The effect of wounding on polyphenol oxidase (PPO), peroxidase (POD), and phenylalanine ammonia-lyase (PAL) was studied in six minimally processed lettuce (Lactuca sativa L.) cultivars upon storage for 7 days at 5 degrees C (Iceberg Mikonos (IM), I. Green Queen (IGQ), I. Asdrúbal (IA), Little Gem Sandra (LGS), Romaine Cazorla (RC), and R. Modelo (RM)). Wounding of lettuce tissue midribs (because of minimal processing) caused an exponential increase in PPO activity due to the activation process from latent to fully active PPO by following first order kinetics in the time range from 3.7 days (LGS) to 6.3 days (RC). However, total PPO activity (active plus latent) remained constant. Isoform pattern of PPO changed upon storage probably because of posttranslational processes. POD activity linearly increased with induction of new POD isoenzymes. PAL activity presented a typical bell-shaped induction pattern in four cultivars. Only IM and IGQ showed a second induction response which has not been previously described in the literature. IM was the cultivar most susceptible to browning and RC was the cultivar least susceptible. However, no clear correlation was observed between browning and any of the biochemical and physiological attributes investigated (PPO, PAL, and POD activities, total and individual phenols accumulation, and ascorbic acid content).  相似文献

Total phenolic content and antioxidant capacity of two tree spinach species (Cnidoscolus chayamansa McVaugh and C. aconitifolius Miller.) were determined in raw and cooked leaf extracts. Antioxidant capacity was assessed by the oxygen radical absorbance capacity (ORAC) assay, and flavonoid glycoside composition was quantified by HPLC and identified by GC. Total phenolics and antioxidant capacity were higher in raw than in cooked leaf extracts. The ORAC values were strongly correlated with total phenolic content (r = 0.926) in all leaf extracts. The major flavonoids isolated from the leaf extracts were kaempferol-3-O-glycosides and quercetin-3-O-glycosides. C. aconitifolius leaves contained more varieties of the flavonoid glycosides than C. chayamansa. Cooking reduced antioxidant activity and phenolic content and resulted in losses of some kaempferol glycoside and quercetin glycoside residues in leaf extracts. The results of this study indicate that tree spinach leaves are a rich source of natural antioxidants for foods.  相似文献

Plasma antioxidant capacity (AC) has been assessed in eight healthy human volunteers after wine intake. Analytical methods used for evaluating AC included the ferric reducing ability of plasma (FRAP) and oxygen radical absorbance capacity using two different fluorescent probes, beta-phycoerythrin (ORAC-PE) and fluorescein (ORAC-FL). In addition, the concentrations of endogenous antioxidants such as uric acid, albumin, and bilirubin were determined. The suitability of analytical methods was evaluated with two different biological matrixes: plasma and serum. Plasma AC was determined before ingestion of 300 mL of red wine (baseline value) and 30, 55, and 120 min afterward. Maximum average increase in AC values was reached at 55 min. Plasma AC determined by ORAC-PE at time zero was significantly correlated with albumin concentration. Plasma AC determined with FRAP at time zero is well correlated with uric acid. Moreover, a good linear correlation was found between uric acid concentration and AC determined by FRAP in each volunteer. The maximum concentration of uric acid occurred after 55 min. Uric acid increase accounts for a nonnegligible part in FRAP values and must be evaluated when using this method for assessing AC in plasma.  相似文献

