Tick-borne fever in goats caused by Ehrlichia (Cytoecetes) phagocytophila was characterised by high fever, dullness, anorexia, tachycardia and a slight to moderate inhibition of rumen motility. The animals developed a gradual decline in the total number of circulating white blood cells. There was a decrease in lymphocytes over a short period, followed by an increase. The number of neutrophils was higher on the 3rd day, causing considerable change in the lymphocyte:neutrophil ratios. The number of eosinophils increased slightly. Serum alkaline phosphatase (ALP) decreased during the febrile episodes, and a marked decline was observed in both plasma zinc and iron concentrations. Furthermore, there was a small but progressive decrease of haemoglobin and haematocrit values. Circulating endogenous pyrogen/leucocyte endogenous mediator could not be detected in plasma from febrile goats. Tick-borne fever was passively transmitted to kids with plasma obtained from these febrile animals. 相似文献

To determine whether removal of 20 mL/kg of blood (approximately 25% of blood volume) resulted in adverse physiological effects in donor horses, we removed this volume of blood from five horses and selected cardiovascular, haematological and biochemical variables measured during collection and for 31 days thereafter. We found that alteration in most variables occurred, although the changes in values usually remained within published reference ranges. Also, recovery of these alterations to pre-collection values was rapid, occurring within 24--48 h in most instances. We concluded that volumes of blood less than or equal to 20 mL/kg when collected appropriately from healthy donor horses result in no adverse acute or chronic physiological changes. These results suggest that horses undergo adequate physiological compensation when approximately 25% of blood volume is removed for the purposes of blood donation or production of plasma. 相似文献

Flurbiprofen, a potent non-steroidal anti-inflammatory and antipyretic agent, was given as an intravenous infusion (2 mg/kg) followed by a bolus injection of 1 mg/kg six hours later. After drug administration body temperature and rumen contractions were slightly depressed, whereas urea values gradually increased; serum sorbitol dehydrogenase (SDH) activity, plasma iron concentration and the number of circulating lymphocytes were significantly lower. Intravenous injection of endotoxin from Escherichia coli O111B4 (0.1 microgram/kg) caused shivering, fever, inhibition of rumen contractions, changes in heart rate, lymphopenia, neutropenia followed by neutrophylic leucocytosis, changes in urea values, hypoferraemia, hypozincaemia and a decline in serum alkaline phosphatase (ALP) activity, whereas gamma-glutamyltranspeptidase, glutamic oxalacetic transaminase, lactic dehydrogenase and SDH values were not significantly altered. Pretreatment with flurbiprofen completely abolished the febrile reactions to endotoxin. The endotoxin-induced inhibition of rumen contractions was only delayed. The drug blocked the initial tachycardia to endotoxin but did not prevent the secondary biphasic increase in heart rate. Flurbiprofen failed to modify the endotoxin-induced decrease in both plasma zinc and serum ALP activity whereas the decline in plasma iron concentration was delayed. After drug pretreatment the changes in circulating white blood cells were more pronounced. These data demonstrate that most of the haematological, blood biochemical and clinical effects of endotoxin cannot be blocked by flurbiprofen, and that these effects are not due to the increase in body temperature alone. Tolerance induced by repetitive daily intravenous administration of endotoxin resulted in an almost complete abolition of all the effects. However, the plasma iron values from tolerant goats were significantly lower than those from non-tolerant animals, which demonstrates that the development of a refractory state can result in modification of this biochemical parameter. 相似文献

Fourteen calves were used to investigate the changes from birth to 83 days of age in the concentrations of serum albumin, alkaline phosphatase, beta-hydroxybutyrate, plasma cortisol, serum creatine kinase, creatinine, iron, plasma fibrinogen, serum gamma-glutamyl transferase, plasma glucose, haptoglobin, serum non-esterified fatty acids, total protein, transferrin, triglycerides, urea and gamma globulin; the haematological variables measured were: basophils, eosinophils, haematocrit, haemoglobin, lymphocytes, mean cell haemoglobin, mean cell haemoglobin concentration, mean cell volume, monocytes, band neutrophils, neutrophils, platelets, red blood cells and white blood cells. The changes are presented as a series of graphs and the values are discussed in relation to the published reference ranges for adult cattle. Two populations of calves were identified which gave rise to a bimodal distribution for some of the variables. Differences in haematocrit, haemoglobin and red blood cell counts were apparent at birth, with raised values for these measurements being associated with an increased white blood cell and neutrophil count between three and 27 days of age. 相似文献

Ovine neutrophils were isolated and characterised by their morphology, biochemical and functional responses. Two major granule types were observed, peroxidase positive and peroxidase negative, which were identified as the ovine equivalent of the human azurophil and specific granules respectively. A third type of granule identified, which was present at low frequency and was peroxidase negative, was possibly the ovine equivalent of the bovine large granule. Superoxide production following stimulation with PMA, A23187, PAF, ConA and opsonized zymosan (ZC), was 20-50% less, compared to bovine and human neutrophils. Coincubation of PMA with either PAF or A23187 enhanced superoxide production by 4 to 5 fold above that of the latter stimulants alone. The amount of beta-glucuronidase was similar to, while myeloperoxidase was more than twice that found in bovine neutrophils. Vitamin B12 binding protein was found in very small amounts, compared to that of bovine or human neutrophils. It was observed that coincubation of PMA with PAF, or A23187 resulted in an inhibition of beta-glucuronidase secretion and an enhancement of myeloperoxidase secretion, respectively. Phagocytic capability of ovine neutrophils was found to be optimal at a neutrophil to ZC ratio of 1:10, and which corresponded with an enhanced myeloperoxidase secretion. 相似文献

