AIM: To detect the expression of WNT5B in normal breast epithelial cells and different breast cancer cell lines, and to investigate the effects of WNT5B over-expression on the viability and apoptosis of human breast cell line MCF-7.METHODS: The mRNA expression of WNT5B was detected by RT-PCR in different breast cancer cells. MCF-7 cells were transfected with plasmid pcDNA3.1/WNT5B or pcDNA3.1, and the expression of WNT5B at mRNA and protein levels was examined in the 2 groups by real-time PCR and Western blotting, respectively. Subsequently, the changes of cell viability and cell apoptosis were analyzed by CCK-8 assay and flow cytometry, respectively. RESULTS: The expression of WNT5B in the breast cancer cell lines was lower than that in MCF10A cells. The WNT5B expression in the MCF-7 cells in experimental group was significantly higher than that in vector group (P<0.05). However, the cell viability in experimental group decreased significantly as compared with vector group (P<0.05). The number of the cells in S-phase obviously increased, while the percentage of the cells in G1-phase and G2/M-phase decreased compared with vector group. The number of apoptotic cells in WNT5B group was significantly higher than that in vector group.CONCLUSION: The expression of WNT5B is decreased in breast cancer cells. WNT5B over-expression significantly inhibits the cell growth and promotes the cell apoptosis in breast cancer MCF7 cells.  相似文献

AIM:To evaluate the expression of SAL-like 4 (SALL4) protein in human prostate cancer cell lines and tissues, and to analyze the relationship between SALL4 expression and the clinicopathological parameters. METHODS:Immunofluorescence, RT-PCR and Western blotting were performed to detect the expression of SALL4 at mRNA and protein levels in 3 common prostate cancer cell lines LNCaP, DU145 and PC-3. The normal prostate epithelial cell line RWPE-1 was used for control. The protein levels of SALL4 in the tissues of benign prostate hyperplasia and prostate cancer tissues were determined by the method of immunohistochemistry. RESULTS:The SALL4 protein was predominantly expressed in the cytoplasm of the cells. The protein levels of SALL4 in 3 common prostate cancer cell lines were significantly higher than that in RWPE-1 cells. However, the mRNA level of SALL4 had no obvious difference among the 4 cell lines. Immunohistochemistry results showed that the expression level of SALL4 in the cancerous tissues was significantly higher than that in noncancerous (benign and normal) prostatic tissues. In addition, we found that the expression level of SALL4 in prostate cancer was significantly correlated with the Gleason score, clinical stage, prognosis estimation and tissue prostate-specific antigen (PSA) expression, but not associated with age, the level of serum total PSA, prostate volume and the expression of androgen receptor in the tissues of the patients. CONCLUSION: The over-expression of SALL4 protein may play an important role in the pathogenesis and progression of prostate cancer, and provides some reference indexes for estimating the malignancy, progression and prognosis of prostate cancer.  相似文献

AIM: To explore the role of Golgi mannosidase Ⅱ(GMⅡ) in the development of gastric carcinoma by analysis of the relationship between differential expression of GMⅡ and differentiation of gastric carcinoma cell lines and tissues. METHODS: Thirty cases of human normal gastric tissues and 38 cases of gastric adenocarcinoma tissues were selected. Three different differentiated gastric carcinoma cell lines (MKN-28, SGC-7901 and BGC-823) and a normal gastric epithelial cell line GES-1 were cultured in vitro. The mRNA levels of GMⅡ were detected by RT-PCR, and the protein expression was detected by immunohistochemistry and Western blotting. RESULTS: GMⅡ was mainly distributed in cytoplasm. The positive rates of GMⅡ in 30 cases of human normal gastric tissues, 8 cases of well-differentiated, 18 cases of moderately-differentiated and 12 cases of poorly-differentiated gastric cancer tissues were 53% (16/30), 63% (5/8), 83% (15/18) and 100% (12/12), respectively. The expression of GMⅡ was gradually increased in normal gastric epithelial cell line and in well, moderately and poorly-differentiated gastric cancer cell lines by cell-attached coverslip. Compared with normal gastric epithelial cell line, 3 gastric carcinoma cell lines showed the higher expression of GMⅡ at mRNA and protein levels (P<0.05). Furthermore, GMⅡ expression in poorly-differentiated gastric carcinoma cell line BGC823 was the highest, and the lowest expression of GMⅡ was the well-differentiated cell line MKN-28. Compared with normal gastric epithelial tissues, gastric carcinoma tissues showed the higher expression of GMⅡ at mRNA and protein levels (P<0.05), and the highest was the poorly-differentiated carcinoma tissues. The expression of GMⅡ at mRNA and protein levels in normal gastric tissues was the lowest. CONCLUSION: GMⅡ is involved in the development and progression of gastric cancer. The expression of GMⅡ is highly related to the poorly-differentiated gastric cancer.  相似文献

