The effects of dietary folic acid on biochemical parameters and gene expression of three heat shock proteins (HSPs) of blunt snout bream (Megalobrama amblycephala) fingerling under acute high temperature stress. Six dietary folic acid groups (0.0, 0.5, 1.0, 2.0, 5.0, and 10.0) mg/kg diets were designed and assigned into 18 tanks in three replicates each (300 l/tank) and were administered for 10 weeks in a re-circulated water system. The fingerlings with an initial weight of 27.0 ± 0.03 g were fed with their respective diets four times daily. At the end of the experiment, samples were collected before challenge, 0, 24, 72 h, and 7 days. Serum total protein (TP), alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), cortisol, glucose, complement C3 (C3), complement C4 (C4, immunoglobulin M (IgM) hepatic superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA), and the expression of heat shock protein 60 (HSP60), 70 (HSP70), and 90 (HSP90) were studied. The results showed that fish fed with dietary folic acid between 1.0, 2.0, and 5.0 mg/kg significantly (P < 0.05) increased serum TP, C3, C4 hepatic SOD, CAT, and the expression of HSP60, HSP70, and HSP90 before and after temperature challenge of 32 °C. Also, serum ALP, cortisol, glucose, and hepatic MDA were significantly (P < 0.05) reduced by supplementation of dietary folic acid level 1.0, 2.0, and 5.0 mg/kg before and after the same temperature challenge of 32 °C. Before stress, 0, 24, 72 h, and 7 days significantly (P < 0.05) affects serum biochemical parameters, immune and antioxidant capacities, and expression level of three HSPs. Furthermore, there was no statistical evidence to show that dietary folic acid inclusion level and temperature duration have significant interactive effect on serum biochemical parameters, antioxidant parameters, and gene expression level (P > 0.05) of the three HSPs. However, there were statistical significant interactive effect between dietary folic acid inclusion level and temperature duration on serum C3 and C4 (P < 0.05) except IgM (P > 0.05). The present results indicate that supplementation of basal diet from 1.0 mg/kg; 2.0 and 5.0 mg/kg can enhance acute high temperature resistance ability in M. amblycephala fingerling to some degree and improve physiological response, immune and antioxidant capacities, and expression level of three HSPs.

The present study was conducted to evaluate the effects of three commercial astaxanthin preparations (100 mg kg?1 diet) with different solubilities in water, from DSM (Dutch State Mines), BASF (Badische Anilin and Soda Fabrik) and Wisdom Company on pigmentation of flesh and antioxidation of flesh, serum and liver in rainbow trout with an initial weight of 52.07 g. After 60 days of feeding, there were no significant differences in growth or flesh proximate composition of rainbow trout among groups (P > 0.05); the Salmo Fan score, redness and astaxanthin content of flesh in rainbow trout fed diets supplemented with astaxanthins were higher than those of the control group (P < 0.05). At 0, 12, 24, 48 and 72 h after thawing, the flesh malondialdehyde (MDA) content of the three astaxanthin groups was lower than that of the control group (P < 0.05). The total antioxidation capacity (T-AOC) of liver in the three astaxanthin groups was significantly higher, but serum catalase (CAT) activities were lower, than that of the control group (P < 0.05). The results indicate that addition of 100 mg kg?1 astaxanthin from DSM, BASF or Wisdom to the diet could improve flesh redness and liver T-AOC, reduce serum CAT, SOD and flesh MDA and extend the shelf life of flesh, in spite of the different solubilities of the three sources in water.

