Until now, there has been no clear knowledge about the effect of dietary carnitine supplementation on lipid metabolism. Therefore, this study was conducted to investigate the effect of a dietary l -carnitine supplementation (500 mg/kg) onx the lipid metabolism of adult rats. Rats fed a hyperlipidaemic basal diet containing 15% lard and 1% cholesterol were used as an animal model. The feeding period was 6 weeks. As parameters of lipid metabolism, the concentrations of individual lipids in plasma, lipoproteins and liver and the fatty acid composition of liver and erythrocyte total lipids were determined. There were no significant differences between the control group and the group receiving the diet supplemented with carnitine on parameters of animal performance (daily body weight gains and feed conversion ratio). As expected, plasma, very low-density lipoproteins (VLDL) and liver exhibited high concentrations of cholesterol. Concentrations of triglycerides and phospholipids in plasma and individual lipoproteins as well as the concentrations of triglycerides, cholesterol and phospholipids in the liver were not significantly altered by dietary carnitine supplementation. The concentration of cholesterol in plasma and liver was increased by dietary carnitine. The fatty acid composition of liver and erythrocyte total lipids was not influenced by dietary carnitine supplementation. In conclusion, this study does not indicate a lipid-lowering effect of dietary carnitine supplementation in hyperlipidaemic rats. Probably, the essential functions of carnitine in metabolism were realized by carnitine which was synthesized endogenously.  相似文献

The objectives of this study were to survey clinics’ guidance about recommended fasting duration (FD) prior to lipoprotein analysis, and to characterize lipoprotein cholesterol and triglyceride concentrations in obese and overweight dogs categorized on the basis of the 5-point body condition score (BCS) scale. A dataset was created from lipoprotein analysis medical records of 1,538 dogs from 75 breeds in 354 clinics from 2012 to 2013. A phone survey was conducted to obtain the clinics’ FD. Two-level linear mixed-effects models were applied to the data. Over 50% of the clinics said they recommended fasting for 12 hr or more. Dogs in clinics with FD 12 hr or more had lower chylomicron triglyceride concentrations than those in clinics with FD less than 8 hr (P=0.05). Mean (± SEM) BCS at sampling was 3.7 ± 0.02. Obese and overweight dogs had higher very low density lipoprotein (VLDL) and high density lipoprotein (HDL) cholesterol and triglyceride concentrations than ideal dogs (P<0.05), but no such difference was found for low density lipoprotein cholesterol and triglyceride concentrations (P≥0.07). Across all BCS, as dog age rose from 0 to 8 years old, HDL cholesterol concentrations decreased by 13.5 mg/dl, whereas VLDL triglyceride concentrations increased by 81.7 mg/dl (P<0.05). In conclusion, FD of 8 hr or less may affect lipoprotein lipid concentrations. Obese and overweight dogs were characterized as having high VLDL and HDL cholesterol and triglyceride concentrations.  相似文献

Plasma lipoproteins were isolated from eight Thoroughbred horses and eight Shetland ponies on the basis of particle size by gel filtration chromatography and according to density using rate-zonal ultracentrifugation. Three major classes corresponding to very low density lipoproteins (VLDL), low density lipoproteins (LDL) and high density lipoproteins (HDL) were identified and characterised by their lipid and apolipoprotein compositions. The particle size distributions of each class were determined by electron microscopy and non-denaturing polyacrylamide gradient gel electrophoresis. HDL was found to dominate the equine lipoprotein spectrum, accounting for 61 per cent of the total plasma lipoprotein mass (VLDL 24 per cent, LDL 15 per cent). The VLDL class was isolated as a single population of particles that were triglyceride rich and cholesterol, phospholipid and protein poor. Equine LDL was characteristically cholesterol rich and was found to be polydisperse comprising three subfractions that were discrete with respect to particle size and lipid composition. The HDL class was composed of homogeneous particles that were typically protein rich. Apolipoprotein (apo) B was the major protein of VLDL and LDL, and presented two components on polyacrylamide gel electrophoresis with molecular weights in the region of human apoB-100 and a third in VLDL similar to that of apoB-48. ApoA-I was the predominant protein in equine HDL. Although there were no breed differences in the physical or chemical properties of each lipoprotein class, the Shetland ponies had higher plasma triglyceride and VLDL concentrations than their Thoroughbred counterparts.  相似文献