AIM: To investigate whether gonadotropin-releasing hormone analogue (GnRHa) affect the sensitivity of breast cancer cells to 5-FU and epirubicin in vitro. METHODS: Two breast cancer cell lines (MCF-7 and MDA- MB-231) were treated with different concentrations of GnRH analogue, triptorelin acetate, or with a GnRHa+5-FU or GnRHa+epirubicin. The cellular growth profiles were determined by CCK-8. The mRNA levels of GnRH receptor, PCNA and MDR1 were measured by RT - PCR. RESULTS: Both cell lines had positive GnRH receptor mRNA expression detected by RT- PCR. GnRHa did not suppress cell growth after GnRHa exposure. IC50 of 5-FU and epirubicin was not changed in the presence of GnRHa. Suppression of cell growth by the exposure to 5-FU and epirubicin was not changed in the presence of GnRHa. GnRHa treatment up-regulated PCNA mRNA expression in MDA-MB-231 cells but not in MCF-7 cells. The expression of MDR1 mRNA was down-regulated by GnRHa in MCF-7 cell lines. No MDR1 mRNA expression in MDA-MB-231 cells was observed. CONCLUSION: The present data suggest that GnRH analogue (triptorelin acetate) does not affect the sensitivity of breast cancer cell lines MCF-7 and MDA-MB-231 to 5-FU and epirubicin. GnRHa may decrease the drug resistance by down-regulating MDR1 mRNA expression.  相似文献

AIM: To study the effect of histone deacetylase 1 (HDAC1) on the apoptosis of breast cancer cells.METHODS: The expression of HDAC1 at mRNA and protein levels in normal mammary epithelial cell line MCF-10A and breast cancer cell lines BT549, MCF-7 and MDA-MB-231 was measured by RT-qPCR and Western blot. HDAC1 siRNA was transfected into MDA-MB-231 cells, and then RT-qPCR and Western blot were used to determine the expression level of HDAC1. The cell viability was measured by MTT assay, and apoptosis was analyzed by flow cytometry. The protein levels of β-catenin, c-Myc, cyclin D1 and cleaved caspase-3 were determined by Western blot. Breast cancer cells with HDAC1 knockdown were treated with Wnt/β-catenin signaling pathway activator, and then the cell viability and apoptosis were measured.RESULTS: The expression of HDAC1 at mRNA and protein levels in BT549, MCF-7 and MDA-MB-231 cells was significantly higher than that in normal mammary epithelial cell line MCF-10A, and the highest expression level of HDAC1 was observed in MDA-MB-231 cells (P<0.05). HDAC1 siRNA reduced the expression of HDAC1 at mRNA and protein levels in the breast cancer cells. The viability of MDA-MB-231 cells was decreased after knockdown of HDAC1 expression, the apoptotic rate was increased, the protein level of cleaved caspase-3 in the cells was elevated, and the protein levels of β-catenin, c-Myc and cyclin D1 were decreased (P<0.05). Wnt/β-catenin signaling pathway activator reversed HDAC1 knockdown-induced apoptosis and decrease in viability of MDA-MB-231 cells, and reduced the protein level of cleaved caspase-3.CONCLUSION: Knockdown of HDAC1 expression induces apoptosis of breast cancer cells by inhibiting the activation of Wnt/β-catenin signaling pathway.  相似文献