An 8-week feeding experiment was conducted to determine the effect of dietary betaine levels on the growth performance, antioxidant capacity, and lipid metabolism in high-fat diet-fed blunt snout bream (Megalobrama amblycephala) with initial body weight 4.3 ± 0.1 g [mean ± SEM]. Five practical diets were formulated to contain normal-fat diet (NFD), high-fat diet (HFD), and high-fat diet with betaine addition (HFB) at difference levels (0.6, 1.2, 1.8%), respectively. The results showed that the highest final body weight (FBW), weight gain ratio (WGR), specific growth rate (SGR), condition factor (CF), and feed intake (FI) (P < 0.05) were obtained in fish fed 1.2% betaine supplementation, whereas feed conversion ratio (FCR) was significantly lower in the same group compared to others. Hepatosomatic index (HSI) and abdominal fat rate (AFR) were significantly high in fat group compared to the lowest in NDF and 1.2% betaine supplementation, while VSI and survival rate (SR) were not affected by dietary betaine supplementation. Significantly higher (P < 0.05), plasma total cholesterol (TC), triglycerides (TG), low-density lipoprotein (LDL), aspartate transaminase (AST), alanine transaminase (ALT), cortisol, and lower high-density lipoprotein (HDL) content were observed in HFD but were improved when supplemented with 1.2% betaine. In addition, increase in superoxide dismutase (SOD), catalase (CAT), and reduced glutathione (GSH) in 1.2% betaine inclusion could reverse the increasing malondialdehyde (MDA) level induced by HFD. Based on the second-order polynomial analysis, the optimum growth of blunt snout bream was observed in fish fed HFD supplemented with 1.2% betaine. HFD upregulated fatty acid synthase messenger RNA (mRNA) expression and downregulated carnitine palmitoyltransferase 1, peroxisome proliferator-activated receptor α, and microsomal triglyceride transfer protein mRNA expression; nevertheless, 1.2% betaine supplementation significantly reversed these HFD-induced effects, implying suppression of fatty acid synthesis, β-oxidation, and lipid transport. This present study indicated that inclusion of betaine (1.2%) can significantly improve growth performance and antioxidant defenses, as well as reduce fatty acid synthesis and enhance mitochondrial β-oxidation and lipid transportation in high-fat diet-fed blunt snout bream, thus effectively alleviating fat accumulation in the liver by changing lipid metabolism.

This study aimed to investigate the optimum dietary carbohydrate/lipid (CHO/L) ratio for fingerling blunt snout bream Megalobrama amblycephala (average initial weight: 6.61 ± 0.03 g). Six isonitrogenous and isoenergetic diets were formulated to contain various CHO/L ratios ranging from 1.62 to 24.20. Each diet was tested in four replicates for 10 weeks. Specific growth rate (SGR), feed conversion ratio and nitrogen and energy retention all improved significantly (< 0.05) as dietary CHO/L ratios decreased from 24.20 to 5.64 but showed little difference (> 0.05) as dietary CHO/L ratios ranged from 2.45 to 5.64. Hepatosomatic index increased significantly (< 0.05) as dietary CHO/L ratios increased, whereas intraperitoneal fat ratio showed an opposite trend (< 0.05). Opposite to moisture content, lipid content of whole body and carcass all increased significantly (< 0.05) as dietary CHO/L ratios decreased. Liver lipid content showed little difference (> 0.05) among all the treatments, while liver glycogen content increased significantly (< 0.05) with increasing CHO/L ratios. High dietary carbohydrate enhanced the activities of liver hexokinase, pyruvate kinase and glucose‐6‐phosphate dehydrogenase but did not induce hyperglycaemia. Based on the second‐order polynomial regression analysis of SGR, the optimal dietary carbohydrate and lipid contents for fingerling blunt snout bream were 291.7 and 81.4 g kg−1, respectively, with a corresponding dietary CHO/L ratio of 3.58.

Lipid peroxidation, protein oxidation and antioxidant activities of muscle, intestine, hepatopancreas and serum in juvenile Jian carp (Cyprinus carpio var. Jian) were investigated after feeding graded levels of biotin (0.010, 0.028, 0.054, 0.151, 0.330, 1.540 and 2.680 mg kg?1 diet) for 63 days. Both malondialdehyde and protein carbonyl content in all studied tissues and serum were the lowest in fish fed diets containing 0.151–0.330 mg biotin kg?1 diet and then increased in fish fed the diet with 2.680 mg biotin kg?1 diet (P < 0.05). Similarly, glutamate–oxaloacetate transaminase and glutamate–pyruvate transaminase activities in serum significantly decreased with biotin levels up to 0.151 mg kg?1 diet (P < 0.05). Conversely, capacities of anti-hydroxyl radical (AHR) and anti-superoxide anion (ASA) in the detected tissues and serum significantly improved with biotin levels up to 0.054–1.540 mg kg?1 diet and then decreased in 2.680 mg biotin kg?1 diet group for muscle and intestinal AHR as well as hepatopancreas ASA (P < 0.05). Activities of superoxide dismutase in all studied tissues and serum significantly elevated with biotin levels up to 0.330 mg kg?1 diet and then decreased when fish fed the diet with 2.680 mg biotin kg?1 diet, except intestine (P < 0.05). Meanwhile, activities of catalase, glutathione peroxidase, glutathione-S-transferase and glutathione reductase and total thiol content in all studied tissues and serum showed the upward trend with biotin supplementations (P < 0.05). These results indicated that biotin improved antioxidant status and depressed lipid peroxidation and protein oxidation in all studied tissues and serum.

