Recent evidence suggests that nitric oxide (NO) signaling plays an important role in plant–pathogen interactions and that aconitase is a major target of NO. In the present study on the signaling role of NO in the elicitation of defense responses in peach fruit against Monilinia fructicola and subsequent effect on brown rot disease, 15 μM NO solution induced disease resistance in harvested peaches. As a potentiated elicitor, NO induced high levels of endogenous NO and superoxide (O2 ?), hydrogen peroxide (H2O2), and NADPH oxidase and Ca2+-ATPase activity in the fruit. Aconitase activity in peach fruit was inhibited by NO. Activity of partially purified aconitase was inhibited in vitro by sodium nitroprusside (SNP) and H2O2; however, the inhibition could be relieved by carboxy-2-phenyl-4,4,5,5-tetramethylimidazoline-1-oxyl 3-oxide (cPTIO) or catalase (CAT), indicating that the defense response and signals induced by NO transduction depend on aconitase and conditions leading to elevated levels of NO; otherwise, H2O2 would inactivate aconitase directly in fruit. Treatment with NO resulted in salicylic acid (SA) accumulating during storage. Higher levels of jasmonic acid (JA) were detected in NO-treated fruit 48 h after the treatment. But after NO was removed, the level of SA and JA were lower than in the control. The results suggest that exogenous NO enhances resistance of harvested peach fruit against the fungus by inducing signals such as endogenous NO, reactive oxygen species (ROS), SA and JA and by inhibiting aconitase activity.  相似文献

The effect of hot water treatment (HWT) to control peach brown rot was investigated. Peaches were dipped in water at 60°C for 60 s and artificially inoculated with Monilinia fructicola conidia. HWT failed to control brown rot if applied before inoculation and microscopic observations revealed a stimulatory effect on germ tube elongation of M. fructicola conidia placed immediately after HWT on the fruit surface, compared to the control. The influence of fruit volatile emission due to HWT was performed on the pathogen conidia exposed to the headspace surrounding peaches. The results showed an increase of M. fructicola conidial germination ranging from 33 to 64% for cultivars Lucie Tardibelle and Red Haven heat‐treated peaches, respectively, compared to the control. The volatile blend emitted from heat‐treated fruit was analysed by solid‐phase microextraction/gas chromatography‐mass spectrometry (SPME/GC‐MS) and proton transfer reaction‐time of flight‐mass spectrometry (PTR‐ToF‐MS). Fifty compounds were detected by SPME/GC‐MS in volatile blends of cv. Lucie Tardibelle peaches and significant differences in volatile emission were observed among heated and control fruit. Using PTR‐ToF‐MS analysis, acetaldehyde and ethanol were detected at levels 15‐ and 28‐fold higher in heated fruit compared to unheated ones, respectively. In vitro assays confirmed the stimulatory effect (60 and 15%) of acetaldehyde (0·6 μL L?1) and ethanol (0·2 μL L?1) on M. fructicola conidial germination and mycelial growth, respectively. The results showed that volatile organic compounds (VOCs) emitted from heat‐treated peaches could stimulate M. fructicola conidial germination, increasing brown rot incidence in treated peaches when the inoculation occurs immediately after HWT.  相似文献

The ability to meet consumers demand for high-quality standard fruit entails the distribution of unblemished safe fruit free of chemical residues on its edible portion. Therefore, this study was focused on investigating the influence of the combined effect of aqueous plant volatiles with half strength prochloraz solution to control anthracnose and stem-end rot in the green-skinned avocado cultivar (Fuerte). This method was applied due to its practicability on bulk fruits in packhouses and the fruits were subjected to stand-alone and combined treatments to assess the development of the disease after cold storage and observe the elicitation of the residual effect of the treatments on the defence related enzymes in ‘Fuerte’. The incidence of stem-end rot was 10% by the combination of prochloraz® (500 μg mL?1; P50) with 0.1% v/v thyme oil compared to the 58.8% incidence exhibited by the untreated fruit during storage at 6.5 °C for 14 days followed by 3 days at retail shelf conditions (15 °C) (preventative application). Citral (0.1% v/v), P50 (500 μg mL?1)?+?0.1% v/v citral and yucca extract alone reduced the stem-end rot incidence to about 25% during storage. More so, thyme oil (0.1% v/v) reduced both anthracnose and stem-end rot incidence to 35% after postharvest storage and P50 (500 μg mL?1)?+?0.1% v/v thyme oil and 0.1% v/v thyme oil effectively induced the activity of phenylalanine ammonia lyase, chitinase and β-1, 3 glucanase in fruit inoculated with Lasiodiplodia theobromae and Colletotrichum gloeosporioides through the improvement of quality and firmness of the fruit after storage.  相似文献

