通过在紫外285nm处测定抗氧化活性物质(蛋白质、芳族胺、酚类等化合物)的含量,在450nm处测定类胡萝卜素的含量,采用DPPH法测定不同溶剂橡子提取物的自由基消除能力,测定羟自由基消除活性及橡子提取物对α-葡萄糖苷酶的抑制作用,研究麻栎橡子抗氧化活性及抗糖尿病活性。结果表明:70%甲醇提取物中的抗氧化活性物质含量最高,而水提取物中最少;70%甲醇提取物和70%乙醇提取物的类胡萝卜素含量较高;70%甲醇提取物的DPPH自由基的消除能力最强;橡子的水提取物对羟自由基的清除作用最强;甲醇提取物的α-葡萄糖苷酶抑制率较高。说明麻栎橡子具有抗氧化活性和抗糖尿病作用。 相似文献

为了明确β-1,3-葡聚糖酶与大豆抗疫霉根腐病的关系,测定了大豆叶片β-1,3-葡聚糖酶的活性和对疫霉菌的抑菌作用,结果表明,在接种大豆疫霉菌后48 h β-1,3-葡聚糖酶活性达到峰值,利用接种48 h提取的β-1,3-葡聚糖酶粗酶液进行抑菌试验,发现β-1,3-葡聚糖酶粗酶液对大豆疫霉菌的菌丝生长和孢子萌发有明显的抑制作用. 相似文献

对小麦抗感白粉病品种苗期、成株期叶片过氧化物酶同工酶和酯酶同工酶进行比较测定。结果表明感病品种苗期过氧化物酶同工酶谱带数多于抗病品种;酯酶同工酶抗感品种无明显差异。成株期过氧化物酶同工酶带数抗感品种无明显差异,但个别酶带有所加强或减弱;感病品种的酯酶同工酶酶带数多于抗病品种。两种同工酶在苗期和成株期测定的酶谱类型均不一致。 相似文献

对小麦叶锈菌侵染前后2个不同抗性小麦品种叶片中的POD、PPO、PAL、β-1,3-葡聚糖酶4种防卫酶的活性变化进行了研究。结果表明:在接种前,抗病品种的POD和PPO活性均高于感病品种,而抗、感品种间PAL和β-1,3-葡聚糖酶的活性差异不大;接种叶锈菌后抗病品种和感病品种小麦叶片中的4种防御酶活性均高于未接种处理,并且抗病品种接种后PPO活性变化幅度明显高于感病品种;抗病、感病的小麦接种叶锈菌处理与未接种处理的POD、PPO、PAL、β-1,3-葡聚糖酶4种防卫酶的活性均升高,POD、PPO、PAL 3种酶活性在接种后3 d达到峰值,β-1,3-葡聚糖酶在接种后4 d达到峰值;相对于其他防御酶,PAL在测定期内活性变化程度不高,表现平稳。 相似文献

枯草芽孢杆菌W-QX-1碱性蛋白酶的性质及其抗烟草花叶病毒活性初步研究 总被引:2,自引:1,他引:1
为发现新的天然源抗植物病毒活性物质,从枯草芽孢杆菌W-QX-1的发酵液中粗提了碱性蛋白酶,并初步测定了其抗TMV活性及其酶学性质。结果表明,W-QX-1碱性蛋白酶作用最适温度为50℃;在硼酸-硼砂缓冲液中的作用最适pH为7.8;在Tris-HCl缓冲液中的作用最适pH为8.0;在pH6~10范围内,45℃以下酶的性质稳定;对干酪素、明胶、牛血清白蛋白和精炼丝素均具有一定的降解能力,其中对干酪素的降解力最强。采用半叶法测定了其抗烟草花叶病毒TMV的活性,发现该酶对TMV的体外钝化作用明显,在酶液浓度为50 mg/L时,其钝化效果即可达到53.40%;而该酶对TMV的初侵染和体内复制增殖也具有一定的抑制能力。试验结果表明,在TMV侵染前24 h施用浓度为200 mg/L碱性蛋白酶液抑制其侵染力的效果达到50.35%,而在TMV侵染后抑制其复制增殖的作用并不明显。可见,该酶是一种性质稳定的广谱性蛋白水解酶,并具有一定的抗TMV活性。 相似文献

采用田间用蚜量比值法划分了抗感性不同的15个小麦品系,用紫外分光光度计测定了各品种(系)旗叶中过氧化氢酶、过氧化物酶、苯丙氨酸解氨酶、几丁质酶的活性.结果表明,苯丙氨酸解氨酶、过氧化物酶、几丁质酶活性与春小麦蚜量比值显著相关,其相关系数分别为r=-0.842 0、r=-0.798 0、r=-0.795 5.抗蚜的春小麦品系在蚜虫取食后苯丙氨酸解氨酶、过氧化物酶、几丁质酶活性急剧上升,感蚜春小麦虽然酶活性有上升,但较为缓慢;而过氧化氢酶活性与春小麦蚜量比值相关性不显著,其相关系数为r=-0.219 6,在抗感性春小麦中无明显的差异. 相似文献

