Lymphocytes from healthy lambs produced vigorous but variable responses to phytolectins when tested in whole blood culture. Differences between groups with low glutathione peroxidase (GSHPx) activities and groups supplemented with selenium were not significant. However, animals with low GSHPx activities and white muscle disease reacted feebly to mitogen. No correlation was found between levels of GSHPx activity and the numbers of leucocytes in the circulation.  相似文献

Crossbred wethers (n = 36; BW = 36.0 kg; SD = 3.4) were used to assess the time-dependent influence of supranutritional organically bound Se on Se accumulation. Four wethers were slaughtered before the trial began (d 0). The remaining wethers were fed diets containing adequate (0.2 microg of Se/g of DM) or supranutritional Se (2.9 microg of Se/g of DM; in the form of high-Se wheat grain) for 14, 28, 42, or 56 d before slaughter (four wethers per Se treatment at each slaughter day). The DMI was set at 3.1% of BW and adjusted weekly based on a targeted ADG of 150 g. Daily Se intake by wethers fed the adequate and supra-nutritional Se diets ranged from 5.3 to 5.9, and 79.0 to 95.0 microg of Se/kg of BW, respectively, and did not differ (P = 0.84 to 0.99) between slaughter day groups within Se treatment. Neither Se treatment nor Se treatment x slaughter day interactions were significant for BW, G:F, or liver, kidneys, and spleen weights (P = 0.06 to 0.84). Within the supranutritional Se treatment, Se contents of most organs and tissues from wethers slaughtered on d 14, 28, 42, and 56 were nearly twice the concentrations (P < 0.01) of wethers slaughtered on d 0. When regressed against the number of days the wethers were fed supranutritional Se, Se concentrations increased (P < 0.001) cubically in kidneys and plasma, quadratically in duodenum, lung, liver, and spleen, and linearly in heart, muscle, and wool. For total Se in kidneys, liver, and spleen, the response was quadratic (P < 0.03). Excluding skeletal muscle, heart, and wool, Se in other organs and tissues reached apparent steady-state concentrations 14 to 28 d after commencement of supranutritional Se diets. Selenium concentrations in skeletal muscle accumulated in a linear manner (P < 0.001) throughout the 56-d feeding period. High-Se grains can be used strategically to deliver supranutritional Se and rapidly enhance Se depots in sheep, a task that does not seem attainable with Se salts. Furthermore, a 100-g portion of uncooked loin (LM) from the wethers fed supranutritional Se contained 196 to 250% of the recommended Se requirement for humans.  相似文献

In trials conducted in 1984 and 1985, the effects of selenium injections on the antibody responses of lambs marginally deficient in selenium to Salmonella dublin were assessed. Control lambs (mean glutathione peroxidase [GSHPx] less than 20 iu/ml) responded well to the vaccine. No difference was observed in the antibody titres of animals treated twice with 5 mg selenium as sodium selenate. During the following season animals given 50 mg selenium as barium selenate showed slightly higher titres than controls. The results show that marginally selenium deficient lambs are able to elicit strong antibody titres to a bacterial antigen and that supplementation with selenium produces, at best, a marginal enhancement of the responses observed.  相似文献

The relationship between the optimum concentration of mitogen which induces lymphocyte blastogenic response and the receptor occupancy by mitogen was investigated. The receptor occupancies which induced maximal blastogenic activity in equine, bovine and canine peripheral blood lymphocytes (PBL) were 31.1 per cent, 26.5 per cent and 38.4 per cent with phytohaemagglutinin-P, and 48.2 per cent, 17.9 per cent and 24.5 per cent with concanavalin A, respectively. The data clearly show that each animal species had its own optimum concentration of mitogen for stimulation of PBL. Optimum concentration for blastogenesis and number of binding sites of each mitogen had a good correlation with each other for all three species.  相似文献

The efficacy of 100-day controlled release anthelmintic capsules containing Se for increasing and maintaining the Se status of lambs was evaluated. Capsules containing either 7.7 or 13.9 mg of Se increased the mean blood Se concentrations from 1.50 +/- 8.4 nmol/l in the unsupplemented lambs to 440 +/- 29.8 and 990 +/- 83.4 nmol/l respectively 100 days after their administration. Blood Se concentrations then declined to 250 +/- 21.5 and 380 +/- 50.1 nmol/l respectively by day 210. The addition of 13.9 mg of Se to a 100-day controlled release anthelmintic capsule is an effective method of protecting lambs from Se deficiency for at least 180 days.  相似文献