Plasma lipid concentrations, lipoprotein composition, and glucose dynamics were measured and compared between mares fed diets containing added water, corn oil (CO), refined rice bran oil (RR), or crude rice bran oil (CR) to test the hypothesis that rice bran oil lowers plasma lipid concentrations, alters lipoprotein composition, and improves insulin sensitivity in mares. Eight healthy adult mares received a basal diet fed at 1.5 times the DE requirement for maintenance and each of the four treatments according to a repeated 4 x 4 Latin square design consisting of four 5-wk feeding periods. Blood samples were collected for lipid analysis after mares were deprived of feed overnight at 0 and 5 wk. Glucose dynamics were assessed at 0 and 4 wk in fed mares by combined intravenous glucose-insulin tolerance tests. Plasma glucose and insulin concentrations were measured, and estimated values of insulin sensitivity (SI), glucose effectiveness, and net insulin response were obtained using the minimal model. Mean BW increased (P = 0.014) by 29 kg (range = 10 to 50 kg) over 5 wk. Mean plasma concentrations of NEFA, triglyceride (TG), and very low-density lipoprotein (VLDL) decreased (P < 0.001) by 55, 30, and 39%, respectively, and plasma high-density lipoprotein and total cholesterol (TC) concentrations increased (P < 0.001) by 15 and 12%, respectively, over 5 wk. Changes in plasma NEFA (r = 0.58; P < 0.001) and TC (r = 0.44; P = 0.013) concentrations were positively correlated with weight gain over 5 wk. Lipid components of VLDL decreased (P < 0.001) in abundance over 5 wk, whereas the relative protein content of VLDL increased by 39% (P < 0.001). Addition of oil to the basal diet instead of water lowered plasma NEFA and TG concentrations further (P = 0.002 and 0.020, respectively) and increased plasma TC concentrations by a greater magnitude (P = 0.072). However, only plasma TG concentrations and VLDL free cholesterol content were affected (P = 0.024 and 0.009, respectively) by the type of oil added to the diet. Mean plasma TG concentration decreased by 14.2 mg/dL over 5 wk in the CR group, which was a larger (P < 0.05) decrease than the one (-5.3 mg/dL) detected in mares that received water. Consumption of experimental diets lowered S(I), but glucose dynamics were not affected by oil supplementation. Addition of oil to the diet altered blood lipid concentrations, and supplementation with CR instead of water specifically affected plasma TG concentrations and VLDL free cholesterol content.  相似文献

1. Plasma lipoprotein metabolism and body composition in lines of chicken selected for high- and low-plasma very low density lipoprotein (VLDL) concentrations were compared to the commercial broiler (meat-type) line from which they were derived. 2. Selection for low-plasma VLDL concentration for 10 generations has reduced the rate of VLDL secretion by at least 50% in males whereas selection for high-VLDL concentration has increased the rate of VLDL secretion over 2-fold. 3. Body fat content was highly correlated with rate of secretion of plasma triglyceride-rich (TGR) lipoproteins (r = 0.88 over the three lines). However, extrapolation of the data suggests that birds secreting no TGR-lipoproteins into the plasma would still have substantial amounts of body fat. 4. Selection for high VLDL has increased the proportion of circulating VLDL-triglyceride taken up by the abdominal fat pad by over 2-fold but there was no difference between high- and low-VLDL lines in the proportion of VLDL-triglyceride taken up by tissues and oxidised to [14C]-carbon dioxide. 5. The results confirm the importance of plasma lipoprotein metabolism in determining body composition in the chicken but suggest there are limits to further reduction in body fat content by manipulation of plasma lipoprotein metabolism.  相似文献