基于高光谱的油麦菜叶片水分CARS-ABC-SVR预测模型   总被引:1,自引:7,他引:1  
为了实现油麦菜生长期间更合理的灌水管理,研究一种基于高光谱技术的精确、快速、有效检测油麦菜叶片水分的新方法。以5种不同水分胁迫水平的油麦菜为研究对象,通过高光谱成像系统获取高光谱图像并利用干燥法测量叶片含水率。采用多项式平滑(Savitzky-Golay,SG)结合标准变量变换(standard normalized variable,SNV)对高光谱数据去噪平滑。利用竞争性自适应加权算法(competitive adaptive reweighted sampling,CARS)进行特征波长选择,并与逐步回归分析(stepwise regression,SR)及连续投影算法(successive projections algorithm,SPA)进行比较,利用支持向量回归机(support vector regression,SVR)分别建立油麦菜叶片全光谱数据、3种特征光谱数据与干基含水率的关系模型。结果表明,基于竞争性自适应加权算法波长选择的支持向量回归模型(CARS-SVR)效果最佳,但预测精度尚不够理想,故引入人工蜂群算法(artificial bee colony,ABC)优化模型的参数惩罚因子和核参数。最终,经人工蜂群算法优化后的模型(CARS-ABC-SVR)的预测集决定系数R2和均方根误差RMSE分别为0.9214和2.95%。因此,利用高光谱技术结合CARS-ABC-SVR模型预测油麦菜叶片水分含量是可行的。  相似文献

Abstract

Leaf lettuce (Lactuca sativa var. crispa L.), radish (Raphanus sativus L.), and spinach (Spinacea oleracea L.) plants were grown in sand culture under variable N concentrations from 0.187 to 48.0 meq/l. The plants were harvested when those grown at 12 meq N/l had attained approximate market maturity. Growth was restricted, and symptoms of N deficiency were evident at the low levels of N. Growth was also restricted at the highest N level. Approximate critical levels for leaf lettuce, radish roots, and spinach are 2000 ppm, 5000 ppm, and 4000 ppm, nitrate‐N respectively, on a dry weight basis.  相似文献

Sweet rowanberry cultivars adapted to northern climates have been developed from rowanberries (Sorbus aucuparia L.) and hybrids of rowanberry with Malus, Pyrus, Aronia, or Mespilus. The rowanberries studied here (cvs. Burka, Dessertnaja, Eliit, Granatnaja, Kubovaja, Rosina, Rubinovaja, Titan, and Zholtaja) have high antioxidant and phenolic contents. The phenolic content varied between 550 and 1014 mg/100 g of fresh weight in sweet rowanberries, whereas 846 and 717 mg were found in the well-characterized bilberry and lingonberry, respectively. Anthocyanins (6-80 mg) were mainly found from berries of hybrid cultivars. Of the other phenolics, chlorogenic (29-160 mg) and neochlorogenic (34-104 mg) acids constituted the major fraction in all rowanberries, the concentrations almost equaling those present in coffee. Antioxidant capacities of rowanberries were high, as measured with FRAP (61-105 micromol of Fe2+/g) and DPPH (21.3-9.7 g/g DPPH) methods. Principal component analysis was able to separate the cultivars of different origin into clusters on the basis of their phenolic profiles.  相似文献

Abstract

The objective of this investigation was to examine the effect of restriction of growing medium volume on the level of tipburn in chervil and leaf lettuce. Chervil: The plants were grown in different sizes of rock wool cubes: a. 31 cm3, b. 64 cm3, c. 135 cm3. The incidence of tipburned leaves decreased by increasing the size of cubes. Leaf lettuce: Plants were grown in different sizes of pots: 12 cm pots with capacity of 0.6 L and 8 cm pots with capacity of 0.25 L. 采用"3414"设计方案设置氮磷钾肥的田间试验,旨在研究不同氮磷钾组合对紫叶莴笋的产量与品质的影响。结果表明,合理施肥对紫叶莴笋具有显著增产效果,其中处理N_2K_2P_2增幅最大,增产量为10 401.3kg/hm~2,并且氮磷钾增产效应为钾氮磷;合理配方施肥处理能显著提高紫叶莴笋叶与茎中氨基酸、还原糖及Vc的含量,降低硝酸盐的含量,提高紫叶莴笋的品质;紫叶莴笋叶花青素含量的增加在一定范围内与施肥量成正比,其中花青素含量最高的为处理N_2P_1K_1,为41.28 mg/100 g,随着氮、磷的增加花青素含量呈现先增加后降低趋势。结合紫叶莴笋产量与品质的多个指标综合评价表明:效果最优的为处理N_2P_1K_2及N_1P_2K_2,氮、磷、钾施肥量分别为276.0、45.0、153.0 kg/hm~2与138.0、90.0、153.0 kg/hm~2。  相似文献

