The ultrastructural differentiation and maturation of the neck cells and the zymogenic cells during physiological cell renewal were investigated in the abomasal oxyntic-gland region of cattle. Immature neck cells of the distal isthmus and proximal neck exhibit transitional morphology to the predominantly mucous isthmus cells. Neck cells confined to the glandular neck are characterized by bipartite peptic-cored mucous secretory granules. In a proximal-distal gradient along the neck, a progressive increase in the peptic granular component and concomitant reduction in mucous components paralleled by proliferation of the rough endoplasmic reticulum creates pre-zymogenic cells in the proximal glandular base. These, in turn, give rise to mature zymogenic cells with pure peptic secretory granules and typical zymogenic cell morphology. In the depth of the gland, older degenerative zymogenic cells are found. Variations in size and number of the zymogenic granules point to different secretory activities of the mature zymogenic-cell population of the glandular base. These results favour the conception of a zymogenic-cell lineage arising within the isthmus and passing through different developmental stages, including neck cells, during their migration down the gland.  相似文献

The chemical content of the secretion of the sheep lacrimal gland was analysed at the light and electron microscope levels by applying histochemical techniques and an ultrastructural histochemical method (periodic acid, thiocarbohydrazide and silver proteinate). Mucosubstance histochemistry demonstrated acidic glycoconjugates, mainly sulphated, in the mucous and seromucous glandular cells and in the apical portion of the cells lining the terminal ducts. Moreover, secretory granules, stained with PA-TCH-SP, showed a different localization of the reaction product. The presence of lysozyme was also found in the glandular serous cells. These histochemical studies demonstrate that the secretion of sheep lacrimal glands is mixed, having serous, mucous and seromucous components, and that an excellent correlation exists between the secretory granule substructure and glycoprotein localization.  相似文献

The effects of varying intensities of histamine stimulation on cells of the western newt fundic stomach epithelium have been studied. Oxyntic, mucous neck and surface mucous cells all secrete. In the oxyntic cell, the main effects are a movement of zymogen granules toward the apex, the discharge of their contents into the lumen and, eventually, their disappearance from the cytoplasm. Additionally, the tubular membrane elements (tubulo- vesicles) fuse with the apical plasma membrane with a corresponding reduction in their numbers and an increase in microvilli, which eventually are so numerous as to virtually occlude the lumina of gastric glands. Total histamine stimulation results in apparent damage to the oxyntic cells. Mucous neck and surface mucous cells release their mucigen droplets by exocytosis and are grossly changed in appearance also.  相似文献

The digestive system of the capybara has been investigated because of its coprofagia habits, important for their absorptive activity. These species present differences in terms of gastrointestinal morphological characters when compared with other rodents. Macroscopiclly, the stomach of the capybara is constituted of the following parts: cardiac, pyloric, body, fundic and gastric diverticulum. It presents two curvatures, one big and another small. Externally, the presence of gastric bands (tenias) is observed. With regards to the volumetric view, the gastric capacity varies from 850 to 2010 ml, with an average of 1498.57 ml. So, the stomach of this animal can be classified as a simple stomach, in the format of a curved sack and similar to an inverted letter 'J'. The gastric mucous membrane presents a surface filled by numerous tortuous gastric folds and longitudinally distributed along all its extension. The mucous tunic also possesses recesses located among the successive gastric folds, which were denoted as gastric parts with numerous openings described as gastric pits. In the cardiac part, a glandular epithelium with cardiac glands is noticed containing a lot of parietal and mucous neck cells. The fundic part, body and gastric diverticulum contain proper gastric glands with main, parietal and mucous neck cells. Finally, the pyloric part has pyloric glands with two cellular types, mucous neck and parietal cells.  相似文献

The development of the gastric gland cells in the ferret was studied with particular reference to maturation during the postnatal period. In the newborn, the glands consist of immature parietal cells with numerous mitochondria, and shallow rudimentary intracellular canaliculus with numerous microvilli, and undifferentiated cells. At one week old, mucous neck cells are present containing scattered mucin granules and the parietal cells contain tubulovesicular components. At two weeks old chief cells can be seen with large secretory granules in their basal cytoplasm. The gastric gland cells subsequently increase considerably in size, and their epithelium undergoes rapid histological maturation towards weaning.  相似文献