AIM: To detect the expression level of wip1 in lung cancer tissue and three lung cancer cell lines, and to explore the relations between the expression level of Wip1 in lung cancer and various clinical and pathological features. METHODS: Real-time PCR was employed to detect the expression of Wip1 mRNA in 44 specimens of non small cell lung cancer tissues and normal tissues. The relations between the expression of Wip1 mRNA and various clinical and pathological features were analyzed. Real-time PCR was also employed to detect the expression of Wip1 mRNA in A549, NCI-1299, NCI-H460 and HBE for relative quantitative analysis.RESULTS: In the 44 specimens, the expressions of Wip1 mRNA in both cancer tissues and normal lung tissues were positive. Wip1 gene was over-expressed in 17 specimens in 44 non small cell lung cancer specimens. The rate was 38.6%. The relative level of Wip1 mRNA in NSCLC tissues was significantly higher than that in normal lung tissues (ratio=2.1644±1.3940, P<0.01). The expression of Wip1 mRNA was also correlated with pathological grading (P<0.05). The relative level of Wip1 mRNA in three kinds of lung cancer cells was significantly higher than that in HBE cells. The difference was statistically significant (P<0.05).CONCLUSION: The Wip1 mRNA is over-expressed in non small cell lung cancer, indicating that Wip1 is related to the tumorigenesis and may become the new target of non small cell lung cancer gene therapy. The expression of Wip1 mRNA is related to tumor cell differentiation and may use for the molecular biological reference index to estimate the malignant degree of cancer.  相似文献

AIM To investigate the inhibitory effect of protocadherin 10 (PCDH10) on proliferation of human breast cancer cells and its possible mechanism. METHODS RT-PCR was used to measure the mRNA expression levels of PCDH10 in 4 human breast cancer cell lines and normal mammary epithelial MCF-10A cells. The breast cancer cell line MDA-MB-231 with stable PCDH10 overexpression was constructed by recombinant lentivirus (pLV-PCDH10) infection, and blank control (blank) group and negative control (pLV-NC) group were also set up. The cell proliferation ability was detected using methyl thiazolyl tetrazolium (MTT) and colony formation experiments. The cell cycle was detected by flow cytometry. Western blot was used to determine the expression of cyclin D1, cyclin-dependent kinase 4 (CDK4), nucear factor-κB p65 subunit (NF-κB p65) and NF-κB inhibitor α (IκBα). RESULTS The mRNA expression levels of PCDH10 in 4 breast cancer cell lines were lower than that in normal mammary epithelial MCF-10A cells (P<0.05). A breast cancer cell line MDA-MB-231 with stable PCDH10 overexpression was constructed successfully. Compared with negative control group, PCDH10 overexpression significantly inhibited cell proliferation, induced cell cycle arrest at G1 phase, down-regulated the expression of cyclin D1 and CDK4, and decreased phosphorylation of NF-κB p65 and IκBα(P<0.05). CONCLUSION PCDH10 inhibits the proliferation and blocks cell cycle progression of breast cancer cells by targeting NF-κB/cyclin D1 signaling pathway.  相似文献

AIM: To investigate the expression and clinical significance of PAK4 in the cell lines and tissues of non-small cell lung cancer (NSCLC). METHODS: PAK4 expression in human bronchial epithelial (HBE) cells, NSCLC cell lines, NSCLC tissues and adjacent non-tumor tissues were assessed by immunohistochemistry, real-time PCR and Western blot. Prognostic value of PAK4 expression was evaluated by Kaplan-Meier analysis and Cox regression. RESULTS: PAK4 was over-expressed in the NSCLC cell lines at both mRNA and protein levels compared with HBE cells (P<0.05). PAK4 was over-expressed in the NSCLC tissues at both mRNA and protein levels compared with adjacent non-tumor tissues (P<0.05). PAK4 was over-expressed in the metastatic NSCLC tissues compared with the primary NSCLC tissues (P<0.05). Higher PAK4 staining scores were positively correlated with differentiation, lymph node metastasis, distant metastasis, and clinical stage. Kaplan-Meier analysis and log-rank test showed that overall survival was significantly different between the patients with up-regulated PAK4 and the patients with down-regulated PAK4(P<0.05). PAK4 over-expression was associated with NSCLC progression.CONCLUSION: Increased PAK4 expression was associated with tumor invasion, metastasis and prognosis in the patients with NSCLC. PAK4 is an important prognostic marker and potential therapeutic target in NSCLC.  相似文献

