The root hemiparasite Striga hermonthica causes very significant yield loss in its dryland staple cereal host, Sorghum bicolor. Striga‐resistant sorghum cultivars could be an important part of integrated S. hermonthica control. For effective resistance breeding, knowledge about the diversity of the parasite is essential. This study aimed (i) to determine the genetic diversity within and between seven S. hermonthica populations from East and West Africa using 15 microsatellite markers and (ii) to assess the virulence and host–parasite interactions of these Striga populations grown on 16 diverse sorghum genotypes in a glasshouse trial. Most of the genetic variance (91%) assessed with microsatellite markers occurred within S. hermonthica populations. Only a small portion (8%) occurred between regions of origin of the populations. A positive correlation (R2 = 0.14) between pairwise geographic and genetic distances reflected the slightly increasing differentiation of S. hermonthica populations with increasing geographic distance. East African S. hermonthica populations, especially those from Sudan, had significantly greater average infestation success across all sorghum genotypes than West African populations. Some specific host–parasite interaction effects were observed. The high genetic variation among individuals of each S. hermonthica population underlines the high potential adaptability to different hosts and changing environments. This points to the need to manage sorghum resistance alleles in space and time and to employ resistant varieties as part of integrated S. hermonthica control, so as to hinder the parasite overcoming resistance.  相似文献

Striga hermonthica is a parasitic weed that poses a serious threat to the production of economically important cereals in sub‐Saharan Africa. The existence of genetic diversity within and between S. hermonthica populations presents a challenge to the successful development and deployment of effective control technologies against this parasitic weed. Understanding the extent of diversity between S. hermonthica populations will facilitate the design and deployment of effective control technologies against the parasite. In the present study, S. hermonthica plants collected from different locations and host crops in Kenya and Nigeria were genotyped using single nucleotide polymorphisms. Statistically significant genetic differentiation (FST = 0.15, = 0.001) was uncovered between populations collected from the two countries. Also, the populations collected in Nigeria formed three distinct subgroups. Unique loci undergoing selection were observed between the Kenyan and Nigerian populations and among the three subgroups found in Nigeria. Striga hermonthica populations parasitising rice in Kenya appeared to be genetically distinct from those parasitising maize and sorghum. The presence of distinct populations in East and West Africa and in different regions in Nigeria highlights the importance of developing and testing Striga control technologies in multiple locations, including locations representing the geographic regions in Nigeria where genetically distinct subpopulations of the parasite were found. Efforts should also be made to develop relevant control technologies for areas infested with ‘rice‐specific’ Striga spp. populations in Kenya.  相似文献

Native fungi of West Africa were evaluated as a means to control Striga hermonthica (Del.) Benth., a troublesome parasitic weed of several gramineous crops. An isolate of Fusarium oxysporum, grown on sorghum straw and incorporated into pots, successfully prevented all emergence of S. hermonthica, and resulted in a 400% increase of sorghum dry weight. The fungus inhibited germination and attachment of S. hermonthica to sorghum roots in Plexiglas root chambers. Several crop species, including sorghum, inoculated with the fungus showed neither any disease symptoms nor any reduction in biomass.  相似文献

The effects of nitrogen and the extent of sorghum root infection by Striga hermonthica on host-parasite association during vegetative growth were studied using a split root system in a 3 × 3 factorial combination of N (37mg on one, 18.5 or 37mg on both root-halves) and Striga (no, one or both root-half infection). High N increased sorghum shoot weight by 22% more than low N, but did not significantly affect Striga growth 64 days after transplanting sorghum (DAP). Striga reduced sorghum stem height and weight by 22% and 25% at 38 DAP, and by 34% and 36% at 64 DAP, respectively. Leaf weight was not affected. Striga stimulated root growth 38 DAP, but not 64 DAP. In partially infected sorghum, 64 DAP, the parasite shoot number, shoot height and shoot dry weight were 36%, 46% and 35%, respectively and host shoot dry matter was 142% of those in fully infected plants, indicating an inverse relationship between the degree of host root infection and the level of resistance. The results suggest that sorghum released resistance-confering substances to the infection points after sensing infection. When infection points are widely distributed as in fully infected sorghum, less of such substances appear to render the host more vulnerable.  相似文献