The histomorphological aspects as well as the histochemical content and distribution of glycoproteins (GPs) in the mucosa of the digestive tract of the white croaker Micropogonias furnieri were studied. The buccopharyngeal cavity and the esophagous showed a squamous stratified epithelium with mucous cells. The stomach presented three portions: cardias, fundus and pylorus. Tubular glands formed by a single type of gland cell were located along the cardias and fundus. Histochemical tests showed that the buccopharyngeal cavity and the esophagous presented the largest amount of the different types of mucosubstances. Both organs showed abundant secretory mucous cells that synthesize large quantities of neutral, sulphated and sialylated GPs. The surface epithelium in the cardias and fundus synthesized and secreted scarce sialylated and neutral GPs whereas the secretions of the apical surface were abundant. The pylorus secreted large amounts of neutral as well as sulphated and sialylated GPs. Gland cells secreted neutral GPs. The ultrastructural features of the gut cells were quite similar to those of other teleosts. The buccopharyngeal cavity and the esophagous surface epithelial cells, identified by their superficial localization, were characterized by cytoplasmic vesicles of different size. Abundant goblet cells with secretory mucous granules were also present. Gastric glands in the stomach contained just one form of cell with a fine structure similar to cells that secrete pepsinogen.  相似文献

Triturus torosus has a resting fundic stomach with gastric pits and glands and cell types similar to previously described amphibian species. Lining the surface and the gastric pits are columnar mucus-secreting cells. They differ in morphology and staining reactions from the mucous-neck cells, the second type of mucus-secreting cells. The latter are confined to a limited region at the necks of gastric glands where they empty into the pits and appear to “isolate” the main oxyntic, acid-secreting cells from the surface mucous cells. The gastric glands are lined by a single cell type, the oxyntic cell, which shows features of both the mammalian chief (zymogenic) and parietal (oxyntic) cells and which presumably secretes both enzymes and acid. This cell has a prominent granular endoplasmic reticulum with prezymogen and zymogen granules, numerous mitochondria, and in the apical cytoplasm an extensive system of tubules and tubulovesicles. The membrane bounding these tubules appears identical to the surface plasmalemma, and occasional continuity between the two can be demonstrated. The oxyntic cells have also a few apical microvilli and intercellular but not intracellular canaliculi. Occasional argentaffin or enterochromaffin cells with few granules are scattered among the oxyntic cells. Like other amphibia, the western newt feeds intermittently and thus is a good experimental animal for the study of gastric secretion.  相似文献

The WTC-deafness Kyoto (dfk) rat is a new mutant characterized by deafness and abnormal, imbalanced behavior. WTC-dfk rats carry an intragenic deletion at the Kcnq1 gene; KCNQ1 plays an important role in K(+) homeostasis, and the mutation of Kcnq1 causes a cardiac long QT syndrome in humans. Here, we studied stomach lesions in these WTC-dfk rats. The most characteristic pathologic feature in the stomach was the appearance of hypertrophic gastric glands in the stomach body. The hypertrophic cells had many eosinophilic granules in their cytoplasm, and these granules were stained red with Azan stain; stained positively for trypsinogen, amylase, and chymotrypsin; and did not stain positively for pepsinogen when using immunohistochemical analysis. These staining results suggested a metaplasia toward a pancreatic acinar cells. Extensive fibrosis was found in the bottom part of the mucosa of 34-week-old WTC-dfk rats, suggesting a progression of stomach lesions with aging. Although cells that were positive for proliferating cell nuclear antigen were restricted in the area of the glandular neck in WTC control rats, positive cells in WTC-dfk rats were scattered throughout the mucosa. The parietal cells in WTC-dfk rats were negative for KCNQ1 immunohistochemical analysis. These findings indicate that a deficiency in rat Kcnq1 provokes an abnormal proliferation and differentiation of gastric glandular cells.  相似文献

To accumulate histological information of cetaceans, the proper gastric gland of Minke whales was examined by light and electron microscopic observation. A small number of mucous neck cells and a large number of chief and parietal cells were observed in the gland. At the body to basal portions of the gland, the ratio of chief cells to other cells seemed to be large compared to the neck portion. Transmission electron microscopy revealed that the chief cell had secretory granules with middle level of electron density, and that the parietal cell contained abundant mitochondria and intracellular canaliculi. The proper gastric gland of the Minke whales may appear to secrete large amounts of digestive enzymes and have high digestive activity.  相似文献