Two trials were undertaken with lambs grazing selenium deficient pasture to determine if copper would enhance liveweight and fleece-weight responses to selenium supplementation. In the first trial, lambs given selenium or selenium plus copper gained significantly more weight and had higher fleece-weights after 260 days than did control lambs or lambs given copper alone. Copper given alone or together with selenium had no significant effect on liveweight or fleece-weight when compared with control lambs and lambs that were given selenium alone respectively. This finding was confirmed in a second trial when growth and fleece-weights of selenium and selenium plus copper treated lambs were compared and no significant differences found. In both trials copper significantly raised liver copper levels. In selenium supplemented but not in selenium deficient sheep, copper significantly increased blood selenium levels.  相似文献

A bioavailability study was conducted in lambs following intravenous and oral administration of sodium selenite (0.4 mg selenium/kg body weight) with and without concurrent oral monensin. Two- or three-compartment open models with first-order absorption after oral administration adequately described plasma selenium disposition irrespective of whether monensin was being administered. No significant differences were observed between groups of lambs receiving intravenous selenium with or without monensin with respect to distribution or elimination half-lives of selenium, areas under the concentration-time curve (AUC), volumes of distribution (Vd(ss)), or clearances (Cl). In lambs given selenium per os, no significant differences were observed with animals receiving monensin as well with respect to absorption and elimination half-lives, Vd(ss), or the time at which peak selenium concentrations occurred (tmax). However, peak selenium levels (Cmax) and AUC values were significantly higher in the group given monensin. The bioavailability of selenium with and without monensin was estimated to be 60% and 43%, respectively.  相似文献

OBJECTIVE: To assess changes in serum concentrations of thyroid hormones associated with selenium deficiency myopathy in lambs. ANIMALS: 35 lambs with selenium deficiency myopathy and 30 healthy lambs. PROCEDURES: Blood samples were collected via jugular venipuncture from lambs with selenium deficiency myopathy and healthy lambs. Activities of markers of selenium deficiency myopathy (erythrocyte glutathione peroxidase [GSH-Px] and plasma creatine kinase [CK]) and serum thyroid-stimulating hormone (TSH) and total thyroxine (tT(4)) and total triiodothyronine (tT(3)) concentrations were assessed; values in affected lambs were compared with those in healthy lambs. Correlations of erythrocyte GSH-Px and plasma CK activities with serum concentrations of TSH, tT(4), and tT(3) were investigated, and the tT(3):tT(4) concentration ratio was evaluated. RESULTS: Compared with findings in healthy lambs, erythrocyte GSH-Px activity, serum tT(3) concentration, and tT(3):tT(4) concentration ratio were significantly decreased and serum concentrations of tT(4) and TSH and the activity of plasma CK were significantly increased in affected lambs. Analysis revealed a significant negative correlation in the affected group between erythrocyte GSH-Px activity and each of the following: plasma CK activity (r = -0.443), serum TSH concentration (r = -0.599), serum tT(4) concentration (r = -0.577), and serum tT(3) concentration (r = -0.621). CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that notable changes in circulating amounts of thyroid hormones develop in association with selenium deficiency in lambs. Such alterations in thyroid hormone metabolism may be involved in the high incidence of disorders, such as stillbirths and neonatal deaths, in selenium-deficient flocks.  相似文献

Lack of genetic variability and apparent susceptibility of cheetahs (Acinonyx jubatus jubatus) to coronavirus infection has lead to speculation that this species may have immune system deficits. To establish a foundation for evaluation of the immune function, cheetah peripheral blood mononuclear cells (PBM) were stimulated by a panel of six mitogens, and responses compared with those of domestic cat PBM. Individual responses in both species were variable, but evenly distributed throughout the range of stimulation for each mitogen. Proliferation by PBM from domestic cats occurred within the same range as that of the cheetahs. However, a significantly lower response to peanut agglutinin (PNA) was observed with domestic cat PBM. Although responses varied between animals, certain individual cheetahs were consistent low responders. The decreased values could not be explained by lack of IL-2 responsiveness since exogenous IL-2 significantly enhanced mitogen-stimulated proliferation in 11 of 12 cheetahs tested. The phenotypic distribution of domestic cat and cheetah lymphocyte subpopulations was similar as assessed by immunofluorescence staining for surface immunoglobulin (sIg) and cytotoxic T (Tc) cells (using a specific monoclonal antibody, FT2). Values for B cells (31.2% sIg+) and Tc (28.7% FT2+) were slightly higher in domestic cats as compared with cheetah PBM (13.3% sIg+; 19.0% FT2+). Even though no species-specific deficits were detected, a significant negative correlation between PHA-stimulated proliferation and percent FT2+ (Tc) cheetah cells was observed. This indicates that proliferation can be used indirectly to assess relative numbers of functional T helper cells in cheetahs. Our studies suggest that these aspects of the cheetah's immune system are comparable with the domestic cat, and establish a basis for in vitro assays evaluating antigen-specific responses.  相似文献