Seven different strains of Trichoderma isolated from avocado roots showed antagonism to Rosellinia necatrix, which is the causal agent of white root rot. We studied these Trichoderma strains on the basis of the secondary metabolites produced in liquid culture. Five different compounds, namely, 6PP (6-pentyl-α-pyrone), Harzianolide (4-hexa-2,4-dienyl-3-(2-hydroxy-propyl)-5H-furan-2-one), T39butenolide (4-hexa-2,4-dienyl-3-(2-oxo-propyl)-5H-furan-2-one), Dehydroharzianolide (4-hexa-2,4-dienyl-3-propenyl-5H-furan-2-one) and Cerinolactone [(3-hydroxy-5-(6-isopropyl-3-methylene-3, 4, 4a, 5, 6, 7, 8, 8a-octahydronaphthalen-2-yl) dihydrofuran-2-one); a recently discovered novel metabolite], were obtained. In vitro studies of the effects of these compounds on different R. necatrix strains isolated from avocado roots and with different virulence demonstrated that 6PP had the strongest effect even at a low concentration. Although unstable, Cerinolactone and T39butenolide also had large effects on R. necatrix, mainly at a concentration of 200 μg. Harzianolide and Dehydroharzianolide exhibited the lowest effects on the pathogen. In vivo studies of Trichoderma metabolites on Lupinus luteus plants demonstrated the delay of white root rot epidemic through preventive application of 6PP or Harzianolide to seeds or plantlets by immersion in solutions of these metabolites at 1 mg l?1 (minimum effective dosage). In contrast, Cerinolactone only was effective at 10 mg l?1 when applied by plantlet immersion. Thus, this study reports the role that these metabolites could play for controlling avocado white root rot caused by R. necatrix.  相似文献

Stem-end rot, caused by Lasiodiplodia theobromae, is an important postharvest disease of papaya in Brazil. The use of fungicides is one of the main disease management measures. However, there are no data available on the sensitivity of L. theobromae to thiophanate methyl (methyl benzimidazole carbamate), the most common fungicide used in papaya orchards in northeastern Brazil. Thus, the effective concentration that results in 50 % of mycelial growth inhibition (EC50) of 109 isolates, representing five populations of the pathogen was estimated in vitro. Seven components of fitness were measured for the 10 isolates with lower and high values of EC50. Of the 109 isolates, 20.2 % were resistant to the fungicide with EC50 values greater than 300 μg ml?1, whereas the remaining 79.8 % were sensitive with an average EC50 of 1.87 μg ml?1. The EC50 values for the resistant isolates were significantly (P?≤?0.05) higher than those for the sensitive isolates. When the fitness components were evaluated, only in relation to the spore production was significant difference among sensitive and resistant isolates, and resistant isolates showed sporulation capacity significantly lower than the S isolates, indicating a fitness cost.  相似文献

The purpose of this study was to determine if exogenous cholesterol availability influenced Pythiaceae resistance to antibiosis. Characterisation of an isolate of Phytophthora erythroseptica and Pythium ultimum for tolerance to antibacterial compounds found that 0.05 g.l?1 chloramphenicol inhibited mycelial growth by 96.6 % and 23.5 % respectively. However, the addition of cholesterol (0.01 g l?1) to potato dextrose agar (PDA) containing 0.05 g l?1 chloramphenicol was found to increase mycelial growth of P. erythroseptica, indicating a role for cholesterol in tolerance to inhibitory antibacterial compounds. To determine if this property extended to suppressive effects of a potential biocontrol agent, P. erythroseptica and P. ultimum were then tested against a cell-free filtrate of diffusible metabolites produced by a suppressive Trichoderma harzianum isolate in the presence and absence of cholesterol in PDA. In the absence of cholesterol, diffusible metabolites of the T. harzianum isolate were found to inhibit mycelial growth of P. erythroseptica and P. ultimum on PDA by 98 % and 63.6 % respectively (P?<?0.0001). However, the inhibitory effect of the metabolites was mitigated when 0.005 g l?1 of cholesterol was present in PDA, with mycelial growth of P. ultimum and P. erythroseptica reduced by only 60.4 % and 41.8 %, respectively (P?<?0.0001), much less inhibition than was observed in the absence of cholesterol. These results demonstrated that access to exogenous cholesterol can influence the sensitivity of Pythiaceae species to antibiosis by positively influencing mycelial growth.  相似文献