Strigolactones, plant‐secreted underground signalling molecules, play an important role in agricultural ecosystems, because they mediate the interaction of crops with symbiotic AM fungi and parasitic weeds like Striga hermonthica. Cereal host plants secret these signalling molecules particularly under nutrient‐deficient conditions and especially when phosphate (P) is limiting. The objective of the present study was to see the potential of P seed priming for Shermonthica management in cereals in relation to strigolactone production. It has been demonstrated that P fertiliser application down‐regulates the production of these signalling molecules in the rhizosphere, which results in lower Shermonthica infection of cereals. The laboratory study showed maximum production of strigolactones from dry and water‐soaked seeds, while seed soaking in P solution reduced their production. Similarly, maximum Shermonthica infection was observed under control treatments with dry sowing or water soaking, while P seed soaking decreased Shermonthica germination, emergence and dry biomass in all cereal crops. Our study shows that P seed priming resulted in lower exudation of strigolactones, which induced less Shermonthica seeds germination and hence may lead to lower Shermonthica infection. P‐based seed priming could prove to be an effective and affordable strategy to reduce Shermonthica infection in cereals. Further research for practical field application is needed.  相似文献

The possibility of reducing Striga hermonthica (Del.) Benth. parasitism in severely infested fields, by means of deep planting - thereby reducing the root length in the upper layers of the soil where Striga seeds are predominantly found - was tested in field trials with maize and sorghum in western Kenya. Sorghum seeds were planted in Striga-infested fields approximately 2.5 cm deep in the soil or at the bottom of conically-shaped plant holes (15-20 cm deep). Depth of plant holes for maize varied from 0 to 30 cm, in un-tilled soil. Deep planting in un-tilled soil gave higher (up to double) grain yields, compared with standard planting in tilled soil. Parasite emergence was related negatively to planting depth of maize (p< 0.05). Deep planting in tilled soil gave 74% more sorghumgrain yield relative to standard planting. In this treatment Striga seed production was not reduced but in un-tilled fields with deeply planted sorghum Striga seed production was completely suppressed. Therefore, a combination of zero-tillage and deep planting seems to be the most effective treatment. The probable mechanism causing these results is avoidance of Striga seed by the host root system, resulting in a delay in the onset of Striga attachment and the formation of smaller numbers of attachments.  相似文献

Striga hermonthica and S. gesnerioides pose serious threats to cereal and cowpea production, endangering peoples' livelihoods on the Abomey plateau, Benin. A 2-year joint experiment was undertaken with farmers in two hamlets to investigate the potential of managing sowing dates of cowpea, sorghum transplanting, and trap cropping as ways of increasing agricultural production and reducing Striga damage. Early sowing of cowpea failed due to dry spells. Late sowing reduced cowpea yield due to water deficiency at the end of the growing season. Transplanting sorghum seedlings raised in fertilised or Striga-free nurseries doubled or tripled cereal yield and substantially reduced S. hermonthica infestation compared to direct early-sown sorghum. Transplanting sorghum from plant hills to fill gaps was unsuccessful. Trap crops such as cowpea and groundnut increased subsequent maize yield. Trap cropping had only a small effect on S. hermonthica infestation. The very poor soils in Somè central were a major constraint upon yield improvement to acceptable levels even after the introduction of the new crop (and Striga) management methods.  相似文献

Ten Sorghum vulgare (Pers.) cultivars varying in tolerance to Striga hermonthica (Del.) Benth. parasitism were grown with or without Striga infection. Endodermal thickening, pericycle lignification and silica crystal deposition were studied microscopically and measured for infected and non-infected sorghum cultivars. Although differences in the root character measurements were statistically significant they were not closely related to the response of the plant to infection. Low stimulant producing cultivars showed low or medium root cell thickening. The cv. Framida had both low stimulant production and high root cell thickening and was the best of the tolerant cultivars. High stimulant producing, tolerant cultivars generally showed heavy or intermediate cell thickening. The high stimulant producing, susceptible cultivar Debaikri also showed intermediate root cell thickening.‘Antibiosis', measured by the content of phenolic compounds in the plant, was then studied. Varietal differences in quality and quantity of phenolic substances in the roots and shoots of sorghum cultivars infected or non-infected with Striga were observed. Infection increased total phenolic contents in both shoot and root extracts. Differences in the total phenolic content in the shoot of non-infected cultivars did not reflect tolerance to Striga infection. The total phenolic acid content of the root extracts was closely related to the response of the host plant to Striga infection, tolerant cultivars having greater total phenolic acid content than susceptible ones.  相似文献

