Relationships of implanted testosterone, dihydrotestosterone and estradiol-17 beta to collagen degradation and intramuscular collagen concentration and stability were determined. Intramuscular collagen content, solubility and shrinkage temperature and serum hydroxyproline were analyzed in groups of six rams, wethers, and wethers implanted with various levels of testosterone or dihydrotestosterone and groups of 10 rams, wethers and wethers implanted with estradiol-17 beta, dihydrotestosterone or a combination of these two steroids. Intramuscular collagen content in both experiments was higher (P less than .05) in muscles of rams than in muscles of wethers. Administration of the highest level of testosterone to wethers raised (P less than .05) total and insoluble intramuscular collagen to concentrations noted in rams. Administration of the testosterone metabolite, dihydrotestosterone, to wethers had no effect on intramuscular collagen. Administration of estradiol-17 beta to wethers tended to raise concentrations of intramuscular collagen so that they were no longer lower (P less than .05) than those in rams. Collagen stability as measured by solubility and thermal shrinkage temperature did not differ among rams, wethers or implanted wethers (P greater than .05). Increases in collagen accretion due to hormone administration were observed to be the result of increases in the insoluble portion of the intramuscular collagen (P less than .05).  相似文献

Twelve blood serum components were determined on 78 barrows and 66 gilts, serially slaughtered at six age groups, starting at 69 d, at 2-wk intervals. One-half of the animals were fed ad libitum and one-half were fed "time restricted". Partial correlations between production and carcass traits and serum components were calculated from the residuals of the least-squares analysis. Calcium, serum protein and albumin were highly (P less than .01) correlated with several production and carcass traits. Creatinine was positively correlated (P less than .05) with backfat, loin fat, and percent dissected muscle and bone. Alkaline phosphatase showed a positive relationship with average daily gain (P less than .05) and percent dissected bone (P less than .01). A stepwise regression analysis was used to examine simultaneously the relative predictive value of the blood serum components. The nitrogenous compounds, which included albumin, total protein, urea and creatinine, were the most important predictors of performance (avg daily gain, feed conversion) and carcass composition. However, most of the serum components were not related to the performance and carcass composition traits.  相似文献

为探讨日粮添加葡萄渣提取物对湖羊生殖相关激素促黄体素(LH)、睾酮(T)和雌二醇(E2)分泌的影响。选择24只体重(22.74±0.23) kg相近的3月龄湖羊公羔,随机分为3组,每组8只。对照组饲喂含有4%亚麻籽油的基础日粮,T1和T2组日粮分别添加0.36%和0.72%葡萄渣提取物。试验预饲期7 d,过渡期14 d,正式期60 d。结果表明:1)添加葡萄渣提取物对宰前活重影响不显著(P>0.05);2)与对照组相比,初情期前湖羊日粮添加亚麻籽油同时添加0.36%葡萄渣提取物对睾丸重量、睾丸体积、睾丸指数和附睾重量影响不显著(P>0.05),添加0.72%的葡萄渣提取物可显著提高睾丸重量[(233.02±32.67) g vs. (347.82±31.82) g,P<0.05]、睾丸体积[(255.50±40.55) mL vs. (365.63±32.41) mL,P<0.05]、睾丸指数[(0.61±0.10) vs. (0.92±0.09), P<0.05]和附睾重量[(45.53±4.00) g vs. (54.38±4.03) g,P=0.088],且T1[(182.76±8.26) μm]、T2[(220.25±6.69) μm]组睾丸曲细精管直径显著高于对照组[(160.02±7.55) μm,P<0.05];3)与对照组相比较,T2组睾丸组织中促黄体素受体(LHR)、类固醇激素合成急性调节蛋白(StAR)基因表达显著上调(P<0.05),芳香化酶CYP19A1基因表达下调(P<0.05);4)T2组外周血液T浓度显著高于对照组[(803.22±145.74) pg·mL-1 vs. (575.09±57.58) pg·mL-1, P<0.05],但是LH浓度下降[(0.05±0.01) IU·L-1 vs. (0.11±0.03) IU·L-1, P<0.05],T1组各激素浓度与对照组相比,差异不显著(P>0.05)。综上所述,在本试验条件下,初情期前湖羊公羔日粮添加0.72%的葡萄渣提取物有利于羔羊睾丸和附睾发育。  相似文献

