A total of 30 British and 30 Spanish Haemophilus parasuis isolates were tested for their susceptibility to 19 of the antimicrobials currently used in swine practice with a broth microdilution method in order to know the emergence of resistance against these compounds in this porcine pathogen. All the British isolates were susceptible to penicillin, ceftiofur, erythromycin, tilmicosin, enrofloxacin, and florfenicol, and most of them were susceptible to the remaining antimicrobials (the highest resistance rate found was of 20% to neomycin). In contrast, all the Spanish isolates were susceptible exclusively to florfenicol, and high proportions of resistance were encountered for penicillin, ampicillin, oxytetracycline, erythromycin, tilmicosin, tiamulin and trimethoprim+sulphamethoxazole; in addition, a bimodal or multimodal distribution, or tailing of Spanish isolates over the MIC range was observed for clindamycin, sulphonamides and tylosine tartrate, suggesting the development of acquired resistance. In addition, several multiresistance patterns were found among the Spanish isolates, 23.3% of them being resistant to at least eight antimicrobials, the same rate as that encountered for those being susceptible to all antimicrobials tested. This study showed that in general British H. parasuis isolates are susceptible to antimicrobial agents routinely used for treatment of porcine respiratory diseases; however, the Spanish isolates need a more continuous surveillance of their susceptibility patterns.  相似文献

In animal breeding programs, deoxyribonucleic acid (DNA) markers can be used to identify sires that are less susceptible to disease. These DNA markers are typically discovered in populations that display differences in susceptibility. To find those differences, it was hypothesized that sires influence their offspring responses to infection with H. parasuis. To identify differences in susceptibility, colostrum-deprived pigs derived from 6 sires were inoculated with a virulent strain of H. parasuis serovar 5. Pigs were infected at 21-d of age and euthanized 1, 2, or 3 days post-infection. Rectal temperatures, bacterial detection, clinical signs, and lesions were measured by comparing disease susceptibility in the offspring from each sire. The effect of the sire on the severity of disease in the offspring was statistically analyzed using to a 2-way ANOVA with sire and test day as fixed effects. Significant differences among sires were found for lesions, rectal temperatures from days 0-1 and 0-2 (P < 0.05) and marginal effects for clinical signs (P = 0.08). On average, the offspring of sire H94 was the most susceptible to challenge. Responses to infection were categorized to determine the clinical responses and analyzed by Chi square. Overall, 10% of all pigs infected were fully resistant to H. parasuis infection. Boar H94 didn't produce any fully resistant offspring. Differences in susceptibility to H. parasuis were observed, and the results support the hypothesis that sires influence their offspring's response to infection. Tissues from this population could be used to identify DNA markers for genetic selection of sires that produce offspring more resistant to H. parasuis infection.  相似文献

Respiratory disease and poor lamb recruitment have been identified as limiting factors for bighorn-sheep populations. Haemophilus somnus (recently reclassified as Histophilus somni) is associated with respiratory disease in American bison, domestic sheep, and cattle. It is also harbored in their reproductive tracts and has been associated with reproductive failure in domestic sheep and cattle. Therefore, reproductive tract and lung samples from bighorn sheep were evaluated for the presence of this organism. Organisms identified as H. somnus were isolated from 6 of 62 vaginal but none of 12 preputial swab samples. Antigen specific to H. somnus was detected by immunohistochemical study in 4 of 12 formalin-fixed lung tissue samples of bighorn sheep that died with evidence of pneumonia. Notably, H. somnus was found in alveolar debris in areas of inflammation. The 6 vaginal isolates and 2 H. somnus isolates previously cultured from pneumonic lungs of bighorn sheep were compared with 3 representative isolates from domestic sheep and 2 from cattle. The profiles of major outer membrane proteins and antigens for all of the isolates were predominantly similar, although differences that may be associated with the host-parasite relationship and virulence were detected. The DNA restriction fragment length profiles of the bighorn-sheep isolates had similarities not shared with the other isolates, suggesting distinct phylogenetic lines. All of the isolates had similar antimicrobial profiles, but the isolates from the bighorn sheep produced less pigment than those from the domestic livestock, and growth of the former was not enhanced by CO2. Wildlife biologists and diagnosticians should be aware of the potential of these organisms to cause disease in bighorn sheep and of growth characteristics that may hinder laboratory detection.  相似文献