Although lettuce may provide relatively low levels of antioxidative phytochemicals which may contribute to human health, lettuce leaf extracts in fact contained compounds with high specific peroxyl radical scavenging activities. After determining the extraction conditions that minimized phenolic oxidation and produced the highest oxygen radical absorbance capacity (ORAC) values, the phenolic compounds from red leaf lettuce were separated by reverse-phase high-performance liquid chromatography (HPLC). The primary phenolic compounds in the leaf tissue extracts were mono- and dicaffeoyltartaric acid (CTA and DCTA), mono- and dicaffeoylquinic acid (CQA and DCQA), quercetin 3-malonylglucoside (QMG), quercetin 3-glucoside (QG), cyanidin 3-malonylglucoside (CMG), and an unknown phenolic ester (UPE). Significant levels of DCQA were only found after wounding. Using the new fluorescein-based ORAC assay procedures, fractions from the HPLC analyses were assayed for peroxyl radical absorbance capacity. Using absorbance to estimate concentration, the decreasing order of contribution to the total ORAC value of an extract from wounded tissue was QMG > DCQA > CMG > DCTA > UPE > QG > CTA. The decreasing order of the specific peroxyl radical scavenging activities was CMG > QG > DCTA > DCQA > QMG > UPE > CQA > CTA. Since the concentrations of plant flavonoid and phenolic acid esters are sensitive to environmental factors, this information may be used to develop pre- and postharvest conditions which increase the dietary benefits of leaf lettuce.  相似文献

Fresh strawberries (Fragaria x ananassa Duch.), raspberries (Rubus idaeus Michx.), highbush blueberries (Vaccinium corymbosum L.), and lowbush blueberries (Vaccinium angustifolium Aiton) were stored at 0, 10, 20, and 30 degrees C for up to 8 days to determine the effects of storage temperature on whole fruit antioxidant capacity (as measured by the oxygen radical absorbing capacity assay, Cao et al., Clin. Chem. 1995, 41, 1738-1744) and total phenolic, anthocyanin, and ascorbate content. The four fruit varied markedly in their total antioxidant capacity, and antioxidant capacity was strongly correlated with the content of total phenolics (0.83) and anthocyanins (0.90). The antioxidant capacity of the two blueberry species was about 3-fold higher than either strawberries or raspberries. However, there was an increase in the antioxidant capacity of strawberries and raspberries during storage at temperatures >0 degrees C, which was accompanied by increases in anthocyanins in strawberries and increases in anthocyanins and total phenolics in raspberries. Ascorbate content differed more than 5-fold among the four fruit species; on average, strawberries and raspberries had almost 4-times more ascorbate than highbush and lowbush blueberries. There were no ascorbate losses in strawberries or highbush blueberries during 8 days of storage at the various temperatures, but there were losses in the other two fruit species. Ascorbate made only a small contribution (0.4-9.4%) to the total antioxidant capacity of the fruit. The increase observed in antioxidant capacity through postharvest phenolic synthesis and metabolism suggested that commercially feasible technologies may be developed to enhance the health functionality of small fruit crops.  相似文献

Antioxidant activity of piper betel leaf extract and its constituents   总被引:6,自引:0,他引:6  
The 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay of the ethanol extracts of three varieties (Bangla, sweet, and Mysore) of Piper betel (pan) revealed the Bangla variety to possess the best antioxidant activity that can be correlated with the total phenolic content and reducing powers of the respective extracts. Column chromatography of the extract of the Bangla variety led to the isolation of chevibetol (CHV), allylpyrocatechol (APC), and their respective glucosides. The HPTLC analyses of the extracts revealed similar chemical profiles in all three P. betel varieties, although the concentrations of CHV and APC were significantly less in the sweet and Mysore varieties. Among the isolated compounds, APC showed the best results in all the in vitro experiments. It could prevent Fe(II)-induced lipid peroxidation (LPO) of liposomes and rat brain homogenates as well as gamma-ray-induced damage of pBR322 plasmid DNA more efficiently than CHV. The superior anti-LPO and radioprotective activities of APC vis-à-vis those of CHV could not be explained by their respective Fe(II) chelation and .OH radical scavenging capacities. The better ability of APC to scavenge O2-. radicals and H2O2 might account for the results.  相似文献

