly reduced by hydroxypropylation and hydroxypropylated waxy rice starch was more effective in retarding the retrogradation than hydroxypropylated waxy corn starch  相似文献

The viscoelastic properties and molecular structure of the starch isolated from waxy (amylose-free) hexaploid wheat (WHW) (Triticum aestivum L.) were examined. WHW starch generally had lower gelatinization onset temperature, peak viscosity, and setback than the starch isolated from normal hexaploid wheat (NHW). Differential scanning calorimetry (DSC) showed that WHW starch had higher transition temperatures (To, Tp, and Tc) and enthalpy (ΔH) than NHW starch. However, when compared on the basis of amylopectin (AP) content, ΔH of WHW starch was almost statistically identical to that of its parental varieties. Typical A-type X-ray diffraction patterns were observed for the starches of WHW and its parental varieties. Somewhat higher crystallinity was indicated for WHW starch. WHW starch was also characterized by having greater retrogradation resistance. The high-performance size-exclusion chromatography (HPSEC) of amylopectin showed that each amylopectin yielded two fractions after debranching. Although WHW amylopectin had somewhat long B chains, little difference was observed in the ratio of Fr.III/ Fr.II between WHW and its parental varieties.  相似文献

The differences in pasting properties involving gelatinization and retrogradation of rice starches from IR24 and Sinandomeng cultivars during heating‐cooling processes were investigated using a Rapid Visco Analyser (RVA)and a dynamic rheometer. The results were discussed in relation to the molecular structure, actual amylose content (AC), and concentration of the starches. Generally, both starches possessed a comparable AC (≈11 wt%), amylose average chain length (CL), iodine absorption properties, and dynamic rheological parameters on heating to 95°C at 10 wt% and on cooling to 10°C at higher concentrations. In contrast to Sinandomeng, IR24 amylose had a greater proportion of high molecular weight species and number‐average degree of polymerization (DPn). IR24 amylopectin possessed a lower DPn and greater CL, exterior CL (ECL), and interior CL (ICL). Comparing the results of RVA analysis and dynamic rheology, the gelatinization properties and higher retrogradation tendencies of IR24 starch can be related to the structural properties and depend on starch concentration. In addition, the exponent n of starch concentration for storage moduli at 25°C (G25Cn) increased linearly with increasing AC.  相似文献

Semolina from four durum wheat genotypes (cvs. Ben, Munich, Rugby, and Vic) were processed into spaghetti that was dried by low (LT), high (HT), and ultrahigh (UHT) temperature drying cycles. Starch was isolated from dried pasta and unprocessed wheat and semolina references. Pasta-drying cycles had no significant effect on the amylose content of starches. Significant increases in enzyme-resistant starch were observed in HT- and UHT-dried pasta (2.27 and 2.51%, respectively) compared with LT-dried pasta (1.68%). Differential scanning calorimetry (DSC) gelatinization characteristics of pasta starches showed a significantly narrow range (Tr), but no changes in onset and peak temperatures (To and Tp, respectively) and gelatinization enthalpy (ΔH1) were observed. When compared with unprocessed reference samples (wheat and semolina), all pasta starches shifted to higher gelatinization To and Tp, with narrow Tr and no changes in δH1. The second endothermic DSC peak indicated no increase in amylose-lipid complexation (δH2) due to drying cycle. Starches isolated from LT and HT pasta exhibited lower peak viscosities than those from UHT-dried pasta. Genotypes Ben and Rugby demonstrated higher pasting temperature and lower peak and breakdown viscosities than Vic and Munich.  相似文献

Although pulsed NMR (PNMR) has been used for qualitative study of starch retrogradation in selected systems, validation is necessary for its application to new systems. PNMR was used to analyze the retrogradation of rice starches in purified form, in rice flour, and in cooked rice grains. The standard curves between the relative solid content (S′, %) by PNMR and the percentage of gelatinized starch (GS, %) were determined for common rice flour, common rice starch, and waxy rice starch at different moisture contents. The coefficients of linear regression for these curves (R2) were all >0.997. Starches with different amylose contents were tested for S′ values at the stages of freshly gelatinized, retrograded (4°C, 18 days), and reheated (90°C, 20 min). The S′ of reheated starch (S′reheat) was similar to the S′ of freshly gelatinized starch (S′0), so we concluded that the increase in S′ during storage corresponded to amylopectin retrogradation. The effect of moisture content on retrogradation of rice starch, rice flour, and cooked rice grains was studied by PNMR, and the data were interpreted using the Avami equation. Decreasing the moisture content increased the rate of retrogradation and led to a higher parameter k and a lower parameter n. For moisture content in the range studied, PNMR can be used to follow amylopectin retrogradation of different rice starch systems.  相似文献