The metabolic origins of equine hyperlipaemia were investigated by analysing the concentration and composition of plasma lipoproteins in 18 ponies with the condition. The mean concentrations of cholesterol, triglyceride and very low density lipoproteins (VLDL) were increased by 4-, 52- and 19-fold, respectively, compared with a control group of 18 healthy ponies. These increases were due to the appearance of a buoyant VLDL fraction (VLDL1) not present in healthy ponies. The mean diameter of VLDL1 particles was 44% greater than control VLDL, and the particles were enriched in triglyceride and free cholesterol and depleted of cholesteryl esters, phospholipid and protein. The apolipoprotein (apo) B-100 content of VLDL1 was reduced and the ratio of apoB-100 to apoB-48 particles was 1:1, compared with 2:1 in control VLDL. The VLDL1 was also enriched in apoE, but had normal complements of apoC-II and apoC-III. The conventional VLDL (called VLDL2), LDL and HDL fractions were moderately enriched with triglyceride, and HDL contained increased amounts of apoE, apoC-II and apoC-III. The activities of lipoprotein lipase and hepatic lipase, the enzymes responsible for the catabolism of VLDL and their remnants, were increased by 2- and 3-fold, respectively, in response to the increased concentrations of their substrates. The composition of VLDL1 suggested that the liver was maximising the secretion of triglyceride by producing larger number of VLDL particles that accommodated a greater mass of triglyceride by having apoB-48 rather than apoB-100 as their structural protein. Plasma free fatty acid (FFA) concentrations were elevated in 17 of the 18 ponies, suggesting that increased FFA flux might be the stimulus for hepatic triglyceride synthesis and VLDL secretion. We conclude that overproduction, rather than defective catabolism, of VLDL was the cause of the hyperlipidaemia and that lipid lowering agents which reduce VLDL synthesis, by decreasing adipose lipolysis and FFA flux, are candidates for the management of hyperlipaemia.  相似文献

OBJECTIVE: To examine whether obese cats, compared with lean cats, have alterations in lipoprotein metabolism that might lead to a decrease in glucose metabolism and insulin secretion. ANIMALS: 10 lean and 10 obese adults cats (5 neutered males and 5 neutered females each). PROCEDURE: Intravenous glucose tolerance tests with measurements of serum glucose, insulin, and nonesterified fatty acid (NEFA) concentrations were performed. Lipoprotein fractions were examined in serum by isopycnic density gradient ultracentrifugation. RESULTS: Obese cats had insulin resistance. Plasma triglyceride and cholesterol concentrations were significantly increased in obese cats, compared with lean cats. Very low density lipoprotein (VLDL) concentrations were increased in obese cats, compared with lean cats; however, the composition of various fractions remained unchanged between obese and lean cats, indicating greater synthesis and catabolism of VLDL in obese cats. Serum high density lipoprotein (HDL) cholesterol concentrations were increased in obese cats, compared with lean cats. Serum NEFA concentrations were only significantly different between obese and lean cats when separated by sex; obese male cats had higher baseline serum NEFA concentrations and greater NEFA suppression in response to insulin, compared with lean male cats. CONCLUSIONS AND CLINICAL RELEVANCE: Lipid metabolism changes in obese cats, compared with lean cats. The increase in VLDL turnover in obese cats might contribute to insulin resistance of glucose metabolism, whereas the increase in serum HDL cholesterol concentration might reflect a protective effect against atherosclerosis in obese cats.  相似文献

OBJECTIVE: To measure and compare concentrations of selected blood lipids before and after thyroidectomy in horses. ANIMALS: 5 healthy adult mares. PROCEDURE: Mares were confirmed to be euthyroid. Thyroidectomy was performed, and hypothyroidism was confirmed. Selected blood lipid variables were measured before hypothyroidism was induced and weekly for 4 weeks after induction. Plasma concentrations of very low-density lipoprotein (VLDL), low-density lipoprotein (LDL), serum triglyceride (TG), total cholesterol (TC), and nonesterified fatty acid (NEFA) were measured. The composition of VLDL and LDL also was examined. RESULTS: Mean plasma concentrations of VLDL and LDL increased significantly after thyroidectomy. By 4 weeks after thyroidectomy, a ninefold increase in mean plasma concentration of VLDL and a threefold increase in LDL, compared with baseline values, were detected. After thyroidectomy, mean percentage of TG in VLDL increased significantly, whereas free cholesterol and cholesterol ester content decreased. Mean percentage of TG in LDL also increased by 3 to 4 weeks after thyroidectomy. Serum concentrations of TG and TC increased, whereas serum NEFA concentration decreased. CONCLUSIONS: Hypothyroidism significantly alters blood lipid concentrations of horses. After thyroidectomy, markedly high VLDL concentration, appearance of TG-rich VLDL, increased serum concentrations of TG and TC, and decreased blood concentration of NEFA were evident. CLINICAL RELEVANCE: Examination of blood lipid concentrations of horses may be useful for detecting naturally acquired hypothyroidism.  相似文献