Buckwheat honey increases serum antioxidant capacity in humans   总被引:11,自引:0,他引:11  
Honey has been known to exert significant in vitro antioxidant activity, in part due to its phenolic content. However, conclusions that the antioxidants in honey are or are not efficacious in the human body cannot be reached if its antioxidant action is not assessed as part of a human study. In the present study, the acute effect of consumption of 500 mL of water, water with buckwheat honey, black tea, black tea with sugar, or black tea with buckwheat honey on serum oxidative reactions was examined in 25 healthy men. Antioxidant capacity of human serum samples was measured using different methods: the oxygen radical absorbance capacity (ORAC) assay, ex vivo susceptibility of serum lipoprotein to Cu(2+)-induced oxidation, and the thiobarbituric acid reactive substances (TBARS) assay. The results showed that the serum antioxidant capacity determined by ORAC increased significantly (p < 0.05) by 7% following consumption of buckwheat honey in water. No significant changes in serum antioxidant capacity could be established after the consumption of any of the other beverages. Ex vivo serum lipoprotein oxidation and TBARS values were not significantly altered after consumption of any of the five beverages. This study provides primary evidence of the in vivo antioxidant activity of buckwheat honey. However, long-term studies and epidemiological data are necessary to investigate whether honey consumption can exert overall antioxidant-related health benefits.  相似文献

【目的】针对目前设施农业中常用的灌溉方式容易造成土表水分蒸发和水肥流失,且不能按照作物所需自动供水供肥的现状,本研究探讨了负压灌溉提高紫叶生菜的产量和品质,以及水分利用效率和土壤微生物群落多样性的机理。【方法】在温室内进行盆栽试验,以紫叶生菜(Lactuca sativa L.)为试验材料,设置3个灌溉处理:常规灌溉、滴灌和负压灌溉。收获后,测定了紫叶生菜的产量,分析了品质(维生素C、可溶性糖、花青素和硝酸盐含量),植株的养分(氮、磷和钾)浓度和吸收量,监测了土壤水分含量动态的变化,计算了水分消耗量和水分利用效率,分析了根际土壤微生物的多样性指数和细菌在门分类上的群落结构组成。【结果】负压灌溉下显著提高紫叶生菜的产量和品质,负压灌溉比常规和滴灌处理的产量分别显著提高了68.1%和29.0%,也提高了维生素C、可溶性糖和花青素的含量,减少了硝酸盐含量。与常规灌溉相比负压灌溉显著提高了紫叶生菜氮、磷、钾的浓度和含量,分别提高13.0%、14.4%、38.4%和90.2%、92.6%、135.5%。紫叶生菜在负压灌溉下耗水量最少,为9900 cm3,比常规和滴灌处理分别减少了23.8%和23.8%;负压下水分利用效率比常规和滴灌分别显著提高了122.2%和70.5%。同时负压灌溉处理下动态的土壤含水量处于10.3%~11.3%之间,变异范围低于常规和滴灌处理9.2%~11.6%。通过高通量测序紫叶生菜根际土壤微生物群落发现,负压灌溉处理下微生物多样性指数最高,表现为OTU、Chao1和Shannon指标的数值显著高于常规和滴灌处理,其数值分别为1808、2437和8.48,分别比常规灌溉处理显著提高了15.2%、15.7%和3.16%。同时也改变了细菌在门分类水平上组成的相对丰度,在负压灌溉处理下比常规和滴灌处理分别提高了放线菌门(Actinobacteria),绿弯菌门(Chloroflexi),疣微菌门(Verrucomicrobia)和浮霉菌门(Planctomycetes)在细菌门分类上的相对丰度。【结论】本试验证明了负压灌溉系统通过土壤水肥平稳供应机制,实现了紫叶生菜高产优质且高效利用水分的目标。因此,负压灌溉系统相比常规和滴灌,显著提高了紫叶生菜的产量和品质、水分利用效率和根际微生物群落的多样性,为设施农业的可持续性发展提供科学依据。  相似文献