Processing conditions similar to traditional nixtamalization are now used by the industry in the production of dry maize flours (DMF). The objective of this investigation was to evaluate the effect of industrial nixtamalization on maize starch. Thus, dent maize grains were sampled from storage silos and the starch isolated (S). From the same batch of maize, DMF was obtained and the starch isolated (S‐DMF). The amylose content in the starches was quite similar (21.5–23.4%) and characteristic of a dent maize. However, nixtamalization increased the calcium content in S‐DMF. The starches investigated exhibited the typical A‐type diffraction pattern after 40 days of storage at 11–84.1% rh. However, the differential scanning calorimetry (DSC) results showed that annealing of maize starch occurred during storage at 30°C. On the other hand, industrial nixtamalization has both a melting and annealing effect on maize starch. Thus, the operative glass transition temperature (Tg), and the DSC parameters that define starch gelatinization (Tp and ΔH) showed that the proportion between crystalline and amorphous regions within the starch granule and the extent of physical damage to starch were modified by nixtamalization. As an example, Tg for S was between 60 and 62.5°C, while the S‐DMF had a Tg of 45–55°C for damaged starch and 65–70°C for annealed starch. Additionally, the extraction of the nonconstitutive starch lipids provided starches with more consistent thermal properties, particularly in the behavior of gelatinization at different water content. This last observation might have important implications in the consistency of starch physicochemical properties and, consequently, in the quality of maize products such as tortillas.  相似文献

Physicochemical properties of starch of three common (Fagopyrum esculentum) and three tartary (F. tataricum) buckwheat varieties from Shanxi Province, China, were compared. Starch color, especially b*, differed greatly between tartary (7.99–9.57) and common (1.97–2.42) buckwheat, indicating that removal of yellow pigments from tartary buckwheat flour may be problematic during starch isolation. Starch swelling volume in water of reference wheat starch (2.8% solids and 92.5°C) was 20.1 mL; for the three common buckwheat starches it was 27.4–28.0 mL; and for the three tartary buckwheat starches it was 26.5–30.8 mL. Peak gelatinization temperature (Tp) in water was 63.7°C for wheat starch, 66.3–68.8°C for common buckwheat and 68.8–70.8°C for tartary buckwheat. Tp of all samples was similarly delayed (by 4.0–4.8°C) by 1% NaCl. Enthalpy of gelatinization (ΔH) was higher for all six buckwheat starches than it was for wheat starch. However, one common buckwheat sample had significantly lower ΔH than the others. Starch pasting profiles, measured by a Rapid Visco-Analyzer, were characteristic and similar for all six buckwheat starches, and very different from the reference wheat starch. A comparison of pasting characteristics of common and tartary buckwheat starches to wheat starch indicated similar peak viscosity, higher hot paste viscosity, higher cool paste viscosity, smaller effect of NaCl on peak viscosity, and higher resistance to shear thinning. Texture profile analysis of starch gels showed significantly greater hardness for all buckwheat samples when compared to wheat starch.  相似文献

Structural characteristics of resistant starch (RS) were investigated. Tuber starches, hydrolyzed with 1N HCl at 35°C for 8 hr followed by autoclaving‐cooling treatment, were heated at 100°C for 16 hr after adjusting the moisture content to 20 or 30%. RS content of the tuber starches ranged from 5.4 to 22.7% depending on the source and type of treatment. Gelatinization parameters of RS isolated from partially acid‐hydrolyzed starch with autoclaving‐cooling followed by heat‐moisture treatment (HMT) showed higher enthalpy (ΔH) values and lower peak temperature (Tp) compared with non‐acid‐hydrolyzed RS. R values, the difference between completion and initial temperatures, and ΔH of RS increased by HMT. The X‐ray diffraction patterns of potato and sweet potato RS isolated from partially acid‐hydrolyzed starch with autoclaving‐cooling showed distinct sharp peaks at 15, 25, 27, and 28°, which decreased by HMT.  相似文献

Gelatinization temperature of rice starch plays an important role in grain quality. We mapped eight loci responsible for the gelatinization parameters of rice: onset temperature (To), peak temperature (Tp), conclusion temperature (Tc) and enthalpy (ΔH) using a population of 242 recombinant inbred lines (RIL). Eight quantitative trait loci (QTL) were mapped onto the chromosome regions where starch‐synthesis related genes are located. The Wx gene (granule‐bound starch synthase [GBSS]), and the region of chromosome 11 played major roles in gelatinization trait determination; and other loci were starch‐branching enzyme (SBE) genes that modify the two major loci by epistasis. High‐level interactions were also detected indicating a trans mode of action of the genes. The genetic basis for these parameters was consistent with the relationship between the thermal properties and the structure of the starch granule. This information can be helpful for the improvement of starch‐producing crops, including the improvement of rice grain quality, both for food and industrial uses.  相似文献