One-day-old turkeys were infected intranasally with Bordetella avium, and tracheas were examined by scanning and transmission electron microscopy at 1 to 5 weeks post-inoculation (PI). The predominant ultrastructural lesions were progressive loss of ciliated epithelium with replacement by nonciliated cells, bacterial colonization of ciliated cells, membrane-bound crystalline inclusions in cytoplasma of epithelial cells, depletion of mucous granules, and distortion of tracheal rings and the mucosal surface. Tracheal surface exudates consisted of mucus, necrotic cells, heterophils, and fibrin. Ciliated cells were replaced by immature cuboidal cells characterized by abundant rough endoplasmic reticulum with small numbers of electron-dense mucous granules in the apical cytoplasm. Bacterial surfaces were rough and contained numerous pleomorphic, knob-like structures, 20-50 nm in diameter. Other changes included enlarged mucosal gland openings, cell extrusion marks, pleomorphic microvilli, and cells with small numbers of short cilia.  相似文献

This study investigated the histomorphological, ultrastructural and morphometrical postnatal developmental changes in the rabbit fundic region, especially during changing of the feeding intake. Seventy-two New Zealand rabbits (V-Line breed) at the ages of 1, 7, 15, 23, 30 and 60 days were obtained for light and electron microscopy and morphometric studies of the fundic region. The newborn rabbit's fundic wall was thin and organized into mucosa, submucosa, musclosa and serosa, with a significant increase in thickness with ageing. The fundic glands were few at the first week of life, then increased in length and diameter compared to the preceding age with prominent zonation at 23 days. The gastric pits appeared wide and deep at the first week of life then became typically narrow and shallow at the third week. The mucous cells were the main cell types lining the fundic glands in the first week of life. These cells showed remoulding with a marked increase in Periodic Acid–Schiff reactivity with age. Parietal cells were differentiated earlier (on the first day of life) than the chief cells and distributed at the neck and basal zones. Chief cells differentiated at 15 days old at the base of the glands, followed by an increase in the number and activity. Few active enteroendocrine cells were first seen at 15 days old and then widely distributed throughout the glands. Conclusion: Pronounced histomorphological changes in the fundic mucosal layer, especially the surface and glandular epithelium, correlate with the postnatal rabbit-feeding intake changes.  相似文献

The development of ectopic gastric, intestinal, or pancreatic tissue in the gastrointestinal tract is extremely rare in rats, although it is fairly common in humans. In this report, we describe an unusual case in which a mixture of different types of ectopic tissue was found in the forestomach of a rat. A solitary white nodular/polypoid structure, which measured 5 mm in size, was detected on the luminal surface of the greater curvature of the forestomach in an 8-week-old female Crl:CD(SD) rat. A histological examination revealed that the lesion contained ectopic glandular gastric tissue, including gastric surface mucous cells, parietal cells, and pyloric gland cells, which was confirmed by immunohistochemistry. Moreover, the lesion also contained villin-positive columnar intestinal absorptive cells and chymotrypsin-positive pancreatic exocrine tissue. To the best of our knowledge, this is the first study to detect a mixture of ectopic glandular gastric, intestinal, and exocrine pancreatic tissue in a rat.  相似文献

The study investigated seasonal changes in the morphology of the seminal vesicles of Eidolon helvum in a typical African tropical environment. Seminal vesicles of forty-eight bats were examined during the early rainy, late rainy and peak dry seasons, using gross anatomical, histological and ultrastructural techniques. The interlobular connective tissue septa from the external capsule demarcated the glandular parenchyma into numerous lobules of tubulo-alveolar glands. A simple cuboidal epithelium of mono- or bi-nucleated cuboidal cells lined the glandular mucosa during the rainy season. This was replaced by a bistratified cuboidal epithelium, whose upper layer of cells showed evidence of degeneration, during the dry season. The PAS-positive secretory products appeared to be numerous during the rainy season, but few and clumped together during the dry season. The mean gross weight, interlobular septal thickness, acini diameter and epithelial height of the glands varied significantly with season. Ultrastructural features of the secretory cells during the rainy season showed well-developed cytoplasmic organelles, numerous electron lucid secretory vesicles and electron-dense granules. Secretory epithelial cells of the seminal vesicles are very active during the rainy season, while regeneration of degenerated epithelium in preparation for the next reproductive cycle occurs during the dry season.  相似文献