Camelina (Camelina sativa) oil was tested as a replacement for fish oil in diets for farmed Atlantic cod (Gadus morhua). Camelina differs from other plant oilseeds previously used in aquaculture with high lipid (40 %), α-linolenic acid (40 %), antioxidants and low proportions of saturated fats. Dietary treatments were fed to cod (19 g fish?1 initial weight) for 9 weeks and included a fish oil control (FO), 40 % (CO40) and 80 % (CO80) replacement of fish oil with camelina oil. There was no effect of replacing fish oil with camelina oil included at levels up to 80 % on the growth performance. Cod fed CO80 stored more lipid in the liver (p < 0.01), including more neutral lipid (p < 0.05) and triacylglycerol (p < 0.05). Cod fed CO80 decreased in total polyunsaturated fatty acids (PUFAs) in muscle compared to CO40 and FO (p < 0.05), increased in monounsaturated fatty acids (p < 0.01), decreased in total ω3 fatty acids (FO > CO40 > CO80; p < 0.01) and increased in total ω6 fatty acids (FO < CO40 < CO80; p < 0.01). In the liver, long-chain (LC) PUFA such as 20:4ω6, 20:5ω3, 22:5ω3 and 22:6ω3 decreased when fish oil was removed from the diet (p < 0.05), and increased in 18-carbon fatty acids (p < 0.01). Camelina oil can reduce the amount of fish oil needed to meet lipid requirements, although replacing 80 % of fish oil reduced LC PUFAs in both tissues. A comparison of BF3 and H2SO4 as catalysts to transmethylate cod liver and muscle lipids revealed small but significant differences in some fatty acid proportions.

As amino acids (AAs) are vital molecules in the metabolism of all living organisms and are the building blocks of enzymes, a 6-week feeding trial was conducted for determining the influence of dietary essential amino acid (EAA) deficiencies on pancreatic, plasma, and hepatic enzyme activities in silvery-black porgy (initial weight 4.7?±?0.01 g) juveniles. Eleven isoproteic (ca. 47%) and isoenergetic (ca. 20.5 MJ kg?1) diets were formulated including a control diet, in which 60% of dietary nitrogen were provided by intact protein (fish meal, gelatin, and wheat meal) and 40% by crystalline AA. The other 10 diets were formulated by 40% reduction in each EAA from the control diet. At the end of the experiment, fish fed with threonine-deficient diet showed the lowest survival rate (P?<?0.05), whereas growth performance decreased in fish fed all EAA-deficient diets, although the reduction in body growth varied depending on the EAA considered. Pancreatic enzymes (trypsin, lipase, α-amylase, and carboxypeptidase A) activities significantly decreased in fish fed the EAA-deficient diets in comparison with fish fed the control diet (P?<?0.05). Fish fed with the arginine-deficient diet had the highest plasma and liver alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase levels (P?<?0.05). Plasma and liver lactate dehydrogenase and superoxide dismutase showed the highest and lowest values, respectively, in fish fed the arginine and lysine-deficient diets (P?<?0.05). Plasma metabolites were significantly affected by dietary EAA deficiencies (P?<?0.05). The results of this study suggesting dietary EAA deficiencies led to reduction in growth performance as well as pancreatic and liver malfunction. Furthermore, arginine and lysine are the most limited EAA for digestive enzyme activities and liver health in silvery-black porgy.

