利用生物膜的高效截留原理,设计了生物强化反应器,在装有弹性纤维载体的反应器内接种具有硝化反硝化作用的复合菌群,经6周左右的人工挂膜后,弹性纤维载体表面出现了肉眼可见的生物膜。试验以循环养殖废水为研究对象,探讨了不同水力停留时间(HRT)和温度(WT)对生物强化反应器净化水质效果的影响。结果显示,HRT和WT对生物强化反应器净化养殖废水效果的影响显著(P<0.05)。对HRT和WT各处理水平下的CODMn、TN和NH 4-N去除率进行差异显著性分析,并综合考虑反应器的运行时间、费用和操作等因素,选取HRT=18 h、WT=30 ℃作为反应器的最佳运行条件,在此条件下反应器稳定运行3周左右。试验期间,CODMn、TN及NH 4-N的平均去除率分别达44.18%、51.31%及82.08%。此外,还对反应器内载体表面的生物膜进行了可培养微生物数量动态监测。强化反应器在正式运行的25 d内,其生物膜上的可培养微生物数量不断增多,氨氧化细菌数量由35.91?104 CFU/mg增长到89.43?104 CFU/mg,硝化细菌数量由25.75?104 CFU/mg增长到61.65?104 CFU/mg,反硝化细菌数量由14.23?104 CFU/mg增长到100.95?104 CFU/mg;氨氧化细菌和硝化细菌在与氮素循环相关的可培养细菌中占据着主导地位(59.95%~81.25%)。脱氮细菌不断地吸附于载体表面,并成功地定殖和繁殖使得生物膜不断成熟和稳定(TN和NH 4-N去除率分别达51.31%及82.08%)。结果表明,生物强化反应器对循环养殖废水中CODMn、TN和NH 4-N的去除效果明显。  相似文献

将生物陶环和珊瑚石两种滤料分别采用半浸没式和全浸没式实验处理养殖废水。结果显示,在2种过滤模式下,生物陶环、珊瑚石生物滤料的生物膜成熟时间分别为8 d和10 d。经过20 d的处理,4个实验组NH4+-N, NO2--N, COD的浓度均显著低于对照组,相比于对照组NH4+-N浓度降低51%以上, COD降低40%以上;半浸没过滤模式的实验组对NH4-N、 NO2--N的去除率均显著高于全浸没实验组;全浸没实验组对COD的去除率显著高于半浸没实验组。半浸没式过滤生物陶环实验组表面AOB数量最高,达4.68×102 cfu/g,全浸没实验组HB数量均显著高于半浸没实验组。结果表明所用4种处理方式处理养殖废水均有较好效果。  相似文献

利用自制的硝化细菌菌剂促进移动床生物膜反应器(Moving bed biofilm reactor,MBBR)的挂膜启动,分析不同载体氨氮负荷、碳氮比条件下反应器运行状况,并进一步进行了实验室模拟循环水养殖草金鱼实验。结果显示,利用自制硝化菌剂能够完成整个移动床反应器的启动过程,在接种15 d后使循环出水氨氮稳定在1 mg/L以下。单位体积载体氨氮负荷实验表明,MBBR能够在100 mg TAN/(L填料·d)条件下,使出水满足一般水产养殖水质要求(氨氮0.5 mg/L,亚硝氮0.1 mg/L)。进水碳氮比在1以内时MBBR能够稳定高效运行。在实验室模拟循环水养殖过程中,经菌剂强化的MBBR能维持循环出水氨氮低于0.5 mg/L,亚硝氮低于0.05 mg/L。  相似文献

采用移动床生物膜反应器(MBBR)处理低浓度氨氮养殖废水,在不同水力停留时间(HRT)和不同曝气条件下,分析MBBR处理人工模拟的低浓度氨氮(2 mg/L左右)养殖废水的进出水氨氮、亚硝酸盐氮和硝酸盐氮的浓度变化,探讨HRT和曝气量对MBBR处理低浓度氨氮养殖废水的影响,并以实际鲟鱼养殖废水(氨氮浓度0.5~1.5 mg/L)和其他研究成果进行验证和比较.结果显示:MBBR的最优HRT为6~8 min,最优曝气量为180 L/h,相应的氨氮去除率为70% ~ 75%,氨氮去除负荷为560~700 g/(m3.d),填料生物膜厚度为26~38 μm;膜表层结构多样,物种丰富,膜生长良好.该反应器对处理低浓度氨氮养殖废水具有的高效能力.  相似文献