A suspension of nonpathogenic Xanthomonas campestris (nXc) strain AZ98101 or AZ98106 was sprayed on peach trees in biological control tests against bacterial spot caused by Xanthomonas arboricola pv. pruni. Treatment with each strain reduced the number of leaves with spots. Meta-analysis of the results from seven field trials showed an integrated risk ratio (the ratio of leaf spot incidence on inoculated leaves to incidence on control leaves) of 0.46 by treatment with AZ98101 and 0.54 by AZ98106, indicating that leaf spot incidence was significantly reduced by each strain. Each also significantly reduced the incidence of fruit spot. The risk ratio with AZ98101 was significantly lower than that with AZ98106. The population of AZ98101R-1, a rifampicin-resistant mutant of AZ98101, was 6.3 × 103 CFU/g (fresh mass) on leaves and 8.0 × 103 CFU/g on fruit at 12 days after inoculation, but was undetectable (<102 CFU/g) by 24 days. This is the first report that nXc strains AZ98101 and AZ98106 effectively control bacterial spot on peach.  相似文献

In 2014 and 2015, a total of 151 tobacco brown spot (Alternaria alternata) isolates were collected from Guizhou Province in China to evaluate their resistance to the benzimidazole thiophanate-methyl, the carbamate diethofencarb, and the dicarboximide procymidone. Resistance to thiophanate-methyl and diethofencarb was observed in all isolates. Resistance to all the three fungicides, thiophanate-methyl, diethofencarb, and procymidone was detected at a frequency of 6.0%. The F167Y single mutation in the β-tubulin gene was found to be associated with resistance to thiophanate-methyl,but no mutation was found in the coiled-coil region of the histidine kinase-encoding gene OS1, a fungal gene for dicarboximide resistance. Procymidone applied at the rate of 20 mg l?1 inhibited spot lesion formation on tobacco leaves with an efficacy of 51.7% for the low resistance (LR) isolates and 74.2% for the procymidone-sensitive isolates. Thiophanate-methyl applied at 100 mg l?1, however, slightly promoted the expansion of disease lesions with an efficacy of ?7.7%. Azoxystrobin applied at 10 and 20 mg l?1 provided efficacies of 91.1 and 100%, respectively, regardless of whether the isolates were thiophanate-methyl resistant or procymidone-LR. Further studies suggested that azoxystrobin exhibited excellent protective activity and good curative activity against A. alternata in plants. The baseline sensitivity to azoxystrobin was then determined. In the presence SHAM, the mean EC50 values for conidial germination inhibition were 0.49?±?0.22 (Mean?±?SD) mg l?1. Interestingly, no resistance was recovered through UV irradiation or Agrobacterium tumefaciens-mediated mutagenesis. This research indicated widespread resistance to thiophanate-methyl and diethofencarb, low frequency of (6.0%) resistance to procymidone in A. alternata populations from tobacco, and suggested that azoxystrobin could potentially constitute a good alternative for the management of tobacco brown spot disease.  相似文献

In order to investigate the effects of flagellin secreted by bacteria associated with pine wood nematode (Bursaphelenchus xylophilus) on pine cells, suspension cells of Pinus thunbergii were treated with flagellin of Pseudomonas fluorescens GcM5-1A, and some physiological indices were determined. Superoxide anions radical (·O2 ?) in treated suspension cells increased with prolongation of treatment time, with maximum content of 1,282 μmol g?1 FW at 30 h, but the content of H2O2 gradually decreased as compared with that in the control group. Maximum content of malondialdehyde (MDA) and free proline, which is 1.72 times and 1.49 times of that in the control, respectively, appeared when suspension cells were treated for 12 h, after that both of the contents decreased gradually. The activities of superoxide dismutase (SOD), peroxidase (POD) and catalases (CAT) also increased at the primary stage of treatment and then decreased compared with those of the control.  相似文献