Prevention of seed input to the seedbank of Striga hermonthica‐infested fields is an important objective of Striga management. In three consecutive years of field experimentation in Mali, Striga reproduction was studied for 10 sorghum genotypes at infestation levels ranging from 30 000 to 200 000 seeds m?2. Host resistance was identified as an important determinant of Striga reproduction, with the most resistant genotypes (N13, IS9830 and SRN39) reducing Striga reproduction by 70–93% compared with the most susceptible genotype (CK60‐B). Seedbank density had a significant effect on Striga seed production. Higher seedbank density resulted in more Striga plants, which led to increased intra‐specific competition and consequently a reduced level of reproduction per plant. For the most susceptible sorghum genotypes, density dependence also occurred in the earlier belowground stages. Striga reproduction continued beyond harvest. At the high infestation level just 8% of the total reproduction was realised after harvest, whereas at the low infestation level 39% was attained after harvest. Even though host‐plant genotype plays a significant role in Striga reproduction, calculations indicated that only at very low infestation levels the use of the most resistant genotype was able to lower the Striga seedbank.  相似文献

For the first time in sterile nutrient media in the absence of the host plant, different forms and rates of nitrogen compounds were screened for their effect onS. hermonthica Benth. shoot development beyond seed germination. There was no shoot formation beyond the inoculation stages whenS. hermonthica germlings were grown in media devoid of nitrogen source. In culture media containing some nitrogen sources, healthy shoots were formed. Increasing concentrations of KNO3, NaNO3, Ca(NO3)2, Mg(NO3)2 and asparagine resulted in a significant increase inS. hermonthica shoot development. Inversely, increasing concentrations of (NH4)2SO4, NH4H2PO4, NH4Cl and urea led to increasing significant reduction ofS. hermonthica shoot development. The amino acids, glycine and asparagine supported reduced shoot development ofS. hermonthica, indicating that organic nitrogen cannot replace inorganic nitrogen forStriga growth. The ammonium nitrogen compounds, (NH4)2SO4 and NH4H2PO4, suppressed further shoot elongation and total dry weight of 20 and 40 days oldStriga hermonthica plants, in sterile culture media. The organic compounds urea, allylthiourea and thiourea had an effect similar to ammonium compounds. Arginine and glycine on the other hand did not suppress the further development of the parasite. The suppressive effect of nitrogen however, was greater when the parasite was 20 days old than when it was 40 days old. This work provides data to show that some nitrogen compounds reduce the severity ofS. hermonthica attack by direct suppression ofStriga growth and development at the post-germination stage and after shoots have been formed.  相似文献

Thirteen fungal species were isolated from diseased plants of Striga hermonthica (Del.) Benth in northern Ghana. The pathogenicity of 12 isolates of the fungal species including Curvularia fallax Boed, Fusarium equiseti (Corda) Sacc., Fusarium equiseti var. bullatum (Sherb.) Wollenw., F. oxysporum Schlecht, F. solani (Mart) Sacc., Macrophonuina phasealina (Tassi) Goidan, and Sclerotium rolfsii (Sacc.) were evaluated against S. hermonthica under controlled environmental conditions. All isolates were pathogenic to S. hermonthica when propagated on wheat (Triticum aestivum L.) grains and incorporated pre-planting into the soil. However, their virulence differed. Two isolates of F. oxysporum and one isolate of F. solani reduced the emergence of S. hermonthica by 88%, 98%, and 76%, respectively. Sorghum (Sorghum bicolor (L.) Moench) yield was increased by 26% when S. hermonthica was controlled. In contrast, in the control treatment with S. hermonthica no yield could be obtained. The F. oxysporum isolates were not pathogenic on sorghum. Germination tests indicated that the F. oxysporum isolates were highly pathogenic to S. hermonthica seeds. Although the F. solani isolate reduced the emergence of S. hermonthica in the pot experiments, it did not influence germination. This indicates that pathogens may attack different stages in the life cycle of S. hermonthica.  相似文献

Castanea sativa is susceptible to Phytophthora spp., a serious root pathogen causing ink disease, while C. crenata and C. mollissima show resistance to infection. Interspecific controlled crosses were established for introgression of resistance genes from the resistant species into the susceptible C. sativa, and two mapping populations were created. Phytophthora cinnamomi resistance of each progeny was evaluated by root and excised shoot inoculation tests. The number of days of survival after root inoculation was the best discriminator of resistance to P. cinnamomi while the percentage of shoots with internal lesions was the symptom most associated with survival. The lesion progression rate in the excised shoot inoculation test was strongly and negatively correlated with survival in the root inoculation test. The excised shoot inoculation test appears to be a reliable approach for screening the resistance of chestnut genotypes to P. cinnamomi. Strong genetic correlations were obtained between survival and ink disease symptoms and among symptoms, indicating that common or linked genes might influence resistance to P. cinnamomi. The most resistant genotypes selected from this study will be tested for other commercial variables, such as ease of vegetative propagation and stock–scion compatibility.  相似文献