The benefits of the inclusion of cobalt and selenium supplements in anthelmintic preparations were demonstrated in a 10 week trial with cobalt- and selenium-deficient blackface wethers. The anthelmintics were based on oxfendazole and on levamisole plus oxyclozanide; three doses provided, in total, 38 mg cobalt and 7.2 or 11.3 mg selenium. Administration of the supplements prevented the weight loss and reduction in food intake observed in unsupplemented animals. Blood glutathione peroxidase activities were restored to normal and increases in serum vitamin B12 levels were observed which were consistent with the prevention of both cobalt and selenium deficiencies.  相似文献

The effect of selenium (Se) on sheep lymphocyte response to mitogens was studied. In an indoor experiment lambs were fed a basal diet containing 0.13 mg Se kg-1, and supplemented with, respectively, 0.1 or 0.5 mg Se kg-1, either as sodium selenite or as selenomethionine. Enhancement of the proliferative response of lymphocytes to phytohaemagglutinin (PHA), pokeweed mitogen (PWM) and concanavalin A was found in lambs following selenium supplementation at the lower levels. The highest dietary selenium content, however, induced decreased mitogen response. Transient increases in lymphocyte response to PHA and PWM by ewes supplemented with selenium was demonstrated in one field study and a combined effect of selenium and vitamin E was seen in another. There was no stimulatory effect on the mitogen response of lymphocytes from sheep supplemented with dietary vitamin E alone.  相似文献

The objective of these studies was to evaluate the effects of dietary restriction and Se on maternal and fetal metabolic hormones. In Exp. 1, pregnant ewe lambs (n = 32; BW = 45.6 +/- 2.3 kg) were allotted randomly to 1 of 4 treatments. Diets contained (DM basis) either no added Se (control), or supranutritional Se added as high-Se wheat at 3.0 mg/kg (Se-wheat), or sodium selenate at 3 (Se3) and 15 (Se15) mg/kg of Se. Diets (DM basis) were similar in CP (15.5%) and ME (2.68 Mcal/kg). Treatments were initiated at 50 +/- 5 d of gestation. The control, Se-wheat, Se3, and Se15 treatments provided 2.5, 75, 75, and 375 microg/kg of BW of Se, respectively. Ewe jugular blood samples were collected at 50, 64, 78, 92, 106, 120, and 134 d of gestation. Fetal serum samples were collected at necropsy on d 134. In Exp. 2, pregnant ewe lambs (n = 36; BW 53.8 +/- 1.3 kg) were allotted randomly to treatments in a 2 x 2 factorial arrangement. Factors were nutrition (control, 100% of requirements vs. restricted nutrition, 60% of control) and dietary Se (adequate Se, 6 microg/kg of BW vs. high Se, 80 microg/kg of BW). Selenium treatments were initiated 21 d before breeding, and nutritional treatments were initiated on d 64 of gestation. Diets were 16% CP and 2.12 Mcal/kg of ME (DM basis). Blood samples were collected from the ewes at 62, 76, 90, 104, 118, 132, and 135 d of gestation. Fetal blood was collected at necropsy on d 135. In Exp.1, dietary Se source and concentration had no effect (P > 0.17) on maternal and fetal serum IGF-I, triiodothyronine (T(3)), or thyroxine (T(4)) concentrations. Selenium supplementation increased (P = 0.06) the T(4):T(3) ratio vs. controls. In Exp. 2, dietary Se had no impact (P > 0.33) on main effect means for maternal and fetal serum IGF-I, T(3), or T(4) concentrations from d 62 to 132; however, at d 135, high-Se ewes had lower (P = 0.01) serum T(4) concentrations than adequate-Se ewes. A nutrition by Se interaction (P = 0.06) was detected for the T(4):T(3) ratios; ewes fed restricted and adequate-Se diets had greater (P = 0.10) T(4):T(3) ratios compared with the other treatments. Nutrient-restricted ewes had lower (P < 0.05) serum IGF-I, T(3), and T(4) concentrations. Fetal serum IGF-I concentrations were lower (P = 0.01) in restricted-vs. control-fed ewes; however, fetal T(3) and T(4) concentrations were unaffected (P > 0.13) by dietary Se or maternal plane of nutrition. These data indicate that dietary Se may alter maternal T(4):T(3) ratios. In addition, nutrient restriction during gestation reduces maternal IGF-I, T(3), and T(4) and fetal IGF-I concentrations.  相似文献