During the breeding season three experiments were conducted to evaluate the LH and testosterone (T) response of rams with male sexual orientation (e.g., male-oriented homosexual rams) to female sheep, to male sheep, and to treatment with LHRH. Male-oriented rams were identified through a series of sexual performance and sexual preference tests. Treatments included exposure to estrous females and to males for 15 min (Exp. 1) and exposure to estrous females and to males for 8 h (Exp. 2). Behavioral responses to stimulus animals were recorded. In Exp. 2 homosexual rams mounted males more than females (P less than .02) and exhibited more flehmen (P less than .002) and investigatory sniffs (P less than .01) when exposed to males vs females. Acts of aggression (butting the stimulus animals) did not differ by gender (P greater than .1). Flehmen and butting were positively correlated to LH secretion (P less than .02) of rams exposed to females but not to males. In Exp. 1, LH concentration determined every 15 min for 6 h was not affected (P greater than .05) by the gender of the stimulus animal. In Exp. 2, LH pulse frequency and concentration were similar (P greater than .05) by treatment. The objective of this study was to determine whether the effect of restraint stress on cortisol, LH, and testosterone varied among sexually inactive and sexually active female- and male-oriented rams, to allow differentiation among ram classes. Restraint stress or no stress was imposed on sexually inactive (n = 7) and sexually active female- (n = 17) and male-oriented (n = 6) rams in a 2 x 3 factorial arrangement. Rams were assigned to restraint or control within each classification. Rams were habituated to wearing halters and being tethered in separate pens, permitting visual, vocal, and olfactory contact with adjacent rams for 7 d before treatment. After 1 d of habituation, rams were fitted with jugular catheters that were checked twice daily for patency. For restraint stress, rams were laid on their side with their legs tied for 1 h. For no stress, rams were tethered with halters and leads, but their legs were not tied. On the treatment day, blood was collected at 30-min intervals for 3 h followed by 15-min intervals for 1 h before restraint, during 1-h restraint, and for 1 h after liberation from restraint. Then blood was collected at 30-min intervals for an additional 2 h. Blood was collected from controls at similar intervals. Control rams were isolated from stressed rams. Cortisol, LH, and testosterone were measured using RIA. Mixed model analyses with repeated measures were used on transformed data. Average prestress data were used as a covariate. Cortisol increased (P < 0.01) within 15 min after restraint and remained increased until 1.5 h after liberation from 1-h of restraint stress. In contrast, in controls cortisol remained unchanged at 5 ng/ mL. Cortisol did not differ over time among ram classes, and the treatment x ram class x time interaction was not significant. For LH and testosterone, the ram class x time interactions appeared to compromise the ability to identify differences in these hormones, indicating that they were not good endocrine candidates for methods of classifying rams. In conclusion, restraint stress increased cortisol in sexually inactive and sexually active female- and male-oriented rams alike, thus not providing a method to differentiate among ram classes.  相似文献

Fifty wethers and 51 spring-born rams were divided into five groups and slaughtered at different seasons of the year at average ages of 271, 361, 459, 557 or 652 d to determine the age and season at which differences in secondary sex characteristics could be detected. Serum testosterone concentrations and testes weights were low in January when the rams were 271 d of age and again in April at 361 d of age. By July, at 459 d of age, testosterone concentrations and testes weights had peaked and then decreased the following November at 557 d and February at 652 d. In contrast with plasma testosterone concentrations and testes weights, buckiness scores, splenius to semimembranosus or semitendinosus muscle ratios, splenius muscle weights and neck and shoulder percentages were not seasonal. All of these measures increased significantly up to July and continued to increase slowly, but not significantly, thereafter. Muscle color and texture scores and rib eye color scores tended to increase in a linear manner for both rams and wethers as age increased. Subcutaneous fat from rams was yellower and softer than that from wethers over all age groups. Ram fat firmness did not change (P greater than .05) with age, and the only significant change in ram fat color was between the groups at 271 and 361 d of age. Overall, season of year coupled with higher levels of serum testosterone was related to initial development of secondary sex characteristics in ram lambs.  相似文献