Rhizopus rot, caused by Rhizopus stolonifer, is one of the main postharvest diseases in stone fruits, but there is little known about the processes of disease development during transport and postharvest storage. The objective of this study was to characterize temporal progress and spatial distribution of the disease in peach fruit. Rhizopus rot development was evaluated using two different fruit arrangements. Only one fruit of each arrangement was inoculated with a R. stolonifer spore suspension. Disease incidence and severity were assessed daily for all the fruit. Nonlinear models were fitted to the quantity of fruit and to the area of fruit that became infected over time and distance in relation to the source of inoculum. Disease‐free fruit placed next to the artificially inoculated peaches showed disease symptoms due to pathogen dissemination by mycelial stolons. The disease incidence and severity progress rates varied from 0.33 to 0.53 day?1 and from 0.30 to 0.49 day?1, respectively. The spatial spread of the disease followed a dispersive wave pattern with increasing speed over time, but decreasing speed with disease severity. For disease severity = 0.5, the velocity at day 3 varied from 0.14 to 0.32 fruit diameter day?1, while it ranged from 0.38 to 1.46 fruit diameter day?1 at day 12.  相似文献

Brinjal shoot and fruit borer Leucinodes orbonalis Guen. is a major pest of brinjal in India. The field collected larvae of L.orbonalis were tested for their susceptibility to three diamide insecticides by fruit dip bioassay technique. Cyantraniliprole and chlorantraniliprole were 5.23 and 2.80 times more toxic to L. orbonalis as compared to flubendiamide. Large variation in the susceptibility of L. orbonalis to cyantraniliprole, chlorantraniliprole, flubendiamide was observed and the LC50 values were 0.084, 0.157 and 0.439 mg a.i. L?1, respectively. In span of two years there was a significant increase in the LC50 values of cyantraniliprole (0.062 to 0.085 mg a.i. L?1), chlorantraniliprole (0.097 to 0.157 mg a.i. L?1), flubendiamide (0.284 to 0.439 mg a.i. L?1) to population of L. orbonalis, which showed 1.35, 1.62 and 1.55 fold resistance, respectively indicating faster development of resistance to diamide insecticides.  相似文献

Seventeen field surveys were done in four commercial orchards during six consecutive fruit-growing seasons from 2006 until 2011 in order to determine the current frequencies of occurrence of M. laxa, M. fructigena, and M. fructicola and their relative contributions to postharvest brown rot in peaches and nectarines in the Ebro Valley. The relative frequencies of occurrence of Monilinia spp. were determined on three sources of primary inoculum and on three sources of secondary inoculum. The major relative frequencies of Monilinia spp. were significantly recorded (P?=?0.05) from mummified fruit on the trees (approx. 42 %) and 7-day-old harvested fruit with brown rot (32 %), followed by that recovered from mummified fruit on the orchard bed (14 %), pruned branches on the orchard bed (8 %) and latent infections of immature fruit (3 %). We found that: (a) the relative frequency of M. fructicola has increased over the years to coexist on the same level as at the time M. laxa, (b) M. fructigena is no longer a cause of brown rot in harvested peaches, (c), a progressive reduction in the time of the first appearance of Monilinia airborne conidia (r?=??0.30, P?=?0.003), and the time of the first latent infection (r?=??0.44, P?=?0.0001) was detected along years after correlation analysis, and (d) these displacements are not associated with an increased incidence of brown rot disease. The M. fructicola increase was due to its significantly increased presence in 7-day-old harvested fruit with brown rot (r?=?0.73, P?=?0.0009), in latent infections of immature fruit (r?=?0.68, P?=?0.002), on pruned branches on the orchard bed (r?=?0.56, P?=?0.018), and on mummified fruit sampled on the trees (r?=?0.53, P?=?0.03). This progressive increase was accompanied by a progressive reduction in the relative frequency of occurrence of M. laxa in 7-day-old harvested fruit with brown rot (r?=??0.55, P?=?0.021) and M. fructigena on mummified fruit sampled on the trees (r?=??0.51, P?=?0.03).  相似文献