Lambs weaned at eight weeks old were compared with control lambs which remained with their dams; both groups grazed the same pasture. Weaning significantly reduced the growth rate, control lambs being, on average, 6 kg heavier than weaned lambs at 15 weeks old. When contamination of pasture with larval parasites was light, both groups of lambs suffered only modest parasitic infections. When lambs were experimentally infected with 5000 Haemonchus contortus and 10,000 Trichostrongylus colubriformis larvae at eight weeks old, the mean faecal egg count for weaned lambs was twice that for controls at 12 weeks old (P less than 0.001) and weaned lambs suffered a significantly greater decline in packed cell volume than controls over the next four weeks. Antibody responses following immunisation with either ovalbumin or Brucella abortus at four and at eight weeks old, did not differ significantly between control and weaned lambs. In contrast serum antibody responses to H contortus and T colubriformis differed significantly between the two groups, with controls responding earlier and more strongly than weaned lambs. The practical significance of these findings is that up to three months old, suckled lambs, when faced with a substantial parasite challenge, have much better prospects than weaned lambs.  相似文献

Influence of supplemental Se on humoral immune response was measured in 60 weaned beef calves with marginal blood Se status. Calves were fed a Se-deficient diet consisting of corn silage, corn grain, and soybean meal. Blood Se concentrations, primary and secondary humoral immune responses to hen egg lysozyme inoculation, and weight gain were determined in a 70-day trial. Calves fed 20 mg of Se/kg of mineral mixture ad libitum had lower antibody responses (P less than 0.02), compared with calves fed 20 mg of Se/kg of mineral mixture and given 0.1 mg of Se and 0.22 IU of vitamin E/kg of body weight, IM, or with calves fed 80, 120, 160, or 200 mg of Se/kg of mineral mixture. Calves fed 80, 120, 160, or 200 mg of Se/kg of mineral mixture had higher (P less than 0.001) blood Se concentrations on day 70, compared with calves fed 20 mg of Se/kg of mineral mixture and given 0.1 mg of Se and 0.22 IU of vitamin E/kg of body weight, IM. Selenium supplementation had no effect on weight gain.  相似文献

Objective To compare isoflurane and sevoflurane in lambs undergoing prolonged anaesthesia for spinal surgery. Study design Prospective randomised clinical study. Animals Eighteen Scottish blackface lambs 3–6 weeks of age and weighing 10–17 kg. Methods After intramuscular medetomidine, anaesthesia was induced and maintained with either isoflurane (group I) or sevoflurane (group S) delivered in oxygen. Meloxicam, morphine, a constant rate infusion of ketamine and atracurium were given intravenously (IV) during surgery. Lungs were ventilated to maintain normocapnia. with peak inspiratory pressures of 20–25 cmH2O. Ephedrine or dextran 40% was administered when mean arterial pressure (MAP) was <55 mmHg. Intrathecal morphine, and IV meloxicam and edrophonium were injected before recovery. Time to loss of palpebral reflex (TLPR) upon induction, cardiorespiratory variables, time at first swallowing and other movement, tracheal extubation, vocalisation, spontaneous head lifting (>1 minute), reunion with the ewe, and the number of MAP treatments were recorded. Statistical analysis utilised anova , Mann–Whitney, t‐test or Pearson’s correlation test as relevant. p < 0.05 was considered significant. Results End‐tidal carbon dioxide (mean ± SD) was significantly lower in group S (5.5 ± 0.6 kPa) than in group I (5.8 ± 0.5 kPa) while MAP (70 ± 11 mmHg) and diastolic arterial blood pressure (60 ± 11 mmHg) were higher in group S than in group I (65 ± 12 and 54 ± 11 mmHg, respectively). No differences were found with TLPR and MAP treatments. Time (median, range) from end of anaesthesia to ewe‐lamb reunion was briefer (p = 0.018) in group S (48, 20–63 minutes). Conclusion Isoflurane and sevoflurane are both suitable for maintaining general anaesthesia in lambs although sevoflurane, as used in this study, allows a more rapid reunion with the ewe. Clinical relevance The principal advantage of sevoflurane over isoflurane during prolonged anaesthesia in lambs is a more rapid recovery.  相似文献