Influence of obesity on plasma lipid and lipoprotein concentrations in dogs   总被引:3,自引:0,他引:3  
OBJECTIVE: To determine effects of obesity and diet in dogs on plasma lipid and lipoprotein concentrations by assaying plasma leptin and ghrelin concentrations and determining total plasma cholesterol and triglyceride concentrations as well as the concentrations of cholesterol and triglycerides in various lipoprotein classes (ie, very-low-density, low-density, and high-density lipoproteins). ANIMALS: 24 Beagles; 12 lean (mean [+/- SEM] body weight, 12.7 +/- 0.7 kg) and 12 chronically obese (21.9 +/- 0.8 kg) dogs of both sexes, between 1 and 9 years old. PROCEDURES: Total plasma cholesterol and triglyceride concentrations; lipoprotein cholesterol and triglyceride concentrations; and plasma ghrelin, leptin, free fatty acids, insulin, and glucose concentrations were measured and compared between lean and obese dogs, both of which were fed a complete and balanced maintenance diet. Chronically obese dogs were subsequently fed a high-protein low-energy diet to evaluate effects of diet composition on plasma lipid and lipoprotein measurements. RESULTS: Chronic obesity resulted in a significant decrease in plasma ghrelin concentration and a significant increase in plasma leptin, cholesterol, and triglyceride concentrations in dogs. High total plasma cholesterol and triglyceride concentrations resulted from increased cholesterol and triglyceride concentrations in all lipoprotein fractions. In obese dogs, modification of diet composition resulted in beneficial effects on plasma lipid and leptin concentrations, even before weight loss was observed. CONCLUSIONS AND CLINICAL RELEVANCE: Correlations exist between obesity and plasma measurements (ie, lipoproteins, leptin, insulin, and ghrelin) commonly associated with obesity. Modification of diet composition to control energy intake improves plasma lipid and leptin concentrations in obese dogs.  相似文献

对708只(公、母各半)5~9周龄黄羽肉鸡进行3个代谢能水平(13.4,12.5,11.6MJ/kg)和3个粗蛋白水平(22.4%,19.4%,16.4%)交叉的饲养试验,以探讨日粮代谢能和粗蛋白水平时黄羽肉鸡腹脂和血脂水平的影响。日粮代谢能和粗蛋白水平对腹脂有明显影响,腹脂随日粮代谢能水平的升高而升高(P<0,01)。随粗蛋白水平的升高而降低(P<0.05)。日粮能量和蛋白水平对板油的影响与腹脂相似。日粮代谢能水平对肌胃脂无明显影响;日粮粗蛋白水平对肌胃脂重无明显影响,但对肌胃脂率有一定影响(P=0.07)。日粮粗蛋白和代谢能水平对血浆极低密度脂蛋白(VLDL)、甘油三酯、胆固醇及总脂浓度均有显著影响(P<8.05),8周龄血浆VLDL浓度随日粮代谢能的升高而降低(P<0.0001);血浆总脂和胆固醇浓度均随日粮代谢能、粗蛋白水平的升高而降低(P≤0.001)。肉鸡8周龄血浆VLDL水平明显存在日粮能量和蛋白的互作效应(P<0.01),而血浆甘油三酯、胆固醇和总脂水平不存在明显的能量和蛋白的互作(P>0.05)。  相似文献