AIM: To explore the effect of ribonucleotide reductase M2 (RRM2) gene knockdown by siRNA on the proliferation and migration of human breast cancer MCF-7 cells and the tumor growth in BALB/c nude mice. METHODS: The mRNA and protein expression leves of RRM2 in human breast cancer cell line MCF-7 and human normal breast cell line MCF-10A were determined by real-time PCR and Western blotting. siRNA-RRM2 was constructed and transfected into MCF-7 cells at different time points and different concentrations. The silencing efficiency of RRM2 gene was detected by real-time PCR. The cell proliferation was measured by CCK-8 assay. The migration was observed using Transwell cell migration system. The effect of siRNA-RRM2 on the tumor growth was determined in nude mice. RESULTS: The mRNA and protein levels of RRM2 were higher in MCF-7 cells than those in MCF-10A cells. siRNA-RRM2 down-regulated the expression of RRM2 in MCF-7 cells in a time-and concentration-dependent manner. The results of CCK-8 assay showed that siRNA-RRM2 inhibited the proliferation ability of MCF-7 cells, but not that of MCF-10A cells. The results of Transwell assay indicated that siRNA-RRM2 inhibited the migration ability of MCF-7 cells. siRNA-RRM2 also inhibited the tumor growth in nude mice. CONCLUSION: RRM2 overexpression is associated with the breast cancer proliferation and migration. Suppression of RRM2 function is a potential therapeutic strategy for treating breast cancer.  相似文献

AIM:To investigate the expression of KDM5B gene in breast cancer tissues and its relationship with clinical data and prognosis of the patients. METHODS:Data sets of breast cancer were collected from The Cancer Genome Atlas (TCGA) database, and KDM5B mRNA expression profiles were downloaded. The mRNA expression of KDM5B in breast cancer tissues and adjacent tissues was detected by real-time PCR. The cases were divided into high expression group and low expression group according to the median expression of KDM5B, and the relationship with clinical data and case characteristics were analyzed. The relationship between KDM5B and prognosis of breast cancer patients was analyzed by Kaplan-Meier plotter. RESULTS:The expression of KDM5B in breast cancer tissues was significantly higher than that in normal breast tissues (P<0.01). In TCGA breast cancer data, the expression of KDM5B was significantly correlated with human epidermal growth factor receptor 2 (HER2), estrogen receptor (ER), age, histopathological type and lymph node metastasis (P<0.01), but not with progesterone receptor (PR), menopause and distant metastasis. The expression of KDM5B was significantly correlated with HER2, age and lymph node metastasis, but not with ER, PR, menopause, pathological type and distant metastasis. The higher the expression of KDM5B, the shorter the total survival time and the disease-free survival time of breast cancer patients. CONCLUSION:KDM5B is over-expressed in breast cancer tissues and correlated with prognosis of the patients. KDM5B expression is significantly correlated with HER2, age and lymph node metastasis. KDM5B may play an important role in the development of breast cancer.  相似文献

AIM: To investigate the effect of microRNA-200a (miR-200a) on the malignant biological beha-viors of breast cancer cells and its regulatory mechanism. METHODS: The expression of miR-200a in human breast can-cer cell lines MDA-MB-231, MDA-MB-468 and MCF-7, and normal human mammary epithelial cell line MCF-10A was detected by RT-qPCR. CCK-8 assay was used to detect the viability of MDA-MB-231 cells after transfection with miR-200a mimic or miR-200a inhibitor. Flow cytometry method and Transwell assay were used to detect the apoptosis and invasive ability of MDA-MB-231 cells after transfection with miR-200a mimic or miR-200a inhibitor. The expression of SIP1, E-cadherin, N-cadherin, Snail, Twist, ZEB1 and ZEB2 at mRNA and protein levels was determined by RT-qPCR and Western blot. RESULTS: Compared with MCF-10A cells, the lowest expression of miR-200a was observed in the MDA-MB-231 cells (P<0.05). Over-expression of miR-200a attenuated the viability of MDA-MB-231 cells (P<0.05), increased apoptosis (P<0.05) and decreased the invasion ability (P<0.05). The expression of SIP1, N-cadherin, Snail, Twist, ZEB1 and ZEB2 at mRNA and protein levels was also significantly down-regulated, while the mRNA and protein expression of E-cadherin was significantly increased (P<0.05). Transfection with miR-200a inhibitor reversed the above results. CONCLUSION: Up-regulation of miR-200a inhibits the viability and invasion ability of MDA-MB-231 cells and promotes the apoptosis of MDA-MB-231 cells. miR-200a may regulate the biological behaviors of breast cancer by inhibiting epithelial-mesenchymal transition.  相似文献