An 8‐week experiment on fingerling black carp Mylopharyngodon piceus was conducted to evaluate the effects of dietary fish oil (FO) supplement on growth, fatty acid composition and non‐specific immunity responses. Five triplicate fingerling groups (initial weight = 2.72 ± 0.35 g) were fed isoenergetic and isonitrogenous diets in which the dietary FO was replaced with rapeseed oil (RO) in graded increments of 25% (0–100%). No significant effects were observed on specific growth rates, survival rates and feed conversion ratios, but there were significant differences in whole body moisture and liver lipid contents (P < 0.05), and the 100% RO replacement diet significantly enhanced hepatosomatic indexes compared to control group (P < 0.05). Other approximate whole body constituents, viscerasomatic ratios and condition factors were not influenced by dietary oil treatments. Fatty acid composition of muscle and liver was influenced by dietary fatty acid input, α‐linoleic acid and γ‐linolenic acid were significantly increased with increasing RO, but eicosapentaenoic acid, docosahexaenoic acid and the n‐3/n‐6 ratio were significantly reduced (P < 0.05). Alternative complement pathway, lysozyme and superoxide dismutase activities were not significantly influenced. These results indicate that black carp fed diets with FO supplement had similar growth and non‐specific immunity to the fish fed diet with RO.

In this study we examined the effects of high-fat diets on alterations in liver histology features and blood biochemistry parameters in blunt snout bream Megalobrama amblycephala. Fish were fed three diets containing 5, 10, and 15 % fat, respectively, for 6 weeks prior to the liver histology examination and blood biochemistry testing. The livers of fish fed the 5 % fat diet showed a normal structure, whereas those of the fish fed the diets with a higher fat content presented nucleus polarization and lipid vacuolization. Transmission electron microscopy examination revealed that the hepatocytes of fish fed the 15 % fat diet had undergone ultrastructural alterations involving the mitochondria, nucleus, and endoplasmic reticulum (ER). Alterations in the blood biochemistry were evaluated to determine whether the blood biochemistry changes could be correlated with the observed alterations in liver histology with the aim of assessing the suitability of the investigated blood parameters as rapid indicators of liver condition. Significantly higher levels of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities and of triglycerides and cholesterol were found in fish fed the 15 % fat diet. On the whole, the results clearly show that high fat intake resulted in fat accumulation and ultrastructural impairments of the mitochondria, nucleus, and ER. We conclude that blood sample measurements (AST, ALT, triglycerides, and cholesterol) could be used as a rapid test for determining liver status in blunt snout bream.

Lipoic acid (LA) is an antioxidant that also favors glucose uptake in mammals. Until now, there are no studies evaluating the potential effect of this molecule on glycemic control in fish. It was evaluated LA effects on glucose uptake in common carp Cyprinus carpio fed with carbohydrate diets from two carbohydrate sources: glucose (GLU) and starch (STA), and supplemented or not with LA, being the diets: +GLU/?LA (GLU); +GLU/+LA (GLU + LA); +STA/?LA (STA); and +STA/+LA (STA + LA). Carp juveniles (6.5 ± 0.1 g) were fed with each diet ad libitum 4 times a day, during 68 days. Muscle glycogen concentration was higher (p < 0.05) in GLU and GLU + LA than in STA and STA + LA groups. On fish fed with starch, muscle cholesterol and triglyceride concentrations were higher (p < 0.05) in fish fed diets supplemented with LA. Muscle protein levels were higher in fish fed with LA, independent of the diet carbohydrate source. Lipid peroxidation was significantly reduced (p < 0.05) in fish muscle on fish fed the STA + LA diets when compared with the STA diet. Our findings indicate that LA modulates lipid, proteins and carbohydrate metabolism together with the well-known antioxidant effect. Also, LA showed to enhance starch utilization taking into account muscle cholesterol and triglyceride levels.

The main indication of cimetidine is being H2-receptor antagonist, but studies suggest that cimetidine may also act as a non-specific stimulant of cell-mediated immunity and immunomodulator. In order to determine the immunomodulatory effect of dietary intake of cimetidine in the common carp (100 ± 10 g), subjects were fed diets containing 0 (control), 50, 100 and 200 mg cimetidine kg?1 of dry diet for a period of 6 weeks. TLC and NBT assays were significantly (P < 0.05) stimulated in cimetidine-supplemented groups displaying the highest value in 200 mg kg?1 group. A decrease (P < 0.05) in cortisol and ACH50 value was recorded in fish treated with cimetidine. Serum protein, albumin and serum globulin levels were not significantly changed. The findings of the present investigation suggest that the incorporation of cimetidine in the diet of common carp enhances the non-specific immunity.