The tobacco cutworm, Spodoptera litura (Lepidoptera: Noctuidae) (Fabricius), is a polyphagous pest which has developed physiological resistance against a number of known insecticides. In order to reduce the load of synthetic insecticides, ecofriendly alternative strategies are needed. The present studies were conducted to evaluate the insecticidal potential of the endophytic fungus Cladosporium uredinicola isolated from Tinospora cordifolia (Thunb.) on S. litura. Development of S. litura was significantly prolonged when larvae were fed on diet amended with ethyl acetate extract of C. uredinicola at concentrations of 1.25–2.00 μl?g?1. The negative effects of fungal toxin on development of S. litura further resulted in a significant reduction in adult emergence, longevity and reproductive potential at higher concentrations. A significantly higher number of adults showed morphological deformities when larvae were fed on diet amended with 2.00 μl?g?1 concentration. The inhibitory effects on growth and development of S. litura indicate toxicity of ethyl acetate extract of C. uredinicola, which is further evidenced by reduced food utilization by larvae. The antifeedant and toxic effects of ethyl acetate extract of C. uredinicola observed in the present studies can be attributed to a bioactive compound produced by the fungus, so this study provides evidence that C. uredinicola isolated from T. cordifolia possesses anti-insect properties and may play an important role in protecting plants against insect pests.  相似文献

This study investigated the effect of calcium nutrition on tomato bacterial wilt caused by Ralstonia solanacearum and the regulation of resistance mechanisms. Plants cultured in nutrient solution with calcium concentrations of 0.5, 5.0, and 25.0 mM, were inoculated with R. solanacearum by the root dip method. Severity of disease development, Ca concentration in tomato root and shoot tissues, hydrogen peroxide (H2O2) concentration, peroxidase (POD, EC 1.11.1.7) and polyphenol oxidase (PPO, EC 1.10.3.2) in tomato leaves were analyzed. Disease severities of low, medium and high Ca treatments were 100 %, 77.1 % and 56.8 % respectively. Plant growth in high Ca treatment was significantly better than those in low Ca treatment in height, stem diameter and biomass. Tomato plants absorbed significantly more Ca in roots and shoots as the level of Ca in the nutrient solution increased. In addition, H2O2 level in high Ca treatment rose faster and reached a higher peak with 10.86 μM gFW?1(31.32 % greater than medium Ca plants). The activities of POD and PPO also have a greater increase in high Ca treatment with 99.09 U gFW?1 and 107.24 U gFW?1 compared to 40.70 U gFW?1 and 77.45 U gFW?1 in low Ca treatment. A negative correlation was found between Ca concentration, level of H2O2, POD, PPO in tomato, and disease severity, indicating that they played an important role in resistance of tomato to this disease. These results suggested that Ca was involved in the regulation of H2O2 concentration, and activity of POD and PPO in tomato.  相似文献

Fungi infection in fruits is an important factor in postharvest losses. The effect of a treatment with a fungal elicitor on the response of tomato fruit to Fusarium oxysporum infection and changes in the phenylpropanoid metabolic pathway was studied. Fungal elicitor retarded for 3 days the development of the Fusarium rot development in tomato, at concentration of 2 g?l ?1 (B2-F treatment). This treatment also induced a 3.11- and 6.03-fold increase of caffeic and chlorogenic acids, respectively, as compared with the control. Furthermore, the flavonoids naringenin-7-O-glucoside, rutin and kaempferol-3-O-glucoside, showed a greater abundance in tomato under the B2-F treatment after 6 days at 20°C. It is concluded that the fungal elicitor reduced the development of Fusarium rot by inducing the biosynthesis of metabolites from the phenylpropanoid metabolic pathway which forms part of the defense response in tomato fruit.  相似文献