为解决循环水养殖生产中硝酸盐积累问题,以凡纳滨对虾(Litopenaeus vannamei)循环水养殖系统为例(养殖负荷5 000 kg),设计了一种移动床生物膜反应器(MBBR)—藻类反应器联用系统作为生物处理单元用以消除养殖水体中无机氮。根据物质平衡原理确定生物处理单元进水流量为453.3m3/h, MBBR尺寸为4 m×4 m×2.8 m(4个),藻类反应器尺寸为6 m×6 m×2.8 m(4个),MBBR水力停留时间(HRT)为0.3 h,藻类反应器HRT为0.36 h,系统新水更新量为0.97m3/h,循环次数为22次/d,系统硝酸盐氮可维持在70 mg/L以下的安全质量浓度范围内。构建中试系统进行验证,发现MBBR-藻类反应器联用相比MBBR,总氮的去除率由3.9%提高至42.8%;藻类可通过光合作用吸收水体磷酸盐,联用系统对总磷的去除率高达66.8%,藻类特定生长率达到3.86~10.35%/d,联用系统有效缩短了去除同量氮磷所需水力停留时间。本研究可为循环水养殖系统中硝酸盐原位消除技术及生物处理单元的建立提供理论参考,助推水...  相似文献

循环水养殖系统启动运行前往往需要经过一段时间的生物膜预培养,使生物膜达到成熟稳定,从而保证系统的水质净化功能。本研究通过养殖试验,研究了生物滤器负荷挂膜的技术方法,以期实现生物膜的快速成熟和系统的快速启动。为此,构建了6组循环水系统组成的养殖车间,建成后立即投入试验生产。试验为期120 d,养殖种类为红鳍东方鲀,初始放养平均体重(632.5±2.26)g。期间,红鳍东方鲀平均增重29.91%,养殖成活率98.7%,养殖密度由(19.34±1.89)kg/m3增加到(32.17±3.40)kg/m3,投饵率由0.2%增加到0.5%–0.7%,每日换水量由50%逐渐减至10%。结果表明,在生物膜的生长期,通过对投饵量及新水补充量的有效调节,可以把养殖水体中的氨氮和亚硝氮浓度控制在安全范围以内,以保证养殖鱼类的生长。生物膜在50天左右达到完全成熟,此后便可依靠生物膜的净化作用将氨氮浓度控制在0.5?1.2 mg/L、亚硝氮浓度控制在0.2?0.5 mg/L、pH值控制在6.5–7.5、COD值低于4 mg/L、细菌总数控制在800–2100 cell/ml的安全范围内。利用生物滤器负荷挂膜技术,在合理调控水质指标的条件下,循环水养殖系统建成后可以立即投入生产,实现生物滤器挂膜与养殖生产的同步进行。  相似文献

人工湿地冬季净化减排水产养殖废水效果   总被引:2,自引:0,他引:2  
根据我国池塘养殖废水排放特点,研究了冬季人工湿地集中处理池塘养殖废水的效果。结果显示,在0.22 m/d水力负荷下,人工湿地对池塘养殖废水中总氮(TN)、总磷(TP)、化学需氧量(COD)和悬浮物(TSS)的去除率分别达到35.0%,38.2%,34.7%,91.6%;水力负荷上升至0.36 m/d后,人工湿地对TN和COD的去除率显著下降为20.0%和27.9%(P<0.05),TP和TSS去除率变化差异不显著(P>0.05),分别为35.7%和和93.2%。经人工湿地处理后出水TN浓度低于1.5 mg/L,TP浓度低于0.2 mg/L,COD浓度低于8 mg/L,TSS浓度低于5 mg/L,均满足水产养殖废水排放要求,表明应用人工湿地技术能有效实现池塘养殖废水的减排。另外,冬季低温使人工湿地对池塘养殖废水中TN和COD的去除率与夏季相比显著降低(P<0.05),但通过对人工湿地进行曝氧能够显著提高其对TN和COD的去除率(P<0.05)。  相似文献