Hyperinsulinaemia is a commonly-observed characteristic of insulin resistance, and a reduction in insulin sensitivity is thought to be either a causative and/or symptomatic feature of equine hyperlipaemia. A positive correlation (r=0·545, P=0·0015) existed between plasma insulin and triglyceride concentrations determined in 31 donkeys with naturally occurring hyperlipidaemia/hyperlipaemia. Greater insulin values tended to occur in the animals with an overweight body score. Inter-animal variation in insulin concentrations, however, prevented the identification of any differences either within hypertriglyceridaemic donkeys (when classified by clinical condition, date of arrival to a sanctuary and eventual outcome after treatment) or between groups of normotriglyceridaemic (n=6) and experimentally fasted hypertriglyceridaemic (n=5) donkeys. Determination of basal plasma insulin concentrations may not provide an accurate assessment of underlying insulin sensitivity. Alternatively, hyperinsulinaemia may be evident only in animals with established insulin resistance.  相似文献

1. The relationship between plasma triglyceride concentrations and body fat content was examined in three groups of male and one of female broiler chickens fed ad libitum on a normal growing diet.

2. Phenotypic correlations between total plasma triglyceride concentration and fat content were + 0.50, + 0.37 and + 0.39 for the three groups of male broilers and +0.33 for the group of females. Triglyceride content of plasma very low density lipoprotein (VLDL) plus low density lipoprotein (LDL) showed similar correlations with fat content.

3. Selection of birds from a flock of 550 male broilers on the basis of high or low plasma VLDL plus LDL triglyceride concentrations produced groups with significant differences in body fat content but little difference in body weight.

4. The results suggest that measurements of plasma triglyceride concentration could be used as an indirect means of selecting for decreased body fat content in broiler breeding programmes.  相似文献

OBJECTIVE: To determine effects of age and sex on plasma lipid and lipoprotein metabolism in cats. ANIMALS: 33 kittens and 16 adolescent, 23 adult, and 10 senior cats. PROCEDURE: Plasma concentrations of cholesterol, triglyceride, and lipoprotein-cholesterol and activities of lipoprotein lipase, hepatic lipase, and lecithin:cholesterol acyl transferase (LCAT) were measured and compared within and among groups. RESULTS: Plasma cholesterol and triglyceride concentrations were significantly higher in 5- and 7-week-old kittens, compared with the same kittens after weaning and cats in the other age groups. Cholesterol concentration was significantly less in 20-week-old kittens, compared with adolescent and adult cats. Lipid and lipoprotein-cholesterol concentrations were not significantly different among the adolescent, adult, and senior groups, nor did sex influence lipid and lipoprotein-cholesterol concentrations in these groups. Activities of lipoprotein and hepatic lipases were significantly less in senior cats, compared with the other groups. Activity of LCAT was highest in 20-week-old kittens and was greater in sexually intact adult and adolescent females, compared with their male counterparts. After castration, activities of hepatic lipase and LCAT significantly decreased in adolescent male cats. CONCLUSIONS AND CLINICAL RELEVANCE: The upper limits of reference ranges for plasma cholesterol and triglyceride concentrations should be increased for kittens < 8 weeks of age. Low cholesterol concentrations in adolescent cats likely reflect high tissue demands for growth and steroidogenesis. Decrease in lipoprotein and hepatic lipase activity in senior cats could predispose this age group to hypertriglyceridemia, particularly in insulin-resistant cats or those fed a high fat diet.  相似文献