Beringite (B) and zerovalent iron grit (Z), singly and in combination (BZ), were added to a loamy sand soil contaminated by trace elements (Reppel, Belgium), mainly by arsenic (As), to reduce As labile fractions and phytoavailability. An uncontaminated sandy soil was studied for comparison. Soils were placed in large lysimeters cultivated with maize and vegetables for 6 years. pH, organic C and total N content increased in amended soils. The Z and BZ treatments reduced the Ca(NO3)2 extractable soil As and As uptake by lettuce. The BZ lettuces had also the lowest foliar Pb, Cd, Zn, and Mn concentrations. All amendments had positive effects on the soil microbial biomass and reduced the qCO2. Glucose mineralization was increased in Z and BZ amended soils. Acid phosphomonoesterase activity was higher in the untreated soil than in the other soils; the alkaline phosphomonoesterase, phosphodiesterase and protease activities were increased by Z and BZ treatments, whereas B amendment had less positive effects. Genetic fingerprinting using Denaturing Gradient Gel Electrophoresis (DGGE) revealed shifts in the composition of eubacterial and fungal communities of the amended soils. Microbial species richness decreased rather than increased in the treated soils, regardless of reduced trace element availability and increased soil microbial biomass and activity.  相似文献

Abstract

A simple and rapid procedure for the determination of nitrate in fresh leaf tissue, suitable for use by relatively untrained operators is presented. The method requires only 4 reagents and can be carried out in less than 20 min from start to finish. A 400 mg sample of fresh leaf tissue is macerated briefly with 5 drops of 10 N sulphuric acid, diluted with 10.0 ml of water and filtered. Nitrate‐N is estimated on an aliquot of the filtered extract by reducing the nitrate to nitrite by shaking with powdered zinc in ammonium hydroxide solution for 3 min. The reduced mixture is allowed to settle (5 min) and an aliquot is withdrawn through a cotton‐wool plug thus removing any particles of the zinc. The filtered aliquot is then reacted with a single colour reagent to yield a pink azo‐dye, the intensity of which is directly proportional to the amount of nitrate.

The method was found to have a coefficient of variation of about 4%. When compared with the phenoldisulphonic acid method for nitrate, on aliquots of the some plant extract, it yielded values which were on average 94.6% of those obtained with the former method. The coefficient of variation between the methods was 7%.  相似文献

在温室中利用负压灌溉装置进行盆栽试验,研究了施钾水平对紫叶生菜的产量、品质、养分吸收、钾肥利用效率、抗氧化能力和根际微生物多样性等指标的影响。结果表明:1)施用钾肥量为300 kg/hm2时紫叶生菜的产量、品质和营养吸收量高于其他处理。2)施肥量300 kg/hm2处理在钾肥贡献率、钾肥农学利用率和钾肥吸收利用率表现最佳。3)225 kg/hm2和300 kg/hm2处理的抗氧化能力要高于其他处理。4)300 kg/hm2处理在OTU、Chao 1和香农指数数值最好,比150 kg/hm2处理分别高出了17%、16%和4%。综合各项指标考虑,在负压灌溉水肥一体化装置下钾肥施用量在300 kg/hm2时,提高了紫叶生菜的产量和品质,促进养分的吸收和抗氧化能力,显著提高了钾肥的利用率和根际土微生物的多样性。  相似文献

Total equivalent antioxidant capacity (TEAC) and phenolic content of 26 common spice extracts from 12 botanical families were investigated. Qualitative and quantitative analyses of major phenolics in the spice extracts were systematically conducted by reversed-phase high-performance liquid chromatography (RP-HPLC). Many spices contained high levels of phenolics and demonstrated high antioxidant capacity. Wide variation in TEAC values (0.55-168.7 mmol/100 g) and total phenolic content (0.04-14.38 g of gallic acid equivalent/100 g) was observed. A highly positive linear relationship (R2= 0.95) obtained between TEAC values and total phenolic content showed that phenolic compounds in the tested spices contributed significantly to their antioxidant capacity. Major types of phenolic constituents identified in the spice extracts were phenolic acids, phenolic diterpenes, flavonoids, and volatile oils (e.g., aromatic compounds). Rosmarinic acid was the dominant phenolic compound in the six spices of the family Labiatae. Phenolic volatile oils were the principal active ingredients in most spices. The spices and related families with the highest antioxidant capacity were screened, e.g., clove in the Myrtaceae, cinnamon in the Lauraceae, oregano in the Labiatae, etc., representing potential sources of potent natural antioxidants for commercial exploitation. This study provides direct comparative data on antioxidant capacity and total and individual phenolics contents of the 26 spice extracts.  相似文献