H. parasuis isolates (n=112) from pigs were tested for antimicrobial susceptibility against 23 antimicrobial agents by the disk diffusion method. All isolates were sensitive to Florfenico and most strains were sensitive to Cefotaxime (103/112; 92%), Ceftazidime (99/112; 88.4%), Chloramphenicol (90/112; 80.4%) and Gentamicin (85/112; 75.9%). High resistance levels to Nalidixic acid (84.8%), TMP (67.9%) and Trimethoprim+Sulfamethoxazole (58%) were observed. Genomic DNA extracted from 52 isolates resistant to at least seven antimicrobial agents was analyzed by PFGE and 46 distinct PFGE patterns identified. Diverse variation was observed between the drug-resistant H. parasuis isolates examined, suggesting that resistance traits were acquired independently by the respective isolates.  相似文献

Clinical signs of severe bronchopneumonia, including anorexia, coughing, nasal discharge, dyspnoea, diarrhoea, distension of the neck, lethargy, recumbency, lameness preceding collapse, and death were observed among a herd of Holstein–Friesian dairy cattle. The outbreak occurred over a 30-day period, and attack and case-fatality rates were 0.4% and 50%, respectively. At necropsy, extensive consolidation in the cranioventral parts of the lungs was observed. Histologically, a severe acute bronchopneumonia with slight pleuritis was present. Both pathological and bacteriological evaluation of the lungs incriminated Histophilus somni (heavy growth). Supplementary laboratory investigations also isolated Clostridium and Klebsiella species (scanty growth) from the lungs. Histophilosis in cattle was confirmed for the first time in Nigeria.  相似文献

为弄清某规模化养猪场感染和流行的副猪嗜血杆菌(HPS)菌株的耐药性、血清型及基因型,本试验自该猪场发生浆膜炎和关节炎病猪的不同组织样品中分离到6株HPS菌株,通过细菌培养特性、生化试验和PCR扩增细菌16S rRNA基因片段并测序对分离株进行鉴定;进而鉴定分离株对不同类别抗生素的耐药性;最后对分离株进行血清分型和RAPD基因分型并分析两者的相关性。结果显示,自病猪不同组织脏器中分离鉴定了6株HPS菌株,体外培养具有典型卫星生长现象;生化试验结果符合HPS反应特性;PCR均可扩增出821 bp的HPS 16S rRNA基因片段,且序列与HPS一致。药敏试验结果显示分离株对头孢菌素类、青霉素类、氨基糖苷类、大环内酯类和四环素类抗生素敏感,而对喹诺酮类、磺胺类、林可霉素类和氯霉素类抗生素产生耐药性。琼脂扩散试验结果显示HPS分离株均为血清5型;RAPD分析显示HPS分离株与15个血清型参考菌株可分为4个基因型。本试验结果为HPS的流行病学调查及防控具有一定的借鉴价值。  相似文献

Histophilus somni (former name: Haemophilus somnus) is a Gram-negative, facultative pathogen bacterium that colonises the mucous membranes of cattle and sheep, however it was also described in American bison and bighorn sheep. It can cause local or generalised diseases and asymptomatic carriers can also occur. The presence and the etiological role of this microorganism have not been confirmed in any other domesticated species yet. The purpose of this study was to prove the presence of H. somni in goats by bacterial isolation. Nasal, vaginal or praeputial swab samples were collected from 205 goats in 10 flocks. H. somni strains were isolated from 2 out of 10 flocks; in one flock 10 H. somni strains were isolated from the genital mucosa of 17 goats, while a single H. somni strain was cultured from a vagina of 26 animals in the other flock. Partial amplification and sequencing of the 16S rRNA gene of three H. somni strains verified the identification. The comparative examination of carbon source metabolism using the Biolog Microstation ID System (Biolog, Ca) showed a close relationship of the caprine strains, while they were less related to H. somni type strain CCUG-36157 of bovine origin. H. somni strains were isolated only in the oestrus season from goat flocks with sheep contact. This is the first paper on isolation of H. somni from goats.  相似文献

The minimum inhibitory concentrations (MICs) of 18 antimicrobial agents were determined for 49 Arcanobacterium pyogenes isolates (42 bovine isolates and 7 porcine isolates). Benzylpenicillin and ampicillin were the most active antibiotics, with MIC ranges of < or = 0.0125-0.05 microgram/ml for both bovine and porcine isolates. All isolates were susceptible to penicillins and cephems. MICs for 90% of the isolates of dihydrostreptomycin, gentamicin and oxytetracycline for bovine isolates were > 100 micrograms/ml, 1.56 micrograms/ml and 25 micrograms/ml, respectively. More resistance to dihydrostreptomycin appeared among porcine isolates (85.7%) than among bovine isolates (52.4%). Resistance to gentamicin occurred in only 3 (7.1%) of the bovine isolates. Resistance to oxytetracycline also appeared more frequent among porcine isolates (85.7%) than among bovine isolates (57.1%). All bovine isolates were susceptible to erythromycin, tilmocosin and lincomycin, but two porcine isolates (28.6%) were simultaneously resistant to these antibiotics. Tiamulin was as active as tilmicosin, with an MIC for 50% of the isolates (MIC50) of 0.05 microgram/ml for both bovine and porcine isolates. The MIC50s of chloramphenicol and its derivatives florfenicol and thiamphenicol were all 1.56 micrograms/ml. The fluoroquinolones enrofloxacin and ofloxacin were not so active as penicillins and macrolides, with MIC50s of 0.78 microgram/ml and 1.56 micrograms/ml, respectively, for both bovine and porcine isolates.  相似文献