为缩短新建海水循环水养殖系统生物过滤器硝化功能构建时间,通过预培养生物膜的方法获得已建立完全硝化功能的2.5 m3过滤材料,将其置于新建系统的生物过滤器中进行硝化细菌接种,系统放养美国红鱼(Sciaenops ocellatusL innaeus)。结果表明:经12 d系统的硝化功能成熟,养殖池氨态氮维持在0.50 mg/L左右,亚硝态氮维持在0.10 mg/L以下,系统运行34 d,硝态氮上升至63.58 mg/L。18~48 d系统稳定运行阶段,养殖池出水口氨态氮平均值0.44 mg/L,进水口氨态氮平均值0.05 mg/L,一次性平均去除率88.64%。系统的日换水量1%。养殖1个月,美国红鱼成活率90.91%,养殖密度达28.65 kg/m3水体。预培养生物膜法有效缩短了海水生物过滤器硝化功能构建的时间,具有操作简单、节约时间、系统运行稳定的特点,将使内陆地区开展海水鱼养殖变为可能。  相似文献

水力条件对反应器内生物膜的生长及流态形式起着决定性作用。实验分别用0.15,0.25,0.35 m/s的水流流速对内径为44 mm的管状生物膜反应器进行水力冲击,观察不同生物滤料的挂膜情况,并利用计算流体力学软件对导流式移动床生物膜反应器流态进行数值模拟。结果显示,在低流速的水力冲击下,生物滤料的挂膜效果最好,平均厚度约为70μm,且不同结构生物滤料的挂膜情况无明显差异;反应器的模拟曝气速度为0.6 m/s时,其内部的综合流动及挂膜效果最佳。因此可知,生物膜的生长情况与同种材质生物滤料的结构形状无关,但与滤料所处的水力情况有关,膜厚度随着水流速度的增大而减小;移动床生物膜反应器的曝气量大小及结构形状是影响其流态的重要因素。本研究可以为此类反应器的设计与高效运行提供基础数据。  相似文献

生物过滤技术是循环水养殖系统中很关键的水处理技术,选择合适的滤料在很大程度上决定着生物过滤效果的好坏和循环水养殖系统能否正常运行。本次试验对一种新型悬浮式滤料在循环水高密度养殖系统中的水处理能力进行了研究,结果表明:(1)该滤料的挂膜性能好,挂膜时间短,且生物膜状态稳定;(2)该滤料的生物过滤能力好,具有较高的氨氮去除率和氨氮负荷,最高分别为57.5%和182.9 g/(m~3·d);其亚硝酸盐去除率平均为45.4%。  相似文献

Previously, we reported 10 PEGE types of 85 tilapia Streptococcus agalactiae(GBS), which shifted from Streptococcus iniae in China, by using PEGE method. Presently, larger and more representative tilapia GBS were isolated, for the ?rst time in China, to characterize their serotypes and genetic diversities more precisely than had done before. 168 GBS strains were distributed in ?ve provinces of China, in which Guangdong, Guangxi and Hainan were the major ones, holding36.9%(62/168), 37.5%(63/168) and 19.6%(33/168), respectively. Serotypes, Ia, Ib and III, were observed in these strains and the most predominant one was Ia(95.2%), which mainly distributed in Guangdong, Guangxi and Hainan. Ia initially occurred in 2009, it shoot up to 32.1% in 2010,but decreased to 16.1% in 2011 before went up to 45.2% in 2012. Ib sporadically occurred during2007–2011, III onlyoccurred in 2012. 14 different PFGE types, including 4 new types(N, O,P and Q), were observed, in which B, D, F and G were the predominant types, holding 83.9%(141/168) of the total GBS strains. Ia corresponded to 11 PFGE types(A–H, N–P), in which type D predominated(51%). Ib represented 3 genotypes(I, J and Q) and III harbored only 2genotypes(N and F). Type N and Fsynchronously presented in Ia and III. In summary, the genetic diversity of tilapia GBS varied by serotypes and changed with geographical locations and years.Although Iastillpredominated, new rareserotypeIII alreadyoccurred in China.  相似文献