The rheological and thermal properties of aged starch gels (15:85 starch-water) from three waxy maize genotypes (wx, wx sh1, and du wx) during storage (4°C for up to 25 days) were studied. After storage, changes of storage modulus (G′) and phase angle (δ) of the gels as a function of temperature were measured using oscillatory rheometry. For the du wx samples, G′ at 25°C increased rapidly during the first four days of storage at 4°C, compared to the gradual increases over the 25-day storage period for the wx and wx sh1 samples. A peak in G′ at 45°C was observed during heating for the du wx samples after 10 days of storage and for the wx sample stored for 25 days. The G′ peak may have been due to syneresis in the gels. Retrogradation of amylopectin of the aged starch samples was examined using differential scanning calorimetry. The du wx starch had greater retrogradation enthalpies than the other two samples (which showed similar retrogradation behavior) throughout the storage. The retrogradation enthalpy of the du wx samples increased rapidly during the first seven days, followed by a slower increase through the rest of storage. For the wx and wx sh1 samples, no endotherm was observed during the first four days of storage, after which the enthalpy increased steadily as a function of storage time. Addition of sucrose delayed the formation of gel networks for all three starches. The greater tendency for gelling and retrogradation of the du wx starch might be attributed to the greater proportion of DP20–30 chains of the amylopectin.  相似文献

The effects of amylose content and other starch properties on concentrated starch gel properties were evaluated using 10 wheat cultivars with different amylose content. Starches were isolated from grains of two waxy and eight nonwaxy wheat lines. The amylose content of waxy wheat lines was 1.4–1.7% and that of nonwaxy lines was 18.5–28.6%. Starch gels were prepared from a concentrated starch suspension (30 and 40%). Gelatinized starch was cooled and stored at 5°C for 1, 8, 16, 24, and 48 hr. The rheological properties of starch gels were studied by measuring dynamic viscoelasticity with parallel plate geometry. The low‐amylose starch showed a significantly lower storage shear modulus (G′) than starches with higher amylose content during storage. Waxy starch gel had a higher frequency dependence of G′ and properties clearly different from nonwaxy starches. In 40% starch gels, the starch with lower amylose showed a faster increase in G′ during 48 hr of storage, and waxy starch showed an extremely steep increase in G′. The amylose content and concentration of starch suspension markedly affected starch gel properties.  相似文献

The increases in storage modulus (G′), retrogradation enthalpy change (ΔH) and ΔH‐related Avrami kinetic parameters of gelatinized rice starch dispersions at 25% (w/w) were investigated with respect to storage period, amylose content (AC), and molecular properties. Three high‐AC and five low‐AC rice cultivars were compared for understanding the multiple influences of AC and molecular properties involved. After refining the results of correlation analysis, the G′ of just‐cooled samples changed positively, mainly with AC and additionally with the average chain length of amylose (CLAM) and the weight ratio of extra‐long plus long chains to short chains of amylopectin (AP) (rAPchain). The developed ΔH on short‐term storage (10 days) elevated with increasing AC and CLAM and decreasing degree of polymerization of AP (DPAP), but after long‐term aging for one to three months with increasing rAPchain, especially for the low‐AC cultivars examined. Greater Avrami rate constants for retrogradation could be attributed to the combination of a lower DPAP and rAPchain or AP chain length and a greater CLAM. The polynomials using these critical factors to describe the retrogradation parameters were elucidated and could account for 85–99.6% of data deviations.  相似文献

Retrogradation of du wx and su2 wx starches after different gelatinization heat treatments was studied by differential scanning calorimetry. Suspensions of 30% (w/w) starch were initially heated to final temperatures of 55–180°C. Gelatinized starch was cooled and stored at 4°C. Starch retrogradation in the storage period was influenced by initial heat treatments. Retrogradation of du wx starch was rapid: when initially heated to 80–105°C, retrogradation enthalpy was ≈10 J/g after one day at 4°C. The retrogradation enthalpy was ≈15 J/g after 22 days of storage, and reached a maximum of 16.2 J/g after 40 days of storage. For du wx starch, application of the Avrami equation to increases in retrogradation enthalpy suggests retrogradation kinetics vary with initial heating temperature. Furthermore, starch retrogradation may not fit simple Avrami theory for initial heating ≤140°C. Retrogradation of su2 wx starch was slow. After 30 days of storage at 4°C, the maximum retrogradation enthalpy for all initial heating temperatures tested was 7.0 J/g, for the initial heating to 80°C. This work indicates that gelatinization heat treatment in these starches is an important factor in amylopectin retrogradation, and that the effect of initial heat treatment varies according to the genotype.  相似文献