We hypothesized that antioxidants in organic amendments would control the rate of their decomposition in soil, such that those with more antioxidant capacity (AOC) would have slower rates of decomposition. This has been tested using materials from an incubation experiment, in which a Vertisol had been amended with a number of organic materials. Green waste compost, wheat straw, sugarcane trash and mineral fibre had been added to the soil and incubated for different periods of up to 84 days. Decomposition had been monitored by measuring cumulative carbon dioxide emissions. One and four m NaOH extracts of soil material from the experiment were analysed for their AOC by the Trolox equivalent method. The same method was used to characterize the organic amendments and the unamended soil. AOCs of the amendments were much greater than that of the soil and therefore we expected that the AOC of amended soils would be greater than the unamended control. However, this was not generally the case. A comparison of the AOC of amended soil measured after 7 days with that calculated from the components showed that the measured values of incubated amended soils were less than the calculated ones, particularly for wheat straw and sugarcane trash amendments. This suggests that some of the extractable antioxidants in the amendments were lost or transformed during the first 7 days of incubation. In the remainder of the incubation period there was a small increase in AOC with time in 4 m NaOH extracts of the amended soils, but this was not the case for 1 m NaOH extracts. There was some evidence that amendments with larger AOCs had slower rates of decomposition, especially during the first 7 days of the incubation. Other factors may have been responsible, but C:N ratio alone could not explain the differences. The results are encouraging, but more data are required to test our hypothesis thoroughly.  相似文献

Ten cranberry (Vaccinium macrocarpon Aiton) cultivars were evaluated for oxygen radical absorbance capacity (ORAC), anthocyanins, and total phenolics contents after three months of storage at 0, 5, 10, 15, and 20 degrees C. The antioxidant capacity of cranberry was affected by cultivars and storage temperatures. Among the 10 cranberry cultivars used in this study, Early Black, Crowley, and Franklin had higher antioxidant capacities than the other cultivars. ORAC values, anthocyanins, and total phenolics contents increased during storage. The highest increases in antioxidant activity, anthocyanin, and phenolics contents occurred at 15 degrees C storage. Fruit stored at 20 degrees C had lower ORAC values than those stored at 15 degrees C. A positive relationship existed between ORAC values and anthocyanin or phenolic content in all 10 cranberry cultivars at different storage temperatures.  相似文献

Lipophilic extracts from 16 species of seaweeds collected along the Qingdao coastline were screened and evaluated for their antioxidant activities (AA) using the beta-carotene-linoleate assay system. The diethyl ether soluble extracts of all selected seaweeds exhibited various degrees of antioxidative efficacy in each screen. The highest antioxidant capacities among the tested samples were observed for Rhodomela confervoides and Symphyocladia latiuscula and were comparable with that of the well-known antioxidant butylated hydroxytoluene and greater than that of propyl gallate. The lipophilic content of all 16 samples and the chemical composition of 4 selected seaweeds, R. confervoidesand S. latiuscula, which had higher AA, Laminaria japonica, which had intermediate AA, and Plocamium telfairiae, which had lower AA, were analyzed by gas chromatography and gas chromatography-mass spectrometry, respectively. Fatty acids and alkanes were found. The present data indicated an increase in antioxidative property with increasing content of unsaturated fatty acid. The result of this study suggests that seaweeds can be considered as a potential source for the extraction of lipophilic antioxidants, which might be used as dietary supplements or in production in the food industry. This is the first report on the antioxidant activities of lipophilic extracts from seaweeds.  相似文献