An 8-week feeding trial was conducted to determine the dietary arginine requirement of fingerling Indian catfish, Heteropneustes fossilis (6.25 ± 1.30 cm, 4.8 ± 0.65 g). Six isonitrogenous (400 g Kg?1) and isoenergetic (17.90 kJ g?1) amino acid test diets were formulated with gradation of 2.5 g Kg?1 containing graded levels of l-arginine (8.5–21.0 g Kg?1, dry diet). Fish were randomly stocked in triplicate groups, in 75-l circular trough with flow-through system and fed experimental diets at 4 % BW/day at 0800 and 1800 h. Maximum live weight gain (277 %), best feed conversion ratio (FCR) (1.52) and protein efficiency ratio (PER) (1.64) were obtained in fish fed diet containing 16.0 g Kg?1 arginine. However, quadratic regression analysis of live weight gain, FCR, PER and body protein deposition (BPD) data indicated requirements for dietary arginine at 16.80, 16.30, 16.11 and 16.10 g Kg?1 of dry diet, respectively. Significantly (p < 0.05) higher whole body protein content, minimum carcass moisture and intermediate carcass fat contents were recorded at 16.0 g Kg?1 dietary arginine diet. Ash content remained insignificantly (p > 0.05) low among all the treatments except at diet I and diet II. Based on the above results, it is recommended that the diet for young H. fossilis should contain arginine at 16.32 g Kg?1, dry diet, corresponding to 40.80 g Kg?1 dietary protein for optimum growth and efficient feed utilization.

In the present study, konjac mannanoligosaccharide (KMOS) was evaluated as a prebiotic in yellow catfish. The fish were fed with diets containing KMOS in four concentrations: 0 g kg?1 (C), 1.0 g kg?1 (KM1), 2.0 g kg?1 (KM2), and 3.0 g kg?1 (KM3) for 49 days, respectively. Another group fed with diets containing 3.0 g kg?1 yeast cell wall mannanoligosaccharide (MOS) (M3) was set as positive control. The results indicated that fish receiving the diets supplemented with KMOS or MOS showed higher relative gain rate (RGR), specific growth rate (SGR), and lower feed conversion ratio (FCR) with significantly differences (P < 0.05) than those fed with the basal diets. Moreover, fish receiving the diets with 2.0 g kg?1 KMOS inclusion showed higher RGR, SGR, and lower FCR (P < 0.05) than that feeding the diets supplemented with 3.0 g kg?1 MOS. The quantities of Bifidobacterium spp. were significantly increased (P < 0.05). Meanwhile, Escherichia coli and Aeromonas spp. were significantly reduced (P < 0.05) in the fish-feeding diets with 2.0 g kg?1 KMOS supplement. Compared with the control group, the significantly enhancement of protease and amylase activity (P < 0.05) in intestine and pancreas was observed in fish fed with diets containing KMOS or MOS. Collectively, an optimum level of KMOS inclusion in diets could modulate intestinal microflora, induce digestive enzyme activity, and improve the growth performance of yellow catfish significantly.

High-fat diets may have favorable effects on growth and cost, but high-fat diets often induce excessive fat deposition, resulting in liver damage. This study aimed to identify the hepatoprotective of a Chinese herb (berberine) for blunt snout bream (Megalobrama amblycephala). Fish were fed with a normal diet (LFD, 5 % fat), high-fat diet (HFD, 15 % fat) or berberine-supplemented diets (BSD, 15 % fat with berberine 50 or 100 mg/kg level) for 8 weeks. After the feeding, histology, oxidative status and mitochondrial function of liver were assessed. The results showed that HFD caused fat accumulation, oxidative stress and apoptosis in hepatocytes of fish. Hepatocytes in HFD group appeared to be hypertrophied, with larger liver cells diameter than these of LFD group. Berberine-supplemented diets could attenuate oxidative stress and hepatocytes apoptosis. HFD induced the decreasing mitochondrial complexes activities and bulk density and surface area density. Berberine improved function of mitochondrial respiratory chain via increasing the complex activities. Moreover, the histological results showed that berberine has the potential to repair mitochondrial ultrastructural damage and elevate the density in cells. In conclusion, our study demonstrated that berberine has attenuated liver damage induced by the high fat mainly via the protection for mitochondria.