Interaction between growth and reproduction occurs in many vertebrates and is particularly obvious at certain stages of the life cycle in fish. Endocrine interactions between the gonadotropic axis and the somatotropic axis are described, the potential role of GH being emphasised. A comparative analysis of these phenomena in mammals, amphibians and fish, suggests a specific role of GH in the physiology of puberty, gametogenesis and fertility. It also shows the original contribution made by studies on the fish model in this field of investigations.  相似文献

Two gonadotropins (GtH; Qa and Qb) were purified by gel filtration and ion exchange chromatography from the pituitaries of Indian walking catfish (Clarias batrachus). The presence of GtH during purification was assessed by in vitro oocyte maturation and in vivo steroidogenic activity, and their identities were determined by elution profiles, molecular weight, biological activities and yield. The molecular weights of Qa and Qb were 37 and 42 kDa, respectively, and composed of distinct subunits (Qa: 20 and 14 kDa and Qb: 26 and 18 kDa). Polyclonal antibodies raised against Qa immunostained Qa, Qb and pituitary GtH cells. A competitive Qa‐ELISA was developed whose sensitivity was 6.25 ng mL?1 (1.25 ng well?1) with intra‐ (3.5%) and inter‐ (12.4%) assay coefficients of variation. Displacement curves parallel to the standard were obtained with plasma and pituitary extracts of catfish, Qb and carp GtHII. The assay was validated by measuring the plasma Qa levels after LHRH treatment and in relation to ovarian growth in the female catfish during different reproductive phases. Based on the results, Qa and Qb corresponded to fish LH and FSH respectively. The findings will increase the knowledge of the mechanisms controlling fish reproduction and identification of sensitive phases in fish in captivity for hormonal manipulation.  相似文献

Tetrahymena is a protozoan parasite, which infects guppy, Poecilia reticulata Peters, and causes substantial economical losses in commercial farms worldwide. Studies of guppy infected by Tetrahymena require standardized infection protocols. The LD50 for Tetrahymena infection of guppies by intraperitoneal (IP) injection was calibrated, and the level obtained was 946 parasites per fish. Guppy infection with Tetrahymena by immersion, imitating the natural route of infection via the integument, was studied under normal or stress conditions. Exposure to cold and netting (CNI) and to cold only (CI) followed by immersion exposure to 10 000 Tetrahymena per mL resulted in 22.5% and 19.2% mortality, respectively, as compared to 14.2% and 10% in groups that were netted only (NI) or non‐stressed (I). Histopathology revealed that immersion infection resulted in a systemic infection. Lysozyme levels, measured 3 weeks after infection, were significantly higher in the CNI group (288 μg per mg protein) compared with CI‐, NI‐ and I‐treated groups (94.5, 64 and 62.3 μg mg?1, respectively). There was no evident parasite immobilization activity in body homogenates, suggesting no development of acquired immunity. Re‐infection by IP injection revealed no increase in protection in any of the treatment groups, mortality range of 56.3–75%, higher than in the non‐exposed control (40.6% mortality).  相似文献

There has been growing concern about the overuse of antibiotics in the ornamental fish industry and its possible effect on the increasing drug resistance in both commensal and pathogenic organisms in these fish. The aim of this study was to carry out an assessment of the diversity of bacteria, including pathogens, in ornamental fish species imported into North America and to assess their antibiotic resistance. Kidney samples were collected from 32 freshwater ornamental fish of various species, which arrived to an importing facility in Portland, Oregon from Colombia, Singapore and Florida. Sixty‐four unique bacterial colonies were isolated and identified by PCR using bacterial 16S primers and DNA sequencing. Multiple isolates were identified as bacteria with potential to cause disease in both fish and humans. The antibiotic resistance profile of each isolate was performed for nine different antibiotics. Among them, cefotaxime (16% resistance among isolates) was the antibiotic associated with more activity, while the least active was tetracycline (77% resistant). Knowing information about the diversity of bacteria in imported ornamental fish, as well as the resistance profiles for the bacteria will be useful in more effectively treating clinical infected fish, and also potential zoonoses in the future.  相似文献