To produce high-affinity monoclonal antibodies against pesticide carbofuran, and the develop immunochemical assays for people's health and environmental protection, the hapten 4-[[(2,3-dihydro-2,2-dimethyl-7-benzofuranyloxy) carbonyl]- amino]-butanoic acid (BFNB) of carbofuran was synthesized and Balb/c mice were immunized by the hapten-carrier (BFNB-bovine serum albumin, BFNB-BSA) conjugates. The splenocytes of immunized mice were fused with Sp2/0 cells and the cultural supernatants of hybridoma cells were screened by the indirect enzyme-linked immunoabsorbent assay (ELISA), based on BFNB-ovoalbumin conjugates (BFNB-OVA). Purified monoclonal antibody (McAb) was obtained from fluids of ascites, deposited by octanoic acid and ammonium sulfate. The affinity and the specificity of McAb were characterized by ELISA or indirect competitive ELISA. A hybridoma cell line (5D3) secreting anti-carbofuran McAb had been established. The titer of culture medium and ascites was up to 1:2.048 × 10^3 and 1:1.024 × 10^6, respectively, and the subtype of the McAb was IgG1. The affinity constant of the McAb was about 2.54 × 10×9 L mol^-1, with an IC50 value of 1.18 ng mL^-1 and a detection limit of 0.01 ng mL^-1. Cross-reactivity studies showed that the McAb was quiet specific for carbofuran, as among the four analogous compounds, they were all hardly recognized (4.59 × 10^-4% for 2,3-dihydro-2,2- dimethyl-7-benzofuranol and less than 3.0 × 10^4% for others). The prepared McAb had a very high affinity and specificity, and it could be used to develop ELISA for rapid determination of carbofuran. 相似文献

为研究芒果皮渣中多糖的提取工艺及其抗氧化活性,在单因素试验的基础上,通过正交试验优化芒果皮渣多糖的提取工艺参数,同时利用清除DPPH·法、·OH法和O2-·法评价其体外抗氧化活性。结果表明,超声波辅助提取芒果皮渣多糖的最佳工艺条件为提取温度80℃、超声功率160 W、提取时间90 min、料液比1∶2 g/m L、提取次数2次,在此条件下多糖提取率可达0.81%。体外抗氧化试验表明,芒果皮渣多糖对DPPH·、·OH和O2-·均有一定的清除能力,随着质量浓度的增加,清除能力逐渐增强。当浓度为1.0 mg/m L时,他们的清除率分别达到93.1%、94.5%和8.89%。本研究建立了新鲜芒果皮渣多糖的提取工艺,并评价了其体外抗氧化活性,可为芒果资源的充分利用、研究开发等提供理论依据。 相似文献

Four widely used in vitro assays for genetic toxicity were evaluated for their ability to predict the carcinogenicity of selected chemicals in rodents. These assays were mutagenesis in Salmonella and mouse lymphoma cells and chromosome aberrations and sister chromatid exchanges in Chinese hamster ovary cells. Seventy-three chemicals recently tested in 2-year carcinogenicity studies conducted by the National Cancer Institute and the National Toxicology Program were used in this evaluation. Test results from the four in vitro assays did not show significant differences in individual concordance with the rodent carcinogenicity results; the concordance of each assay was approximately 60 percent. Within the limits of this study there was no evidence of complementarity among the four assays, and no battery of tests constructed from these assays improved substantially on the overall performance of the Salmonella assay. The in vitro assays which represented a range of three cell types and four end points did show substantial agreement among themselves, indicating that chemicals positive in one in vitro assay tended to be positive in the other in vitro assays. 相似文献

幼龄绵羊血清中T_3含量的RIA测定表明,年平均含量1.246±0.49ng/ml。冬季昼夜平均含量最高为1.748±0.13ng/ml,极明显地高于其它三个季度。春季昼夜平均值1.567±0.20ng/ml,也极明显地高于秋、夏季。秋、夏季的昼夜平均值较接近,为0.851±0.06ng/ml和0.816±0.07ng/ml,并无明显差异,但昼夜升降变化,恰好相反。年平均值的昼夜变化及春季含量的昼夜变化,仅见最高值显著高于最低值。其余季度的昼夜含量变化,无明显差异。 相似文献

为了探讨绵羊卵巢卵泡颗粒细胞体外培养时FSH(Invitrogen)与胰岛素的最佳质量浓度,试验在无血清条件下培养绵羊卵泡颗粒细胞,并向其中加入不同质量浓度的FSH与胰岛素,体外培养7 d后测定细胞数目以及培养液内雌二醇浓度。结果表明,当培养液内FSH质量浓度为25 ng/mL、胰岛素质量浓度为100ng/mL时,颗粒细胞的数目最多;当FSH质量浓度为1 ng/mL、胰岛素质量浓度为10 ng/mL时,培养液内雌激素质量浓度最高。 相似文献

以贵州省草海湖泊沉积物孔隙水为研究对象,实测其溶解态甲基汞和溶解态无机汞的质量浓度,并根据质量浓度梯度和菲克第一定律,初步估算了沉积物-水界面溶解态无机汞和甲基汞的扩散通量.结果表明:不同季节沉积物孔隙水中无机汞、甲基汞与上覆水体中的无机汞、甲基汞之间均存在质量浓度梯度,其溶解态无机汞的质量浓度梯度在枯水期和丰水期分别为8.6 ng/L和25.1 ng/L;溶解态甲基汞的质量浓度梯度分别为0.46 ng/L和2.7 ng/L.在枯水期和丰水期沉积物孔隙水中无机汞的释放通量分别为31.4 ng/(m2.d)和52.8 ng/(m2.d);甲基汞的释放通量分别为1.7 ng/(m2.d)和3.9 ng/(m2.d).沉积物-水界面无机汞和甲基汞的扩散通量远低于同背景区的阿哈湖、红枫湖,并存在明显的季节性变化,其变化与孔隙水中汞的质量浓度的季节性变化一致,均为在丰水期的扩散通量大于在枯水期. 相似文献