AIM: To investigate the clinicopathological significance of the protein expression of phosphorylated ezrin at threonine 567 (pEZRThr567) in lung squamous cell carcinoma, adjacent tissues and normal tissues. METHODS: pEZRThr567 protein was detected in lung squamous carcinoma, adjacent and normal tissues by the method of immunohistochemistry. The correlation of pEZRThr567 expression with clinicopathological parameters of lung squamous carcinomas was also analyzed. The localization of pEZRThr567 was detected by immunofluorescence staining in lung squamous cell line EBC-1. RESULTS: The protein expression of pEZRThr567 in lung squamous carcinoma was significantly higher than that in the adjacent and normal lung tissues (P<0.01). pEZRThr567 mainly localized on the cell membrane, and its over-expression signi-ficantly correlated with the differentiation, clinical stage and lymph node metastasis in lung squamous carcinoma. CONCLUSION: pEZRThr567 may be an effective biomarker for prediction of malignant potential and poor prognosis of lung cancer.  相似文献

AIM: To investigate the expression of CUG-binding protein 1 (CUGBP1) in breast cancer tissues, and to explore the effect of CUGBP1 gene silencing on the viability and invasion ability of human breast cancer MCF-7 cells. METHODS: A total of 96 cases of patients with breast cancer undergoing surgical treatment were selected in the Second Affiliated Hospital of Zhengzhou University from March 2015 to September 2017. Immunohistochemical staining was used to detect the protein expression of CUGBP1 in the breast cancer and adjacent tissues. MCF-7 cells were cultured and divided into CUGBP1 interference sequence group, control sequence group and blank group. Western blot was used to detect the protein expression of CUGBP1, Twist, E-cadherin and vimentin in the cells. The cell viability was measured by MTT assay. The cell invasion ability was detected by Transwell assay. RESULTS: The positive expression rate of CUGBP1 protein in the breast cancer tissues was higher than that in the adjacent tissues (χ2=28.900, P<0.001). The differences of CUGBP1 protein expression in the breast cancer tissues among TNM staging, histological grading and lymph node metastasis were statistically significant (P<0.05). The relative protein expression levels of CUGBP1, Twist and vimentin in CUGBP1 interference sequence group were lower than those in control sequence group and blank group, while the relative protein expression of E-cadherin was higher than that in control sequence group and blank group (P<0.05). The cell viability at 24 h, 48 h, 72 h and 96 h in CUGBP1 interference sequence group was lower than that in control sequence group and blank group (〖P<0.05). The invasive cells in CUGBP1 interference sequence group were less than those in control sequence group and blank group (P<0.05). CONCLUSION: CUGBP1 protein is highly expressed in the breast cancer tissues. Specific silencing of 〖STBX〗CUGBP1〖STBZ〗 gene expression in breast cancer MCF-7 cells effectively inhibits the cell viability and invasiveness, and its mechanism may be related to inhibiting the process of epithelial-mesenchymal transition.  相似文献

AIM: To investigate the expression of MAT1 protein in pancreatic cancers and the relationship between MAT1 and clinicopathological features of pancreatic cancers. METHODS: 94 surgical specimens, including 70 pancreatic cancers, 10 pancreatic benign tumors, 14 chronic pancreatitis and 10 autopsy normal pancreas tissues, were analyzed immunohistochemically, and then MAT1 expression and clinicopathological features were compared. RESULTS: MAT1 was expressed mainly in the cancer cells,and also in the fibroblasts, where it was localized within the cytoplasm and nuclear envelope. MAT1 expression was found in 75.7% (53/70) of the cancers, but not detected or weakly expressed in control tissues. There was a significant difference in expression of MAT1 among the above four tissues (P<0.01). Furthermore, MAT1 expression showed a significant correlation with TNM stage (P<0.05), but no correlation with other clinicopathological features. CONCLUSION: The MAT1 protein that is up-regulated in the pancreatic cancer may involve in pancreatic carcinogenesis, and MAT1 in primary lesion may be an useful marker for evaluating prognosis in patients with pancreatic cancer.  相似文献