Centaurea stoebe is native to Europe and Western Asia and was introduced into North America in the late 19th century, where it has become highly invasive. In its native range, C. stoebe occurs in two cytotypes, namely diploids (2n = 18) and tetraploids (2n = 36), but only the tetraploid form has been identified in the invaded range. We used special growth pouches to determine whether diploid and tetraploid cytotypes from the native range differed in root growth and architecture. We grew seeds from five populations of each cytotype in growth pouches during a period of sixteen days and measured root growth traits both by hand and using a root‐scanning software package (WinRHIZO). Tetraploid cytotypes had significantly larger total root length, taproot length, surface area, root volume, above‐ and below‐ground biomass and root to shoot ratios than diploid cytotypes. We suggest that increased early root growth of tetraploid cytotypes as compared with diploids may be one factor that pre‐adapted them towards the colonisation of warmer and drier climates in Europe and North America, where tetraploids are currently expanding and invasive respectively.  相似文献

Economically feasible strategies to cope with invasive species are urgently needed. Plant density can be increased to reduce competitive effects on target plants. This study indicates that increasing native plant density can be used to reduce the effect of invasive Ageratina adenophora. Seedlings of an indigenous tree species, Schima wallichii, were grown in pots containing uninvaded or invaded soil, with or without A. adenophora leaf litter on the soil surface. Schima wallichii seedlings were also grown at four densities under four levels of A. adenophora leaf litter. Root and shoot biomass and length were measured as response parameters in both bioassays. Schima wallichii growth was inhibited by A. adenophora leaf litter and invaded soil. High litter levels reduced S. wallichii root length and dry weight at low plant densities. The inhibition disappeared at high S. wallichii plant densities. As A. adenophora did not inhibit S. wallichii growth at high plant densities, adjustments of seedling density should be studied as a possible management strategy for invasion by A. adenophora and potentially by other exotic plant species. As density‐dependent growth inhibition is the key characteristic of chemical interference, we propose that phytotoxins contribute to A. adenophora invasion particularly at low densities of native seedlings.  相似文献

以22份山西历年审定(鉴定、登记)的高粱品种及预登记的高粱品种为参试材料,通过自然干旱(DS)和正常灌溉(NI)两种处理方式,采用大田自然鉴定法,调查农艺性状、品质性状和光合性状3类主要性状指标。结合隶属函数(U)和干旱敏感指数(S)的聚类分析法及热图法对材料进行分类,对抗旱指标进行综合评价。结果表明:在干旱胁迫下,高粱品种除了穗柄伸出长平均增长5.26 cm和单宁含量平均增加0.09%,其余性状指标均出现下降;除净光合速率外,其余性状指标之间均存在一定程度的相关性。利用以上两种方法,筛选出极抗品种3份(S9、S10和S7即晋杂108、晋杂22号和晋杂101),极弱品种1份(S17即晋杂4号)。根据干旱敏感指数计算,主成分分析为9个性状,主成分累计贡献率84.662%,产量和单穗粒重是决定第一主成分的主要性状,贡献率为17.999%。干旱胁迫对高粱品种(系)花后主要性状指标均有显著影响。不同高粱材料间抗旱性差异显著,应用两种抗旱性评价方法,可以较为准确地评价参试高粱品种(系)的抗旱性。产量、单穗粒重、株高和倒二叶叶宽可以作为评价高粱花后抗旱性的主要形态指标。  相似文献

The goal of the present study was to determine whether grafting of watermelon on gourd rootstocks could improve alkalinity tolerance and to investigate the physiological and morphological response mechanisms of the grafted plants under different pH levels. The experiment was carried out in a climate chamber to investigate growth, leaf chlorophyll content (SPAD), leaf area, stem length, shoot and root dry weight, root length, electrolyte leakage, leaf mineral composition, total chlorophyll and carotenoid contents. Two watermelon cultivars (Crimson tide, CT, and Crisby) were grafted onto three commercial Cucurbita maxima?×?C. moschata hybrid rootstocks under climate-chamber conditions (Strong tosa, ST, Ercole and Nun 9075). The grafted seedlings were transplanted onto 8?L continuously aerated pots containing nutrient solution with two different pH levels (8.5 and 6.5) and replicated three times. The results showed that in both grafted and non-grafted plants, there was a substantial reduction in shoot and root biomass production at high pH levels. At high pH level, the significantly highest leaf area, stem length, SPAD, concentration of P, Ca, S and Mn in leaf tissues were recorded in graft combination ‘Crisby/Ercole’, whereas the significantly highest concentration of Fe in leaf tissues, shoot dry weight were recorded in graft combination ‘Crisby/Nun 9075’. Moreover, at high pH, the significantly highest concentration of Mg and Cu in shoot under high pH levels was significantly found in graft combination of ‘CT/ST’. These results suggest that the use of interspecific Cucurbita maxima?×?C. moschata hybrid rootstocks can improve crop performance in watermelon plants under alkaline conditions.