OBJECTIVE: To measure and compare the concentration and composition of very low-density lipoprotein (VLDL) in plasma and selected lipids in serum of horses fed mixed grass hay ad libitum or denied feed for 36 hours. ANIMALS: 4 healthy adult mares. PROCEDURE: Mares were either fed mixed grass hay ad libitum or denied feed for 36 hours beginning at 8:00 AM. Blood samples were collected every 2 hours during the study period and analyzed for nonesterified fatty acid (NEFA), triglyceride (TG), VLDL, and glucose concentrations and composition of VLDL. RESULTS: Withholding feed significantly increased mean serum concentrations of NEFA. By 36 hours, a 16-fold increase in mean serum NEFA concentration and 2-fold increase in mean plasma VLDL concentration, compared with baseline values, were detected. Mean plasma TG concentrations significantly increased with time in feed-deprived horses. Significantly lower overall mean plasma glucose concentrations were detected in feed-deprived horses. Mean percentage of protein in VLDL was significantly lower in feed-deprived horses. Plasma VLDL concentrations varied widely among horses in response to withholding feed. Plasma TG and VLDL concentrations remained unaltered in 2 horses. CONCLUSIONS AND CLINICAL RELEVANCE: Withholding feed significantly increases blood lipid concentrations in horses, but individual horses respond differently. Serum NEFA concentrations were increased in all 4 horses denied feed, indicating mobilization of tissue triglyceride stores. Variation in plasma VLDL concentration in response to withholding feed suggests that its metabolism is strongly influenced by other, as yet undetermined, factors in horses. Differences in the plasma VLDL concentrations among horses in response to withholding feed may be used as an indication of susceptibility to the hyperlipemic syndrome of Equidae.  相似文献

Blood samples were obtained from 12 Iranian fat-tailed sheep during 7 weeks pre-partum, at parturition and 7 weeks post-partum. The lipids measured were cholesterol, triglyceride, total lipid, high-density lipoprotein (HDL)-cholesterol, low-density lipoprotein (LDL)-cholesterol, and very low-density lipoprotein (VLDL)-cholesterol. The concentrations of cholesterol, triglyceride, HDL-cholesterol and VLDL-cholesterol during the 7 weeks pre-partum, at parturition and the 7 weeks post-partum were significantly different (P < 0.05). One week before parturition, the concentrations of cholesterol, triglyceride, HDL-cholesterol and VLDL-cholesterol were higher (P < 0.05) than at other periods. The lowest concentrations of these parameters were observed 2-3 weeks after parturition. In this study, significant positive correlations were observed between the time of sampling (pre-partum, parturition and post-partum) and serum cholesterol (r = 0.22; P < 0.01) and HDL-cholesterol (r = 0.25; P < 0.01).  相似文献

BACKGROUND: Lipid transport systems in animals have been evaluated both as experimental models for lipid and lipoprotein metabolism in humans and to gain insight into the lipid metabolism of specific breeds. The Turkman horse is a pure-blooded horse that lives on the margins of the Caspian Basin; information about serum lipids and lipoproteins in this breed is lacking. OBJECTIVE: The purpose of this study was to evaluate serum lipid and lipoprotein concentrations in clinically healthy Turkman horses. METHODS: Concentrations of triglyceride (TG), total cholesterol (TC), high density lipoprotein cholesterol (HDLC), low density lipoprotein cholesterol (LDL-C), and very low density lipoprotein cholesterol (VLDL-C), and lipoprotein electrophoretic patterns were determined in serum samples from 48 Turkman horses of both sexes and 0-12 years of age. Results were analyzed by 1-way ANOVA. RESULTS: In both sexes and all age groups, the relative percentages of LDL-C (44.2-64.3%) were higher than those of HDL-C (29.7-47.1%) and VLDL-C (5.9-9.0%). Serum concentrations of TG and TC were significantly higher in male (0.45 +/- 0.12 mmol/L; 2.70 +/- 0.45 mmol/L) than in female (0.34 +/- 0.1 mmol/L; 2.15 +/- 0.3 mmol/L) horses in most age groups. The relative percentages of alpha- and beta-lipoproteins in electrophoretic tracings were 62.77 +/- 5.05% and 36.67 +/- 4.29%, respectively. CONCLUSION: Serum lipid and lipoprotein values in Turkman horses differ from those of other equine breeds, and may be useful for evaluating metabolic diseases in this species.  相似文献