Experiments were performed to determine whether serum concentrations of total cholesterol (TC) undergo cyclic changes during the estrous cycle of dairy heifers and to assess the relationship between serum concentrations of TC and ovarian steroid hormones. The effects of a hypercholesterolemic diet upon luteal progesterone secretion also were determined. Experiment 1 involved five dairy heifers exhibiting normal estrous cycles. Serum concentrations of TC, progesterone, testosterone and estradiol-17 beta were determined in blood samples collected throughout a complete estrous cycle. A transient decline in TC was observed during the luteal phase of all heifers beginning on d 2 and reached a nadir 6 d after estrus. Highest mean concentrations of TC occurred between d -2 and +2 (96.3 +/- 8.2 mg/dl), which were markedly higher (P less than .05) than the lowest mean concentrations (76.3 +/- 10.3 mg/dl) observed on d 6. Concentrations of serum TC were negatively correlated (r = -.40; P less than .01) with progesterone between d 2 and 9. Serum TC was not correlated with testosterone or estradiol-17 beta. In Exp. 2, seven cycling Holstein heifers were fed a control diet for 70 d (Stage I), a diet containing 15% whole sunflower seed as a source of supplemental dietary lipid for 70 d (Stage II) and then the control diet for 70 d (Stage III). Postpartum, presuckle, colostrum samples were collected from 100 mares. Colostral specific gravities significantly correlated (r = 0.9) with colostral immunoglobulin (Ig)G concentrations. Foal serum IgG concentrations highly correlated (r = 0.82) with specific gravities of the colostrum each foal ingested. Eight of 48 foals (17%) had serum IgG concentrations less than 400 mg/dl. The dams of these 8 foals had colostral sp gr less than 1.06 and colostral IgG concentrations less than 3,000 mg/dl. Foals had serum IgG concentrations greater than 520 mg/dl 24 hours after parturition, when the colostral specific gravity of the dam was greater than or equal to 1.06. Effects of breed on colostral specific gravity, colostral IgG concentrations, foal serum IgG concentrations, and mare serum IgG concentrations were not significant.  相似文献

The objective was to determine effects of a single parenteral dose of melatonin on testicular blood flow indices, testicular echogenicity and plasma testosterone concentrations in rams during the physiological breeding season. We hypothesized that melatonin enhances testicular blood flow, echogenicity and plasma testosterone concentrations during the breeding season in rams. During the breeding season, 12 sexually mature Ossimi rams were randomly allocated to either a melatonin group (n = 8) that received 18 mg of melatonin in 1 ml of corn oil (injected SC) or a control group (n = 4) that received 1 ml corn oil only. Blood collection and ultrasonographic assessment of the testes and supratesticular arteries were conducted immediately before treatment (W0) and once weekly for 6 weeks after melatonin injection (W1-W6). Mean plasma testosterone concentrations were greater (p < .05; at least 1 ng/ml) in the melatonin-treated group compared to the control group from W4 to W6 after treatment. A decrease (p < .05) in both resistive index (RI) and pulsatility index (PI) began 1 week after melatonin injection (W1) and persisted until the end of the experiment, with mean RI and PI values in the melatonin group lower (p < .05) than those in the control group on W3 and W4. Furthermore, plasma testosterone concentrations in melatonin-treated rams were inversely correlated to both RI and PI (r = −.7 and −.6, respectively, p < .01). Testicular echogenicity decreased (p < .05) 1 week after melatonin injection (W1) and remained lower (p < .05) in the melatonin-treated group compared to the control group until the end of the study (W6). In conclusion, melatonin administration significantly altered testicular blood flow and echogenicity and increased plasma testosterone concentrations in Ossimi rams during the breeding season.  相似文献

The objective of this study was to identify rams exhibiting high (HP) and low (LP) levels of sexual performance and to determine whether their respective behavioral responses to ewes in estrus were related to changes in serum testosterone (T) and LH concentrations. Rams were selected on the basis of standardized serving capacity tests. Plasma T and LH concentrations in rams were measured in three experiments: 1) after 15 min of exposure to estrous ewes, 2) after an injection of 500 ng of LHRH, and 3) during an 11-h exposure to estrous ewes. During 15 min of exposure to ewes, HP rams were sexually active, whereas LP rams showed no sexual interest. Secretion of LH was similar (P greater than .05) between ram groups. Sexual arousal, copulation, and ejaculation of HP males were not related (P greater than .05) to LH secretion. Exposure to estrous ewes for 11 h, however, stimulated LH pulse frequency and elevated basal LH and T concentrations in HP but not LP rams (P less than .001). Luteinizing hormone secretion was positively correlated to the frequency of mounts (r = .19; P less than .01) and ejaculation (r = .17; P less than .03). Aggressive behavior of rams directed at ewes was negatively correlated to LH (r = -.22 P less than .003). Concentrations of LH and T after LHRH injection were similar between HP and LP rams (P greater than .05). These results show that the effects of the ewe on LH secretion of rams depend on length of the exposure period and sexual activity of the male.  相似文献