The grapevine (Vitis vinifera L.) cultivars ‘Agiorgitiko’ and ‘Malagouzia’, naturally infected with Grapevine rupestris stem pitting-associated virus (GRSPaV), were subjected to in vitro chemotherapy using the antiviral inosine 5′-monophosphate dehydrogenase inhibitors tiazofurin (TR), ribavirin (RBV) and mycophenolic acid (MPA). The chemotherapy lasted 80 days and was carried out as two consecutive treatments. Severe phytotoxicity, estimated after 40 days of culture, was observed in drug-treated explants, especially when high doses of TR were used. Phytotoxicity exhibited a cultivar- and chemical compound-dependent profile. The virus eradication status of the survived plantlets was determined by nested RT-PCR using total RNA templates, after 80 days of drug treatment and one year later, after the passage of one dormancy period, in potted plants grown in a greenhouse. Data indicated that the highest GRSPaV elimination in ‘Agiorgitiko’ was obtained with 10 μg? ml?1 TR, 30 μg ?ml?1 RBV and 20 μg? ml?1 MPA. The eradication rates were lower in the case of ‘Malagouzia’, where the highest ones were achieved after treatments with 15 μg ml?1 TR and 80 μg ml?1 MPA. This is the first report on GRSPaV elimination in grapevine following treatment with antiviral compounds, which could provide an alternative to the traditional methods of virus eradication through meristem culture and thermotherapy.  相似文献

From 2003 to 2007 surveys have been conducted in different stone fruit growing regions in southwest Germany to detect European stone fruit yellows (ESFY) disease in Germany. Samplings have been done regularly in selected reference orchards in the regions Neuwieder Becken, Rheinhessen, Vorderpfalz and Südpfalz in summer on trees showing ESFY typical symptoms as well as on branches of trees with unspecific symptoms. All samples have been analysed by PCR for infection with Candidatus Phytoplasma prunorum. The phytoplasma could be detected in all investigated regions on the cultivated Prunus species P. armeniaca, P. persica and P. domestica. No infection was found in wild Prunus species. The main spread of the disease appeared on apricot while peach and European plum were less affected. A good correlation between symptoms and molecular detection of the pathogen could be shown for the typical symptoms in summer and winter for apricot as well as for peach. During regular psyllid captures in the reference orchards the population dynamics of Cacopsylla pruni could be described in southwest Germany for several years. By PCR-testing all collected insects individually a yearly natural infection rate of about 1–2% of all individuals of C. pruni could be calculated.  相似文献

In this study peroxynitrite (ONOO?) is proposed as an important player in defence responses during the interaction of potato (Solanum tuberosum) and the oomycete pathogen Phytophthora infestans. The potato–avr P. infestans model system exhibited a transient programme of boosted ONOO? formation correlated in time with the burst of nitric oxide (NO) and superoxide during the first 6 h post‐inoculation (hpi). The early ONOO? over‐accumulation was not accompanied by TPx gene expression. In contrast, the compatible interaction revealed a 24 h delay of ONOO? formation; however, an enhanced level of NO and superoxide correlated with TPx up‐regulation was recorded within the earlier stages of pathogen infection. Peroxynitrite over‐accumulation in the susceptible potato coincided with an enhanced level of protein tyrosine nitration starting from 24 hpi. Surprisingly, the nitroproteome profile of the resistant potato did not show any visible difference after inoculation, apart from one band containing subtilisin‐like protease‐like proteins, which appeared 48 h after pathogen attack. An additional pharmacological approach showed that treatment of the susceptible genotype with ONOO? followed by inoculation with P. infestans contributed to slowing down of the colonization of host tissues by the pathogen via a faster and stronger up‐regulation of the key defence markers, including the PR‐1 gene. Taken together, the results obtained indicate that a precise control of emitted NO and superoxide in cooperation with thioredoxin‐dependent redox sensors in sites of pathogen ingress could generate a sufficient threshold of ONOO?, triggering defence responses.  相似文献

The performance of postharvest treatments with sodium propylparaben (SPP), alone or combined with low doses of the fungicide imazalil (IMZ), against citrus green (GM) and blue (BM) molds was evaluated on several citrus species and cultivars artificially inoculated with Penicillium digitatum and P. italicum, respectively, and incubated at 20 °C or cold-stored at 5 °C. Effectiveness of 100 mM SPP dips at 20 °C for 60 s was higher on oranges than on mandarins, with GM and BM incidence reductions of up to 60–90 % after 7 days at 20 °C. Irrespective of citrus cultivar and storage condition, SPP generally improved the curative action of 25 μl l?1 IMZ to control Penicillium molds. In additional tests, 100 mM SPP dips at 20 °C for 60 s only prevented GM on ‘Valencia’ oranges inoculated 24 h after treatment when combined with IMZ. It can be concluded that postharvest SPP treatments show promise as an effective alternative to be considered in citrus postharvest disease control programs.  相似文献