AIM: To evaluate the expression of Wnt/β-catenin signaling pathway-related proteins in breast cancer and the significance. METHODS: The patients with breast cancer (n=150) in our hospital from January 2015 to January 2017 were selected as study object. The tumor tissue samples of these patients were obtained from paraffin section of breast cancer by surgical resection with complete clinicopathological data. The corresponding paracancerous tissue sam-ples were taken from the non-tumor tissue samples from the above breast cancer patients, which were 0.5~1 cm away from the tumor tissue. The methods of real-time PCR and Western blot were performed to examine the expression of Wnt-1 and β-catenin at mRNA and protein levels. Human breat cancer MCF-7 cells were divided into 3 groups:control group (MCF-7 cells without treatment), agonist group[MCF-7 cells+Wnt3a (1 mg/L)] and antagonit group[MCF-7 cells+DKK1 (16 μmol/L)]. The expression of Wnt-1 and β-catenin at mRNA and protein levels was detected by real-time PCR and Western blot. RESULTS: Compared with the paracancerous tissues, the expression levels of Wnt-1 and β-catenin were higher in tumor tissues at mRNA and proteins levels (P<0.05). Notably, the positive expression rates of Wnt-1 and β-catenin were significantly higher in tumor tissues than that in the paracancerous tissues. Furthermore, Wnt-1 expression was associated with tumor metastasis (χ2=5.352, P=0.021), tumor stage (χ2=9.412, P=0.002) and tumor size (χ2=9.412, P=0.002). In addition, β-catenin expression was also associated with tumor metastasis (χ2=9.851, P=0.002) and tumor stage (χ2=5.661, P=0.017). Compared with control group, the expression of Wnt-1 and β-catenin at mRNA and protein levels in agonist group was increased (P<0.05),while that in antagonist group was decreased (P<0.05). CONCLUSION: The expression levels of Wnt-1 and β-catenin related with Wnt/β-catenin signaling pathway are increased in the breast cancer, which are closely related to the malignant state of the tumor.  相似文献

AIM:To investigate the expression of miR-183 in breast cell lines and tissue specimens and its effects on the biological behaviors of breast cancer cells. METHODS:Human breast cell lines and clinical tissue specimens of breast diseases were used in the study. The expression of miR-183 was determined by real-time PCR. The characteristics of cell proliferation, invasion and migration were examined after miR-183 was transfected by lipofection. RESULTS:Altered expression of miR-183 was found in the highly invasive breast cancer cells as compared with weakly invasive breast cells. The expression of miR-183 was also significantly decreased in the breast cancer tissues as compared with that in the benign breast disease tissues. The capacities of breast cancer cell invasion and migration were significantly increased after transfection of miR-183 inhibitor and were decreased after transfection of miR-183 mimic. No significant change of cell proliferation was observed after miR-183 transfection. CONCLUSION:miR-183 may play an important role in breast cancer progression, especially in the cell invasion and migration.  相似文献