An 8‐wk feeding trial was conducted to evaluate the influence of different proportions of dietary plant ingredients on the growth of juvenile blunt snout bream, Megalobrama amblycephala. A 2 × 3 factorial arrangement was set for this trial with three ratios of cottonseed meal (CSM) and canola meal (CM) (2:1, 1:1, or 1:2) and three proportions of soybean meal (SBM) (2, 1, or 0). The results showed that the specific growth rate and protein efficiency ratio both significantly decreased with decreasing amounts of SBM in the diets of blunt snout bream (P < 0.05), while the feed efficiency ratio and feed intake increased significantly (P < 0.05). Meanwhile, the growth performance and feed utilization were shown to improve with increasing amounts of CSM, although the effects were not significant (P > 0.05). The interaction of these two factors had no effect on either growth or feed utilization (P > 0.05). Diet composition was not seen to affect body composition (P > 0.05). The ratio of CSM to CM significantly affected gut amylase activity, liver alanine aminotransferase content, and plasma cholesterol levels (P < 0.05). The interaction between the two main effects significantly influenced the plasma glucose level (P < 0.05). Therefore, the suitable proportion of plant feed for blunt snout bream should include high levels of SBM and CSM, with a SBM : CSM : CM ratio of 2:2:1.

Anaerobically digested piggery waste was used as a substitute protein source at 10% and 20% levels on an isonitrogenous and (as far as possible) isocalorific basis, and compared to a control diet of soyabean meal and fish meal for fingerling mirror carp (Cyprinus carpio). The fish fed on the substitute diets showed significantly higher (P < 0.05) specific growth rates, but the fish on the 20% diet had significantly lower (P < 0.05) fat contents, and significantly higher (P < 0.05) ash contents compared to both the control and the 10% diet, at the end of the experiment. Raw animal wastes do not perform well when substituted for conventional protein sources in fish diets. With processing, the performance is improved. Anaerobic digestion seems to be an appropriate processing technique for the upgrading of piggery wastes. A substitution level of 20% of the processed waste for conventional materials may be obtained without adversely affecting growth performance.

The effects of three artificial diets (S1, S2 and S3) on survival, growth and biochemical composition of one-year-old pearl oyster Pinctada martensii were investigated. Six experimental groups (EG1, EG2, EG3, EG4, EG5 and EG6) and one control group (CG) were set up. EG1, EG2 and EG3 were solely fed on S1, S2 and S3, respectively. EG4, EG5 and EG6 were fed on mixed diets, as follows: S1 and Platymonas subcordiformis; S2 and P. subcordiformis; and S3 and P. subcordiformis, respectively. CG was fed on only P. subcordiformis. All groups were continuously fed for 60 days. Survival, growth and biochemical composition of soft tissues were compared across the groups. Results showed that survival rate, the absolute growth rate (AGR) and relative growth rate (RGR) of shell length did not differ significantly across the groups (p > 0.05). The AGR and RGR of total weight differed significantly among the groups (p < 0.05). AGR and RGR of shell length and total weight were the highest in EG5 and the lowest in EG1. Gross fat content showed insignificant differences among the groups (p > 0.05). However, gross protein content and ash content showed significant differences across the groups (p < 0.05). The gross protein of the groups solely fed on artificial diets was lower than those of the groups fed on mixtures of artificial diets and microalgae or single microalgae. The contents of other amino acids, total amino acids (TAAs) and essential amino acids (EAAs) showed significant differences across the groups (p < 0.05). TAA, EAA and delicious amino acids of the groups fed solely on artificial diets were lower than those of the groups fed on mixtures of artificial diets and microalgae or single microalgae. Results indicated that the artificial diet (S2) can serve as substitutes of microalgal diets for P. martensii.