Interrelationships among concentrations and maturation of intramuscular collagen, serum concentration of hydroxyproline and testosterone and meat tenderness were determined in growing bulls and steers. Sixty-four Charolais X Angus bulls were assigned to sex treatment groups (intact or castrate) and slaughter groups (9, 12, 15 or 18 mo of age). Animals were bled at 30-min intervals via intrajugular catheters between 0600 and 1400 beginning 48 h before slaughter. Serum concentrations of testosterone were determined in each sample from bulls and from four samples from steers; serum hydroxyproline was determined in the last sample from both sexes. Testosterone mean values for the collection period were calculated. Samples of the longissimus, semitendinosus and infraspinatus muscles secured within 45 min postmortem were analyzed for intramuscular collagen concentration, percent soluble collagen and collagen thermal shrinkage temperature. Tenderness of loin steaks was determined by Warner-Bratzler shear test. Serum concentrations of hydroxyproline and testosterone were higher (P less than .01) in bulls than steers. Age effects were noted for both hydroxyproline (P less than .01) and testosterone (P less than .06). Total intramuscular collagen was greater (P less than .01) in bulls than steers and was different (P less than .01) among muscles, but the muscle differences were not uniform over all ages (P less than .05). Percent soluble collagen declined (P less than .01) with age and was different (P less than .01) among muscles. In the present study, the correlations among age, body weight, scrotal circumference (SC), semen quality and peripheral testosterone and estradiol-17beta (E(2)) concentrations were investigated in pubertal (n=5) and postpubertal (n=7) groups of Holstein bulls over a 6 week period. There were significant positive correlations (P<0.01) among age, body weight and SC in both groups, and similar significant correlations between sperm motility and SC in pubertal bulls (P<0.01) and between sperm concentration and SC in postpubertal bulls (P<0.05). The sperm motility after collection (P<0.05) and after freezing and thawing (P<0.01) of the postpubertal bulls correlated positively with the E(2) concentration. Estrogen may be important for the function of postpubertal bull testes, in which it may regulate spermatozoa motility in vivo.  相似文献

Groups of Merino wethers treated with 2 doses of zeranol (6 mg and 12 mg), or testosterone cyclopentyl propionate (150 mg) and untreated controls were grazed at 2 sites, one an oestrogenic subterranean clover (Trifolium subterraneum) pasture and the other a low oestrogen medic (Medicago truncatula) pasture. The influence of oestrogenic subterranean clover on these treatments was assessed by measuring changes in teat length, bulbourethral gland weight and pathology, bodyweight, carcase weight, dressing percentage and greasy and clean fleece weights. Teat lengths were increased by all treatments except 6 mg of zeranol where increases were not significant, and although increased by exposure to oestrogenic pasture this effect were not additive. Bulbourethral gland weights were increased by both of the zeranol treatments and by oestrogenic pasture, and these effects appeared to be additive. Differences observed histologically indicated that testosterone protected whereas zeranol exacerbated the influence of oestrogen. The bodyweights of all treated groups were heavier than the controls, but carcase weights were not significantly different. However an effect was seen in the group given the 6 mg dose of zeranol on the low oestrogen site, where the dressing percentage was significantly lower than in the control and testosterone treated groups. Differences in greasy and clean fleece weights were not significant except that the washing yield of the testosterone-treated group was significantly lower at the low oestrogen site.  相似文献

Diagnosing high intracranial pressure by clinical and diagnostic imaging is particularly challenging for chronic or slow‐growing lesions. The aim of this prospective case‐control study is to determine whether the neuroscore and brain magnetic resonance imaging (MRI) are related to the direct measurement of intracranial pressure in sheep affected by intracranial slow‐growing lesions due to chronic cerebral coenurosis (Coenurus cerebralis). Seventeen affected and 10 control sheep were included. All animals underwent a neurological examination, MRI of the brain, and direct measurement of intracranial pressure. The severity of clinical signs and MRI findings were scored. Data were statistically analyzed. The invasive intracranial pressure value was higher in affected animals. A severely altered neuroscore is related to an increased intracranial pressure beyond the normal threshold (< 0.05). The volume of the calvarium was larger in affected animals than in control animals (P = 0.0001) and was positively influenced by the presence and volume of the parasitic cyst (r = 0.7881, P < 0.01). Several degrees of deviation and deformation of both the ventricular system and brain parenchyma were detected by MRI. Subjective MRI findings were not associated with intracranial hypertension. In conclusion, this study shows that in sheep affected by slow‐growing lesions, severe alterations in the neuroscore and the results of objective MRI are related to an increased intracranial pressure beyond the normal threshold.  相似文献