Mast cells in the equine respiratory mucosa were studied at both light--and transmission electron--microscope levels. Mast cells were identified at all levels of the tract, with the greatest cell density in the nasopharynx. The majority (57 to 94 per cent) of this cell population were located within the connective tissue of the lamina propria. Up to 20 per cent of these cells were associated with the mucosal glandular tissue, whilst small numbers were present within the surface epithelium and in association with nodular lymphoid tissue. In the peripheral lung tissue 20 per cent of the mast cell population was associated with the small airways, 25 per cent with the pleura, and 32 per cent with blood vessels. The fine and ultrastructural features of these mast cells resemble those described in other species, and their granules consist of two types which resemble human mast cells.  相似文献

Rats heavily infected with larval Taenia taeniaeformis show hyperplasia of the gastric mucosa accompanied by mucous cell proliferation, increase in the level of intragastric pH and hypergastrinemia. Sixty one rats were divided into 2 groups designed as infected (36 rats) and control (25 rats) group. These rats were examined with time course of the infection histopathologically and physiopathologically, during 14-112 days postinfection (DPI). In the infected rats, gastric mucosal hyperplasia began to be observed at 56 DPI, and the structural disturbance of zymogenic units in the corpus and mucous units in the antrum had increased with time. However, the degree of these changes in the antrum was weaker than those in the corpus. Alcianblue and/or PAS-positive cells increased in their numbers with time, and 4 types of cells other than typical surface mucous cell and mucous neck cell were observed by electron-microscopy. However, zymogenic and parietal cells decreased in their number after 56 DPI. Further, the infected rats showed changes in the serum concentration of alanine aminotransferase, aspartate aminotransferase, blood urea nitrogen, glucose and total protein. Some similarities with Menetrier's disease were discussed.  相似文献

The stomach of the Pacific white-sided dolphin is divided into three parts: forestomach, proper gastric gland portion, and pyloric chamber. The histological features of the dolphin stomach are similar to those of terrestrial mammal stomachs, although the distribution of glycoconjugates in mucosal cells of the dolphin stomach is unknown. To learn about glycoconjugates in cetacean gastric mucosa, the glycoconjugate distribution in the mucous epithelium of the Pacific white-sided dolphin was studied using 21 lectins. Among the lectins tested, GSL-I and DBA specifically labelled the superficial layer of the forestomach epithelium. GSL-I, SBA, RCA-I, VVA, GSL-II, DSL, LEL, STL, s-WGA, WGA, PNA, and Jacalin labelled the luminal surface of the chief cells in the proper gastric gland. GSL-I, SBA, RCA-I, DSL, LEL, STL, s-WGA, PNA, and LCA labelled tubular structures in the cytoplasm of parietal cells. The surface portion of the pits in the pyloric chamber strongly reacted with RCA-I, GSL-II, WGA, PNA, LCA, PHA-L, and UEA-I, whereas the neck portion reacted weakly. Although lining one tubular portion, individual secretory cells in the pyloric gland displayed a heterogeneous reaction. This is the first report on the lectin histochemistry of a cetacean stomach and reveals GSL-I and DBA as specific marker lectins for the cornified stratified squamous epithelium cells of the Pacific white-sided dolphin. The stomachs of cetaceans and terrestrial mammals have similar histological features and mucous glycoconjugate content.  相似文献

应用透射电镜观察了恒河猴肾上腺髓质含有的 3种嗜铬细胞 ,即明嗜铬细胞、暗嗜铬细胞和小颗粒嗜铬细胞的超微结构特征。明嗜铬细胞卵圆形或略微不规则形 ,电子密度较低 ,胞核圆形 ;胞质内含有大量的分泌颗粒。分泌颗粒多为圆形 ,直径 130~ 35 0 nm,根据其超微结构 ,可分为 2型 : 型颗粒 ,电子密度高 ,核芯呈均质状 ,有的核芯偏于颗粒一侧 ; 型颗粒 ,电子密度中等或较低 ,核芯为细粒状 ,不偏位。暗嗜铬细胞多边形或不规则形 ,电子密度较高 ,胞核不规则形 ;分泌颗粒与明嗜铬细胞相似。小颗粒嗜铬细胞与明嗜铬细胞相似 ,但分泌颗粒较小 ,直径 10 0~ 2 5 0 nm。  相似文献