An experiment was conducted to examine the effect of dietary protein on the pathophysiology iof acute haemonchosis in Finn Dorset/Dorset horn lambs. Erythrokinetic and metabolic studies using 51Cr-red cells, 59Fe-trasferrin and 125I-albumine and digestive efficiency and nitrogen balance measurements wer conducted between 19 and 39 days after infection. Identical radioisotopic and nutritional studies were conducted later on pair-fed control lambs. The results demonstrated that lambs on a law protein diet (88 g crude protein (CP) kg−1 dry matter (DM)) were less able to withstand the pathyphysiological consequences of infection with 350 Haemonchus contortus larvae kg−1 body weight than lambs given a high protein (170 g CP kg−1 DM) diet, despite similar worm budens. Erythrokinetic studies indicated that abomasal blood loss was similar in both groups of infected lambs, and circulating red cell volumes were reduced and plasma volumes were expanded to a similar degree in both groups. The majority of infected lambs in both dietary groups responded equally well to the gastric haemorrhage by increasing the rate of red cell production and their absorption of dietary iron was greater than in the control lambs. Albumin catabolism was increased in both groups of infected lambs, particularly in those given the low protein diet. Within each dietary group there were no differences in the digestibilities of the various proximate fractions of the experimental diets and no differences in the urinary of faecal nitrogen losses between infected lambs and their pair-fed control. However, loss of appetive was a major feature of the disease, particularly in the lambs given the low protein diet and this feature appeared to be crucial in determining the ability of the infected sheep to withstand the pathophysiological effects of the parasite. 相似文献

The aim of this study was to examine the coagulation profile in peste des petits ruminant (PPR) in kids. Five kids from a
group of 150 animals (72 goats and 78 kids) were brought to the Veterinary Medical Teaching Hospital from a farm in Burdur
province (Turkey) with nasal and ocular discharges and diarrhea. Fifteen goats and 41 kids had died due to diarrhea and three
kids were presented to the Department of Pathology for diagnosis. Blood samples were taken from 12 ill animals (infected group)
for haematological and biochemical analysis. In addition, five healthy kids were examined from another healthy flock (control
group). Leukocyte and lymphocyte numbers of infected group showed significant declinations in comparison to control group
(≤0.001). Haemorrhages in all organs of digestive system and small haemorrhagic areas in liver were caused to decrease in
erythrocyte and haematocrit values ( p ≤ 0.001) in infected group. Concentrations of blood urea nitrogen (BUN) ( p ≤ 0.01) and creatinine ( p ≤ 0.001) in infected group were significantly higher than control group. Compared to control group, significant increases
were determined in serum concentrations of alkaline phosphatase (ALP) ( p ≤ 0.01), aspartate aminotransferase (AST) ( p ≤ 0.001) and alanine aminotransferase (ALT) ( p ≤ 0.001) in the infected group. No significant differences were observed between the infected and control groups for serum
gamma glutamyl-transferase (GGT) concentration value. In our study, thrombocytopenia ( p ≤ 0.001) together with prolonged activated partial thromboplastin time (APTT; p ≤ 0.01) and prothrombin time (PT; p ≤ 0.001) may show that disseminated intravascular coagulopathy which can occur in kids with PPR. 相似文献

The biochemical properties of ovine placental lactogen (oPL) have been previously determined following purification, which has yielded various results. To clarify the properties of oPL prior to purification, oPL was examined in solubilized fetal cotyledonary tissue (d 100 of gestation) or conditioned culture medium by electrophoretic, immunoblotting and immunoprecipitation techniques. In cotyledonary tissue or conditioned culture medium, oPL has an apparent molecular weight (Mr) of 22,000 with an isoelectric point (pI) of 9.2. Incorporation of [3H]-glucosamine or [3H]-mannose into immunoreactive oPL could not be detected, nor did the presence of tunicamycin in explant culture medium alter the apparent Mr of oPL. In vitro translation of d 100 fetal cotyledonary mRNA, followed by immunoprecipitation, provided evidence that pre-oPL has an apparent Mr of 25,000. The size of oPL mRNA was determined to be approximately 1,350 base pairs by Northern hybridization procedures using an oligonucleotide probe which was generated from oPL amino acid sequence data. These experiments suggest that the only intracellular processing oPL undergoes is removal of a amino-terminal signal sequence. We conclude that oPL is synthesized and secreted as a single nonglycosylated-basic protein, at a time during gestation when circulating oPL is elevated. 相似文献

A significant decrease in plasma iron and increase in plasma zinc concentrations were observed. After 3 daily i.m. injections of ACTH (10 micrograms-1 day-1) decreases were seen in both serum Alkaline phosphatase (ALP) activity and plasma trace metal concentrations; heart rate was significantly higher. Intraveneous injection of E. coli endotoxin (0.1 microgram kg-1) caused shivering, fever, inhibition of rumen contractions, changes in heart rate, lymphopenia, neutropenia followed by neutrophilic leucocytosis, hypoferraemia, hypozincaemia, hypoglycaemia and a decline in serum ALP activity. ACTH, given i.m. for 3 days, reduced the febrile responses to E. coli endotoxin, modified the changes in heart rate, intensified the inhibition of rumen contractions, and induced a more marked decrease in the number of circulating neutrophils. ACTH pretreatment did not affect the endotoxin-induced decrease in blood glucose concentrations nor the drop in plasma zinc and iron values. These results suggest that glucocorticosteroids are not primarily involved in the fall in plasma iron and zinc concentrations during E. coli endotoxin-induced fever, the effects of endotoxin released glucocorticosteroids on white blood cells and blood glucose are masked by some other effect(s) of endotoxin, and in dwarf goats, ACTH has antipyretic properties without influencing normal body temperature.