The effects of irrigation on verticillium wilt in olive, in terms of morphological, biomass and physiological parameters were evaluated on pot‐grown trees maintained in the field for 3 years. Plants inoculated and noninoculated with Verticillium dahliae were irrigated to high and low range of soil water content (HR and LR) at daily (DF; about 2 days/event), weekly (WF) and daily‐weekly (DWF) drip‐irrigation frequency. Morphological parameters, relative biomass and biomass water‐use efficiency were higher at LR than at HR (with few exceptions) and at DF than at other frequencies in noninoculated and inoculated plants, but the fungus reduced those parameters by 17.0–38.5%. Lower root weight ratio, relative biomass and shoot length as area originated at HR in noninoculated plants, could be favourable to the accumulation of root infections and the amount of fungus per tissue length in inoculated plants because higher infection was known at HR. Moreover, higher aerial biomass and length promoted by irrigation at DF could prevent the more severe expression of symptoms, which occurred at WF and DWF in the presence of Verticillium. Negative correlations were found between indicated parameters and disease. Lower water stress (SΨ), and higher stomatal conductance (gs) and net photosynthesis at DF in noninoculated plants could limit the disease by improving water status, as SΨ was increased by the fungus only at WF and DWF, and gs and disease were negatively correlated. LR‐DF treatment minimized the disease and kept the growth, water‐use efficiency and physiological parameters in inoculated plants to levels close to noninoculated plants.  相似文献

Schoenoplectus juncoides is one of the most harmful weeds found in East Asian paddy fields. Recent emergence of biotypes that are resistant to the herbicide sulfonylurea (SU) has made weed control difficult. To examine the effect of the evolution of this herbicide resistance on genetic diversity within local populations, we investigated microsatellite variability within and among paddy field populations of S. juncoides in Kinki, Japan. In vivo assay of acetolactate synthase activity and root elongation assay in the presence of SU revealed that of 21 populations, five were sulfonylurea‐susceptible (SU‐S) and eight were completely sulfonylurea‐resistant (SU‐R). The remaining eight populations were a mixture of SU‐S and SU‐R individuals. The average gene diversity for SU‐R populations (HS = 0.168) was lower than those for SU‐S (HS = 0.256) and mixed (HS = 0.209) populations, but the difference was not significant. This indicates that positive selection for SU‐R phenotype did not cause a genome‐wide reduction in genetic diversity. Genetic differentiation among S. juncoides populations was higher than that observed for most weed species studied previously. Although populations in neighbouring paddy fields showed a high level of differentiation, Bayesian clustering analyses suggested that some level of gene flow occurs among them and that the genetic exchange or colonisation between neighbouring populations could contribute to the geographical expansion of the resistant allele.  相似文献

Sonchus arvensis is a problematic perennial weed species causing problems in many crops. Control measures often rely on soil tillage or mowing; both methods are used to exhaust the root system. This approach fails during periods of restrictions in sprouting capacity or shoot regrowth. The aim of this study was to develop a simulation model for prediction of shoot sprouting capacity in underground buds of S. arvensis. We hypothesised restricted regrowth to be induced as a function of photoperiod and temperature sum and released as a function of cold temperature sum. The model was parameterised and calibrated against observations of shoot regrowth from an experiment on intact roots of S. arvensis. Subsequently, it was validated against observations of regrowth from another experiment on intact roots and sprouting from fragmented roots from a third experiment. Restriction in regrowth was attributed to reduction in the photoperiod, observed when the day length decreased below a threshold of 16.5 h, but also to a daily mean temperature below 16.8°C. Resumed regrowth occurred in late autumn at a daily mean temperature threshold of 4.7°C. The pattern of changes over time in shoot regrowth from an intact root system was well predicted by the model. The temporal change in sprouting capacity from buds on fragmented roots followed the same pattern, but with a quantitative difference between the model prediction and the observed sprouting capacity. We conclude that the model can be used to predict the pattern over time of sprouting capacity in intact roots of S. arvensis.  相似文献

Thidiazuron at 0.1 to 10 mg 1?1 induced concurrent germination and haustorium initiation in Striga asiatica (L.) Kuntze and S. hermonthica (Del.) Benth, but it had no effects on S. gesnerioides (Willd.) Vatke. Both millet and sorghum strains of S. hermonthica were equally responsive. The response of Striga seeds to thidiazuron increased with conditioning. Early applications of the compound induced some germination, but had adverse effects on the conditioning process.