This study investigated the efficiency of iodophor disinfection (135 ppm active iodine for 15–30 min) of non‐hardened Salmo trutta eggs against different groups of bacteria and against fungus. Egg samples were taken from non‐disinfected and from disinfected eggs, microorganisms were cultured on specific nutrient media and their mass was measured by turbidimetric methods. Bacteria and fungus mass of non‐hardened eggs could be reduced but not eliminated by iodophor disinfection with 135 ppm active iodine for 15 min. The extent of reduction was 47–65% (Experiment 1). The efficiency of disinfection increased with disinfection time as the reduction in bacteria and fungus mass was 40–55% after 15 min and 58–74% after 30 min (Experiment 2). Disinfection efficiency of iodophor solution diluted in water (reduction 49–57%) and of iodophor solution diluted in sodium chloride solution iso‐osmolar to the oocytes (reduction 52–61%) was similar (Experiment 3). The reduction in bacteria and fungus mass was persistent as it was 39–72% lower in embryos deriving from disinfected eggs than in embryos deriving from non‐disinfected ones (Experiment 4). In conclusion, the tested disinfection method is inadequate to eliminate pathogens completely but it could positively influence immune defence of eggs and embryos.  相似文献

The endemic, anadromous cyprinidChalcalburnus tarichi is the only fish species known to occur in alkaline Lake Van (Eastern Anatolia, Turkey). EightC. tarichi were maintained individually in Lake Van water (17 – 19°C; pH 9.8; 153 mEq·I–1 total alkalinity; 22 total salinity) and tank water samples analyzed for 24 h in 2 to 4 h intervals. At zero time, < 1µM ammonia was present and urea was undetectable in the tank water; at 24 h, total ammonia and urea made up 114±32 and 35±25µM, respectively. Over the experimental period, ammonia-N and urea-N excretion averaged 1041±494 and 607±169moles·kg–1 fish·h–1, respectively. The extent of urea excretion was highly variable between specimens. Uric acid excretion was not detectable.Urea was present at high concentrations in all tissues and plasma (25 – 35moles·g–1·ml–1) of freshly caughtC. tarichi; total ammonia content of the tissues was by a factor of 1.9 (liver) to 3.0 (brain) lower. High arginase activity (2.4±0.2 U·min–1·g–1) was detected in the liver ofC. tarichi but ornithine carbamoylphosphate transferase, a key enzyme of the ornithine-urea-cycle, was absent. Ureagenesis is likely through degradation of arginine and/or uricolysis. High glutamine synthetase activity (11±0.6 U·min–1·g–1) and low ammonia content in brain suggest that, like other teleosts,C. tarichi has an efficient ammonia detoxification in the brain, but in no other tissue.Nitrogenous waste excretion at alkaline pH is discussed. The ability ofC. tarichi to excrete high levels of ammonia at extremely alkaline pH is unique among teleosts studied so far. The mechanism of ammonia excretion under Lake Van conditions remains to be elucidated.  相似文献

The indiscriminate use of antibiotics and chemicals in shrimp hatcheries has led to biomagnification and that in turn could lead to rejection of a whole consignment. The application of the bioencapsulation technique as a tool for curative treatment in shrimp larvae was investigated. Herbs having antibacterial properties such as Solanum trilobatum, Andrographis paniculata and Psoralea corylifolia (methanolic extracts) were bioencapsulated in Artemia and fed to Penaeus monodon post larvae PL 1–25. The post larvae were reared in a medium inoculated with pathogenic bacteria such as Pseudomonas aeruginosa, Staphylococcus aureus, Salmonella typhi and Vibrio sp. Post larvae reared in the non-inoculated water and fed with non-enriched Artemia exhibited 90% survival, highest specific growth rate (12.43%) and reduced bacterial load. P. monodon reared in the bacterial inoculated water and fed with the non-enriched Artemia exhibited the lowest survival (10–30%), specific growth rate (8.42–9.1%) and increased bacterial load (2.86 × 103 to 3.76 × 105 cfu/g). The methanolic extracts of the herbs helped to increase survival and specific growth rate and reduced bacterial load in the P. monodon culture system. Among the three herbal extracts, P. corylifolia enriched Artemia fed post larvae showed the tendency to higher survival (>50%), growth rate (11.5 averaged) and low bacterial load (1.12 × 105 cfu/g).