Higher MICs were observed with oxytetracycline, doxycycline, tilmicosin and thiamphenicol for porcine isolates than for bovine isolates. Enrofloxacin was the most active, with an MIC for 90% of the isolates (MIC90) of 0.05 microg/ml for both bovine and porcine isolates. Aspoxicillin exhibited the same excellent activity against penicillin-susceptible isolates as ceftiofur, with MICs ranging from < or = 0.025 to 0.1 microg/ml. Histophilus somni is a Gram-negative bacterium and member of the Pasteurellaceae that is responsible for respiratory disease and other systemic infections in cattle. One of the bacterium’s virulence factors is antigenic phase variation of its lipooligosaccharide (LOS). LOS antigenic variation may occur through variation in composition or structure of glycoses or their substitutions, such as phosphorylcholine (ChoP). However, the role of ChoP in the pathogenesis of H. somni disease has not been established. In Haemophilus influenzae ChoP on the LOS binds to platelet activating factor on epithelial cells, promoting bacterial colonization of the host upper respiratory tract. However, ChoP is not expressed in the blood as it also binds C-reactive protein, resulting in complement activation and killing of the bacteria. In order to simulate the susceptibility of calves with suppressed immunity due to stress or previous infection, calves were challenged with bovine herpes virus-1 or dexamethazone 3 days prior to challenge with H. somni. Following challenge, expression of ChoP on the LOS of 2 different H. somni strains was associated with colonization of the upper respiratory tract. In contrast, lack of ChoP expression was associated with bacteria recovered from systemic sites. Histopathology of cardiac tissue from myocarditis revealed lesions containing bacterial clusters that appeared similar to a biofilm. Furthermore, some respiratory cultures contained substantial numbers of Pasteurella multocida, which were not present on preculture screens. Subsequent biofilm experiments have shown that H. somni and P. multocida grow equally well together in a biofilm, suggesting a commensal relationship may exist between the two species. Our results also showed that ChoP contributed to, but was not required for, adhesion to respiratory epithelial cells. In conclusion, expression of ChoP on H. somni LOS contributed to colonization of the bacteria to the host upper respiratory tract, but phase variable loss of ChoP expression may help the bacteria survive systemically.  相似文献

A total of 31 isolates of Haemophilus parasuis obtained from Australian pigs were serotyped by the Kielstein-Rapp-Gabrielson scheme. The isolates were assigned to serovar 1 (1 isolate), serovar 2 (1 isolate), serovar 4 (4 isolates), serovar 5 (7 isolates), serovar 9 (2 isolates), serovar 10/7 (4 isolates), serovar 12 (1 isolate) and serovar 13 (6 isolates). The remaining 5 isolates could not be assigned to a serovar. Two different serovars (5 and 13) were detected in one herd. The only 2 isolates obtained from clinically normal pigs (from the same herd) were serovar 9. The common serovars were isolated from pigs with pneumonia as well as from pigs with conditions of the Glässer's disease type. The serological heterogeneity amongst Australian isolates of H parasuis has important implications for the use of vaccines to control Glässer's disease.  相似文献

We examined the 12 antimicrobial susceptibilities of 175 E. coli isolates from sick cattle and pigs by an agar dilution method. Resistance was found in 78.3% of isolates for oxytetracycline, 70.3% of isolates for dihydrostreptomycin, and 49.1% of isolates for ampicillin. When compared with healthy animals reported by Kijima-Tanaka et al., resistance rates for 11 antimicrobial agents were higher in sick cattle than in healthy cattle, and resistance rates for all antimicrobial agents were higher in sick pigs than in healthy pigs. Comparing cattle and pigs, resistance rates to colistin was higher in porcine isolates than in bovine isolates, but was lower in porcine isolates than in bovine isolates for cefazolin. With regard to the association of virulence factors, higher resistance rates to colistin and enrofloxacin were observed in STEC (61 strains) than in non-STEC (57 strains) among porcine isolates, while there were no significant differences in bovine isolates. In conclusion, these results can be considered helpful for adequate selection and prudent use of antimicrobial agents for several types of colibacillosis.  相似文献