We evaluated the effects of supplementing betaine on growth performance, fat deposition and lipid metabolism status in broilers kept under chronic heat stress. Five hundred and forty chicks were randomly divided into six groups and the two normal temperature groups were held at normal ambient temperature and fed the basal diet (CONT) and basal diet plus 0.1% betaine, respectively. Heat stressed (HS) broilers were held at 32 ± 1°C from days 22 to 42 and fed the basal diet containing variable levels of betaine. Broilers were examined at days 28, 35 and 42 for body weight, feed consumption, fat deposition and serum lipids. The CONT and betaine‐supplemented groups showed higher (P < 0.01 or P < 0.05) feed consumption, body weight gain, and lower feed : gain ratio compared with the HS‐CONT group. Meanwhile, heat stress increased abdominal, intermuscular and subcutaneous fat deposition, whereas the supplemental betaine significantly decreased those compared with the HS‐CONT group. Additionally, betaine supplementation significantly decreased triglyceride, free fatty acids, low‐density lipoprotein cholesterol and high‐density lipoprotein cholesterol compared with HS‐CONT. This study was designed to compare the effects of a fat-supplemented diet versus a traditional diet on the lipoprotein (LP) content in horses. In the first of two trials, eight two-year old horses were fed a basal diet, in which 80% of the digestible energy was supplied as chopped alfalfa hay. One group of four horses (fat-supplemented group, FS) was fed the remaining 20% of the digestible energy as corn oil, while the other group of four horses (control) was fed rolled corn to complete their diet. Blood samples were collected at the start of the experiment and every 2.5 weeks thereafter for 10 weeks. Total serum lipids were measured in both groups of horses and the lipoproteins were fractionated into very low density LP (VLDL), low density LP (LDL), and high density LP (HDL) using ultracentrifugation and agarose-column chromatography. Each LP fraction was measured for protein, cholesterol (CH) and triglyceride TG) content. Total serum lipids were increased in the FS group above the control (9.16 vs. 4.65 mg/ml, week 5). Serum cholesterol and triglyceride concentrations were elevated in the FS group, but were highly variable. Variations in lipid concentrations may have been due to variation in time of sampling during the day. In the second trial, four of the eight horses were used and divided into the same two groups; FS vs. control. After an initial sample, postprandial serial blood samples (1 hour intervals for 8 hours) were drawn at 2, 4 and 6 weeks. Upon examination of the data, Hour 3 post-feeding was chosen to represent postprandial LP values. In both trials, the horses were able to adapt to the added dietary fat. Maintenance of body weight and increased speed of lipid clearance from the blood by the end of each trial in the FS group support this statement. In the FS group, there was an increase in VLDL TG concentration, but not in LDL. This indicates an increase in VLDL TG clearance from the circulation, presumably by increased lipoprotein lipase (LPL) activity. Cholesterol concentrations were also increased in the FS horses in the LDL and HDL. The rise in cholesterol may be attributed to endogenous recycling of liver products such as bile salts, which aid in digestion and absorption and are cholesterol based. In addition, there were greater quantities of LDL and HOL produced in the FS horses as supported by the increased protein concentrations as well as larger peaks for the eluate from the gel filtration column.  相似文献

OBJECTIVE: To determine effects of dietary lipid and protein on plasma lipoprotein and free fatty acid concentrations and hepatic fatty acid synthesis during weight gain and rapid weight loss in cats. ANIMALS: 24 ovariohysterectomized cats. PROCEDURE: Cats were fed a high energy diet until they gained 30% of their ideal body weight and then randomly assigned to receive 1 of 4 weight reduction diets (6 cats/diet) at 25% of maintenance energy requirements. Diets contained a low or high quality protein source and a lipid source deficient or sufficient in long chain essential fatty acids. Plasma samples and liver biopsy specimens were obtained before and after weight gain and during and after weight loss for determination of free fatty acid, triglyceride, and lipoprotein concentrations. Synthesis of these substances was measured by use of isotope enrichment. RESULTS: Plasma total cholesterol concentration and concentration of lipoprotein fractions increased after weight gain, compared with baseline values. Weight loss resulted in a significant decrease in concentrations of all lipoprotein fractions except high density lipoprotein. High density lipoprotein concentration was significantly greater in cats fed diets containing an oil blend, compared with cats fed diets containing corn oil. Fatty acid synthesis after weight loss was below the detection limit of the measurement technique. CONCLUSIONS AND CLINICAL RELEVANCE: In cats undergoing rapid weight loss there is neither increased triglyceride synthesis nor decreased transport of very low density lipoproteins from the liver, suggesting that their involvement in the development of hepatic lipidosis may be minimal.  相似文献