AIM: To investigate the expression of sodium channel modifier 1 (SCNM1) in hepatitis B-related hepatocellular carcinoma (HCC) and its relationship with clinicopathological features and prognosis.METHODS: The specimens were collected from 108 patients with hepatitis B-related HCC who were treated in the Third Affiliated Hospital of Sun Yat-sen University from January 2013 to December 2015. All patients signed the informed consent and met the requirements of medical ethics. The mRNA expression level of SCNM1 in hepatitis B-related HCC tissues and tumor-adjacent tissues was detected by RT-qPCR, and the relationship between the mRNA expression of SCNM1 and the clinicopathological characteristics of hepatocellular carcinoma was analyzed. The relationship between SCNM1 expression and the prognosis of the patients was analyzed by Kaplan-Meier plotter.RESULTS: The data from TCGA database, Human Protein Atlas database and Oncomine database showed that the expression of SCNM1 in hepatocellular carcinoma tissues was significantly higher than that in the normal liver tissues (P<0.01). SCNM1 was mainly distributed in the nucleus. The results of RT-qPCR showed that the median mRNA expression of SCNM1 in hepatitis B-related HCC tissues was significantly higher than that in the matched tumor-adjacent tissues (t=8.082, P<0.01). The mRNA expression of SCNM1 was correlated with cirrhosis, alanine aminotransferase and tumor size (P<0.05), but not with sex, age and tumor envelope. The total survi-val time of the HCC patients with high expression of SCNM1 was shorter than that of the patients with low expression of SCNM1 (HR=1.53, P=0.016), and that of the patients with hepatitis B-related HCC was even shorter (HR=2.41, P=0.015).CONCLUSION: SCNM1 is highly expressed in hepatitis B-related HCC and may play an important role in the development of hepatitis B-related HCC.  相似文献

AIM:To investigate the expression of microRNA-187* (miR-187*) in human colon cancer cell lines and normal colon tissues, and to determine the effects of miR-187* up-regulation on the proliferation and cell cycle of human colon cancer cell line HCT116. METHODS:The expression profiling of miRNAs in 3 colorectal adenocarcinoma samples and their matched normal tissue samples was performed using miRNA microarray chip. Total RNA was isolated from 8 colon cancer cell lines and 10 normal colon tissues. The miR-187* level was detected by Taqman real-time RT-PCR. B-cell-specific Moloney murine leukemia virus integration site 1 (BMI-1), the possible target of miR-187*, was also detected. Synthetic miR-187* mimics were transfected into HCT116 cell line by LipofectamineTM 2000. The mRNA expression of miR-187* and BMI-1 in HCT116 cell line was measured by real-time RT-PCR. Cell growth and cell cycle were assayed by MTS method and flow cytometry. RESULTS:miR-187* was found to be differentially expressed between colorectal adenocarcinoma and normal tissues. The expression of miR-187* in 8 colon cancer cell lines was down-regulated, while BMI-1 mRNA was up-regulated. Compared with blank control group, miR-187* expression was remarkably increased after transfection with miR-187* mimics, and ectopic expression of miR-187* significantly inhibited the mRNA expression of BMI-1. The cell growth was inhibited in miR-187* mimics group, and proliferating cell nuclear antigen (PCNA) mRNA expression was decreased. The cells at G2/M phase in miR-187* mimics group were significantly increased. CONCLUSION: miR-187* is down-regulated in human colon cancer cell lines. Up-regulation of miR-187* not only inhibits the proliferation but also influence the cell cycle of HCT116 cells, which might act as a tumor suppressor in colorectal cancer by inhibiting the expression of BMI-1.  相似文献

AIM: To investigate the expression profile of myosin VI in various human carcinomas and adjacent normal tissues. METHODS: A piece of cancer profiling array containing 154 matched cDNA pairs from 19 tumors and the adjacent normal tissues and 10 diverse tumor cell lines were separately hybridized with radiolabeled probes for myosin VI and housekeeping gene ubiquitin. Immunohistochemistry (IHC) was used to confirm the expression profile of myosin VI in 40 cases of ovarian adjacent normal tissues, 8 cases of cystadenoma, 16 cases of borderline tumors and 52 cases of endometrioid carcinoma by tissue microarray. RESULTS: Myosin VI was expressed in all the tissues and cell lines. The expression of myosin VI was significantly higher in ovarian and colon cancer tissues than that in matched normal tissues. The results of IHC confirmed that myosin VI expression rates were 100% (52/52), 81.3% (13/16) and 10.4% (5/48) in the ovarain carcinoma, boderline tumor and nomal ovarian epithelium or cystadenoma, respectively. The expression of myosin VI protein was significantly higher and stronger in ovarain carcinoma than that in the borderline tumor, benign tumor or normal ovary tissues. A significant correlation was also found between the expression of myosin VI and the tumor grade of ovarain carcinoma. CONCLUSION: Differentiated expression of myosin VI is found in diverse malignant tumor tissues and cell lines, suggesting myosin VI plays an important role in the tumor development and progression.  相似文献