Levels of serum serotonin and serum lipids--triglyceride, total cholesterol, low-density lipoprotein, high-density lipoprotein and very low-density lipoprotein, were determined in normal horses and horses diagnosed with aggression on the basis of a questionnaire survey. Blood serotonin levels in aggressive horses were found to be significantly lower than in non-aggressive horses (P < 0.01), but no association was found with respect to blood lipids.  相似文献

We hypothesized that oscillating dietary CP would improve N retention by increasing the uptake of endogenous urea N by portal drained viscera (PDV), compared with static dietary CP regimens. Chronic indwelling catheters were surgically implanted in the abdominal aorta, a mesenteric vein, a hepatic vein, and the portal vein of 18 growing Dorset x Suffolk wethers (44.6 +/- 3.6 kg of BW). Wethers had ad libitum access to the following diets in a completely randomized block design: 1) Low (9.9% CP), 2) Medium (12.5% CP), or 3) Low and High (14.2% CP) diets oscillated on a 48-h interval (Osc). Dry matter intake was greater (P = 0.04) for the Osc diet (1,313 g/d) than the Low diet (987 g/d) and was intermediate for the Medium diet (1,112 g/d). Nitrogen intake was not different between the wethers fed the Osc (25.4 g/d) and Medium diets (22.2 g/d), but was lower (P < 0.01) in wethers fed the Low diet (16.0 g/d). Wethers fed the Osc diet (6.7 g/d) retained more (P < 0.04) N than did those fed the Medium diet (4.0 g/d). Hepatic arterial blood flow was not different (P = 0.81) between wethers fed the Osc (31 L/h) or Medium diet (39 L/h) but was greater (P = 0.05) in wethers fed the Low diet (66 L/h). Net release of alpha-amino N by the PDV did not differ (P = 0.90) between the Low (37.8 mmol/h) and Medium diets (41.5 mmol/h) or between the Osc (53.0 mmol/h) and Medium diets (P = 0.29). Net PDV release of ammonia N was less (P = 0.05) for the Low diet than for the Medium diet, and this was accompanied by a similar decrease (P = 0.04) in hepatic ammonia N uptake. Urea N concentrations tended to be (P = 0.06) less in arterial, portal, and hepatic blood in wethers fed the Low diet compared with those fed the Medium diet. Wethers fed the Osc diet tended (P = 0.06) to have a greater PDV uptake of urea N than did those fed the Medium diet, but there was no difference between the Osc and Medium diets (P = 0.72) in hepatic urea N release. Net PDV uptake of glutamine tended to be greater (P < 0.07) in wethers fed the Low diet (6.7 mmol/h) than those fed the Medium diet (2.7 mmol/h). These data indicate that oscillating dietary protein may improve N retention by increasing endogenous urea N uptake by the gastrointestinal tract.  相似文献

Breeding activity (libido), testicular size, testosterone levels and seminal characteristics of 10 DLS adult rams (a population of 1/2 Dorset, 1/4 Leicester and 1/4 Suffolk selected for extended breeding season) were compared with those of 10 adult Suffolk rams. The study lasted 18 mo during which the data were recorded over 30 collection periods. All characteristics varied significantly with the season of the year, being lowest during the summer and highest during the fall. Suffolk rams had higher libido but lower testosterone levels than the DLS rams throughout the study. Testosterone increased earlier and decreased later in the breeding season in DLS compared with Suffolk rams, the interaction breed X period being significant. Testosterone concentration was highly associated with testicular size and libido, accounting for 65 and 28% of the variation in the Suffolk and 37 and 18% in the DLS rams, respectively. Seminal output and quality (motility and percentage of live spermatozoa) were highest in October and November and lowest in April and May. Rams within breeds differed significantly in the seminal characteristics studied. Results of the study showed that selecting the DLS ewes for extended breeding season may have altered the reproductive performance of the rams of that breed.  相似文献