The development of the gastric glands of the cat (Felis silvestris catus) There are five stages in the development of the cat's gastric glands: 1. During the stage of the indifferent epithelium from day 19 to day 24, the anlage of the stomach develops with all layers; 2. The stage of gland formation from day 24 to day 41 is the beginning of the gland buds. They develop in connection with endocrine cells on day 34 into primitive oxyntic and primative mucous cells. The latter form the basis for all other cells, including the surface mucous cells; 3. During the stage of gland evagination from day 42 to 55, the anlagen are separated into primitive pits and tubules, while the cells continue to differentiate and the first intermediate cells are seen; 4. The stage of gland branching from day 56 to birth is characterized by the formation of additional glands at the bottom of the pits which change the ordinary anlagen into branched glands. During this stage, the cardiac glands are formed; 5. In the stage of gland maturation from birth to the 9th week, the peptic cells are formed and the glands start functioning. The oxyntic cells show carbonic-anhydrase activity and signs of acid secretion, and, between the weeks 4 and 8, the peptic cells contain pepsinogen, producing a negative reaction to PAS and a positive reaction to HID. Mucous cells and mucous neck cells produce PAS- and AB-positive mucin.  相似文献

The ultrastructural and cytochemical properties of peripheral blood cells of Gymnocypris eckloni were investigated by transmission electron microscopy and a range of cytochemical techniques to provide clear insight into the structure and function of blood cells from this fish. Ultrastructurally, erythrocytes, leucocytes (neutrophils, eosinophils, lymphocytes, monocytes), thrombocytes and plasma cells were identified in the peripheral blood of G. eckloni. The most special ultrastructural characteristics of blood cells in this fish were that neutrophils exhibited only one type of cytoplasmic granules containing an eccentric, spherical or oval electron‐dense core, and eosinophils presented two types of granules with non‐uniform electronic density and without crystalloids in their cytoplasm. Neutrophils, eosinophils, lymphocytes, monocytes and thrombocytes were positive for periodic acid–Schiff and α‐naphthyl acetate esterase staining. Intense peroxidase positive staining was observed in neutrophils and monocytes, but not in eosinophils, lymphocytes and thrombocytes. Neutrophils, eosinophils and monocytes were stained positively for acid phosphatase, whereas lymphocytes and thrombocytes did not stain. Leucocytes and thrombocytes were negative for alkaline phosphatase and Sudan black B staining. Erythrocytes were negative for all cytochemical staining. The cytochemical and ultrastructural features of peripheral blood cells of G. eckloni were similar to those of other fish species. However, some important differences were identified in G. eckloni.  相似文献

The microstructural and ultrastructural features of the equine lingual tonsil were studied in five young horses. Located at the root of the tongue it presented an irregular surface with rounded elevations, numerous folds and crypts. Stratified squamous non-keratinized epithelium lining its outer surface was modified by heavy infiltration of lymphoid cells to form reticular epithelium within the crypt. The latter implies a role in initiating and maintaining immune responses to incoming infectious agents and antigens. Lamellated structures resembling Hassall's corpuscle were observed towards the outer surface epithelium. Microplicae were visible by scanning electron microscopy on the surface of both the outer and reticular epithelia. No microvillus cells resembling M cells were observed. The stratum superficiale of the reticular epithelium showed strong affinity for Soybean (SBA), Phosphocarpus tetragonolobus 1 (WBA 1), Ulex europaeus (UEA) and Griffonia simplicifolia 1 isolectin-B4 (GS1-B4). The characteristic lectin binding patterns may be useful for embryological and microbiological investigations. Vimentin filaments were not detected consistent with absence of M cells. Mucus glandular acini in the deeper lamina propria mucosae contained glycogen, acidic, neutral and weakly sulphated mucopolysaccharides. Transmission electron microscopy revealed that the layers of the outer surface and reticular epithelia shared characteristic features except the stratum superficiale, which had nuclei of varying shapes and an abundance of cell organelles. A few mast cells with electron lucent granules and myelinated nerve fibres were localized in the deeper portion.  相似文献
